The induction of parthenocarpic fruit set was investigated using the apple cvs. Golden Delicious and Jonagold. The gibberellins GA3, GA4, GA5 and GA7 and the synthetic phenylurea-type cytokinin CPPU (N-(2-chloro-4-pyridyl)-N-phenylurea), were applied alone and in combination to unpollinated flowers at the end of petal fall. Gibberellins induced only a marginal final set of parthenocarpic fruits. CPPU sprays were more effective, particularly in the first year. When applied in combination, CPPU and gibberellins had a positive synergistic effect on parthenocarpic fruit set and fruit size, but a negative effect on flower induction the next year. After CPPU + GA sprays, percent fruit set was similar, or greater, compared to natural pollinated trees. The parthenocarpic fruits induced by CPPU + GA had an increased length to diameter ratio. CPPU stimulated, and GA4 and GA7 reduced, the russeting of the parthenocarpic fruits. The internal quality of the fruits was hardly affected, but Ca-deficiency symptoms occurred more frequently in parthenocarpic fruits.  相似文献

In maturing fruits ofPhaseolus coccineus a soluble glucosyltransferase activity occurs which converts gibberellins into their O-glucosides. The enzyme glucosylates GA3 and structurally closely related gibberellins (GA7 and GA30) to their 3-O-glucosides by transfer of glucose preferentially from UDP-glucose. From cell suspension cultures ofLycopersicon peruvianum cytosolic glucosyltransferases were isolated which in the presence of UDP-glucose converted GA7 and GA9 to the corresponding glucosyl esters. In both cases numerous other gibberellins failed to serve as substrates. Thus, the enzymes are UDP-glucose: gibberellin glucosyltransferases of considerable substrate specificity.  相似文献

Following harvest, Kinnow mandarin (Citrus nobilis × Citrus deliciosa) fruits were variously treated with gibberellins (GA) and cytokinins. Ethylene caused marked chlorophyll (Chl) degradation and its effect was partially reversed by kinetin and benzylaminopurine (BAP) and to a lesser extent by GA3 and GA4+7. No appreciable accumulation of carotenoids (Car) occurred in these fruits irrespective of treatment. The loss of Chl during natural maturation was significantly reduced by cytokinins. Treatments with gibberellins alone or in combination with cytokinins were much less effective. The reduced loss of Chl in response to exogenously applied cytokinins may be probably related to a decline in its endogenous levels.  相似文献

Growth Regulators and the Phytochrome-Mediated Dormancy of Celery Seeds   总被引:2,自引:0,他引:2  
Seeds of five celery (Apium graveolens L.) cultivars germinated at 15°C in the light or dark but at 22°C only in the light. This light requirement was overcome by treatment with a mixture of the gibberellins GA4 and GA7 (GA4/7) but interactions of cytokinins, daminozide, ethephon, EDTA and N-phenyl-N′-4-pyridylurea (NC5392) with GA4/7 were observed. Varietal differences in response to GA4/7 concentration and the requirement for cytokinins were related to the upper temperature limits for germination of the different cultivars. Seeds of cultivars responding to low concentrations of GA4/7 appeared to contain less natural inhibitor than those requiring either high concentrations of GA4/7 or cytokinin in addition to low GA4/7. The cytokinin requirement for germination was partially removed by leaching the seeds with water. Interaction studies with applied hormones indicated that in seeds incubated in the light inhibition by abscisic acid was partially alleviated by N6-benzyladenine but not by GA4/7 application. The implications of these results are discussed in relation to the involvement of natural plant hormones in the dormancy mechanism of celery seeds.  相似文献

Seed dormancy of a highly-dormant cultivar of celery (Apium graveolens L.) was broken by combinations of plant-derived smoke extract or N6-benzyladenine (BA) and gibberellins A4/7 (GA4/7) in the dark at temperatures between 18 and 26°C. A less dormant cultivar which responded to GA4/7 alone showed no additional response to smoke extract or BA. Neither smoke extract nor BA affected either cultivar in the dark in the absence of GA4/7. The partial dormancy-breaking effect of short exposures to red-light was also enhanced by smoke extracts in this highly-dormant cultivar. Imbibition of celery (Apium graveolens L.) seeds at 32°C for up to 96 h lowered the upper temperature limit for germination. If this high temperature treatment was given in the light, these seeds germinated slightly earlier than those treated in the dark although the final percentage germination was similar for both treatments. The inhibitory effect of the high temperature treatment was completely removed by allowing the seeds to imbibe in a mixture of the gibberellins A4 and A7 (GA4/7) and partially removed by the cytokinin N6-benzylaminopurine (BA). GA4/7 was less effective when added before rather than after the high temperature treatment, whereas the opposite was true of BA. At constant temperatures more GA4/7 was required to promote germination as the temperature was raised but addition of BA reduced the concentration of GA4/7 required. A model is proposed for the control of celery seed germination by light and temperature through the action of endogenous cytokinins and gibberellins.  相似文献

Satsuma (Citrus unshiu [Mak] Marc.) and Clementine (Citrus reticulata [Hort.] Ex. Tanaka, cv Oroval) are two species of seedless mandarins differing in their tendency to develop parthenocarpic fruits. Satsuma is a male-sterile cultivar that shows a high degree of natural parthenocarpy and a high fruit set. Seedless Clementine varieties are self-incompatible, and in the absence of cross-pollination show a very low ability to set fruit. The gibberellins (GAs) GA53, putative 17-OH-GA53, GA44, GA17, GA19, GA20, GA29, GA1, 3-epi-GA1, GA8, GA24, GA9, and GA4 have been identified from developing fruits of both species by full-scan combined gas chromatography-mass spectrometry. Using selected ion monitoring with [2H2]- and [13C]-labeled internal standards, the levels of GA53, GA44, GA19, GA20, GA1, GA8, GA4, and GA9 were determined in developing ovaries at anthesis and 7 days before and after anthesis, from both species. Except for GA8, levels of the 13-hydroxy-GAs were higher in Satsuma than in Clementine, and these differences were more prominent for developing young fruits. At petal fall, Satsuma had, on a nanograms per gram dry weight basis, higher levels of GA53 (10.4x), GA44 (13.9x), GA19 (3.0x), GA20 (11.2x), and GA1 (2.0x). By contrast, levels of GA8 were always higher in Clementine, whereas levels of GA4 did not differ greatly. Levels of GA9 were very low in both species. At petal fall, fruitlets of Satsuma and Clementine contained 65 and 13 picograms of GA1, respectively. At this time, the application of 25 micrograms of paclobutrazol to fruits increased fruit abscission in both varieties. This effect was reversed by the simultaneous applications of 1 microgram of GA3. GA3 alone improved the set in Clementine (13x), but had little influence on Satsuma. Thus, seedless fruits of the self-incompatible Clementine mandarin may not have adequate GA levels for fruit set. Collectively, these results suggest that endogenous GA content in developing ovaries is the limiting factor controlling the parthenocarpic development of the fruits.  相似文献

The radio-labeled gibberellins GA1, GA3,GA4, and GA7 were applied to intact developing applefruits (Malus domestica Borkh. cv. Jonagold) during theperiod when GAs are suggested to inhibit flower bud induction for the followingyear. Radioactivity from these compounds was found to be transported intoadjacent tissues as there are pedicels and bourses (4%). Application topedicels, after removal of the fruits, enhanced the transport into adjacentbourses up to 11%. The bud-carrying lateral bourse shoots contained onlyminor amounts of radioactivity on average 0.4% in both cases. Theseexport rates were identical, 1 or 5 days after application.After application of the corresponding deuterium-labeled GAs and analyses bymass spectrometry the specific metabolization of GA1 toGA1 13-O-glucoside and of GA3 to GA313-O-glucoside was demonstrated. Additional metabolites of GA1 andGA3 were not detected. After fruit application of GA3 theratio of GA3 to GA3 13-O-glucoside was found to be 1:2 inthe fruit. Pedicel application led to ratios of 1:4 and 1:5, respectively, inthe pedicel and in the adjacent bourse. After the application of GA4and GA7, neither glucosylation products nor other GA-like metabolitescould be identified.This is the first report of the metabolism of GAs to GA 13-O-glucosides indeveloping apple fruits. The possible function of the GAs as a signal in flowerbud formation for the following year is discussed.  相似文献

Although fruit set and development are induced by applications of gibberellins, final fruit weight of gibberellin-induced parthenocarpic fruit is often less than that of pollinated fruit. We examined changes in the activities of sucrose-metabolizing enzymes and sugar accumulation in developing fruits of cultivated blueberry (Vaccinium ashei Reade) and their correlation with fruit growth upon pollination or exogenous applications of gibberellic acid (GA3). The objective was to determine if differences in fruit growth could be attributed to differences in enzyme activities and subsequent sugar accumulation in fruits. The fruit development period of GA3-treated fruits was 15 days longer than that of pollinated fruits. At maturity, GA3-treated fruit accumulated an average of 180 mg dry weight while pollinated fruit accumulated 390 mg dry weight. Dry weight accumulation in nonpollinated fruits was negligible and these fruits abscised by 45 days after bloom (DAB). The total carbon (C) cost (dry weight C + respiratory C) for fruit development was 109 and 244 mg C fruit-1 for GA3-treated and pollinated fruits, respectively. Hexose concentration increased to 100 mg (g fresh weight)-1 at ripening in both GA3-treated and pollinated fruits. Nonpollinated fruits reached a maximum hexose concentration at 45 DAB. Sucrose phosphate synthase (EC 2.4.1.14) and sucrose synthase (EC 2.4.1.13) activities reached a maximum of ≤5.0 μmol (g fresh weight)-1 h-1 in both GA3-treated and pollinated fruits. Soluble acid invertase (EC 3.2.1.26) activity increased to about 60 μmol (g fresh weight)-1 h-1 in both GA3-treated and pollinated fruits at ripening, while in nonpollinated fruits, a maximum soluble acid invertase activity of 0.12 μmol (g fresh weight)-1 h-1 was measured at 24 DAB. Insoluble acid invertase activity declined during the early stages of fruit growth and remained relatively low throughout fruit development. Neutral invertase activity was low throughout development, increasing to 5 μmol (g fresh weight)-1 h-1 at ripening in GA3-treated and pollinated fruits. Our studies demonstrate that blueberry fruit development does not appear to be limited by sucrose metabolizing enzyme activity and/or the ability to accumulate sugars in either GA3-treated or pollinated fruits.  相似文献

During the overall germination process in the light-requiring seeds of Chenopodium album L. two sites of hormonal action can be distinguished. The start of visible growth is induced in darkness by the gibberellins 4 and 7 (GA4+7) or by ethylene. Added cytokinins are inactive although 8-14C-kinetin shows rapid penetration into the seed and rapid turnover. The induction of growth by GA4+7 and the uptake of 8-methylene-14C-GA3 are enhanced at lower pH values. Prolonged incubation in darkness results in a decreased responsiveness of the seeds towards GA4+7 and ethylene. A second site of hormonal action is located during the progress of growth inside the covering structures. ABA inhibits radicle growth, while GA4+7, GA3, kinetin, zeatin and ethylene reduce the inhibition.  相似文献

To interpret the metabolism of radiolabeled gibberellins A12-aldehyde and A12 in shoots of pea (Pisum sativum L.), the identity of the radiolabeled peaks has to be determined and the endogenous presence of the gibberellins demonstrated. High specific activity [14C]GA12 and [14C]GA12-aldehyde were synthesized using a pumpkin endosperm enzyme preparation, and purified by high performance liquid chromatography (HPLC). [14C]GA12 was supplied to upper shoots of pea, line G2, to produce radiolabeled metabolites on the 13-OH pathway. Endogenous compounds copurifying with the [14C]GAs on HPLC were analyzed by gas chromatography-mass spectrometry. The endogenous presence of GA53, GA44, GA19 and GA20 was demonstrated and their HPLC peak identity ascertained. The 14C was progressively diluted in GAs further down the pathway, proportional to the levels found in the tissue and inversely proportional to the speed of metabolism, ranging from 63% in GA53 to 4% in GA20. Calculated levels of GA20, GA19, GA44, and GA53 were 42, 8, 10, and 0.5 nanograms/gram, respectively.  相似文献

The level of gibberellin(GA)-like material in cotyledons of soybean (Glycine max L.) was highest at mid-pod fill—about 10 nanograms GA3 equivalents per gram fresh weight of tissue, assayed in the immersion dwarf rice bioassay. This amount is about 1000-fold less than levels in Pisum and Phaseolus seed, other legume species whose spectrum of endogenous gibberellins (GAs) is well known. The metabolism of [14C]-GA12-7-aldehyde (GA12ald)—the universal GA precursor—by intact, mid-pod-fill, soybean cotyledons and their cell-free extracts was investigated. In 4 hours, extracts converted GA12ald to two products—[14C]GA12 (42% yield) and [14C]GA15 (7%). Within 5 minutes, intact embryos converted GA12ald to [14C]GA12 and [14C]GA15 in 15% yield; 4 hour incubations afforded at least 22 products (96% total yield). The putative [14C]GA12 was identified as a product of [14C]GA12ald metabolism on the basis of co-chromatography with authentic GA12 on a series of reversed and normal phase high pressure liquid chromatography (HPLC) and thin-layer chromatography (TLC) systems, and by a dual feed of the putative [14C]GA12 and authentic [14C]GA12 to cotyledons of both peas and soybeans. The [14C]GA15 was identified as a metabolite of [14C]GA12ald by capillary gas chromatography (GC)-mass-spectrometry-selected ion monitoring, GC-radiocounting, HPLC, and TLC. By adding the [14C] metabolites of [14C]GA12ald to a different and larger extract (about 0.2 kg fresh weight of soybean reproductive tissue) and purifying endogenous substances co-chromatographing with these metabolites, at least two GA-like substances were obtained and one identified as GA7 by GC-mass spectrometry. Since [14C]GA9 was not found as a [14C]metabolite of [14C]GA12ald, soybean embryos might have a pathway for biosynthesis of active, C-19 gibberellins like that of the cucurbits; GA12ald → GA12 → GA15 → GA24 → GA36 → GA4 → GA7.  相似文献

The germination of seeds of celery (Apium graveolens L.) becomes progressively thermoinhibited on incubation in the dark at high temperatures, the inhibitory temperature being dependent on the cultivar used. In two high-dormancy cultivars of celery, the production of germination inhibitors in seeds incubated in the dark at 26°C gradually increased over a 7-day period. Inhibitor production was measured by incubating seeds of the low-dormancy cultivar Florida 683 in homogenates of the thermoinhibited seeds of the high-dormancy cultivars and recording germination either in the light or with the gibberellins A4 and A7 (GA4/7) in the dark. Most Florida 683 seeds which failed to germinate in the homogenates after 15 days were induced to germinate by addition of N6-benzyladenine (BA). The presence of BA in addition to GA4/7 throughout incubation in the dark completely overcame the inhibitory effects of homogenates. This indicates that thermoinhibition of celery seeds is associated with the accumulation of a germination inhibitor which interacts with cytokinins. The gibberellin (GA) content of the reproductive organs ofCitrus sinensis (L.) Osb., cv. Bianca Comuna and the seedless variety, Salustiana, were examined by combined gas chromatography-mass spectrometry (GC/MS) at different stages of development. Gibberellins A1, A20, and A29 were identified in the reproductive buds of both cultivars 21 days prior to anthesis and in fruits 35 days after anthesis by comparison of their mass spectra and Kovats retention indices with those of standards. In addition, three uncharacterized isomers of GA1 were detected, one in buds and two in fruits. The presence of GA4 in both tissues, and of GA8 in the reproductive buds, was indicated by the occurrence of characteristic ions at the expected retention times, although their spectra were too weak for full identification. Vegetative shoots of cv. Salustiana contained gibberellins A1, A19, A20, and A29, and the unidentified isomer of GA1 present in reproductive buds. The presence of trace amounts of gibberellins A8 and A17 was also indicated. Although the two varieties did not differ qualitatively in the GAs present during flower and fruit development, the seedless variety contained slightly greater amounts. The concentrations of gibberellins A1, A4, and A20 were determined by gas chromatography-selected ion monitoring (GC/SIM) throughout ovary development and early fruit growth. In both varieties, the maximum GA1 concentration occurred at anthesis. Highest concentrations of gibberellins A20 and A4 were found in fruit 35 days after anthesis, although the GA1 concentration at this stage remained low.  相似文献

Summary The effect of gibberellins A1 through A9 on stem elongation and flower formation in five plants was tested. The plants wereMyosotis alpestris and a biennial strain ofCentaurium minus (cold-requiring plants),Silene armeria andCrepis parviflora (long-day plants), andBryophyllum crenatum (a long-short-day plant). The two former plants were maintained on non-inductive temperatures and long days, the three latter on short days, InMyosotis, flower formation was only obtained with GA7 and GA1, the latter being relatively less active. InCentaurium GA3 was the most effective, followed by GA1, GA4 and GA7 and perhaps GA5 and GA9. InSilene, flower formations was induced only by GA7. InCrepis, the most effective gibberellins were GA4 and GA7, inBryophyllum, GA3, GA4 and GA7. Thus, the different gibberellins exhibited considerable differences in their activity with respect to flower induction, and different plants exhibited in this respect certain specific differences in their sensitivity to the various gibberellins. Except inCrepis, flower initiation as a result of gibberellin treatment was always preceded by substantial stem or internode elongation; however, the correlation between the effect of the different gibberellins on stem elongation and flower induction was not in all cases complete. Methyl jasmonate (JA-Me) inhibited or retarded germination of Amaranthus caudatus seeds in darkness at 24°C, Ethephon, ACC and gibberellins (GA3 or GA4+7) partially or completely reversed this inhibition depending on the concentration of JA-Me applied. Both ethephon and the gibberellins were more effective than ACC. Abstract

Gibberellins are a classic example of the production of plant growth regulators by microorganisms. They are important biotechnological products and are increasingly used in agriculture and horticulture.

This article intends to assemble information on the history of the identification of gibberellins (GA) and producing microorganisms, especially Gibberella fujikuroi (Saw.) Wr. Furthermore, the biosynthesis of gibberelins through the isoprenoid biosynthetic pathway will be described. The main product of GA biosynthesis in Gibberella fujikuroi is gibberellic acid (GA3), which is formed from GA4 via GA7. Both the amount and the type of gibberellins produced by the fungus are dependent on the genetic constitution of the strain and the fermentation conditions.

Mutation and selection for increased product formation are probably the most important factors in improving the yield of gibberellins. Some publications concerning methods of parasexual recombination will also be summarized. Beside strain improvement of wild strains, medium development and appropriate cultivation techniques (batch, fed-batch-, continuous-, and solid state-fermentation) are very important prerequisites for successful economy of gibberellin production. Furthermore, the most important ways of gibberellin recovery and purification are described. Continuing reductions in the costs make gibberellins more attractive for existing applications and open possibilities for further applications of GA3 and some other active gibberellins like GA4, Ga7, and GA9  相似文献

One-bud nodal segments of Ilex paraguariensis (yerba mate) were cultured in vitro in a sugar-rich medium with different gibberellins or an inhibitor of their synthesis. Bud sprouting, shoot length (assessed as shoots of less or more than 5 mm) and bud abscission were evaluated after 45 d of culture in a growth chamber at 27±2 °C, with a 14 h photoperiod of white fluorescent light. There was a differential effect of the two types of gibberellins used; the double bond ring-A gibberellins (GA3 and GA7) inhibited shoot length, while the non double bond-ring A gibberellins (GA1 and GA4) stimulated shoots with a length of more than 5 mm. Prohexadione-Ca (Bx-112; a late step gibberellin biosynthesis inhibitor), at high doses, restrained bud sprouting up to 75%, but lower doses promoted shoot lengthening.  相似文献

In de-rooted seedlings of Amaranthus caudatus L., betacyanin synthesis induced by white light or cytokinin was inhibited by abscisic acid (ABA) or a mixture of gibberellins A4 and A7 (GA4/7). The GA4/7 and ABA effects were additive. Thus ABA inhibited the cytokinin action but had no effect on the gibberellin response.  相似文献
