一株蒽降解细菌的分离及降解特性研究   总被引:1,自引:1,他引:0

【目的】从盐碱土壤中筛选蒽降解菌株并分析其降解特性。【方法】采用极度稀释结果流式细胞检测法筛选分离纯化菌株,通过16S rRNA基因序列分析对菌株进行初步鉴定,采用气质联用仪(GC-MS)分析蒽的降解特性。【结果】从盐碱土壤中筛选出一株高效蒽降解菌株。经过16S rRNA基因序列分析,鉴定该菌株为Demequina salsinemorus BJ1。菌株可以利用蒽作为唯一碳源生长,降解率可达92%。在一定浓度范围内,随着蒽浓度的降低,细菌生长速率变快,降解率升高。添加外加碳源后,细菌生长速率明显变快,而对蒽降解率变低。对萃取中间代谢产物的质谱分析表明,降解蒽的中间代谢产物主要有9,10-anthracenedione (9,10-蒽醌)和Phthalic acid (邻苯二甲酸)等,说明它可能通过邻苯二甲酸途径降解蒽。【结论】筛选得到一株新的耐盐碱蒽降解菌,该菌降解效率高,对修复石油污染的土壤有一定的现实意义。  相似文献

【背景】粪臭素是畜牧堆肥中有机污染物的主要成分,造成养殖场及周边环境恶化,粪臭素污染问题亟待解决,利用微生物降解粪臭素是一种环保节能的有效方法。【目的】分离鉴定粪臭素高效降解菌株,研究其降解特性,为粪臭素降解提供高效的菌种资源,为该菌株应用于臭味污染环境的净化提供基础。【方法】以粪臭素为唯一碳源的无机盐培养基作为培养基质,从猪粪堆肥样品中分离筛选粪臭素高效降解菌株,通过形态特征和16S rRNA基因序列分析进行分离菌株的初步鉴定,分析其生长规律及粪臭素降解特性,并利用气相色谱质谱联用(GC-MS)对菌株代谢粪臭素的产物进行分析。【结果】从样品中分离获得一株能以粪臭素为唯一碳源的细菌YKSW-6菌株,形态学和16S rRNA基因序列分析初步鉴定该菌株为戈登氏红球菌(Rhodococcus gordoniae)。接种量为10%时,该菌培养14 h对100 mg/L的粪臭素降解率达到100%。其能够利用D-山梨醇、溴-丁二酸等18种碳源,对亚碲酸钾、溴酸钾等13种化学敏感物具有抗性。菌株YKSW-6在5%接种量、温度30-42℃和pH值为6.0-9.0时对100 mg/L的粪臭素降解效率均能达到100%,菌株生长和降解粪臭素的最佳条件为:pH 7.2,温度37℃,转速180 r/min。GC-MS结果表明,粪臭素在菌株的作用下C2先被氧化,转变为3-甲基羟基吲哚,随后进一步被氧化为N-(2-乙酰基苯基)甲酰胺。同时中间产物还有苯乙醛和苯乙酸。【结论】红球菌YKSW-6为目前已报道的降解粪臭素能力较强的菌株,丰富了粪臭素降解菌种的资源库,为实际环境微生物修复应用提供了理论参考。  相似文献

【背景】烟草在生产和加工中会产生高浓度的尼古丁废弃物,对环境造成较大的污染。【目的】筛选降解尼古丁的微生物菌种并解析其降解尼古丁的代谢途径,理解微生物如何降解尼古丁。【方法】用常规分离筛选方法、结合形态学观察和分子鉴定手段分离和鉴定菌株类别,进而利用单因素试验方法,通过设置不同的尼古丁浓度、温度和pH确定菌株降解尼古丁的最适发酵条件和降解率,利用气相色谱-质谱联用技术检测菌株在尼古丁降解过程中的主要代谢产物。【结果】获得一株以尼古丁为唯一碳源和氮源的节杆菌属(Arthrobacter)菌株,编号为D4;该菌株降解尼古丁的最适温度和pH分别为30.0℃和7.0;在1 g/L的尼古丁浓度下具备较快的尼古丁降解速率,培养18 h时尼古丁降解率可达到90%以上;尼古丁浓度≥4 g/L时菌株生长受到明显抑制;与目前报道的节杆菌属降解途径不同,该菌株降解尼古丁过程中产生了新的终产物N-甲基吡咯烷酮、可替宁及中间产物麦斯明。【结论】本研究分离鉴定到一株具有较快尼古丁降解速率的节杆菌,该菌株很可能存在新的尼古丁降解途径。  相似文献

【目的】筛选能够降解聚乳酸的微生物, 提高聚乳酸的降解速率并鉴定聚乳酸降解酶种类。【方法】以明胶为唯一碳源, 筛选能够降解聚乳酸的微生物; 通过优化明胶添加浓度、聚乳酸添加量以及金属离子种类提高聚乳酸的降解速率; 通过分析降解过程中代谢产物和酶活力变化明确聚乳酸降解酶类别。【结果】筛选获得一株聚乳酸降解菌株, 鉴定为Lentzea waywayandensis; 经优化培养, 聚乳酸在25 d后失重84.8%; 降解过程中, 检测到乳酸的产生, 体外酶活实验仅检测到蛋白酶活力。【结论】明胶作为唯一碳源适用于聚乳酸降解菌株的筛选; 明胶作为碳源的同时可以作为诱导剂诱导菌株L. waywayandensis降解聚乳酸; 此外, 研究表明蛋白酶在聚乳酸降解过程中发挥重要作用。  相似文献

【目的】以丙烯腈为目标污染物,利用实验室已筛选获得的一株高效腈降解菌Rhodococus rhodochrous BX2,研究其对丙烯腈的降解特性,优化降解条件以提高菌株对丙烯腈的降解能力。【方法】通过单因素试验和响应面分析相结合的方法优化Rhodococus rhodochrous BX2对丙烯腈的降解条件。考察外加碳、氮源对BX2的生长及丙烯腈降解的影响,并确定其在丙烯腈合成废水中对丙烯腈的处理效果。【结果】菌株BX2优化后的最佳降解条件为:底物浓度403.51 mg/L、p H 7.44、温度34.46°C,在此条件下丙烯腈的降解率为95.1%。外加碳源为葡萄糖,或外加氮源为氯化铵对菌株生长及丙烯腈降解有明显的促进作用。菌株Rhodococus rhodochrous BX2能够高效降解合成废水中的丙烯腈,在30 h时其丙烯腈降解率可达89.4%。【结论】降解条件优化以及外源物质的添加强化了菌株对丙烯腈合成废水的处理效果,为生物法处理丙烯腈废水新方法的开发提供技术支持。  相似文献

以合肥钢铁厂焦化废水处理厂的活性污泥为对象,从中驯化筛选得到降酚菌株并利用离子束对出发菌株进行诱变筛选,获得高效降解菌株KE2,它与出发菌株KE相比较,降解苯酚的能力显著提高。在温度为30℃左右,pH值为7时,KE2菌株生长和降解苯酚情况最佳,适量的外加碳源(葡萄糖)对KE2的生长和降酚率有促进作用。  相似文献

【目的】研究Sphingobacterium bambusaue及其紫外诱变菌株的石油降解功能。【方法】紫外诱变后筛选石油降解高效菌株; 以不同石油浓度、pH值及盐浓度优化培养条件, 用重量法检测高效菌株石油降解率。【结果】发现菌株S. bambusaue在石油降解培养基中培养5 d的石油降解率为25.86%, UV诱变高效菌株IBFC2009-S3培养5 d的石油降解为42.85%, 比始发菌株提高65.7%; UV诱变菌株IBFC2009-S3的优化培养条件为石油浓度0.5 g/L、pH值7.0以及NaCl质量浓度为10 g/L, 其石油降解率可达50.51%。【结论】首次报道S. bambusaue具有石油降解功能; 紫外诱变获得的菌株S3的石油降解能力较强。  相似文献

光合细菌PSB-1D不能利用2-氯苯酚(2-CP)作为唯一的碳源和能源.选用苹果酸、丙酸钠、乙酸钠、柠檬酸钠、苯酚、葡萄糖和可溶性淀粉等7种不同碳源作为光合细菌PSB-1D降解2-CP的共代谢基质,考察了在黑暗好氧培养条件下,不同共代谢基质对PSB-1D生长及降解2-CP效果的影响.结果表明:葡萄糖能够很好地促进PSB-1D的大量繁殖,提高降解效果,缩短降解周期,为最佳共代谢基质.对葡萄糖的投加浓度进行了优化,当葡萄糖的投加浓度为3 g·L-1时,菌株PSB-1D培养168 h后的菌体生长浓度△D560为1.749,2-CP的半衰期为3.9 d,降解速率常数为0.00864 h-1.采用SDS-PAGE对微生物全细胞蛋白质进行分析发现,在共代谢过程中当菌株PSB-1D利用葡萄糖作为底物提供能源和碳源时,可诱导产生2-CP特异性降解酶.  相似文献

【目的】采用复合诱导的方法,选育出能在低温条件下性状稳定、生长良好且具有工业生产价值的低温乳酸菌菌株。【方法】使用紫外照射和亚硝基胍处理的方法对分离的Q1菌株进行诱变处理,将选育得到的低温菌株Q1-4-6与出发菌株Q1在15°C、20°C、37°C下,比较其生长速率、产酸量和糖降解量。【结果】从玛曲牧民自制酸奶中分离得到一株乳酸菌Q1,根据其形态学特征、生理生化特性,同时结合16SrDNA序列分析结果,将其鉴定为肠球菌属棉籽糖肠球菌(Enterococcus raffinosus)。经过多轮诱变选育出一株能在低温下良好生长、遗传性状稳定的菌株Q1-4-6,将出发菌株与诱变菌株在15°C、20°C比较其生长、产酸和糖降解量发现,诱变菌株均好于出发菌株。在37°C的自然条件下,诱变菌株和出发菌株的生长速率差异不大,但均略高于出发菌株。在15°C培养发现,诱变菌株生长速率、产酸量均高于购买菌株干酪乳杆菌(CL04)和嗜热链球菌(CL05)。【结论】通过UV+NTG复合诱变,最终选育出一株在低温下生长良好、遗传性状稳定的菌株Q1-4-6。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. Rasmussen’s encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and 16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis, intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8+ T cells. Our results suggest that EBV and HHV6 infection and the activation of CD8+ T cells are involved in the pathogenesis of RE.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献
