We examined the responses of some gustatory neurons in various contact-chemoreceptor sensilla of second-instar larvae of the spruce budworm. These included the L1 and L2 sensilla on the maxillary palp, and the LST and MST sensilla on the galea. Our objective was to determine whether there were differences in the physiological characteristics of individual neurons between the early and late larval instars. Changes were observed in both some sugar-sensitive and amino acid-sensitive neurons. We also confirmed the presence of a water-sensitive neuron in the L2 sensillum. Our findings are discussed in relation to changes that occur during the development of both the host plant and the insect. To our knowledge, this is the first paper to examine the responses from contact-chemoreceptor sensilla of very young second-instar caterpillar larvae.  相似文献

The tip of the maxillary palp in the spruce budworm and other lepidoptera larvae contains a cluster of eight basiconic sensilla. Except for a few electrophysiological recordings from the entire group of these sensilla, no information is available on the response characteristics of any individual one. Using a compound microscope at 320x magnification, with a long working-distance objective, it has been possible to view individual sensilla and to record electrical responses from three of these to some gustatory stimuli in preliminary tests. One of them, sensillum (L1), contains a sugar-sensitive neuron whose responses to a range of sucrose concentrations are reported here. The physiological characteristics of this neuron differ from those of the known sugar-sensitive neuron in the lateral styloconic sensillum on the galea of these same insects. Input of chemosensory information about the same gustatory stimulus by more than one neuron in different mouthpart sensilla is discussed in relation to the feeding habits of the spruce budworm.  相似文献

The blowfly, Phormia regina, has sensilla with four contact-chemoreceptor cells and one mechanoreceptor cell on its labellum. Three of the four chemoreceptor cells are called the sugar, the salt and the water receptor cells, respectively. However, the specificity of the remaining chemoreceptor cell, traditionally called the "fifth cell", has not yet been clarified. Referring to behavioral evaluation of the oral toxicity of monoterpenes, we measured the electrophysiological response of the "fifth cell" to these compounds. Of all the monoterpenes examined, D-limonene exhibited the strongest oral toxicity and induced the severest aversive behavior with vomiting and/or excretion in the fly. D-Limonene, when dispersed in an aqueous stimulus solution including dimethyl sulfoxide or an odorant-binding protein (OBP) found in the contact-chemoreceptor sensillum, the chemical sense-related lipophilic ligand-binding protein (CRLBP), evoked impulses from the "fifth cell". Considering the relationship between the aversive effects of monoterpenes and the response of the "fifth cell" to these effects, we propose that the "fifth cell" is a warning cell that has been differentiated as a taste system for detecting and avoiding dangerous foods. Here we suggest that in the insect contact-chemoreceptor sensillum, CRLBP carries lipophilic members of the noxious taste substances to the "fifth cell" through the aqueous sensillum lymph. This insect OBP may functionally be analogous to the von Ebner's grand protein in taste organs of mammals.  相似文献

On the basis of 1H and 13C NMR spectroscopy studies, the proportion of pyranose and furanose forms of 6-deoxyheptoses in water solution was determined. Water solution of 6-deoxyheptoses contains all possible furanose and pyranose forms (except 6-deoxy-gluco-heptose for which only pyranose was found), although pyranose is dominant.  相似文献

Using comparative ion-exchange chromatography on Dowex 1X4, the product of dephosphorylation of fructose 2,6-bisphosphate with purified yeast fructose-2,6-bisphosphate 6-phosphohydrolase, was shown to be identical to the furanose form of fructose 2-phosphate prepared by chemical synthesis according to Pontis and Fischer [Biochem. J. 89, 452-459 (1963)]. As expected for the furanose form of fructose 2-phosphate, the enzymatically formed product consumes 1 mol periodate/mol fructose 2-phosphate, whereas the chemically synthesized pyranose form consumes 2 mol periodate/mol. In addition, it is shown that the enzymatic product behaves identically to the furanose, not the pyranose, form of fructose 2-phosphate in hydrolysis of the ester bond at pH 4 and 37 degrees C, as described previously for the chemically synthesized compounds [Pontis and Fischer (1963) vide supra].  相似文献

Insect repellents are widely used to protect against insect bites and thus prevent allergic reaction and the spread of disease. To gain insight into the mosquito’s response to chemicals repellents, we investigated the interaction between the olfactory system of the mosquito Culex quinquefasciatus Say and chemical repellents using single sensillum recording. The interactions of 50 repellent chemicals with olfactory receptor neurons were measured in six different types of mosquito sensilla: long sharp trichoid (LST), short sharp trichoid (SST), short blunt trichoid I (SBT-I), short blunt trichoid II (SBT-II), short blunt trichoid-curved (SBT-C), and grooved peg (GP). A single olfactory neuron reacted to the chemical repellents in each of the sensilla except for SBT-I and SBT-II, where two neurons were involved. Other than LST and GP, which showed no or very weak responses to the repellents tested, all the sensilla showed significant excitatory responses to certain types of repellents. Terpene-derived chemicals such as eucalyptol, α-pinene, and camphor, stimulated olfactory receptor neurons in a dose-dependent manner and mosquitoes responded more strongly to terpene-derived chemical repellents than to non-terpene-derived chemicals such as dimethyl phthalate. Mosquitoes also exhibited a similar response to stereoisomers of chemicals such as (−)-β-pinene versus (+)-β-pinene, and (−)-menthone versus (+)-menthone. This study not only demonstrates the effects of chemical repellents on the mosquito olfactory system but also provides important information that will assist those screening new mosquito repellents and designing new mosquito control agents.  相似文献

A 3 min treatment of a single sugar receptor with 0·5 mM p-chloromercuribenzoate (PCMB) did not affect its response to d-fructose, but depressed completely its response to d-glucose. This is the first direct evidence of the presence of two different sites in the sugar receptor of the fly.No specific protection by d-glucose on PCMB treatment suggested that PCMB did not react at a glucose-binding site but did react at a specific site indispensable to simulation by d-glucose.Various sugars were examined and classified into two groups according to the effects of PCMB treatment on the sugar receptor. They correspond to those effective in the furanose and pyranose forms, respectively. The pyranose group was further divided into two subclasses according to the presence or absence of three successive equatorial hydroxyl groups regardless of their positions. The results are discussed in relation to the structures that are common to furanose stimulating sugars.  相似文献

Abstract. Inhibition of peripheral chemoreceptors by micromolar concentrations of alkaloids may account for feeding deterrence. Earlier work showed a reduction in both the proboscis extension response and sensory activity recorded from sugar-sensitive cells in tarsal D and labellar 'largest' hairs when the stimulating solution was sucrose mixed with alkaloids. In the present work a similar behavioural effect was also seen when alkaloids were mixed with pyranose and furanose sugars as well as with water and deuterium oxide. Behavioural deterrence continued after the stimulus was removed. Complete recovery occurred after 120 s. To check against the possibility of a central inhibitory state (CIS) having been established two sets of experiments were run in which quinine was applied to a single tarsus followed by a sucrose post-test to the contralateral tarsus. In one half of the tests sugar was applied first (S1Q2), and in the other half quinine first (Q1S2). Comparable tests were run with contralateral labellar hairs. No CIS was set up by Q1; however, a central excitatory state (CES) was set up by S1. In order to test (a) whether quinine had different effects on other known or unknown receptors and (b) post-ingestive effects, volumes imbibed in single drinks of sucrose with and without quinine were compared. Volumes of the mixture imbibed were not significantly different from what was predicted based on the known reduction in sensory acitivty.  相似文献

Ribose is transported into cells in its pyranose form and must be rearranged to its furanose form for further utilization. Ribose pyranase RbsD catalyzes the conversion of ribose from the pyranose to furanose form. This is the key step for substrate supply to ribokinase RbsK, which converts ribose to ribose-5-phosphate for further metabolism. Sequence analysis indicated Sa240 from Staphylococcus aureus was a ribose pyranase homolog. Here we showed that Sa240 formed dimeric structure both in solution and in crystal. S240-ribose complex structure showed a ribose binding site formed by an incomplete active site compared with RbsD. Because the catalytic activity of ribose pyranase depends on its oligomeric state, we propose Sa240 is catalytically inactive in its dimeric structure.  相似文献

Responses of a single sugar receptor to oligosaccharides, such as turanose, palatinose, cellobiose, trehalose, maltotriose, melezitose, and raffinose, were depressed almost completely after 3 min treatment with 0·5 mM p-chloromercuribenzoate (PCMB). In the same preparation, responses to d-glucose were depressed in the same manner, but those to d-fructose were hardly affected after PCMB treatment. This may indicate that these oligosaccharides do not react with a furanose site but react only with a pyranose site. The stereospecificity for these oligosaccharides of the sugar receptor are discussed.The response to 4 M d-mannose, a very weak stimulative sugar, was almost completely depressed after PCMB treatment, which suggests that a stimulative fraction of d-mannose reacts with the pyranose site in spite of its inhibitory effect on fructose stimulation.  相似文献
