Stearoyl-acyl carrier protein-desaturase (SACPD)-catalyzed synthesis of oleic acid (18:1) is an essential step in fatty acid biosynthesis. Arabidopsis mutants (ssi2) with reduced SACPD activity accumulate salicylic acid (SA) and exhibit enhanced resistance to multiple pathogens. We show that reduced levels of 18:1 induce similar defense-related phenotypes in soybean. A Bean pod mottle virus (BPMV)-based vector was employed to effectively silence soybean SACPDs. The silenced plants contained reduced 18:1 and increased stearic acid, developed spontaneous cell death lesions, increased SA accumulation, and constitutively expressed pathogenesis-related genes. These plants also expressed elevated levels of resistance-like genes and showed resistance to bacterial and oomycete pathogens. Exogenous application of glycerol induced similar phenotypes, mimicking the effect of silencing SACPDs in healthy soybean plants. Overexpression of a soybean SACPD increased 18:1 levels in ssi2 but not in wild-type Arabidopsis plants, suggesting that the soybean enzyme was under feedback regulation similar to that of the Arabidopsis isozymes. These results suggest that soybean and Arabidopsis respond similarly to 18:1-derived cues by inducing a novel broad-spectrum resistance-conferring pathway, even though they differ significantly in their lipid biosynthetic pathways. We also demonstrate the efficacy of BPMV-induced gene silencing as a tool for functional studies in soybean.  相似文献

BAK1 is a co-receptor of brassinosteroid (BR) receptor BRI1, and plays a well-characterized role in BR signalling. BAK1 also physically interacts with the flagellin receptor FLS2 and regulates pathogen resistance. The role of BAK1 in mediating Nicotiana attenuata's resistance responses to its specialist herbivore, Manduca sexta, was examined here. A virus-induced gene-silencing system was used to generate empty vector (EV) and NaBAK1-silenced plants. The wounding- and herbivory-induced responses were examined on EV and NaBAK1-silenced plants by wounding plants or simulating herbivory by treating wounds with larval oral secretions (OS). After wounding or OS elicitation, NaBAK1-silenced plants showed attenuated jasmonic acid (JA) and JA-isoleucine bursts, phytohormone responses important in mediating plant defences against herbivores. However, these decreased JA and JA-Ile levels did not result from compromised MAPK activity or elevated SA levels. After simulated herbivory, NaBAK1-silenced plants had EV levels of defensive secondary metabolites, namely, trypsin proteinase inhibitors (TPIs), and similar levels of resistance to Manduca sexta larvae. Additional experiments demonstrated that decreased JA levels in NaBAK1-VIGS plants, rather than the enzymatic activity of JAR proteins or Ile levels, were responsible for the reduced JA-Ile levels observed in these plants. Methyl jasmonate application elicited higher levels of TPI activity in NaBAK1-silenced plants than in EV plants, suggesting that silencing NaBAK1 enhances the accumulation of TPIs induced by a given level of JA. Thus NaBAK1 is involved in modulating herbivory-induced JA accumulation and how JA levels are transduced into TPI levels in N. attenuata.  相似文献

greenhouse experiment with factorial arrangement based on randomized complete block design with four replications was conducted in 2015 to evaluate the effects of salicylic acid (SA) (1 mM) and jasmonic acid (JA) (0.5 mM) on oil accumulation and fatty acid composition of soybean oil (Glycine max L.) under salt stress (Non-saline, 4, 7, and 10 dS/m NaCl). Oil percentage of soybean seeds declined, while oil content per seed enhanced with increasing seed filling duration. Foliar application of SA improved oil content per soybean seed at different stages of development under all salinity levels. Although JA treatment enhanced seed oil percentage, oil yield of these plants decreased as a result of reduction in seed yield per plant. In contrast, the highest oil yield was recorded for SA treated plants, due to higher seed yield. Salinity had no significant effects on percentage of palmitic acid and stearic acid, but treatment with JA significantly reduced stearic acid percentage. Oleic acid content of seeds increased, but percentages of linoleic acid, linolenic acid and unsaturation index (UI) of soybean oil decreased with increasing salinity. Foliar application of SA and JA improved oil quality of soybean seeds by reducing oleic acid and enhancing linoleic acid, linolenic acid contents and UI. Exogenous application of SA had the most beneficial effects on soybean seeds due to enhancing oil yield and quality under saline and non-saline conditions.  相似文献

We report here that disruption of function of the ω-3 FATTY ACID DESATURASE7 (FAD7) enhances plant defenses against aphids. The suppressor of prosystemin-mediated responses2 (spr2) mutation in tomato (Solanum lycopersicum), which eliminates the function of FAD7, reduces the settling behavior, survival, and fecundity of the potato aphid (Macrosiphum euphorbiae). Likewise, the antisense suppression of LeFAD7 expression in wild-type tomato plants reduces aphid infestations. Aphid resistance in the spr2 mutant is associated with enhanced levels of salicylic acid (SA) and mRNA encoding the pathogenesis-related protein P4. Introduction of the Naphthalene/salicylate hydroxylase transgene, which suppresses SA accumulation, restores wild-type levels of aphid susceptibility to spr2. Resistance in spr2 is also lost when we utilize virus-induced gene silencing to suppress the expression of NONEXPRESSOR OF PATHOGENESIS-RELATED PROTEINS1 (NPR1), a positive regulator of many SA-dependent defenses. These results indicate that FAD7 suppresses defenses against aphids that are mediated through SA and NPR1. Although loss of function of FAD7 also inhibits the synthesis of jasmonate (JA), the effects of this desaturase on aphid resistance are not dependent on JA; other mutants impaired in JA synthesis (acx1) or perception (jai1-1) show wild-type levels of aphid susceptibility, and spr2 retains aphid resistance when treated with methyl jasmonate. Thus, FAD7 may influence JA-dependent defenses against chewing insects and SA-dependent defenses against aphids through independent effects on JA synthesis and SA signaling. The Arabidopsis (Arabidopsis thaliana) mutants Atfad7-2 and Atfad7-1fad8 also show enhanced resistance to the green peach aphid (Myzus persicae) compared with wild-type controls, indicating that FAD7 influences plant-aphid interactions in at least two plant families.  相似文献

Phytohormones and fatty acids play a significant role in developmental stages of plant growth and defense against biotic and abiotic stresses. The purpose of this study was to determine the spermidine (Spd)-induced phytohormones and fatty acids changes involve the acclimation of cucumber plants against salt stress. Plants exposed to salt stress had significant reduction in their growth. Exogenously applied Spd increased the shoot length and protein content in salt-stressed plants. The accumulation of total phenol and malondialdehyde was higher in salt-affected plants than in their controls and these detrimental effects were mitigated by Spd treatment. Moreover, salt stress caused a significant increase in salicylic acid (SA) and jasmonic acid (JA); while Spd treatment ameliorated these salt stress effects by reducing SA and JA content. The marked accumulation of total free fatty acid was observed in salt-stressed plants, while the application of Spd to salt-stressed plants reduced the total free fatty acid content. In addition, Spd inhibited the stearic acid, linoleic acid and linolenic acid in salt-stressed plants. The results of current study suggest that exogenous application of Spd-induced phytohormones and fatty acids changes would be a reason for increasing the acclimation of cucumber plants under salt stress condition.  相似文献

以遗传背景一致的抗二氯喹啉酸型和敏感型稗草为供试材料,分析了经外源茉莉酸甲酯(MeJA)预处理后,稗草种群间应对二氯喹啉酸胁迫的抗性水平及4种内源激素含量的变化。结果表明: 外源MeJA处理可显著提高抗性稗草对二氯喹啉酸的抗性,而对敏感稗草的抗性影响不显著。供试稗草间植株体内生长素(IAA)、脱落酸(ABA)、水杨酸(SA)和茉莉酸(JA)的含量及其变化幅度存在显著差异。二氯喹啉酸处理显著增加了稗草ABA、SA和JA的含量,且敏感稗草ABA和JA含量升高幅度明显高于抗性稗草;外源MeJA预处理能显著增强二氯喹啉酸对稗草ABA和JA含量的诱导,且敏感稗草变化幅度较大。表明敏感稗草较快的激素变化不利于其应对除草剂胁迫,抗性稗草相对平缓的激素变化赋予了其对除草剂的适应性,且MeJA处理增强了其对二氯喹啉酸的抗性。稗草植株体内ABA和JA激素信号在其抵抗二氯喹啉酸胁迫过程中可能发挥重要的生物学功能。  相似文献

Microsomal omega-3 fatty acid desaturase is an essential enzyme in the production of the n-3 polyunsaturated fatty acid alpha-linolenic acid during the seed developing stage. We have constructed a chimeric gene consisting of a maize Ubi1-P-int and a soybean GmFAD3 cDNA, which was introduced into rice plants by Agrobacterium-mediated transformation. Ten transformants containing the chimeric gene were established and expression subsequently confirmed by Northern blotting. Furthermore, alpha-linolenic acid content of the T(1) seeds increased dramatically up to tenfold that of the control, and this phenotype was also stably inherited in the T(2) and T(3) progenies. These results demonstrate that the alpha-linolenic acid content of rice seed oil can easily be altered using the combination of a high-activity promoter and a GmFAD3 gene.  相似文献

The Arabidopsis mutants ssi2 and fab2 are defective in stearoyl ACP desaturase, which causes altered salicylic acid (SA)- and jasmonic acid (JA)-mediated defense signaling. Both ssi2 and fab2 plants show spontaneous cell death, express PR genes constitutively, accumulate high levels of SA, and exhibit enhanced resistance to bacterial and oomycete pathogens. In contrast to constitutive activation of the SA pathway, ssi2 and fab2 plants are repressed in JA-mediated induction of the PDF1.2 gene, which suggests that the SSI2-mediated signaling pathway modulates cross talk between the SA and JA pathways. In this study, we have characterized two recessive nonallelic mutants in the ssi2 background, designated as rdc (restorer of defective cross talk) 2 and rdc8. Both ssi2 rdc mutants are suppressed in constitutive SA signaling, show basal level expression of PR-1 gene, and induce high levels of PDF1.2 in response to exogenous application of JA. Interestingly, while the rdc8 mutation completely abolishes spontaneous cell death in ssi2 rdc8 plants, the ssi2 rdc2 plants continue to show some albeit reduced cell death. Fatty acid (FA) analysis showed a reduction in 16:3 levels in ssi2 rdc8 plants, which suggests that this mutation may limit the flux of FAs into the prokaryotic pathway of glycerolipid biosynthesis. Both rdc2 and rdc8 continue to accumulate high levels of 18:0, which suggests that 18:0 levels were responsible for neither constitutive SA signaling nor repression of JA-induced expression of the PDF1.2 gene in ssi2 plants. We also analyzed SA and JA responses of the fab2-derived shs1 mutant, which accumulates levels of 18:0 over 50% lower than those in the fab2 plants. Even though fab2 shs1 plants were morphologically bigger than fab2 plants, they expressed PR genes constitutively, showed HR-like cell death, and accumulated elevated levels of SA. However, unlike the ssi2 rdc plants, fab2 shs1 plants were unable to induce high levels of PDF1.2 expression in response to exogenous application of JA. Together, these results show that defective cross talk in ssi2 can be restored by second site mutations and is independent of morphological size of the plants, cell death, and elevated levels of 18:0.  相似文献

Signaling induced upon a reduction in oleic acid (18:1) levels simultaneously up-regulates salicylic acid (SA)-mediated responses and inhibits jasmonic acid (JA)-inducible defenses, resulting in enhanced resistance to biotrophs but increased susceptibility to necrotrophs. SA and the signaling component Enhanced Disease Susceptibility1 function redundantly in this low-18:1-derived pathway to induce SA signaling but do not function in the repression of JA responses. We show that repression of JA-mediated signaling under low-18:1 conditions is mediated via the WRKY50 and WRKY51 proteins. Knockout mutations in WRKY50 and WRKY51 lowered SA levels but did not restore pathogenesis-related gene expression or pathogen resistance to basal levels in the low-18:1-containing Arabidopsis (Arabidopsis thaliana) mutant, suppressor of SA insensitivity2 (ssi2). In contrast, both JA-inducible PDF1.2 (defensin) expression and basal resistance to Botrytis cinerea were restored. Simultaneous mutations in both WRKY genes (ssi2 wrky50 wrky51) did not further enhance the JA or Botrytis-related responses. The ssi2 wrky50 and ssi2 wrky51 plants contained high levels of reactive oxygen species and exhibited enhanced cell death, the same as ssi2 plants. This suggested that high reactive oxygen species levels or increased cell death were not responsible for the enhanced susceptibility of ssi2 plants to B. cinerea. Exogenous SA inhibited JA-inducible PDF1.2 expression in the wild type but not in wrky50 or wrky51 mutant plants. These results show that the WRKY50 and WRKY51 proteins mediate both SA- and low-18:1-dependent repression of JA signaling.  相似文献

Soil contamination with nickel (Ni) is a persistent threat to crop production worldwide. The present study examined the putative roles of jasmonic acid (JA) in improving Ni tolerance in soybean. Our findings showed that priming of soybean seeds with JA significantly improved the growth performance of soybean when grown under excessive Ni. The enhanced Ni tolerance of soybean prompted by JA could be ascribed to its ability to regulate Ni uptake and accumulation, and to decrease Ni-induced membrane damage as evidenced by reduced levels of reactive oxygen species (ROS), malondialdehyde, lipoxygenase activity, and electrolyte leakage in Ni-stressed plants. JA also boosted redox states and antioxidant capacity in Ni-stressed plants by maintaining increased levels of ascorbate and glutathione, and enhanced activities of ROS-detoxifying enzymes compared with Ni-stressed alone plants. Additionally, methylglyoxal detoxification system was significantly upregulated in JA-primed and “JA-primed?+?Ni-stressed” plants, indicating an alleviating effect of JA on Ni-induced methylglyoxal toxicity. Our results conclude that JA-mediated regulation of Ni uptake and accumulation, and enhanced ROS metabolism by activating antioxidant defense and glyoxalase systems contributed to improved performance of soybean under excessive Ni, thereby suggesting JA as an effective stress regulator in mitigating Ni toxicity in economically important soybean, and perhaps in other crops.

α-Linolenic acid (ALA, C 18:3Δ9,12,15) has many important biological functions. ω-3 Fatty acid desaturase (FAD3), existing in cytosolic and plastidic compartments of higher plants, catalyzes linoleic acid (LA) desaturation to produce ALA. GmFAD3A-2 and GmFAD3C genes encoding cytosolic FAD3 from Qihuang 29 soybean were cloned and inserted into p416 vector and expressed in K601 yeast strain. Gas chromatography showed that the transformed yeast strains could produce ALA. The ALA accumulation levels for the strains transformed with GmFAD3A-2 or GmFAD3C genes were 0.77 ± 0.1 and 4.13 ± 0.4% of total fatty acids, respectively, while, as compared with that of the control, the contents of LA decreased from 14.34 ± 0.8 to 10.93 ± 0.0 and 7.85 ± 0.1%, respectively, implying that the GmFAD3C enzyme is more vigorous or stable, than GmFAD3A-2. Mitogen-activated protein kinase (MAPK) cascades play important roles in disease resistance in model plant species such as Arabidopsis (Arabidopsis thaliana) and tobacco (Nicotiana tabacum). However, the importance of MAPK signaling pathways in the disease resistance of crops is still largely uninvestigated. To better understand the role of MAPK signaling pathways in disease resistance in soybean (Glycine max), 13, nine, and 10 genes encoding distinct MAPKs, MAPKKs, and MAPKKKs, respectively, were silenced using virus-induced gene silencing mediated by Bean pod mottle virus. Among the plants silenced for various MAPKs, MAPKKs, and MAPKKKs, those in which GmMAPK4 homologs (GmMPK4s) were silenced displayed strong phenotypes including stunted stature and spontaneous cell death on the leaves and stems, the characteristic hallmarks of activated defense responses. Microarray analysis showed that genes involved in defense responses, such as those in salicylic acid (SA) signaling pathways, were significantly up-regulated in GmMPK4-silenced plants, whereas genes involved in growth and development, such as those in auxin signaling pathways and in cell cycle and proliferation, were significantly down-regulated. As expected, SA and hydrogen peroxide accumulation was significantly increased in GmMPK4-silenced plants. Accordingly, GmMPK4-silenced plants were more resistant to downy mildew and Soybean mosaic virus compared with vector control plants. Using bimolecular fluorescence complementation analysis and in vitro kinase assays, we determined that GmMKK1 and GmMKK2 might function upstream of GmMPK4. Taken together, our results indicate that GmMPK4s negatively regulate SA accumulation and defense response but positively regulate plant growth and development, and their functions are conserved across plant species.  相似文献

Plant defenses against pathogens and insects are regulated differentially by cross-communicating signal transduction pathways in which salicylic acid (SA) and jasmonic acid (JA) play key roles. In this study, we investigated the molecular mechanism of the antagonistic effect of SA on JA signaling. Arabidopsis plants unable to accumulate SA produced 25-fold higher levels of JA and showed enhanced expression of the JA-responsive genes LOX2, PDF1.2, and VSP in response to infection by Pseudomonas syringae pv tomato DC3000, indicating that in wild-type plants, pathogen-induced SA accumulation is associated with the suppression of JA signaling. Analysis of the Arabidopsis mutant npr1, which is impaired in SA signal transduction, revealed that the antagonistic effect of SA on JA signaling requires the regulatory protein NPR1. Nuclear localization of NPR1, which is essential for SA-mediated defense gene expression, is not required for the suppression of JA signaling, indicating that cross-talk between SA and JA is modulated through a novel function of NPR1 in the cytosol.  相似文献
