Two experiments were conducted to evaluate use of chromium mordanted neutral detergent residue (Cr-NDr) and cobalt EDTA (Co-EDTA) as predictors of dry matter intake (DMI) in high producing grazing dairy cows. The first experiment was conducted with 10 lactating Holstein cows individually fed a total mixed ration (TMR) in confinement, and dosed with Cr-NDr and Co-EDTA twice daily at milking times for 12-days to validate the markers used for the second experiment. The Cr-NDr accounted for 96% of the variation (r2) in DMI, while Co-EDTA underpredicted DMI by 43% (r2=0.65). The second experiment was conducted on a pasture-based dairy farm, to evaluate the use of Cr-NDr to predict DMI of grazing dairy cows. 15 and 14 high producing dairy cows in trial 1 and 2, respectively, were dosed twice a day at milking times with Cr-NDr for 12-days. Mean total DMI estimated from marker recoveries were unrealistically high (5.95 and 5.52% of body weight for trials 1 and 2, respectively). It was concluded that either diurnal variation in fecal excretion of the marker or a failure in the technique of collecting pasture samples that reflected the cows’ true grazing selection in order to determine pasture composition occurred.  相似文献

Wild white-faced sakis (Pithecia pithecia) ingest primarily seeds that provide a diet that is lipid-rich and moderately high in dietary fiber. Although little anatomical information is available on sakis, evidence from other vertebrate seed predators suggests that such a diet is correlated with adaptations in gut morphology or physiology. Milton [1984] reported a 20 hr transit time (TT=transit time or time of first appearance in feces) of a particulate marker for a single monk saki (Pithecia monachus). This suggests that TT for Pithecia sakis may be four to five times longer than what has been reported for soft-fruit-eating platyrrhines, such as Ateles and Cebus. During a captive study, we calculated an average TT of 14.7+/-0.4 hr (n=5 trials started in the evening) for a chromic oxide (Cr(2)O(3)) marker that follows liquid digesta and TTs of 14.5 hr (trial started the previous evening) and 23.0 hr (trial started the previous morning) for two trials using particulate markers. Mean retention time (MRT) for the liquid marker ranged from 15.3 hr to 37.7 hr in four trials that lasted longer than 90 hr. Marker recovery was incomplete for the particulate markers in these trials, and thus MRT could only be determined for the liquid phase marker. Three 5-day trials on a low-fiber, blended diet revealed high fiber fraction digestibilities (neutral detergent fiber (NDF)=77.4% and acid detergent fiber (ADF)=74.4%). Data collected for this study and nutritional data from wild sakis suggest that pitheciin seed predators may have a potential for fiber digestibility that is intermediate between ripe-pulp frugivores and folivores.  相似文献

Members of the Bovini genus are classified as grazers. Smaller species of ruminants are not expected to be able to digest particularly fibrous diets and are more often classified as intermediate feeders or browsers. Anoas (Bubalus spp.) are interesting in this respect as they are the smallest representatives of the Bovini, being only 10–20% of the body weight of other species of the same genus. A feeding trial was carried out with four lowland anoas (Bubalus depressicornis) at London Zoo, investigating diet digestibility by total fecal collection and passage rates by the simultaneous administration of a fluid (Co‐EDTA) and a particle (Cr‐mordanted fibre <2 mm) marker. The diet consisted of legume hay, dairy cow pellets, browse, fruits, and vegetables. The achieved digestibility coefficients averaged 70±4% for dry matter and 57±7% for cell walls (NDF). Mean retention times for the total gastrointestinal tract were 25±4.1 hr for fluid and 39±6.7 hr for particles, respectively. The ratio of forestomach particle:fluid retention was 2.14±0.40. Additional information regarding anoa diets in captivity was collected through a survey targeting all institutions that have anoas in their collection currently. Suitability of the provided diet was evaluated using the ratio of unstructured:structured feeds (unstructured feeds pellets, grains, produce; structured feeds=roughage, browse) on a dry matter basis and an assumed complete consumption of offered unstructured diet items, with only the remaining intake capacity being met by structured items. The use of this ratio reliably predicted one facility that reported chronic diet‐related problems. As other ruminants, anoas should receive a diet with restricted amounts of concentrates and fruits. The comparatively high fibre digestibility and the high selective particle retention in the forestomach suggest a classification of an intermediate/grazing ruminant. The digestibility and passage of an experimental diet was used to compare the digestive physiology of two Propithecus species: P. verreauxi and P. tattersalli. Though both animals have a similar feeding ecology, the captive status of P. verreauxi is considered more stable than that of P. tattersalli. The test diet included a local tree species, Rhus copallina, at 15% of dry matter intake (DMI) and Mazuri Leafeater Primate Diet at 85% of DMI. The chemical composition of the diet (dry matter basis) was 25% crude protein, 34% neutral detergent fiber (NDF), and 22% acid detergent fiber (ADF) with a gross energy of 4.52 kcal/g. After a 6 week acclimation to the experimental diet, animals were placed in research caging. After a 7 day adjustment period, animals were dosed with chromium mordant and Co‐EDTA as markers for digesta passage and all feed refusals and feces were collected at timed intervals for 7 days. Digestibility values, similar for both species, were approximately 65% for dry matter, crude protein, and energy, and 40% and 35% respectively, for NDF and ADF. Transit times (17–18.5 hr) and mean retention times (31–34 hr) were not significantly different between species, and there was no difference between the chromium mordant and Co‐EDTA. Serum values for glucose, urea, and non‐esterified fatty acids (NEFA) were obtained during four different time periods to monitor nutritional status. While there was no change in serum glucose, serum urea increased over time. The NEFAs increased across all four time periods for P. verreauxi and increased for the first three periods then decreased in the last period for P. tattersalli. Results obtained indicate no difference in digestibility nor digesta passage between species, and that both Propithecus species were similar to other post‐gastric folivores. Am. J. An important component of digestive physiology involves ingesta mean retention time (MRT), which describes the time available for digestion. At least three different variables have been proposed to influence MRT in herbivorous mammals: body mass, diet type, and food intake (dry matter intake, DMI). To investigate which of these parameters influences MRT in primates, we collated data for 19 species from trials where both MRT and DMI were measured in captivity, and acquired data on the composition of the natural diet from the literature. We ran comparative tests using both raw species values and phylogenetically independent contrasts. MRT was not significantly associated with body mass, but there was a significant correlation between MRT and relative DMI (rDMI, g/kg(0.75)/d). MRT was also significantly correlated with diet type indices. Thus, both rDMI and diet type were better predictors of MRT than body mass. The rDMI-MRT relationship suggests that primate digestive differentiation occurs along a continuum between an "efficiency" (low intake, long MRT, high fiber digestibility) and an "intake" (high intake, short MRT, low fiber digestibility) strategy. Whereas simple-stomached (hindgut fermenting) species can be found along the whole continuum, foregut fermenters appear limited to the "efficiency" approach.  相似文献

1. Mean retention times (MRTs) of fluid (marked with Co-EDTA), fine particles (mordanted with Yb) and large particles (mordanted with Cr) were measured in brushtail possums (Trichosurus vulpecula), ringtail possums (Pseudocheirus peregrinus) and laboratory rabbits fed semipurified diets. 2. In brushtail possums there were no significant differences in MRT among the three digesta markers. 3. In ringtail possums MRTs of the fluid and fine particle markers were approximately twice that of the large particle marker, indicative of selective retention of both fluid and fine particles in the caecum. 4. In the rabbit MRT of fine particles was also greater than that of large particles, again indicative of selective retention of fine particles in the caecum. 5. Fibre digestibility was greater in the rabbits than in the ringtail possums, and greater for neutral-detergent fibre (including agar) but less for acid-detergent fibre in the rabbits than in the brushtails. Differences in fibre digestibility between brushtails and rabbits were explained by differences in patterns of digesta flow. However, the higher digestibilities of fibre in the rabbits than in the ringtail possums could not be explained on a similar basis.  相似文献

Common (Callithrix jacchus) and pygmy (Cebuella pygmaea) marmosets and cotton‐top tamarins (Saguinus oedipus) share broadly similar diets of fruits, insects, and tree exudates. Marmosets, however, differ from tamarins in actively gouging trees with their anterior dentition to elicit tree exudates flow. Tree gouging in common marmosets involves the generation of relatively wide jaw gapes, but not necessarily relatively large bite forces. We compared fiber architecture of the masseter and temporalis muscles in C. jacchus (N = 18), C. pygmaea (N = 5), and S. oedipus (N = 13). We tested the hypothesis that tree‐gouging marmosets would exhibit relatively longer fibers and other architectural variables that facilitate muscle stretch. As an architectural trade‐off between maximizing muscle excursion/contraction velocity and muscle force, we also tested the hypothesis that marmosets would exhibit relatively less pinnate fibers, smaller physiologic cross‐sectional areas (PCSA), and lower priority indices (I) for force. As predicted, marmosets display relatively longer‐fibered muscles, a higher ratio of fiber length to muscle mass, and a relatively greater potential excursion of the distal tendon attachments, all of which favor muscle stretch. Marmosets further display relatively smaller PCSAs and other features that reflect a reduced capacity for force generation. The longer fibers and attendant higher contraction velocities likely facilitate the production of relatively wide jaw gapes and the capacity to generate more power from their jaw muscles during gouging. The observed functional trade‐off between muscle excursion/contraction velocity and muscle force suggests that primate jaw‐muscle architecture reflects evolutionary changes related to jaw movements as one of a number of functional demands imposed on the masticatory apparatus. In captivity, langurs (foregut fermenting primates) often suffer from digestive disorders, and in particular display soft stools on diets with a high proportion of fruits, vegetables, and grain products. In this study, we tested whether the improvement in feces consistency expected after an omission of vegetables from a conventional diet was also accompanied by a change in the excretion pattern as measured by the mean retention time (MRT). Two adult Javan langurs were kept together and offered a conventional diet of pellets, browse, vegetables, or a diet consisting of pellets and browse only. MRT were measured with cobalt-EDTA as the fluid and chromium oxide as the particle marker before, during, and after the test diet, collecting the feces of both animals together. MRTs of fluids and particles were longer (47/49 h) on the test diet as compared to the usual diet (42/43 h). Feces consistency improved during the test period. The test period was marked by an increased fiber intake; however, on the conventional diet, dietary water intake exceeded the calculated water flux for these animals distinctively, due to the high proportion of vegetables. Therefore, it cannot be decided whether the increase of fiber level alone or also a reduction of the excessive water intake was responsible for the changes observed. Comparing the results of this study to other studies on ingesta retention in foregut fermenting primates, no correlation between the body mass of the animals and the MRTs measured is evident, which is in accordance with observations in other groups of foregut fermenters.  相似文献

Processing of ingesta particles plays a crucial role in the digestive physiology of herbivores. In the ruminant forestomach different sized particles are stratified into a small and a large particle fraction and only the latter is regurgitated and remasticated to smaller, easier-to-digest particles. In contrast, it has been suggested that in non-ruminating foregut fermenters, such as hippopotamuses, larger particles should be selectively excreted since they tend to be digested at a slower rate and hence can be considered intake-limiting bulk. In our study we determined the mean retention time (MRT) of fluids and different sized particles (2 mm and 10 mm) in six pygmy hippos (Hexaprotodon liberiensis) and six banteng (Bos javanicus) on a diet of fresh grass at two intake levels. We used cobalt ethylendiamintetraacetate (Co-EDTA) as fluid and chromium (Cr)-mordanted fibre (2 mm) and cerium (Ce)-mordanted fibre (10 mm) as particle markers, mixed in the food. Average total tract MRT for fluid, small and large particles at the high intake level was 32, 76 and 73 h in pygmy hippos and 25, 56 and 60 h in banteng, and at the low intake level 39, 109, and 105 h in pygmy hippos and 22, 51 and 58 h in banteng, respectively. In accordance with the prediction, large particles moved faster than, or as fast as the small particles, through the gut of pygmy hippos. In contrast, large particles were excreted slower than the small particles in the ruminant of this study, the banteng. Pygmy hippos had longer retention times than the banteng, which probably compensate for the less efficient particle size reduction. Although the results were not as distinct as expected, most likely due to the fact that ingestive mastication of the larger particle marker could not be prevented, they confirm our hypothesis of a functional difference in selective particle retention between ruminating and non-ruminating foregut fermenters.  相似文献

There are limited data on the diet dry matter digestibility (DMD) of captive African elephants. Although the total fecal collection method is the standard for determining DMD, it is labor‐intensive, time‐consuming, and expensive. The acid‐insoluble ash (AIA) marker technique has been used successfully to determine DMD in ruminants and monogastrics. The objective of this study was to assess how accurately the AIA marker technique could estimate the DMD of captive African elephants (Loxodonta africana). Three mature male African elephants at Disney's Animal Kingdom in Florida were used in this study. The animals were offered a Bermuda grass hay‐based ration, and the total dry matter intake (DMI) and total fecal output were measured daily over a 7‐day period to determine the total collection DMD. The feed ingredients and fecal samples were also analyzed for AIA. Although there were differences (P<0.05) in total DMI and total fecal outputs, the DMD values did not (P>0.05) differ (35.1±0.72 vs. 37.1±0.72 for total collection and AIA, respectively). There was a linear (y=0.9461x; R2=0.74) relationship between the total collection and AIA marker technique DMD values. Direct measurement of individual animal dry matter intake (DMI) remains a fundamental challenge to assessing dairy feed efficiency (FE). Digesta marker, is currently the most used indirect technique for estimating DMI in production animals. In this meta-analysis we evaluated the performance of marker-based estimates against direct or observed measurements and developed equations for the prediction of FE (g energy-corrected milk (ECM)/kg DMI). Data were taken from 29 change-over studies consisting of 416 cow-within period observations. Most studies used more than one digesta marker. So, for each observed measurement of DMI, faecal dry matter output (FDMO) and apparent total tract dry matter digestibility (DMD), there was one or more corresponding marker estimate. There were 924, 409 and 846 observations for estimated FDMO (eFDMO), estimated apparent total tract DMD (eDMD) and estimated DMI (eDMI), respectively. The experimental diets were based mainly on grass silage, with soya bean or rapeseed meal as protein supplements and cereal grains or by-products as energy supplements. Across all diets, average forage to concentrate ratio on a dry matter (DM) basis was 59 : 41. Variance component and repeatability estimates of observed and marker estimations were determined using random factors in mixed procedures of SAS. Between-cow CV in observed FDMO, DMD and DMI was, 10.3, 1.69 and 8.04, respectively. Overall, the repeatability estimates of observed variables were greater than their corresponding marker-based estimates of repeatability. Regression of observed measurements on marker-based estimates gave good relationships (R2=0.87, 0.68, 0.74 and 0.74, relative prediction error =10.9%, 6.5%, 15.4% and 18.7%for FDMO, DMD, DMI and FE predictions, respectively). Despite this, the mean and slope biases were statistically significant (P<0.001) for all regressions. More than half of the errors in all regressions were due to mean and slope biases (52.4% 87.4%, 82.9% and 85.8% for FDMO, DMD, DMI and FE, respectively), whereas the contributions of random errors were small. Based on residual variance, the best model for predicting FE developed from the dataset was FE (g ECM/kg DMI)=1179(±54.1) +38.2(±2.05)×ECM(kg/day)−0.64(±0.051)×BW (kg)−75.6(±4.39)×eFDMO (kg/day). Although eDMD was positively related to FE, it only showed a tendency to reduce the residual variance. Despite inaccuracy in marker procedures, eFDMO from external markers provided a reliable determination for FE measurement. However, DMD estimated by internal markers did not improve prediction of FE, probably reflecting small variability.  相似文献

Whether differences in digestive physiology exist between different ruminant feeding types has been an ongoing debate. In this regard, potential differences in ingesta retention have been understood to be of particular importance. We analyzed a data pool in which only mean retention time (MRT) data for the ruminoreticulum (RR) were collated that were obtained using comparable techniques with either chromium or cobalt EDTA as a fluid marker and/or with chromium-mordanted fiber of less than 2 mm in size as a particle marker. Data were compared using one averaged value per species. In general, the paucity of species in such a collection is striking and does not allow—in contrast to earlier statements—any final conclusions regarding the influence of body weight (BW) or feeding type on ruminant MRTs. In particular, there was no significant correlation between MRTparticlesRR or MRTfluidRR and BW, neither in the interspecific nor in the intraspecific comparisons, and no difference between the feeding types. The trend that indicates longer MRTparticlesRR in grazers is based on too few species to be conclusive. Small browsers seemed to have shorter MRTfluidRR than similar-sized grazers. In contrast, there was a trend for large grazers to have shorter MRTfluidRR than large browsers. In direct pair-wise comparisons between cattle and the browsers giraffe, moose, and okapi, the latter difference was significant. Cattle also had the highest relative RR fluid outflow rates among the species investigated. This is in accord with the observation that grazers have larger omasa, a major function of which is water-reabsorption distal to the RR. Grazers seem to have longer MRTparticlesRR per unit MRTfluidRR, and cattle are particular outliers in this respect. It is hypothesized that potentially shorter MRTfluidRR in large grazers and higher relative outflow rates are linked to a higher saliva production and a lesser viscosity of both saliva and RR fluids. A constant supply of a fluid phase of low viscosity is proposed to be the prerogative for the physical mechanisms of flotation and sedimentation that result in the stratification of RR contents and its selective particle retention typical for large grazing species.  相似文献

Intake measurements were carried out in 22 tapirs from seven UK zoological collections. Dry matter intake (DMI) ranged from 48 to 86 g/kg0.75/d. Across collections, the highest proportion of the ingested diet consisted of pelleted feeds (including grains and bread) at 46±17% DMI, followed by commercial produce at 26±12% DMI, roughage (excluding browse) at 17±11% DMI, and browse at 11±11% DMI. The proportion of roughage, crude protein, crude fiber, and neutral detergent fiber levels in the diets investigated were well below levels recommended for domestic horses and other ungulates. Intakes of digestible energy (DE) as estimated from food nutrients using of a standard equation for domestic horses ranged from 0.58 to 0.88 MJ/DE/kg0.75/d, with many individuals exceeding the assumed maintenance requirement of 0.6 MJ/DE/kg0.75/d for hindgut fermenters. At values exceeding this DE intake, animals had higher than ideal body condition scores (BCS). Animals with higher BCS (i.e. more obese animals) generally had higher fecal scores (FS) (i.e. softer feces), and both BCS and FS were positively correlated to DMI and calculated DE intake. This suggests that the population studied was generally overfed, with resulting obesity and softer fecal consistency. The use of highly digestible feeds such as commercial produce and pelleted feeds should be restricted in the diets of these animals and roughage intake promoted in order to attempt to achieve normal BCS and FS in this captive population. The objective of this study was to determine the effects of varying forage particle length on chewing activity, sorting behavior, rumen pH and rumen fill in late lactation and dry dairy cattle, fed rations with similar physically effective NDF but different mean particle length. Treatments consisted of three diets differing only in geometric mean length of forage: hay (5.40, 8.96 and 77.90 mm, for short (S), medium (M) and long (L) diets, respectively) for Experiment 1 (E1), and straw (10.16, 24.68 and 80.37 mm) for S, M and L diets, respectively, for Experiment 2 (E2). Hay or straw comprised the sole source of forage (50% and 75% of ration dry matter (DM) for E1 and E2, respectively). Both experiments used three rumen cannulated Holstein dairy cows, in late lactation for E1 and dry in E2, with 3 × 3 Latin square designs with 14 day periods. In E1, DM intake (DMI; 18.3 ± 2.1 kg/day; mean ± s.e.), pH (6.4 ± 0.1), time spent eating (280 ± 22.5 min/day), time spent ruminating (487 ± 17 min/day), and total time spent chewing (767 ± 34 min/day) were not different, whereas eating minutes per kilogram of DMI and NDF intake (NDFI) tended to increase linearly as forage length increased. Rumen digesta volume (l; 113.3 S, 117.8 M and 114.4 L ± 17.1) had a quadratic response, and rumen digesta weight tended to respond quadratically; however, differences were numerically small. In E2, DMI (8.3 ± 1.3 kg/day), pH (6.7 ± 0.1), time spent eating (236 ± 23.5 min/day), time spent ruminating (468 ± 45.2 min/day), total time spent chewing (704 ± 67.7 min/day) and minutes per kilogram NDFI were not different, whereas minutes per kilogram of DMI had a trend for a quadratic effect. Rumen digesta volume (111 ± 18.8 l) and weight (103 ± 17.4 kg) were not different. In both experiments, cows sorted against longer particles as determined by a particle length selection index; this behavior increased linearly as particle length increased. Greater forage particle length increased sorting behavior, but had no effect on rumen fermentation or chewing behavior.  相似文献

The orientation to and landing on a source of human odour by female Culex quinquefasciatus Say (Diptera: Culicidae) is observed in a wind tunnel without an airflow or with a laminar airflow of 0.2 m s?1. Odours from human feet are collected by ‘wearing’ clean glass beads inside a stocking and presenting beads in a Petri dish in a wind tunnel. Mosquitoes are activated by brief exposure to a 1 L min?1 jet of 4% CO2 positioned 10 cm from the release cage. In moving air at 0.2 m s?1, a mean ± SE of 3.45 ± 0.49 landings are observed in 10‐min trials (five mosquitoes per trial), whereas 6.50 ± 0.96 landings are recorded in still air. Furthermore, 1.45 ± 0.31 mosquitoes are recorded on beads at any one time in moving air (a measure of individuals landing versus one landing multiple times) compared with 3.10 ± 0.31 in still air. Upwind flight to beads in moving air is demonstrated by angular headings of flight immediately before landing, whereas approaches to beads in still air are oriented randomly. The mean ± SE latency until first landing is 226.7 ± 17.98 s in moving air compared with 122.5 ± 24.18 in still air. Strategies used to locate a prospective host at close range in still air are considered.  相似文献

In contrast to the domestic horse, whose digestive physiology has been thoroughly investigated, knowledge on the digestive physiology of wild equids is scarce. Comparisons between the domestic horse and the domestic donkey suggest that wild asses might achieve higher digestibilities. This could derive from longer retention times or a greater difference in the mean retention time (MRT) of particles vs. fluid (the selectivity factor (SF)). Here, we measured MRT of a solute (fluid; MRTsolute) and a particle (<2 mm; MRTparticle) marker in five captive male Somali wild asses (Equus africanus somaliensis) fed a diet of 95% grass hay. At a mean dry matter intake of 94 ± 3 g kg?0.75 day?1, MRTsolute was 33.3 ± 5.4 h and MRTparticle 39.6 ± 3.9 h, resulting in a SF of 1.21 ± 0.14. For their food intake, Somali wild asses appeared to have slightly higher MRTparticle than expected based on domestic equid data, in contrast to Grevy zebras (Equus grevyi), potentially indicating higher capacities of the digestive tract. However, considering data on domestic horses, donkeys, and zebra, there was no evident difference in the SF of wild equids compared to domestic ones. Together with an absence of reported anatomical differences in the digestive tract of wild and domestic equids, the data suggest a general similarity in the digestive physiology of equid species that contrasts with the diversity in the digestive physiology of ruminants, and that might be one contributing factor to a lack of sympatric, niche-differentiated equid species.  相似文献

Teleost retinal pigment epithelial (RPE) cells contain pigment granules within apical projections which undergo actin‐dependent, bi‐directional motility. Dissociated RPE cells in culture attach to the substrate and extend apical projections in a radial array from the central cell body. Pigment granules within projections can be triggered to aggregate or disperse by the presence or absence of 1 mM cAMP. Aminated, fluorescent latex beads attached to the dorsal surface of apical projections and moved in the retrograde direction, towards the cell body. Bead rates on RPE cells with aggregating or fully aggregated pigment granules were 2.2 ± 0.5 and 2.6 ± 0.2 μm/min (mean ± SEM), respectively, similar to rates of aggregating (retrograde) pigment granule movement (2.0 ± 0.4 μm/min). Bead rates were slightly slower on cells with fully dispersed or dispersing pigment granules (1.5 ± 0.1 and 1.5 ± 0.4 μm/min). Movements of surface‐attached beads and aggregating pigment granules were closely correlated in the distal portions of apical projections, but were more independent of each other in proximal regions of the projections. The actin disrupting drug, cytochalasin D (CD), reversibly halted retrograde bead movements, suggesting that motility of surface‐attached particles is actin‐dependent. In contrast, the microtubule depolymerizing drug, nocodazole, had no effect on retrograde bead motility. The similar characteristics and actin‐dependence of retrograde bead movements and aggregating pigment granules suggest a correlation between these two processes.  相似文献

Quantifying cortisol concentration in hair is a non‐invasive biomarker of long‐term hypothalamic‐pituitary‐adrenal (HPA) activation, and thus can provide important information on laboratory animal health. Marmosets (Callithrix jacchus) and capuchins (Cebus apella) are New World primates increasingly used in biomedical and neuroscience research, yet published hair cortisol concentrations for these species are limited. Review of the existing published hair cortisol values from marmosets reveals highly discrepant values and the use of variable techniques for hair collection, processing, and cortisol extraction. In this investigation we utilized a well‐established, standardized protocol to extract and quantify cortisol from marmoset (n = 12) and capuchin (n = 4) hair. Shaved hair samples were collected from the upper thigh during scheduled exams and analyzed via methanol extraction and enzyme immunoassay. In marmosets, hair cortisol concentration ranged from 2,710 to 6,267 pg/mg and averaged 4,070 ± 304 pg/mg. In capuchins, hair cortisol concentration ranged from 621 to 2,089 pg/mg and averaged 1,092 ± 338 pg/mg. Hair cortisol concentration was significantly different between marmosets and capuchins, with marmosets having higher concentrations than capuchins. The incorporation of hair cortisol analysis into research protocols provides a non‐invasive measure of HPA axis activity over time, which offers insight into animal health. Utilization of standard protocols across laboratories is essential to obtaining valid measurements and allowing for valuable future cross‐species comparisons.  相似文献

Objective: Obesity has been proposed to negatively impact cardiac function in overweight (OW) individuals. The relationship between diastolic dysfunction and oxygen uptake (V?o 2) kinetics is equivocal. This exploratory investigation evaluated the relationship between resting left ventricular function and V?o 2 kinetics during cycle ergometry in OW and non‐overweight (NO) children and adolescents. Research Methods and Procedures: Fourteen OW (>85 percentile for BMI for age and gender) children, 10 boys and 4 girls (age, 11.7 ± 1.9 years; body mass, 80.6 ± 45.5 kg) and 10 NO children (4 boys, 6 girls) volunteered to participate in the study (age, 12.5 ± 2.1 years; body mass, 45.8 ± 13.8 kg). Resting cardiovascular structure and function were assessed using spectral Doppler echocardiography. All subjects underwent two sub‐maximal exercise stages on a cycle ergometer (3 minutes unloaded and 5 minutes at 50 W, both at a cadence of 50 rpm). Respiratory data were measured on a breath‐by‐breath basis at both workloads and the mean response time (MRT) was calculated. Results: Analysis of the MRT data demonstrated that there were no significant differences between OW and NO (OW, 52.6 ± 11.7 seconds vs. NO, 45.6 ± 7.4 seconds). Significant correlations (p < 0.05) were obtained between MRT V?o 2 and echocardiographic‐derived mitral valve inflow pressure half‐time (r = 0.55) and between MRT V?o 2, and mitral valve inflow deceleration time (r = 0.55). Discussion: The evidence from this research suggests a possible link between left ventricular diastolic function at rest and oxygen uptake kinetics during sub‐maximal exercise in OW and NO children and adolescents.  相似文献

The endocrine control mechanisms for female mammalian aggression have been largely unstudied. Although it has been proposed that androgens may modulate female aggressive behavior in a similar manner to males, very little conclusive evidence exists. Previous work in male marmosets found that post‐encounter increases in testosterone (T) were dependent on the intensity of aggression displayed during the aggressive encounter. We exposed female marmosets (Callithrix kuhlii), a monogamous and biparental primate, to aggressive interactions with unfamiliar intruders. Individual female marmosets exhibited changes in T and estradiol (E2) that are associated with aggressiveness dependent on the intensity of aggression displayed as well as their role during the encounter. Resident females exhibited increased E2 immediately following an encounter in which they displayed high rates of aggression. If resident females received high rates of aggression from the intruder, the resident displayed increased T 24 hr following the encounter. Interestingly, if the female was an intruder in the encounter, the intensity of her aggression was associated with increased cortisol immediately following the trials, whereas received aggression was associated with increased T and E2 immediately following the trial. Female primates do exhibit situation‐dependent changes in gonadal steroids in association with aggression that may serve to prime them for future aggressive interactions. Am. J.
