Summary Attempts to measure the nesting density or territory size of bald eagles led to a fundamental difficulty, inherent in all such measurements on organisms which are distributed along an irregular boundary, such as a coastline. The length of such a boundary is not a meaningful measure, and neither can a meaningful area be associated with each nest. Mandelbrot's (1983) fractal geometry applies to the problem, but has not previously supplied practical units of measurement for fractals such as coastlines or rugged surfaces. A practical method is given for measuring the extent of such fractals, introducing a unit of variable dimension, the metron, which includes the existing SI units of length, area and volume as special cases. A linear measure, the spacing allows densities on fractals of different dimensions to be compared directly. The method is applied to the distribution of bald eagle nests along the coastlines of two islands in the Aleutians, and an extension of the method to handle distributions on mountainsides and island surfaces is indicated.  相似文献

Phosphatidylcholine (PtdCho) can provide lipid second messengers involved in signal transduction pathways. As a measure of phospholipid turnover in response to extracellular stimulation, we investigated differential enhancement of [3H]choline incorporation into PtdCho by phorbol esters. In C6 rat glioma and SK-N-SH human neuroblastoma cells, [3H]PtdCho synthesis was 2–4 fold stimulated by -12-O-tetradecanoylphorbol-13-acetate (-TPA) when [3H]choline was incubated simultaneously with, or 15 min prior to, -TPA treatment. By contrast, in N1E-115 mouse and SK-N-MC human neuroblastoma cells, phorbol esters had no appreciable effect on [3H]choline incorporation; however, in all cells, 200 M oleic acid enhanced PtdCho synthesis, indicating a stimulable process. Alterations by thymeleatoxin (TMT), an activator of conventional PKC isoforms (, and ), were similar to -TPA. We investigated whether expression of specific PKC isoforms might correlate with these effects of phorbol esters on PtdCho synthesis. All cell lines bound phorbol esters, had PKC activity that was translocated by phorbol esters and differentially expressed isoforms of PKC. Northern and western blot analyses, using specific cDNA and antibodies for PKC-,-,-,-,-, and-, revealed that expression of -isoform predominated in C6 and SK-N-SH cells. In contrast, TPA-responsive -isoform predominated in SK-N-MC cells. -PKC was not detected in any cells and only in C6 cells was PKC- present and translocated by -TPA treatment. PKC- was not detected in SK-N-MC cell lines but translocated with TPA treatment in the other three cell lines. PKC- was present in all cells but was unaltered by TPA treatment. A gene encoding a thermo-stable endo--1,4-glucanase was isolated from the thermophilic fungus, Thermoascus aurantiacusIFO9748, and designated as eg1. Induction of this gene expression at 50°C was stronger than at 30°C. The deduced amino acid sequence encoded by eg1 showed that it belongs to the glycoside hydrolase family 5. The cloned gene was expressed in Saccharomyces cerevisiae and the gene product was purified and characterized. No significant activity loss was detected over 2 h at 70°C and the product was stable from pH 3–10. The enzyme was optimally active at 70°C over 20 min and the optimal pH was 6.  相似文献

Anadromous and resident forms of pond smelt, Hypomesus nipponensis, were found to occur in sympatry in Lake Ogawara, Japan. Profiles of Sr: Ca ratios from individuals could be grouped to two patterns (1) a resident pattern with low Sr: Ca ratios from core to edge and (2) an anadromous pattern with relatively low Sr: Ca ratios near the core with abrupt increases in ratios at a location approximately 0.3mm from the core. Spawners smaller than 60mm standard length (SL) were resident, between 60 to 80mm were mixed resident and anadromous, and larger than 80mm were anadromous. Anadromous individuals first migrated after 40 to 82 days from hatching (mean±sd, 59.1±13.5d) and 14.6 to 30.9mm SL (22.2±5.3mm). There was no difference in SL between resident and anadromous individuals during age at first migration, suggesting that size may not be the mechanism for divergence of alternative life history styles.  相似文献

This work is based on a previous observation and on a related mathematical modeling regarding the linear growth of a colony of microorganisms under given conditions. We had previously shown that the growth rate of the colony is merely proportional to the individual exponential multiplication constant, , of the microorganisms.Tiny colonies of penicillium are subjected to different doses of irradiation. The subsequent observation of the colonies' growth rate beautifully furnishes a measure of how the multiplication constant, , of the microorganism is affected by irradiation.The plot of with respect to the irradiation dose, shows a linear interdependence between the two quantities. The extrapolation of this plot easily yields the radiation pasteurization dose of the microorganisms in hand.  相似文献

Summary A negative feedback interaction between luminosity type horizonatal cells (HCs) and green-sensitive cones generates the long-wavelength-sensitive depolarizing response in biphasic chromaticity type HCs. This interaction is suppressed in the dark and is potentiated by light adaptation of the retina. HCs are morphologically plastic; during light adaptation, their dendritic terminals within cone pedicles extend, giving rise to spinules. This paper examines whether there is a quantitative correlation between the time course of light-dependent formation of the spinules and enhancement of the feedback interaction. The strength of the feedback interaction in isolated retinac of the roach was determined as the neutral wavelength at which reversal of spectral response polarity occurred in biphasic HCs. A good correlation was found between the neutral wavelength and the spinule/ribbon ratios of retinae. Biphasic HCs were intracellularly stained with horseradish peroxidase and the correlative ultrastructure of the contacted pedicles was examined. Neutral wavelength was found to be correlated with the spinule number, weighted according to the number of synaptic contacts mediating feed-forward transmission. The latter was estimated from the total number of labelled Cb/H2 HC processes (central and lateral) at synaptic triads. A model in which spinules mediate the negative feedback interaction of HCs in the retina of cyprinid fish is presented.  相似文献

Photolyzed rhodopsin was phosphorylated in bovine rod outer segments incubated at –10 C. In the experiment in which urea-treated outer segments and rhodopsin kinase were incubated with ATP in the presence of 30% glycerol, the extent of phosphate incorporation at –10 C was about 30% of that at 37 C. Separation of phosphorylated rhodopsin by isoelectric focusing indicated that a limited number of sites were phosphorylated at –10 C. The partially phosphorylated pigment incorporated more phosphates when the temperatures was raised to 37 C. This was partly due to decreased inhibition of phosphorylation by glycerol at higher temperature. Since the maximum phosphorylation at –10 C (at which metarhodopsin II is stable) occurred at a pH value (6.0) lower than the pKa for metarhodopsin I-metarhodopsin II transition, metarhodopsin II was suggested to be the preferred substrate for rhodopsin kinase at –10 C. Limited proteolysis with thermolysin of rhodopsin phosphorylated at 37 C released peptides containing about 50% of the total phosphate incorporated. In contrast, proteolytic digestion of rhodopsin phosphorylated at –10 C released negligible amounts of phosphate-containing peptides. The results were taken to suggest that the incorporation of phosphates at metarhodopsin II level under the present condition occurred in the residues other than those removed by thermolysin digestion.Based on material presented at the Fifth International Congress of Eye Research, Eindhoven, October 1982  相似文献

Cultured hypocotyl explants of tomatillo (Physalis ixocarpa Brot.), were evaluated with regard to their morphogenic responses to combinations of benzyladenine (BA, 0–5 M) with either naphthaleneacetic acid (NAA, 0–50 M) or 2,4-dichlorophenoxyacetic acid (2,4-D, 0–50 M). The induction of shoots or roots was dependent on the cytokinin/auxin combination.Hypocotyl explants failed to form shoots when they were grown on media containing either a cytokinin or an auxin alone. The highest frequency of shoot formation was observed on media containing 12.5–25 M BA and 5 M NAA. Likewise the highest frequency of root formation was observed on media supplemented with 1 M BA and 1 M NAA. Complete plants were regenerated and transferred to soil, where they reached maturity.  相似文献

I examine the branch of evolutionary epistemology which tries to account for the character of cognitive mechanisms in animals and humans by extending the biological theory of evolution to the neurophysiological substrates of cognition. Like Plotkin, I construe this branch as a struggling science, and attempt to characterize the sort of theory one might expect to find this truly interdisciplinary endeavor, an endeavor which encompasses not only evolutionary biology, cognitive psychology, and developmental neuroscience, but also and especially, the computational modeling of artificial life programming; I suggest that extending Schaffner's notion of interlevel theories to include both horizontal and vertical levels of abstraction best fits the theories currently being developed in cognitive science. Finally, I support this claim with examples drawn from computational modeling data using the genetic algorithm.  相似文献

Zebra seabream, Diplodus cervinus cervinus, caught off the Canary Islands is characterized by protogynous hermaphroditism. The male:female ratio is in favour of females (1:2.16). The reproductive season extends from spring to summer, with a peak in spawning activity in May–June. Males reach maturity at a larger total length, 327mm (5 years old) than females 273mm (4 years old). Recruitment occurs from late October to January in shallow waters of 0.5–8m depth along the coastal line. The recruits are located over rocky substrates with an important algae vegetation forming schools lower than 2m2. During the spawning season, schools of adults from 3 to 8 fish are observed. The schools are formed by one large individual and a few moderate size individuals. Mating takes place in small groups formed by one dominant male and a group of several females (polygamy). Moderate size individuals are often observed mixed in large schools (up to 30 individuals) of Diplodus sargus cadenati. Subadults form groups of a few fish (<5 individuals) or more commonly mixed groups (>15 individuals) with individuals of species of similar size. Otoliths age readings indicate that the population consists of 18 age groups, including a very high proportion of individuals between 2 and 4 years old. The von Bertalanffy growth parameters for the whole population are: L = 603mm, k = 0.149 year–1, and t0 = –0.22 year. The mean rates of total, natural and fishing mortality are 0.551, 0.215 and 0.336 year–1, respectively. The length at first capture is 183mm. The exploitation rate indicates that the stock is overfished. The direct effects of fishing on the population result in changes in the abundance, with a reduction to 85% of the unexploited equilibrium level. The length at first capture by the commercial fishery is less than the length at maturity. With 58% of the total catch below this length there is a danger of recruitment overfishing.  相似文献

Summary The white-rot fungus Ganoderma australis selectively degrades lignin in the ecosystem palo podrido. Using conditions that simulate those of palo podrido in the laboratory, it was found that low nitrogen content and low O2 tension stimulate the productio of manganese peroxidase and lignin degradation, and depress cellulose degradation and cellulase production. The inverse is found at high nitrogen concentration and high O2 tension. This agrees with previous results indicating that low O2 tension and low nitrogen stimulate selective lignin degradation by this fungus. Correspondence to: J. Eyzaguirre  相似文献

Summary The incidence of myosatellite cells associated with white and red muscle fibres of the parietal muscle and red fibres of the craniovelar muscle was estimated by quantitative electron microscopy in the Atlantic hagfish (Myxine glutinosa, L.). Myosatellite cell nuclei constitute 3, 11 and 23 % of the total number of nuclei inside the basal lamina of the three types of muscle fibres, respectively. However, the total number of nuclei is highest in white fibres, most of the nuclei belonging to striated muscle cells. Myosatellite cell profiles in transverse sections constitute 23, 41 and 61 % of the number of muscle fibre profiles of the three types, respectively. The intervals between adjacent myosatellite cells are 135 m in white fibres, 55 m in red parietal fibres, and only 25 m in craniovelar fibres. Since craniovelar fibres are also comparatively thin, myosatellite cells constitute a significant fraction of the volume inside the basal lamina in these fibres. The myosatellite cells are 30–50 m long and up to 5 m thick. Some myosatellite cells possess few organelles, whereas others appear to contain many free ribosomes, granular endoplasmic reticulum, prominent Golgi apparatus and lysosome-like bodies.This investigation was supported by the Norwegian Research Council for Science and the Humanities (NAVF grant No. C20.30–37). 4-Methylumbelliferyl 6-O-benzyl--d-lactoside (6Bn-MU-Lac) and some related compounds were synthesizedvia different selective reactions including phase-transfer glycosylation. Their suitability as substrates for a fluorometric assay of ceramide glycanase (CGase) was evaluated. Among others, the 6Bn-MU-Lac, which is resistant to exogalactosidase, was found to be a suitable substrate for routine assay of the CGase activity. For American leech CGase, theK m value is 0.232 mM at pH 5. Flora, vegetation and seed bank were studied in a dry Afromontane forest in the Kondoa Irangi Hills, Tanzania with the objective to obtain information necessary for the promotion of conservation of this forest, which is rapidly degenerating due to human pressure. A preliminary checklist of 104 vascular plants occurring in the forest is provided. Based on 27 plots, each of 400m2, the forest had the following ecological features: mean tree density 408 stems·ha–1; basal area 66.56m2·ha–1; Shannon and Wiener diversity 2.6406; evenness 0.7585 and species richness 31 tree species per 1.08ha. The seed bank was dominated by woody species, including mature forest tree species, and had several species in common with the standing vegetation. The forest contains species that are phytogeographically linked to Madagascan, Western and Southern African floras and also species disjunctly distributed in this forest and the Eastern Arc Mountains of Tanzania. Correspondence Analysis of tree species based on size classes showed that the timber species had declining populations, indicating the possibility of genetic erosion, while other species showed expanding or interrupted populations. Some DBH-size classes (e.g. 145.0–189.9cm) have disappeared from the forest, presumably due to their selective removal. We recommend detailed integrated floristic and faunistic studies of the forest, targeting the ecologically sensitive indicators of habitat change such as orchids, birds, reptiles and amphibians. Finally, a proposal to conserve the forest under a community-based forest conservation scheme is put forward.  相似文献

Summary A new cytochemical technique is proposed for side by side localization of acetylcholine and of acetylcholinesterase activity of motor end-plate at ultrastructural level. The technique is based on the simultaneous ionic fixation of vesicular acetylcholine and of histochemical copper thiocholine precipitate with phosphomolybdic acid: the molybdic heteropolyanion forms insoluble salts with these two quaternary ammonium cations, providing in situ acetylcholine phosphomolybdate and copper thiocholine phosphomolybdate. Both of them are osmium resistant; the electron dense precipitates allow for a fine localization of acetylcholine and acetylcholinesterase activity at electron microscopic level.  相似文献

Summary A detailed account of the ultrastructure and behaviour of the spindle pole body (SPB) of the entomophthoraceous fungusErynia neoaphidis is presented for the first time.The SPB consists of extranuclear (ENC) and intranuclear (INC) components. The ENC is a saucepan-shaped structure which lies in a pocket of the nuclear envelope. It is composed of a forked, fibrillar handle and a shallow, cylindrical pan. The pan has a wall of two layers, both of which are thickened with a regular periodicity so that they appear to be beaded. It is postulated that the pan is formed from rough endoplasmic reticulum and that it synthesizes the amorphous, electron-dense material coating the ENC.The INC is a saucer-shaped, electron-dense plaque in which the ends of the spindle microtubules terminate. During metaphase, a clear zone separates the INC from the nuclear envelope and persists until telophase. The roles of the amorphous, electron-dense material and the clear zone as well as the method of SPB replication are discussed.  相似文献

The use of bioreactors may provide an efficient and economic tool for mass clonal propagation of plants if technical problems can be solved. In this paper, we report the results of experiments aimed at optimising conditions for apple rootstock M26 grown in RITA containers using the temporary immersion principle. We tested different types and sizes of explants, different concentrations of plant growth regulators (BAP, kinetin and IBA) in the multiplication and elongation phases, and medium exchange during the shoot elongation period. The results show that the higher concentrations of cytokinins were required during the shoot multiplication phase, while the lower concentrations were better during the shoot elongation phase. Hyperhydricity was increased with increasing concentration in of cytokinins during both shoot multiplication and shoot elongation phases. The best shoot production in terms of shoot number and shoot quality was obtained using 4.4mol BAP and 0.5mol IBA during the shoot multiplication phase and 1.1mol BAP and 0.25mol IBA during the shoot elongation phase. Medium exchange twice during the shoot elongation phase resulted in higher shoot production compared with no exchange of the medium. However, it also resulted in increased hyperhydricity. Immersion frequency of 16 times per day gave a higher multiplication rate and longer shoots than 8 times per day. The explant size of 0.5cm or 1cm resulted in a significantly higher shoot production rate compared with that of 1.5cm, but shoot length and hyperhydricity were not affected by the explant size. Shoot cultures from the liquid media rooted normally in the RITA containers with more than 90% rooting and the rooted plantlets acclimatised well in the greenhouse.  相似文献

Summary The functional R6K origin is composed of two DNA elements, one of 580 bp carrying the origin sequences and the other of 277 bp containing the seven 22 bp direct repeats previosly identified as also required for and origin activity. These two genetic elements are separated by approximately 3,000 bp of R6K sequences which are dispensable for origin activity. The function of the origin depends on the presence in cis of the 580 bp and the 277 bp fragments and requires that they be oriented as in the intact R6K. Activation of the origin depends on the R6K replication initiation protein .Within the 580 bp of the origin, there is a sequence of 98 bp which appears as an inverted repeat of 96 bp in the replicon. Deletion of the 96 bp or 98 bp results in inactivation of the and the origins respectively. These long repeats are palindromic and it is suggested that these may serve as the recognition signals for initiation of DNA replication in the and the origins of R6K. DNA homology analysis performed on , and origin sequences, also reveals 10–23 bp sequences in the and the origins that are related to the family of 22 bp direct repeats in the origin which were shown previously to be binding sites for the protein.  相似文献

Summary TheLap-D locus ofDrosophila melanogaster was investigated. The mode of inheritance of the alleles at this locus was confirmed and a new allele was discovered. The enzymes are not exclusively pupal enzymes but are found throughout the life cycle. Amino acid naphthylamides other than leucine naphthylamide were found to be effective substrates for these enzymes. It is suggested, therefore, that the restrictive designation leucine aminopeptidase be abandoned in favor of the more general amino acid naphthylamidase or aminopeptidase.This work was supported by the U.
