The roles of Na+ and K+ (Rb+) uptake were further studied in a NaCl-tolerant strain of Ceratopteris richardii containing the stl2 mutation by direct comparison with the wild-type strain. In addition to Na+ tolerance, stl2 also confers tolerance to Mg2+ and sensitivity to K+. In addition to higher K+ (Rb+) uptake at concentrations commonly associated with low-affinity K+ transport, stl2 maintained higher uptake down to 0·1 mol m–3 Rb+. Up to a 25-fold excess of Na+ had little effect in either genotype on K+ (Rb+) uptake at low concentrations, i.e. 0·2 and 0·5 mol m–3 RbCl. Pretreatment with K+ (20 mol m–3) inhibited uptake of K+ (Rb+) in the wild type, whereas concurrent inclusion of K+ inhibited uptake of Rb+ more in stl2. In the absence of K+, Na+ uptake (0·01–60 mol m–3) was nearly identical in the wild type and stl2. K+ inhibited Na+ uptake more effectively in stl2 than the wild type, especially at 60 mol m–3 Na+. Greater inhibition of K+ uptake in stl2 occurred with MgCl2 or TEA (tetraethylammonium chloride) preincubation or with simultaneous inclusion of Al3+ (Al2SO4). The higher effective velocity of K+ uptake at a wide range of concentrations and the enhanced selectivity for K+ and against Na+ contribute to the preservation of higher cytosolic K+ and lower Na+ under salinity stress.  相似文献

In order to identify physiological components that contribute to salinity tolerance, we compared the effects of Na+, Mg2+ and K+ salts (NaCl, Na2SO4, MgCl2, MgSO4, KCl and K2SO4), Ca2+ (CaSO4), mannitol and melibiose on the wild type and the single-gene NaCl-tolerant mutants stl1 and stl2 of Ceratopteris richardii. Compared with gametophytic growth of the wild type, stl2 showed a low level of tolerance that was restricted to Na+ salts and osmotic stress. stl2 exhibited high tolerance to both Na+ and Mg2+ salts, as well as to osmotic stress. In response to short-term exposure (3 d) to NaCl, accumulation of K+ and Na+ was similar in the wild type and stl1. In contrast, stl2 accumulated higher levels of K+ and lower levels of Na+. Ca2+ supplementation (1.0 mol m?3) ameliorated growth inhibition by Na+ and Mg2+ stress in wild type and stll, but not in stl2. In addition, under Na+ stress (175 mol m?3) wild-type, stll and stl2 gametopbytes maintained higher tissue levels of K+ and lower levels of Na+ when supplemented with Ca2+ (1.0 mol m?3). stl2 gametophytes were extremely sensitive to K+ supplementation. Growth of stl2 was greater than or equal to that of the wild type at trace concentrations of K+ but decreased substantially with increasing K+ concentration. Supplementation with K+ from 0 to 1.85 mol m?3 alleviated some of the inhibition by 75 mol m?3 NaCl in the wild type and in stl1. In stl2, growth at 75 mol m?3 NaCl was similar at 0 and 1.85 mol m?3 K+ supplementation. Although K+ supplementation above 1.85 mol m?3 did not alleviate inhibition of growth by Na+ in any genotype, stl2 maintained greater relative tolerance to NaCl at all K+ concentrations tested.  相似文献

Previous work showed that in hamster red cells the amiloride-sensitive (AS) Na+ influx of 0.8 mmol/liter cells/hr is not mediated by Na-H exchange as in other red cells, but depends upon intracellular Mg2+ and can be increased by 40-fold by loading cells with Mg2+ to 10 mm. The purpose of this study was to verify the connection of AS Na+ influx with Na-dependent, amiloride-sensitive Mg2+ efflux and to utilize AS Na+ influx to explore that pathway.Determination of unidirectional influx of Na+ and net loss of Mg2+ in parallel sets of cells showed that activation by extracellular [Na+] follows a simple Michaelis-Menten relationship for both processes with a K m of 105–107 mm and that activation of both processes is sigmoidally dependent upon cytoplasmic [Mg2+] with a [Mg2+]0.5 of 2.1–2.3 mm and a Hill coefficient of 1.8. Comparison of Vmax for both sets of experiments indicated a stoichiometry of 2 Na: l Mg. Amiloride inhibits Na+ influx and Mg2+ extrusion in parallel (K i = 0.3 mm). Like Mg2+ extrusion, amiloride-sensitive Na+ influx shows an absolute requirement for cytoplasmic ATP and is increased by cell swelling. Hence, amiloride-sensitive Na+ influx in hamster red cells appears to be through the Na-Mg exchange pathway.There was no amiloride-sensitive Na+ efflux in hamster red cells loaded with Na+ and incubated with high [Mg2+] in the medium with or without external Na+, nor with ATP depletion. A mutant in Saccharomyces cerevisiae required one hundred times more K+ than wild type for the same half maximal growth rate. Mutant cells and wild type cells grown at millimolar K+ did not show significant differences in Rb+ transport. In the mutant, a rapid K+ loss induced by azide or incubation (4 h) in K+-free medium decreased the Rb+ transport K m by one half; in the wild type, those treatments decreased the Rb+ K m twenty and one hundred times, respectively. Mutant and wild type did not show significant differences in Na+ transport and in the Na+ inhibition of Rb+ transport, either in normal-K+ cells or in K+-starved cells. Summary Kinetic properties of Na+–Ca2+ exchange in a renal epithelial cell line (LLC-MK2) were assessed by measuring cytosolic free Ca2+ with fura-2 and45Ca2+ influx. Replacing external Na+ with K+ produced relatively small increases in free Ca2+ and45Ca2+ uptake unless the cells were incubated with ouabain. Ouabain markedly increased cell Na+ and strongly potentiated the effect of replacing external Na+ with K+ on free Ca2+ and45Ca2+ uptake.45Ca2+ influx in 140mm K+ or N-methyl-d-glucamine minus influx in 140mm Na+ was used to quantify Na+–Ca2+ exchange activity of Na+-loaded cells. The dependence of exchange on cell Na+ was sigmoidal; theK 0.5 was 26±3 mmol/liter cell water space, and the Hill coefficient was 3.1±0.2. The kinetic features of the dependence of exchange on cell Na+ partly account for the small increase in Ca2+ influx when all external Na+ is replaced by K+. Besides raising cell Na+ ouabain appears to activate the exchanger. Magnesium competitively inhibited exchange activity. The potency of Mg2+ was 8.2-fold lower with potassium instead of N-methyl-d-glucamine or choline as the replacement for external Na+. Potassium also increased theV max of exchange by 86% and had no effect on theK m for Ca2+. The exchanger does not cause detectable22Na+–Mg2+ exchange and does not appear to require K+ or transport86Rb+. Although exchange activity was plentiful in the epithelial cells from monkey kidney, others from amphibian, canine, opossum, and porcine kidney had no detectable exchange activity. All of the measured kinetic properties of Na+–Ca2+ exchange in the renal epithelial cells are very similar to those of the exchanger in rat aortic myocytes.  相似文献

Effects of interrupted K+ supply on different parameters of growth and mineral cation nutrition were evaluated for spring wheat (Triticum aestivum L. cv. Svenno). K+ (2.0 mM) was supplied to the plants during different periods in an otherwise complete nutrient solution. Shoot growth was reduced before root growth after interruption in K+ supply. Root structure was greatly affected by the length of the period in K+ -free nutrient solution. Root length was minimal, and root branching was maximal within a narrow range of K+ status of the roots. This range corresponded to cultivation for the last 1 to 3 days, of 11 in total, in K+ -free nutrient solution, or to continuous cultivation in solution containing 0.5 to 2 mM K+. In comparison, both higher and lower internal/external K+ concentrations had inhibitory effects on root branching. However, the differing root morphology probably had no significant influence on the magnitude of Ca2+, Mg2+ and Na+ uptake. Uptake of Ca2+ and especially Mg2+ significantly increased after K+ interruption, while Na+ uptake was constant in the roots and slowly increased in the shoots. The two divalent cations could replace K+ in the cells and maintain electroneutrality down to a certain minimal range of K+ concentrations. This range was significantly higher in the shoot [110 to 140 μmol (g fresh weight)?1] than in the root [20 to 30 μmol (g fresh weight)?1]. It is suggested that the critical K+ values are a measure of the minimal amount of K+ that must be present for physiological activity in the cells. At the critical levels, K+ (86Rb) influx and Ca2+ and Mg2+ concentrations were maximal. Below the critical K+ values, growth was reduced, and Ca2+ and Mg2+ could no longer substitute for K+ for electrostatic balance. In a short-term experiment, the ability of Ca2+ to compete with K+ in maintaining electroneutrality in the cells was studied in wheat seedlings with different K+ status. The results indicate that K+, which was taken up actively and fastest at the external K+ concentration used (2.0 mM), partly determines the size of Ca2+ influx.  相似文献

Detached internodes of Chara corallina survived in solutions containing 100 mol m?3 NaCl when the external concentration of Ca2+ was greater than 1 mol m?3. Na+ influx was roughly proportional to external Na+ up to 100 mol m?3 NaCl. Na+ influx involved two components: a Ca2+-insensitive influx which allowed the passage of Na+ independently of external Ca2+; and a Ca2+-inhibitable mechanism where Na+ influx was inversely proportional to external Ca2+. The Ca2+-inhibitable Na+ influx was similar to the Ca2+-inhibitable K+ influx. Mg2+ and Ba2+ were able to substitute for Ca2+ in partially inhibiting Na+ influx in the absence of external Ca2+. The effect of Ca2+ appears specific to Na+ and K+ influx since the effects of a Ca2+-free solution on the influx of some other cations, anions and neutral compounds is small. It is suggested that Na+ influx via the Ca2+-inhibitable mechanism represents Na+ leakage through K+ channels and that cell death at high salinity occurs due to a cytotoxic Na+ influx via this mechanism.  相似文献

Synechococcus R-2 is a unicellular blue-green alga. The cells will grow on Rb+ as a substitute for K+ but at a slower rate (t2~ 15 h versus 12 h). Potassium is not, strictly speaking, an essential element for Synechococcus. Rubidium duxes (using 86Rb+) are much slower than those of potassium, about 1 nmol m?2 s?1 in the light (0.35 mol m?3 Rb+). 86Rb+ fluxes in the dark are about 0.1 nmol m?2 s?1. These fluxes are very slow compared to those of Na+ and other ions. Isotopic influx of Rb+ can supply sufficient Rb+ to keep up with the demands for growth, but the net dux needed to keep up with growth in the light is a large proportion of the total observed dux. Kinetic studies of Rb+ uptake versus [Rb+] show two uptake phases consistent with a high-affinity and a low-affinity system. Both systems appear to be light-activated. Transport of Rb+ appears to be passive at pHo 10 in the light and dark. There is no case for active transport of Rb+ at pHo 7.5 in the light, but a marginal case for active uptake in the dark (about 3 kJ mol?1). There is only a small effect of Na+ upon Rb+ transport. 86Rb+ should not be used in place of 42K+ in K+ nutrition studies as the details of Rb+ transport are different to those of K+ transport.  相似文献

Right-side-out plasma membrane vesicles were isolated from wheat roots using an aqueous polymer two-phase system. The purity and orientation of the vesicles were confirmed by marker enzyme analysis. Membrane potential (Ψ)-dependent 22Na+ influx and sodium/proton (Na+/ H+) antiport-mediated efflux across the plasma membrane were studied using these vesicles. Membrane potentials were imposed on the vesicles using either K+ gradients in the presence of valinomycin or H+ gradients. The ΔΨ was quantified by the uptake of the lipophilic cation tetraphenylphosphonium. Uptake of Na+ into the vesicles was stimulated by a negative ΔΨ and had a Km for extrav-esicular Na+ of 34.8 ± 5.9 mol m3. The ΔΨ-dependent uptake of Na+ was similar in vesicles from roots of hexaploid (cv. Troy) and tetraploid (cv. Langdon) wheat differing in a K+/Na+ discrimination trait, and was also unaffected by growth in 50 mol m?3 NaCl. Inhibition of ΔΨ-dependent Na+ uptake by Ca2+ was greater in the hexaploid than in the tetraploid. Sodium/proton antiport was measured as Na+-dependent, amiloride-inhibited pH gradient formation in the vesicles. Acidification of the vesicle interior was measured by the uptake of 14C-methylamine. The Na+/H+ antiport had a Km, for intravesicular Na+ of between 13 and 19 mol m?3. In the hexaploid, Na+/H+ antiport activity was greater when roots were grown in the presence of 50 mol m?3NaCl, and was also greater than the activity in salt-grown tetraploid wheat roots. Antiport activity was not increased in a Langdon 4D chromosome substitution line which carries a trait for K+/Na+ discrimination. It is concluded that neither of the transport processes measured is responsible for the Na+/K+ discrimination trait located on the 4D chromosome of wheat.  相似文献

The stl1 and stl2 mutations confer low and high levels of NaCl tolerance to gametophytes of the fern Ceratopteris richardii, respectively. As an initial characterization of these mutations, the levels of various organic solutes, tissue ion content and water relations were examined in the wild-type and mutant strains in the absence and presence of 60 mol m-3 NaCl stress (a level which results in a 20, 15 and 0% reduction in gametophyte growth in the wild-type, stl1 mutant and stl2 mutant, respectively). All strains exhibited major changes in organic and inorganic solute levels and water relations in response to 60 mol m-3 NaCl stress. Differences in organic solute levels and water relations between the wild-type and mutant strains in the absence and in response to 60 mol m-3 NaCl stress were minimal. Analysis of tissue ion content showed that stl1 was associated with a slight reduction in Na+ accumulation during 60 mol m-3 NaCl stress. stl2 was associated with (1) higher constitutive levels of K+ and (2) continued selective accumulation of K+ and reduced accumulation of Na+ during 60 mol m-3 NaCl stress. A K+/Na+ ratio close to 1 was observed in the wild-type during 60 mol m-3 NaCl stress, while higher ratios were detected in stl1 and stl2 (1·7 and 4·0, respectively). The findings of this study suggest that the tolerance imparted by stl1 and stl2 is associated with altered ion accumulation during NaCl stress, rather than an enhanced ability to accumulate organic solutes to be used for osmotic adjustment of the cytoplasm.  相似文献

It has been shown that addition of phosphate to phosphate deficient yeast gives rise to an immediate increase in the rate of Na+ uptake and an immediate decrease in the rate of Rb+ uptake. In addition, phosphate uptake is enhanced specifically by Na ions presumably by a process with a very high affinity for phosphate with a Km of about 2 × 10−6M at pH 7.2, whereas the Km for phosphate uptake of the Na+ independent process amounts to 1.3 × 10−4M.  相似文献

Abstract: Bovine chromaffin secretory vesicle ghosts loaded with Na+ were found to take up Ca2+ when incubated in K+ media or in sucrose media containing micromolar concentrations of free Ca2+. Li+- or choline+loaded ghosts did not take up Ca2+. The Ca2+ accumulated by Na+-loaded ghosts could be released by the Ca2+ ionophore A23187, but not by EGTA. Ca2+ uptake was inhibited by external Sr2+, Na +, Li +, or choline +. All the 45Ca2+ accumulated by Na+-dependent Ca2+ uptake could be released by external Na +, indicating that both Ca2+ influx and efflux occur in a Na+-dependent manner. Na + -dependent Ca2+ uptake and release were only slightly inhibited by Mg2+. In the presence of the Na+ ionophore Monensin the Ca2+ uptake by Na +-loaded ghosts was reduced. Ca2+ sequestered by the Na+-dependent mechanism could also be released by external Ca2+ or Sr2+ but not by Mg2+, indicating the presence of a Ca2+/Ca2+ exchange activity in secretory membrane vesicles. This Ca2+/Ca2+ exchange system is inhibited by Mg2+, but not by Sr2+. The Na + -dependent Ca2+ uptake system in the presence of Mg2+ is a saturable process with an apparent Km of 0.28 μM and a Vmax= 14.5 nmol min?1 mg protein?1. Ruthenium red inhibited neither the Na+/Ca2+ nor the Ca2+/Ca2+ exchange, even at high concentrations.  相似文献

In frog red blood cells, K-Cl cotransport (i.e., the difference between ouabain-resistant K fluxes in Cl and NO3) has been shown to mediate a large fraction of the total K+ transport. In the present study, Cl-dependent and Cl-independent K+ fluxes via frog erythrocyte membranes were investigated as a function of external and internal K+ ([K+] e and [K+] i ) concentration. The dependence of ouabain-resistant Cl-dependent K+ (86Rb) influx on [K+] e over the range 0–20 mm fitted the Michaelis-Menten equation, with an apparent affinity (K m ) of 8.2 ± 1.3 mm and maximal velocity (V max ) of 10.4 ± 1.6 mmol/l cells/hr under isotonic conditions. Hypotonic stimulation of the Cl-dependent K+ influx increased both K m (12.8 ± 1.7 mm, P < 0.05) and V max (20.2 ± 2.9 mmol/l/hr, P < 0.001). Raising [K+] e above 20 mm in isotonic media significantly reduced the Cl-dependent K+ influx due to a reciprocal decrease of the external Na+ ([Na+] e ) concentration below 50 mm. Replacing [Na+] e by NMDG+ markedly decreased V max (3.2 ± 0.7 mmol/l/hr, P < 0.001) and increased K m (15.7 ± 2.1 mm, P < 0.03) of Cl-dependent K+ influx. Moreover, NMDG+ Cl substitution for NaCl in isotonic and hypotonic media containing 10 mm RbCl significantly reduced both Rb+ uptake and K+ loss from red cells. Cell swelling did not affect the Na+-dependent changes in Rb+ uptake and K+ loss. In a nominally K+(Rb+)-free medium, net K+ loss was reduced after lowering [Na+] e below 50 mm. These results indicate that over 50 mm [Na+] e is required for complete activation of the K-Cl cotransporter. In nystatin-pretreated cells with various intracellular K+, Cl-dependent K+ loss in K+-free media was a linear function of [K+] i , with a rate constant of 0.11 ± 0.01 and 0.18 ± 0.008 hr−1 (P < 0.001) in isotonic and hypotonic media, respectively. Thus K-Cl cotransport in frog erythrocytes exhibits a strong asymmetry with respect to transported K+ ions. The residual, ouabain-resistant K+ fluxes in NO3 were only 5–10% of the total and were well fitted to linear regressions. The rate constants for the residual influxes were not different from those for K+ effluxes in isotonic (∼0.014 hr−1) and hypotonic (∼0.022 hr−1) media, but cell swelling resulted in a significant increase in the rate constants. Abstract The freshwater Charophyte Chora corallina dies when subjected to 70 molm?3 NaCl if the Ca2+ concentration is 0.1 mol m ?3. This stress is accompanied by a depolarization of the cell to a membrane potential more positive than EK, a net influx of Na+ into the vacuole, and a net loss of K+ from the vacuole. Raising the Ca2+ concentration to 7 mol m ?3 in the presence of elevated Na+ restores the Na+ to Ca2+ ratio to 10: 1 as in the control solution, and results in enhanced survival even though turgor is not regulated. Mg2+ is not a good substitute for Ca2+. It is suggested that the main reason that C. corallina fails to occupy saline habitats is its failure to regulate turgor, not sensitivity to Na +, since the latter is similar to that seen in C. buckellii, which is found in saline habitats.  相似文献

Steady state kinetics were used to examine the influence of Cd2+ both on K+ stimulation of a membrane-bound ATPase from sugar beet roots (Beta vulgaris L. cv. Monohill) and on K+(86Rb+) uptake in intact or excised beet roots. The in vitro effect of Cd2+ was studied both on a 12000–25000 g root fraction of the (Na++K++Mg2+)ATPase and on the ATPase when further purified by an aqueous polymer two-phase system. The observed data can be summarized as follows: 1) Cd2+ at high concentrations (>100 μM) inhibits the MgATPase activity in a competitive way, probably by forming a complex with ATP. 2) Cd2+ at concentrations <100 μM inhibits the specific K+ activation at both high and low affinity sites for K+. The inhibition pattern appears to be the same in the two ATPase preparations of different purity. In the presence of the substrate MgATP, and at K+ <5 mM, the inhibition by Cd2+ with respect to K+ is uncompetitive. In the presence of MgATP and K+ >10 μM, the inhibition by Cd2+ is competitive. 3) At the low concentrations of K+, Cd2+ also inhibits the 2,4-dinitrophenol(DNP)-sensitive (metabolic) K+(86Rb+) uptake uncompetitively both in excised roots and in roots of intact plants. 4) The DNP-insensitive (non metabolic) K+(86Rb+) uptake is little influenced by Cd2+. As Cd2+ inhibits the metabolic uptake of K+(86Rb+) and the K+ activation of the ATPase in the same way at low concentrations of K+, the same binding site is probably involved. Therefore, under field conditions, when the concentration of K+ is low, the presence of Cd2+ could be disadvantageous.  相似文献

A microsomal (Na++ K++ Mg2+)ATPase preparation from sugar beet roots was used. The activation by simultaneous addition of Na+ and K+ at different levels was examined in terms of steady state kinetics. The observed data can be summarized in the following way: 1. The apparent affinity between the enzyme and the substrate MgATP depends on the ratio between Na+ and K+. At low Na+ concentration (below 5 mM), the apparent Km decreases with increasing concentrations of K+ (1–20 mM). At 5 mM Na+, the K+ level does not change the apparent Km, while at Na+ levels above 10 mM, the apparent Km between enzyme and substrate increases with increasing concentration of K+. 2. When the MgATP concentration is kept constant, homotropic cooperativity (concerning one type of ligand) and heterotropic cooperativity (concerning different types of ligands) exist in the activation by Na+ and K+. The Na+ binding is cooperative with different Km values and Hill coefficients (n) in the presence of low and high concentration of K+. At low Na+ level (< 5 mM). a negative cooperativity exists for Na+ (nNa < 1) which is more pronounced in the presence of high [K+]. When the concentration of Na+ is raised the negative cooperativity disappears and turns into a positive one (nNa > 1). Only K+ binding in the presence of low [Na+] shows cooperativity with a Hill coefficient that reflects changes from negative to positive homotropic cooperativity with increasing concentrations of K+ (nK < 1 → nK > 1). In the presence of [Na+] > 10 mM, the changes in nk are insignificant. 3. A model is proposed in which one or two different K sites and one or two Na sites control the catalytic activity, with multiple interactions between Na+, K+ and MgATP. 4. In the presence of Na+ (< 10 mM), K+ is probably bound to two K sites, one of which translocates K+ through the membrane by an antiport Na+/K+ mechanism. This could be connected with an elevated K+ uptake in the presence of Na+ and could therefore explain some field properties of sugar beets.  相似文献

ATP and adenylylimidodiphosphate (AdoPP[NH]P) bind to (Na+ + K+)-ATPase in the absence of Mg2+ (EDTA present) with a homogeneous but 15-fold different affinity, the Kd values being 0.13 μM and 1.9 μM, respectively. The binding capacities of the two nucleotides are nearly equal and amount to 3.9 and 4 nmol/mg protein or 1.7 and 1.8 mol/mol (Na+ + K+)-ATPase, respectively. The Kd value for ATP is equal to the Km for phosphorylation by ATP (0.05–0.25 μM) and the binding capacity is equivalent to the phosphorylation capacity of 1.8 mol/mol (Na+ + K+)-ATPase. Hence, the enzyme contains two high-affinity nucleotide binding and phosphorylating sites per molecule, or one per α-subunit. Additional low-affinity nucleotide binding sites are elicited in the presence of Mg2+, as shown by binding studies with the non-phosphorylating (AdoPP[NH]P). The Kd and binding capacity for AdoPP[NH]P at these sites is dependent on the Mg2+ concentration. The Kd increases from 0.06 mM at 0.5 mM Mg2+ to a maximum of 0.26 mM at 2 mM Mg2+ and the binding capacity from 1.5 nmol/mg protein at 0.5 mM Mg2+ to 3.3 nmol/mg protein at 4 mM Mg2+. Extrapolation of a double reciprocal plot of binding capacity vs. total Mg2+ concentration yields a maximal binding capacity at infinite Mg2+ concentration of 3.8 nmol/mg protein or 1.7 mol/mol (Na+ + K+)-ATPase. The Kd for Mg2+ at the sites, where it exerts this effect, is 0.8 mM. The Kd for the high-affinity sites increases from 1.5–1.9 μM in the absence of Mg2+ to a maximum of 4.2 μM at 2 mM Mg2+ concentration. The binding capacity of these sites (1.8 mol/mol enzyme) is independent of the Mg2+ concentration. Hence, Mg2+ induces two low-affinity non-phosphorylating nucleotide binding sites per molecule (Na+ + K+)-ATPase in addition to the two high-affinity, phosphorylating nucleotide binding sites.  相似文献

Rubidium uptake in potassium-starved cells followed biphasic kinetics in the micromolar and millimolar range and was independent of the temperature. In contrast, Rb+ uptake in normal-K+ cells followed a monophasic kinetics in the millimolar range and increased at temperatures higher than 30°C. Differences in the K m values and in the Arrhenius plots of Rb+ uptake suggest different uptake systems in K+-starved and in normal-K+ cells. In addition, the substantial inhibition of Rb+ uptake caused by carbonyl cyanide-m-chlorophenyl hydrazone indicates that these systems are strongly dependent on membrane voltage. Lithium (sodium) tolerance, influx, and efflux were separately studied. F. oxysporum was shown to be very tolerant to sodium, while lithium caused a specific toxic effect. Li+ uptake in K+-starved cells exhibits a monophasic kinetics with low affinity. Li+ efflux was not affected by external pH or addition of potassium to the medium, suggesting that a Na+/cation antiporter is not involved in this process. Red cells of hibernating species have a higher relative rate of Na+–K+ pump activity at low temperature than the red cells of a mammal with a typical sensitivity to cold. The kinetics of ATP stimulation of the Na+–K+ pump were determined in guinea pig and ground squirrel red cells at different temperatures between 5 and 37°C by measuring ouabain-sensitive K+ influx at different levels of ATP. In guinea pig cells, elevation of intracellular free Mg2+ to 2 mmol·l-1 by use of the divalent cation ionophore A23187 caused the apparent affinity of the pump for ATP to increase with cooling to 20°C, rather than to decrease, as occurs in cells not loaded with Mg2+. In ground squirrel cells raising intracellular free Mg2+ had little effect on apparent affinity of the pump for ATP at 20°C. ATP affinity rose slightly with cooling both in Mg2+-enriched and in control ground squirrel cells. Increased intracellular free Mg2+ in guinea pig cells stimulated Na+–K+ pump activity so that at 20°C the pump rate was the same in the Mg2+-enriched guinea pig and control ground squirrel cells. Pump activity in Mg2+-enriched guinea pig cells at 5°C was significantly improved but still lower than pump activity in control cells from ground squirrel. Thus, loss of affinity of the Na+–K+ pump for ATP that occurs with cooling in cold-sensitive guinea pig red cells can be, at least partially, prevented by elevating cytoplasmic free Mg2+. At 0°C, when Na+ was the only cation present in the incubation medium, increasing the Na+ concentration from 3 to 10 mM enhanced the affinity of [3H]l-[2-(di-phenylmethoxy)ethyl]-4-(3-phenyl-2-propenyl)piperazine ([3H]GBR 12783) for the specific binding site present in rat striatal membranes without affecting the 5max. For higher Na+ concentrations, specific binding values plateaued and then slightly decreased at 130 mM Na+. In a 10 mM Na+ medium, the KD and the Bmax were, respectively, 0.23 nM and 12.9 pmol/mg of protein. In the presence of 0.4 nM [3H]GBR 12783, the half-maximal specific binding occurred at 5 mM Na+. A similar Na+ dependence was observed at 20°C. Scatchard plots indicated that K+, Ca2+, Mg2+, and Tris+ acted like competitive inhibitors of the specific binding of [3H]GBR 12783. The inhibitory potency of various cations (K+, Ca2+, Mg2+, Tris+, Li+ and choline) was enhanced when the Na+ concentration was decreased from 130 to 10 mM. In a 10 mM Na+ medium, the rank order of inhibitory potency was Ca2+ (0.13 mM) > Mg2+ > Tris+ > K+ (15 mM). The requirement for Na+ was rather specific, because none of the other cations acted as a substitute for Na+. No anionic requirement was found: Cl-, Br-, and F- were equipotent. These results suggest that low Na+ concentrations are required for maximal binding; higher Na+ concentrations protect the specific binding site against the inhibitory effect of other cations.  相似文献
