The estimates of genetic diversity in populations of chum salmon Oncorhynchus keta from different regions of Sakhalin Island, Iturup Island, and the Anadyr’ River were obtained on the basis of analysis of allozyme variability. These estimates together with our published earlier data on chum salmon from the Amur River basin and the rivers of the northern coast of the Sea of Okhotsk demonstrate pronounced regional genetic differentiation in the Asian part of the fish range. The intraregional level of interpopulation genetic diversity was maximum on Sakhalin Island (G ST = 6.6%) and was small on Iturup Island (G ST = 0.9%) and the northern coast of the Sea of Okhotsk (G ST = 0.6%). Interpopulation genetic diversity of Sakhalin chum salmon was almost commensurable to the diversity of the whole pool of studied Asian populations (G ST = 7.6%) and would be presented more completely in baselines assigned for genetic identification of mixed stocks. It was demonstrated that the character and degree of genetic differentiation between populations of chum salmon from the main hatcheries situated in different regions of the Sakhalin oblast and connected to one another by numerous transplantations of fertilized eggs did not change significantly during an approximately 20-year period of our observation, and this fact suggests low efficiency of such transplantations.  相似文献

The nucleotide sequences of a fragment of the mitochondrial DNA cytochrome b gene were determined in 12 chum salmon populations from the Russian Far East. The level of genetic diversity in the chum salmon populations from the Iturup Island, northern coast of the Sea of Okhotsk, and Anadyr’ River was found to be higher than in the populations from Kamchatka and Sakhalin, which may be related to the history of their origin and dispersal. The proportions of intrapopulation genetic variability (F CT) and interpopulation genetic variability within the groups (F SC) account for 90.87 and 0.9%, respectively, and the intergroup component (F ST) comprises 8.23%. The predominance of one haplotype, B1, which is common for all populations studied, and a low share of intergroup variability suggest the beginning of colonization by the species of the given region from a common source (group of founders) and a relatively recent time of divergence of the chum salmon populations from the region examined.  相似文献

Based on the data of the analysis of distribution of juvenile chum salmon Oncorhynchus keta that were marked at salmon hatcheries in the southern part of the Sakhalin Island and Japan it has been established that their stocks in the Sea of Okhotsk coastal waters off South Sakhalin and Japan are of mixed origin. One part of the stock is composed of juveniles from hatcheries located in the southern part of Sakhalin, and the other part is composed of migrants from the waters adjacent to Japan. The first part is formed by juveniles both from hatcheries on the Sea of Okhotsk and Sea of Japan coasts of South Sakhalin. The second part is formed by juveniles reared at hatcheries in all regions of Japan: the Pacific and Sea of Okhotsk coasts of Honshu and Hokkaido, and from the Sea of Okhotsk coast of Hokkaido and Izmena (Nemuro) Strait. In July, in the Sea of Okhotsk coastal waters off South Sakhalin, the length and weight of most juvenile chum salmon of Japanese origin exceed 10 cm and 10 g, and juveniles of Sakhalin origin are always smaller than 10 cm and 10 g, which makes it possible to differentiate juveniles in trawl catches.  相似文献

The efficiency of reproduction of Pacific salmon (Oncorhynchus) in the rivers of the northeastern Sakhalin coast is characterized based on long-term observations. The rivers of the northern, central, and southern parts differ in the type of their channel, faunistic composition of aquatic biota, and prevalent salmon species. The northern rivers have a plain type of channel with rather low spawning efficiency of salmons and relatively poor freshwater ichthyofauna. The central area houses the largest rivers (Tym’ and Nabil’ Rivers) running to the Nyiskii and Nabil’skii Gulfs; the rivers there have developed channels of a plain type, high diversity of resident ichthyofauna, low spawning efficiency of the pink salmon Oncorhynchus gorbuscha, and high (in the past) abundance of the fall chum salmon O. keta. Characteristic of the southern rivers with their prevalently mountain–foothill type of channels are poor fish population and high spawning efficiency of the pink salmon. The population dynamics of the salmonids in the northeastern coast of Sakhalin is described. The total number of breeders entering the rivers of the northeastern coast that provides an efficient reproduction is estimated as 3.5?9.0 million individuals for the pink salmon and 0.04–0.60 million individuals for the chum salmon (fall race).  相似文献

The complete mitochondrial genome of the Sakhalin sturgeon Acipenser mikadoi and two mitogenomes of the Amur sturgeon A. schrenckii were sequenced using Roche 454 technology. The mitogenomes of the green sturgeon A. medirostris (obtained from GenBank) and the Sakhalin sturgeon differ as much as the mitogenomes of two mtDNA haplogroups (SM and BG) found in the same population of the Amur sturgeon: 0.0042 ± 0.0006 and 0.0036 ± 0.0005 substitutions per site (Tamura–Nei distance, TrN), respectively. The differences of these mitogenome pairs from mitogenomes of sister species (kaluga A. dauricus and white sturgeon A. transmontanus) are 3–6 times larger: 0.0260 ± 0.0015 and 0.0102 ± 0.0008, respectively. Thus, the differences between the mitogenomes of the Sakhalin and green sturgeons can be attributed to the variability at the intraspecific level. The time that has passed since the divergence of the Sakhalin and green sturgeons is considered to be much shorter than was previously believed: approximately 0.16 rather than 9.60 million years.  相似文献

The efficiency of reproduction of Pacific salmons of the genus Oncorhynchus in the rivers of the northwestern coast of Sakhalin Island is characterized on the basis of long-term data (1989?2014). Differences between the rivers in the northern and southern parts of the region under study have been revealed with respect to the channel type, faunistic composition of the aquatic biota, and prevailing species of salmons. A high spawning efficiency of pink salmon O. gorbuscha, high densities of filling of spawning grounds, and a relatively poor composition of the freshwater ichthyofauna have been recorded in the rivers of the northern part with the mountain–piedmont type of channel. Varied fish population and the high spawning efficiency of chum salmon O. keta have been recorded in the rivers of the southern part with the mainly lowland channel. The salmon spawning area in the rivers of the northern and southern parts has been estimated. The data on the dynamics of the abundance of salmons on the northwestern coast of Sakhalin are given. The total value of the passage of spawners to the rivers of northwestern Sakhalin that is required for effective reproduction has been estimated, ranging from 0.672 to 1.639 million specimens for pink salmon and approximately 0.43 million specimens for chum salmon.  相似文献

Previously, a very low level of divergence between the species of the genus Salamandrella—S. keyserlingii and S. schrenckii—was detected on the basis of variability of the nucleotide sequences of three genes of the nuclear genome (BDNF, POMC, and RAG1). Fixed interspecific differences were detected only in one nucleotide position of the RAG1 gene, and the level of interspecific divergence for this gene was only 0.07%. In this paper, we present the results of a study of the variability of the ENC1, MGAT4C, and RAG2 nuclear genes. The level of interspecific divergence for the MGAT4C gene was 0.14%, and for the RAG2 gene, it was 0.8%. The results of a phylogenetic analysis of the nucleotide sequences of the RAG2 gene in representatives of the family Hynobiidae indicate that the separation of the Salamandrella branch, which is basal for the genera Batrachuperus, Liua, Hynobius, and Pseudohynobius, occurred approximately 55 million years ago. The time of divergence between species of the Salamandrella genus was approximately 21 million years ago.  相似文献

Long-term changes in the sex ratio have been studied in pink salmon (Oncorhynchus gorbuscha and chum salmon (O. keta) populations of Kamchatka and Sakhalin. It has been demonstrated that these changes are an adaptation to population dynamics: an increase in the population size is accompanied by a shift towards a higher proportion of males; a decrease in population size, by a shift towards a higher proportion of females. The correspondence between morphological and molecular characters in populations of the two species has been analyzed in order to determine the mechanism of sex ratio control. In some pink salmon and chum salmon populations, there is a discrepancy between sex identifications based on morphological characters and molecular markers. This discrepancy is assumed to be accounted for by sex inversion mechanisms, which may be population-or region-specific. In two cases, it has been found that the sex ratio discrepancy in populations is related to the numbers of fish in subsequent generations. These findings suggest that sex inversion may be related to population size control.  相似文献

RFLP analysis of three amplified mtDNA fragments (Cytb/D-loop, ND1/ND2, and ND3/ND4L/ND4) was performed in the following taxa: Parahucho perryi, Hucho taimen, Brachymystax lenok, B. tumensis, Salmo salar, Salvelinus leucomaenis, and S. levanidovi. For mtDNA of P. perryi, a substantial decrease in the haplotype and nucleotide diversity was observed as a result of random genetic drift, caused by a reduction in the effective population size. Nucleotide divergence estimates between the mtDNA haplotypes were determined. Sakhalin taimen P. perryi was found to be approximately equally diverged from S. salar and from the charrs of the genus Salvelinus, by 11.0 and 10.0%, respectively. The divergence between P. perryi and H. taimen constituted 14.6%, between P. perryi and lenoks of the genus Brachymystax, 14.2%, and between H. taimen and Brachymystax, 7.7%. The analysis of possible phylogenetic relationships of the mtDNA from P. perryi among the group of taxa examined confirmed validity of the genus Parahucho. Phylogenetic reconstructions performed showed that robustness of the trees constructed for the complex of phylogenetically informative characters over three mtDNA fragments was considerably higher than that of the trees constructed for individual genes.  相似文献

The phylogenetic relationships and phylogeography of two relatively rare Macaronesian Lactuca species, Lactuca watsoniana (Azores) and L. palmensis (Canary Islands), were, until this date, unclear. Karyological information of the Azorean species was also unknown. For this study, a chromosome count was performed and L. watsoniana showed 2n = 34. A phylogenetic approach was used to clarify the relationships of the Azorean endemic L. watsoniana and the La Palma endemic L. palmensis within the subtribe Lactucinae. Maximum parsimony, Maximum likelihood and Bayesian analysis of a combined molecular dataset (ITS and four chloroplast DNA regions) and molecular clock analyses were performed with the Macaronesian Lactuca species, as well as a TCS haplotype network. The analyses revealed that L. watsoniana and L. palmensis belong to different subclades of the Lactuca clade. Lactuca watsoniana showed a strongly supported phylogenetic relationship with North American species, while L. palmensis was closely related to L. tenerrima and L. inermis, from Europe and Africa. Lactuca watsoniana showed four single-island haplotypes. A divergence time estimation of the Macaronesian lineages was used to examine island colonization pathways. Results obtained with BEAST suggest a divergence of L. palmensis and L. watsoniana clades c. 11 million years ago, L. watsoniana diverged from its North American sister species c. 3.8 million years ago and L. palmensis diverged from its sister L. tenerrima, c. 1.3 million years ago, probably originating from an African ancestral lineage which colonized the Canary Islands. Divergence analyses with *BEAST indicate a more recent divergence of the L. watsoniana crown, c. 0.9 million years ago. In the Azores colonization, in a stepping stone, east-to-west dispersal pattern, associated with geological events might explain the current distribution range of L. watsoniana.  相似文献

Phylogeographic analysis on the basis of individual marker variability provides insight into the history and mechanisms of the range formation of widely distributed species. A preliminary study of the mtDNA cytochrome b gene in Siberian rubythroat Luscinia calliope revealed the existence of three well-differentiated haplogroups, including one western and two eastern haplogroups. Continuing the study of the genetic markers of the species, we found that, in western part of the range, represented by the nominative geographic race, there were almost exclusively haplotypes of western group. In eastern populations of Khabarovsk krai, Chukotka, Kamchatka, and Sakhalin, haplotypes of all groups are mixed in different proportions. At the same time, the populations of Hokkaido and Iturup islands are exclusively represented by individuals with eastern haplotypes. Comparison of the identified nuclear copies of mitochondrial genes and construction of the phylogenetic network of haplotypes on the basis of cloned and initial sequences showed that two groups of eastern haplotypes (one of which geographically corresponded to L. c. anadyrensis and L. c. camtschatkensis and the second corresponded to L. c. sachalinensis) originated from nuclear pseudogenes of L. c. calliope through intergenomic recombination. In this regard, we propose a new hypothesis for the establishment of the modern range of this species, according to which the Siberian rubythroat dispersal from South Siberia occurred in two stages. At first, the species expanded its range to the northeast in the direction of the Kolyma and Koryak uplands. During the settling of these areas of northeastern Asia, a recombination between the mitochondrial and nuclear DNA took place, which led to the forming of a new haplotype, which was widespread in the emerging breeding populations. Birds with recombinant haplotypes populated the territories of Chukotka and Kamchatka, and then gradually occupied the Kuril Islands and, eventually, reached Hokkaido. At the next stage, Siberian rubythroat, probably, appeared in Sakhalin Island during spring migration, where some individuals stopped for breeding. Settling of the island was accompanied by similar intergenomic recombination and rapid fixation of a new recombinant haplotype with its subsequent spread across Sakhalin. The insular way of dispersal is completely repeated by modern migrants.  相似文献

The crane fly Tipula (Platytipula) moiwana (Matsumura, 1916) occurs in the southern Russian Far East, including Sakhalin Island and the Kuril Islands, in Japan (Hokkaido), and in northeastern China. Sakhalin taimen (Parahucho perryi) range from the Russian Far East mainland along the Sea of Japan coast, and Sakhalin, Kuril, and Hokkaido Islands and are considered to primarily be an anadromous species. We used otolith strontium-to-calcium ratios (Sr/Ca) to determine the chronology of migration between freshwater and saltwater and identify migratory contingents of taimen collected from the Koppi River, Russia. In addition, we examined taimen from the Sarufutsu River, Japan and Tumnin River, Russia that were captured in marine waters. Transects of otolith Sr/Ca for the Sarufutsu River fish were consistent with patterns observed in anadromous salmonids. Two fish from the Tumnin River appeared to be recent migrants to saltwater and one fish was characterized by an otolith Sr/Ca transect consistent with marine migration. Using these transects as benchmarks, all Koppi River taimen were classified as freshwater residents. These findings suggest more work is needed to assess life history variability among locations and the role of freshwater productivity in controlling migratory behavior in taimen.  相似文献

Sprat (Sprattus sprattus) and small herring (Clupea harengus) are the dominant prey fish of Atlantic salmon (Salmo salar) in the Baltic Sea. If the fatty acid (FA) proportions of sprat and herring differ, the dietary history of ascending salmon could be determined from their FA profiles. Therefore, we investigated the FA composition of several age groups of whole sprat and small herring, caught from the three main feeding areas of salmon in autumn and spring. Oleic acid (18:1n-9) was the most prevalent FA in sprat and characteristic of this species. In herring, palmitic acid (16:0) was the most common FA, but herring lipid was characterized by n-6 polyunsaturated FAs, and moreover, by palmitoleic acid (16:1n-7) and vaccenic acid (18:1n-7). Due to the higher lipid content of sprat, the concentrations of all other FAs, excluding these, were higher in sprat than in herring. The concentration of docosahexaenoic acid (DHA, 22:6n-3) increased with an increase in the lipid content and was consequently highest in the youngest specimens, being in young sprat almost double that of young herring, and 2.6 times higher in the sprat biomass than in that of herring. As a result of a decrease in the DHA concentration with age, the ratio thiamine/DHA increased with respect to age in both species, and was lower in sprat than in herring. It is concluded that an abundance of DHA in the diet of salmon most likely increases oxidative stress because of the susceptibility of DHA to peroxidation, and thus decreases thiamine resources of fasting, prespawning salmon. Because the FA composition of sprat and herring differs, and the relative abundancies of prey fish differ between the feeding areas of salmon, the feeding area of ascending salmon can most probably be derived by comparing their FA profiles.  相似文献

Wood mice of the genus Apodemus belong to the most frequent and epidemiologically important rodents of Europe and adjacent regions. Previous studies showed that in the Middle East region species of this genus exhibit extraordinary morphological similarity precluding their proper determination without application of molecular characters. In order to determine the species of the studied populations and to obtain an insight into their phylogeographic history, we analyzed their genetic variation. We sequenced 1139 bp fragment of the mitochondrial DNA control region and flanking tRNA genes in samples collected from six localities. Phylogenetic analyses revealed presence of distinct clades corresponding to species A. uralensis and A. witherbyi. In most localities we confirmed presence of both species which suggests their large sympatric and syntopic occurrence. We recognized an extensive genetic variability, 38 specimens of A. uralensis belong to 32 distinct haplotypes, while 19 specimens of A. witherbyi to 14 haplotypes. We confirmed presence of several distinct haplotypes that may originate from multiple wood mouse colonization waves from distinct geographic regions.  相似文献

During investigation of the mycobiota in the Sea of Okhotsk, three new Penicillium species were isolated from subaqueous soils collected in the Sakhalin Gulf and in the northeastern part of the Sakhalin shelf, near the Piltun Bay. According to Raper and Thom’s classification, the newly described species P. piltunense, P. ochotense, and P. attenuatum belong to subsection Divaricata. Two analyses were performed to reveal the phylogenetic relationships of the putative new species with other Penicillium species. Phylogenetically, the new species are related to the members of the P. canescens group and share some morphological and physiological features with them. However, they differ in having a larger colony diameter that makes them similar to P. atrovenetum, P. coralligerum, and P. antarcticum. A detailed analysis based on ITS, combined β-tubulin and calmodulin datasets, and morphological features revealed that the new species formed a distinct group inside the P. atrovenetum subclade. Moreover, P. attenuatum differs from the other two species macro- and micromorphologically and may represent a distinct phylogenetic lineage.  相似文献

For the first time, genetic analysis of the cytochrome b gene fragment haplotypes encoding the identical and the most common cytochrome b polypeptide (F1) in M. rutilus from eastern and Beringian maternal lineages was carried out. The F1 frequencies for the vole populations from Northern Priokhotye and the Kolyma basin were calculated. Considerable polymorphism of the cytochrome b F1 haplotypes within two major phylogroups of red-backed vole was supported by high molecular diversity indices for these clades. The proportion of genetic variation between the maternal lineages of F1 red-backed vole individuals (60.71%) was considerably higher than inter(24.44%) and intrapopulation (14.85%) components. The data obtained make it possible to advance a hypothesis on the convergence of the cytochrome b polypeptide structure upon sequence divergence of the corresponding gene.  相似文献

The gastropod Hexaplex trunculus is widely distributed in a relatively large range of habitats, but has no dispersal stage. We investigated its genetic structure across its distribution range, from Mediterranean Sea to adjacent Atlantic coasts, by sequencing mitochondrial DNA portions of the NADH dehydrogenase gene ND2 (420 pb) and the internal transcribed spacer ITS2 (450 pb). Our results suggested a significant genetic variability of ND2 (π = 0.009 and Hd = 0.629) and low variability of the ITS2 sequences. A strong phylogeographic break, separated the Aegean populations from those of Western/Eastern Mediterranean and the Atlantic ones, was founded. The tow lineages may have been separated by vicariance events due to the Peloponnese break that separates the Aegean populations from other populations and was maintained until now by the quasi-circular anticyclonic front associated to the straits of Cretan Arc of the Peloponnesian Peninsula. Tunisian coasts appear particularly diverse since the two divergent lineages co-occured. These results may have management consequences since H. trunculus is a high commercial value harvested species.  相似文献

Some of the views on the marine ecology of Pacific salmon (Oncorhynchus spp.) that were popular in the second half of the 20th century are discussed critically: the absolutization of the influence of sea surface temperature on distribution of salmon and strength of their year classes, as well as the conclusions on the shortage of food (particularly in winter) and the fierce competition for food, the “suppression” of other salmon species and own adjacent broodline by pink salmon, the limited carrying capacity of the pelagic zone of subarctic ocean waters for salmon, the distortion of the structure of epipelagic communities in ecosystems of the North Pacific due to the large-scale stock enhancement of chum salmon, etc. Most of these ideas have not been confirmed by the data of long-term monitoring conducted in the form of complex marine expeditions by the Pacific Research Fisheries Center (TINRO Center) in the Far-Eastern Seas and adjacent North Pacific waters since the 1980s. The data show that Pacific salmon are ecologically very flexible species with a wider temperature range of habitat than was previously believed. Salmon are able to make considerable vertical migrations, easily crossing zones of sharp temperature gradient and different water masses. Having the wide feeding spectra and being dispersed (as non-schooling fish) when feeding in the sea and ocean, they successfully satisfy their dietary needs in vast areas even with relatively low concentrations of prey organisms (macroplankton and small nekton). The total biomass of all the Pacific salmon species in the North Pacific is not greater than 4–5 million t (including 1.5–2.0 million t in Russian waters), whereas the biomass of other common species of nekton is a few hundreds of millions of tons. Salmon account for 1.0–5.0% of the total amount of food consumed by nekton in the epipelagic layer of the western Bering Sea, 0.5–1.0% in the Sea of Okhotsk, less than 1% in the ocean waters off the Kuril Islands, and 5.0–15.0% in the ocean waters off East Kamchatka. Thus, the role of Pacific salmon in the trophic webs of subarctic waters is rather moderate. Therefore, neither pink nor chum salmon can be considered as the species responsible for the large reorganization in ecosystems and the population fluctuations in other common nekton species.  相似文献

The variation in mitochondrial DNA (mtDNA) structure among chinook salmon Oncorhynchus tschawytscha Walbaum populations from Kamchatka was inferred from restriction length polymorphism analysis using eight restriction endonucleases. The nucleotide sequence variation in three amplified mtDNA regions was examined at seven polymorphic restriction sites in 579 fish from 13 localities. Based on the frequencies of 11 combined haplotypes and the number of nucleotide substitutions, the among-and within-population variation was estimated. The heterogeneity test showed highly significant differences among all the populations. The estimated maximum time of independent divergence of the asian chinook salmon populations, whose differences was about 0.02% nucleotide substitutions, did not exceed 10 000–20 000 years. Apparently, the retreat of the late Pleistocene glacier triggered spreading, recolonization, and formation of the present-day pattern of the species subdivision into structural components.  相似文献
