绵羊是一种短光照型生殖动物,具有明显的生殖季节性。人们普遍认为,绵羊生殖周期与环境光周期的同步变化是通过褪黑素作用于下丘脑-垂体-性腺轴系统来实现的。近年来的不少研究结果表明,绵羊对光周期的感受性还受褪黑素受体的调控。现从生态学(光周期的生殖效应)、神经内分泌学和分子生物学(褪黑素受体)等领域的研究进展出发,对褪黑素调控绵羊自然季节性生殖的作用及机制进行系统综述。  相似文献

目的探讨褪黑素(Melatonin,MLT)对小鼠前胃癌细胞(murine foregastric carcinoma,MFC)的生存素survivin和半胱天冬酶-3(cysteinyl aspartate-specific protease-3,caspase-3)表达的影响。方法使用不同浓度褪黑素处理培养的胃癌细胞,运用细胞免疫组化、实时荧光定量PCR、免疫印迹法、分光光度法等方法检测褪黑素在抑制小鼠MFC前胃癌细胞增殖过程中对细胞survivin、caspase-3的表达影响。结果与空白对照组相比,6mmol/L浓度褪黑素干预组的胃癌细胞survivin表达下调,caspase-3蛋白表达上调,并呈剂量依赖性。结论褪黑素能够降低胃癌细胞survivin的表达,进而激活caspase-3的活性,是褪黑素体外抑制肿瘤细胞增殖的作用机制之一。  相似文献

哺乳动物松果腺是视觉系统的一个退化器官。主要的明-暗周期与松果腺褪黑素生成的昼夜波动同步。明-暗周期的信息通过中枢和外周交感神经的一系列复杂神经元从眼传递到松果腺。在松果腺内,神经信号以褪黑素合成和释放在夜间升高为标志的内分泌信号表现出来。褪黑素是一种高活力的激素,它在调制光照周期对各种哺乳动物的季节性生殖中起着重要作用。褪黑素除影响生殖外,在机体内作用广泛。  相似文献

褪黑素对谷氨酸钠致痫大鼠海马5-色胺水平的影响   总被引:1,自引:0,他引:1  
目的观察褪黑素(Melatonin,MT)对谷氨酸钠(Glutamate,Glu)致痫大鼠海马5-羟色胺(5-hydroxytryptamine,5-HT)水平的影响,研究其抑制癫痫的作用机制。方法40只健康雄性SD大鼠随机分为4组(每组10只),分别为生理盐水对照组(NS组);谷氨酸钠致痫组(Glu组);褪黑素+谷氨酸钠组(MT+Glu组);Luzidole+褪黑素+谷氨酸钠组(Luz+MT+Glu组)。观察并记录大鼠行为学及脑电图改变,用免疫组织化学方法检测大鼠海马内5-HT含量变化。结果行为学观察和EEG显示,NS组无痫样发作和痫样放电,Glu组和Luz+MT+Glu组痫样发作重(Ⅲ—Ⅴ级),脑电图显示频发高幅的痫样波,TM+Glu组无或仅有轻微发作(0-Ⅱ级),脑电图上无或偶见散在单个微小痫样波;免疫组织化学分析结果显示,Glu组和Luz+MT+Glu组大鼠海马内5-HT含量与对照组比较均减少,差异性明显(P〈0.05),MT+Glu组较Glu组和Luz+MT+Glu组5-HT含量升高,差异性明显(P〈0.05)。结论MT对谷氨酸钠致痫大鼠痫样发作程度、痫样放电有抑制作用,其机制之一是经由其特异性的膜受体,通过某种机制增强5-HT作用,进而发挥抑痫效应。  相似文献

目的:研究褪黑素通过大电导Ca2+激活K+(BKCa)通道介导大脑中动脉张力变化的作用机制。方法:8周龄雄性Wistar大鼠,麻醉后取大脑中动脉,酶消化法急性分离脑中动脉平滑肌细胞,采用膜片钳技术全细胞记录模式检测细胞外液加入褪黑素前后BKCa通道和电压门控钾(KV)通道的电流密度,褪黑素受体抑制剂2-苯基-N-乙酰色胺(luzindole)孵育后,全细胞记录模式记录加入褪黑素后BKCa通道电流幅值和贴附式单通道记录模式记录加入褪黑素后BKCa通道Po值,内面向外记录模式检测加入褪黑素后BKCa通道电导(G),开放概率(Po),平均开放时间(To)和关闭时间(Tc)。结果:①褪黑素(100 μmol/L)显著增加全细胞BKCa通道电流密度,但对KV通道电流密度无显著影响;②luzindole (1 μmol/L)显著抑制褪黑素引起的BKCa通道电流密度增加;③贴附式单通道记录模式下,褪黑素(100 μmol/L)增加BKCa单通道Po值,luzindole (1 μmol/L)显著抑制褪黑素引起的Po增加;④内面向外单通道记录模式下,褪黑素(1 μmol/L,100 μmol/L)缩短BKCa单通道的To和Tc值,且Tc较To显著缩短;结论:褪黑素通过受体依赖和非受体依赖途径激活BKCa通道,介导大脑中动脉血管舒张。  相似文献

为探究外源褪黑素对棉花抗枯萎病的影响及作用机理,以海岛棉新海14号为材料,于叶面喷施不同浓度褪黑素(0、10、25、50、75、100 μmol/L)后接种棉花枯萎病菌,对枯萎病菌侵染后棉花幼苗进行抗病性鉴定、抗病相关酶活性及基因表达分析。结果表明,外源褪黑素对棉花枯萎病菌的生长无抑制作用,10-100 μmol/L褪黑素均能在一定程度上提高棉花对枯萎病的抗性,其中50 μmol/L褪黑素处理效果最好。与对照相比,50 μmol/L褪黑素预处理能有效减少接菌后棉花叶片中的过氧化氢(H2O2)含量和超氧阴离子(O2·-)的产生速率,增强过氧化物酶(POD)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、抗坏血酸过氧化物酶(APX)、几丁质酶(CHT)、β-1,3葡聚糖酶(GLU)、多酚氧化酶(PPO)、苯丙氨酸解氨酶(PAL)活性,并显著提高木质素代谢途径相关基因(GbPALGb4CLGbCAD)和类黄酮代谢途径关键基因(GbCHIGbDFRGbTT7)的表达量。表明50 μmol/L 的褪黑素处理能提高接菌后棉花抗氧化能力,增强防御酶活性,调控木质素、类黄酮代谢途径相关基因的表达,从而提高棉花对枯萎病的抗性。  相似文献

褪黑素在季节性繁殖动物的生殖细胞发育与性腺功能调控中发挥重要作用,然而褪黑素如何通过下丘脑-垂体-性腺(HPG)轴实现其调控功能目前仍不清楚。因此,本研究选取典型的长日照动物——高原鼠兔(Ochotona curzoniae)作为研究对象,使用酶联免疫吸附法测定繁殖期与非繁殖期雌雄鼠兔的血清褪黑素水平昼夜变化,利用实时荧光定量PCR分析褪黑素受体基因Mtnr1a与Mtnr1b在下丘脑、垂体与性腺中的表达水平,并通过免疫荧光染色进一步确认两种受体在性腺不同类型细胞中的定位。结果显示,非繁殖期雄性鼠兔的血清褪黑素含量始终高于繁殖期,且呈现不同的昼夜变化模式;雌性鼠兔的血清褪黑素含量远低于雄性鼠兔,且在繁殖期与非繁殖期并不存在显著差异。Mtnr1a与Mtnr1b在下丘脑、垂体与性腺中均有表达,雄性鼠兔在下丘脑与垂体中表现出繁殖期与非繁殖期基因表达的显著差异,而雌性鼠兔基因表达的差异主要出现在垂体与性腺中。两种受体蛋白在雄性性腺生殖细胞和支持细胞中均有分布,但MTNR1A更局限于精原细胞内,MTNR1B则在管腔内生殖细胞中表达。雌性性腺中MTNR1A在卵母细胞胞质内有表达,但更集中表达于颗粒细胞;MTNR1B在颗粒细胞以及卵母细胞核质内均有表达,且在生长卵泡的卵泡膜细胞中呈现高表达。以上结果表明,褪黑素调控雌雄高原鼠兔季节性繁殖的模式并不相同,其作用不仅限于通过下丘脑-垂体-性腺轴的间接调控,也可能通过性腺内靶向受体直接影响生殖细胞与体细胞命运。  相似文献

褪黑素对调节季节性繁殖哺乳动物的生殖具有重要调节作用。其受体MTNR1a(Melatonin receptor 1a,褪黑素受体1a)主要参与昼夜节律和生殖调控,MTNR1b(Melatonin receptor 1b,褪黑素受体1b)与多种疾病发生密切相关。为了探讨褪黑素受体基因的生物学功能,本实验对牦牛不同组织中MTNR1a、MTNR1b基因的表达与定位情况进行了研究。采用qRT-PCR (Quantitative Real-Time PCR, qRT-PCR) 检测成年雄性牦牛各组织及不同发育阶段(30日龄,2岁、4岁、6岁和8岁龄)牦牛睾丸组织中MTNR1a、MTNR1b mRNA的表达规律,并运用免疫组化技术对不同年龄牦牛睾丸中MTNR1a、MTNR1b蛋白进行了定位研究。结果发现,MTNR1a mRNA在松果体组织中表达量最高,肺脏、肌肉和睾丸次之;随着年龄增加,MTNR1a mRNA在睾丸中的表达量逐渐升高,到4岁后表达量趋于平稳;MTNR1a蛋白在不同发育阶段牦牛睾丸组织中均有表达,圆形精子呈现较强的免疫阳性,其次为初级精母细胞;MTNR1b mRNA在松果体表达量最高(P<0.05),肾脏、肝脏和下丘脑次之;在不同年龄牦牛睾丸中MTNR1b mRNA均有表达,且随着年龄的增加表达量逐渐增加,在8岁时表达量最高;MTNR1b蛋白主要定位在圆形精子细胞中。MTNR1a、MTNR1b基因在牦牛不同组织及不同发育阶段睾丸中的广泛表达,揭示了其在雄性牦牛生殖等生理过程中的重要作用。  相似文献

Efficient reproduction using natural mating and reproduction technology [in vitro fertilization (IVF) and embryo transfer (ET)] was investigated in IRS2 deficient mice with C57BL/6JJcl genetic background (Irs2(-/-) mice) as a typical type 2 diabetes model. From the results using various combinations of Irs2(-/-) and Irs2(-/+) mice, the combination of female Irs2(-/+) x male Irs2(-/-) was found to be more efficient than other combinations. In applications of reproduction technology using IVF and ET, the combination of female Irs2(-/+) x male Irs2(-/-) involves the possibility of Irs2(-/-) production by repeats using female Irs2(-/+) mice. However, reproductive continuity using this combination is difficult because of dependence on human technique and the cost of ET. Therefore, we concluded that Irs2(-/-) mice should be produced by embryo transfer using Irs2(-/-) mice from a colony consisting of female Irs2(-/+) x male Irs2(-/-).  相似文献

To study the evolutionary role played by differential male and female fertility (sexual asymmetry) both between individuals and over the life span within single individuals, the terms "intrinsic male fertility" and "intrinsic female fertility" are introduced. With the help of these terms, the concept of sexual asymmetry can be made precise and its effect on the establishment and maintenance of genetic polymorphisms can be analyzed. The main conclusions are: (1) any mutant causing a modification of the male fertility parameters which result in an increased intrinsic male fertility becomes established; (2) a corollary of this is that age-specific sexual asymmetry, as results from alternating degrees of female and male flowering in successive reproduction cycles, for example, has only secondary effects on the initial growth rate; (3) under the biologically reasonable premise that modifications of life histories result from reallocation of fixed net reproduction resources (defined as constant total female and male net reproduction output), a shift of net reproduction (whether female, male, or both in arbitrary proportions) to earlier ages is evolutionarily successful in growing but not in declining populations; shifts of net reproduction to later ages have opposite consequences.  相似文献

The beta1-3 N-acetylglucosaminyltransferase-1 (B3gnt1) gene encodes a poly-N-acetyllactosamine synthase which can initiate and extend poly-N-acetyllactosamine chains [Gal(beta1-4)GlcNAc (beta1-3)(n)]. Previous investigations with heterozygous and homozygous null mice for this gene have revealed the importance of poly-N-acetyllactosamine chains for the formation of olfactory axon connections with the olfactory bulb and the migration of gonadotropin releasing hormone neurons to the hypothalamus. The possible long-term effects of these developmental defects, however, has not yet been studied. Here we have examined a reproductive phenotype observed in B3gnt1-null mice. Whereas the B3gnt1 null females were fertile, the B3gnt1 null males were not able to sire litters at the expected rate when mated to either wildtype or B3gnt1-null females. We assessed male sexual behavior as well as male reproduction parameters such as testes size, spermatogenesis, sperm number, morphology, and the development of early embryos in order to identify the source of a reduced rate of reproduction. Our findings show that the B3gnt1 null male reproductive organs were functional and could not account for the lower rate at which they produced offspring with wildtype conspecifics. Hence, we propose that the phenotype observed resulted from an impaired sexual response to female mating partners.  相似文献

We measured carbon (respiration, photosynthesis, and production) and nitrogen allocation to male and female cones of Rocky Mountain Douglas-fir (Pseudotsuga menziesii var. glauca) to quantify gender-specific: (1) resource allocation to reproduction, and (2) contribution to carbon costs of reproduction via photosynthesis. We also measured foliar photosynthesis and nitrogen concentration ([N]) near and far from female cones to examine the relationship between reproduction and foliar physiology. Over one growing season, male cones required only 8% of all carbon allocated to reproduction, with females consuming the remaining 92%. Female cones, however, had maximum instantaneous refixation rates of 54%, which, integrated over the season, offset 6% of their total carbon requirements, while male cones were completely dependent on vegetative tissues for carbon. Male cones received 22% of all nitrogen allocated to reproduction and female cones received the remaining 78%. Foliage near female cones had elevated photosynthesis during the early stages of cone development and consistently lower [N] than foliage far from cones. Although female cones may photosynthesize, the annual sum of carbon fixed by reproductive structures is minor in comparison to the total carbon allocated to production and respiration.  相似文献

? Many plants combine sexual reproduction with vegetative propagation, but how trade-offs between these reproductive modes affect fitness is poorly understood. Although such trade-offs have been demonstrated at the level of individual shoots (ramets), there is little evidence that they scale up to affect genet fitness. For hermaphrodites, reproductive investment is further divided between female and male sexual functions. Female function should generally incur greater carbon costs than male function, which might involve greater nitrogen (N) costs. ? Using a common garden experiment with diclinous, clonal Sagittaria latifolia we manipulated investment in reproduction through female and male sex functions of 412 plants from monoecious and dioecious populations. ? We detected a 1?:?1 trade-off between biomass investment in female function and clonal reproduction. For male function, there was no apparent trade-off between clonal and sexual reproduction in terms of biomass investment. Instead, male function incurred a substantially higher N cost. ? Our results indicate that: trade-offs between investment in clonal propagation and sexual reproduction occur at the genet level in S.?latifolia; and sexual reproduction interferes with clonal expansion, with investment in female function limiting the quantity of clonal propagules produced, and investment in male function limiting the nutrient content of clonal propagules.  相似文献

Theoretical models of life-history evolution assume trade-offs between present and future reproduction and/or survival. Models of the evolution of sex assume trade-offs between male function and female function. Generally, experiments designed to evaluate the cost of reproduction on other functions tend to ignore male function. The present work on Gladiolus takes into account simultaneously the different primary functions of the plant and separates sexual reproduction into one male component (pollen production) and one female component (seed production). The study of environmental (within-clone), between-clone and genetic correlations using strains of Gladiolus and principal component analysis show that trade-offs exist between male function, female function and survival, including both characters of plant vigour, perennation (corm production) and vegetative propagation (cormel production). Phenotypic correlations, using different species and species-hybrids, have been obtained which confirm these results. In particular, these results underline the importance of the impact of pollen production on the other functions.  相似文献

One of the most studied life-history trade-offs is that resulting from the cost of reproduction: a trade-off arises when reproduction diverts limited resources from other life-history traits. We examine the cost of reproduction in male, and the effect of male mating status on female Callosobruchus maculatus seed beetles. Cost of reproduction for male C. maculatus was manifested as reduced longevity. There was also a positive relationship between male body size and male longevity. Females mated to males that had already copulated twice did not live as long as females mated to males that had copulated once or not at all. The third copulation of males also lasted longer than the two previous ones. We conclude that even though the cost of reproduction for males has been studied much less than that in females, there is growing evidence that male reproductive effort is more complex than has traditionally been thought.  相似文献

在多父本等量花粉混合授粉的交配实验设计基础上,利用SSR分子标记对其子代进行父本分析,研究鹅掌楸的配子选择与雄性繁殖适合度。参试亲本为鹅掌楸2个种的5个单株(鹅掌楸(Liriodendron chinense)2株,即FY和LS;北美鹅掌楸(L.tulipifera)3株,即LYS、MSL和NK)。结果表明:鹅掌楸的配子选择个体间差异较大。作为母本,NK和LYS倾向于选择异种雄配子,而MSL和LS则倾向于选择同种雄配子;在同种雄配子的选择中,NK、LYS和LS倾向于自交,而MSL则倾向于异交。以北美鹅掌楸为母本时,北美鹅掌楸与鹅掌楸的雄性繁殖贡献率分别为45.5%和54.5%,北美鹅掌楸的雄性繁殖适合度为鹅掌楸的0.556倍。以鹅掌楸为母本时,二者的繁殖贡献率分别为15.6%和84.4%,北美鹅掌楸的雄性繁殖适合度为鹅掌楸的0.123倍。总体上看,鹅掌楸的雄性繁殖适合度高于北美鹅掌楸,鹅掌楸与北美鹅掌楸均表现为自交亲和。  相似文献
