抗褐飞虱水稻品种的培育及其抗性表现   总被引:2,自引:0,他引:2  
褐飞虱Nilaparvata lugens(Stl)是危害水稻的主要虫害之一,发掘和利用新的抗褐飞虱基因培育抗性品种是目前防治褐飞虱最经济有效的方法之一。抗褐飞虱基因来自药用野生稻的抗虫品种B5,对褐飞虱生物型1和2具有高度抗性,B5携带的抗性基因Bph14被定位在第3染色体上。本研究以B5-10为抗源,以优良杂交稻亲本扬稻6号为受体亲本,通过复交和回交,利用与Bph14紧密连锁的分子标记MRG2329在后代中进行分子标记辅助选择,通过苗期分子标记检测和成株期农艺性状选择,最后育成恢复系R476和杂交组合广两优476。采用苗期群体鉴定技术对R476和广两优476的褐飞虱抗性进行了鉴定,R476和广两优476的抗性水平分别为中抗和中感。广两优476在稻飞虱发生较重的稻田进行试种示范,与对照品种扬两优6号和两优培九相比,广两优476对稻飞虱表现出明显的抗性。研究结果表明在育种过程中利用分子标记辅助选择Bph14基因是培育抗褐飞虱水稻品种的有效途径之一。  相似文献

褐飞虱(Nilaparvata lugens St(a)l)、白背飞虱(Sogatella furcifera Horvath)和灰飞虱(Laodelphax striatellus Fallén)是水稻生产上的重要害虫,给我国水稻生产造成了严重的经济损失.培育和利用抗虫品种是防治稻飞虱经济有效的措施.抗性遗传和抗性基因研究是进行抗虫育种的基础.目前,有关水稻抗褐飞虱基因的遗传与定位研究取得了较大进展,包括21个主基因、50余个QTLs和许多褐飞虱抗性相关基因被发掘、定位与克隆,而白背飞虱和灰飞虱抗性基因尚有待进一步发掘和鉴定.此外,今后应加强稻飞虱抗性基因在生产上的应用.  相似文献

稻飞虱是水稻生产最严重的害虫之一。野生稻拥有丰富的抗虫基因资源,导入系是鉴定和利用野生稻有利基因的有效途径。本研究通过对371份小粒野生稻导入系进行抗褐飞虱和白背飞虱接虫鉴定,分别筛选出了11份抗、72份中抗褐飞虱的材料和7份抗、45份中抗白背飞虱的材料,其中有5份材料兼抗褐飞虱和白背飞虱,这是从小粒野生稻中鉴定出抗白背飞虱材料的首次报道。通过对2份抗性导入系材料与感虫亲本杂交构建的F1和F2群体的抗虫鉴定和分析表明:K41对褐飞虱和白背飞虱的抗性受2对显性抗虫基因通过互补作用所控制;P114对褐飞虱和白背飞虱的抗性都是由1对主效的隐性基因控制。这些结果必将有利于小粒野生稻抗稻飞虱的基因定位和育种利用。  相似文献

褐飞虱是水稻生产中最严重的害虫之一,从野生稻中发掘抗虫基因,有利于培育具有抗虫能力强的水稻新品种。该研究通过对云南野生稻进行温室和大田抗虫鉴定以及9个已知抗褐飞虱基因的PCR鉴定,发现云南野生稻对褐飞虱表现出不同程度的抗性,尤其疣粒野生稻和药用野生稻对褐飞虱表现出高抗,可作为抗虫基因发掘的优良抗源材料;不同褐飞虱抗性的云南野生稻中含有的抗褐飞虱基因差异很大,3种野生稻中均不含Bph1和Bph18(t)抗病基因,景洪普通野生稻和元江普通野生稻可能含bph2基因,东乡普通野生稻可能含bph2、Bph15和Bph27(t)基因,疣粒野生稻中可能含bph2和bph19(t)基因,药用野生稻和药用野生稻(宽叶型)中可能含bph2和Bph6基因,药用野生稻F1中可能含bph2、Bph14和bph20(t)基因,药用野生稻F2中可能含bph2和Bph27(t)基因或者其同源基因。该研究为快速发掘利用云南野生稻中的抗虫基因奠定了理论基础。  相似文献

褐飞虱Nilaparvata lugens St(a)l是对水稻最具破坏性的害虫之一,OsLecRK1是水稻Bph3基因簇中对褐飞虱抗性贡献最大的基因.本文对RHTd(含Bph3)等材料进行了褐飞虱抗性评价,克隆并构建了OsLecRK1过量表达突变体水稻,利用该突变体分析了OsLecRK1基因对褐飞虱若虫存活率、若虫发育历期等生物学参数的影响.结果 表明,含Bph3基因水稻RHTd对褐飞虱的抗性明显地强于含Bph1基因水稻Mudgo和bph2基因水稻ASD7,RHTd水稻的褐飞虱受害指数仅为Mudgo和ASD7水稻的53.5%和24.1%.过量表达OsLecRK1基因能显著地增加水稻对褐飞虱的驱避性和抗生性,褐飞虱雌成虫偏好于在野生型水稻上产卵;突变体水稻上的褐飞虱若虫存活率显著地降低,仅为野生型水稻上若虫存活率的75.2% ~81.8%,且若虫发育历期显著地延长,羽化率和初羽化雌成虫体重均显著地降低;此外,褐飞虱在突变体水稻上取食分泌的蜜露量只有野生型上的40.3% ~ 60.9%,褐飞虱单雌产卵量只为野生型51% ~61.2%,卵孵化率只有野生型的52.2%~56.7%,均显著地减少.结果 表明,含Bph3基因水稻RHTd对褐飞虱的抗性明显地高于分别含Bph1、bph2的水稻Mudgo和ASD7;水稻Bph3基因座的OsLecRK1单个基因过量表达即可显著增加水稻对褐飞虱的抗性,OsLecRK1协同影响褐飞虱的多个生物学参数降低褐飞虱的适合度.  相似文献

从198份广西药用野生稻中筛选出3份对褐飞虱高抗(0级)的材料,用高抗材料与感虫材料杂交,结果表明其抗性是受一对显性基因控制;与14个已知抗褐飞虱基因品种杂交、回交和自交,对BC2F2后代材料进行了抗性遗传分析,等位性测定结果表明,抗源材料与已知的14个抗性基因不等位,结果发现广西药用野生稻与国外药用野生稻的抗性基因(隐性基因)不同。初步认为它是一个新的抗性基因。在利用方面,通过幼胚离体培养获得绿苗,经多代回交和自交,成功地将CC染色体组中的抗性基因转育到栽培稻中,获得了BC4F5高世代、异源附加系。  相似文献

利用回交重组自交群体检测水稻抗褐飞虱数量性状基因座   总被引:10,自引:1,他引:9  
利用由98个系组成的Nipponbare/Kasalath//Nipponbare回交重组自交系(Backcross Inbred lines,BILs)作图群体(BC1F9),进行水稻抗褐飞虱数量性状基因座(Quantative Trait Locus,QTL)的检测和遗传效应分析。采用苗期集团鉴定方法,并以死苗率作为抗褐飞虱表型值,分析亲本和98个BILs抗褐飞虱表现。共检测到3个苗期抗褐飞虱QTL,分别位于2、10和12染色体上,各QTL的LOD值为2.01~2.33,贡献率为10.4%~16.6%,可解释群体总表型变异的39.0%。这3个数量性状基因座对褐飞虱的抗性均来自抗虫亲本Kasalath。与这些数量性状基因座连锁的分子标记可望应用于聚合多个抗性基因,培育对褐飞虱具有水平抗性水稻新品种的育种实践中。  相似文献

褐飞虱(Nilapavata lugens,brown planthopper,BPH)是水稻生产中最重要的害虫.水稻与褐飞虱互作机制的研究为培育新的水稻品种做出了贡献.本文综述了栽培稻和野生稻中分离定位的抗褐飞虱基因,以及褐飞虱唾液蛋白、水稻和褐飞虱代谢物和褐飞虱共生菌在水稻-褐飞虱互作关系中的作用.目前从栽培稻和野生稻中鉴定了 40个抗褐飞虱基因,褐飞虱取食的信号转导开启水稻抗性基因的表达和防御机制的改变,包括筛管封闭、次生代谢产物的产生,以及蛋白酶抑制剂的诱导等.褐飞虱的唾液蛋白以及体内存在多种共生微生物对褐飞虱的生长发育和适应抗性都起到一定作用,但目前提出的水稻-褐飞虱分子相互作用的模型仍有许多方面有待研究.  相似文献

在半自然状态下 ,用生命表方法初步比较了褐飞虱Nilaparvatalugens和白背飞虱Sogatellafurcif era在 4个抗褐飞虱水稻品种ASD7,IR3 6,JX89和Mudgo上的种群参数。初步的结果表明 ,白背飞虱在 4个抗褐飞虱水稻品种上具有较高的繁殖率 ,较短的发育时间和较大的种群增长。对褐飞虱各种群参数影响最大的是JX89,对白背飞虱影响最大的是Mudgo。与感性品种TN1比较 ,这些抗褐飞虱的水稻品种对 2种稻飞虱均具有不同程度的抗性。这些结果说明 ,抗褐飞虱水稻品种对白背飞虱同样具有一定的抗性 ,但白背飞虱对这种抗性具有较强的适应性 ,如果在一个地区长期推广抗褐飞虱的水稻品种 ,势必会导致白背飞虱的大发生  相似文献

Thirteen near-isogenic lines (NILs) of japonica rice were developed via a backcross method using the recurrent parent Chucheong, which is of good eating quality but is susceptible to Magnaporthe grisea, and three blast resistant japonica donors, Seolak, Daeseong and Bongkwang. The agro-morphological traits of these NILs, such as heading date, culm length, and panicle length, were similar to those of Chucheong. In a genome-wide scan using 158 SSR markers, chromosome segments of Chucheong were identified in most polymorphic regions of the 13 NIL plants, and only a few chromosome segments were found to have been substituted by donor alleles. The genetic similarities of the 13 NILs to the recurrent parent Chucheong averaged 0.961, with a range of 0.932-0.984. Analysis of 13 major blast resistance (R) genes in these lines using specific DNA markers showed that each NIL appeared to contain some combination of the four R genes, Pib, Pii, Pik-m and Pita-2, with the first three genes being present in each line. Screening of nine M. grisea isolates revealed that one NIL M7 was resistant to all nine isolates; the remaining NILs were each resistant to between three and seven isolates, except for NIL M106, which was resistant to only two isolates. In a blast nursery experiment, all the NILs proved to be more resistant than Chucheong. These newly developed NILs have potential as commercial rice varieties because of their increased resistance to M. grisea combined with the desirable agronomic traits of Chucheong. They also provide material for studying the genetic basis of blast resistance.  相似文献

A series of introgression lines (ILs) were generated from repeated backcrossing between the exotic hexaploid wheat genotype Am3 and the common wheat genotype Laizhou953. Am3 was synthesized by crossing Triticum carthlicum with Aegilops tauschii and was used as the donor parent in this study, and Laizhou953 was used as the recurrent parent. Two hundred and five SSR markers showing polymorphism between the two parents were used to identify the introgressed Am3 chromosome segments in 97 BC4F3 ILs. The introgressed segments in each line and the length of the introgressed segments were estimated according to the wheat SSR consensus map. The introgressed segments from Am3 in the 97 lines covers 37.7% of the donor genome. The introgressed segments were most found on 2D, 3B, 6B, and 1D with coverage of 59.8, 59.5, 59.1, and 59% of the chromosomes, respectively. None of the 97 lines tested contained chromosome 4D segments introgressed from Am3. Introgressed segments for each of the chromosomes were mapped using the consensus wheat linkage map. Nine agronomic traits from BC4F3 lines were evaluated and the phenotype showed most lines have the tendency to be more similar to the recurrent parent. There were lines showing better agronomic traits than the recurrent parent, which indicated the introgression of favorable alleles from the exotic hexaploid wheat into the elite cultivar Laizhou953. Marker and phenotype data were used to identify quantitative trait loci (QTLs) controlling these nine traits. In total, 38, 33, and 28 putative QTLs were detected for seven of the nine traits in 2003, 2004, and 2005, respectively. Some of these agronomic important QTLs were detected in more than one season. Controlling the brown planthopper (BPH), Nilaparvata lugens (Stål) (Hemiptera: Delphacidae), is a difficult task in rice (Oryza sativa L.) production. We focused on vitellogenins (Vg), which are the major yolk protein precursors of vitellins and play an important role in the reproduction of oviparous species, including insects. We studied the accumulation of Vg mRNA and protein in a virulent BPH strain, Nagasaki-03, and a nonvirulent strain, Hatano-66, after rearing them on four rice lines. The rice lines used were two single resistance gene introgression lines, Norin-PL3 (Bph1 carrier) and Norin-PL4 (bph2 carrier), a pyramided line in which both genes were combined, and a susceptible japonica recurrent parent Tsukushibare. RT-PCR and quantitative RT-PCR analyses showed that the Vg mRNA level decreased greatly in Hatano-66 on the resistant lines. In contrast, the level of reduction on the resistant lines was much less in Nagasaki-03. Immunoblot analysis showed that Nagasaki-03 retained comparable levels of 175 kDa Vg protein on both the susceptible and resistant lines, whereas in Hatano-66, no Vg protein was detected on the resistant lines. Our results showed that BPH resistance genes caused differential reduction in the accumulation of Vg mRNA and protein, leading to the retardation of BPH reproduction on the resistant host rice plants.  相似文献

水稻抗稻瘟病基因Pi25是一个遗传传递能力强的广谱抗性基因。本研究以携带抗稻瘟病基因Pi25的BL27为抗源供体,与优质、配合力强、感稻瘟病的水稻保持系臻达B为受体亲本进行杂交、回交创制水稻抗病保持系新种质,再与臻达A测交和回交进行不育系转育,结合分子标记辅助选择和农艺性状筛选,获得3个抗性基因纯合、农艺性状和开花习性均与臻达A相似的改良不育系株系。利用福建省近年来致病性代表的22个稻瘟病菌株对3个改良不育系及其15个杂交种进行抗性鉴定,3个改良不育系的抗性频率为95.45%~100%,15个杂交种的抗性频率均达75%以上,而原始对照臻达A及其杂交种的抗性频率仅为54.55%和40.91%~63.64%。自然病圃诱发鉴定表明,3个改良不育系的叶瘟和穗颈瘟均为0级,表现高抗,而对照臻达A的叶瘟为5级,穗颈瘟为7级,表现感病;15个杂交种均表现良好的稻瘟病抗性。进一步分析比较15个杂交种的产量、农艺性状和稻米品质表现,结果表明臻达A-Pi25-3改良不育系的综合性状表现最优,继续回交转育,于2015年育成了稻瘟病抗性强、配合力好、群体整齐和性状稳定的不育系,命名为157A。研究表明,抗稻瘟病基因Pi25不仅在水稻不育系臻达A的遗传背景下的抗性表达完全,且在不同水稻恢复系测交种的背景下同样表现出较高水平的抗性,说明抗性基因Pi25对不育系稻瘟病改良的效果明显。创制的新不育系157A的稻瘟病抗性显著提高,还基本保留了原来不育系高配合力等优良特性,为选育高产、优质、抗病杂交稻新品种提供了不育系新种质。  相似文献

The brown planthopper (Nilaparvata lugens Stål; BPH) is one of the most serious rice pests worldwide. Growing resistant varieties is the most effective way to manage this insect, and wild rice species are a valuable source of resistance genes for developing resistant cultivars. BPH27 derived from an accession of Guangxi wild rice, Oryza rufipogon Griff. (Accession no. 2183, hereafter named GX2183), was primarily mapped to a 17-cM region on the long arm of the chromosome four. In this study, fine mapping of BPH27 was conducted using two BC1F2 populations derived from introgression lines of GX2183. Insect resistance was evaluated in the BC1F2 populations with 6,010 individual offsprings, and 346 resistance extremes were obtained and employed for fine mapping of BPH27. High-resolution linkage analysis defined the BPH27 locus to an 86.3-kb region in Nipponbare. Regarding the sequence information of rice cultivars, Nipponbare and 93-11, all predicted open reading frames (ORFs) in the fine-mapping region have been annotated as 11 types of proteins, and three ORFs encode disease-related proteins. Moreover, the average BPH numbers showed significant differences in 96–120 h after release in comparisons between the preliminary near-isogenic lines (pre-NILs, lines harboring resistance genes) and BaiR54. BPH growth and development were inhibited and survival rates were lower in the pre-NIL plants compared with the recurrent parent BaiR54. The pre-NIL exhibited 50.7 % reductions in population growth rates (PGR) compared to BaiR54. The new development in fine mapping of BPH27 will facilitate the efforts to clone this important resistant gene and to use it in BPH-resistance rice breeding.  相似文献

Bacterial Blight (BB) caused by Xanthomonas oryzae pv. oryzae is a major disease of rice in tropical Asia. Since all the Basmati varieties are highly susceptible and the disease is prevalent in the entire Basmati growing region of India, BB is a severe constraint in Basmati rice production. The present study was undertaken with the objective of combining the important Basmati quality traits with resistance to BB by a combination of phenotypic and molecular marker-assisted selection (MAS). Screening of 13 near-isogenic lines of rice against four isolates of the pathogen from Basmati growing regions identified the Xa4, xa8, xa13 and Xa21 effective against all the isolates tested. Two or more of these genes in combination imparted enhanced resistance as expressed by reduced average lesion length in comparison to individual genes. The two-gene pyramid line IRBB55 carrying xa13 and Xa21 was found equally effective as three/four gene pyramid lines. The two BB resistance genes present in IRBB55 were combined with the Basmati quality traits of Pusa Basmati-1 (PB-1), the most popular high yielding Basmati rice variety used as recurrent parent. Phenotypic selection for disease resistance, agronomic and Basmati quality characteristics and marker-assisted selection for the two resistance genes were carried out in BC1F1, BC1F2 and BC1F3 generations. Background analysis using 252 polymorphic amplified fragment length polymorphism (AFLP) markers detected 80.4 to 86.7% recurrent parent alleles in BC1F3 selections. Recombinants having enhanced resistance to BB, Basmati quality and desirable agronomic traits were identified, which can either be directly developed into commercial varieties or used as immediate donors of BB resistance in Basmati breeding programs.  相似文献

本文报道近十多年来广东省褐稻虱Nilaparvata lugens(Stal)生物型变异动态监测的研究结果,针对当前褐稻虱优势生物型及发展趋势,推荐优良的抗褐稻虱品种资源。监测结果表明;根据苗期抗性反应,分蘖期生存率测定,田间系统调查监测,广州褐稻虱田间种群对带有Bph1抗性基因的代表品种IR26的致害力不断增强,1992年以来抗性反应达生物型2的水平。1992~1994年在全省7个不同生态类型地区取样测定及验证试验结果表明,生物型 2已上升为广东褐稻虱田间主害代的优势种群。多年的抗性鉴定结果证实,外引品种IR56、IR50404同其对应亲本PTB33、Babawee一样,对褐稻虱的抗性稳定,已推荐用作进一步抗 性育种的优良抗源。粳籼89等23个品种能抗褐稻虱生物型1和2,可及时替代带即BPh1抗性 基因的品种。  相似文献

Rice hoja blanca virus (RHBV) is a major virus disease of economic importance affecting rice in northern South America, Central America and the Caribbean. This is the first report of transgenic resistance to RHBV and the transformation of an indica rice variety from Latin America. Rice transformed with the RHBV nucleocapsid protein ( N) gene had a significant reduction in disease development. Several reactions were observed that ranged from susceptible to completely resistant plants (immunity). The resistant reactions were characterized by the production of local lesions like a hypersensitive reaction or a recovery phenotype with the emergence of symptom-less new leaves. These transgenic RHBV-resistant rice lines expressed the N gene RNA at low levels that were below the detection limit by Northern blots and only resolved by RT-PCR. The nucleocapsid protein could not be detected in any of the transgenic plants either by Western or ELISA tests. These results suggest that the resistance encoded by the N gene in these plants appears to be mediated by RNA. When challenged with RHBV, the resistant transgenic lines showed a significant increased performance for important agronomic traits including the number of tillers, the number of grains per plant and the yield as compared to the susceptible control. Furthermore, upon inoculation some of the most-resistant transgenic lines showed agronomic traits similar to the uninoculated non-transgenic Cica 8 control. Using both agronomic traits and disease severity as criteria, several of the most-resistant lines were followed through the R(4) generation and demonstrated that the N gene and RHBV resistance was inherited in a stable manner. These transgenic rice lines could become a new genetic resource in developing RHBV-resistant cultivars.  相似文献
