Summary The types of Q-variant bands were determined by a combination of numerical designations setting five levels for both the size of bands and the intensity of fluorescence. This scoring system was used in a study of the frequencies of Q variants in 400 Japanese individuals: variant bands were observed in seven specific autosome pairs of Nos. 3,4,13,14,15,21, and 22. The number of variants per individual ranged from 0 to 8, and the mean was 3.83±1.86. The incidence of Q variants according to the types of variant bands was determined in specific chromosomes.A low frequency of No. 3 chromosome variants and a high frequency of a long Y in males seems to be characteristic for Japanese populations.Variation in the length of the long arm of Y (Yq) was analyzed in a total of 157 men. The relative length of Yq, which was determined by a ratio of Yq/21q, ranged from 0.98 to 2.27, with an average of 1.56±0.25. The length of pale band Yq11 was relatively constant between individuals, with an average of 0.64±0.08. Therefore, it was clear that the variation in the Yq length was the result mainly of a variation in the length of the brilliant band Yq12. However, a slight tendency for the length of band Yq11 to increase in proportionally to the total length of the Yq was revealed. In this study special consideration was paid to the reliable analysis of Q-band heteromorphism, and the factors or obstacles preventing such analysis have been discussed briefly.  相似文献

Chromosomal analyses were performed from peripheral blood samples from 20 adult rabbit males from Line R. This line has been selected for growth rate after weaning through 25 generations. Seminal characteristics and reproductive outcomes from these males were also examined. The chromosomal analysis results showed that one male had an aneuploidy rate of 16% (male A). The aneuploidies found represented both hypo- and hyper- haploidy. Differences between the male A and the contemporary males (males N), in fertility at 12 days post-insemination (44% vs. 66%) and at birth (31% vs. 59%) were observed. The male A also showed a high percentage of pregnancy losses (29% vs. 12%). For seminal characteristics, the percentage of abnormal spermatozoa was statistically different (P < 0.05) between male A and males N (34 ± 4 vs. 18 ± 2), and concentration differed significantly between males, showing the male A reduced fewer spermatozoa than males N (67 ± 23 vs. 172 ± 10, x106/ml). Motility and kinematic parameters revealed no differences between males. Differences between male reproductive performance found in this study could be explained by the effect of aneuploidy on spermatogenesis and its deleterious effect on male reproduction.  相似文献

In a population of Melanoplus femur-rubrum 13.9% of the males and 9.1% of the females sampled possessed a metacentric B chromosome (B). In crosses of females with one B (1 B females) and 0 B males 0.82 ± 0.05 of the offspring received the B. The value expected from Mendelian segregation is 0.5. In crosses of 1 B males and 0 B females the frequency of offspring receiving the B was 0.53 ± 0.02. The B is heterochromatic during prophase I of spermatogenesis but is euchromatic during prophase I of oogenesis. The observation that in 1 B females only one B was present in metaphase I of oogenesis suggested strongly that the high rate of transmission of the B by the females resulted from preferential segregation of the B into the secondary oocyte. Summary The correlated responses in male and female sterility to 50 generations of individual selection for pupa weight in Tribolium were analyzed. Two replicate lines (S-lines) were selected for heavier pupa weight and stabilizing selection for pupa weight was practiced in two replicate control lines (C-lines). There was close agreement between replicates in both sets of lines for direct and correlated responses. The rate of inbreeding has been constant for all lines (approximately 0.5% per generation).Regression of generation means for pupa weight on generation of selection indicated a significant linear regression in the direct response for both lines. The linear increases of 46 and 55 g. per generation in the S-lines accounted for 98% of the variation among generations and the linear decreases of 5 and 10 g. per generation in the C-lines accounted for 70–90% of the variation in the generation means.Maximum likelihood estimators were used to calculate the frequency of male and female sterility for each generation and line. Average sterility in the base population ranged from about 4 to 12% for both sexes. Polynomial regressions of percent sterility on generation of selection showed that quadratic and higher order regressions were occasionally significant but accounted for a relatively small fraction of the total variation. In the two S-line replicates, linear regression coefficients of percent sterility on generation number were 0.16±.09 and 0.20±.07 for males and 0.72±.08 and 0.54±.08 for females, suggesting a larger correlated response in female than in male sterility. In the C-lines, linear regression coefficients were 0.02±.08 and –.12±.05 for males in the two replicates and –.05±.05 and –.05±.05 for females. Estimates of realized genetic correlations between pupa weight and sterility in the S-lines ranged from 0.04 to 0.14 for males and from 0.14 to 0.37 for females when the heritability of sterility was allowed to take on values from 0.05 to 0.25.Supported by NSF Grants G-1238 and GB-5987, NIH Grant GM-16074 and NIH Fellowship 1 FO2 GM4 5130-01.  相似文献

Males with the SR X chromosome show the "sex-ratio" (sr) phenotype in which they produce almost entirely daughters. The few sons (about 1%) are invariably sterile X/O males and result entirely from nullo-XY sperm. The "male-sex-ratio" (msr) phenotype is a modified form of sr in which SR/Y males produce a higher frequency of sterile X/O sons. The msr trait is due to the presence of the SR X-chromosome in males which are also homozygous for one or more autosomes from the L116 strain. Here the frequency of nullo-3 and diplo-3 sperm from msr males was measured by crossing to a compound-3 strain and found to be 13.8% and 3.2%, respectively, of the total viable sperm. The sr males produced very low levels of nullo-3 sperm at a frequency not different from control X/Y males and a slightly elevated frequency of diplo-3 sperm over X/Y males. The msr males were found to have only 12% the fecundity of sr males and in matings to cause a high frequency of brown inviable eggs. These results indicate that high rates of autosomal aneuploidy are not restricted to chromosome 3 but also occur for chromosomes 2, 4 and 5. The overall frequency of autosomal aneuploid sperm is estimated to be approximately 50%. Microscopic studies of meiosis in testes from msr males indicates meiotic nondisjunction and meiotic chromosome loss are responsible for the msr phenotype. Last, microscopic studies of sperm cysts from msr males reveal high levels of spermiogenic failure.  相似文献

We assessed behaviors involved with depositing and receiving scent in three captive heterosexual pairs of red-bellied tamarins (Saguinus labiatus). The frequencies of scent deposition and scent investigatory behaviors differed between the sexes. Females scent marked exclusively by the anogenital gland. Males deposited 95.8% of scent marks via the anogenital gland and 4.2% via the sternal gland. Females scent marked at a significantly higher rate than that of males (0.9 ± 0.1 versus 0.3 ± 0.1 per 20 min, respectively). Males investigated the scent of their opposite-sex partners whereas females investigated no male scent. Mean ± SEM latency for males to investigate female scent was 208.7 ± 65.0 sec. Around 9% of all scent marks were overmarked within 8 min and there was a nonsignificant trend for males to overmark the scent of their female partners than vice versa. We discuss the sex differences in olfactory communication in red-bellied tamarins in terms of sexual selection theory.  相似文献

There is a lack of biomarkers or indices that can be used to predict the quality of fish semen samples following the freezing and thawing cycle. In the present study, a series of semen indices were tested to assess if they could accurately forecast the cryopreservation potential of Atlantic cod (Gadus morhua) semen. Fresh and frozen-thawed sperm activity variables were compared, and relationships between frozen-thawed sperm activity and fertilization success were examined. In comparison with fresh sperm, activity variables of frozen-thawed spermatozoa were reduced. Of the 18 males examined, mean (± SEM) spermatocrit of fresh sperm was 40.72 ± 4.23%, osmolality of the seminal plasma 366.32 ± 4.95 mOsmol/kg, pH 8.32 ± 0.04, protein concentration 1.05 ± 0.08 mg/mL, anti-trypsin activity 153.83 ± 19.25 U/L, and total antioxidant capacity 0.15 ± 0.03 μmol Trolox equivalents/mL. Frozen-thawed fertilization success was highly variable among males with values ranging from 18.5 to 90.2%. Regressions yielded significant positive relationships between frozen-thawed motility, velocity, track crossing frequency, and subsequent fertilization success. Sequential multiple regressions explained up to 95% of the variation in frozen-thawed sperm activity. Spermatocrit and pH of fresh semen were negatively related, whereas osmolality and antioxidant capacity were positively related to frozen-thawed motility and velocity. Each of these indices can be measured within minutes of collecting a fresh sample of semen and are thus early indicators of the capacity of semen samples to withstand cryopreservation. These results have many benefits for conservation of wild stocks, aquaculture production, and for understanding semen biology and cryobiology of fishes.  相似文献

Summary Over-winter torpor patterns of Richardson's ground squirrels hibernating in southern Alberta were monitored with temperature-sensitive radiocollars to determine if these patterns differed between males and females in a manner related to the greater costs of mating effort by males than females. The hibernation season (from immergence to emergence) was composed of three periods: post-immergence euthermy, heterothermy, and pre-emergence euthermy. The hibernation season was shorter for juveniles than adults both among males (< 150 versus 234 days) and females (185 versus 231 days), a reflection of the later immergence into hibernation by juveniles. However, regardless of the absolute duration of hibernation, heterothermy accounted for a smaller proportion of the hibernation season of males (93±5%) than females (98±1%) and, within the heterothermal period, males had shorter torpor bouts and longer inter-torpor arousals. Overall, males spent a smaller proportion of the hibernation season in torpor (85±6%) than females (92±1%). This sexual difference was largely attributable to the longer duration of preemergence euthermy for males than females. Males terminated torpor in January and February, when hibernacula were at their coldest, then remained euthermic for 8.8 days (range 0.5–25.0 days) before emergence. In contrast, females terminated torpor in March, when hibernaculum temperatures were increasing, then remained euthermic for only 1.1 days (range 0.5–2.0 days) before emergence. Males lost less mass per euthermic day during hibernation than females (7.0 versus 9.3 g/day). Males and females hibernated at similar depths (56 cm), but males had larger chambers than females (18 versus 16 cm3/g). Many males, but no females, cached seeds in the hibernaculum. Males met the costs of thermogenesis and euthermy from a combination of fat reserves and food caches, whereas females relied solely on fat. Access to food caches permitted males to terminate torpor several weeks in advance of emergence, during which time they recouped mass and developed sperm in preparation for the forthcoming mating season.  相似文献

A total of 1,000 lymphocyte interphase nuclei per proband from 90 females and 138 males age 1 wk to 93 years were analyzed by in situ hybridization for loss of the X and Y chromosomes, respectively. Both sex chromosomes showed an age-dependent loss. In males, Y hypoploidy was very low up to age 15 years (0.05%) but continuously increased to a frequency of 1.34% in men age 76-80 years. In females, the baseline level for X chromosome loss is much higher than that seen for the Y chromosome in males. Even prepubertal females show a rate of X chromosome loss, on the order of 1.5%-2.5%, rising to approximately 4.5%-5% in women older than 75 years. Dividing the female probands into three biological age groups on the basis of sex hormone function (< 13 years, 13-51 years, and > 51 years), a significant correlation of X chromosome loss versus age could clearly be demonstrated in women beyond age 51 years. Females age 51-91 years showed monosomy X at a rate from 3.2% to 5.1%. In contrast to sex chromosomal loss, the frequency of autosomal monosomies does not change during the course of aging: Chromosome 1 and chromosome 17 monosomic cells were found with a constant incidence of 1.2% and 1%, respectively. These data also indicate that autosome loss in interphase nuclei is not a function of chromosome size.  相似文献

Mixed populations of the twospotted spider mite (TSM),Tetranychus urticae (Koch), and the Banks grass mite (BGM),Oligonychus pratensis (Banks), occur on corn and sorghum plants in late summer in the Great Plains. Interspecific matings between these arrhenotokous species occur readily in the laboratory but yield no female offspring. The effect of interspecific mating on female: male sex ratios was measured by examining the F1 progeny of females that mated with both heterospecific and conspecific males in no-choice situations. TSM females that mated first with BGM males and then with TSM males produced a smaller percentage of female offspring than TSM females that mated only with TSM males (43.1±5.8 and 78.9±2.8% females, respectively). Similarly, BGM females mated with heterospecific males and then with conspecific males produced fewer female offspring than females mated only with BGM males (55.7±5.2 and 77.5±2.5%, respectively). Lower female: male sex ratios were produced also by BGM females that mated with TSM males after first mating with conspecifics (62.4±3.4%). In mixed populations containing males of both species, females also produced lower female: male sex ratios, but these ratios were not as low as expected based on mating propensities and progeny sex ratios observed in no-choice tests. These data suggest that interspecific mating may substantially reduce female fitness in both mite species by reducing the output of female offspring, but in mixed populations this effect is mitigated by unidentified behavioral mechanisms.  相似文献

We estimated queen mating frequency, genetic relatedness among workers, and worker reproduction in Vespa crabro flavofasciata using microsatellite DNA markers. Of 20 colonies examined, 15 contained queens inseminated by a single male, 3 colonies contained queens inseminated by two males, and 2 colonies contained queens inseminated by three males. The genetic relatedness among workers was estimated to be 0.73±0.003 (mean±SE). For this high relatedness, kin selection theory predicts a potential conflict between queens and workers over male production. To verify whether males are derived from queens or workers, 260 males from 13 colonies were genotyped at four microsatellite loci. We found that all of the males were derived from the queens. This finding was further supported by the fact that only 33 of 2,990 workers dissected had developed ovaries. These workers belonged to 2 of the 20 colonies. There was no relationship between queen mating frequency and worker reproduction, and no workers produced male offspring in any of the colonies. These results suggest that male production dominated by queens in V. crabro flavofasciata is possibly due to worker policing.  相似文献

A recent conceptual model of spatial organization in vertebrates, based upon changes in home range overlap with habitat quality, ‘the space-use model’, predicts large and strongly overlapping home ranges and absence of territorial behaviour in habitats with poor food availability. We investigated whether the model can be extended to predict intra-population variation in space-use in a habitat with strong temporal variation in resource abundance. We studied space use of Eurasian red squirrels (Sciurus vulgaris) in subalpine conifer forests over 4 years with strong fluctuations in seed production. Norway spruce dominated forests are resource limited habitats for squirrels, and tree seed availability is gradually depleted in the year following a poor seed-crop. Male and female squirrels responded to spruce seed-crop failure in 2000 by strongly increasing their home ranges and core-areas in the following year (mean MCP ± SE in summer and autumn, respectively: 2001, 122.9±14.1 ha and 84.3±9.4 ha, against 43.0±6.4 ha and 20.8±3.8 ha in 2002, and 22.7±1.5 ha and 21.4±2.0 ha in 2003). In 2001, half of the animals, those with the poorest quality pre-dispersal home ranges, emigrated to areas with more larch. Residents had multi-nuclear core-areas. Also, intra-sexual core-area overlap (males by males, females by females) was higher in 2001 and summer 2002, than in autumn 2002 and in 2003 (means ± SE in summer and autumn, respectively: males by males, 2001, 52±8 and 44±7%, 2002, 80±30 and 15±11%, 2003, 37±19 and 26±14%; females by females 2001, 52±10 and 112±32%, 2002, 55±27 and 0±0%, 2003, 12±8 and 0.1±0%). Red squirrels responded to food shortage by moving to patches with other food resources and abandoned the spacing pattern of reduced core-area overlap among males and nearly exclusive core-areas among females, found in less variable habitats. After richer seed-crops in 2001 and 2002, it took squirrels about a year to reduce the size of their home ranges and core-areas and return to a spacing pattern of stable home ranges and intra-sexual territoriality of adult females. These results are consistent with the space-use model and show that spacing behaviour in red squirrels is a plastic, conditional strategy with individuals adapting the size and/or location of their home ranges in relation to local distribution and abundance of food resources.Co-ordinating editor: R. Summary. The genetic variance of queen mating frequency was studied in honeybees (Apis mellifera carnica). Worker offspring (N = 966) of 28 naturally mated half sister-queens (r = 0.25) from seven unrelated breeding lines were genotyped at four DNA microsatellites. The mating frequencies of the queens were derived from the offspring genotypes. The number of observed matings per queen ranged from 10 to 28 with an average of 17.32 ± 1.10 (number of estimated matings: 24.94 ± 2.51; number of effective matings: 20.09 ± 1.73). Half-sib analyses of the breeding lines were used to estimate heritability. Heritability was h2 = 0.449 ± 0.135 for the estimated number of matings and h2 = 0.262 ± 0.103 for the number of effective males, which are both significantly different from zero. We conclude that a high genetic variance for polyandry in honeybees can be favored by balanced selection between individual queen and colony level.Received 16 October 2003; revised 4 May 2004; accepted 4 May 2004.  相似文献

Development, survival and fecundity for Scatella (Teichomyza) fusca Maquart (Diptera: Ephydridae) were studied at 20 ± 1 °C and 85 ± 10% r.h. Mean (± S.E.) developmental times of eggs, larvae and pupae were 4.0 ± 0.06, 14.9 ± 0.19 and 14.6 ± 0.11 days, respectively, the mean (± S.E.) survival of the original egg cohort to the start of larval, pupal and adult stage being 77.2 ± 3.2%, 54.5 ± 2.6% and 47.9 ± 3.5%, respectively. Females and males displayed approximately straight survivorship curves during adult life, implying constant mortality rates. Mean (± S.E.) adult longevity was 41.6 ± 2.98 days for females and 51.2 ± 3.91 days for males. Assuming a stable age distribution the population consisted of 56% eggs, 31% larvae, 6% pupae and 7% adults. Oviposition peaked when females were 25 days old, and the highest reproductive values (RVx) (mean ± S.E.) ranged from 129.1 ± 7.57 to 138.5 ± 6.83 for individuals 17–27 days old. A female deposited a mean (± S.E.) of 614.7 ± 35.9 female eggs over a maximum life span of 93 days. The basic reproductive rate (R 0) (mean ± S.E.) was 173.0 ± 14.2 female offspring per female and the intrinsic rate of natural increase of female individuals (r) (mean ± S.E.) was 0.088 ± 0.001 day-1. The mean (± S.E.) generation time (T) was 57.8 ± 0.78 days. In cultures with equal numbers of first instar S. fusca larvae and predacious third instar larvae of Hydrotaea aenescens (synonymous Ophyra aenescens) Wiedemann, mean (± S.E.) survival to the adult stage of S. fusca (16.7 ± 8.8%) was significantly lower than in controls with S. fusca alone (58.3 ± 7.4%). The potential significance of predation by H. aenescens on S. fusca in pig farms is discussed.  相似文献

Sperm cryobanking could be a good alternative to breeding in captivity in order to preserve genetic diversity. Sperm from two well-characterized brown trout populations originating from two river basins in the Northwest of Spain (Esla and Duerna), both threatened by extinction, was cryopreserved. In order to determine whether a sperm cryobank is the best option for preserving genetic profiles, cell viability, chromatin fragmentation, fertility and genetic variability of the offspring obtained with fresh and frozen sperm, were analyzed. Sperm viability was not reduced by freezing (87.0 ± 3.32% to 77.9 ± 3.59% and 77.6 ± 6.53% to 76.6 ± 2.61% in fresh and frozen sperm from Esla and Duerna, respectively). The percentage of fragmented DNA increased after freezing in spermatozoa from Esla males (from 4.7 ± 0.23% to 6.0 ± 0.28%), but not those from Duerna males.After freezing/thawing, the percentage of eyed embryos drops from 66.8 ± 6.77% to 16.1 ± 3.46% and from 50 ± 8.97% to 11.5 ± 2.50% in the Esla and Duerna basins, respectively. This reduction indicates that many spermatozoa have lost their ability to contribute to embryo development and this loss is not related to either spermatozoa viability or the DNA integrity. Genotypic determination by microsatellite analysis showed that frozen/thawed sperm provided offspring with a similar genetic profile to unfrozen milt, demonstrating the accuracy of the cryopreservation procedure.Taking into account the prolificacy of this species, even a low rate of success of fry after cryopreservation, could provide enough individuals to recover stable populations without altering the genetic profiles of the preserved strains. Therefore, cryopreservation is considered a safe, simple and cheap technology for gene banking in the analyzed brown trout populations.  相似文献

The objective was to establish a protocol for the collection and evaluation of epididymal sperm in agoutis. Eight males (1-2 y old) underwent left orchidectomy and epididymal sperma were collected by retrograde flush. Average values were flush volume 32 μL, pH 6.9, sperm concentration 748 x 106 sperm/mL, with motility 86.5% and vigor 4.6. Viable sperm were present in all flush samples; 66% of sperm were alive, and 41.9% of sperm responded positively to the hypoosmotic test (using distilled water). There were 21.1% morphologically abnormal sperm, of which 2.0 and 19.1% were primary and secondary defects, respectively. The acrosome was intact in 99.5% of sperm. The sperm head was 4.89 ± 0.41 μm long and 3.13 ± 0.35 μm wide, with an area of 13.01 ± 2.01 μm2. Midpieces were 5.33 ±0.44 μm long and 0.98 ± 0.13 wide, sperm tails were 29.91 ± 2.29 μm, and overall sperm length was 40.12 ± 2.44 μm. In conclusion, epididymal sperm collection from agoutis was satisfactory; the collected sperm has the potential to be stored, facilitating development of other reproductive biotechnologies for this species.  相似文献

Although numbers were low in winter, males of Unionicola intermedia were found throughout the year. A mean of 2.5 (±0.26 SE) males per mussel was found with a range of 0 to 20. Females had a mean of 13.07 (±0.87 SE) per mussel and a range from 0 to 57, with the highest mean in August. The incidence of males was much lower than that of females. Nymphs and females were highly aggregated and males showed aggregation at a lower level of significance for nine months of the year. Although the primary sex ratio is unknown, the tertiary sex ratio is 86% female and on a monthly basis ranged from 68.9% to 96.7% female. It is speculated that reproduction is arrhenotokous. The sex ratio and the significant seasonal variations of the active stages may be influenced by the dispersal pattern of this species and by the biology of its host.  相似文献

The aim of this study was to assess the following parameters in prepubertal goat oocytes of different follicle diameter (≥3 mm, <3 mm, control): oocyte diameter, early (Annexin-V) and late (TUNEL) apoptosis, embryo development and chromosomal ploidy of these blastocysts using Fluorescence In Situ Hybridization (FISH). Before in vitro maturation, oocytes were measured and stained with Annexin-V or TUNEL. The rest of the oocytes were matured, fertilized, and cultured in vitro for 8 days. Oocytes from follicles of ≥3 mm showed greater mean oocyte diameter (128.27 ± 7.20 μm vs. 125.35 ± 7.59 μm), higher percentages of TUNEL positive (42.86 vs. 24.23%), higher cleavage (47.85 ± 3.98 vs. 23.07 ± 2.44 %) and blastocyst rates (19.77 ± 3.04 vs. 4.11 ± 1.10 %) than oocytes from follicles of <3 mm.. Blastocyst mean cell numbers did not show differences between follicular groups (123.83 ± 49.62 vs. 104.29 ± 36.09 for follicles of ≥3 mm and <3 mm, respectively). A total of 54 blastocysts with 7084 nuclei were hybridized with specific probes to chromosomes X and Y. Ninety-eight percent (98%) of the embryos presented at least one cell carrying an abnormal number of chromosomes, but 78% of them presented less than 25% of chromosomal abnormal cells. No differences in the percentage of blastocysts with abnormal ploidy were found in embryos produced from oocytes of different follicle diameter.  相似文献

The objective of this study was to describe the clinical findings, semen characteristics, and testicular histopathology in male dromedary camels affected with impotentia generandi (IG). According to the history, 82.6% (38/46) of the cases were classified as primary-IG (P-IG; never been able to impregnate a female), whereas 17.4% (8/46) were classified as secondary-IG (S-IG; acquired infertility). Only one scrotal testis was observed in four cases, and no scrotal testis was observed in one case. Overall, testicular length, width, and depth were 6.46 ± 0.2, 3.41 ± 0.1, and 2.8 ± 0.08 cm, respectively. Within the P-IG males, 42.2% of the testes were classified as small, 47.9% as normal, and 9.9% as large. Within the S-IG males, 0.0% of the testes were classified as small, 80% as normal, and 20% as large. Ejaculate volume, total sperm number in the ejaculate, and sperm motility, viability, and abnormal morphology were 4.4 ± 0.3 mL, 25.7 ± 1.0 × 106, 18.7 ± 3.1%, 25.2 ± 3.4%, and 46.6 ± 3.7%, respectively. Azoospermia was observed in 30.4% of the cases, asthenospermia was observed in the 25% of the cases, and necrospermia was observed in 10% of the cases. The proportion of abnormal sperm was between 20% and 50%, and between 60% and 94% in 56.2% and 34.4% of the cases, respectively. Hypospermatogenesis, arrested spermatogenesis, Sertoli cell–only syndrome, and testicular degeneration were the main histopathological findings. In conclusion, IG in male dromedary camels appears to be related mainly to testicular dysfunction, which alters semen quality and reduces fertility.  相似文献
