非使用价值是环境与资源价值的重要组成部分,非使用价值的支付动机研究是环境与资源价值研究的重要内容,遗产价值、选择价值、存在价值是非使用价值的三大支付动机.传统的自上而下或自下而上非使用价值支付动机研究方法会出现顺序效应等问题.根据社会心理学理论,消费者支付意愿的大小与其支付动机强度成正比,为此提出了基于认知水平的非使用价值支付动机分析方法,将对支付动机的直接评估转为对动机认知水平的评估.论文以鼓山风景区为例进行了实证分析,并与传统方法进行比较,结果显示,基于认知水平的支付动机研究方法可大大减低受访者回答的难度,并能精确地量化受访者3种支付动机间的差异,同时克服了顺序效应问题,从而提高了研究结果的科学性.  相似文献

Seven improved cultivars of pigeon pea (Cajanus cajan (L.) Mill sp.) were evaluated at 0 (original Zn deficient soil), 5 and 50 ppm Zn levels under greenhouse conditions. Plants were harvested at 6 weeks after sowing and at maturity. Responses to 5 ppm Zn in shoot at 6 weeks of growth, and in leaf, stem, pod-hull and grain at maturity ranged from 63 to 387, 37 to 116, 15 to 73,9 to 145 and 51 to 200%, respectively. Application of 50 ppm Zn in most of the cultivars did not markedly affect the yield of different plant parts. Zinc concentration at 0 Zn level in shoot at 6 weeks of growth and in leaf, stem, pod-hull and grain of different genotypes varied from 9.8 to 14.5, 13.7 to 21.2, 10.8 to 16.7, 4.17 to 5.83 and 9.2 to 16.7 ppm, respectively, and the increase in concentration with 5 ppm applied Zn ranged from 28 to 248, 28 to 89, 27 to 85, 20 to 142, and 105 to 254 per cent, respectively. The concentration further increased with an increase in Zn level to 50 ppm. There was less variation in the yield and tissue Zn concentration of different genotypes after Zn application. Phosphorus concentration at 0 Zn level in shoot at 6 weeks of growth, and in leaf, stem, pod-hull and grain of different genotypes varied from 0.50 to 0.71, 0.18 to 0.31, 0.11 to 0.24, 0.15 to 0.20 and 0.43 to 0.58% respectively. Zinc decreased P in all plant parts but relative decrease was more in vegetative parts than in grain. The variability in Zn response among pigeon pea genotypes could partly be attributed to the maintenance of proper P/Zn balance in metabolically active plant parts, such as, leaf, and partly to their capacity to exploit soil Zn and to translocate it to the above-ground parts.Contribution from the Department of Soils, Haryana Agricultural University, Hissar (India).  相似文献

On a global scale, changing climates are affecting ecological systems across multiple levels of biological organization. Moreover, climates are changing at rates unprecedented in recent geological history. Thus, one of the most pressing concerns of the modern era is to understand the biological responses to climate such that society can both adapt and implement measures that attempt to offset the negative impacts of a rapidly changing climate. One crucial question, to understand organismal responses to climate, is whether the ability of organisms to adapt can keep pace with quickly changing environments. To address this question, a syntheses of knowledge from a broad set of biological disciplines will be needed that integrates information from the fields of ecology, behavior, physiology, genetics, and evolution. This symposium assembled a diverse group of scientists from these subdisciplines to present their perspectives regarding the ability of organisms to adapt to changing climates. Specifically, the goals of this symposia were to (1) highlight what each discipline brings to a discussion of organismal responses to climate, (2) to initiate and foster a discussion to break barriers in the transfer of knowledge across disciplines, and (3) to synthesize an approach to address ongoing issues concerning biological responses to climate.  相似文献

Human papillomavirus type 16 (HPV16) is the primary etiologic agent of cervical carcinoma, whereas bovine papillomavirus type 1 (BPV1) causes benign fibropapillomas. However, the capsid proteins, L1 and L2, of these divergent papillomaviruses exhibit functional conservation. A peptide comprising residues 1 to 88 of BPV1 L2 binds to a variety of cell lines, but not to the monocyte-derived cell line D32, and blocks BPV1 infection of mouse C127 cells. Residues 13 to 31 of HPV16 L2 and BPV1 L2 residues 1 to 88 compete for binding to the cell surface, and their binding, unlike that of HPV16 L1/L2 virus-like particles, is unaffected by heparinase or trypsin pretreatment of HeLa cells. A fusion of HPV16 L2 peptide 13-31 and GFP binds (K(d), approximately 1 nM) to approximately 45,000 receptors per HeLa cell. Furthermore, mutation of L2 residues 18 and 19 or 21 and 22 significantly reduces both the ability of the HPV16 L2 13-31-GFP fusion protein to bind to SiHa cells and the infectivity of HPV16 pseudovirions. Antibody to BPV1 L2 peptides comprising residues 115 to 135 binds to intact BPV1 virions, but fails to neutralize at a 1:10 dilution. However, deletion of residues 91 to 129 from L2 abolishes the infectivity of BPV1, but not their binding to the cell surface. In summary, L2 residues 91 to 129 contain epitopes displayed on the virion surface and are required for infection, but not virion binding to the cell surface. Upon the binding of papillomavirus to the cell surface, residues 13 to 31 of L2 interact with a widely expressed, trypsin- and heparinase-resistant cell surface molecule and facilitate infection.  相似文献

Oligosaccharide patterns obtained by gel filtration of the urine of GM1-gangliosidosis Type 1 patients are quite different from those of GM1-gangliosidosis Type 2. Neurodegeneration or Neuroprotection: The Pivotal Role of Astrocytes   总被引:11,自引:0,他引:11  
Formation of nitric oxide (NO), by astrocytes, has been suggested to contribute, via impairment of mitochondrial function, to the neurodegnerative process. Thus co-culture of neuronal cells with NO-generating astrocytes leads to a loss of mitochondrial function, as reflected by diminished activities of complexes IV and II+III. However, such damage may in the first instance be limited due to upregulation of neuronal glutathione metabolism as a result of metabolic trafficking of glutathione from the astrocyte to neurone. Furthermore, exposure of astrocytes to NO leads to increased glutathione metabolism resulting in the preservation of glutathione precursors for neuronal utilization. Failure of glutathione trafficking could render neuronal cells particularly susceptible to NO, leading to cell death. In addition, depletion with time of the nitric oxide synthase cofactor, tetrahydrobiopterin, may result in the astrocytic generation of more potent oxidizing species, which could contribute to the neurodegenerative process.  相似文献

安徽省啮齿动物区系情况,解放前Allen(1940)、Gee(1930)、Tate(1947),阿部余四男(1944)曾有过一些零星报道,解放后盛和林等(1963)、王岐山等(1966)、黄文几等(1978)也进行过调查,但就啮齿动物区系而言,尚缺乏完整的系统材料。我站从1958年起,开始对本省啮齿动物进行调查,重点主要在平原及丘陵地带,对皖南及大别山区调查较少。近年来,我们在皖南山区的祁门、休宁、绩溪、黄山、宁国、宣城、泾县、歙县、青阳及大别山区的金寨、霍山、岳西、潜山、舒城等山地进行了补点调查。现将调查结果,结合以往的工作和文献作初步整理,并就啮齿动物的地理分布和优势种,综合自然地理等因素进行啮齿动物的地理区划。  相似文献

Nasonovia ribisnigri, a main pest of salad crops, has developed resistance to various insecticides in southern France, including the carbamate pirimicarb and the cyclodiene endosulfan, two insecticides widely used to control this aphid. Here we have investigated the mechanisms of resistance to these two insecticides by studying cross-resistance, synergism, activity of detoxifying enzymes, and possible modifications of the target proteins. Resistance to pirimicarb was shown to be mainly due to a decreased sensitivity of the target acetylcholinesterase; this modification conferred also, resistance to propoxur but not to methomyl and the two tested organophosphates (acephate and paraoxon). Endosulfan resistance was associated with a moderate level of resistance to dieldrin, and resistance to both insecticides was due, in part, to increased detoxification by glutathione S-transferases (GST). The endosulfan resistant strain displayed the same amino acid at position 302 of the Rdl gene (GABA receptor) as susceptible aphids (e.g. Ala), indicating that the Ala to Ser (or to Gly) mutation observed among dieldrin resistant strains of other insect species was not present.  相似文献

The article discusses the relationship between the notion of a moral right to personal self-determination, the notion of a moral right to know and the notion of a moral right not to know. In particular, the author asks under what conditions, if any, the right to self-determination implies a right to have information or a right not to have information. The conclusions he defends are theoretical in character rather than concrete norms and directions, and they are intended to be relevant in many contexts, not only in the context of information concerning, say, medical decisions. Yet the author restates the question concerning the right to have and especially the right not to have genetic knowledge about one's own genes on the basis of a right to self-determination. He argues that a right (not) to have genetic information can be defended on the ground of a right to self-determination, but that this kind of defense presupposes several normative, and controversial, arguments.  相似文献

Effects on reproduction of dystocia, stillbirth, abortion, milk fever, retained placenta, metritis, cystic ovaries, anestrus, ketosis, displaced abomasum, locomotor disorders, and mastitis were reviewed. Papers were considered if they provided quantitative estimates of diseases on days to first estrus, days to first service, conception rate at first service, days from first service to conception, days to conception or days open, calving interval, conception rates at various days post partum (dpp), and number of services per conception or per cow. Only papers in English in peer-reviewed journals were selected for analysis of post 1960 data from intensive dairy regions. Seventy papers fulfilled the selection criteria. Summary estimates of disease effects were calculated according to meta-analysis methods, and study designs were described in detail to identify possible heterogeneity of the results. Stillbirth, milk fever, displaced abomasum and mastitis had no effect on reproduction. Clinical ketosis, dystocia and retained placenta were associated with 2 to 3 more days to first service and with a 4 to 10% lower conception rate at first service, resulting in 6 to 12 more days to conception. Locomotor disorders were associated with an average increase of 12 d to conception, with wide variation depending on lesions and stage of occurrence. Metritis was associated with 7 more days to first service, 20% lower conception rate at first service, resulting in 19 more days to conception. Cystic ovaries were associated with 6 to 11 more days to first service and with 20 to 30 more days to conception. Anestrus was associated with 26 more days to first service and with an 18% lower conception rate at first service, resulting in 41 more days to conception. Abortion was associated with 70 to 80 more days to conception.  相似文献

In earlier work in which electrorotation was used to apply external torque to tethered cells of the bacterium Escherichia coli, it was found that the torque required to force flagellar motors backward was considerably larger than the torque required to stop them. That is, there appeared to be substantial barrier to backward rotation. Here, we show that in most, possibly all, cases this barrier is an artifact due to angular variation of the torque applied by electrorotation, of the motor torque, or both; the motor torque appears to be independent to speed or to vary linearly with speed up to speeds of tens of Hertz, in either direction. However, motors often break catastrophically when driven backward, so backward rotation is not equivalent to forward rotation. Also, cells can rotate backward while stalled, either in randomly timed jumps of 180 degrees or very slowly and smoothly. When cells rotate slowly and smoothly backward, the motor takes several seconds to recover after electrorotation is stopped, suggesting that some form of reversible damage has occurred. These findings do not affect the interpretation of electrorotation experiments in which motors are driven rapidly forward.  相似文献

应对气候变化和保护生物多样性是2大全球性热点环境问题。气候变化导致物种多样性丧失、生态系统服务降低和区域生态安全屏障功能受损,威胁到中国国土生态安全格局和生态脆弱区域的可持续发展,给生物多样性保护带来新的挑战。做好生物多样性保护适应气候变化的风险管理工作,既是生物多样性应对气候变化风险的必要措施,也是减缓气候变化的重要途径。结合爱知目标10的实现情况,分析了欧盟、澳大利亚、美国等发达国家发布的生物多样性适应气候变化技术政策制定情况、中国生物多样性应对气候变化进展情况,剖析了中国生物多样性保护适应气候变化存在的问题,包括生物多样性应对气候变化的科学认知亟待提高、生物多样性保护适应气候变化的能力建设不足、自然保护地之间缺乏适应气候变化的生态廊道网络、生物多样性保护适应气候变化的技术标准缺乏。研究提出了中国生物多样性应对气候变化的适应性管理策略,包括制定《中国生物多样性保护协同应对气候变化的国家方案》、加强生物多样性保护适应气候变化的能力建设、开展自然保护区适应气候变化的风险管理试点、强化生物多样性应对气候变化的科技支撑,以期为推进纳入气候变化风险管理的生物多样性保护工作提供决策依据。  相似文献

Methodology consitutes the study of methods considered as < manners of leading its thought, of establishing or of showing truths according to certain principles and with a certain order >. We will see that medical < truths >, knowledge, could be of an extremely different nature: factual knowledge is a simple observation, often with operational vocation\; theoretical knowledge belongs to a coherent whole, it fascines and facilitates our representations of pathologies\; the causal knowledge is often the subject of extreme interrogations in medical practice\; it is of primarily metaphysical nature. These distinctions are not without consequence in methodological terms. Theories are useful because they facilitate the choice of the assumptions to be tested, the variables to be measured and the interpretation of the results of experiments. The risk however exists to be unable to think apart from their framework, and thus to neglect any source of knowledge which they could not integrate. The cause is inaccessible to science\; it will be necessary, in practice, to be limited to the search for variables of upstream on which action of prevention or cure could be undertaken. To establish, to show this medical knowledge, it is necessary to resort to the scientific method by formulating refutable assumptions by reproducible experiments, and that this process involves a belief in the found results. It appears thus that for the same experimental plan, the level of belief and thus the level of proof of the results is a function of the type of measurement carried out (commonplace measurement or of high technicality), of the medical discipline and the media character or not of studied pathology, even of stakes of being able. The medical reproducibility of experiments is problematic since results of these experiments are in general random. This results from the considerable number of variables entering in the determination of human pathologies, and that the majority of these variables are either unknown, or impossible to control. Randomisation makes it possible to give a probabilistic character to this risk: it is then possible to resort to mathematics to carry out statistical tests, to calculate confidence intervals and to profit from a possibility of making inferences with a known margin of error. In the absence of randomisation, it is possible to resort to models\; they imply however the knowledge of statistical methods, reserve, experience and a great intellectual honesty during the interpretation of the results, finally a replication of the experiments is here, more than elsewhere, essential.  相似文献

In order to promote cell attachment, fibronectin must first undergo activation by a suitable substrate. In this study, 52 materials have been surveyed for their ability (a) to bind fibronectin, (b) to activate the cell-adhesive property of fibronectin, and (c) to support the growth of cells. Many plastics, polysaccharides, metals, and ceramics were found to support cell growth as well as the fibronectin-dependent attachment of cells. Several other substrates have been identified that were inactive in promoting either cell attachment or growth. Hydrophobic substrates were found to be active in fibronectin activation, whereas hydrophilic substrates were found to be inactive. Since fibronectin binds to substrata of extremely varied chemical composition, it is clear that the binding of fibronectin to such substrata is nonspecific in nature. Since protein pretreatment of all substrata, except collagen and poly(L-lysine), abolished the physical binding of fibronectin, the binding of fibronectin to artificial substrata is probably ascribable to a nonspecific hydrophobic protein-substratum interaction. In contrast, several lines of evidence indicate that the interaction between fibronectin and collagen displays biological specificity. Poly(hydroxyethylmethacrylate)(poly(HEMA)), which has previously been shown to be nonadhesive for cells, is demonstrated here to be unique in its inability to bind fibronectin. Addition of one part per million of an adhesive polymer to poly(HEMA) permits fibronectin binding to occur.  相似文献

Farming is the basis of our civilization yet is more damaging to wild nature than any other sector of human activity. Here, we propose that in order to limit its impact into the future, conservation researchers and practitioners need to address several big topics--about the scale of future demand, about which crops and livestock to study, about whether low-yield or high-yield farming has the potential to be least harmful to nature, about the environmental performance of new and existing farming methods, and about the measures needed to enable promising approaches and techniques to deliver on their potential. Tackling these issues requires conservationists to explore the many consequences that decisions about agriculture have beyond the farm, to think broadly and imaginatively about the scale and scope of what is required to halt biodiversity loss, and to be brave enough to test and when necessary support counterintuitive measures.  相似文献

One of the main causes of failure of fluorescence in situ hybridization with rRNA-targeted oligonucleotides, besides low cellular ribosome content and impermeability of cell walls, is the inaccessibility of probe target sites due to higher-order structure of the ribosome. Analogous to a study on the 16S rRNA (B. M. Fuchs, G. Wallner, W. Beisker, I. Schwippl, W. Ludwig, and R. Amann, Appl. Environ. Microbiol. 64:4973-4982, 1998), the accessibility of the 23S rRNA of Escherichia coli DSM 30083(T) was studied in detail with a set of 184 CY3-labeled oligonucleotide probes. The probe-conferred fluorescence was quantified flow cytometrically. The brightest signal resulted from probe 23S-2018, complementary to positions 2018 to 2035. The distribution of probe-conferred cell fluorescence in six arbitrarily set brightness classes (classes I to VI, 100 to 81%, 80 to 61%, 60 to 41%, 40 to 21%, 20 to 6%, and 5 to 0% of the brightness of 23S-2018, respectively) was as follows: class I, 3%; class II, 21%; class III, 35%; class IV, 18%; class V, 16%; and class VI, 7%. A fine-resolution analysis of selected areas confirmed steep changes in accessibility on the 23S RNA to oligonucleotide probes. This is similar to the situation for the 16S rRNA. Indeed, no significant differences were found between the hybridization of oligonucleotide probes to 16S and 23S rRNA. Interestingly, indications were obtained of an effect of the type of fluorescent dye coupled to a probe on in situ accessibility. The results were translated into an accessibility map for the 23S rRNA of E. coli, which may be extrapolated to other bacteria. Thereby, it may contribute to a better exploitation of the high potential of the 23S rRNA for identification of bacteria in the future.  相似文献

Disturbances (pulse, press and ramp) constitute a major force influencing, even determining, the structure and functions of ecological components – populations, communities and ecosystems. The capacity to weather a disturbance without loss is defined as resistance, whereas resilience is the capacity to recover from a disturbance after incurring losses, which may be considerable. This article seeks to resolve differences in the ecological definition of resistance and of resilience and to examine the importance of resilience as applied to ecological restoration. In restoration, interventions are designed and implemented with the aim of strengthening the resilience, that is, the capacity to recover, of degraded systems. In response to restorative measures, degraded systems may have both resistance and negative resilience to remain in the degraded state. The key aim of restoration is to overcome the resistance and negative resilience of the degraded state by strengthening positive resilience, the capacity to recover to the intact undegraded state. Restoration may be hindered by a lack of knowledge of acting disturbances (both past and present), of the previous intact condition and of appropriate interventions to implement. Resilience to a disturbance is determined after the disturbance has ceased. Thus, for current ongoing press and ramp disturbances, resilience can be hard to determine.  相似文献
