In terms of the quality of minimally processed fruit, flesh browning is fundamentally important in the development of an aesthetically unpleasant appearance, with consequent off-flavours. The development of browning depends on the enzymatic action of the polyphenol oxidase (PPO). In the ‘Golden Delicious’ apple genome ten PPO genes were initially identified and located on three main chromosomes (2, 5 and 10). Of these genes, one element in particular, here called Md-PPO, located on chromosome 10, was further investigated and genetically mapped in two apple progenies (‘Fuji x Pink Lady’ and ‘Golden Delicious x Braeburn’). Both linkage maps, made up of 481 and 608 markers respectively, were then employed to find QTL regions associated with fruit flesh browning, allowing the detection of 25 QTLs related to several browning parameters. These were distributed over six linkage groups with LOD values spanning from 3.08 to 4.99 and showed a rate of phenotypic variance from 26.1 to 38.6%. Anchoring of these intervals to the apple genome led to the identification of several genes involved in polyphenol synthesis and cell wall metabolism. Finally, the expression profile of two specific candidate genes, up and downstream of the polyphenolic pathway, namely phenylalanine ammonia lyase (PAL) and polyphenol oxidase (PPO), provided insight into flesh browning physiology. Ceratitis capitata (Wiedemann), the Mediterranean fruit fly, is one of the key pest species affecting deciduous fruit orchards along the Mediterranean coasts. Because of global warming, C. capitata is gradually spreading north and is becoming a major pest of apples. Determining the susceptibility of the main apple varieties grown in the region will serve as a cornerstone to the management of this pest. In this study, we show the results of a field and laboratory no‐choice test conducted to determine the Medfly preferences on different apple cultivars. The seven main varieties of apples (Gala, Red Delicious, Golden Delicious, Granny Smith, Kanzi, Morgen Dallago and Fuji) were tested. The results demonstrate that C. capitata lays eggs on all apple cultivars in both field and laboratory conditions. The Granny Smith, Red Delicious and Morgen Dallago varieties showed the lowest susceptibility in laboratory conditions, (0.75, 1.55, 2 oviposition punctures/fruit, respectively), with significant differences in oviposition compared to the Golden Delicious, Kanzi and Fuji (3.27, 3.31, 3.1 oviposition punctures/fruit, respectively) varieties, which were shown to be the most susceptible to Medfly attack in laboratory conditions. On the other hand, only slight and not statistically significant differences emerged from the field trials. In relation to the physico‐chemical characteristics, the apple cultivars showing the lowest susceptibility (Granny Smith, Red Delicious and Morgen Dallago) had harder peels and pulps and lower sugar contents than the most susceptible cultivars (Golden Delicious, Fuji and Kanzi). These results were also confirmed through evaluation of larval development on different varieties. In fact, Granny Smith, Red Delicious and Morgen Dallago were the three varieties that did not allow adequate larval and adult development and reduced the possibility of the emergence of a new generation.  相似文献

The rate of entry by neonate larvae of the frugivorous codling moth, Cydia pomonella (L.) (Lepidoptera: Tortricidae), into fruit material was investigated. We used no‐choice bioassays in climate‐controlled rooms to assay larval entry across four host plant species (apple, pear, quince, walnut) and three varieties within a single fruit species (apple). Larvae successfully entering apples were reared to adulthood, and we collected tissue samples from apples which were successfully colonized in order to determine sucrose concentrations. This information was used to evaluate differences in adult moth size, development time, and pulp sucrose concentration due to apple variety. Four important findings emerged: (1) neonate larvae had the highest frequency of entry (86% of larvae) into apple fruits, compared with pear (78%), quince (56%), and walnut (32%); (2) the frequency of larval entry into immature apples differed across apple varieties, and larval entry rate was highest in variety Golden Delicious (72%), compared with Granny Smith (46%) and Red Delicious (64%); (3) on average, adult moths were larger and development times were shorter on the variety with the highest entry frequency (Golden Delicious); and (4) apple pulp sucrose concentrations were higher for Golden Delicious (17.5 μg mg?1) than for either Granny Smith (15.9 μg mg?1) or Red Delicious (15.1 μg mg?1) varieties, which correlates positively with entry and development data. We conclude that host fruit species and varietals within a species affect the entry rate of neonate codling moth larvae in no‐choice assays. We hypothesize that larval development is influenced by mean sucrose concentrations or other phytochemical differences associated with host fruit varieties.  相似文献

Host specificity tests of the moth,Microthrix inconspicuella Ragonot in Australia, indicated that larvae could feed and develop on young apple leaves. Additional tests in South Africa on leaves and fruit of the 4 apple varieties, Jonathan, Starking (Red Delicious), Granny Smith and Golden Delicious, showed that apples were not a preferred food. Little feeding occurred and pupation happened infrequently. No 2nd generation resulted whenM. inconspicuella colonies were confined on apple fruit or leaves.   相似文献

The sequence of events which occurs during the ripening of the Passe-Crassane pear fruit have been previously studied. In this work, we have investigated the ripening of another climacteric fruit (Pyrus malus L. cv Golden Delicious) and of a nonclimacteric fruit (Prunus avium L. cv Bigarreau Napoléon). We show that both climacteric fruits exhibit the same preclimacteric sequence of events. Differences exist, however, between the Golden Delicious apple and the Passe-Crassane pear in that the protein synthesis capacity of the two fruits is not the same during the over-ripening period. On the other hand, a nonclimacteric fruit, the Bigarreau Napoléon cherry, does not show an increase in its protein synthesis capacity during the over-ripening period.  相似文献

Cacopsylla melanoneura is a univoltine psyllid vector of ‘Candidatus Phytoplasma mali’, the etiological agent of apple proliferation (AP), a severe disease in European apple orchards. The influence of ‘Ca. P. mali’ on the fitness of C. melanoneura was studied. In the spring of 2007, male-female pairs of field-collected adults were exposed to ‘Ca. P. mali’-infected or healthy ‘Golden Delicious’ apple shoots. Exposure to these diseased shoots did not affect the life span of the adult psyllids. However, significantly fewer eggs were laid on the diseased shoots. Furthermore, fewer of the eggs that were laid on the infected plants hatched. Data suggest a detrimental effect of AP phytoplasma on the fitness of C. melanoneura.  相似文献

Carposina sasakii Matsumura (Lepidoptera: Carposinidae), is one of the most serious fruit-boring pests in over ten species of fruit trees, and is especially damaging apples in the northern of China. The application of new planting systems, i.e., high-density and dwarfing rootstock orchard systems with mixed apple varieties, makes it important to study the fitness of C. sasakii on these apple varieties to gain fundamental knowledge for use in pest management involving this insect. In this study, life table data of C. sasakii were collected using Golden Delicious and Red Fuji apples as hosts. The egg-larva duration of male C. sasakii reared on Golden Delicious apples (22.81 d) was significantly shorter than that reared on Red Fuji apples (24.27 d). The egg-larva mortality in Golden Delicious apples (59.00%) was lower than that in Red Fuji apples (72.49%). The mortality of the pupal stage, however, was higher in Golden Delicious (10.51%) than in Red Fuji (0%). The total oviposition period (TPOP) on Golden Delicious apples (32.94 d) was significantly shorter than in individuals reared on Red Fuji apples (34.19 d). The intrinsic rate of increase (r = 0.0581 d−1), net reproductive rate (R0 = 7.57 offspring), and finite rate of increase (λ = 1.0598 d−1) were all higher on Golden Delicious than those on Red Fuji. When the net reproductive rate (R0) was used, the harvest rate of pupae was higher (0.8678) when reared on Golden Delicious apples than when reared on Red Fuji apples (0.8398). When a large cohort size (n = 200) was used for effective bootstrap sample, the PE values for C. sasakii reared on Golden Delicious apples and Red Fuji apples were both almost equal to 1. For C. sasakii culturing purposes, Golden Delicious apples would be more productive than Red Fuji.  相似文献

Four of five apple cultivars (Golden Delicious, Red Delicious, McIntosh, Macoun, and Melrose) inoculated with Escherichia coli O157:H7 promoted growth of the bacterium in bruised tissue independent of the date of harvest (i.e., degree of apple ripening) or the source of the apple (i.e., tree-picked or dropped fruit). Apple harvest for this study began 4 September 1998 and ended 9 October, with weekly sampling. Throughout this study, freshly picked (<2 days after harvest) McIntosh apples usually prevented the growth of E. coli O157:H7 for 2 days. Growth of E. coli O157:H7 did occur following 6 days of incubation in bruised McIntosh apple tissue. However, the maximum total cell number was approximately 80-fold less than the maximum total cell number recovered from Red Delicious apples. When fruit was stored for 1 month at 4 degrees C prior to inoculation with E. coli O157:H7, all five cultivars supported growth of the bacterium. For each apple cultivar, the pH of bruised tissue was significantly higher and degrees Brix was significantly lower than the pH and degrees Brix of undamaged tissue regardless of the source. In freshly picked apples, changes in the pH did not occur over the harvest season. Bruised Golden Delicious, McIntosh, and Melrose apple tissue pHs were not significantly different (tree-picked or dropped), and the degrees Brix values of McIntosh, Macoun, and Melrose apple tissue were not significantly different. Single-cultivar preparations of cider did not support growth of E. coli, and the cell concentration of inoculated cider declined over an 11-day test period. The rate of decline in E. coli cell concentration in the McIntosh cider was greater than those in the other ciders tested. The findings of this study suggested that the presence of some factor besides, or in addition to, pH inhibited E. coli growth in McIntosh apples.  相似文献

We report the isolation of a ripening-related apple cDNA which is complementary to a mRNA which may be involved in ethylene production. Poly(A)+ RNA was extracted from cortical tissue of ripe apple fruit (Malus domestica Borkh cv. Golden Delicious) and a cDNA library constructed in the plasmid vector pSPORT. The library was screened with pTOM13, a tomato cDNA clone thought to code for ACC oxidase in that fruit. An apple cDNA clone (pAP4) was isolated and sequenced. The 1182 bp cDNA insert includes an open reading frame of 942 bp, and shows strong homology with reported tomato and avocado sequences, both at the nucleic acid and amino acid levels. The polypeptide has a calculated molecular mass of 35.4 kDa and a calculated pI of 5.15. In apple cortical tissue, expression of pAP4-complementary RNA increased with ethylene production by the fruit during ripening. Expression was also enhanced in both ethylene-treated and wounded fruit.  相似文献

Extreme weather events like high solar radiation can cause stress in apple fruits (Malus domestica Borkh.). The aim of the study was to make a screening of individual phenols and peroxidase activity in apple peel as a response to sunburn and different sun-exposures in the period when weather conditions are suitable for sunburn occurrence. Apple fruits of ‘Golden Delicious’ and ‘Braeburn’ were sampled. Fruit temperature and color were measured prior HPLC–MS2 and peroxidase activity analyses. Sunburned peel was darker and more yellow-red in comparison to healthy peel, which appeared yellow-green. Fruit temperature, total as well as individual flavonols and dihydrochalcones, total hydroxycinnamics and perixodase activity were highest in sunburned peel in comparison with healthy sun-exposed peel, furthermore both were different than shaded sides of both fruits and peel of apples inside the tree crown; moreover in sunburned peel dihydrochalcones were determined for the first time. Chlorogenic acid was up to 2.5 times higher, 3-hydroxy-phloretin-2′-O-xyloglucoside was up to 10 times higher and quercetin-3-galactoside was up to 33 times higher in sunburned peel, comparing to shaded sided peels. Flavanols did not show a distinct pattern. A deeper insight in phenolic response against environmental stress caused by high solar radiation and high air temperatures has been made.  相似文献

Jasmonates are a class of oxylipins that induce a wide variety of higher-plant responses. To determine if jasmonates play a role in the regulation of climacteric fruit ripening, the effects of exogenous jasmonates on ethylene biosynthesis and color, as well as the endogenous concentrations of jasmonates were determined during the onset of ripening of apple (Malus domestica Borkh. cv. Golden Delicious) and tomato (Lycopersicon esculentum Mill. cv. Cobra) fruit. Transient (12 h) treatment of pre-climacteric fruit discs with exogenous jasmonates at low concentration (1 or 10 μM) promoted ethylene biosynthesis and color change in a concentration-dependent fashion. Activities of both 1-aminocyclopropane-1-carboxylic acid (ACC) oxidase and ACC synthase were stimulated by jasmonate treatments in this concentration range. The endogenous concentration of jasmonates increased transiently prior to the climacteric increase in ethylene biosynthesis during the onset of ripening of both apple and tomato fruit. The onset of tomato fruit ripening was also preceded by an increase in the percentage of the cis-isomer of jasmonic acid. Inhibition of ethylene action by diazocyclopentadiene negated the jasmonate-induced stimulation of ethylene biosynthesis, indicating jasmonates act at least in part via ethylene action. These results suggest jasmonates may play a role together with ethylene in regulating the early steps of climacteric fruit ripening. Received: 14 August 1997 / Accepted: 4 October 1997  相似文献

Development of powdery mildew (Podosphaera leucotricha) on five popular cultivars of apple, viz., Scarlet Gala, Golden spur, Mollies Delicious, Red Fuzi and Red Chief was studied to determine incidence–severity relationship. The disease was confined primarily to the vegetative terminal shoots early in the season which also traversed later onto other leaves. Several biochemical changes occur in the trees due to fungal/microbial infection. We studied the qualitative/quantitative changes in phenolic acids in apple-powdery mildew pathosystems. Scarlet Gala and Red Chief are very rich in phenolic acids, and had shown resistances to the pathogen but those with low amount of phenolic acids, viz., Golden spur, Mollies Delicious, and Red Fuzi, were highly susceptible. Thus, the quantity of phenolic acids (secondary metabolites) has been taken as a biochemical parameter in screening apple cultivars for resistance/susceptibility against powdery mildew of apple.  相似文献

Experiments with the growth regulator ethephon (2-chloroethylphosphonic acid) were made to stimulate the flower differentiation in the ‘Carola’ and ‘Golden Delicious’ apple cultivars in Pillnitz (GDR) and ‘Jonathan’ and ‘Golden Delicious’ in Ujfehértó (Hungaria). A twofold application of 150 ppm ethephon in June proved most effective. Our results indicate that it is possible to promote flower differentiation in apple trees without growth retardation and fruit thinning.  相似文献

Applied diamines and polyamines inhibited the incorporation of radioactively labeled leucine and uridine into trichloroacetic acid-insoluble material in apple (Malus domestica Borkh, cv Golden Delicious) fruit tissue. The inhibitory effect was in general more pronounced with the higher molecular weight amines. Putrescine at 5 millimolar inhibited leucine incorporation by 37% and uridine by 44%. Spermidine and spermine at the same concentration inhibited uridine incorporation by 60%. The polyamines at concentrations between 0.1 and 1.0 millimolar inhibited leucine incorporation by 55 to 90%. The inhibitory effect of 0.1 to 10 millimolar polyamines on dark- and wound-induced senescence or ethylene production, is discussed in the light of interference with macromolecular synthesis.  相似文献
