Micrococcus luteus cells cultivated in medium containing lead salts exhibited a sequence of changes in the quantity of total cellular lipids with essentially no changes from normal cellular yields. The lipid composition of cells cultivated one to four times was moderately decreased (phase I) whereas that of cells cultivated five to six times was reduced by as much as 50% (phase II). Cells cultivated more than six times in lead-containing media had progressively greater quantities of lipid (phase III) approaching that found in control cells. These cells with reestablished lipid contents showed no further effects from more prolonged exposure to lead salts. Chromatographic studies of total lipids of cells of each lipid phase revealed relatively complete lipid compositions. These results indicated that lead is apparently affecting a common biochemical parameter in the biosynthesis of lipids of lipid phase II cells. Changes in the relative quantities of individual components were observed in both the nonpolar and polar lipids in each lipid phase. The most notable changes were the decrease in aliphatic hydrocarbons with concomitant increases in the diglycerides and components identified as a complex family of ketones. Microscopy examinations of control and lead-treated cells revealed electron dense inclusion bodies in membrane fragments in only lead-treated cells.  相似文献

Previous studies from our laboratory have shown that male C57BL/6J mice excrete into the urine multilamellar lysosomal bodies that contain specific neutral glycosphingolipids. These mice excrete approximately 20-30% of their kidney glycolipids each day. The significance and function of this secretion of multilamellar lysosomal organelles is unknown. To characterize these excreted bodies further, we report here their neutral lipid and phospholipid composition. The bodies were collected by differential centrifugation, extracted with chloroform-methanol, and lipids were fractionated into neutral lipids, glycolipids, and phospholipids. The neutral lipids consisted primarily of cholesterol, dolichol, and ubiquinone. The phospholipid fraction consisted primarily of a single molecular species of phosphatidylcholine. This lipid which comprises more than 90% of the total phospholipids was found to contain 16:0 ether and C22:6 n-3 fatty acid as determined by gas-liquid chromatography-mass spectrometry. The glycosphingolipids as reported previously consisted primarily of galabiosylceramides and globotriaosylceramides. This membrane lipid composition is different from any previously reported cellular organelle.  相似文献

The effect of growth temperature on the lipid and fatty acid composition of Streptococcus faecium has been studied. No differences in the qualitative composition of S. faecium lipids were observed. In all isolated fractions (neutral lipids, glycolipids, and phospholipids plus other polar lipids), the major fatty acids were palmitic (C-16:0), palmitoleic (C-16:1), octadecenoic (C-18:1), and cyclopropane (C-19:0). Changes in the fatty acid composition of the different fractions were observed which depended on growth temperature; the most significant one was the decrease of octadecenoic acid and the increase of palmitic acid in glycolipids and polar lipids as the temperature increased. The level of cyclopropane C-19:0 was approximately eightfold lower at 8 degrees C than at the other temperatures tested (20, 30, and 45 degrees C).  相似文献

The green alga Nannochloropsis sp. QII was cultivated in media with sufficient and growth-limiting levels of nitrogen (nitrate). Nitrogen deficiency promoted lipid synthesis yielding cells with lipids comprising 55% of the biomass. The major lipids were triacylglycerols (79%), polar lipids (9%) and hydrocarbons (2.5%). The polar lipids consisted of a broad range of phospholipids, glycolipids and sulfolipids. Other lipids identified were pigments, free fatty acids, saponifiable and unsaponifiable sterol derivatives, various glycerides, a family of alkyl-1, 4-dioxane derivatives and a series of alkyl- and hydroxyalkyl-dimethyl-acetals. Experiments in which 14CO2 was provided at different times in the growth cycle demonstrated that enhanced lipid biosynthesis at low nitrogen levels resulted principally from de novo CO2 fixation.  相似文献

The content and composition of lipids in saliva of healthy caries-free squirrel monkeys were investigated. The dialyzed and lyophilized saliva on extraction with chloroform/methanol yielded 8.0 +/- 0.9 mg of lipids/100 ml of saliva. Following fractionation on silicic acid column, 30.9% of lipids were found in the neutral lipid fraction, 58.8% in the glycolipid fraction, and 10.3% in the phospholipid fraction. The neutral lipids exhibited high content of free fatty acids (58.8%) and triglycerides (23.3%), the glycolipids consisted mainly of neutral and sulfated glyceroglucolipids (95%), while the phospholipids were rich in sphingomyelin and phosphatidylcholine. The results show that squirrel monkey saliva, while displaying lipid content similar to that of caries-susceptible humans, contains 50% less lipids than saliva of periodontal disease-prone marmoset.  相似文献

The composition of hydrocarbons, fatty acids, and of total, polar, and neutral lipids was studied in freshwater Potamogetonaceae grasses collected in two different regions of the Volga river. More than 40 fatty acids and hydrocarbons were separated and identified by chromato-mass spectrometry. The variability of lipid characteristics of plants of the same family is discussed.  相似文献

Lipid class composition of Dunaliella viridis Teodoresco was analysed using thin layer chromatography coupled with flame ionisation detection (TLC/FID technique). D. viridis was cultured under four different photon fluence rates and in darkness, and under two different conditions of CO2 supply (atmospheric and 1%) with and without nitrogen sufficiency. Nine lipid classes were identified and quantified. Total lipids per cell and acetone-mobile polar lipids decreased with light, while the percentage of sterols and triglycerides increased with increasing irradiance. Total phospholipids increase was related with growth rate while hydrocarbons, wax esters and sterol esters accumulated in darkness. There were almost no changes in total lipids per cell because of nitrogen limitation; however, nitrogen limitation led to higher changes in lipid class composition under 1% CO2 than under atmospheric CO2 levels. The main reserve lipid, triglycerides, accumulated in high amounts under 1% CO2 and nitrogen limitation, increasing from 1% to 22% of total lipids. The ratio sterols/acetone-mobile polar lipids could be an index of the 'light status' independently of nitrogen limitation, while the ratio triglycerides/total phospholipids could indicate any physiological stress uncoupling C and N metabolism and affecting the growth rate. Deuterium nuclear magnetic resonance was used to investigate the structure of different lipid fractions isolated from the anaerobic bacteria Clostridium butyricum and Clostridium beijerinckii. The fractions isolated from C. butyricum were (1) phosphatidylethanolamine/plasmenylethanolamine and (2) the glycerol acetal of plasmenylethanolamine, and from C. beijerinckii similar fractions containing principally (1) phosphatidyl-N-monomethylethanolamine, along with its plasmalogen, and (2) the glycerol acetal of this plasmalogen were isolated. The third fraction from both species consisted largely of the acidic lipids phosphatidylglycerol and cardiolipin along with plasmalogen forms of these lipids. Palmitic acid with deuterium labels at C-2, C-3, or C-4 or oleic acid with deuterium labels at C-2 and C-9,10 was added to the growth medium and incorporated to various extents in the lipid fractions. Biochemical analysis showed that palmitic acid and oleic acid were preferentially bound to the sn-2 and sn-1 positions, respectively, of the glycerol backbone when both fatty acids were added to the medium. From the 2H NMR spectra, the hydrocarbon chain ordering near the lipid-water interface could be determined and appeared to be similar for all three lipid fractions. The deuterium quadrupole splitting and order parameter were low at the C-2 segment and increased by almost a factor of 2 at positions C-3 and C-4 for cells fed with deuterated palmitic acid along with unlabeled oleic acid. These results agree with previous findings on pure diacyl lipids in which the sn-2 chain was found to adopt a bent conformation at the carbon segment C-2. Experiments were conducted to determine the effects of whitefly parasitoids on the cuticular lipid composition of the silverleaf whitefly, Bemisia argentifolii Bellows and Perring [=sweetpotato whitefly, Bemisia tabaci (Gennadius), Biotype B] nymphs. The cuticular lipids of B. argentifolii nymphs that had been attacked by parasitic wasps, either Eretmocerus mundus Mercet or Encarsia pergandiella Howard, were characterized by capillary gas chromatography and CGC-mass spectrometry and the results compared with the cuticular lipids of unparasitized nymphs. Previous studies with B. argentifolii nymphs had shown that wax esters were the major components of the cuticular lipids with lesser amounts of hydrocarbons, long-chain aldehydes, and long-chain alcohols. No appreciable changes in lipid composition were observed for the cuticular lipids of E. pergandiella-parasitized nymphs as compared to unparasitized controls. However, the cuticular lipids from nymphs parasitized by E. mundus contained measurable quantities of two additional components in their hydrocarbon fraction. Analyses and comparisons with an authentic standard indicated that the two hydrocarbons were the even-numbered chain length methyl-branched alkanes, 2-methyltriacontane and 2-methyldotriacontane. The occurrences and possible functions of 2-methylalkanes as cuticular lipid components of insects are discussed and specifically, in regard to host recognition, acceptance, and discrimination by parasitoids. Abstract Non-polar and polar lipids were isolated from Methanopyrus kandleri . Non-polar lipids accounted for 50% w/w of total lipids, with a high proportion of 2,3-di- O -geranylgeranyl-sn-glycerol, 2,3-di- O -phytanyl-sn-glycerol and geranylgeraniol. The core lipids prepared by mild acid methanolysis consisted exclusively of 2,3-di- O -phytanyl-sn-glycerol. Two-dimensional TLC showed mostly glycolipids, and minor amounts of aminophospholipids, phosphoglycolipids and phospholipids. The purification yielded three diglycosyl-lipids (50% of total polar lipids), one triglycosyl-lipid (5%) and six glycosyl-lipids with five glycosyl-groups (36%), which consisted of glucose, galactose and mannose. The lipid analysis supports the unique position of Methanopyrus kandleri within the 16S rRNA-based phylogenetic tree.  相似文献

The lipids of six strains of Bacteroides ruminicola were characterized. The nonpolar lipid accounted for 6 to 24% of the total lipid and was composed of diglycerides, triglycerides, and free fatty acids. The phospholipid fraction contained phosphatidylethanolamine, phosphosphingolipids, and trace quantities of phosphatidic acid. In three strains the phosphosphingolipid fraction made up more than half of the total lipid. The fatty acids in the nonpolar, acyl- and phosphosphingolipid consisted of a homologous series of branched and normal chains from 12 to 19 carbons. The long-chain base isolated from the phosphosphingolipids consisted of a homologous series of branched and normal chains from 14 to 24 carbons.  相似文献

The colonial microalga Botryococcus braunii accumulates large quantities of hydrocarbons mainly in the extracellular space; most other oleaginous microalgae store lipids in the cytoplasm. Botryococcus braunii is classified into three principal races (A, B, and L) based on the types of hydrocarbons. Race B has attracted the most attention as an alternative to petroleum by its higher hydrocarbon contents than the other races and its hydrocarbon components, botryococcenes and methylsqualenes, both can be readily converted into biofuels. We studied race B using fluorescence and electron microscopy, and clarify the stage when extracellular hydrocarbon accumulation occurs during the cell cycle, in a correlation with the behavior and structural changes of the lipid bodies and discussed development of the algal colony. New accumulation of lipids on the cell surface occurred after cell division in the basolateral region of daughter cells. While lipid bodies were observed throughout the cell cycle, their size and inclusions were dynamically changing. When cells began dividing, the lipid bodies increased in size and inclusions until the extracellular accumulation of lipids started. Most of the lipids disappeared from the cytoplasm concomitant with the extracellular accumulation, and then reformed. We therefore hypothesize that lipid bodies produced during the growth of B. braunii are related to lipid secretion. New lipids secreted at the cell surface formed layers of oil droplets, to a maximum depth of six layers, and fused to form flattened, continuous sheets. The sheets that combined a pair of daughter cells remained during successive cellular divisions and the colony increased in size with increasing number of cells.  相似文献

The ability to oxidize n-alkanes was studied with various species of fungi belonging to the Cunninghamella genus. These fungi are able to assimilate hydrocarbons and to accumulate up to 1.5 g/litre of biomass. The most active strain was Cunninghamella elegans (-) 1204. The amount of lipids formed, and their composition, depended on the length of the carbon chain of oxidized alkane. The content of fat in the cells increased with the length of the hydrocarbon chain. The following lipid fractions have been detected: phospholipids, monoglycerides, diglycerides, triglycerides, sterols, free fatty acids, sterol esters, and hydrocarbons. The qualitative composition of the fractions depended, to a considerable extent, on the n-alkane utilized. Investigation of the fatty-acid composition of intracellular lipids has shown that fatty acids with an even number of carbon atoms are formed from hydrocarbons with an even number of these atoms, while fatty acids both with an even and odd number of carbon atoms are synthesized from hydrocarbons with an odd number of these atoms. The relative content of the acids with the same number of carbon atoms as that of the alkane being utilized increased with the length of the carbon chain.  相似文献

The lipids of mammalian stratum corneum are known to be important regulators of skin permeability. Since the human stratum corneum displays remarkable regional variations in skin permeability, we assessed the total lipid concentration, the distribution of all major lipid species, and the fatty acid composition in Bligh-Dyer extracts from four skin sites (abdomen, leg, face, and sole) that are known to display widely disparate permeability. Statistically significant differences in lipid weight were found at the four sites that were inversely proportional to their known permeability. In all four sites, among the polar lipids, the stratum corneum contained negligible phospholipids, but substantially more cholesterol sulfate (1-7%) than previously appreciated. As in the stratum corneum from other mammals, the bulk of the lipids consisted of neutral (60-80%) and sphingolipids (15-35%). Of the neutral lipids, free sterols (4- to 5-times greater than esterified sterols), free fatty acids, triglycerides, and highly nonpolar species (n-alkanes and squalene) predominated. n-Alkanes, which were present in greater quantities than previously appreciated, comprised a homologous series of odd- and even-chained compounds ranging from C19 to C34. The sphingolipids comprised over 80% ceramides vs. lesser quantities of glycosphingolipids. In all four sites, the sphingolipids were the major repository of long-chain, saturated fatty acids. The neutral lipid:sphingolipid ratio generally was proportional to the known permeability of each site: higher neutral lipids and lower sphingolipids generally were associated with superior barrier properties. These studies provide: 1) the first detailed, quantitative analysis of human stratum corneum lipids and 2) information about the variability in lipid composition at four skin sites with known differences in permeability. The latter results suggest that variations in neutral lipids, rather than sphingolipids, may underlie local variations in skin permeability.  相似文献

The lipid fraction of the green alga Botryococcus cultured in a batch mode was found to contain polar lipids (more than 50% of the total lipids), di- and triacylglycerides, steroids and their esters, free fatty acids, and hydrocarbons. In senescent culture, the content of polar lipids somewhat decreased and that of triacylglycerides increased by more than four times. The content of hydrocarbons in the algal biomass did not exceed 0.9% and depended little on the culture age. Intracellular lipids contained saturated and unsaturated (mono-, di-, and trienoic) fatty acids. The maximum content of C16:3 and alpha-C18:3 fatty acids (up to 35% of the total fatty acids) was detected in the phase of active growth. The extracellular and intracellular lipids of the alga differed in the proportion of particular lipids and in the fatty acid pattern.  相似文献

The lipid composition of the non-photosynthetic marine diatom, Nitzschia alba, has been quantitatively determined. Triglycerides accounted for 20% of the cell dry weight and 87% of the total lipids. Smaller amounts of 1,2- and 1,3-diglycerides, free sterol (24-methylene cholesterol), hydrocarbons and an unknown component were the remaining neutral lipids detected. Phosphatidylsulfocholine (phosphatidyl S,S-dimethylmercaptoethanol), present in amounts of 0.8% of cell dry weight (35% of total polar lipids), was the major polar lipid component. Other phospholipids were lysophosphatidylsulfocholine, phosphatidylglycerol, phosphatidylinositol and cardiolipin, but both phosphatidylcholine and phosphatidylethanolamine were completely absent. Another novel sulfolipid, deoxyceramide sulfonic acid, as well as the sulfate ester of the free sterol, were also present. Considerable amounts of the four lipids often associated with photosynthetic organisms, mono- and di-galactosyl diglycerides, sulfoquinovosyl diglyceride and phosphatidylglycerol, were identified in N. alba. However, the fatty acid components of the glycosyl diglycerides did not show the high amounts of polyunsaturated acids (18 : 2, 18 : 3) normally found in photosynthesizing organisms. All polar lipids were found to be associated with various cell membrane fractions in N. alba.  相似文献

Summary Candida guilliermondii strain 1 was grown on solar as a sole carbon source for 14 days, and the lipid classes were investigated. The yeast showed high affinity towards hydrocarbons of short chain length, and within 6 days the cellular lipid classes represented 39.69%, 27.50%, 15.35%, 2.23%, 16.20% of hydrocarbons, neutral lipids, free fatty acids, sterols and polar lipids respectively. Undecanoic and hexadecanoic acids were the major fatty acids of the cellular neutral lipids and oleic acid was the major component of the polar lipids.  相似文献

The composition and positional distribution of fatty acids in the polar lipids from 4 strains of Chlorella differing in chilling susceptibility and frost hardiness were analyzed by enzymatic hydrolysis and gas-liquid chromatography. Analysis of the polar lipids from chilling-sensitive, chilling-resistant and chilling-sensitive revertant strains of Chlorella ellipsoidea IAM C-102 showed that the sum of palmitic and trans -3-hexadecenoic acid in phosphatidylglycerol (PG) is about 60% for the sensitive strains and 53% for the resistant strain. The sum of dipalmitoyl and 1-palmitoyl-2-( trans -3-hexadecenoyl) PG as estimated from the positional distribution of their fatty acids, is about 10% in the case of each of the three strains. The contents of unsaturated fatty acids in phosphatidylcholine (PC) and phosphatidylethanolamine (PE) were higher in the resistant than in the sensitive strain. This suggests that unsaturation of fatty acids in not only PG but also PC and PE is involved in chilling sensitivity of Chlorella . On the other hand, lipid changes during the development of frost hardiness of C. ellipsoidea IAM C-27, a frost hardy strain, were examined. The results showed that fatty acids in most lipid classes are unsaturated in the hardening process but their degree of unsaturation is not greatly different from that of the chilling-resistant strain, suggesting that not only unsaturation of fatty acids in lipids but also other factors are necessary for the development of frost hardiness.  相似文献

Isolation and biochemical characterization of goldfish retinal photoreceptor outer segment membranes are described. The lipid fraction is composed primarily of phospholipids (68 mol%) with substantial amounts of neutral lipids (32 mol%). Sterols account for only about 2 wt% of the membranes (about 9 mol% of the total lipids). The phospholipid class composition and fatty acid composition are similar to those of other vertebrate photoreceptor membranes. Two novel findings were the high levels of free fatty acids (21 mol% of the total lipids, primarily palmitic and docosahexaenoic acids) and the presence of relatively significant amounts of a C-32 diacylglycerol molecular species.  相似文献

Cuticular lipid compositions of all life stages of the stored product moth Plodia interpunctella have been determined. Eggs and adults of P. interpunctella have cuticular lipids consisting solely of hydrocarbons. The composition of eggs and adult females is qualitatively nearly identical with ca. 86 hydrocarbons (11 n-alkanes, 39 monomethyl alkanes, 19 dimethyl alkanes, 11 trimethyl alkanes and 6 monoenes) except females lack the 2-methyl alkanes found in eggs. Adult males have a hydrocarbon composition qualitatively nearly identical to females with the exception that they lack the monoenes. Larval and pupal cuticular lipids are dominated by a mixture of ca. 20 previously described 2-acyl-1,3-cyclohexanediones, with only minute amounts of n-alkanes on the larvae and pupae. The 2-acyl-1,3-cyclohexanediones are continuously secreted onto their silk webbing and food particles by the paired mandibular glands found in all larvae. Extracts from dissected mandibular glands have a qualitatively identical composition to larval cuticular extracts. The pupal stage (which does not have mandibular glands) is enclosed in a silk cocoon also coated with 2-acyl-1,3-cyclohexanediones laid down while the wandering stage larvae spin the cocoon. The 2-acyl-1,3-cyclohexanediones have physical properties which closely mimic those of cuticular hydrocarbons, including melting point and boiling point range and hydrophobicity. This is the first report of an insect with a life stage that does not use conventional cuticular lipids for conservation of water.  相似文献
