Testicular sperm motility and methods for the cryopreservation of spermatozoa in the freshwater goby Rhinogobius sp. CB (Cross Band type) were examined. Spermatozoa were almost immotile upon dilution with 300 mOsm kg−1 of NaCl, KCl and mannitol solutions but began to swim in solutions with concentrations <200 mOsm kg−1. The highest percentage and longest duration of motility was obtained in the 0 and 100–200 mOsm kg−1 solutions, respectively. The highest post-thaw motility, c. 50% of motility before cryopreservation, was obtained when spermatozoa were diluted with an extender of 10% methanol and 90% artificial seminal plasma, cooled at −10·0 ± 1·1° C min−1 (mean ± s . e .) to −50° C and plunged into liquid nitrogen. Spermatozoa were cryopreserved in a 50 μl acrylic haematocrit tube to store the small amount of milt. As the cryopreservation method described above was applicable to the endangered Rhinogobius sp. BI (Bonin Island type), it is probable that this method can be used for other species of freshwater gobies.  相似文献

Abstract The apple leaf miner Phyllonorycter ringoniella (Matsumura) (Lepidoptera: Gracillariidae) overwinters as a diapausing pupa. The diapause rate reaches 100% in early October. Diapause intensity decreases gradually from early October and diapause terminates in early February. The fresh body weight of diapausing pupae is 1.6 times that of non-diapausing pupae. The main cryoprotectant in P. ringoniella pupae is trehalose. Three stages are distinguishable as indicated by the correlations between diapause intensity, levels of cold hardiness and the trehalose content: diapause induction occurred in October, diapause development from November to December, and post-diapause quiescence from January to April. During diapause induction, the pupae accumulate low levels of trehalose and do not survive exposure to −15 °C. During diapause development, the pupae gradually accumulate more trehalose and show some ability to survive exposure to −15 °C, but not to −20 °C. During post-diapause quiescence, the pupae accumulate relatively more trehalose and cold hardiness fully develops, but decreases quickly in April. The trehalose content in pupae sampled in December is unaffected by acclimation temperatures in the range 0–30 °C, but decreases in pupae sampled in March after acclimation at temperatures from 5 to 15 °C. These results suggest that overwintering pupae of P. ringoniella have the ability to accumulate trehalose and develop a high level of cold hardiness during diapause development.  相似文献

Turbot Scophthalmus maximus maximum oxygen uptake following feeding and exhaustive exercise increased from 107 mg O2 kg−1 h−1 at 6° C to c . 218 mg O2 kg−1 h−1 at 18° C, then increased slightly from 18 to 22° C to 224 mg O2 kg−1 h−1. Standard oxygen uptake increased exponentially as a function of temperature from 11 mg O2 kg −1 h−1 at 6° C to 66 mg O2 kg−1 h−1 at 22° C. Gradual reduction in oxygen concentration to 87–90% air saturation at 6, 10. 18° C and <80% at 14 and 22° C limited the maximum metabolic rate but, supersaturation (>100% saturation) had little effect. Metabolic scope attained a maximum of 176 mg O2 kg−1 h−1 at 18° C. Interpolation of the results showed that this value changed little between 16 and 20° C. It is suggested that this temperature range is optimal for turbot of c . 500 g. A comparison with a previous study on feeding demand in intensive farming conditions showed a linear relationship between appetite and metabolic scope. It is concluded that the ability of a fish to supply energy (including the energy requirement of digestive metabolism) above a standard level is a limiting factor in the manifestation of its feeding demand.  相似文献

Snakeheads were adapted to fresh water (1 mOsm kg−1), 25% sea water (230 mOsm kg−1), 33% sea water (320 mOsm kg−1) and 40% sea water (380 mOsm kg−1) for 20 days. Exposure to salt water resulted in tissue dehydration, elevations of plasma osmolality, Na+, Mg2+, Cland protein concentrations and stimulation of branchial Na+-K+-ATPase activity. These changes were accompanied by concomitant decline of the hepatosomatic index and liver glycogen concentration. The routine rate of oxygen consumption was increased in snake-heads adapted to 33% sea water. These data were taken to indicate a stressful effect of salinity to the snakehead despite documentation of its ability to penetrate into brackish waters.  相似文献

Routine oxygen consumption rates of bonnethead sharks, Sphyrna tiburo , increased from 141·3±29·7 mg O2 kg−1 h−1 during autumn to 218·6±64·2 mg O2 kg−1 h−1 during spring, and 329·7±38·3 mg O2 kg−1 h−1 during summer. ABSTRACT. Supercooling points (SCP) and low temperature tolerance were determined for larval, pupal and adult stages of Sarcophaga crassipalpis Macquart (Diptera: Sarcophagidae). No stage tolerates tissue-freezing. Ontogenetic changes in SCP profiles are similar for comparable developmental stages of diapause and non-diapause groups. Feeding larvae have SCPs near -7°C which decrease to -11°C in the postfeeding wandering phase of the final larval instar. The lowest SCPs are recorded for pupae at -23°C. The capacity to survive at -17°C varies with age of the diapausing pupae: 10-day-old pupae are less cold tolerant than pupae that have been in diapause for 45–80 days. Although the SCP of non-diapausing pupae is as low as in diapausing pupae, non-diapausing pupae are extremely sensitive to low temperature exposure and do not survive to adult eclosion when exposed to -17°C for as little as 20 min. The use of hexane to break pupal diapause has no effect on SCPs or low temperature tolerance.  相似文献

The spawning induction activity of dissolved and pelleted (D-Ala6, Pro9 N ethylamide) luteinizing hormone-releasing hormone analogue (LHRHa) stored for various periods was assessed in mature female sea bass. The spawning response of mature fish was reduced significantly after injection of dissolved LHRHa (20 μg kg−1) stored for more than 90 days in a refrigerator (4–10°C) or for more than 30 days at room temperature (28–30°C). Similar to fish administered fresh preparations of LHRHa, fish spawned successfully after injection of a solution of LHRHa previously frozen, subjected to alternate freezing and thawing, exposed to sunlight or implanted pelleted LHRHa (50 ng kg−1) stored at room temperature for 30–120 days. Loss of hormone bioactivity after prolonged storage may have been due to bacterial growth in solubilized preparations. Injection or implantation of stored LHRHa did not influence egg production among treated sea bass. These results demonstrated the relatively prolonged shelf life of stored LHRHa.  相似文献

Aims:  To isolate and identify a benefic bacterium, Bacillus subtilis E20, from natto (fermented soybeans), and incorporate it into shrimp feed to promote shrimp growth performance.
Methods and Results:  A protease-producing bacterium, E20, isolated from natto was identified as B. subtilis by an API 50 CHB kit and the 16S rDNA sequence. B. subtilis E20 was able to grow at a broad range of temperatures (10–50°C), pH values (5–10), and NaCl levels (0–9%). The best culture conditions for B. subtilis E20 to produce the protease were 40°C, a pH of 6–8 and 0% NaCl. No shrimp died after being injected with B. subtilis E20 [up to 109 colony-forming units (CFU) per shrimp]. Bacillus subtilis E20 was incorporated in diets at the levels of 0 (control), 106, 107, and 108 CFU kg−1 for shrimp grow-out culture, and results showed that after feeding on B. subtilis E20-containing diets (108 CFU kg−1 of diet), shrimp had excellent growth performance and production compared to the control because protease activities in the digestive tract were improved by B. subtilis E20.
Conclusions:  Bacillus subtilis E20 isolated from natto is a great protease producer and is able to improve shrimp growth performance through increasing the digestibility of food.
Significance and Impact of the Study:  Results suggest that B. subtilis E20 is a potential candidate for use as a probiotic to improve shrimp growth performance, and consequently reduce feed costs.  相似文献

In diapause adults of Graphosoma lineatum overwintering in a field-cage, high chill-tolerance (CT) developed gradually, within 5 months from August to December. In laboratory-acclimation experiments, the diapause state appeared to be an essential pre-condition for successful cold-acclimation and overwintering. First, diapause prevented elevation of the median supercooling point (SCP) by about 5.5°C that accompanies the onset of reproductive activity in non-diapause specimens. Second, diapause allowed subsequent physiological changes resulting in cold-acclimation during a gradual (18-day) decrease of temperature from 25 to 0°C. No, or very modest, cold-acclimation was observed in non-diapause specimens. Decrease of temperature led to a rapid loss of ca. 1/3 of the body water in both non-diapause and diapause specimens. Approximately 0.1 M of trehalose accumulated in tissues of diapause specimens only, and haemolymph osmolality rose from 347 mOsm (at 25°C) to 444 mOsm after an 18-day cold-acclimation and to 764 mOsm during further storage at 0°C for 100 days. Upon transfer of cold-acclimated diapause specimens back to 25°C for one week (de-acclimation), the high CT was lost, the SCP elevated by about 2.5-3°C, and the levels of trehalose, water content and haemolymph osmolality returned to pre-acclimation or non-diapause levels.  相似文献

After stimulation in a hypotonic solution (9.4 mOsm kg−1), inseminated eggs of the chum salmon Oncorhynchus keta initiate cleavages in isotonic salmon Ringer's solution (267.3 mOsm kg−1) containing 3.2 mM Ca2+ ions. Blastomeres of these eggs, however, separate from each other and the enveloping layer is not observed at the blastula stage. An increase in external divalent cations rescues the separation; the concentration of CaCl2 in the external medium should be 25 mM or more to induce close contact of blastomeres and the formation of an enveloping layer in isotonic salt solutions. The effectiveness of Ca2+ ions can be substituted by Mg2+, Sr2+ and Zn2+ ions; the same results are obtained in isotonic MgCl2 and SrCl2 solutions (100 mM) or in isotonic salmon Ringer's solution containing Zn ions (6.2 mM). The close contact of blastomeres and the formation of an enveloping layer are also observed in a low Ca2+ concentration (< 0.1 mM) in a hypotonic salt solution (9.4 mOsm kg−1). The Ca2+ level in the external medium to induce the enveloping layer formation seems to be correlated with the salinity of the incubation medium. It is suggested that adhesion molecules on the surface of blastomeres in the chum salmon eggs are different in properties from those found in sea urchin and other fish species.  相似文献

Three experiments of spawning induction in shi drum, Umbrina cirrosa L., were performed in six different commercial Italian hatcheries from May to August (water temperatures: 19–29 °C; salinity: 21–37 p.p.t.). In the first experiment, 119 females (1–4.7 kg), subdivided into 29 lots, were injected with a single dose (2, 5, 8, 10, 15 and 20  μ g kg−1 body weight) of short-acting gonadotropin- releasing hormone agonist (GnRHa-S), des-Gly10,[D-Ala6]-LH-RH ethylamide. In the other two experiments, 85 females (0.7–5.8 kg), subdivided into 22 and four lots, were treated with one (40 or 80  μ g kg−1) or three doses (40  μ g kg−1) of long-acting GnRHa (GnRHa-L), respectively. GnRHa-S stimulated spawning in 69% of the 29 treated lots; the number of eggs laid reached a maximum of 130 000 and a weighted mean of 29 200 total eggs kg−1. GnRHa-L elicited a spawning response in 95% of the 22 one-dose treated lots; the number of laid eggs was higher than with GnRHa-S, reaching a maximum of 213 100 and a weighted mean of 59 400 total eggs kg−1. The yield of developing embryos in 67% of the single GnRHa-L treatments was higher (sometimes up to three times) than with GnRHa-S. Triple treatments of the four lots of females with GnRHa-L always resulted in spawning responses; the best result corresponded to a number of total laid eggs of 358 900 eggs kg−1 with a yield of 177 300 developing embryos.  相似文献

At 14° C, standard metabolic rate (75·1 mg O2 h−1 kg−1), routine metabolic rate (108.8 mg O2 h−1 kg−1), active metabolic rate ( c . 380 mg O2 h−1 kg−1), critical swimming speed (Ucrit 1·7 BL s−1), heart rate 47 min−1), dorsal aortic pressure (3·2 kPa) and ventilation frequency (63 min−1) for triploid brown trout Salmo trutta were within the ranges reported for diploid brown trout and other salmonids at the same temperature. During prolonged swimming ( c . 80% U crit), cardiac output increased by 2·3-fold due to increases in heart rate (1·8-fold) and stroke volume (1·2-fold). At 18° C, although standard and routine metabolic rates, as well as resting heart rate and ventilation frequency increased significantly, active metabolic rate and certain cardiorespiratory variables during exercise did not differ from those values for fish acclimated to 14° C. As a result, factorial metabolic scope was reduced (2·93-fold at 18° C v . 5·13-fold at 14° C). Therefore, it is concluded that cardiorespiratory performance in triploid brown trout was not unusual at 18° C, but that reduced factorial metabolic scope may be a contributing factor to the mortality observed in triploid brown trout at temperatures near 18° C.  相似文献

The resting metabolic rate (RMR) of juvenile European sea bass Dicentrarchus labrax L. (47·5±1·5 g, 15–18 cm) was 126·2±2·5 mgO2 kg−1 h−1, and temporal patterns of oxygen consumption were not affcted by visual contact or social interaction with conspecifics. The results suggest that a group effect is not present in juvenile D. labrax , thus no selective advantage of shoaling is gained through lowered metabolism in this facultative schooling species.  相似文献

Abstract.  The effect of long-term (seasonal) acclimation and rapid cold hardening is investigated on the cold torpor temperature ( CT min) of adult grain aphids, Sitobion avenae, reared at 20 or 10 °C for more than 6 months before experimentation. Rapid cold hardening is induced by exposing aphids reared at 20 to 0 °C for 3 h and aphids reared at 10 to 0 °C for 30 min (acclimation regimes previously found to induce maximum rapid cold hardening). The effect of cooling aphids from the same rearing regimes from 10 to −10 °C at 1, 0.5 and 0.1 °C min−1 is also investigated. In the 20 °C acclimated population, rapid cold hardening and cooling at 0.1 °C min−1 both produce a significant decrease in CT min from 1.5 ± 0.3 to –0.9 ± 0.3 and –1.3 ± 0.3 °C, respectively. Rapid cold hardening also results in a significant reduction in CT min of the population reared at 10 °C from 0.8 ± 0.1 to –0.9 ± 0.2 °C. However, none of the cooling regimes tested reduces the CT min of the winter-acclimated (10 °C) population. The present study demonstrates that rapid cold-hardening induced during the cooling phase of natural diurnal temperature cycles could lower the movement threshold of S. avenae , allowing insects to move and continue feeding at lower temperatures than would otherwise be possible.  相似文献

The diel rhythms in metabolic rate ( MR ) and activity level ( AL ) were measured for single post-hatching dogfish (weight range, 2.76–10.61 g) at 15° C by the indirect calorimetric method of rate of oxygen consumption ( V O2) and by video-observation respectively, over a period of 72 b. The mean VO 2 increased from 62.0 (s.e. 2.9) mg O2 kg−1 h−1 in the daylight hours to 85.5 (s.e. 3.1) mg O2 kg−1 h−1 during the dark (light regíme, 12 h L: 12 h D). The simultaneous measurement of A L also showed mean night elevation from 0.6 (s.e. 0.2) min h−1 in the light phase to 14.5 (s.e. 1.6) min h−1 during the darkness. Bimodal nocturnal activity (BNA) was exhibited by the post-hatching dogfish within the 12 h dark period, with V O2 increasing from 71.4 (s.e. 2.8) mg O2 kg−1 h−1 before 01.00 hours to 99.5 (s.e. 4.2) mg O2 kg−1 h−1 after 01.00 hours. Similarly, A L also increased from 8.9 (s.e. I.7)min h−1 before 01.00 hours to 21.1 (s.e. 2.8) min h−1 after 01.00 hours. The importance of the results presented to the natural behavioural ecology of the hatching dogfish are discussed.  相似文献

Standard metabolic rate (SMR), active metabolic rate (AMR) and critical oxygen saturation ( Scrit ) were measured in Atlantic cod Gadus morhua at 5, 10 and 15° C. The SMR was 35.5, 57.0 and 78.2 mg O2 kg−1 h−1 and Scrit was 16.5, 23.2 and 30.3%, at 5, 10 and 15° C, respectively. Previously reported SMR for Atlantic cod from arctic waters at 4° C was twice that measured at 5° C in the present study. A possible intraspecific latitudinal difference in the SMR is discussed. The AMR was 146.6, 197.9 and 200.4 mg O2 kg−1 h−1 and the critical swimming speed ( Ucrit ) was 1 6, 1.7 and 1.9 at 5, 10 and 15° C, respectively. The maximum oxygen consumption was found to be associated with exercise, rather than recovery from exercise as previously reported in another Study of Cod metabolism.  相似文献

The Cape golden mole, Chrysochloris asiatica is an insectivore which excavates superficial foraging burrows as it searches for its food. It has a mean (±S.D.) resting metabolic rate (RMR) when newly captured of 1–17±0.17 cm3 O2g-1 h-1 ( n = 14), within the thermoneutral zone (TNZ) of 30–32°C.
The body temperature (Tb) of the mole in the TNZ is low 32.9 ± 0.36 ( n = 14) and remains stable at ambient temperatures (Tas) from 28–32°C. Above 32°C (range 34–37°C), Tb increases albeit slightly to 36 ± 1.75°C ( n = 14). The conductance is high 0.27 ± 006cm3 O2g-1 h-l°C-1 ( n = 46) at the lower limit of thermoneutrality. The mean RMR at 9°C (the lowest Ta tested) was 4.82±11 cm3 O2g-1h-1, which is 4.1 times that of the RMR in the TNZ.
At an ambient temperature of 9°C, three of the golden moles entered a state of torpor where the RMR was reduced from 5.9±0.56 to 10 1.0 ± 0.69cm3O2g-1h-1.  相似文献

The growth properties of juvenile spotted wolffish Anarhichas minor reared at 4, 6, 8 and 12° C, and a group reared under 'temperature steps', (T‐step) i.e . with temperature reduced successively from 12 to 9 and 6° C were investigated. Growth rate and feed efficiency ration was significantly influenced by temperature and fish size. Overall growth rate was highest at 6° C (0·68% day−1) and lowest at 12° C (0·48% day−1), while the 4 and 8° C, and the T‐step groups had similar overall growth rates, i.e . 0·59, 0·62 and 0·51% day−1 respectively. Optimal temperature for growth ( T opt G ) and feed efficiency ratio (Topt FCE) decreased as fish size increased, indicating an ontogenetic reduction in T opt G and T opt FCE. The results suggest a T opt G of juvenile spotted wolffish in the size range 135–380 g, dropping from 7·9° C for 130–135 g to 6·6° C for 360–380 g juveniles. The T opt FCE dropped from 7·4° C for 120–150 g to 6·5° C for 300–380 g juveniles. A wider parabolic regression curve between growth, feed efficiency ratio and temperature as fish size increased, may indicate increased temperature tolerance with size. Individual growth rates varied greatly at all time periods within the experimental temperatures, but at the same time significant size rank correlations were maintained and this may indicate stable size hierarchies in juvenile spotted wolffish.  相似文献
