细胞质雄性不育水稻幼穗和花粉发育期的蛋白酶活性变化   总被引:1,自引:0,他引:1  
水稻细胞质雄性不育系珍汕97A,在花粉母细胞形成期以后可溶性蛋白含量迅速降低。单核期仅为花粉母细胞形成期的56%,二核期和三核期分别为花粉母细胞形成期的36.4%和30.3%。保持系珍汕97B虽在花粉母细胞形成期后可溶性蛋白含量降低,但在二核期则有所增高,表现为鞍形变化过程。在花粉母细胞形成期后,珍汕97A的酪蛋白水解酶活性增高,单核期为前一时期的1倍,而珍汕97B的酶活性增幅较小。第一次枝梗分化期珍汕97A的内肽酶活性较珍汕97B高,而雌雄蕊形成期两者的酶活皆降低。整个发育期中珍汕97A的内肽酶活性较珍汕97B高15%至55.5%。珍汕97A在幼穗第一次枝梗分化期时氨肽酶活性较珍汕97B高。第二次枝梗分化期后,不育系和保持系的氨肽酶活性均下降。但花粉母细胞形成期后,珍汕97A的氨肽酶活性迅速增高,三核期达最高水平,这可能意味着此时大量蛋白质被水解。蛋白酶类抑制剂的实验表明,在雌雄蕊形成期后不育系和保持系含有半胱氨酸型氨肽酶和约10%含金属型氨肽酶;而在三核期,保持系的氨肽酶则主要为半胱氨酸型。不育系三核期含有的BAPA内肽酶包括半胱氨酸型、丝氨酸型,以及部分含金属酶类;而保持系三核期的内肽酶仅为丝氨酸型。不育系肽酶类型的变化,可能反映酶基因编码的去抑制。  相似文献

利用RAPD技术,从水稻野败型细胞质雄性不育系珍汕97A的保持系珍汕97B中得到一个特异的扩增片段PWP-13。该片段全长808bp,1-142区段为正常的cob基因片段;143-372、406-707区段与一个报告的嵌合cob基因的同源性分别为98%、100%,但由于碱基缺失,所推测的氨基酸序列差异较大;373-405区段为PWP-13所特有的重复序列,708-808区段为未知序列。序列内含有3组长度分别为9、31、27bp的小重复序列。  相似文献

以珍汕 97A为材料 ,通过不同剂量的 6 0 Co- γ射线和 Na N3药剂对珍汕 97A进行复合处理 ,分析研究它们的诱变效应。结果表明 ,随着辐射剂量的增加 ,M1群体中的育性恢复突变率有所下降 ;而分离出不育株的 M2 株系占育性恢复突变 M1株系的比例逐渐增加 ;与珍汕97A相比 ,M2 分离出的不育株有的仅发生农艺性状的变异 ,有的则在不育性的表达和农艺性状上均发生变异 ;与珍汕 97B相比 ,育性恢复的 M2 群体部分农艺性状发生了多种变异 ,并在M2 群体中出现了无花粉型不育株及弯秆突变体。此外 ,还讨论了诱变技术对不育系的改良与创新的有关问题。  相似文献

水稻(Oryza sativaL.)雄性不育系珍汕97A,保持系珍汕97B和恢复系测64三系小孢子发生过程的研究表明;恢复系测64小孢子母细胞细胞质浓,有明显的微管荧光围绕着细胞核。小孢子母细胞经两次减数分裂形成四分体。四分体和小孢子的微管从细胞核表面向胞质周缘延伸,形成放射性排列格局,花粉发育正常。细胞质中有少量点状微管荧光,保持系珍汕97B小孢子发生过程的细胞形态和微管结构与恢复系测64相似。但细胞质中的点状微管荧光多一些。雄性不育系珍汕97A小孢子发生早期,小孢子母细胞内出现液泡,核中染色质凝集,微管荧光很弱,没有清晰的微管丝结构。细胞质中有许多点状微管荧光等不正常现象。小孢子母细胞经过减数分裂形成的四分体也没有清晰的丝状微管结构。随后,所有的小孢子迅速败育,雄性不育系珍汕97A在小孢子母细胞发生的很早时期,微管结构就明显不正常。  相似文献

水稻雄性不育系珍汕97A是我国应用最大,使用最广泛的不育系,利用抽穗期基因型明确的秋光(e1e1e2e2e3e3se-1^eSe-1^e),越光(E1E1E2E2e3e3Se-1^eSe-1^e),日本晴(E1E1e2e2e3e3Se-1Se-1)和日光(E1E1E2E2e3e3Se-1Se-1)作测验品种,分析了水稻珍汕97B的抽穗期基因型,结果表明,珍汕97B的抽穗期感光基因型为:e1e1e2e2E3E3Se-1Se-1,同时还存在1对隐性感光抑制基因i-Se-1,进一步用QTL近等基因系NIL(Hd1),HIL(Hd2),NIL(Hd3),NIL(Hd5)和NIL(Hd6)进行的实验也验证了珍汕]97B 在1个显性的主效感光基因Se-1,以及其他感光修饰基因,如E3,Hd3(En-Se-1),Hd5和Hd6的基因的作用。因此,推测珍汕97A带有主效感光基因是其配制的灿型杂交稻抽穗期超亲表现的内因。  相似文献

本实验对水稻雄性不育系珍汕97A及其保持系在幼苗(2叶或3叶)和孕穗期(抽穗前8天)分别进行一次热激处理,并比较了其同工酶组成、花粉育性以及自交结实性的变化。同工酶分析结果表明,热激处理能使不育系与保持系花药的POX和Est的表达发生不同的改变,导致两系在这两种同工酶组成上的原有差异缩小;育性观察结果显示,在正常温度条件下,热激处理对珍汕97A的育性表达几乎无影响,而在有高温作用(穗分化发育阶段的日平均温度高于30℃)的条件下,3叶期幼苗往后的热激处理似乎能不同程度地提高珍汕97A的育性稳定性。简要讨论了热激处理与不育系育性转变间的关系。  相似文献

红莲型CMS是在我国杂交水稻生产中被广泛利用的雄性不育细胞质之一。为了同时定位红莲型CMS育性恢复主效和微效QTL,利用红莲型CMS不育系粤泰A(YTA)与“Lemont/特青”RIL群体测交,结合1张含有198个DNA分子标记的高密度遗传图谱,对测交F1群体的小穗育性和花粉育性进行复合区间作图。在对YTA的育性恢复性方面,该。RIL群体的2个亲本之间具有明显差异,特青的恢复性较强,其测交F1的小穗育性和花粉育性分别为72%和51%;而Lemont测交F1的小穗育性和花粉育性分别为32%和9%。复合区间作图定位到4个育性恢复QTL,分别位于水稻第1、2和10号染色体上,单个QTL的贡献率在5%~24%之间。其中,除1个QTL的增效基因来源于Lemont外,其余3个QTL的增效基因均来源于特青。效应最大的QTL为qRF-10-1,该QTL位于10号染色体RM258-C16标记区间,对小穗育性表型变异的贡献率为24%,对花粉育性的贡献率为17%,且该QTL被检测到的LOD值显著较高,因此是1个主效QTL,其增效基因来源于特青。除了主效QTLqRF-10-1外,其它3个QTL对性状的贡献率均在10%以下(5%~8%)。由此表明,该RIL群体对红莲型CMS的育性恢复由1个主效QTL控制,并受其它几个微效QTL的影响。该QTL定位结果与小穗育性在测交F1群体中呈连续的双峰分布的结果相一致。与主效QTL qRF-10-1紧密连锁的SSR标记为RM258,该主效QTL可作为分子标记辅助育种的操作目标之一,用于杂交稻分子育种中培育红莲型CMS的强恢复系。  相似文献

以籼稻恢复系圭630与粳型广亲和品种02428的F1代花药培养,获得81个双单倍体(DH),构建了有233个RFLP标记的分子图谱。用籼稻野败型不育系珍汕97A测定各DH系的恢复性,并将恢复性作为数量性状进行QTL的区间作图分析,鉴别出8个基因座位,其中有2个基因座位,Rfi-3和尾Rfi-4,单个QTL的基因贡献值分别是49.6%和35.4%,对育性恢复起主要作用,定为主效基因座位,位于第三和四染色体上,其它6个基因座位对育性恢复亦有一定的影响。表明野败型雄性不育恢复性是受主效基因和微效基因共同控制的性状。  相似文献

The improvement of grain quality, such as protein content (PC) and amino acid composition, has been a major concern of rice breeders. We constructed a population of 190 recombinant inbred lines (RILs) from a cross between Zhenshan 97 and Nanyangzhan to map the quantitative trait locus or loci (QTL) for amino acid content (AAC) as characterized by each of the AACs, total essential AAC, and all AAC. Using the data collected from milled rice in 2002 and 2004, we identified 18 chromosomal regions for 19 components of AAC. For 13 of all the loci, the Zhenshan 97 allele increased the trait values. Most QTL were co-localized, forming ten QTL clusters in 2002 and six in 2004. The QTL clusters varied in both effects and locations, and the mean values of variation explained by individual QTL in the clusters ranged from 4.3% to 28.82%. A relatively strong QTL cluster, consisting of up to 19 individual QTL, was found at the bottom of chromosome 1. The major QTL clusters identified for two different years were coincident. A wide coincidence was found between the QTL we detected and the loci involved in amino acid metabolism pathways, including N assimilation and transfer, and amino acid or protein biosynthesis. The results will be useful for candidate gene identification and marker-assisted favorable allele transfer in rice breeding programs.  相似文献

QTL analysis of fertility restoration in cytoplasmic male sterile pepper   总被引:8,自引:0,他引:8  
Fertility restoration of Petersons cytoplasmic male-sterility in pepper (Capsicum annuum L.) is quantitative and environment-dependent. QTL analysis of fertility restoration was performed based on the test-cross progeny of 77013A (a strict cytoplasmic-genetic male sterile line) and a doubled haploid population of 114 lines obtained from an F1 hybrid between Yolo wonder (a sterility maintainer line) and Perennial (a fertility-restorer line). The fertility of the test-crossed lines was assessed under greenhouse and open field conditions using three criteria related to pollen or seed production. One major QTL for fertility restoration was mapped to chromosome P6. It was significant in all the environments and for all the traits, accounting for 20–69% of the phenotypic variation, depending on the trait. Four additional minor QTLs were also detected on chromosomes P5, P2, and linkage groups PY3 and PY1, accounting for 7–17% of the phenotypic variation. Most of the alleles increasing fertility originated from the restorer parent, except for two alleles at minor QTLs. Phenotypic analysis and genetic dissection indicated that breeding pepper for complete sterility of female lines and high hybrid fertility requires complex combinations of alleles from both parents and a strict control of the environment.  相似文献

A better understanding of the genetics of seedling characteristics in rice could be helpful in improving rice varieties. Zhenshan 97 and Minghui 63, the parents of Shanyou 63, an elite hybrid developed during the last decade in China, vary greatly with respect to their physiological and morphological traits at the seedling growth stage. In this study, we used a population of 240 recombinant inbred lines derived from a cross between Zhenshan 97 and Minghui 63 to identify quantitative trait loci (QTL) for seedling characteristics. All plant material was grown in hydroponic culture. Data for the following characters were collected at 30 days and 40 days post-sowing: plant height, shoot dry matter weight (SDW), maximum root length, root dry weight (RDW), total dry weight , and root-shoot ratio (the ratio of SDW to RDW). Analysis using composite interval mapping detected 16 QTL for the six traits in 30-day-old seedlings. Of these 16 QTL, Minghui 63 alleles increased trait values at only two of them. The QTL in the vicinity of R3166 on chromosome 5 simultaneously influenced PH, SDW, MRL, RDW, and TDW in the same direction. Twenty QTL were detected for the same traits in the 40-day-old seedlings. However, at this stage Minghui 63 alleles increased trait values at eight QTL. The QTL linked to R3166 also affected PH, SDW, MRL, RDW, and TDW. Only four QTL were common to the two stages. These results clearly indicate that different genes (QTL) control the same traits during different time intervals. Zhenshan 97 alleles had positive effects during the first 30 days of seedling growth, but thereafter the positive effects of Minghui 63 alleles on seedling growth gradually became more pronounced.  相似文献

A set of rice (Oryza sativa L.) recombinant inbred lines from a cross between Zhenshan 97 (indica) and HR5 (indica) was planted for four different growing seasons in two locations at three nitrogen (N) fertilization levels (N300, 300 kg urea/ha; N150, 150 kg urea/ha; and N0, 0 kg urea/ha). Grain yield and its components were evaluated, including grain yield per plant (GYPP), panicle number per plant (PNPP), grain number per panicle (GNPP), filled grains per panicle (FGPP), spikelet fertility percentage (SFP) and 100-grain weight (HGW). Correlation and path analysis indicated that SFP had the greatest contribution to GYPP at the N300 and N150 levels, but FGPP contributed the most to GYPP at the N0 level. Quantitative trait loci (QTL) were mapped based on a mixed linear model; genetic components (main effects, epistatic effects and QTL-by-environment interactions) were estimated separately. Six to 15 QTL with main effects were detected for each trait except SFP. Clusters of main-effect QTL associated with PNPP, GNPP, SFP and HGW were observed in regions on chromosomes 1, 2, 3, 5, 7 and 10. The main-effect QTL (qGYPP-4b and qGNPP-12) were only detected at the N0 level and explained 10.9 and 10.2% of the total phenotypic variation, respectively. A total of 33 digenic interactions among grain yield and its components were also identified. The identification of genomic regions associated with yield and its components at different nitrogen levels will be useful in marker-assisted selection for improving the nitrogen use efficiency of rice.  相似文献

The cytoplasmic male sterility (CMS) of wild-abortive (WA) cytoplasm has been widely used for breeding hybrid rice. Two restorer genes for the CMS have been found by traditional genetic analysis. To tag the restorer genes we used a set of near-isogenic lines (NILs) of Zhenshan 97 carrying different genotypes for fertility restoration from IR24, to perform RAPD analysis. From the survey of 720 random primers, six RAPD markers were identified to be associated with Rf-3. Three of these OPK05-800, OPU10-1100 and OPW01-350, were mapped on chromosome 1. Two populations from the crosses between Zhenshan 97 A and a near-isogenic restorer line ZSR21 and between Zhenshan 97 A and IR24 were used for mapping Rf-3. The three RAPD markers and three RFLP markers, RG532, RG140 and RG458, were found to be closely linked to Rf-3 in the two populations. The same location of Rf-3 was also found in a population from the cross of IR58025 A//IR36/IR58025 B. At the RG532 locus, different alleles were found between two CMS lines, Zhenshan 97 A and IR58025 A, and between two restorer lines, IR24 and IR36. 分别在两年收集珍汕97/明恢63的重组自交系群体的表现数据,运用混合线性模型的QTL定位方法,联合分析穗部5个性状的QTLs7及QTL与环境互作关系。每穗颖花数、每穗实粒数、结实率、穗长和穗着密度分别检测到10、3、6、8和7个QTLs分别解释各性状变异的29.13%、19.2%、29.46%、26.39%和35.76%。对于同一性状,高值亲本和低值亲本中均存在增效和减效QTL。相关性状QTL的位置表现相同或相似,高值亲本和低值亲本中均存在增效和减效QTL。相关性状QTL的位置表现相同或相似,成族分布。1个穗长QTL,2个每穗颖花数QTL3,3个结实率QTLs表现与环境显著互作,QTL与环境互作效应的贡献率比相应的QTL贡献率略大。遗传力稍高的每穗实粒数和穗着粒密度的DQTL与环境不互作。  相似文献
