以牛粪污水为研究对象, 通过高通量测序技术研究三种微生物除臭菌剂在牛粪污水除臭过程中对细菌多样性的影响, 并对部分菌群功能进行预测分析。对分别添加布氏乳杆菌(Lactobacillus buchneri )CC1、甲基营养型芽孢杆菌(Bacillus methylotrophicus )Q3、苍白杆菌(Ochrobactrum )FSB进行除臭处理的牛粪污水细菌多样性及群落结构进行分析对比。结果表明: (1)分别添加甲基营养型芽孢杆菌Q3和苍白杆菌FSB的处理可使粪水的微生物群落丰富度和多样性先升后降, 而添加布氏乳杆菌 CC1的处理微生物群落丰富度和多样持续升高。(2)在属水平, 不同处理的组间差异性较大, 微生物的丰富度、多样性、相似性、优势菌群均有不同。与除臭和降解功能相关的脱硫微菌属(Desulfomicrobium)、纤维素分解菌属(Ruminiclostridium)、Prolixibacter菌属和Desulfomicrobium菌属在分别添加菌剂处理的F、C、Q组与对照组K,CK差异明显并随着发酵时间的变化而变化, 其中随着处理时间的延长, 纤维素分解菌属(Ruminiclostridium)的丰度增加明显, 菌剂处理组增幅明显大于对照组, 发酵15天时在处理Q中, 其丰度由0增加到13.01%, F处理和C处理分别由0增加到4.33%, 而对照组仅由0增加到1.69%; 具有除NH3功能的菌属Prolixibacter的丰度在所有处理中发酵3天时丰度最大, 15天时为0; 与产NH3功能相关的菌属Paracoccus随着发酵进程逐渐降低, 在15天时完全消失; 产生H2S的菌属Desulfomicrobium也随着发酵进程逐渐降低, 由对照的5.22%下降至3%以下, 并且添加菌剂组下降幅度大于不加菌剂组; 病原菌Pesudomonas和 Acholeplasma的数量呈现先升高后降低的趋势, 由0天的1.54%和2.8%降至15天的1.38%和1.87%。(3)在门水平含量最多的是变形菌门(Proteobacteria), 接下来依次为厚壁菌门(Firmicutes)、拟杆菌门(Bacteroidetes)、放线菌门(Actinobacteria)、互养菌门(Synergistetes)、热脱硫杆菌门(Spirochaetae)、疣微菌门(Verrucomicrobia)和纤维杆菌门(Fibrobacteres)。其中, 变细菌门在K2处理中丰度最高, 达43.6%, 在C1处理中最低, 为27.5%, 并且在所有处理中表现出发酵15天的丰富度大于3天的变化趋势。厚壁菌门在处理F、Q、C中也表现出发酵15天的含量大于3天的变化趋势。拟杆菌门和放线菌门在所有处理中都表现出随着发酵时间的延长而含量降低的现象, 并且所有处理的含量均低于空白对照; 在Q和F处理中, 与纤维素降解相关的纤维杆菌门的含量明显升高。(4)功能菌群总体变化趋势为: 分别添加三种菌剂组中纤维素分解菌群、有机物降解菌群、固氮菌群、H2S/NH3去除菌群种类和丰度明显高于对照组, 而H2S/NH3产生菌低于对照组, 病原菌和土著菌株经过发酵后含量明显降低或消失。  相似文献

【目的】提出一种合成微生物组的理性构建策略,用于构建郫县豆瓣蚕豆醅初始发酵的微生物组合菌剂。【方法】采取自下而上的合成微生物组理性构建策略,以相对丰度、频率和特征向量中心度作为核心微生物属的选择指标,分析确定蚕豆醅发酵核心微生物;设计模拟原位体系的全合成培养基,并利用该培养基快速、稳定地检测核心微生物包括产香性能在内的发酵特征。基于核心微生物的产香互补性能进行双菌组合发酵实验,结合核心微生物之间的生长相互作用,设计三菌组合发酵菌剂并验证其发酵性能。【结果】本研究确定并分离了郫县豆瓣蚕豆醅发酵过程中的 9种核心微生物。检测核心微生物产生的挥发性风味化合物,发现酵母菌类、乳酸菌类和其他类微生物之间存在产香互补关系。然后,结合微生物间的生长抑制关系设计了由乳酸片球菌、肉葡萄球菌及异变假丝酵母组成的三菌组合菌剂。与企业原位发酵样品相比,三菌组合菌剂产生的挥发性化合物数量达到原位样品的63.1%,化合物种类结构较为相似。与原位样品相比,组合菌剂样品氨基酸态氮浓度提升了21.8%。【结论】本研究提出了一种自下而上的合成微生物组理性构建策略,基于此策略设计了郫县豆瓣蚕豆醅发酵组合菌剂。使用该组合菌剂作为起始发酵剂发酵的郫县豆瓣蚕豆醅具有良好的风味谱和优异的氨基酸态氮水平。本研究在合成微生物组构建与发酵食品工艺改造方面具有较大的科学与应用价值。  相似文献

利用PCR-DGGE方法研究了接种外源微生物菌剂对鲜猪粪高温好氧堆肥过程中细菌群落多样性的影响.结果表明:接种外源微生物菌剂可以促进堆肥的顺利进行,比不接种处理的高温期提前2 d.DGGE图谱分析表明,堆肥中优势细菌群落组成发生了明显的更迭现象,不同堆肥时期细菌群落的Shannon-Wiener指数呈显著差异.目的条带克隆测序结果表明,整个堆肥过程Clostridium stercorarium subsp. thermolacticum sp.一直是优势菌属,不经培养细菌、Bacillus coagulans sp.、Clostridium thermocellum sp.在接种外源微生物菌剂处理的第10、16天成为优势菌属,不经培养的Firmicutes sp.和不经培养的 delta proteobacterium分别在未接种外源微生物菌剂处理堆肥发酵的第5天和第16天成为优势菌属.非优势菌属Ureibacillus thermosphaericus、不经培养的Silvimonas sp.出现在堆肥腐熟后期,不经培养的土壤细菌主要出现在堆肥初期和高温初期.UPGMC聚类分析表明,接种外源微生物菌剂明显影响了堆肥不同时期的细菌群落结构组成.堆肥化过程中细菌DGGE图谱主成分分析表明,细菌群落变化主要受外源接种微生物菌剂的影响.  相似文献

【背景】传统的酸马奶是由鲜马奶自然发酵而成,鲜马奶中的微生物在很大程度上决定了酸马奶的品质。然而,鲜马奶中的微生物组成却鲜有报道。【目的】解析鲜马奶中细菌多样性,比较分析鲜马奶和酸马奶中微生物的差异,探究马奶发酵过程中微生物的变化。【方法】采用PacBio SMRT测序技术,对6份采集自内蒙古锡林浩特牧区的鲜马奶中细菌16SrRNA基因序列全长进行测序,在种水平解析鲜马奶样品的细菌组成,并与前期同一地区的酸马奶菌群组成作对比。【结果】鲜马奶中包含大量的乳酸菌,主要为乳酸乳球菌(12.43%)、格氏乳球菌(3.53%)、肠膜明串珠菌(1.77%)等。此外还有部分板崎肠杆菌等条件致病菌的存在。马奶发酵前后菌群变化较大,鲜马奶中微生物多样性显著高于酸马奶,其中乳酸乳球菌和瑞士乳杆菌为鲜马奶和酸马奶的共有细菌。鲜马奶中的肠膜明串珠菌与乳酸乳球菌、格氏乳球菌呈正相关关系,与嗜水气单胞菌等有害菌为负相关关系,表明在马奶发酵过程中乳酸菌可能会抑制有害菌的生长。【结论】在马奶的发酵过程中,乳酸菌的大量生长抑制了马奶中有害菌生长,降低了马奶中微生物的多样性,提升了酸马奶的品质。为酸马奶的工业化生产提供了参考。  相似文献

【目的】传统发酵乳制品是一类未经任何处理自然发酵而成的,其微生态环境未遭破坏,从而乳酸菌的生物学特性和基因多样性得到了很好的保留,具有开发和利用价值。自然发酵酸驼乳常用来治疗多种疾病且效果良好,与其中丰富的乳酸菌资源有着密不可分的联系。然而,目前有关自然发酵酸驼乳微生物菌群及多样性相关研究甚少。因此进一步挖掘内蒙古地区双峰驼自然发酵酸驼乳微生物群落结构和多样性是至关重要的。【方法】本研究采用IlluminaMiseq测序技术,测定了苏尼特和阿拉善双峰驼的自然发酵酸驼乳中微生物16S rRNA V3–V4区序列,并对群落结构和多样性进行了比较分析。【结果】多样性分析表明,苏尼特双峰驼酸驼乳中微生物群落丰富度和种群差异性比阿拉善双峰驼酸驼乳大,细菌多样性也高。在门水平上,苏尼特和阿拉善双峰驼酸驼乳中的菌群均以厚壁菌门(Firmicutes)和变形菌门(Proteobacteria)为主。在属水平上,苏尼特双峰驼酸驼乳主要以乳杆菌属(Lactobacillus)和乳球菌属(Lactococcus)为优势菌群,阿拉善双峰驼酸驼乳以乳杆菌属(Lactobacillus)和醋酸杆菌属(Acetobacter)为优势菌属。此外,肠杆菌属(Enterobacter)、拉乌尔菌属(Raoultella)和明串珠菌属(Leuconostoc)等的含有食源性致病菌和环境污染菌的菌属被检出。综上所述,不同地区不同品种酸驼乳的乳酸菌种类及优势菌群有较大差异,存在显著的地理差异。【结论】通过本研究,不仅对苏尼特和阿拉善双峰驼自然发酵酸驼乳乳酸菌的组成和种类有了明确的认知,为评估发酵酸驼乳微生物群落对消费者身体健康的影响提供了数据基础的同时为今后筛选优势菌群和挖掘新型益生菌奠定基础。  相似文献

Composts prepared from a variety of feedstocks were tested for their ability to suppress seedling and root diseases of creeping bentgrass caused by Pythium graminicola. Among the most suppressive materials in laboratory experiments were different batches of a brewery sludge compost and a biosolids compost from Endicott, N.Y. Batches of these composts that were initially not suppressive to Pythium damping-off became more suppressive with increasing compost age. Leaf, yard waste, food, and spent mushroom composts as well as certain biosolids, cow manure, chicken-cow manure, and leaf-chicken manure composts were not suppressive to Pythium damping-off. In some cases, turkey litter, chicken manure, chicken-leaf, and food waste composts were inhibitory to creeping bentgrass seed germination in laboratory experiments. Microbial populations varied among all of the composts tested. Bacterial populations were high in all composts except the turkey litter compost, in which populations were 1,000- to 10,000-fold lower than in the other composts tested. Among the highest populations of heterotrophic fungi and antibiotic-producing actinomycetes were those found in all batches of the brewery sludge compost, whereas the lowest populations were found in turkey litter, chicken manure, and food waste composts. Heat treatment of suppressive composts reduced populations of bacteria, fungi, and actinomycetes in all composts tested. Disease suppressiveness was also reduced or eliminated in heated composts. Amending heated composts with small amounts of nonheated compost restored suppressive properties and partially restored microbial populations to wild-type levels. A strong negative relationship between compost microbial activity (as measured by the hydrolysis of fluorescein diacetate) and Pythium damping-off severity was observed. When composts were applied to creeping bentgrass in field experiments, a significant level of suppressiveness was evident with some composts when disease pressure was high (i.e., disease ratings high in uninoculated plots). A 1991 batch of turkey litter compost and the 1990 batch of Endicott biosolids were consistently suppressive to foliar symptoms of Pythium root rot on creeping bentgrass. This study indicates that suppression of Pythium diseases of creeping bentgrass in batches of brewery sludge and Endicott biosolids composts, and possibly in other suppressive composts examined in less detail in this study, is related directly to the microbial activities in the composts. On the other hand, the mechanisms of Pythium suppression in turkey litter and perhaps other poultry-based composts is not related directly to the compost microbial activity. Although turkey litter showed a lack of suppressiveness in laboratory bioassays and low microbial populations and activity, it resulted in a significant and consistent level of suppressiveness in field experiments. Therefore, the microbiological properties of Pythium-suppressive composts may differ substantially, and measurements of microbial populations and activity may not be predictive of the level of disease suppression in all composts.  相似文献

Compost prepared from wheat straw and cattle/chicken mature was inoculated with the lignocellulolytic fungus, Penicillium expansum. Compared to uninoculated compost, the inoculated compost exhibited a 150% higher germination index, more than 1.2 g kg(-1)-dw of changes in NH(4)(+)-N concentrations, a ca. 12.0% higher humus content and a lignocellulose degradation that proceeded 57.5% faster. Culture-based determinations of microbial populations demonstrated that aerobic heterotrophic bacteria and fungi were about 1-2 orders of magnitude higher in inoculated than in uninoculated compost. The number of ammonifying, ammonium-oxidizing, nitrite-oxidizing, denitrifying bacteria and cellulose-decomposing bacteria was 6.1-9.0 log(10) CFU g(-1)-dw, 1.2-4.3 log(10) MPN g(-1)-dw, 3.5-6.8 log(10) MPN g(-1)-dw, 3.58-4.34 log(10) MPN g(-1)-dw, 1.4-3.8 log(10)MPN g(-1)-dw, and 4.2-8.8 log(10) CFU g(-1)-dw higher in the compost inoculated with P. expansum.  相似文献

Twenty-two species of thermophilic fungi were isolated from mushroom compost. Scytalidium thermophilum was present in the compost ingredients, fresh straw, horse droppings, and drainage from compost and dominated the fungal biota of compost after preparation. Of 34 species of thermophilic fungi tested, 9 promoted mycelial growth of Agaricus bisporus on sterilized compost: Chaetomium thermophilum, an unidentified Chaetomium sp., Malbranchea sulfurea, Myriococcum thermophilum, S. thermophilum, Stilbella thermophila, Thielavia terrestris, and two unidentified basidiomycetes. These species will be considered for future experiments on inoculation and more controlled preparation of compost.  相似文献

在浙江省安吉县采集了相邻的天然灌木林和板栗林土壤,分析土壤水溶性碳(WSOC)、微生物生物量碳(MBC)、易氧化碳(ROC)、水溶性有机氮(WSON)和微生物生物量氮(MBN),并利用核磁共振方法分析土壤总有机碳的波谱特征,研究天然灌木林改造成板栗林对土壤碳库和氮库的影响.结果表明: 天然灌木林改造成板栗林后,土壤中的碱解氮、有效磷和速效钾显著增加,而WSOC、MBC、ROC、WSON和MBN显著下降.天然灌木林和板栗林土壤有机碳以烷基碳和烷氧碳为主.天然灌木林改造成板栗林后,土壤有机碳中的烷氧碳和羰基碳比例显著下降,而烷基碳和芳香碳比例以及A/O-A值和芳香度均显著增加.天然灌木林改造成板栗林并长期集约经营后,土壤活性碳库和氮库含量均显著下降,而土壤碳库的稳定性显著增加.  相似文献

土壤微生物多样性能反应土壤的肥力,不同的施肥措施对土壤微生物的种群和功能多样性也会产生重要的影响。以山东德州连续两年小麦季和玉米季收获后土壤为研究对象,利用Biolog技术研究了6种不同施肥处理对土壤微生物群落功能多样性的影响。结果表明:其中各个施肥处理的平均颜色变化率(average well color development,AWCD)差异显著,常规氮磷钾肥+全量秸秆还田+秸秆腐熟剂(FS)处理代谢活性最高;物种丰富度指数(H)和均匀度指数(E)也表明各施肥方式均能够维持微生物种群的多样性,其中FS和30%猪粪+70%常规氮磷钾肥(OF)处理物种丰富度指数(H)和均匀度指数(E)最高;PCA及RDA分析显示,OF和FS处理微生物功能多样性相似,且其微生物功能多样性与有机质(Soil organic matter,SOM)、全氮(Total N,TN)、速效磷(Available P,AP)和速效钾(Available K,AK)密切相关。猪粪堆肥有机无机复合肥3 600 kg/hm2(OI2)处理与猪粪堆肥有机无机复合肥1 800 kg/hm2(OI1)处理相似,其功能多样性比常规施肥(CF)处理稍高。综上所述,OF处理和FS处理的土壤微生物群落功能多样性程度高于其他处理,说明秸秆还田+秸秆腐熟剂和有机肥部分替代氮磷钾肥能够显著提高土壤微生物功能多样性,有利于保护土壤微生态。  相似文献

The relationship between compost amendment, plant biomass produced, and bacterial root colonization as measured by fluorescence in situ hybridization was examined following plant growth in mine tailings. Mine tailings can remain devoid of vegetation for decades after deposition due to a combination of factors that include heavy metal toxicity, low pH, poor substrate structure and water-holding capacity, and a severely impacted heterotrophic microbial community. Research has shown that plant establishment, a desired remedial objective to reduce eolian and water erosion of such tailings, is enhanced by organic matter amendment and is correlated with significant increases in rhizosphere populations of neutrophilic heterotrophic bacteria. Results show that for the acidic metalliferous tailings tested in this study, compost amendment was associated with significantly increased bacterial colonization of roots and increased production of plant biomass. In contrast, for a Vinton control soil, increased compost had no effect on root colonization and resulted only in increased plant biomass at high levels of compost amendment. These data suggest that the positive association between compost amendment and root colonization is important in the stressed mine tailings environment where root colonization may enhance both microbial and plant survival and growth.  相似文献

Ammonia suppressants are applied to chicken litter to decrease ammonia levels. And mushroom (Agaricus bisporus) producers use poultry litter to increase the nitrogen in the compost. To determine the influence of ammonia suppressants used in poultry litter on compost preparation and mushroom production, four mushroom crops were cultivated from compost prepared using litter treated with PLT, Barn Fresh and Impact-P at 25.22 kg/100 m2, 40 kg/100 m2, and 0.49 kg/100 m2, respectively, during the poultry production process. In general, no significant differences (P>0.05) were noted between treatments for total nitrogen, ammonia, pH, EC, ash, and moisture when compost or the headspace air was sampled during compost preparation throughout all stages. Nor were mushroom yields or counts significantly affected (P>0.05) by the presence of ammonia suppressants in the poultry litter. Thus, the mushroom industry can confidently use poultry litter amended with PLT, Impact-P, and Barn Fresh when used at the recommended rates.  相似文献
