The study investigates the change in osmolality and haemolymph constituents in marron Cherax cainii and yabbies Cherax destructor associated with moult stages, body weights and their feeding status. A total of 582 haemolymph samples from 5 moult stages (postmoult-AB, intermoult-C, and premoult stages – D0, D1, D2), two body weight classes (2–15 g and 61–75 g) and nutritional status were used for analysis of osmolality, protein, glucose, and ionic concentrations of potassium and chloride following the standard biochemical procedures. The haemolymph protein, glucose, potassium and chloride levels were highest at intermoult and early premoult stages, and lowest at postmoult in both crayfish species. Except protein, no significant differences were seen in analyzed parameters between various weight classes and two species. Haemolymph osmolality, protein and glucose were significantly higher in fed crayfish, whereas no variations in haemolymph potassium and chloride concentrations were observed between the fed and unfed crayfish. Maximum osmolality was recorded at 7–8 h after feeding in both crayfish species. The results showed that the biochemical changes in the haemolymph of marron and yabbies are related to moult stages, body weight and feeding and thus can be used as tools for determining suitable diets.  相似文献

The presence of the yabbie Cherax destructor in a number of wild aquatic systems in the Pilbara and Southwest Coast Drainage Divisions of Western Australia is documented. This is of great concern as all native freshwater crayfishes in Western Australia are endemic and restricted to the southwest, while the Pilbara Division has no native species. An introduced population of C. destructor was sampled monthly from the Hutt River (Pilbara Drainage Division) for determination of life-history and reproductive biology in a wild aquatic system in Western Australia for the first time. Proliferation in that system was attributed to specific traits including: attaining first maturity at the end of its first year of life; a protracted spawning period (July–January); relatively high mean ovarian fecundity of 210.2 (±9.24 S.E.); and a rapid growth rate (curvature parameter K = 0.78 and asymptotic orbital carapace length OCL = 51.25 mm ascertained from a seasonal von Bertalanffy growth curve) that was comparable to the larger sympatric marron Cherax cainii in this system. The life-history characteristics of C. destructor in the Hutt River were typical of many other invasive crayfish species and it has the potential to impact the unique aquatic ecosystems and the endemic freshwater crayfish species of the region.  相似文献

The responses of individuals of Cherax destructor and Euastacus armatus to water containing stressed or unstressed crayfish were tested in the laboratory. Individuals of E. armatus showed only an increase in locomotion when stressed crayfish water was introduced. Individuals of C. destructor showed a decrease in the time spent in a lowered posture, an increase in the time spent in the raised posture and in grooming behaviour when stressed crayfish water was introduced. Individuals of C. destructor responded more strongly to heterospecific animals (Cherax rotundus and E. armatus) than to conspecifics. The levels of responses to the two heterospecific species were similar.  相似文献

1. This paper investigates moult stage and size-specific changes in whole body composition during growth in juvenile crayfish in order to better describe the nature of growth and energy use. 2. Composition is described in terms of moult stage and size-specific wet, dry and ash-free dry weight, water, carbon, protein nitrogen, non-protein nitrogen, ash and energy. Dry weight and ash-free dry weight (AFDW) peaked in the middle and in the later stages of premoult. Both peaks were about 2–2.3 times postmoult weight. Losses in tissue weight during ecdysis were substantial in the smallest crayfish but declined with size. Water was taken up between late premoult and early postmoult. Tissue accumulation occurred primarily between B and D1.3, with further weight gain largely the result of fluid uptake. Ash increased immediately postmoult with a major peak occurring during intermoult. Mean organic carbon varied between 33 and 35.5% of the body and 49% of the exuvia. Chitin varied between 9 and 17% of body AFDW and made up about 50.5% of the exuvia. Protein content varied between 47 and 62% of body AFDW and about 25% of the exuvia. Carbon, chitin and protein were not affected by moult stage but protein declined with ocular carapace length (OCL) in larger crayfish. Mean caloric content varied between 19 and 22 J mg-1 AFDW depending on size and moult stage. Caloric content increased with OCL during premoult and early intermoult then declined with size until part of the way through premoult. 3. Relationships between protein, chitin and remaining carbon (organic carbon minus chitin) were examined. It is suggested that protein and some carbon are catabolized during the moulting process, possibly to fuel metabolism. Models are presented showing changes in proximate composition over the moult cycle for two sizes of crayfish, and tissue and energy accumulation and loss over a series of moult cycles and sizes from 3.1 mm to 17 mm OCL.  相似文献

Structural changes in the spermatophore of the freshwater ‘red claw’ crayfish Cherax quadricarinatus (Von Martens, 1898) (Decapoda, Parastacidae). —Acta Zoologica (Stockholm) 88 : 000–000 The structure of the spermatophore was studied in Cherax quadricarinatus. Pieces of the distal vas deferens and transferred spermatophore from the females were fixed, cut and stained. Within the distal vas deferens, the primary layer and the secondary layer of the spermatophore were distinguishable. In the latter, two components were detected: cytoplasmic droplets and a homogeneous matrix. During the first 10 minutes post‐extrusion the cytoplasmic droplets drastically changed from looking like ‘empty droplets’; at this time the spermatophore changed from a liquid stage to a sticky one. One hour after extrusion the spermatophore began to harden and within the first 24–48 h post‐mating it was a solid and intense white structure tightly attached to the female; after 72 h it acquired a softer aspect, completely dehiscing between 96 and 120 h post‐mating. Histologically, the primary layer maintained its integrity surrounding the spermatozoa while the secondary layer lost the cytoplasmic droplets. The spermatophore began to hydrate between 24 and 48 h and by 72–96 h many sections of the sperm cord began to coalesce. From 48 h post‐mating some fissures appeared within the matrix that enlarged between 72 and 120 h. We propose that both manipulation by the female and hydration are the mechanisms involved in the release of the spermatozoa from the spermatophore.  相似文献

The neurotransmitter dopamine (DA) modulates many physiological processes in decapod crustaceans, including reproduction. This study was aimed at evaluating whether the long‐term injection of DA affects ovarian growth in the freshwater crayfish Cherax quadricarinatus. Three experiments were performed with females of different mean initial size: 4.15 g (Group A); 9.65 g (Group B); and 13.98 g (Group C). Treated females were injected with DA and control females with physiological saline twice a week for 90 (Group B), 105 (Group A) and 120 (Group C) days. At the end of the experiments, the animals were killed, and the stage of ovarian development, gonadosomatic index, and mean oocyte diameter were determined. DA had a differential effect according to female size: it negatively affected ovarian growth of females in a weight range of 4–14 g and had no effect on ovarian maturation when injected to females with an initial weight of 14 g. The results are compared with previous reports in freshwater decapod crustaceans.  相似文献

The Australian Yabby Cherax destructor voluntarily emerges from water to breathe air with increased frequency as water PO2 decreases. When the water PO2 declined below 2.7 kPa the crayfish spent >50% of time breathing air. The respiratory gas transport, acid-base, ionic and energetic status were quantified in simulations of this emersion behaviour to determine the benefits that the crayfish may gain from switching to air-breathing. C. destructor initially showed an elevated O2 uptake rate on emerging from hypoxic water, but after 1 h the O2 uptake rate was not different from that of crayfish in normoxic water. During 3 h of air breathing, subsequent to 2.7 kPa aquatic hypoxia, the haemolymph PO2 increased while oxygen content was essentially unchanged, although cardiac output increased 5-fold. The haemolymph PCO2 increased from 0.44 to 1.21 kPa after 3 h while the CO2 content increased from 3.47 to 8.66 mmol · l−1 and the pH decreased from 7.73 to 7.57 after 1 h in air. In air C. destructor eventually achieved an O2 uptake rate similar to that achieved in water. A general hyperglycaemia occurred without anaerobiosis. In air-breathing C. destructor, small changes in lactate appear to offset the decrease in haemocyanin-O2 affinity caused by acid Bohr shift. During air-breathing, decreased haemocyanin-O2 affinity assisted in maintaining O2 diffusion into the tissues, but the ATP content of the tail muscle decreased so that after 3 h in air the energy charge was only 0.59. The data are consistent with a specific depression of the Emden-Meyerhof pathway, preventing either lactate formation or oxidative phosphorylation in the tail muscle, despite a concomitant glycogenolysis. Six isonitrogenous and isocalorific diets supplemented with five different levels of beta –1,3 – glucan (0.08%, 0.1%, 0.2%, 0.4% and 0.8%) were formulated and tested for marron (Cherax tenuimanus) growth, survival, organosomatic indices, osmoregulatory capacity and immunological parameters (total and differential haemocyte counts, haemolymph clotting time and bacteraemia). The sixth diet without any beta – 1,3 – glucan was used as a control. Each diet was provided to 18 marron (0.47 ± 0.02 g initial weight) replicated 3 times in individual 250 L fiberglass cylindrical tanks. Each tank was provided with a biological filtration recirculating water system. After 84 days of culture, the survival and yield were higher in the marron fed 0.1% beta glucan supplemented diet. The different levels of beta glucan did not alter any of the physiological parameters of marron. However, dietary supplementation with beta glucan resulted in significantly higher (P < 0.05) total haemocyte count (THC) and granular cells. The bacteraemia rank was lower in all diets having beta glucan supplemented with more than or equal to 0.1% compared to the control and 0.08% beta glucan supplemented diets. Results suggest that dietary beta – 1,3 – glucan at a minimum concentration of 0.1–0.2% can improve the immune system of marron.  相似文献

Larvae of an estuarine grapsid crabChasmagnathus granulata Dana 1851, from temperate and subtropical regions of South America, were reared in seawater (32/%.) at five different constant temperatures (12, 15, 18, 21, 24 °C). Complete larval development from hatching (Zoea I) to metamorphosis (Crab I) occurred in a range from 15 to 24 °C. Highest survival (60% to the first juvenile stage) was observed at 18 °C, while all larvae reared at 12 °C died before metamorphosis. The duration of development (D) decreased with increasing temperature (T). This relationship is described for all larval stages as a power function (linear regressions after logarithmic transformation of bothD andT). The temperature-dependence of the instantaneous developmental rate (D −1) is compared among larval stages and temperatures using the Q10 coefficient (van't Hoff's equation). Through all four zoeal stages, this index tends to increase during development and to decrease with increasingT (comparing ranges 12–18, 15–21, 18–24 °C). In the Megalopa, low Q10 values were found in the range from 15 to 24 °C. In another series of experiments, larvae were reared at constant 18 °C, and their dry weight (W) and respiratory response to changes inT were measured in all successive stages during the intermoult period (stage C) of the moulting cycle. Both individual and weight-specific respiration (R, QO 2) increased exponentially with increasingT. At each temperature,R increased significantly during growth and development through successive larval stages. No significantly differentQO 2 values were found in the first three zoeal stages, while a significant decrease with increasingW occurred in the Zoea IV and Megalopa. As in the temperature-dependence ofD, the respiratory response to changes in temperature (Q10) depends on both the temperature range and the developmental stage, however, with different patterns. In the zoeal stages, the respiratory Q10 was minimum (1.7–2.2) at low temperatures (12–18 °C), but maximum (2.2–3.0) at 18–24 °C. The Megalopa, in contrast, showed a stronger metabolic response in the lower than in the upper temperature range (Q10=2.8 and 1.7, respectively). We interpret this pattern as an adaptation to a sequence of temperature conditions that should typically be encountered byC. granulata larvae during their ontogenetic migrations: hatching in and subsequent export from shallow estuarine lagoons, zoeal development in coastal marine waters, which are on average cooler, return in the Megalopa stage to warm lagoons. We thus propose that high metabolic sensitivity to changes in temperature may serve as a signal stimulating larval migration, so that the zoeae should tend to leave warm estuaries and lagoons, whereas the Megalopa should avoid remaining in the cooler marine waters and initiate its migration to wards shallow coastal lagoons.  相似文献

This study investigated the effects of food quality on digestive enzyme activities, in vitro protein digestibility and histological traits of the midgut gland in juvenile crayfish Cherax quadricarinatus. Animals of a wide weight range were fed different diets: two commercial diets with high or low lipid content (high lipid and low lipid, respectively) and were compared with a reference diet (RF) previously formulated for this species. Proteinase, lipase and amylase activities were significantly influenced by diet and weight. Specific trypsin activity was significantly higher for crayfish fed with the HL diet. Trypsin activity depended on diet and weight. Protein digestibility showed that HL was the most digestible diet and RF the least. The weight of the animals did not affect protein digestibility. Structural disorganization, hypertrophy of B‐cells and presence of large vacuoles in R‐cells were mainly observed in juveniles fed with HL, indicative of malnutrition. Thus, our data suggest that the HL diet would not be the most appropriate for C. quadricarinatus, while RF diet would be more convenient for culture of this species.  相似文献

Effects of feeding and starvation on the moult cycle and on the ultrastructure of hepatopancreas cells were studied in Stage I lobster larvae (Homarus americanus Milne-Edwards). The relative significance of yolk and first food was quite different in larvae originating from two females. This difference was evident also in the amounts of stored lipid in the R-cells of the larval hepatopancreas. Most larvae from one hatch were, in principle, able to develop exclusively with yolk reserves (without food) to the second instar. The larvae from the second hatch showed lecithotrophic development only to the transition between late intermoult and early premoult (Stages C/D0 of Drachs's moult cycle) of the first larval instar. When initial starvation in this group lasted for 3 days or more, the point of no return (PNR) was exceeded. After the PNR, consumption of food was still possible, but development ceased in the transition C/D0 or in late premoult (D3–4). It is suggested that these stages of the moult cycle are critical points were cessation of development and increased mortality are particularly likely in early larval lobsters under nutritional stress. Examination of hepatopancreas R-cells suggested that the PNR is caused by an irreversible loss of the ability to restore lipid reserves depleted during initial starvation. Initial periods of starvation ending before the PNR prolonged mainly Stage D0 of the same instar (I). During this delay, structural changes in the R-cells caused by the preceding period of starvation were reversed: reduced lipid inclusions, swollen mitochondria, an increased number of residual bodies indicating autolysis, and a reduction of the microvillous processes. Continually starved larvae which showed lecithotrophic development throughout the first instar and were then re-fed after moulting successfully, had later a prolonged intermoult (Stage C) period in the second instar. This shows that, despite occasional lecithotrophy, food is an important factor in early larval development of the lobster.  相似文献

All intertidal gastropods for which cold tolerance strategies have been assessed have been shown to be freeze tolerant. Thus, freeze tolerance is considered an adaptation to the intertidal environment. We investigated the cold tolerance strategies of three species of subtropical and temperate snails (Gastropoda: Littorinidae) to determine whether this group is phylogenetically constrained to freeze tolerance. We exposed dry acclimated and wet rehydrated snails to low temperatures to determine temperature of crystallisation (Tc), lower lethal temperature and LT50 and to examine the relationship between ice formation and mortality. Tc was lowest in dry Afrolittorina knysnaensis (–13.6±0.4 °C), followed by dry Echinolittorina natalensis (–10.9±0.2 °C) and wet A. knysnaensis (–10.2±0.2 °C). The Tc of both A. knysnaensis and E. natalensis increased with rehydration, whereas Tc of dry and wet Afrolittorina africana did not differ (–9.6±0.2 and –9.0±0.2 °C respectively). Wet snails of all species exhibited no or low survival of inoculative freezing, whereas dry individuals of A. knysnaensis could survive subzero temperatures above –8 °C when freezing was inoculated with ice. In the absence of external ice, Afrolittorina knysnaensis employs a freeze-avoidance strategy of cold tolerance, the first time this has been reported for an intertidal snail, indicating that there is no family-level phylogenetic constraint to freeze tolerance. Echinolittorina natalensis and A. africana both showed pre-freeze mortality and survival of some internal ice formation, but were not cold hardy in any strict sense.  相似文献

Summary

The effects of spiperone and domperidone (dopaminergic antagonists), as well as naloxone (enkephalinergic antagonist) on ovarian growth of the crayfish Cherax quadricarinatus, were evaluated during the non-reproductive period. In vivo assays were carried out by administering these compounds by adding them to the crayfish food, and subsequently measuring the oocyte diameter (oocytes in secondary vitellogenesis). Only naloxone produced a significantly (p <0.05) higher oocyte diameter than that of the controls, suggesting that this antagonist was blocking the effects of endogenous enkcephalins on the secretion of one or more neurohormones involved in crustacean reproduction. To test this hypothesis, an in vitro incubation of ovarian pieces with thoracic ganglion (TG), a source of the gonad stimulating hormone (GSH), or eyestalk tissue (ET), the source of the gonad inhibiting hormone (GIH), with or without naloxone added to the incubation medium, was done. A significant increase of ovarian growth was observed when TG was present. Moreover, the addition of naloxone to this preparation was able to further increase the ovarian growth, presumably by preventing enkephalinergic inhibition of GSH secretion.  相似文献

Summary

Concurrent morphological, anatomical and physiological changes took place during the first reproductive cycle in the Australian red-claw crayfish Cherax quadricarinatus, which prepared the female for spawning and holding of the newly deposited eggs. The endopod became longer and wider than the exopod and developed a mixture of plumose and long thin simple (ovigerous) setae. Small oocytes (0.24±0.05 mm) were present in the immature ovary. The growing ovary contained two distinct oocyte populations: one consisted of small (0.55±0.07 mm), barely growing oocytes, while the other consisted of large oocytes, which increased in size continuously (0.73 to 2.55 mm) until egg laying took place. A gradual change in the relative abundance of ovarian polypeptides occurred until the late vitellogenic stage (large oocytes < 1.8 mm). Three predominant female-specific, SDS-PAGE separated, polypeptides were observed (103, 78 and 73 kDa) that may represent vitellin subunits. The most abundant carotenoid in the ovary was astaxanthin, while β-carotene was present at a lower concentration. The strong correlation between the increasing diameter of the oocyte and the concentration of astaxanthin in the ovary and in the hemolymph suggested an association of astaxanthin with vitellin and vitellogenin.  相似文献

The Red-Claw crayfish, Cherax quadricarinatus von Martens, is native to freshwater habitats of northern Australia and Papua New Guinea. Owing to its large size and suitability for aquaculture, C. quadricarinatus has been widely translocated around the world. Unfortunately, C. quadricarinatus is also recognised as invasive, having already established feral populations in South Africa, Mexico, Jamaica and Puerto Rico. The hardiness and conspicuous colouration of C. quadricarinatus has also made it popular in the aquarium trade worldwide, including Singapore. Here, we report the establishment of feral populations of C. quadricarinatus in the water supply catchments of Singapore.  相似文献

Moult cycle and morphogenesis in larval instars (zoea I, zoea II, megalopa) of the spider crabHyas araneus (L.) were studied in the laboratory. Changes in the epidermis and cuticle were documented photographically at daily intervals to characterize the stages of the moult cycle. Stage A (early postmoult) is a very short period during which the larva takes up water. During late postmoult (B) and intermoult (C) the endocuticle is secreted, and there is conspicuous epidermal tissue condensation and growth. The onset of early premoult (D0) is characterized by epidermal apolysis, occurring first at the bases of the setae in the telson of zoeal instars or in the rostrum of the megalopa, respectively. Intermediate premoult (D1) is the main period of morphogenesis, in particular of setogenesis: in the setae of the zoeal telson and carapace there is invagination or (in the zoea II) degeneration of epidermal tissues. Formation of new setae in the interior of epidermal tubules was observed in zoeal maxillipeds and in the antennae of the zoea II and megalopa instars. During late premoult (Stages D2–4) part of the new cuticle is secreted, and the results of morphogenesis become clearly visible. For technical reasons (rigid exoskeleton) only a preliminary account of the moult cycle in the megalopa can be given. A time schedule is suggested for the stages of the moult cycle. It is estimated that postmoult (A–B) takes ca 9 to 15 % of total instar duration, intermoult (C) ca 22 to 37 %, and premoult (D) ca 48 to 69 %. There is an increasing trend of relative portions of time (% of total instar duration) from instar to instar in Stages A–C (mainly in the latter) and a decreasing trend in Stage D (mainly in D0 and D2–4).  相似文献

Ecdysteroid titer in whole extracts of female O. gammarellus varies during the course of the moult cycle: it is minimal at the beginning of the intermoult cycle (stages A, B and Ca) and in D0, and maximal in D2. In hemolymph, the hormone content presents a single peak level in D2; this peak is constituted by ecdysterone, as revealed by HPLC. Ovaries, at the end of vitellogenesis, contain ecdysterone and low polarity substances. Ecdysteroids are also present in eggs after laying.

Variations in ecdysteroid levels seem to influence oocyte growth; vitellogenin synthesis, however, is independent of hormonal fluctuations.  相似文献

Summary

The Australian redclaw crayfish Cherax quadricarinatus hatches at an advanced stage. Two post-hatching stages rely on yolk reserves before the organism becomes a juvenile. After a juvenile appears, the yolk is depleted and young crayfish might spend a few days among female pleopods before starting to feed independently. During this phase, the juvenile organism needs alternative energy sources. Offspring from three females were analyzed with histological and histochemical techniques, from hatching until reaching the 2-week juvenile stage, to trace digestive system development and internal organ lipid and glycogen accumulation. Neutral lipids are stored in specialized cells of the hepatopancreatic tissue until the juvenile stage is reached. An increasing rate of accumulation was found. Glycogen was stored in hepatopancreas and tail muscle cells until post-embryo II, and no increasing or decreasing rate was found in storage functions.  相似文献

The influence of body size on the consumption of live zooplankton (Daphnia spp.) by freshwater crayfish was examined using yabbies (Cherax destructor) ranging from 5 to 45 g. Food preference between live zooplankton and inert pellets was also assessed under experimental conditions. In experimental tanks, yabbies of four size classes (<15, 15–24.9, 25–34.9 and 35–45 g) were presented with live Daphnia. All yabbies were held in separate tanks with five animals per size class. In yabbies less than 15 g, the feeding mode on zooplankton involved rapid searching and probing with the first two pairs of walking legs. Once a prey was located, the chelae on the end of these walking legs would grasp the zooplankton and then rapidly move it towards the mouthparts. Yabbies larger than 25 g tended to use their walking legs to push the Daphnia nearer to their third maxilliped which would then force or scoop the zooplankton towards the mouthparts. A short-term feeding trial showed that there was no significant difference between size classes in regards to zooplankton consumption (P > 0.05). Capture efficiency of live Daphnia by yabbies less than 15 g was significantly lower (76%, P = 0.008) than the three larger size classes (93.6%). Yabbies less than 15 g consumed a significantly (P < 0.001) higher percentage (5.2%) of their body weight than the other size classes (1.1%, 0.8%, and 0.6%, respectively). In the presence of both live zooplankton and a pellet diet, yabbies spent significantly (P = 0.005) more time feeding on zooplankton (85%) than on inert pellets (15%). This was the first study to quantify zooplankton consumption by yabbies and the results provide insights into understanding the trophic role of freshwater crayfish in structuring zooplankton communities and the husbandry management of crayfish farming.
