Seeds of the crisp lettuce cultivar Pennlake were germinated using all combinations of six ‘initial’ solutions of polyethylene glycol 6000 (PEG) with osmotic potentials ranging from 0 to -8 bars and seven ‘secondary’ solutions of PEG with osmotic potentials ranging from 0 to -10 bars, to which seeds were moved after 24 or 48 h in the ‘initial’ solution. The number of seeds germinating decreased at more negative osmotic potentials of both ‘initial’ and ‘secondary’ solutions but there was an interaction between germination temperature and the osmotic potential of the ‘initial’ solution. At an ‘initial’ solution osmotic potential of 0 bars germination at 20°C exceeded that at 10°C. As the osmotic potential of the ‘initial’ solution decreased germination at 20°C decreased more than at 10°C so that at the more negative osmotic potentials germination at 10°C exceeded that at 20°C. However seeds ungerminated after 14 days germinated normally when transferred back to water, so that the average final germination was 99.5%. The results suggest that major fluctuations in soil water potential in a seedbed are unlikely to influence seed germination per se provided that a period of 24 to 48 h at 0 bars tension is available at some time. The timing of such a period relative to sowing will have a considerable effect on the time of germination and hence the time of emergence. It is concluded that factors other than the direct effect of soil moisture content on germination are involved in reducing seedling emergence under fluctuating soil moisture conditions in the field.  相似文献

Under stress integrated germination test (SIGT), seeds undergo osmo-saline stresses, which enable to detect differences in vigour of long-term stored seeds with high germination percentage (G%). The quality of Brassica villosa subsp. drepanensis seeds stored in a genebank (at ? 20°C for 16 years) was compared with seeds at harvest by standard germination tests (GT), SIGT and cytogenetic analysis. No differences were detected in G% and mean germination time under GT. Conversely, SIGT performed with NaCl ? 0.9 MPa osmotic potential did not influence G% at harvest but reduced that of stored seeds, SIGT at ? 1.4 MPa reduced G% of both. Cytogenetic analysis showed reduction of mitotic index, appearance of chromosomal aberrations and smaller nucleoli in stored seeds compared with harvest seeds germinated in water. SIGT at ? 0.9 MPa had no effect on mitotic index, but increased chromosome aberrations and nucleoli number. SIGT at ? 1.4 MPa inhibited G% of harvest and stored seeds, reduced mitoses in harvest and completely prevented it in stored seeds. The results indicate that GT does not faithfully reflect the quality of stored seeds, with misinterpretation of their vigour, whereas SIGT and cytogenetical parameters are sensitive, reliable and inexpensive methods for early prediction of genetic erosion in germplasm banks.  相似文献

The effects of reduced water potential (ψ) on seed germination at 25 and 15 °C in unprimed (UP) and primed (P) seeds of two cultivars of sweet sorghum (cv. Keller and cv. Makueni local), were analyzed through the hydrotime model. Six ψ (from 0 to ?1.0 MPa) in polyethylene glycol 6000 (PEG) solutions were used for the tests. Seeds were primed in 250 g/L PEG solution at 15 °C for 48 h. Decreasing ψ of imbibition solution reduced and delayed germination. At 15 °C seeds germinated less and slower than at 25 °C at any ψ. Seeds of cv. Makueni local exhibited a greater sensitivity to water stress in terms of germination percentage, than seeds of cv. Keller, but they were faster in germination. Osmopriming was beneficial for seed germination, both in terms of final percentage and rate, at any temperature and ψ. The hydrotime analysis revealed that predicted θ H constant was increased when temperature was reduced to 15 °C and at this temperature median base water potential [ψ b(50)] for germination was higher (less negative) than at 25 °C. Seed priming shifted ψ b(50) towards more negative values and reduced θ H requirements for germination. At 25 °C the two cultivars behaved similarly while at 15 °C cv. Keller exhibited a ψ b more negative but required a greater θ H to germinate, indicating a greater water-stress tolerance but a slower germination, than cv. Makueni local. The application of the model allows to identify water stress tolerant cultivars during germination, to include into breeding programs for the selection of well-performing cultivars under stress conditions.  相似文献

In a climate variability context, knowledge about Alpha grass (Stipa tenacissima) seed germination and seedling establishment requirements is a key factor given the relevance of this species in wide areas of North Africa and southern Europe. Such knowledge can help to collect information about current S. tenacissima populations allowing the conservation and restoration in these ecosystems. According to this objective, we conducted a series of laboratory studies to investigate the effects of several ecological factors as temperature, light, salinity, burial depth and drought, in relation to germination capacity and development of new seedlings. The main results revealed that germination was delayed with increasing drought conditions generated by osmotic solutions. Seeds germinated at all the concentrations of NaCl solutions, but germination was completely inhibited at a PEG 6000 solution of ?1.6 MPa. Further, an osmotic potential of ?0.8 MPa inhibited root and shoot growth. Variations in temperature also promoted variable germination rates (GR50). The base germination temperature (Tb (50)) was less than 2 °C for the different osmotic potentials. Burial depth was another limiting factor required for establishment. Despite significant seed production, this study has identified several key limiting points affecting seed germination and seedling establishment which can affect the viability of their populations in the near future.  相似文献

Switchgrass (Panicum virgatum L.) is a warm perennial grass with valuable characteristics as a biofuel crop. To avoid competition with food crops, biofuel crops will be likely relegated to less productive soils such as marginal lands. Consequently, the salinity and water scarcity problems that commonly affect marginal lands compromise biofuel crop germination, emergence, and seedling establishment. The aims of this study were to study the germination and seedling growth of switchgrass under salinity and water stress and to describe the morpho-anatomical responses of the roots and leaves in the seedlings to these stress conditions. The effect of salt and water stress was assessed using sodium chloride (NaCl) and polyethylene glycol (PEG) 8000 at the same water potentials of ??0.8, ??1.0, and ??1.2 MPa. Seeds were moist prechilled for 7 days at 5 °C and germinated at 30/15 °C (8 h light/16 h dark). NaCl treatments (??0.8 and ??1.0 MPa) delayed germination rates but did not reduce the final germination percentage, whereas at a lower potential (??1.2 MPa), the final germination percentage was diminished. The effects of PEG (??1.0 and ??1.2 MPa) on the germination rate and final percentage were more detrimental than those induced by isosmotic concentrations of NaCl. PEG and NaCl reduced significantly the vigor index of ??0.8 to ??1.2 MPa. The morpho-anatomical changes such as the reduction in the root cross-sectional area and the thickening of the endodermis walls for both stress conditions and aerenchyma formation in the cortex under salinity could significantly contribute in the survival and tolerance during the early seedling stages.  相似文献

The spatial and temporal fluctuations of water availability can be an obstacle for recruitment of many species in the restinga and might restrict seed germination and seedling growth in specific regeneration safe-sites. Clusia hilariana is one of the most dominant species of Restinga de Jurubatiba. This species has a high proportion of seedling establishment occurring inside the tanks of soil bromeliads underneath vegetation patches. Given the thin seed coats, the fast germination time and seed dispersal of C. hilariana during the dry season, we hypothesized that their major regeneration niche (the tanks of soil bromeliads) is related to susceptibility of seed germination and also seedling growth to low water availability. To test this hypothesis, seeds were germinated under decreasing water potentials using PEG 6000 solutions and seedlings were grown under varying water regimes. The percentage of seed germination progressively decreased at lower water potentials. After 38 days in ?1.0 MPa no seeds germinated. However, approximately 90% of seeds germinated when transferred to Ψ = 0 MPa. The relative growth rates of seedlings of C. hilariana did not differ between water treatments. Thus, the major regeneration niche of C. hilariana is not a consequence of a high sensitivity of seeds and seedlings to water shortage. Nonetheless, C. hilariana showed an array of seed and seedling traits that may help to overcome establishment constraints of the harsh environment of restingas.  相似文献

Seeds with efficient antioxidant defence system show higher germination under stress conditions; however, such information is limited for the halophyte seeds. We therefore studied lipid peroxidation and antioxidant responses of a leaf-succulent halophyte Salsola drummondii during seed germination under different salinity levels (0, 200 and 800 mM NaCl), temperature (10/20, 20/30 and 25/35°C) and light regimes. Seeds absorbed water and germinated in less than 1 h in non-saline control while increases in salinity decreased the rate of water uptake as well as seed germination. Non-optimal temperatures (10/20 and 25/35°C) and complete dark condition reduced seed germination in comparison to those seeds germinated under optimal temperature (20/30°C) and 12-h photoperiod, respectively. Generally, higher lipid peroxidation and antioxidant enzyme activities were observed in seeds at non-optimal temperature and in those seeds germinated in dark. Decrease in reduced ascorbic acid content was found in highest salinity and temperature treatments, while reduced glutathione content did not change significantly with changes in salinity, temperature and light regimes. These results indicate variation in temperature and light but not salinity enhances antioxidant enzyme activities in germinating seeds of Salsola drummondii.  相似文献

Germination studies are important for collecting information on field seedling recruitment, plant conservation and restoration. This study investigated the role of light, temperature, nitrogen, water stress and burial depth in controlling germination of Stipa bungeana seeds. S. bungeana seeds are photo-inhibited; light significantly decreased seed germination regardless of temperature and water conditions. Seeds germinated at 10–30° C, and the highest germination was 72 % and 88 % at 20° C in light and dark, respectively. Thermal model analysis showed that presence of light significantly increased average thermal requirement [θ T (50)] from 105°Cd to 186°Cd at sub-optimal temperature, implying that light delays seed germination. Hydrotime model analysis showed that presence of light caused a shift in the median base water potential [Ψ b(50)] from ?0.68 to ?0.26 MPa, which partly explains why light decreased both percentage and speed of germination, even at optimal conditions. As burial depth increased, seedling emergence initially increased and then decreased; the highest seedling emergence recruitment was 43 %, for seeds buried at a depth of 1 cm. Field observations showed that seedling emergence occurred primarily from July to September, and scarcely occurred from April to June. These results suggest that the light inhibitory effect is an adaptive mechanism that prevents S. bungeana seeds from germinating on the soil surface. To attain highest seedling establishment, seeds of S. bungeana should be sown at a soil depth of 1 cm prior to the rainy season, using seeds stored for 1 year.  相似文献

In this study, we investigated the influence of cold stratification on seed germination in S. × niederederi, a hybrid between the North American S. canadensis and the European S. virgaurea, using fruit samples collected in 2016 in Poland. We aimed to test the hypothesis that the low temperature exposure decreases the final percentage and speed of seed germination in the hybrid and its parental species. For each species, sets of 100 achenes in three replications were mixed with dry sand and stored in Petri dishes in darkness for 12 weeks, at ?18 °C and?+?4 °C, and?+?25 °C. The seeds were incubated for 21 d at room temperature (+25 °C), under the 12 h photoperiod (630 lx). We showed a lack of significant differences in: (i) the final percentage of germinated seeds of studied species stored at the same conditions, (ii) the final percentage of germinated seeds between the applied stratification conditions in the hybrid and its parental species, and (iii) the mean values of Timson’s index, mean germination time, and coefficient of velocity of germination between the stratification conditions in each species. The statistically significant inter-specific differences in the mean germination time parameter after the +25 °C treatment suggest that the seeds of S. × niederederi are able to germinate faster than the seeds of its parental species. However, to improve our knowledge of naturalization and invasion abilities of S. × niederederi by sexual reproduction, the seed germination and seedling survival of the hybrid should be tested in the field.  相似文献

Dioon edule seedling mortality is mostly attributed to dehydration by prolonged drought, even when they present xeromorphic characteristics like the adult plants. The effect of germination date (GD) and soil water deficit on seedling tolerance to water stress was assessed. The seedlings germinated and grown from mature seeds every month from December to April GD were selected to evaluate the leaf area, photosynthetic pigment content, crassulacean acid metabolism (CAM) activity, stomatal conductance (gs) and leaflet anatomy at soil water potential (Ψs) of 0.0 MPa (day 1), ?0.1 MPa (day 40), ?1.0 MPa (day 90), ?1.5 MPa (day 130), and a control (0.0 MPa at day 130) to recognize differences due to leaf development. The seedlings shifted from C3 to CAM cycling when exposed to water stress at Ψs of ?1.0 MPa, like adult plants. The March–April GD seedlings with undeveloped sclerified hypodermis and stomata, presented reduced leaf area, lower Chlorophyll a/b ratio, higher CAM activity and midday partial stomatal closure when reached Ψs of ?1.0 MPa. These have higher probability of dehydration during severe drought (February–April) than those of the December–February GD with similar Ψs. Plants used for restoration purposes must have full leaf development to increase the survival.  相似文献

为了阐明干旱胁迫与不同储藏时间对杠柳种子萌发的影响机理,在室内采用不同浓度的聚乙二醇(PEG)模拟干旱胁迫条件,对3种不同储藏时间的杠柳种子进行了萌发期对比试验,观测了种子的总萌发率、发芽势、活力指数和胚根的生长情况,分析了抗旱特性,结果表明:不同程度的干旱胁迫与储藏时间对杠柳种子萌发均有一定的影响,当PEG-6000浓度为5%,对储藏1年杠柳种子的萌芽率和发芽势起促进作用,对储藏2年种子萌芽率轻度抑制,对储藏3年种子抑制影响较大,当PEG-6000浓度>5%,3种种子的萌芽率和发芽势均呈下降趋势,但当胁迫浓度增大到25%时,种子基本没有萌发。在不同的水分胁迫处理中,3种种子的活力指数和胚根生长速度一直呈下降趋势,尤其3年种子的下降幅度最大。不同储藏时间杠柳种子对干旱胁迫的适应性有差异,储藏1~2年的杠柳种子在PEG-6000浓度为5%~15%中萌发效果较好,且储藏1年种子好于2年种子,3年种子较差。3种种子的耐旱能力由强到弱依次为:1年种子>2年种子>3年种子。  相似文献

Abstract

Limonium mansanetianum is catalogued as critically threatened (CR) species and it is included in Valencian Catalogue of Threatened Plant Species. Limonium mansanetianum is a gypsicolous species, which only lives in a restricted area to south-centre of Valencia province (Spain). The species is a low-branched woody shrub with summer flowering. The influence of incubation temperature (10°, 15°, 20° and 25°/20?°C) and salinity (0%–3.0% NaCl) on seed germination of L. mansanetianum was studied. Best seed germination was obtained in distilled water controls. Seed germination decreased with an increase in salinity and few seeds germinated at 2.5% and 3.0% NaCl. Optimal temperature regime for germination was 15?°C where germination in 0.5% and 1.0% NaCl was not affected. Recovery and hypersaline conditions experiments showed that L. mansanetianum seeds displayed a greater tolerance to high salinity and temperature stress before germination.  相似文献

Periploca sepium Bunge is a native and widespread shrub on the Loess Plateau, an arid and semi-arid region in China. To understand the adaptability of its seed germination to dry environments, we investigated the germination rate, water relations, lipid peroxidation, antioxidant capacity and accumulation of major organic solutes during seed germination under water deficit conditions. Results showed that seeds pre-treated by hydration–dehydration or ?0.9 MPa PEG germinated faster than control seeds, indicating strong resistance of P. sepium to drought condition. The re-dried seeds showed higher proline, total free amino acids (TFAA) and soluble proteins (SP) contents than control dry seeds, indicating the maintenance of physiological advancement when dehydrated. Osmotic stress made seed germination stay on the plateau phase (phase II). However, germinating seeds moved into phase III immediately once transferred into distilled water. Large increases in SP and soluble sugars (SS) of both re-dried and osmotic stressed seeds help themselves to resist drought stress. The re-hydrated seeds showed significantly higher levels of proline, TFAA, SP and SS than control seeds. The largely accumulated SS during osmotic stress declined sharply when transferred into distilled water. Our data demonstrate that P. sepium’s tolerance to drought stresses during germination is associated with enhanced activity of antioxidant enzymes and accumulation of some compatible solutes. Seed physiological advancement progressed slowly under low water conditions and it was maintained when seeds were air dried. This strategy ensures high and more rapid seed germination of P. sepium under drying and wetting conditions in drought-prone regions.  相似文献

Responses of seed germination to air temperature, water potential, light, and smoke were studied in the laboratory for seeds of the invasive bunch grass Pennisetum ciliare (L.) Link (syn. Cenchrus ciliare L.; buffel grass). First introduced to North America during the mid-twentieth Century for establishing pastures, this African bunch grass has become an invasive species of concern. Across all the experiments conducted, a low germination was observed for P. ciliare fascicles that never exceeded 30 % at 21 days after sowing. Optimal day/night air temperatures for germination, controlled with an environmental chamber, were 25/15 and 30/20 °C, while extreme temperatures of 15/5 and 45/35 °C inhibited germination. By sowing seeds of P. ciliare under different water potentials, created with aqueous solutions of polyethylene glycol, an optimum of ?0.03 MPa led to the highest germination, while no germination was observed at ?1.0 MPa. Monochromatic optical filters were utilized to germinate seeds under various wavelengths, of which red (650 nm) and far red (730 nm) led to the highest germination. In addition, seeds that were incubated in the dark had higher germination than those incubated under white light. Incubation in smoke water, which can stimulate germination of pyrophytic species, resulted in a marginal inhibition of germination compared with imbibition with distilled water.  相似文献

Three seedlots of crisp lettuce cv. Saladin were ‘primed’ in 1% tri-potassium orthophosphate (K3PO4) or in water for 6 to 12 h and then dried at room temperature. Two of the seedlots were separated into fractions of different mean weight before ‘priming’. The effects of ‘priming’ and grading on crop uniformity, germination at 35°C and root length in a slant test were evaluated. In general, ‘priming’ increased root length in a slant test but there were no consistent beneficial effects of ‘priming’ on seedling size, head weight or uniformity of head weight. Larger seed produced longer roots in a slant test than small seed and gave larger seedlings but seed size had no effect on head weight or cv. of head weight. ‘Priming’ had no effect on germination at 20°C but significantly increased germination at 35°C. ‘Priming’ in K3PO4 was more effective than in water. It is suggested that the response of seeds to ‘priming’ varies with seedlot and individual seed weight.  相似文献

Few plants are habitat-indifferent halophytes (i.e., grow well in both saline and non-saline soils). These plants offer a good opportunity to study drought and salinity tolerances during germination for seeds developed and matured in soils differ in salinity. Here, we assessed drought tolerance during germination, as simulated with PEG, and response of germination to light and temperature for Suaeda vermiculata, a habitat-indifferent shrub. Seeds matured in saline and non-saline soils were germinated in six PEG concentrations (0 to ? 1.0 MPa) and put in three incubators adjusted at different temperatures in both light and dark regimes. Drought tolerance was greater for seeds of the non-saline than those of saline soils, especially at higher temperatures. Seeds of the saline soils germinated in the lowest osmotic potentials (? 0.8 and ??1.0 MPa) only at lower temperatures, but seeds of the non-saline soils germinated to higher levels at all temperatures. Tolerances to drought and high temperatures were greater in light for seeds of saline soils, but in darkness for seeds of non-saline soils. Germination rate index did not differ significantly between seeds of the two soil types in higher osmotic potentials, but was significantly greater in seeds of non-saline at lower osmotic potentials. Most seeds that failed to germinate in the PEG concentrations recovered their germination when transferred to distilled water. Germination recovery levels and speeds increased with the decrease in osmotic potentials. Seeds of the saline soil postpone their germination until arrival of suitable temperatures and effective rainfalls that ensure seedling survival in salty habitats of the arid unpredictable deserts.  相似文献

渗透胁迫下水稻种子萌发特性及抗旱性鉴定指标研究   总被引:30,自引:0,他引:30  
用聚乙二醇(PEG)-6000作为渗透剂模拟干旱胁迫,探讨在4种不同浓度的渗透胁迫下水稻种子的萌发特性,并确立间接评价水稻苗期抗旱性的萌发期鉴定指标。结果表明,高浓度渗透液对种子萌发的抑制作用明显大于低浓度,萌发率的干旱胁迫反应指数随渗透液浓度的降低和萌发进程而增加。在不同浓度的渗透胁迫处理中,20%浓度下萌发6d后的萌发率表现较大的变异幅度和变异系数以及较均衡的品种频率分布,较好地反映了水稻种质对渗透胁迫的不同反应,认为20%/6d条件较适合作为萌发率鉴定的渗透液模拟干旱胁迫;经反复干旱后的幼苗存活率与萌发胁迫指数和20%胁迫浓度下萌发8d后芽鞘长的干旱胁迫反应指数(DRI)表现极显著或显著正相关,认为萌发胁迫指数和芽鞘长的DRI可作为评价水稻苗期抗旱性的间接鉴定指标。  相似文献

Effects of temperature, light, NaCl and polyethylene glycol(PEG)-6000 on seed germination and radicle growth in a halophyticshrub, Kalidium caspicum(L.) Ung.-Sternb. were investigated.When seeds were incubated in deionized water at constant temperaturesbetween 10 and 30°C, the percentage germination in the darkexceeded 75%; light suppressed seed germination at alternatingtemperatures. Incubating seeds with a hypersaline solution ofNaCl for 30 d had no adverse effect on their germinability.The percentage germination of seeds incubated with a –0.8MPa NaCl solution was 73, 80 and 54% at 10, 20 and 30°C,respectively, but all radicles died before their length exceeded5 mm. In contrast, when seeds were incubated with a –0.8MPa PEG solution at 20°C, 68% of seeds germinated, and 95%of the emerging radicles survived beyond 5 mm. Orchid is a major floral crop around the world and Dendrobium hybrids are considered to be one of the most popular orchids. In vitro germination of hybrid seeds is a common practice among orchid growers, however, in many cross pollinations the embryos may not develop to maturity, leading to poor seed germination. The effect of seed maturity and sucrose concentration were investigated via asymbiotic germination of nobile Dendrobium hybrids. Capsules were harvested from two hybrids (Den. Lucky Girl × Den. Second Love ‘Kirameki’ and Den. Lucky Girl × Den. Hamana Lake ‘Kumi’) and one selfing of Den. Second Love ‘Kirameki’ at 2, 3, 4, and 5 months after pollination and immature seeds were taken. Immature seeds from 3- to 5-month old capsules could be successfully germinated on Hyponex based medium. Immature seeds from 4-month old capsules showed greatest germination rate of tested treatments, whereas 3-month old immature seeds showed the least germination. After 6 weeks of in vitro culture, protocorms derived from embryos developed on every concentration of sucrose, but germination was greater at lower concentrations. Greater concentration of sucrose decreased normal-developed protocorms.  相似文献

模拟水分胁迫对不同种源麻楝种子萌发能力的影响   总被引:3,自引:0,他引:3  
以麻楝6个种源种子为试验材料,用不同浓度的聚乙二醇(PEG)溶液模拟干旱胁迫,探讨干旱胁迫对种子发芽率、发芽势、发芽指数、活力指数以及幼苗苗高和胚根长及根苗比的影响,为麻楝的引种和推广种植提供依据。结果显示:(1)不同水势胁迫处理均降低了麻楝种子的发芽率和发芽势,当水势为-0.40MPa时延缓了种子萌发进程;种子的发芽率、发芽势、发芽指数和活力指数均随干旱胁迫强度的增加呈明显下降的趋势;当胁迫水势为-0.86MPa时,干旱胁迫处理的种子在试验结束时仍未能萌发,即-0.86MPa是麻楝种子萌发的临界水势。(2)当胁迫水势高于-0.40MPa时,麻楝幼苗的胚根长度与对照组差异不显著且长于对照组,说明高于-0.40MPa的水势有利于麻楝种子胚根的生长;麻楝幼苗苗高生长则是随着PEG浓度的升高而逐渐减缓。(3)适当的干旱胁迫可以增大各种源麻楝幼苗根苗比,且在胁迫水势高于-0.20MPa时都达到最大值。研究表明,麻楝种子具有一定的抗干旱胁迫的萌发能力,并以来自缅甸的Khin Aye Pale和泰国的Phu Wiang材料较强,来源于中国三亚和马来西亚Ulu Tranan的较弱。  相似文献
