为了探究浙贝母(Fritillaria thunbergii)药效成分积累量与生物碱合成相关基因表达水平之间的关系,该研究采用UPLC-MS、qPCR技术分别测定不同产地11个样品中总生物碱(贝母素甲和贝母素乙之和)含量和3个参与生物碱合成途径相关基因(HMGRFPSDXR)的表达量,同时运用生物统计学方法分析成熟期鳞茎生物碱含量与各基因表达量之间的相关性。结果表明:不同产地浙贝母成熟期鳞茎总生物碱含量存在显著差异(P<0.05),为0.2105% ~ 0.4612%; HMGRFPS基因在盛花期组织表达、盛花期至成熟期鳞茎表达变化趋势同生物碱含量变化趋势基本一致; DXR基因在成熟期鳞茎中表达量最高,盛花期组织表达、盛花期至成熟期鳞茎表达变化趋势同生物碱含量变化趋势大体不一致; HMGRFPS基因表达量分别与贝母素甲、贝母素乙和总生物碱含量呈显著或极显著正相关性(P<0.05或P<0.01),以FPS基因表达量与生物碱含量相关系数为最高,相关系数分别为0.672,0.631,0.664,DXR基因与生物碱含量呈低度相关性。由此可以推断,生物碱的积累受MVA途径中HMGRFPS基因协同调控或者修饰作用明显,受MEP途径中DXR基因调控作用不明显。  相似文献

目的 探讨医护人员心理资本、应对方式与工作倦怠的关系。方法 采用心理资本量表、简易应对方式量表和Maslach工作倦怠量表通用版对河北省某省级三甲医院101名医护人员进行调查。结果 (1)医生自我效能得分高于护士,成就感低落得分低于护士(P<0.05)。(2)心理资本与工作倦怠(乐观与成就感低落相关除外)呈显著负相关(P<0.01 or P<0.05),消极应对与成就感低落呈显著正相关(P<0.01)。(3)希望对情绪衰竭和玩世不恭有明显负向预测(P<0.01,P<0.001),自我效能(P<0.001)和消极应对(P<0.05)对成就感低落分别有负向和或正向预测作用。结论 提升心理资本水平,调整消极应对方式能有效预防和矫治医护人员工作倦怠;医院管理层应关注护士群体的心理状态。  相似文献

为明确施肥和刈割对蒙古半干旱草地四种植物化学计量特征的影响,该文于2014年8月以内蒙古半干旱草地的四种植物冷蒿(Artimesia frigida)、菊叶委陵菜(Potentilla tanacetifolia)、羊草(Aneurotepidimu chinense)、扁蓿豆(Melissitus ruthenica)为研究对象,利用方差分析及多重比较分析对照、施氮肥、割草、割草和施氮肥处理下的植物碳、氮、磷含量的差异以及不同处理下碳氮比和氮磷比的变化。结果表明:施氮肥与割草均未影响植物碳含量。施氮肥显著提高了植物氮含量(P<0.05),进而降低了碳氮比(P<0.05),提高了氮磷比(P<0.05)。割草处理后扁蓿豆的氮含量显著提高了0.18%(P<0.05),羊草的氮含量显著降低了0.13%(P<0.05),冷蒿与菊叶委陵菜的氮含量无显著变化。割草显著提高了冷蒿和菊叶委陵菜的磷含量(P<0.05),但是未影响羊草和扁蓿豆的磷含量。该研究证实了氮添加或割草会影响部分草地物种的化学计量特征,但是氮添加或割草对植物化学计量特征的影响存在种间差异。  相似文献

【目的】探索研究反刍动物胃肠道微生物合成维生素B12的方法,并评估植物乳酸菌或博落回提取物对断奶山羊回肠食靡微生物合成维生素B12的影响。【方法】选取体重相近年龄相仿的断奶黑山羊20只,随机分为对照组(CON, n=7)、乳酸菌组(LAC, n=7)和博落回组(MAC, n=6)。CON组饲喂普通的日粮,LAC组饲喂基础日粮+10 g/d的植物乳酸菌(Lactobacillus plantarum P-8 strains, 4.0×109 CFU/g),MAC组饲喂基础日粮+0.3 g/d的博落回提取物(Macleaya cordata 3.75%)。试验结束后,采集回肠中段食靡样品。利用宏基因组测序技术,比对最新功能基因数据库VB12Path和公共数据库KEGG,分析植物乳酸菌和博落回提取物对山羊回肠食靡微生物合成维生素B12的影响。【结果】结果显示,比对VB12Path数据库共注释到55个与维生素B12合成相关的基因。与CON组相比,LAC组和MAC组中合成维生素B12基因的丰富度和均匀度降低(P<0.05)。3组间基因的β多样性也有显著的差异(P<0.05);比对KEGG数据库共注释到49个与维生素B12合成相关的基因,LAC组的多样性与CON组没有差异,但MAC组的α多样性显著降低(P<0.05)。值得注意的是,比对VB12Path数据库和KEGG数据库均发现LAC组和MAC组中参与前咕啉2合成途径、参与无氧合成途径、有氧合成途径、参与重排转换途径以及腺苷钴胺素合成途径的部分基因(gltXcbiTcobTbtuD等)的丰度均显著地高于CON组(P<0.05)。【结论】2个数据库比对后的相似结果表明博落回提取物在对断奶山羊回肠微生物合成维生素B12相关代谢上与植物乳酸菌的作用相似,均可以通过改变其多样性和提高部分关键基因的丰度,从而影响微生物合成维生素B12的潜能,为后期博落回提取物和植物乳酸菌在畜牧养殖中的运用提供一定的理论支撑。此外,2个数据库比对的差异提示未来研究胃肠道微生物维生素B12相关代谢时,应用多个数据库比对,能更全面精确地进行评价,为后期分析过程奠定研究基础和提供新的思路。  相似文献

肠道黏膜微生物在调控宿主生理功能方面发挥重要作用,其结构组成受到多种因素影响。性别被认为是塑造肠道微生物的因素之一。然而,性别对肠道黏膜菌群的差异影响还不清楚。目的以江山乌猪为研究对象,探究性别差异对其肠道黏膜微生物组成及功能的影响。方法选取性成熟的雌性和雄性江山乌猪各8头,利用16S rRNA基因高通量测序技术分析回肠和结肠黏膜菌群。结果在回肠黏膜中,雄性江山乌猪菌群的Chao1指数和Shannon指数显著高于雌性(P<0.05),在结肠黏膜中,不同性别江山乌猪菌群Chao1指数和Shannon指数无显著差异(P>0.05)。菌群差异分析显示,回肠黏膜中,沙雷氏菌属(Serratia)和埃希氏志贺菌属(Escherichia_Shigella)在雌性组中的相对丰度显著高于雄性组(P<0.05),雄性组中Oscillospiraceae UCG-005、拟普雷沃氏菌属(Alloprevotella)、布劳特氏菌属(Blautia)和Prevotellaceae_NK3B31_group相对丰度显著高于雌性组(P<0.05);结肠黏膜中,雌性组中unclassified_MuribaculaceaeRikenellaceae_RC9_gut_group和Prevotellaceae UCG-003相对丰度显著高于雄性组(P<0.05),Oscillospiraceae UCG-005、Lachnospiraceae_NK4A136_group和unclassified_Lachnospiraceae在雄性组中相对丰度更高(P<0.05)。功能预测发现,雄性乌猪回肠黏膜菌群显著富集了氨基酸代谢、碳水化合物代谢和能量代谢等功能途径(P<0.05);结肠黏膜菌群主要富集了膜转运相关的ABC转运蛋白和信号转导相关的双组分系统等功能途径(P<0.05)。结论不同性别江山乌猪肠黏膜菌群结构及功能具有明显差异。这些结果揭示了不同性别江山乌猪肠道黏膜菌群的差异特征,为了解和挖掘我国地方畜禽品种肠道微生物资源提供部分参考。  相似文献

为研究溶藻弧菌Ⅲ型分泌系统VcrV基因的功能和生物学特性,采用同源重组技术成功构建了溶藻弧菌缺失株ΔVcrV,并用PCR检测其遗传稳定性。结果显示,缺失株遗传稳定;与野生株相比,生长和自凝集能力无显著变化;但生物膜形成能力降低,半致死剂量升高16.5倍;游动性和涌动性极显著升高,细胞粘附能力极显著降低(P<0.01),对H2O2和NaCl的耐受性降低;对头孢呋辛、麦迪霉素、克林霉素抗生素敏感度上升,对丁胺卡那、多粘菌素B抗生素敏感度降低;活性氧含量极显著降低(P<0.01),脯氨酸、肽聚糖、β-内酰胺酶、过氧化氢酶、超氧化物歧化酶、谷胱甘肽过氧化物酶指标均极显著升高(P<0.01)。缺失株生物学特性表明,VcrV基因参与溶藻弧菌Ⅲ型分泌系统性的致病性和多种生物学功能。  相似文献

为分析马缨杜鹃(Rhododendron delavayi)花开花至凋谢过程中的代谢产物差异及其通路,该文采用LC-MS/MS技术对其花苞期、开裂期、传粉期、盛开期、衰老期和凋谢期的化学成分进行非靶向代谢组学分析。结果表明:(1)共鉴定到973种代谢物,主要包含黄酮类、有机酸、酚酸类、氨基酸及其衍生物、脂类、生物碱等。(2)主成分分析(PCA)表明样本间代谢物存在差异,结合正交偏最小二乘判别分析(OPLS-DA)、t检验的P值和单变量分析的差异倍数(fold-change)筛选差异代谢物(VIP>1,P<0.05,Fc>2或Fc<0.5),涉及591种,在马缨杜鹃花期进入衰老期和凋谢期后差异代谢物数量和表达量显著上升,其中花苞期至开裂期差异代谢物的表达主要呈现下调,而进入衰老期和凋谢期后差异代谢物的表达主要呈现上调。(3)KEGG注释到68条代谢通路,其中差异代谢物极显著富集(P < 0.01)通路3条,包括苯丙素类生物合成、植物激素的生物合成和类黄酮生物合成。(4)结合苯丙素类、黄酮类等有效成分生物合成通路共筛选到10种代谢物包括苯丙氨酸(L-phenylalanine)、反式肉桂酸(trans-cinnamic acid)、查耳酮(chalcone)、柚皮素(naringenin)、对香豆酰基莽草酸(p-coumaroyl shikimic acid)、阿魏酸(ferulic acid)、松柏醇(coniferyl alcohol)、芥子酸(sinapic acid)、紫丁香苷(syringin)、槲皮素(quercetin)。此外,有效成分的差异代谢物表明苯丙素类生物合成代谢活动随马缨杜鹃花的发育逐渐增强,而黄酮类化合物生物合成逐渐减弱,这些关键差异代谢物可能对马缨杜鹃花的发育有重要的调控作用。该研究为马缨杜鹃花开花至凋谢进程中的有效成分代谢途径活性物质的研究提供了代谢组学基础,为进一步研究马缨杜鹃花花期调控的分子机理提供参考。  相似文献

【背景】研究发现PrtV基因编码含多囊肾病(polycystic kidney disease)结构域的金属蛋白酶,其在多种细菌的致病过程中具有重要作用。拟态弧菌是一种感染多种水生动物的重要病原菌,PrtV基因在拟态弧菌致病中的作用尚不清楚。【目的】探究PrtV基因对拟态弧菌致病相关生物学特性的影响。【方法】采用自然转化的方法构建拟态弧菌PrtV基因缺失株(ΔPrtV),同时通过基因与质粒重组后电转化导入缺失株构建回补株(ΔPrtV/pPrtV),对突变株的生长特性、生化特征、生物被膜形成、自聚集能力、胞外产物卵磷脂酶和蛋白酶活性,以及致病性和细胞毒性等进行分析。【结果】与野生株相比,缺失株的生长特性、生物被膜形成、自聚集能力和卵磷脂酶活性无变化,但分解尿素、甘氨酸、香豆酸盐、鸟氨酸和赖氨酸的理化特性改变;胞外产物蛋白酶活性显著降低(P<0.05),细胞毒性显著下降(P<0.05),对杂交鲇的致病力下降10倍。【结论】PrtV基因与拟态弧菌的细胞毒性及致病性等多种生物学特性有关。该结果为进一步解析拟态弧菌PrtV基因功能及其致病机制提供了依据。  相似文献

目的 探讨对脑卒中患者应用连续性护理的效果。方法 构建连续性护理模式,从入院开始进行连续性护理干预,通过患者住院天数、住院费用、再入院率、护理服务满意度、BI指数等指标评价应用效果。结果 连续性护理干预减少了患者住院天数、住院费用、再入院率(P<0.01,P<0.01,P<0.05),提高了患者对护理服务的满意度及BI指数(P<0.05)。结论 连续性护理是脑卒中患者护理的有效途径,值得推广应用。  相似文献

The effects of an arsenic-rich fish diet and selenium (Se) supplementation on blood arsenic (As), Se, and thyroid hormones were studied in 32 women divided into four equal groups. Groups 1 and 4 received 400 μg Se-methionine daily, group 2 received 400 μg selenite daily, and group 3 received placebo tablets for 15 wk. In addition, groups 1–3 increased their fish intake, eating at least three fish dinners weekly. Mean blood Se concentrations (initially 1.68 ± 0.24 μmol/L) increased twofold in the Se-methionine groups (p < 0.0001) and by 32% in the selenite group (p < 0.01). Group means of blood As concentrations increased by 63% (p < 0.01), 50% (p < 0.01), 106% (p < 0.01), and 29% (p < 0.05) in the four groups, respectively. Analyzed As intake from duplicate portions of consumed fish correlated with final blood As concentrations (r=0.85, p < 0.001, n=32). In the group not receiving Se, there was a positive correlation between final blood As concentrations and plasma T4 : T3 ratio (r=0.80, p < 0.02, n=8). Initially, blood As concentrations correlated negatively with both T3 and T4 in plasma, but this correlation disappeared upon Se supplementation. The results demonstrate that increased intake of fish may influence blood As concentrations and that circulating thyroid hormones may be influenced by Se-As interactions.  相似文献

蛋白质是构成生命系统的基本元件之一,是大部分生物学功能的执行者。蛋白质丰度与其生物学功能息息相关,其丰度受基因表达过程中各环节严格精密的调控。其中,蛋白质丰度与其相应mRNA丰度存在较强的相关性,蛋白质丰度差异的40%可由mRNA丰度来解释。茉莉酸信号途径调节巴西橡胶树中的天然橡胶生物合成,但相关基因彼此间的表达丰度差异尚待阐明。该文比较了S/2D d3割胶制度下,15个橡胶生物合成调控相关基因COI1、JAZ1、JAZ2、JAZ3、MYC1、MYC2、MYC3、MYC4、MYC5、GAPDH、HMGR1、SRPP、REF、HRT1、HRT2以及2个常用内参基因18S、ACTIN1在10个橡胶树种质胶乳中的表达丰度差异;将ACTIN1的表达丰度设定为1,以此为标准计算出样品中其他基因的表达丰度。结果表明:相同个体中不同基因的转录丰度差异明显,不同个体中相同基因集的丰度大小排序存在一定差异;同一基因在不同个体中的转录丰度差异明显,这16个基因的最大丰度分别是最低丰度的9.43、6.04、10.02、12.29、18.82、9.22、38.46、112.83、121.36、15.34、19.09、13.54、10.05、19.80、24.83、11.82倍,他们的变异系数分别为73.05%、55.19%、69.09%、67.37%、66.59%、53.87%、83.25%、122.02%、166.34%、59.89%、70.59%、75.67%、74.20%、68.34%、84.23%、78.59%;总的来说,在群体水平上,16个基因的转录丰度从高到低依次为18S SRPPHMGR1REFMYC2/HRT1COI1MYC1/MYC4GAPDH/JAZ1/MYC5JAZ2HRT2/MYC3/JAZ3,他们的群体平均丰度依次为ACTIN1的28 382.26、43.64、11.39、7.16、5.47、5.10、1.07、0.75、0.74、0.45、0.42、0.33、0.12、0.06、0.06、0.04倍。值得注意的是,无论在个体水平还是群体水平上,18S的丰度毫无疑问是最大的,在mRNA中,SRPP的丰度最大,JAZ1大于JAZ2和JAZ3,MYC2大于MYC1、MYC3、MYC4、MYC5,HRT1大于HRT2。综上结果表明,结构基因和功能基因的丰度高于调控基因。在基因相对表达分析中,常对目的基因和内参基因作均一化处理,从而掩盖了不同基因间的真实丰度差异,因此,在基因表达分析中,既要关注基因的相对表达量,也要关注基因间的丰度差异,这有助于更全面地理解基因的功能。  相似文献

割胶促进橡胶树合成天然橡胶与激活乳管细胞的茉莉酸信号途径密切相关,但茉莉酸信号途径关键环节的基因表达水平与干胶产量的相关性尚不清楚。为了找到与产量相关的分子标记,该研究采用qPCR技术,分析了割胶条件下茉莉酸信号途径关键环节的9个相关基因在5个橡胶树魏克汉种质和5个1981’IRRDB种质乳管细胞中的表达。结果表明:大多魏克汉种质的株次干胶产量显著高于1981’IRRDB种质。在9个基因中,除了HbMYC4和HbMYC5,其余7个基因在大多橡胶树魏克汉种质中的表达量均显著高于1981’IRRDB种质,尤其是HbMYC3基因表达差异性好,与干胶产量相关性高,有望作为橡胶树产量育种的一个分子标记。这对育种周期长的橡胶树产量育种具有实际应用价值。  相似文献

Degradation of rubber particles from tyre treads, having diameters from 0.8 to 2.3 mm, was achieved using Nocardia sp. 835A-Rc, a mutant strain with strong rubber-degrading ability. The entire surface of the particles was uniformly attacked by the organism either without stirring of the culture medium or at a very low stirring rate of 40 rpm. At a higher rate of stirring, however, a small number of large microbial colonies were formed on the rubber surface and separate deep semi-spherical cavities were observed after the removal of microbial cells by washing. The number of microbial colonies decreased with increasing stirring rate but each one of the colonies became larger at the same time. As the result of these two counteracting effects of stirring on microbial activity, the weight loss of the particles increased when the stirring rate was raised from 0 to 40 rpm but decreased when the rate was increased from 40 to 70 or 150 rpm. At the stirring rate of 40 rpm, the weight losses of the particles with mean diameters of about 0.8, 1.1 and 2.3 mm were 57, 50 and 36%, respectively, after 8 weeks. The rate of microbial degradation increased again when the stirring was raised from 150 to 300 rpm.  相似文献

Colletotrichum gloeosporoides has been described as the causal agent of Colletotrichum leaf disease of rubber in Sri Lanka and other parts of the world since 1905. A study carried out on vegetative and reproductive characteristics of 52 isolates from Colletotrichum leaf disease lesions on Hevea brasiliensis in Sri Lanka revealed that only 18 isolates belong to Colletotrichum gloeosporioides. The remaining 34 isolates represented C. aculatum indicating that C. acutatum is the main cause of Colletotrichum leaf disease in Sri Lanka.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

Embryogenic cell suspensions of rubber derived from immature inflorescences and inner integuments of immature fruits released 3.1 ± 0.2 × 107 protoplasts g-1 f. wt. (mean ± s.e.m, n = 10) and 3.2 ± 0.2 × 107 protoplasts g-1 f. wt., with mean viabilities of 83 ± 2% and 77 ± 8%, respectively. Sustained mitotic division was observed only when protoplasts were cultured in KPR liquid medium on nitrocellulose membranes overlying the same semi-solid medium containing Lolium multiflorum nurse cells. Protoplast-derived cell colonies were produced within 2 months of culture. Protoplast-derived cell colonies proliferated, upon subculture to MS-based regeneration medium, with 40% of the protoplast-derived calli developing somatic embryos. Powdery mildew of rubber tree caused by Oidium heveae is an important disease of rubber plantations worldwide. Identification and classification of this fungus is still uncertain because there is no authoritative report of its morphology and no record of its teleomorphic stage. In this study, we compared five specimens of the rubber powdery mildew fungus collected in Malaysia, Thailand, and Brazil based on morphological and molecular characteristics. Morphological results showed that the fungus on rubber tree belongs to Oidium subgen. Pseudoidium. Nucleotide sequence analysis of the ribosomal DNA internal transcribed spacer (ITS) region and the large subunit rRNA gene (28S rDNA) were conducted to determine the relationships of the rubber powdery mildew fungus and to link this anamorphic fungus with its allied teleomorph. The results showed that the rDNA sequences of the two specimens from Malaysia were identical to a specimen from Thailand, whereas they differed by three bases from the two Brazilian isolates: one nucleotide position in the ITS2 and two positions in the 28S sequences. The ITS sequences of the two Brazilian isolates were identical to sequences of Erysiphe sp. on Quercus phillyraeoides collected in Japan, although the 28S sequences differed at one base from sequences of this fungus. Phylogenetic trees of both rDNA regions constructed by the distance and parsimony methods showed that the rubber powdery mildew fungus grouped with Erysiphe sp. on Q. phillyraeoides with 100% bootstrap support. Comparisons of the anamorph of two isolates of Erysiphe sp. from Q. phillyraeoides with the rubber mildew did not reveal any obvious differences between the two powdery mildew taxa, which suggests that O. heveae may be an anamorph of Erysiphe sp. on Q. phillyraeoides. Cross-inoculation tests are required to substantiate this conclusion.  相似文献

Colonization and degradation of rubber pieces by Nocardia sp.   总被引:1,自引:0,他引:1  
The growth of a Nocardia sp. occurs essentially on the insoluble rubber substrate and the cells are tightly bound to the rubber in the initial stage of the growth in spite of vigorous stirring of the cultures. The colonization of rubber pieces was followed by staining with Schiff reagent, and it was revealed that not only the thickness of rubber pieces, but also their length and width greatly influenced microbial colonization and degradation of natural rubber products. Among rubber pieces of various shapes, long strips were most rapidly covered by many microbial colonies and experienced the highest rate of rubber degradation. The rate of degradation (expressed by % weight loss) of the long strips of rubber was a linear function of surface area per unit weight of rubber. Thin and wide films of rubber were also rapidly colonized and degraded, while the colonization and degradation of short and narrow pieces were substantially slower and less extensive.  相似文献

Cellulose production byAcetobacter pasteurianus was investigated in static culture using four bioreactors with silicone rubber membrane submerged in the medium. The shape of the membrane was flat sheet, flat sack, tube and cylindrical balloon. Production rate of cellulose as well as its yield on consumed glucose by the bacteria grown on the flat type membranes was approximately ten-fold greater than those on the non-flat ones in spite of the same membrane thickness. The membrane reactor using flat sacks of silicone rubber membrane as support of bacterial pellicle can supply greater ratio of surface to volume than a conventional liquid surface culture and is promising for industrial production of bacterial cellulose in large scale.  相似文献
