Gallbladder carcinomas are rare but highly lethal neoplasms. We examined the expression of five cell-cycle-related molecules (p53, RB, cyclin D1, p27, Ki-67), and MSH2, in 46 carcinomas, 14 adenomas, 15 low-grade dysplasias, 9 intestinal metaplasias and 20 normal gallbladder epithelia. The expression of these molecules was altered in gallbladder carcinomas and adenomas. In gallbladder carcinomas we observed increased expression of p53, cyclin D1, Ki-67, and MSH2 together with decreased expression of RB and p27 protein. Aberrant expression of cyclin D1 and reduced expression of RB were noted in adenomas, and expression of cyclin D1 was elevated in low-grade dysplasias. However, there was no change in the levels of these cell-cycle molecules in metaplasia. Expression of p53, p27, Ki-67, and MSH2 was correlated with clinical stage (P<0.05) and there was also a correlation between the expression of Ki-67 and MSH-2 and patient age (P<0.05). These results suggest that altered expression of cell-cycle molecules p53, cyclin D1, RB, p27, and of MSH-2 is involved in the progression of gallbladder carcinomas.  相似文献

Heat shock proteins (HSPs) can be induced by various stresses and play an important role in cell cycle progression. HSP70 has been shown to act as an inhibitor of apoptosis. We studied HSP70 expression in bronchial epithelial cells of C57BL/6 mice and homozygous HPS70 knockout mice (hsp70.1-/-) exposed to chronic hypoxic stress. We also investigated changes in cellular proliferation and apoptosis in relation to HSP70. Lungs were removed from mice after a three-week period of exposure to 10 % O(2). Immunoblots for HSP70 and immunohistochemical staining for HSP70 and Ki-67 were performed. Apoptosis was assessed using the TUNEL assay. The three-week period of hypoxic stress did not change HSP70 levels in total lung tissue, but a significant reduction in HSP70 expression was observed in bronchiolar epithelial cells. In wild type mice, both HSP70 and Ki-67 expression were significantly reduced in bronchiolar epithelial cells. In homozygous HPS70 knockout mice (hsp70.1-/-), apoptosis of bronchiolar epithelial cells was significantly increased. Our results suggest that HSP70 may exert anti-apoptotic effects in mouse bronchiolar epithelial cells.  相似文献

Heat-shock proteins (HSPs) are important in spermatogenesis. This study investigated developmental changes in the expression of major HSPs in porcine testis. The testis from five immature (mean age 2.9+/-0.1 months) and five mature boars (35.7+/-14.0 months) were examined. Two-dimensional polyacrylamide gel electrophoresis was conducted and proteins were identified by Western blotting and/or matrix-assisted laser desorption/ionization mass spectrometry. Moreover, the 90, 70, and 60 kDa HSPs, 70 kDa heat-shock cognate protein (HSC 70), tubulin, and actin were quantified on two-dimensional gels. Protein spots were quantified by densitometry, combined with a computer-assisted image analysis system. Immunohistochemistry was performed to analyze the expression pattern of major HSPs and beta-tubulin in testis. One isoform of HSP 90 (HSP 90 alpha), two isoforms of HSC 70 (HSC 70a and HSC 70c), one isoform of HSP70 (HSP 70e), and tubulin increased after sexual maturation (P<0.05). A testis-specific HSP70 (P70t) was markedly increased in the testes of sexually mature boars. Meanwhile, levels of actin and some isoforms of HSPs including 60 kDa HSP remained similar in both groups. These observations were further confirmed by immunohistochemistry; therefore, the upregulation of protein expression in the adult testis could be attributed to a higher level of protein expression and the number of cells that were HSPs-positive already resided in the immature testis. The differential expression of major HSPs suggested that they may be important in porcine spermatogenesis.  相似文献

Expression of antisense RNA against eukaryotic translation initiation factor 4E (eIF-4E) in HeLa cells causes a reduction in the levels of both eIF-4E and eIF-4 gamma (p220) and a concomitant decrease in the rates of both cell growth and protein synthesis (De Benedetti, A., Joshi-Barve, S., Rinker-Schaffer, C., and Rhoads, R. E. (1991) Mol. Cell Biol. 11, 5435-5445). The synthesis of most proteins in the antisense RNA-expressing cells (AS cells) is decreased, but certain proteins continue to be synthesized. In the present study, we identified many of these as stress-inducible or heat shock proteins (HSPs). By mobilities on sodium dodecyl sulfate-polyacrylamide gel electrophoresis and by reactivity with monoclonal antibodies generated against human HSPs, four of these were shown to be HSP 90, HSP 70, HSP 65, and HSP 27. The steady-state levels of HSP 90, 70, and 27 were elevated in relation to total protein in AS cells. Pulse labeling and immunoprecipitation indicated that HSP 90 and HSP 70 were synthesized more rapidly in AS cells than in control cells. The accelerated synthesis of HSPs in the AS cells was not due, however, to increased mRNA levels; the levels of HSP 90 and 70 mRNAs either remained the same or decreased after induction of antisense RNA expression. Actin mRNA, a typical cellular mRNA, was found on high polysomes in control cells but shifted to smaller polysomes in AS cells, as expected from the general decrease in translational initiation caused by eIF-4E and eIF-4 gamma depletion. HSP 90 and 70 mRNAs showed the opposite behavior; they were associated with small polysomes in control cells but shifted to higher polysomes in AS cells. These results demonstrate that HSP mRNAs have little or no requirement in vivo for the cap-recognition machinery and suggest that these mRNAs may utilize an alternative, cap-independent mechanism of translational initiation.  相似文献

OBJECTIVE: To investigate p53 protein expression and proliferative activity in imprints of tumor biopsies from superficial transitional cell carcinoma of the bladder in relation to the histologic grade of malignancy and recurrence status. STUDY DESIGN: The study group consisted of 70 cases of superficial transitional cell carcinoma of the bladder. In order to investigate p53 protein expression and Ki-67 expression, an immunocytochemical avidin-extravidin complex technique was performed using monoclonal antibodies p53 D0-7 and proliferating cells correspondingly. RESULTS: Thirty-seven percent of superficial transitional cell carcinoma cases showed positive expression of p53 protein. No correlation was found between p53 protein expression and grade of malignancy (P = .45). p53 Protein expression was statistically correlated with a high Ki-67 labeling index (LI) (P < .001) and recurrence status (P < .001). Forty-seven percent of cases showed a Ki-67 LI > 25%. No correlation was found between a high Ki-67 LI and grade of malignancy (P = .703). A significant difference in high Ki-67 LI between recurrent and nonrecurrent tumors of the same grade (P < .001) and between recurrent and nonrecurrent tumors was found independently of grade (P < .001). CONCLUSION: These results on cytologic material could provide useful information on the biologic behavior of superficial transitional cell carcinoma of the bladder at the time of diagnosis.  相似文献

Background  

Heat shock proteins (HSPs), including mainly HSP110, HSP90, HSP70, HSP60 and small HSP families, are evolutionary conserved proteins involved in various cellular processes. Abnormal expression of HSPs has been detected in several tumor types, which indicates that specific HSPs have different prognostic significance for different tumors. 肺癌中金属硫蛋白的表达及其与细胞增殖、凋亡的关系   总被引:4,自引:0,他引:4  
目的 探讨肺癌中金属硫蛋白 (Metallothionein ,MT)的表达及其与临床病理学参数、Ki 6 7、P5 3和凋亡的关系。方法 免疫组织化学SP法对 5 6例石蜡包埋的肺癌组织分别进行MT、Ki 6 7、P5 3蛋白的检测并对 11例石蜡包埋的癌旁正常支气管组织进行MT蛋白的检测。TUNEL法对 19例MT蛋白阴性和弱阳性表达及 2 8例MT蛋白中等阳性和强阳性表达的肺癌标本进行原位细胞凋亡检测。结果 MT蛋白在肺癌中的表达高于其在正常支气管组织中的表达 ,但无显著差异 (P =0 5 6 1) ;肺癌中MT蛋白的表达与患者的性别、肿瘤的组织学类型及分化程度有关 (均为P <0 0 5 ) ,与患者的年龄、肿瘤大小、TNM分期和淋巴结转移与否无关 (均为P >0 0 5 ) ,与Ki 6 7蛋白的表达呈正相关 (r=0 2 78,P <0 0 5 ) ,与凋亡呈负相关 (r=- 0 319,P <0 0 5 ) ,与P5 3蛋白的表达无显著相关 (r=0 16 7,P >0 0 5 )。结论 MT蛋白的表达与肺癌的分化程度、Ki 6 7及凋亡显著相关 ,MT可作为判断肺癌细胞分化程度、增殖活性及凋亡等的指标。  相似文献

目的检测P27kip1与EGFR在星形胶质瘤中的表达,探讨其对增殖活性的影响,为治疗提供一定的依据.方法采用免疫组化S-P法检测星形胶质瘤EGFR、P27kip1的表达,每一例肿瘤用ki-67LI表示其增殖活性,应用SAS8.0统计软件进行结果分析.结果 1.P27kip1表达水平与星形胶质瘤病理分级负相关(rs=-0.487,P<0.0001).2.对EGFR、P27kip1及Ki-67LI联合分析显示:P27kip1( )EGFR(-)组的ki-67LI(5.0%)明显低于P27kip1( )EGFR( )、P27kip1(-)EGFR( )、P27kip1(-)EGFR(-)组(10.4%、10.4%、10.8%)(P<0.05).结论 1.P27kip1在星形胶质瘤的发生、发展过程中可能起抑制作用.2.P27kip1、EGFR对星形胶质瘤增殖活性起相互拮抗的作用.  相似文献

目的:分析新疆维吾尔族妇女宫颈鳞癌组织中Smad7和Ki-67表达及相关性。方法:采用免疫组化SP法检测宫颈炎组织88例(汉族42例,维吾尔族46例)以及宫颈鳞癌组织120例(汉族50例,维吾尔族70例)中Smad7、Ki-67蛋白的表达。结果:Smad7、Ki-67蛋白在宫颈鳞癌中的阳性表达率(78.3%、76.7%)均分别显著高于宫颈炎组织(15.9%、23.9%),差异有统计学意义(P0.05);维吾尔族妇女宫颈鳞癌中Smad7的阳性表达率(87.1%)显著高于汉族(66%)(P0.05)。维吾尔族宫颈鳞癌患者中Smad7、Ki-67的阳性表达率呈显著正相关性(x2=1.93,r=0.138,P0.05)。结论:宫颈鳞癌组织中Smad7、Ki-67蛋白的阳性表达率显著高于宫颈炎组织,宫颈鳞癌中Smad7蛋白的表达存在民族差异,Smad7、Ki-67等多种蛋白的联合检测可能会间接提高新疆维吾尔族妇女宫颈鳞癌诊断的准确度和精确度。  相似文献

Heat shock proteins (HSPs) expression is commonly used as indicators of cellular stress in animals. However, very little is known about either the expression patterns of HSPs or their role in the stress-tolerance phenomenon in early life stages of fish. To this end, we examined the impact of food-deprivation (12 h), reduced oxygen levels (3.5 mg/L for 1 h) and heat shock (HS: + 5 °C for 1 h) on HSP70 and HSP90 protein expression in early life stages of the gilthead sea bream (Sparus aurata), a warm-water aquaculture species. Also, we investigated HSP70 and HSP90 response to food-deprivation (7 days) in early life stages of rainbow trout (Oncorhynchus mykiss), a cool-water aquaculture species, and the tolerance of this larvae to heat shock (either + 5 or + 10 °C for 1 h). Our results clearly demonstrate that food-deprivation enhances HSP70 and HSP90 protein expression in larvae of both species. In gilthead sea bream larvae, the stressors-induced HSP70 and HSP90 (only in the reduced oxygen group) protein expression returned to unstressed levels after 24 h recovery. In fed trout larvae, a + 5 °C heat shock did not elevate HSP70 and HSP90 expression, whereas 100% mortality was evident with a + 10 °C HS. However, food-deprived trout larvae, which had higher HSP70 and HSP90 protein content, survived HS and showed HS-dependent increases in HSP70, but not HSP90 expression. Overall, HSP70 and HSP90 protein expression in early life stages of fish have the potential to be used as markers of nutritional stress, while elevation of the tissue HSPs content may be used as a means to increase stress tolerance during larval rearing.  相似文献

Earlier studies identified the role of bta-mir-2898 in bovine. Our earlier study identified that, bta-mir-2898 can be over expressed in crossbred cattle during heat stress. Nevertheless the differential expression of bta-mir-2898 among native vs crossbred cattle during summer stress along with it's correlation with different heat shock proteins (HSPs) is not yet studied. In the present context, we studied the differential expression of bta-mir-2898 among Frieswal (Bos indicus x Bos taurus) and Sahiwal (Bos indicus) breeds of cattle during a range of environmental air temperatures and further investigated the correlation of bta-mir-2898 with different HSPs (HSP70, HSP90, HSP60. HSF, HSPB8 and HSP27). It was observed that, at peak air temperature the relative miRNA expression level (p < 0.05) of bta-mir-2898 was 3.4 ± 0.41 and 0.79 ± 0.22 among Frieswal and Sahiwal, respectively. We also observed significant levels (p < 0.05) of mRNA abundance of HSP70, HSP90, HSPB8 and HSP27 among the breeds. In all the cases Sahiwal found to exhibited higher level of HSPs in comparison to Frieswal. Studies revealed that the expression profile of bta-mir-2898 was negatively correlated with the expression of all the HSPs during thermal stress in post anti-mir2898 treated PBMC invitro cultured model originated from both Frieswal and Sahiwal cattle breeds. However, significantly (p < 0.05) higher negative correlations were observed between bta-mir-2898 and HSP70, HSP60 and HSPB8. Present findings highlighted the preliminary role of overexpressed bta-mir-2898 in cattle during thermal stress and its impact on different heat shock proteins.  相似文献

The aim of the present study was to examine the effect of selenium (Se) deficiency on the expression of selenoproteins in chicken muscular stomach and to detect the correlation of selenoproteins with muscular stomach injuries. One-day-old broiler chickens were maintained for 55 days on a normal diet (0.2 mg/kg) or a Se-deficient diet (0.033 mg Se/kg). The expression levels of 25 selenoproteins, heat shock proteins (HSPs), and inflammatory factors were then examined by real-time PCR. Following this, the correlation between selenoproteins, HSPs, and inflammatory factors was analyzed by principal component analysis (PCA). The results showed that Se deficiency decreased the expression of 25 selenoproteins (P < 0.05), but increased the expression of HSP27, HSP40, HSP60, HSP70, and HSP90, and NF-κB, iNOS, TNF-α, COX-2, and HO-1 (P < 0.05). Selenoproteins showed a high negative correlation with HSPs and inflammatory factors. Thus, the results suggested that Se deficiency induced muscular stomach injuries by decreasing the expression of selenoproteins. In addition, selenoproteins play an important role in regulating HSPs and inflammatory response. The muscular stomach is a key target of Se deficiency and may play a special role in response to Se deficiency.  相似文献

Mammalian cells exhibit increased sensitivity to hyperthermic temperatures of 38-43 degrees C after an acute high-temperature heat shock; this phenomenon is known as the stepdown heating (SDH) effect. We characterized the SDH effect on (1) the synthesis of major heat shock proteins, HSP110, 90, 72/70, 60 (35S-amino acids label), (2) on heat-induced protein glycosylation (3H-D-mannose label), and (3) on thermotolerance expression, using cell survival as an endpoint. Partitioning of label between soluble and insoluble cell fractions was separately examined. Synthesis of high molecular weight HSPs (HSP110, 90, and 72/70) was increased both by acute (10 min, 45 degrees C) and chronic (1-6 h, 41.5 degrees C) hyperthermia, primarily in the soluble cytosol fraction. SDH (10 min, 45 degrees C + 1 to 6 h, 41.5 degrees C) completely inhibited labeling of HSP110, partially inhibited HSP90 labeling, and had virtually no effect on HSP72/70 synthesis, when compared with chronic hyperthermia alone. At the cell survival level, SDH increased sevenfold the rate of cell killing at 41.5 degrees C, but reduced the expression of thermotolerance by only a factor of two. This suggests that SDH sensitization did not result from changes in HSP72/70 synthesis, nor solely from inhibition of thermotolerance. 35S-labeled HSP60 and HSP50 were found primarily in the cellular pellet fraction after both acute and chronic hyperthermia. SDH completely inhibited 35S-labeling of both HSP60 and HSP50. Labeling of GP50 with 3H-D-mannose was also completely inhibited by SDH. Moreover, SDH progressively reduced N-acetylgalactosaminyl-transferase activity. The data demonstrate that heat sensitization by SDH is accompanied by complex and selectively inhibitory patterns of HSP synthesis and protein glycosylation. Profound inhibition of HSP110, HSP60, and HSP50/GP50 labeling suggests that these may be associated with mechanisms of SDH sensitization.  相似文献

Cultured mouse lymphocytes respond to a brief incubation at an elevated temperature (41-43 degrees C) with the new and (or) enhanced synthesis of a select group of polypeptides (known as heat-shock proteins, HSPs) having relative molecular masses of 110, 100, 90, 70, and 65 kilodaltons (kDa). Expression of these HSPs is dependent on new RNA synthesis. Because the synthesis of any particular HSP is dependent on the temperature and the length of time cells remain at a particular elevated temperature, synthesis of each HSP is not necessarily coordinated with the synthesis of the other HSPs. Cultured mouse lymphocytes treated with arsenite or ethanol exhibit new and (or) enhanced synthesis of HSPs with molecular masses of 110, 90, 70, and 65 kDa but do not exhibit enhanced synthesis of the 100-kDa HSP. Short-term concurrent exposure of mouse lymphocytes to an elevated temperature and a level of ethanol, which individually do not induce detectable HSP synthesis, results in the pronounced synthesis of HSPs similar to those seen following exposure to higher levels of either stress applied separately. Thus, in this study we demonstrate that hyperthermia and ethanol stress can act synergistically to affect a dramatic change in the gene expression of mouse lymphocytes.  相似文献

Neuroendocrine tumours of the gastrointestinal tract are relatively uncommon neoplasms with, in spite of their characteristic morphology, relatively unpredictable biological behaviour. In some sites, notably the appendix, these tumours are largely benign whereas at other localisations, such as the small bowel, metastases occur and the outcome is less favourable. Given the lack of discriminative power of histological parameters, immunohistochemical parameters have been proposed. Of these the Ki-67 index, as an indicator of proliferative activity, has shown some promise. In order to assess their proliferative activity and the potential contribution of this parameter to defining biological behaviour, we performed Ki-67 immunostaining of a series of 64 well differentiated neuroendocrine tumours of the gut (stomach, small bowel, appendix, colon and rectum). Ki-67 labeling index, based upon counting of up to 5000 cells, ranged between 0 and 6.1%. No difference was found according to age, gender, size, location or TNM classification. Ki-67 labeling index of midgut endocrine tumours of long term surviving patients did not differ from patients that died. We conclude that Ki-67 labeling index as an indicator of proliferative activity of well differentiated neuroendocrine tumours of the digestive tract does not correlate with size nor site nor stage. Even though only small numbers of tumours could be analysed, which hampered appropriate statistical analysis, it seems unlikely that proliferative activity has potential as an independent prognostic parameter for this type of tumour.  相似文献

Few studies have focused on the expression of heat shock proteins (HSPs) after chronic heat stress. The objective of this study was to investigate the effect of chronic high temperature–humidity index treatment on the expressions of HSP60, HSP70, HSP90, HSPA2 and HSC70, in the Rex rabbit testis and the expressions of these proteins after recovery from the chronic heat shock. Thirty mature male rabbits of the same age were randomly divided into three groups: control, heat stress, and recovery. The western blot results showed that the expressional levels of HSP60, HSP90, and HSC70 increased significantly and HSPA2 was elevated slightly after a 9-week heat treatment. HSP70 was absent in the control testis and had a high level of expression after heat stress. All of these proteins partially reverted back to normal levels after a 9-week recovery. The immunohistochemical results indicated that the expression patterns of HSP60, HSP90, HSPA2, and HSC70 did not change.  相似文献
