Euplotes is diversified into dozens of widely distributed species that produce structurally homologous families of water‐borne protein pheromones governing self‐/nonself‐recognition phenomena. Structures of pheromones and pheromone coding genes have so far been studied from species lying in different positions of the Euplotes phylogenetic tree. We have now cloned the coding genes and determined the NMR molecular structure of four pheromones isolated from Euplotes petzi, a polar species which is phylogenetically distant from previously studied species and forms the deepest branching clade in the tree. The E. petzi pheromone genes have significantly shorter sequences than in other congeners, lack introns, and encode products of only 32 amino acids. Likewise, the three‐dimensional structure of the E. petzi pheromones is markedly simpler than the three‐helix up‐down‐up architecture previously determined in another polar species, Euplotes nobilii, and in a temperate‐water species, Euplotes raikovi. Although sharing the same up‐down‐up architecture, it includes only two short α‐helices that find their topological counterparts with the second and third helices of the E. raikovi and E. nobilii pheromones. The overall picture that emerges is that the evolution of Euplotes pheromones involves progressive increases in the gene sequence length and in the complexity of the three‐dimensional molecular structure.  相似文献

Oregon‐R, +3, and crossbred strains of Drosophila melanogaster were tested for their response to selection for abdominal bristle number. Various subsidiary tests, consisting of heritability estimations, testing for lethal second and third chromosomes, and chromosome assays were conducted on the selection replicates, which had undergone 14 generations of selection. Evidence showed that a plateau which occurred very early in the +3 high selection replicates was due to fixation of a few additive genes with large effects, thus accounting for the low phenotypic and additive genetic variance, the slight regression in abdominal bristle number on relaxation of selection, the absence of directional dominance, and the low frequency of recessive lethals.

High frequencies of second and third chromosome lethals were found in the Oregon‐R high and low replicates and in the +3 low replicates. That these lethals were not selected for heterozygote superiority for extreme bristle effect was indicated by the slight regression of these replicates on relaxation of selection, and by the absence of high, fluctuating phenotypic variances.

From chromosome assays it appears that the two parental strains had different arrays of genes affecting high bristle number, with these genes located mostly in chromosome II in the Oregon‐R high line but in chromosome III in the +3 high line. In the Crossbred high line, high bristle factors were located in both the second and third chromosomes. The low bristle factors were located mainly in the second chromosome in all three low selection lines.

It appears that the original cross had combined different genes favouring high bristle number, thus allowing greater response in the Crossbred high selection line. The same did not occur for low selection; the response from the Crossbred low line was similar to that of the parental low lines, suggesting that the gene arrays affecting low bristle number in the two original populations were comparable.  相似文献

Genes encoding the enzyme methionine sulfoxide reductase type B, specific to the reduction of the oxidized methionine-R form, were characterized from the expressed (macronuclear) genome of two ecologically separate marine species of Euplotes, i.e. temperate water E. raikovi and polar water E. nobilii. Both species were found to contain a single msrB gene with a very simple structural organization encoding a protein of 127 (E. raikovi) or 126 (E. nobilii) amino acid residues that belongs to the group of zinc-containing enzymes. Both msrB genes are constitutively expressed, suggesting that the MsrB enzyme plays an essential role in repairing oxidative damages that appear to be primarily caused by physiological cell aging in E. raikovi and by interactions with an O2 saturated environment in E. nobilii.  相似文献

ABSTRACT. Studies of the bristle (dorsal) cilia of Euplotes minuta. E. aediculatus, and Stylonychia mytilus by light and electron microscopy indicate that these cilia do not beat metachronously in any of the species. The bristle cilia in Stylonychia may beat actively, but those in Euplotes stand erect or are bent in different directions with the flow of water. The duration and degree of bending appear correlated with the duration and velocity of the water current. The fine structure of the bristle complex is similar in both Euplotes species and like other reports of Euplotes in the literature. The complex consists of paired kinetosomes, the anterior bearing a short cilium containing four to six rows of fibrous balls (lasiosomes) oriented along the anterior surface of the axoneme, the posterior lacking a cilium but with a small cap. Microtubular ribbons are associated with the paired kinetosomes, and a collar with a pronounced alveolar ring underneath the pellicular membrane tightly surrounds the cilium at the opening of the bristle pit. The bristle complex in S. mytilus differs from that of Euplotes and other hypotrichs in that it has a single kinetosome in interphase cells and, attached to the kinetosome, a prominent fibrous structure (parakinetosomal body). Microtubules are attached to the parakinetosomal body. As in Euplotes, the bristle unit is surrounded by mucocyst-like organelles (ampules). Observations of behavior and fine structure suggest that the dorsal bristles may be sensory, perhaps responding to stimuli from water currents, although other functions are possible, too.  相似文献

Redundant encoding of local and global spatial cues is a common occurrence in many species. However, preferential use of the each type of cue seems to vary across species and tasks. In the current study, pigeons (Columba livia) were trained in three experiments on a touch screen task which included redundant local positional cues and global spatial cues. Specifically, pigeons were required to choose the middle out of three choice squares, such that the position within the array provided local information and the location on the screen provided global information. In Experiment 1, pigeons were trained and tested on vertically aligned arrays. In Experiment 2, pigeons were trained and tested on horizontally aligned arrays, and in Experiment 3, pigeons were trained and tested with vertical, horizontal and diagonally aligned arrays. The results indicate that preference for cue type depends upon the type of spatial information being encoded. Specifically, on vertical and diagonally aligned arrays, pigeons preferred global cues, whereas on horizontally aligned arrays, pigeons preferred local cues.  相似文献

Ventoplagia gen. n. is described, with the type species Ventoplagia brevirostris sp. n. The frontal bristles extending only to the base of the pedicel, 2+3 dorsocentral bristles, 0+2 intraalar bristles, the absence of prealar bristle, the short and fine anepimeral (pteropleural) bristle, the scutellum without lateral bristles, and the welldeveloped posteroventral bristle of the hind tibia indicate that the new genus belongs to tribe Minthoini. Ventoplagia gen. n. is closely related to the genus Palmonia Kugler. The characters distinguishing these genera are given.  相似文献

The chromatographic pigment arrays of nine diploid species (G. arboreum, G. anomalum, G. herbaceum, G. stocksii, G. sturtii, G. thurberi, G. gossypioides, G. raimondii and G. klotzschianum) were studied. Among the Old World cottons, G. sturtii of Australia was very different from the species analyzed. The two species of the Herbacea section (G. herbaceum and G. arboreum) were found to have very similar pigment arrays. Both G. anomalum and G. stocksii were more like the Herbacea species than any other species in the genus, but both G. anomalum and G. stocksii had unique pigment characteristics. Although the evidence obtained so far from pigmentation patterns suggests that some pairs of species are closely related, the pigment arrays do not support the classification of the New World diploids into more than one section. From analysis of pigments of interspecific hybrids and their parents, it was found that with a hybrid and one parent species the pigment array of the other parent species could be predicted. Using this approach, the pigment arrays of three New World diploid species were predicted.  相似文献

Francisella are facultative intracellular bacteria causing severe disease in a broad range of animals. Two species are notable: Francisella tularensis, the causative organism of tularemia and a putative warfare agent, and Francisella noatunensis, an emerging fish pathogen causing significant losses in wild and farmed fish. Although various aspects of Francisella biology have been intensively studied, their natural reservoir in periods between massive outbreaks remains mysterious. Protists have been suspected to serve as a disguised vector of Francisella and co-culturing attempts demonstrate that some species are able to survive and multiply within protozoan cells. Here, we report the first finding of a natural occurrence of Francisella sp. as a protist endosymbiont. By molecular and morphological approaches, we identified intracellular bacteria localized in a strain of the marine ciliate Euplotes raikovi, isolated from the coast of Adriatic Sea. Phylogenetic analysis placed these endosymbionts within the genus Francisella, in close but distinct association with F. noatunensis. We suggest the establishment of a novel subspecies within F. noatunensis and propose the cytoplasmatic endosymbiont of E. raikovi as “Candidatus F. noatunensis subsp. endociliophora” subsp. nov.  相似文献

Mesosternal (MS) bristles in Drosophila are a pair of machrochaetae found at the sternal end of the sternopleural (STP) microchaetae, and are thought to be invariable. In a closely related drosophilid genus, Zaprionus, their number is four and, in contrast to Drosophila, they show interspecific and intraspecific variability. The genetic basis of MS bristle number variability was studied in Z. indianus, the only cosmopolitan species of the genus. The trait responded rapidly to selection and two lines were obtained, one lacking any bristles (0-0) and the other bearing the normal phenotype (2-2). Other symmetrical phenotypes, (1-1) and (3-3), could also be selected for, but with lesser success. By contrast, STP bristle number did not vary significantly between the two lines (0-0) and (2-2), revealing its genetic independence from MS bristle number. Reciprocal crosses between these two lines showed that MS bristle number is mainly influenced by a major gene on the X chromosome (i.e. F1 males always resembled their mothers) with codominant expression (i.e. heterozygous F1 females harboured an average phenotype of 2 bristles). However, trait penetrance was incomplete and backcrosses revealed that this variability was partly due to genetic modifiers, most likely autosomal. The canalization of MS bristle number was investigated under different temperatures, and the increased appearance of abnormal phenotypes mainly occurred at extreme temperatures. There was a bias, however, towards bristle loss, as shown by a liability (developmental map) analysis. Finally, when ancestral and introduced populations were compared, the latter were far less stable, suggesting that genetic bottlenecks may perturb the MS bristle number canalization system. MS bristle number, thus, appears to be an excellent model for investigating developmental canalization at both the quantitative and the molecular level.  相似文献

The feeding apparatus of Kalablepharis ovalis (isolated from a freshwater impoundment in Colorado) and Katablepharis clone G-2 (isolated from the littoral of the Black Sea near Yalta in the Crimea) consists of inner and outer oval-shaped arrays of microtubules that begin at the anterior end of the cell and pass into the posterior of the cell. Each array of microtubules contains groups of microtubules with two to eight microtubules per group depending on the position of the array in the cell. A specialized area of the plasma membrane, the mouth, occurs at the anterior end of the cell. The mouth is oval with the long axis oriented dorsoventrally and consists of a raised ridge surrounding a central depression. The anterior end of the microtubules of the inner and outer arrays supports the raised ridge of the mouth. In freeze-fracture replicas, the protoplasmic face of the plasma membrane contains intramembrane particles on the raised ridge of the mouth. Three small membrane-cisternae occur on the protoplasmic side of the plasma membrane in the area of the mouth. Katablepharis clone G-2 also has five or six additional large membrane-cisternae associated with the inner microtubular array in the anterior portion of the cell. These larger membrane-cisternae do not occur in K. ovalis. Vesicles with electron-dense contents occur in association with the microtubular arrays. Katablepharis ovalis has a second type of vesicle containing a single-membrane profile associated with the microtubule arrays. The structure of the microtubular arrays in Katablepharis is compared with similar structures in suctorian ciliates and dinoflagellates.  相似文献

The evolutionary history of a 160-bp tandem satellite array, originally described from Microtus chrotorrhinus and called MSAT-160, was examined in related species of arvicolid rodents by sequence analyses, quantitative dot blotting, and Southern blotting. Results indicate that MSAT-160 is present in 12 of the 20 species and subspecies of Microtus assayed, but not in species belonging to any of the eight other genera examined. DNA from each species containing MSAT-160 was digested with 12 restriction endonucleases and restriction patterns were obtained reflecting the variable extent of homogenization of any given variant in different species. For example, with MboI digestion, M. chrotorrhinus produced a type A ladder pattern where most monomers contain the restriction site, M. ochrogaster generated a type B pattern where most monomers lack the site, and M. agrestis yielded a pattern intermediate between the A and B types. Further, dot blotting revealed copy-number differences between species. These findings indicate that changes in the periodic structure and amount of satellite DNA have occured since these species last shared a common ancestor. In addition, various species-pacific patterns were documented, illustrating that mechanism other than genomewide homogenization, such as stochastic mutation, out-of-register crossing over, deletion, and random amplification also play a role in structuring tandem arrays. Stochastic mutation and homogenization rates in satellite DNA, levels of species diversity, and magnitudes of chromosomal divergence differ significantly in Microtus, Mus, and Ctenomys, the three rodent lineages examined.  相似文献

Numerous strains of different mating types of the marine ciliate Euplotes raikovi have been found to be autonomous excreters into the surrounding medium of specific mating-inducing factors (gamones) (Luporini, P et al., J exp zool 226 (1983) 1 [9]). The gamone from the mating type represented by strain 13 has been isolated and identified as a glycoprotein with a molecular weight (MW) of about 12 kD and a pI of 4. It has been termed euplomone r 13. At a concentration of 3 × 10−12 M, euplomone r 13 specifically induces cells of a complementary mating type to unite in conjugation within 2 h.  相似文献

The chromosomal arrangement of different transgenic repeat arrays inserted at various chromosomal positions was tested by FISH in Arabidopsis 2C leaf and root nuclei. Large lacO (∼10 kb) but not tetO (4.8 kb) or small lacO (∼2 kb) arrays were, in general, more often spatially associated with heterochromatic chromocenters (CC) than flanking regions (that either overlap the array insert position or are between 5 and 163 kb apart from the insert site). Allelic and ectopic pairing frequencies of lacO arrays were significantly increased only in nuclei of lines with two large lacO arrays inserted at different positions on the same chromosome arm. Within the same lines, root nuclei showed a significantly lower increase of pairing frequencies at the insert position compared to leaf nuclei but still a higher frequency than in the wild-type situation. Thus, the frequencies of homologous pairing and association with heterochromatin of transgenic repeats may differ with the construct, the chromosomal insertion position, the cell type and with the number and repetitiveness of inserts. Strong CpG methylation is correlated with a high frequency of homologous pairing at large repeat array loci in somatic cells but has no impact on their association with CCs. These results show that single low-copy arrays apparently do not alter interphase chromatin architecture and are more suitable for chromatin tagging than multiple high copy arrays.  相似文献

Summary The arrangement of bristles on a leg segment of the fruitflyDrosophila melanogaster was studied in various mutants that have abnormal numbers of bristles on this segment. Eighteen mutations at six different genetic loci were analyzed, plus five double or triple mutant combinations. Recessive mutations at theachaete-scute locus were found to affect distinct groups of bristles:achaete mutations remove mechanosensory bristles, whereasscute mutations remove mainly chemosensory bristles. Mechanosensory bristles remain uniformly spaced along the longitudinal axis unless their number decreases below a certain threshold, suggesting that spacing is controlled by cell interactions that cannot function when bristle cells are too far apart. Above a certain threshold, bristle spacing and alignment both become irregular, perhaps due to excessive force from these same interactions. Chemosensory bristles occupy definite positions that are virtually unaffected by removal of individual bristles from the array. Extra chemosensory bristles develop only near the six normal sites. At two of the six sites the multiple bristles tend to exhibit uniform longitudinal spacing — a property confined to mechanosensory bristles in wild-type flies. To explain the various mutant phenotypes the following scheme is proposed, with different mutations directly or indirectly affecting each step: (1) spots and stripes are demarcated within the pattern area, (2) one bristle cell normally arises within each spot, multiple bristle cells within each stripe, (3) incipient bristle cells inhibit neighboring cells from becoming bristle cells, and (4) the bristle cells within each stripe become aligned to form rows and then repel one another to generate uniform spacing.  相似文献

Responses of three locally endemic (Eucalyptus brevistylis, Eucalyptus jacksonii and Eucalyptus guilfoylei) and three co‐occurring regional eucalypts (Eucalyptus marginata, Eucalyptus diversicolor and Corymbia calophylla) to moderate‐ and high‐intensity fires were examined in granitic terrain of the Tingle Mosaic, south‐western Australia. Significant associations between diameter distributions and community type (CT) for each species (P < 0.001) suggest that fire response will also vary according to the habitat/fire interaction. None of the species were fire sensitive, although responses differed both within and between species, and with CT. All species examined predominately consisted of several cohorts of regeneration within a forest stand. Each species had thick bark and re‐sprouted from crown epicormics following 100% scorch of the mature tree. The quantity and type of regeneration in relation to gaps created by individual dead trees following fire differed between species; for example, E. guilfoylei regeneration was strongly associated with gaps, and C. calophylla with non‐gaps. However, regeneration of the two tall open‐forest species, E. jacksonii and E. diversicolor were not most associated with either gaps or non‐gaps. The very low levels of regeneration of E. brevistylis following fire and the high proportion of stems of E. jacksonii that were hollow butted (40% of stems > 1 m DBHOB) may be factors associated with narrow endemism of these species and may affect the vulnerability of these eucalypts to fire. The interaction of seed availability, intense fires and subsequent rainfall may be critical in the long term survival of these species. Eucalyptus guilfoylei, by contrast, appears well adapted to the increasing levels of disturbance likely in the region where these species occur. The vulnerability of a locally endemic species in a fire‐prone environment is likely to reflect differences to the prevailing adaptations of the dominant species rather than an inherent ability of the species to survive or respond. Management regimes must account for variations in species responses to fire in different CTs if the long‐term survival chances of local endemics are to be enhanced.  相似文献

We assessed the combined effects of varying the relative density and the relative floral morphological complexity of plant species on the behaviour of their bumblebee pollinators. Three species of bumblebee (Bombus pascuorum, B. terrestris and B. hortorum) were observed foraging on experimental arrays consisting of pair-wise combinations of four plant species: Borago officinalis, Phacelia tanacetifolia (both with simple flowers), Antirrhinum majus and Linaria vulgaris (both with complex flowers). Plant arrangements consisted of either two simple-flower species, a simple with a complex species or two complex species. The number of plants in each array was constant, while the frequency of each species was manipulated so that it was either rare, equal or common compared with its competitor. Contrary to predictions, rare plants were actually at an advantage in terms of the number of bees attracted per plant. However, rare plants were at a disadvantage in terms of pollen wastage because foragers more often went to a flower of another species after visiting a rare plant. The behaviour of bees on each plant species was further affected by plant floral complexity and the identity of the other species in the array. The three bumblebee species were markedly different in their foraging behaviour and in their responses to varying floral density and complexity. Each species preferred particular flower species. The development of the female gametophytes of Epipactis atrorubens, E. helleborine, and E. palustris have been investigated using confocal scanning laser microscopy. These species have T-tetrads and eight-nucleate female gametophytes in common. The development of the female gametophyte is monosporic in accordance with the Polygonum type. Furthermore, the outer integument grows slowly and reaches to the middle of the inner integument, when the female gametophyte is mature. In the first two species, T-tetrads develop prior to anthesis, which may be correlated with autogamy. The development of the female gametophyte of E. palustris appears to be similar in plants from different localities.  相似文献

  • Three synchronopatric Cactaceae species, Echinopsis rhodotricha, Harrisia balansae and Praecereus saxicola, have mostly nocturnal anthesis and similar flowers, characteristics that motivated us to perform a comparative study of reproductive ecology.
  • Reproductive phenology was sampled monthly from December 2014 to November 2015. We describe floral biology, breeding system via pollination treatments and evaluate floral visitors from focal and filming observations. Pollen grains found on moth proboscis were compared among cactus species under light microscopy. We used fluorescent dye particles to test intra‐ and interspecific pollen flow.
  • These three species have extended flowering with greater intensity in the wet season, causing high overlap. They have white and hypocrateriformis flowers that open at twilight or nightfall and last about 15 h. H. balansae seems to be self‐incompatible, while E. rhodotricha presented self‐compatibility. P. saxicola presented self‐fertility, but most of the population seems to be self‐incompatible. We suggest sphingophily for the three species, but only P. saxicola was visited by Manduca rustica (Sphingidae). However, we observed pollen grains of all three species on the proboscis of moths, especially M. rustica and M. sexta. Prolonged anthesis allowed bees (herein considered as secondary pollinators) to visit flowers of E. rhodotricha and P. saxicola.
  • It can be concluded that the studied species share nocturnal and diurnal pollinators, suggesting interspecific pollen flow, which, however, could not be detected with fluorescent dye particles. In view of the low frequency of primary pollinators, it appears that these three species have different reproductive strategies, ensuring the fruiting and production of viable seeds.

Complex bristle types formed by species in the genus Mallomonas include those with helmet or lance-shaped apices. The ornamentation on each side of the helmet has been thought to be equivalent or symmetrical, whereas on a lance-shaped bristle an expanded portion folds over one side of the shaft to form an asymmetrical structure. We describe, for the first time, helmet bristles with a distinctly asymmetrical design, also formed by the folding of a siliceous membrane over one side of the helmet. We postulate that the asymmetrical helmet represents a structure that combines the formation of a symmetrical helmet and a lance-shaped design on the same bristle. Further, we report structurally similar asymmetrical helmet bristles, lance-shaped bristles and scales that are unambiguously assigned to Mallomonas asmundiae in Middle Eocene sediments from a maar lake in northern Canada, supporting the hypothesis that scale and bristle morphology in the Synurophyceae has undergone extensive prolonged evolutionary stasis. Given differences in scale morphology and the presence of asymmetrical helmet bristles, we transfer the North American endemic Mallomonas acaroides var. muskokana to the rank of species. Further, we formally describe Mallomonas dispar and M. lancea, fossil species with asymmetrical helmet bristles and lance-shaped bristles, respectively. The taxonomic and biogeographic significance of asymmetrical and lance-bearing bristles is discussed.  相似文献
