The microstructure of aptychi (bivalved calcareous coverings on lower jaws) of three genera of Late Cretaceous Ancyloceratina, Baculites, Polyptychoceras and Jeletzkytes is described for the first time on the basis of well-preserved and in situ material from the Western Interior of the USA and Hokkaido, Japan. Optical and scanning electron microscope observations of aptychi on polished median and cross-sections reveal some variation in their relative size, shape and microstructure among the three genera. The aptychus of Baculites is composed of two calcitic layers: one with tilted lamellae and the other one with horizontal lamellae, whereas those of Polyptychoceras and Jeletzkytes consist of a thin layer with horizontal lamellae. Comparison with aptychi (e.g. Laevaptychus) of Jurassic Ammonitina shows that the aptychi of Ancyloceratina differ from those of Jurassic Ammonitina in the smaller number of layers and the absence of a sponge-like structure. We propose for the first time growth models for a sponge-like aptychus of Jurassic Ammonitina and the lamellar aptychus of Cretaceous Ancyloceratina. The remarkable microstructural variation of aptychi observed in Mesozoic Ammonoidea is probably related to the diversity of their modes of feeding and the secondary function of the lower jaws as opercula.  相似文献

Aptychi are calcitic coverings on the outer surface of organic ammonite lower jaws. They are similar in shape to that of the corresponding ammonite apertures. This observation and additional features of many aptychi are in harmony with their former interpretation as protective opercula. We suggest that they served as opercula in addition to functioning as jaws. The primary function of the lower jaws was thus secondarily extended to that of protective shields when they acquired their calcitic covering, while as lower jaws their importance dwindled to that of a more passive abutment. To date, bivalve calcitic plates which cover the outer chitinous lamella of the lower jaws of Jurassic and Cretaceous ammonoids (aptychi sensu stricto) have been classified into several morphotypes (form genera) based on shape, surface sculpture and internal microstructure. However, previous works on aptychi microstructure focused mainly on thick morphotypes (e.g. Laevaptychus and Lamellaptychus), whereas little was investigated for thin Praestriaptychi. In this study, the microstructure of Praestriaptychi of Upper Oxfordian Perisphinctes and of recently discovered aptychi of aspidoceratid microconch Mirosphinctes (both belonging to the superfamily Perisphinctoidea) is described. These aptychi are compared with Laevaptychi of Upper Oxfordian macroconchs Euaspidoceras (a dimorphic counterpart of Mirosphinctes). This study demonstrated that the aptychi of Perisphinctes and Mirosphinctes differ from each other and from Laevaptychi in their microstructure, number of calcitic layers and in growth patterns. The aptychi of both aspidoceratid dimorphs are similar in terms of growth pattern, but differ in microstructure and the number of calcitic layers. Neither aptychi of Perisphinctes nor of Mirosphinctes have a tubular layer with a honeycomb surface pattern, which is typical for Laevaptychus. This indicates that aptychi were extremely diverse and their microstructure varied significantly not only within the superfamily, but even within a dimorphic pair of aspidoceratid ammonites. A lack of a tubular layer in Praestriaptychus indicates that it developed independently in Lamellaptychus of Haploceratoidea and Laevaptychus of Aspidoceratidae.  相似文献

The Scallop theorem states that reciprocal methods of locomotion, such as jet propulsion or paddling, will not work in Stokes flow (Reynolds number=0). In nature the effective limit of jet propulsion is still in the range where inertial forces are significant. It appears that almost all animals that use jet propulsion swim at Reynolds numbers (Re) of about 5 or more. Juvenile squid and octopods hatch from the egg already swimming in this inertial regime. Juvenile jellyfish, or ephyrae, break off from polyps swimming at Re greater than 5. Many other organisms, such as scallops, rarely swim at Re less than 100. The limitations of jet propulsion at intermediate Re is explored here using the immersed boundary method to solve the 2D Navier-Stokes equations coupled to the motion of a simplified jellyfish. The contraction and expansion kinematics are prescribed, but the forward and backward swimming motions of the idealized jellyfish are emergent properties determined by the resulting fluid dynamics. Simulations are performed for both an oblate bell shape using a paddling mode of swimming and a prolate bell shape using jet propulsion. Average forward velocities and work put into the system are calculated for Re between 1 and 320. The results show that forward velocities rapidly decay with decreasing Re for all bell shapes when Re<10. Similarly, the work required to generate the pulsing motion increases significantly for Re<10. When compared to actual organisms, the swimming velocities and vortex separation patterns for the model prolate agree with those observed in Nemopsis bachei. The forward swimming velocities of the model oblate jellyfish after two pulse cycles are comparable to those reported for Aurelia aurita, but discrepancies are observed in the vortex dynamics between when the 2D model oblate jellyfish and the organism. This discrepancy is likely due to a combination of the differences between the 3D reality of the jellyfish and the 2D simplification, as well as the rigidity of the time varying geometry imposed by the idealized model.  相似文献

Evolutionary constraints which limit the forces produced during bell contractions of medusae affect the overall medusan morphospace such that jet propulsion is limited to only small medusae. Cubomedusae, which often possess large prolate bells and are thought to swim via jet propulsion, appear to violate the theoretical constraints which determine the medusan morphospace. To examine propulsion by cubomedusae, we quantified size related changes in wake dynamics, bell shape, swimming and turning kinematics of two species of cubomedusae, Chironex fleckeri and Chiropsella bronzie. During growth, these cubomedusae transitioned from using jet propulsion at smaller sizes to a rowing-jetting hybrid mode of propulsion at larger sizes. Simple modifications in the flexibility and kinematics of their velarium appeared to be sufficient to alter their propulsive mode. Turning occurs during both bell contraction and expansion and is achieved by generating asymmetric vortex structures during both stages of the swimming cycle. Swimming characteristics were considered in conjunction with the unique foraging strategy used by cubomedusae.  相似文献

Morton, Nicol & Nixon, Marion 1987 07 15: Size and function of ammonite aptychi in comparison with buccal masses of modem cephalopods.
Previous impressions that the size of ammonite aptychi is unusually large, thereby posing a problem to their interpretation as part of the jaw apparatus, are shown to be incorrect. The relative length of the aptychus for at least the Jurassic ammonite groups studied, at approximately 15% of the length of the body chamber, is remarkably constant in different taxonomic groups and sizes. This is well within the range for buccal mass length as percentage of mantle length of living cephalopods, being most similar to Octopus and Sepia and much smaller than Nautilus. The height and width of aptychi relative to whorl height and width are larger but again ammonites are probably not significantly different from modern cephalopods. The size and design, with no obvious structures for biting or crushing, suggest that ammonites were adapted to a particular type of relatively unspecialised feeding in which mostly small animals were ingested, possibly with some external digestion. The large shovel-like lower jaw may have functioned like a scoop for collecting large quantities of water and small prey, and movement of the buccal complex with the upper jaw almost closed against the lower jaw could have expelled water while retaining captured prey. Calcification of aptychi may have been protective, but more likely acted to weight the buccal mass for nektobenthic feeding and to make it more rigid.  相似文献

We tested the hypothesis that a combination of coagulant and ballast could be efficient for removal of positively buoyant harmful cyanobacteria in shallow tropical waterbodies, and will not promote the release of cyanotoxins. This laboratory study examined the efficacy of coagulants [polyaluminium chloride (PAC) and chitosan (made of shrimp shells)] alone, and combined with ballast (lanthanum modified bentonite, red soil or gravel) to remove the natural populations of cyanobacteria collected from a shallow eutrophic urban reservoir with alternating blooms of Cylindrospermopsis and Microcystis. PAC combined with ballast was effective in settling blooms dominated by Microcystis or Cylindrospermopsis. Contrary to our expectation, chitosan combined with ballast was only effective in settling Cylindrospermopsis-dominated blooms at low pH, whereas at pH  8 no effective flocculation and settling could be evoked. Chitosan also had a detrimental effect on Cylindrospermopsis causing the release of saxitoxins. In contrast, no detrimental effect on Microcystis was observed and all coagulant-ballast treatments were effective in not only settling the Microcystis dominated bloom, but also lowering dissolved microcystin concentrations. Our data show that the best procedure for biomass reduction also depends on the dominant species.  相似文献

Filtration of ballast water was investigated as a means of minimizing the introduction of nonindigenous zooplankton and phytoplankton by ships visiting the North American Great Lakes-St. Lawrence Seaway system (GLSLSS). An automatic backwash screen filtration (ABSF) system with nominal filtration options of 25, 50 or 100 μm was mounted on the deck of an operating Seaway-sized dry bulk carrier, the MV Algonorth. Water was pumped through the ABSF with a deck mounted pump at 341 m3 hr−1 during routine ship operations in the GLSLSS, and effectiveness of the various screen pore sizes at removing taxonomic categories of zooplankton and phytoplankton was measured using matched treatment and control ballast tanks. The smallest pore sizes (25 and 50 μm) performed better than the 100 μm pore size at removing biological material. There was no difference in the filtration efficiency of the 25 and 50 μm screens relative to macro- or microzooplankton in these tests, but this result was probably due to low densities of macrozooplankton, and soft-bodied (aloricate) characteristics of the microzooplankton present. The 25 and 50 μm pore sizes were subjected to more controlled tests on board a stationary barge platform equipped with triplicate 700 L catchment bins moored in Duluth Harbor of Lake Superior. In these tests, filter pore size, organism size and rigidity influenced zooplankton removal efficiency by the ABSF. The 25 μm screen reduced both macrozooplankton and microzooplankton significantly more than the 50 μm screen. Zooplankton width was more determinative of filtration performance than length, and both filters removed loricate species of rotifers significantly more efficiently than aloricate species of the same length and width size classes. The 25 and 50 μm ABSF also significantly reduced algal densities, with the exception of colonial and filamentous green algae (50 μm only). Filter efficiency relative to algal particles was influenced by filter pore size, organism morphology and structure, and intake density, while algal particle size was not determinative. This research provides compelling evidence that 25 or 50 μm filtration is a potentially powerful means of reducing densities of organisms discharged by ships operating in the Great Lakes but an additional treatment step would be necessary to effectively minimize risk and meet the International Maritime Organization's discharge standards associated with organisms of all sizes in the water column.  相似文献

The jaws of the ammonite generaEleganticeras, Hildoceras, Normannites, Scaphites, Physodoceras, andQuenstedtoceras (ofQuenst. lower jaws only) are described. Lower jaws and aptychi of these genera are shown to be identical. They consist of an inner layer of organic material, which corresponds to the complete anaptychi of liassic ammonites, and an outer layer of calcitic material deposited on both flancs of the lower jaw, giving rise, after decay of the organic layer, to the aptychi. Previously published interpretations of aptychi as opercula or hoods are discussed and rejected. It is concluded that they certainly functioned as jaws or possibly shovel-like devices.  相似文献

One proposed solution to the problem of ballast-mediated aquatic invasions involves chemically treating ballast water to kill key target organisms. Here, we examine the efficacy of three commercially available ballast water biocides using vegetative microalgae, dinoflagellate resting cysts and bacteria as test organisms. Chemicals tested were the ballast water biocides SeaKleen® and Peraclean® Ocean, and the chlorine dioxide biocide Vibrex®. Results demonstrate that the applicability of each of the three chemical biocides as a routine ballast water treatment is limited by factors such as cost, biological effectiveness and possible residual toxicity of the discharged ballast water (assessed on the basis of impact on motility of vegetative marine microalgae). Of the three biocides tested, Peraclean® Ocean appears to hold the most potential; however its effectiveness in shipboard trials is yet to be proven. Peraclean® Ocean was biodegradable within 2–6 weeks (initial concentration of 200 ppm), could effectively inactivate resting cysts of the marine dinoflagellates Gymnodinium catenatum, Alexandrium catenella and Protoceratium reticulatum at 400 ppm, could control bacterial growth of Escherichia coli, Staphylococcus aureus, Listeria innocua and Vibrio alginolyticus at 125–250 ppm, and could eliminate vegetative dinoflagellate cells at a concentration of 100 ppm. SeaKleen® eliminated vegetative microalgae at 2 ppm and could control resting cysts of the dinoflagellates G. catenatum and P. reticulatum at a concentration of 6 and 10 ppm, respectively, when exposed for a period of 2 weeks. SeaKleen® did not inactivate resting cysts of A. catenella at a concentration of 10 ppm and was found to degrade at a rate that could result in the discharge of residual toxic water into the marine environment. Together with the poor bactericidal properties of SeaKleen® (100–200 ppm required), this may limit the use of this biocide as a routine treatment option. Vibrex® is not a suitable ballast water treatment option due to the need for hydrochloric acid as an activator, however it was found to be the most effective against bacteria (complete inhibition at 15 ppm) indicating that onboard chlorine dioxide generators may provide an effective bacterial treatment option. The performance of these biocides was adversely influenced by a variety of factors including low water temperatures (6 °C compared to 17 °C), light versus dark conditions, and the presence of humus-rich seawater and ballast water sediments.  相似文献

In the autumn (October–December), Finnish Landrace ewes aged 1 year (yearling), 4–5 years (mature) and 7–8 years (older) were fed forages with no supplemented (control) or supplemented daily with 150 g (diet 150) or 300 g (diet 300) of concentrates per ewe in order to determine the effect of energy supply to initiate flushing on ewe productivity. Concurrently, in the summer (August–September), yearling and mature ewes were fed 0, 300 or 450 g of concentrates daily for 17 or 34 days. Concentrate supplement prior to and during mating had little effect on the live weight, however mature ewes gained weight in the summer. In the autumn, increasing proportion of concentrate in the diet reduced the length of the mating period in the yearling ewes (P=0.03) however, diet combination had no significant effect of age on conception. In August and early September, flushing did not hasten the onset of estrus and only 25% of the young ewes and 83% of the adult ewes were mated successfully. The flushing diet had no effect on the prolificacy in yearling and older ewes. In contrast, prolificacy and number of lambs weaned increased (P<0.05) for mature ewes on diet 150. In the autumn, there was a significant influence of flushing on the litter weight at birth and live weight gain from birth to weaning for older ewes however, diet 300 and the control were similar. Although litter weight at birth and weaning tended to be higher in the summer for ewes supplemented with concentrate for 34 days, the number of ewes required for a significant influence due to diet and length of flushing combination were not adequate. It was concluded that little is to be gained in lamb production by flushing ewes of the Finnish Landrace breed.  相似文献

Schistosomiasis is a globally important helminthic disease of both humans and animals, and is the second most common parasitic disease after malaria. Although praziquantel is extensively used for treatment of parasitic diseases, drug resistance has been reported. Therefore, new drugs and effective vaccines are needed for continuous control of schistosomiasis. Eggs produced by schistosomes are responsible for the occurrence and spread of schistosomiasis. Revealing the reproductive mechanism of schistosomes will help to control this disease. In this study, the proteomic profiles of single-sex infected female worms and bisexual infected mature female worms of Schistosoma japonicum at 18, 21, 23 and 25 days p.i. were identified with isobaric tags for relative quantitation-coupled liquid chromatography–tandem mass spectrometry. Differentially expressed proteins were subsequently used for bioinformatic analysis. Six highly expressed differentially expressed proteins in mature female worms were selected and long-term interference with small interfering RNA (siRNA) was conducted to determine biological functions. SiRNA against S. japonicum translationally controlled tumour protein (SjTCTP) resulted in the most significant effect on the growth and development of MF worms. Sjtctp mRNA expression gradually increased over time with a high level of expression maintained at 25–42 days p.i., while levels were significantly higher in mature female worms than male and SF worms. The subsequent animal immune protection experiments showed that recombinant SjTCTP (rSjTCTP) reduced the number of adults by 44.7% (P < 0.01), average egg burden per gram of liver by 57.94% (P < 0.01), egg hatching rate by 47.57% (P < 0.01), and oviposition of individual females by 43.16%. rSjTCTP induced higher levels of serum IgG, IL-2, and IL-10 in mice. Collectively, these results show that SjTCTP is vital to reproduction of female worms and, thus, is a candidate antigen for immune protection.  相似文献

The occurrence of freshwater harmful algal bloom toxins impacting the coastal ocean is an emerging threat, and the potential for invertebrate prey items to concentrate toxin and cause harm to human and wildlife consumers is not yet fully recognized. We examined toxin uptake and release in marine mussels for both particulate and dissolved phases of the hepatotoxin microcystin, produced by the freshwater cyanobacterial genus Microcystis. We also extended our experimental investigation of particulate toxin to include oysters (Crassostrea sp.) grown commercially for aquaculture. California mussels (Mytilus californianus) and oysters were exposed to Microcystis and microcystin toxin for 24 h at varying concentrations, and then were placed in constantly flowing seawater and sampled through time simulating riverine flushing events to the coastal ocean. Mussels exposed to particulate microcystin purged the toxin slowly, with toxin detectable for at least 8 weeks post-exposure and maximum toxin of 39.11 ng/g after exposure to 26.65 μg/L microcystins. Dissolved toxin was also taken up by California mussels, with maximum concentrations of 20.74 ng/g after exposure to 7.74 μg/L microcystin, but was purged more rapidly. Oysters also took up particulate toxin but purged it more quickly than mussels. Additionally, naturally occurring marine mussels collected from San Francisco Bay tested positive for high levels of microcystin toxin. These results suggest that ephemeral discharge of Microcystis or microcystin to estuaries and the coastal ocean accumulate in higher trophic levels for weeks to months following exposure.  相似文献

The thermal environment can induce substantial variation in important life-history traits. Experimental manipulation of the thermal environment can help researchers determine the contribution of this factor to phenotypic variation in life-history traits. During the reproductive season, we kept female northern grass lizards, Takydromus septentrionalis (Lacertidae), in three temperature-controlled rooms (25, 28 and 32 °C) to measure the effect of the maternal thermal environment on reproductive traits. Maternal thermal environment remarkably affected reproductive frequency and thereby seasonal reproductive output, but had little effect on reproductive traits per clutch or hatchling traits. Females kept at 32 °C produced more clutches and thus had shorter clutch intervals than females from 28 to 25 °C. Clutch size, clutch mass, relative clutch mass, egg size and hatchling traits did not vary among the three treatments. The eggs produced by the females were incubated at 27 °C and the traits of hatchlings were measured. The result that egg (offspring) size was independent of maternal thermal environments is consistent with the prediction of the optimal egg size (offspring) theory. The eggs produced by low temperature females (28 and 25 °C) took longer time to complete their post-oviposition development than did eggs produced by high temperature females (32 °C). This suggests that the eggs from low temperatures might have been laid when the embryos were at relatively early stages. Therefore, maternal thermal environment prior to oviposition could affect post-oviposition development in T. septentrionalis.  相似文献

Epidemiological studies and healthy eating guidelines suggest a positive correlation between ingestion of whole grain cereal and food rich in fibre with protection from chronic diseases. The prebiotic potential of whole grains may be related, however, little is known about the microbiota modulatory capability of oat grain or the impact processing has on this ability. In this study the fermentation profile of whole grain oat flakes, processed to produce two different sized flakes (small and large), by human faecal microbiota was investigated in vitro. Simulated digestion and subsequent fermentation by gut bacteria was investigated using pH controlled faecal batch cultures inoculated with human faecal slurry. The different sized oat flakes, Oat 23’s (0.53–0.63 mm) and Oat 25’s/26’s (0.85–1.0 mm) were compared to oligofructose, a confirmed prebiotic, and cellulose, a poorly fermented carbohydrate. Bacterial enumeration was carried out using the culture independent technique, fluorescent in situ hybridisation, and short chain fatty acid (SCFA) production monitored by gas chromatography. Significant changes in total bacterial populations were observed after 24 h incubation for all substrates except Oat 23’s and cellulose. Oats 23’s fermentation resulted in a significant increase in the BacteroidesPrevotella group. Oligofructose and Oats 25’s/26’s produced significant increases in Bifidobacterium in the latter stages of fermentation while numbers declined for Oats 23’s between 5 h and 24 h. This is possibly due to the smaller surface area of the larger flakes inhibiting the simulated digestion, which may have resulted in increased levels of resistant starch (Bifidobacterium are known to ferment this dietary fibre). Fermentation of Oat 25’s/26’s resulted in a propionate rich SCFA profile and a significant increase in butyrate, which have both been linked to benefiting host health. The smaller sized oats did not produce a significant increase in butyrate concentration. This study shows for the first time the impact of oat grain on the microbial ecology of the human gut and its potential to beneficially modulate the gut microbiota through increasing Bifidobacterium population.  相似文献

We characterized the physical/chemical conditions and the algal and bacterial assemblages in ballast water from 62 ballast tanks aboard 28 ships operated by the U.S. Military Sealift Command and the Maritime Administration, sampled at 9 ports on the U.S. West Coast and 4 ports on the U.S. East Coast. The ballast tank waters had been held for 2–176 days, and 90% of the tanks had undergone ballast exchange with open ocean waters. Phytoplankton abundance was highly variable (grand mean for all tanks, 3.21 × 104 viable cells m−3; median, 7.9 × 103 cells m−3) and was unrelated to physical/chemical parameters, except for a positive relationship between centric diatom abundance and nitrate concentration. A total of 100 phytoplankton species were identified from the ballast tanks, including 23 potentially harmful taxa (e.g. Chaetoceros concavicornis, Dinophysis acuminata, Gambierdiscus toxicus, Heterosigma akashiwo, Karlodinium veneficum, Prorocentrum minimum, Pseudo-nitzschia multiseries). Assemblages were dominated by chain-forming diatoms and dinoflagellates, and viable organisms comprised about half of the total cells. Species richness was higher in ballast tanks with coastal water, and in tanks containing Atlantic or Pacific Ocean source waters rather than Indian Ocean water. Total and viable phytoplankton numbers decreased with age of water in the tanks. Diversity also generally decreased with water age, and tanks with ballast water age >33 days did not produce culturable phytoplankton. Abundance was significantly higher in tanks with recently added coastal water than in tanks without coastal sources, but highly variable in waters held less than 30 days. Bacterial abundance was significantly lower in ballast tanks with Atlantic than Pacific Ocean source water, but otherwise was surprisingly consistent among ballast tanks (overall mean across all tanks, 3.13 ± 1.27 × 1011 cells m−3; median, 2.79 × 1011 cells m−3) and was unrelated to vessel type, exchange status, age of water, environmental conditions measured, or phytoplankton abundance. At least one of four pathogenic eubacteria (Listeria monocytogenes, Escherichia coli, Mycobacterium spp., Pseudomonas aeruginosa) was detected in 48% of the ballast tanks, but toxigenic strains of Vibrio cholerae were not detected. For ships with tanks of similar ballasting history, the largest source of variation in phytoplankton and bacteria abundance was among ships; for ships with tanks of differing ballasting histories, and for all ships/tanks considered collectively, the largest source of variation was within ships. Significant differences in phytoplankton abundance, but not bacterial abundance, sometimes occurred between paired tanks with similar ballasting history; hence, for regulatory purposes phytoplankton abundance cannot be estimated from single tanks only. Most tanks (94%) had adequate records to determine the source locations and age of the ballast water and, as mentioned, 90% had had ballast exchange with open-ocean waters. Although additional data are needed from sediments that can accumulate at the bottom of ballast tanks, the data from this water-column study indicate that in general, U.S. Department of Defense (DoD) ships are well managed to minimize the risk for introduction of harmful microbiota. Nevertheless, abundances of viable phytoplankton with maximum dimension >50 μm exceeded proposed International Maritime Organization standards in 47% of the ballast tanks sampled. The data suggest that further treatment technologies and/or alternative management strategies will be necessary to enable DoD vessels to comply with proposed standards.  相似文献

Th1 and Th2 cytokines play key role in protection from and pathogenesis of mycobacterial infection and their dynamic changes may predict clinical outcome of the patient. Patients with tuberculosis (TB) have a poorer cellular immune response to recombinant 32-kDa antigen (Ag) of Mycobacterium bovis (r32-kDa M. bovis) than do healthy volunteers. The basis for this observation was studied by evaluating the Th1 (gamma interferon [IFN-γ]) produced in response to the r32-kDa Ag M. bovis by peripheral blood mononuclear cells (PBMC) from patients with pulmonary TB (n = 20), extra-pulmonary TB (n = 13) and from healthy volunteers (n = 9). Recombinant 32-kDa M. bovis stimulated PBMC from TB patients produced significantly lower levels of IFN-γ at 0 month, and increased at 2–4, and 6 months of treatment and were highly significant (p < 0.000) compared to the responses in controls. The ratios of IFN-γ to IL-10 were low in patients newly diagnosed and improved both during and after treatment. The present study concludes that the levels of in vitro response to M. bovis BCG r32-kDa Ag leading to the specific release of IFN-γ increased after anti-tuberculosis treatment and seems to reflect the clinical status of the patient, thus reiterating the utility of this antigen in T cell based assays as a surrogate marker of cell mediated responses.  相似文献

A milestone in understanding a globally significant mechanism of marine bioinvasions was published 25 years ago. The transformative paper on ballast water provided a baseline of patterns, processes and predictions of marine introductions, underpinned a dramatic increase in research on the topic, and presented a foundational insight for an international approach to vector management. The 25 year anniversary of JT Carlton’s ‘Transoceanic and interoceanic dispersal of coastal marine organisms: the biology of ballast water’ coincides with an International Maritime Organization (IMO) convention for management of ballast water that has not been fully ratified. The emergence of ship biofouling as a vector of management concern worldwide has also prompted renewed efforts to reduce species transfers by ships through new policy initiatives.  相似文献

Glucose regulated protein (GRP) located in endoplasmic reticulum (ER) was a member of heat shock protein (Hsp) family. The protective mechanism adapted to ER stimuli was closely related to GRP. GRP78, known as BiP, was one of central regulator responded to stress in ER. Grass carp (Ctenopharyngodon idella) GRP78 (CiGRP78) was up-regulated in almost tissues, especially in liver, under heat shock (34 °C), cold stress (4 °C) or lead nitrate (0.25 mmol/L) stress. In order to understand the function of CiGRP78 in cellular protection, CiGRP78 ORF cDNA was inserted into the plasmid of pET-32a(+) or pEGFP-C1 respectively, then the recombinant plasmids were transformed or transfected into Escherichia coli cells, mouse myeloma cells (SP2/0) or grass carp kidney cells (CIK). In the cells, CiGRP78 was over-expressed following thermal, cold or Pb2+ stress. Results showed that CiGRP78 not only contributed to protecting prokaryotic cells against thermal or cold extremes, but also played the same role in SP2/0 and CIK cells. After treatment with heat stress at 42 °C for 1 h, although the viability of the cells declined a lot, CIK cells with pEGFP-C1/CiGRP78 exhibited a higher survival rate (28%) than wild-type cells (7%) or cells with only pEGFP-C1 (5.1%). When the time lag extended to 2.5 h, the survival rates were 19%, 5.7%, 4.8% respectively. In addition, CiGRP78 would also provide a transient cytoplasm protection against Pb2+ stress in a dose- and time-dependent manner. After treatment with lead nitrate at concentration of 10 μmol/L for 12 h, 24 h or 36 h, the survival rates of cells with pEGFP-C1 or wild-type cells were 46.7% or 46.7% (12 h), 25% or 22% (24 h), 10% or 11% (36 h) respectively. When the cells were treated with lead nitrate at the concentration of 25 μmol/L, the survival rates of cells with pEGFP-C1 or wild-type cells were 45.5% or 30% (12 h), 16.7% or 25% (24 h), 6.5% or 8% (36 h), respectively. CiGRP78 provided a distinct protection in CIK cells at the low concentration for 24 h. The survival rates of CIK cells with pEGFP-C1/CiGRP78 treated with lead nitrate at concentration of 10 μmol/L or 25 μmol/L were 65.9% or 58.8% respectively. When the cells were treated with lead nitrate at concentration of 50 μmol/L for 24 h, the survival rate of the CIK cells was only about 30%. If the process-time was extended to 36 h, CiGRP78 could not provide any cytoplasm protection for CIK cells.  相似文献

AimTo compare the radiotherapy technique used in a randomised trial with VMAT and an in-house technique for prostate cancer.BackgroundTechniques are evolving with volumetric modulated arc therapy (VMAT) commonly used. The CHHiP trial used a 3 PTV forward planned IMRT technique (FP_CH). Our centre has adopted a simpler two PTV technique with locally calculated margins.Materials and methods25 patients treated with FP_CH to 60 Gy in 20 fractions were re-planned with VMAT (VMAT_CH) and a two PTV protocol (VMAT_60/52 and VMAT_60/48). Target coverage, conformity index (CI), homogeneity index (HI), monitor units (MU) and dose to the rectum, bladder, hips and penile bulb were compared.ResultsPTV coverage was high for all techniques. VMAT_CH plans had better CI than FP_CH (p   0.05). VMAT_60/52/48 plans had better CI than VMAT_CH. FP_CH had better HI and fewer MU than VMAT (p   0.05). More favourable rectum doses were found for VMAT _CH than FP_CH (V48.6, V52.8, V57, p   0.05) with less difference for bladder (p   0.05). Comparing VMAT_CH to VMAT_60/52/48 showed little differences for the bladder and rectum but VMAT_CH had larger penile bulb doses (V40.8, V48.6, mean, D2, p   0.05). Femoral head doses (V40.8) were similarly low for all techniques (p = ≥ 0.05).ConclusionVMAT produced more conformal plans with smaller rectum doses compared to FP_CH albeit worse HI and more MU. VMAT_60/52 and VMAT_60/48 plans had similar rectal and bladder doses to VMAT_CH but better CI and penile bulb doses which may reduce toxicity.  相似文献
