中华绒螯蟹卵巢差减cDNA文库的构建   总被引:17,自引:0,他引:17  
应用抑制性差减杂交技术构建中华绒螯蟹卵巢两个发育时期的差减cDNA文库。正向差减杂交以Ⅲ期卵巢为试验方、Ⅱ期卵巢为驱动方,反向差减杂交以Ⅱ期卵巢为试验方、Ⅲ期卵巢为驱动方;将所获差减cDNA片段克隆入质粒表达载体,转化大肠杆菌JM109。最后获得的正、反向差减文库分别含863、360个重组子。PCR扩增鉴定正、反向差减cDNA文库的插入片段平均大小分别为360和160bp,表明所构建的差减言语库适合进一步研究中华绒螯蟹卵巢发育相关基因。  相似文献

应用抑制性差减杂交技术,已经获得了中华绒螯蟹卵巢发育过程中差异表达基因的部分cDNA序列。为了进一步获得基因的全长cDNA序列,运用SMART技术,成功构建了中华绒螯蟹卵巢(Ⅲ期)RACE cDNA文库。琼脂糖凝胶电泳结果表明,文库所含全长cDNA的长度主要集中在500—2000bP之间,RACE PCR结果表明,所用基因特异性引物与接头引物皆能扩增出产物,说明所构文库的质量较好,适于用RACE方法从中分离中华绒螯蟹卵巢发育相关基因的全长cDNA。  相似文献

以紫外线灭活的dsRNA病毒草鱼出血病病毒(GCHV)诱导和模拟诱导的牙鲆胚胎细胞为材料,利用抑制性差减杂交(SSH)技术,成功构建了双链RNA病毒诱导的牙鲆胚胎细胞(FEC)差减cDNA文库.以管家基因α-tublin作为差减指标,经检测,该文库差减效率达2 10倍,表明经病毒诱导后某些差异表达基因也得到了相应倍数的富集.将获得的cDNA片段连接到pGEM-T载体,PCR检测显示差减片段在250bp~2 000bp之间.该差减cDNA文库的构建为从分子水平研究牙鲆培养细胞对dsRNA病毒的免疫反应、以及进一步鉴定和克隆差异表达基因打下了坚实基础.  相似文献

黄萎病菌诱导的海岛棉抗病反应的SSH文库构建及分析   总被引:10,自引:0,他引:10  
在Pima90幼苗接种黄萎病病原菌后2、4、8、12、24、48、72和96h分批取幼根以抽提棉花总RNA。以未接种病原的棉花cDNA为驱动方,利用SSH法对接种病原后的棉花cDNA进行差减杂交。对杂交后的cDNA进行T/A克隆并转化到大肠杆菌中完成文库构建,共获得了534个克隆。以M13引物对扩增插入片段进行PCR扩增,电泳结果显示插入片段大小从0.2—1.2kb不等,平均大小为0.5kb。将克隆中的插入片段点种于尼龙膜上,分别以病原诱导前后的总cDNA为探针进行反向Northern杂交。对78个在海岛棉的抗病反应中呈上升表达的克隆进行了测序并对测序结果去载体后在NCBI上利用Blastn和Blastx进行序列相似性分析。结果表明,大部分阳性克隆与拟南芥等多个物种不同逆境条件下诱导的相关基因或表达序列相同或同源,如棉花病程相关蛋白家族10,拟南芥抗病反应家族蛋白等。SSH文库的构建和分析将有助于了解棉花抗黄萎病反应的分子机制。  相似文献

鲫鱼低氧相关基因差减cDNA文库的构建与分析   总被引:1,自引:0,他引:1  
鲫鱼对低氧具有极强的耐受性。在低氧状态下鲫鱼鳃瓣表面积增加,无氧代谢增强,能量消耗降低。但是人们对鲫鱼产生这些低氧反应的分子机理还缺乏了解。本研究以1%低氧处理24h的鲫鱼囊胚细胞(CAB)作为检测子(Tester),常氧条件下培养的CAB细胞作为驱赶子(Driver),分别提取总RNA,利用SMART cDNA 技术合成双链cDNA,经差减杂交和抑制性PCR扩增获得差减PCR产物。然后将差减PCR产物连接到pGEM-T载体上,构建差减cDNA文库。以管家基因β-actin作为指标检测差减效率,发现该文库差减效率达28倍。随机挑选阳性克隆进行PCR检测,显示差减片段在0.1–2kb之间。挑取含有插入片段的1300个克隆进行测序,通过生物信息学分析获得267个基因序列(e≤0.001;Identity>40%)。该差减cDNA文库的成功构建对克隆鱼类耐低氧相关基因和深入认识鱼类耐受低氧的分子机制有非常重要的意义。  相似文献

嗜水气单胞菌感染的中华鳖主要器官差减cDNA文库的构建   总被引:1,自引:0,他引:1  
以致病性嗜水气单胞菌(Aeromonas hydrophila)人工感染的中华鳖(Trionyx sinensis)肝、脾、肾组织为材料,应用抑制性差减杂交(SSH)技术,构建了嗜水气单胞菌感染组织的差减cDNA文库。以中华鳖管家基因-βactin作为差减指标检测该文库差减效率达210倍,表明感染细菌后某些差异表达基因得到了相应倍数的富集。将获得的cDNA片段连接到pMD18-T载体并转化大肠杆菌DH5α感受态细胞。PCR阳性检测显示差减片段在150—800bp之间。该差减cDNA文库的构建为快速分离和鉴定中华鳖与细菌感染相关的免疫基因及从分子水平探讨中华鳖的病理和抗感染免疫机制奠定了基础。  相似文献

应用计算机工具、GenScan软件预测中国美利奴绵羊MHC Class I区段的BAC文库中453oⅡ克隆的基因数目、特性及结构,建立一种可以从cDNA文库中简便有效获取表达基因的技术方法。选取4个预测基因作研究对象设计引物,应用PCR技术,对已构建好的cDNA文库进行PCR扩增,回收"目的基因"片段并连接pGEM-T载体,转DH5α大肠杆菌中扩增后测序。琼脂糖凝胶电泳检测PCR扩增产物,cDNA文库中有目的条带,测序结果与GenBank进行Blast分析,分析结果表明这些基因与羊的基因均具有99%以上的相似性。因此应用基因预测分析与PCR结合技术可简便迅速的从cDNA文库中获取表达基因。  相似文献

以Hoagland溶液培养的梭梭幼苗(H)为对照群体,甘露醇处理的梭梭幼苗(M)为目标群体,进行抑制差减杂交。用经过H cDNA差减的M cDNA构建了一个含有大约400个独立克隆的差减文库;采用差减前的H cDNA和M cDNA以及正向/反向差减杂交后的cDNA为模板标记探针,对随机挑取的100个重组质粒进行差示筛选,获得了21个阳性侯克隆,从这些阳性候选克隆中随机挑取了8个进行Northern blot分析。证实其中3个候选克隆代表了在M中特异表达或表达增强的基因,序列分析和同源性比较表明它们与逆境胁迫有关;而另外5个候选克隆无Northern杂交信号,推测它们为低丰度转录本。  相似文献

以小麦抗叶锈病近等基因系TcLr41和感病亲本Thatcher的叶片cDNA分别作为试验方和驱动方,利用抑制差减杂交技术,构建了一个包含2544个克隆的差减文库。随机提取阳性克隆质粒DNA后经PCR检测,插入片段大部分集中在200~1000bp之间,证明所构建的文库符合要求。在功能已知的基因中,推测过氧化氢酶(catalasc)基因、抗秆锈病基因(rust resistance gene)、铜蓝结合蛋白(blue copper—binding protein)基因、锌指蛋白(ring zinc finger protein)基因、胁迫反应蛋白(stress responsive protein)基因等可能是TcLr41中抗病相关差异表达基因。  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

A complex analysis of seasonal fluctuations of the mean group parameters of the system of regulation of lipid peroxidation has been performed in liver of Balb/c mice. Association of lipid characteristics and morphophysiological parameters is studied in the Balb/c mouse liver. An inter-connection is revealed between the liver index and the amount of lysoforms of phospholipids, the scale and character of the interconnection differing essentially depending on proportion of phos-phatidylcholine in mouse liver phospholipids.  相似文献

Seven strains of aerobic carbon monoxide-oxidizing bacteria (carboxydebacteria) when growing on CO as sole source of carbon and energy had doubling times which ranged from 12–42 h. The activity profiles obtained after discontinuous sucrose density gradient centrifugation indicated that the CO-oxidizing enzymes are soluble and the hydrogenases are membrane-bound in all strains examined. The CO-oxidizing enzymes of Pseudomonas carboxydohydrogena, Pseudomonas carboxydoflava, Comamonas compransoris, and the so far unidentified strains OM2, OM3, and OM4 had a molecular weight of 230,000; that of Achromobacter carboxydus amounted to 170,000. The molecular weights of the CO-oxidizing and H2-oxidizing enzymes turned out to be identical. The cell sonicates were shown to catalyze the oxidation of both CO and H2 with methylene blue, thionine, phenazine methosulfate, toluylene blue, dichlorophenolindophenol, cytochrome c or ferricyanide as electron acceptors. Methyl viologen, benzyl viologen, FAD+, FMN+, and NAD(P)+ were not reduced. The spectrum of electron acceptors was identical for all strains tested. Neither free formate, hydrogen nor oxygen gas were involved in the CO-oxidation reaction. Methylene blue was reduced by CO at a 1:1 molar ratio. The results indicate that CO-oxidation by carboxydobacteria is catalyzed by identical or similar enzymes and that the reaction obeys the equation CO+H2OCO2+2H++2e- as previously shown for Pseudomonas carboxydovorans.Dedicated to Otto Kandler remembering almost three decades of enjoyable cooperation  相似文献

Since the introduction of the concepts of allostery about four decades ago, much advancement has been made in elucidating the structure-function correlation in allostery. However, there are still a number of issues that remain unresolved. In this review we used mammalian pyruvate kinase (PK) as a model system to understand the role of protein dynamics in modulating cooperativity. PK has a triosephosphate isomerase (TIM)(α/β)8 barrel structural motif. PK is an ideal system to address basic questions regarding regulatory mechanisms about this common (α/β)8 structural motif. The simplest model accounting for all of the solution thermodynamic and kinetic data on ligand-enzyme interactions involves two conformational states, inactive ET and active ER. These conformational states are represented by domain movements. Further studies provide the first evidence for a differential effect of ligand binding on the dynamics of the structural elements, not major secondary structural changes. These data are consistent with our model that allosteric regulation of PK is the consequence of perturbation of the distribution of an ensemble of states in which the inactive ET and active ER represent the two extreme end states. Sequence differences and ligands can modulate the distribution of states leading to alterations of functions. The future work includes: defining the network of functionally connected residues; elucidating the chemical principles governing the sequence differences which affect functions; and probing the nature of mutations on the stability of the secondary structural elements, which in turn modulate allostery.  相似文献

对我国花型Hemibarbuspattern鱼类作了骨骼系统比较,结果表明,此类型鱼类脑颅较长,副蝶骨平直或稍弯曲,眶蝶骨腹纵嵴发达(铜鱼Coreius septentrionalis例外),下颞窝和咽突中等大,基枕骨后突发达;脑颅中的上筛骨的后突、侧筛骨的外筛突,蝶耳骨的外突、上耳骨的后突、围眶骨和后颞窝等均有明显的差异;咽颅中的舌颌骨、尾舌骨、鳃盖骨和下咽齿的列数等又有显著的区别;附肢骨骼中的腰带骨、脊椎骨中的复合神经骨和第4椎骨腹侧的悬器等也有不同之处。据此,这些差异和区别可作为属间或种间的分类依据。  相似文献
