高等真核细胞的染色体DNA通过基质结合区(MAR)不时地与核基质特异性结合而组织成一种空间环状结构。为了研究以DNA套环形式附着于核基质上的DNA序列的特性,从处于泌乳期的乳腺组织中克隆了多个MAR DNA序列。体外结合实验表明,这些序列能够同核基质蛋白共结合成不溶性的复合物,这些复合物可较容易的通过离心去除。其中,两个MAR序列中包含有TL、CA—和GA—阻断以及ATTA基序。这两个序列中含有多个复制/转录因子的结合位点、增强子基序、多个完全的和非完全的反向重复序列以及潜在的DNA弯曲核心序列样结构。同一DNA序列中存在不同元件的组合可能说明在控制一系列细胞的发育过程中,它们可能发挥有正的或负的调控元件的功能。  相似文献

The permanent DNA attachment sites to the nuclear matrix in the domain of chicken alpha-globin genes originally found in erythrocyte nuclei are shown to exist in sperm and cultured fibroblast cells too. Short fragments of permanently attached to the nuclear matrix DNA have been cloned and sequenced. A primary structure of a 1.7 k.b. fragment from 5'-region of chicken alpha-globin gene domain containing both replication origin and permanent attachment site has been determined. A region possessing homologies with papovaviral replication origins and putative mammalian ARS elements has been found on the 1.7 k.b. fragment. A region containing short internal repeats and GC-rich motifs has also been found. Similar motifs were observed in several of the cloned short fragments of DNA permanently attached to the nuclear matrix.  相似文献

Vertebrates have developed multiple mechanisms to coordinate the replication of epigenetic and genetic information. Dnmt1 encodes the maintenance enzyme DNA-methyltransferase, which is responsible for propagating the DNA methylation pattern and the epigenetic information that it encodes during replication. Direct sequence analysis and bisulfite mapping of the 5' region of DNA-methyltransferase 1 (dnmt1) have indicated the presence of many sequence elements associated with previously characterized origins of DNA replication. This study tests the hypothesis that the dnmt1 region containing these elements is an origin of replication in human cells. First, we demonstrate that a vector containing this dnmt1 sequence is able to support autonomous replication when transfected into HeLa cells. Second, using a gel retardation assay, we show that it contains a site for binding of origin-rich sequences binding activity, a recently purified replication protein. Finally, using competitive polymerase chain reaction, we show that replication initiates in this region in vivo. Based on these lines of evidence, we propose that initiation sites for DNA replication are located between the first intron and exon 7 of the human dnmt1 locus.  相似文献

A comparison was made between the DNA sequences in two regions of the mouse and the human apolipoprotein B genes: the 5'-flanking sequence and the region between the first exon and the second intron. Considerable homology was observed, particularly in the immediate 5' region and in the second intron. Because promoter and enhancer elements have been previously localized to these regions in the human apolipoprotein B gene, it is proposed that regions of conserved base sequence delineate binding regions for regulatory proteins. In some cases, contiguous regions of homology are longer than expected for regions designed as recognition sites for individual nuclear proteins, and may define regions recognizable by a cluster of interacting proteins. Both the human and mouse genes contain repetitive elements and a hypervariable dinucleotide repeat.  相似文献

The Yersinia enterocolitica prophage PY54 replicates as a linear DNA molecule with covalently closed ends. For replication of a circular PY54 minimal replicon that has been derived from a linear minireplicon, two phage-encoded loci are essential in Escherichia coli: (i) the reading frame of the replication initiation gene repA and (ii) its 212-bp origin located within the 3' portion of repA. The RepA protein acts in trans on the origin since we have physically separated the PY54 origin and repA onto a two-plasmid origin test system. For this trans action, the repA 3' end carrying the origin is dispensable. Mutagenesis by alanine scan demonstrated that the motifs for primase and for nucleotide binding present in the protein are essential for RepA activity. The replication initiation functions of RepA are replicon specific. The replication initiation proteins DnaA, DnaG, and DnaB of the host are unable to promote origin replication in the presence of mutant RepA proteins that carry single residue exchanges in these motifs. The proposed origins of the known related hairpin prophages PY54, N15, and PKO2 are all located toward the 3' end of the corresponding repA genes, where several structure elements are conserved. Origin function depends on the integrity of these elements.  相似文献

Polycomb group (PcG) proteins repress homeotic genes in cells where these genes must remain inactive during development. This repression requires cis-acting silencers, also called PcG response elements. Currently, these silencers are ill-defined sequences and it is not known how PcG proteins associate with DNA. Here, we show that the Drosophila PcG protein Pleiohomeotic binds to specific sites in a silencer of the homeotic gene Ultrabithorax. In an Ultrabithorax reporter gene, point mutations in these Pleiohomeotic binding sites abolish PcG repression in vivo. Hence, DNA-bound Pleiohomeotic protein may function in the recruitment of other non-DNA-binding PcG proteins to homeotic gene silencers.  相似文献
