Although maize endosperm undergoes programmed cell death during its development, it is not known whether this developmental feature is common to cereals or whether it arose inadvertently from the selection process that resulted in the enlarged endosperm of modern maize. Examination of wheat endosperm during its development revealed that this tissue undergoes a programmed cell death that shares features with the maize program but differs in some aspects of its execution. Cell death initiated and progressed stochastically in wheat endosperm in contrast to maize where cell death initiates within the upper central endosperm and expands outward. After a peak of ethylene production during early development, wheat endosperm DNA underwent internucleosomal fragmentation that was detectable from mid to late development. The developmental onset and progression of DNA degradation was regulated by the level of ethylene production and perception. These observations suggest that programmed cell death of the endosperm and regulation of this program by ethylene is not unique to maize but that differences in the execution of the program appear to exist among cereals.  相似文献

Regulation of programmed cell death in maize endosperm by abscisic acid   总被引:26,自引:0,他引:26  
Cereal endosperm undergoes programmed cell death (PCD) during its development, a process that is controlled, in part, by ethylene. Whether other hormones influence endosperm PCD has not been investigated. Abscisic acid (ABA) plays an essential role during late seed development that enables an embryo to survive desiccation. To examine whether ABA is also involved in regulating the onset of PCD during endosperm development, we have used genetic and biochemical means to disrupt ABA biosynthesis or perception during maize kernel development. The onset and progression of cell death, as determined by viability staining and the appearance of internucleosomal DNA fragmentation, was accelerated in developing endosperm of ABA-insensitive vp1 and ABA-deficient vp9 mutants. Ethylene was synthesized in vp1 and vp9 mutant kernels at levels that were 2–4-fold higher than in wild-type kernels. Moreover, the increase and timing of ethylene production correlated with the premature onset and accelerated progression of internucleosomal fragmentation in these mutants. Treatment of developing wild-type endosperm with fluridone, an inhibitor of ABA biosynthesis, recapitulated the increase in ethylene production and accelerated execution of the PCD program that was observed in the ABA mutant kernels. These data suggest that a balance between ABA and ethylene establishes the appropriate onset and progression of programmed cell death during maize endosperm development.  相似文献

We present cellular- and ultracellular-level studies here to show developmental programmed cell death (PCD) of placento-chalazal (P-C) cell layers in maternal pedicel tissue in developing caryopses of normal seed (Mn1) and in the invertase-deficient miniature (mn1) seed mutant in maize (Zea mays). PCD was evidenced by loss of nuclei and all subcellular membranous organizations in many P-C layers. The terminal deoxynucleotidyl transferase-mediated X-dUTP nick-end labeling (TUNEL) stain that is diagnostic of apoptotic-like PCD identified spatially and temporally two distinctive subdomains, which coincided with nucellar and integumental P-C layers based on their developmental origins. The early phase of PCD in the nucellar P-C was TUNEL negative and was specific to only the fertilized caryopses, indicating that the signaling for PCD in these maternal cells originated in the zygotic tissues. In fact, the initiation of PCD coincided with endosperm cellularization and was rapidly and coordinately completed prior to the beginning of the major storage phase in endosperm. Cell shape in these cell layers was also influenced by the genotype of filial endosperm. The later phase of PCD was restricted to the integumental P-C layers underneath the nucellar cells and was TUNEL positive in both genotypes. The two subdomains of the P-C layers were also distinguishable by unique cell wall-associated phenolic compounds. Based on collective evidence, we infer that the nucellar PCD may have osmolytic etiology and may lead to activation of the post-phloem transport function of the P-C layer, whereas the integumental PCD was senescent related, in particular, protecting the maturing seed against microbes that may be transported from the maternal tissue.  相似文献

Programmed cell death (PCD) is the genetically regulated disassembly of cells, and occurs in the endosperm of cereals during seed maturation. Since PCD determines the lifetime of cells, it can affect endosperm growth and, therefore, cereal yield. However, the features and mechanisms of PCD in the developing starchy endosperm in the Poaceae remain unclear. In the present study, we investigated the characteristics of PCD in developing starchy endosperm of rice (Oryza sativa L.) by fluorescence microscopy, focusing on the spatial and temporal progress of PCD-associated responses. Cell death commenced in the central region of starchy endosperm, and then spread to the peripheral region. PCD-associated responses, such as mitochondrial membrane permeabilization and activation of the protease that cleaves the amino acid sequence VEID, showed similar spatial patterns to that of cell death, but preceded cell death. Degradation of nuclear DNA could not be detected in developing starchy endosperm by the TUNEL assay. These results indicated that PCD in developing starchy endosperm of rice proceeds via a highly organized pattern. In addition, these results suggested that PCD in developing starchy endosperm of rice is characterized by the involvement of mitochondrial signaling and the activity of a caspase-like protease that cleaves the VEID sequence.  相似文献

After fertilization, filial grain organs are surrounded by the maternal nucellus embedded within the integuments and pericarp. Rapid early endosperm growth must be coordinated with maternal tissue development. Parameters of maternal tissue growth and development were analysed during early endosperm formation. In the pericarp, cell proliferation is accomplished around the time of fertilization, followed by cell elongation predominantly in longitudinal directions. The rapid cell expansion coincides with endosperm cellularization. Distribution of TUNEL (terminal deoxynucleotidyl transferase-mediated dUTP nick end labelling)-positive nuclei reveals distinct patterns starting in the nucellus at anthesis and followed later by the inner cell rows of the pericarp, then spreading to the whole pericarp. The pattern suggests timely and spatially regulated programmed cell death (PCD) processes in maternal seed tissues. When the endosperm is coenocytic, PCD events are only observed within the nucellus. Thereby, remobilization of nucellar storage compounds by PCD could nourish the early developing endosperm when functional interconnections are absent between maternal and filial seed organs. Specific proteases promote PCD events. Characterization of the barley vacuolar processing enzyme (VPE) gene family identified seven gene members specifically expressed in the developing grain. HvVPE2a (known as nucellain) together with closely similar HvVPE2b and HvVPE2d might be involved in nucellar PCD. HvVPE4 is strongly cell specific for pericarp parenchyma. Correlative evidence suggests that HvVPE4 plays a role in PCD events in the pericarp. Possible functions of PCD in the maternal tissues imply a potential nutritive role or the relief of a physical restraint for endosperm growth. Programmed cell death (PCD) is an active, genetically controlled process that ultimately leads to elimination of unnecessary or damaged cells from multicellular organism. It occurs during normal growth and development or in response to a variety of environmental triggers and is indispensable for survival of the organism. In Echinocystis lobata the endosperm, an ephemeral tissue in angiosperm plants, undergoes distinct cytological, physiological and molecular changes during seed development and maturation. As a result, mature seeds are deprived of this tissue. The endosperm was analyzed at the consecutive stages of seed development. The morphological changes of cells were studied at light and electron microscope levels. In this paper we report that endosperm cells undergo morphological and biochemical changes characteristic of apoptosis, a particular type of PCD, i.e. cell shrinkage, chromatin condensation, nuclear fragmentation, and cytoplasm degradation, while the ultrastructure of mitochondria seems to be less changed. Furthermore, the progression of DNA degradation has been shown by agarose gel electrophoresis (ladder pattern of DNA fragmentseparation), TUNEL and comet assay. It isconcluded that during seed maturation, endosperm degradation process is accompanied by typical PCD-related changes of cell morphology and internucleosomal DNA cleavage.  相似文献

Endosperm in cereals such as wheat, is a part of the mature seeds and a valuable source of key substances for humans and animals. For this reason, the development of immature endosperm tissues in planta was the focus of this research. However, it is commonly known that tissue culture conditions can alter the developmental pathway of plant cells and can expose their potency. There is scarce information about research on isolated endosperm in wheat. The development of isolated immature endosperm in the winter bread wheat variety ‘Kobra’, depending on the media composition, is presented in this study. Abscisic acid (ABA) is a key plant growth regulator for proper seed development. The addition of exogenous ABA had a positive impact on the size and ultrastructural features in isolated endosperm, especially of the outer aleurone-like cells. Furthermore, the content of starch in the endosperm cultured on a medium with ABA did not significantly differ from that of caryopsis at the same age, in contrast to soluble carbohydrates. Fluorescein diacetate (FDA) staining and confocal microscopy observation confirmed the viability of the cells from the outer layers. The analysis of internucleosomal fragmentation of DNA in the explants suggests the induction of programmed cell death (PCD) and DNA degradation typical of necrosis. We concluded that the development of isolated immature endosperm in bread wheat depends on the composition of the media. Thus, it could be a model for in vitro studies of this specific storage tissue and its response to culture conditions in bread wheat.

Programmed cell death (PCD) is a crucial phenomenon in the life cycle of cereal grains. In germinating grains, the scutellum allows the transport of nutrients from the starchy endosperm to the growing embryo, and therefore it may be the last grain tissue to undergo PCD. Thus, the aim of this work was to analyse whether the scutellum of wheat grains undergoes PCD and to perform a morphological and biochemical analysis of this process. Scutellum cells of grains following germination showed a progressive increase of DNA fragmentation, and the TUNEL assay showed that PCD extended in an apical-to-basal gradient along the scutellum affecting epidermal and parenchymal cells. Electron-transmission microscopy revealed high cytoplasm vacuolation, altered mitochondria, and the presence of double-membrane structures, which might constitute symptoms of vacuolar cell death, whereas the nucleus appeared lobed and had an increased heterochromatin content as the most distinctive features. An acid- and Zn(2+)-dependent nucleolytic activity was identified in nuclear extracts of scutellum cells undergoing PCD. This nuclease was not detected in grains imbibed in the presence of abscisic acid, which inhibited germination. This nucleolytic activity promoted DNA fragmentation in vitro on nuclei isolated from healthy cells, thus suggesting a main role in nucleus dismantling during PCD.  相似文献

小麦淀粉胚乳在发育过程中经历程序性细胞死亡(PCD).小麦淀粉胚乳的DNA在发育的特定阶段呈现梯状电泳条带,用乙烯处理使DNA片段化发生的时间提前,而且ABA处理虽然不能推迟DNA片段化的发生时间,但能减弱DNA片段化的程度.小麦淀粉胚乳细胞在PCD过程中出现某些动植物细胞凋亡的共同的结构变化特征,但也有一些独特的结构变化.如染色质凝聚后仅少数染色质块发生趋边化;细胞核在PCD过程中最先开始衰退,细胞核解体时胞质中有丰富的细胞器,细胞核解体后细胞并未死亡,在胞质中仍在合成和积累淀粉和储藏蛋白,直到细胞被淀粉充满,细胞才死亡;不形成凋亡小体,死亡的淀粉胚乳细胞成为营养物质的储藏库.因此小麦淀粉胚乳细胞的PCD是一种特殊形式的PCD.  相似文献
