Fucoxanthin–chlorophyll proteins (FCP) are the major light-harvesting proteins of diatom algae, a major contributor to marine carbon fixation. FCP complexes from representatives of centric (Cyclotella meneghiniana) and pennate (Phaeodactylum tricornutum) diatoms were prepared by sucrose gradient centrifugation and studied by means of electron microscopy followed by single particle analysis. The oligomeric FCP from a centric diatom were observed to take the form of unusual chain-like or circular shapes, a very unique supramolecular assembly for such antennas. The existence of the often disputed oligomeric form of FCP in pennate diatoms has been confirmed. Contrary to the centric diatom FCP, pennate diatom FCP oligomers are very similar to oligomeric antennas from related heterokont (Stramenopila) algae. Evolutionary aspects of the presence of novel light-harvesting protein arrangement in centric diatoms are discussed.  相似文献

Natural phytoplankton populations have been grown in outdoor continuous cultures at three dilution rates (D = 0.5, 0.25, and 0.1 · day?1) under nitrogen (N) or silicon (Si) limitation and two light intensities. At a high specific nutrient flux (high dilution rate) under N limitation an assemblage of primarily small, fast growing centric diatoms such as Skeletonema costatum (Grev.) Cleve and Chaetoceros spp. dominated with a low percentage of flagellates. At a low specific nutrient flux, a mixture of larger, slower growing centric diatoms, small flagellates, and pennate diatoms was obtained. Similar trends were observed under silicate limitation. Decreasing the light intensity at the lowest dilution rate selected for an assemblage similar to that observed at the high dilution rate and high light intensity.The results of these competition experiments suggest that specific nutrient flux (dilution rate) is an important factor in determining between group dominance (e.g., centric and pennate diatoms and small flagellates). Successful competitors representing broad phytoplankton groups can be arranged along a resource gradient of specific nutrient flux (dilution rate), with groups such as centric and pennate diatoms, represented as high and medium flux species, respectively.  相似文献

The rarely studied large diatom Stictocyclus stictodiscus (Grun.) R. Ross has been investigated by light and electron microscopy. Confusion over the classification of this diatom has been reduced, and it has been assigned to a new family, Stictocyclaceae, though the relationship of this to the overall classification of centric diatoms is still not clear.  相似文献

The diatoms are one of the best characterised algal groups. Despite this, little is known of the evolution of the group from the earliest cell to the myriad of taxa known today. Relationships among taxa at the family or generic level have been recognised in some diatoms. However, relationships at higher taxonomic levels are poorly understood and have often been strongly influenced by the first appearances of key taxa in the fossil record. An independent assessment of relationships among the diatoms at these higher taxonomic levels has been made using rRNA sequence data to infer phylogenetic relationships. In this paper we present an analysis of 18S rRNA data from several chosen centric, araphid and raphid pennate taxa. The phylogenetic inferences from these 18S rRNA sequences are supported by evidence from the fossil record and evidence from ontogenetic data. Ribosomal RNA data indicate that both the centric and araphid pennate lineages may not be monophyletic.  相似文献

Diatoms are highly diversified microeukaryotes with a taxonomy that has been under perpetual revision, particularly by means of molecular analyses. In this study, we evaluated typical genes to find the best molecular marker for the DNA taxonomy of centric diatoms, including Chaetoceros Ehrenberg, Cyclotella Kützing ex de Brébisson, Discostella Houk & Klee, Stephanodiscus Ehrenberg, and Thalassiosira Cleve. Our test genes included nuclear ribosomal RNA (e.g. small subunit, 5.8S, and large subunit [LSU]), and chloroplast genes (e.g. ribulose-1, 5-biphoshate carboxylase oxygenase and D1 protein of the photosystem II reaction centre core complex [psbA]). Calculated genetic divergence was highest for LSU ribosomal RNA D1-D5 (p-distance of 12.3), followed by 5.8S (7.7), ribulose-1, 5-biphoshate carboxylase oxygenase (7.4), small subunit (6.6), and psbA (3.7). The phylogenetic trees for individual genes effectively separated taxonomically tested centric diatoms with different phylogenetic resolutions; however, psbA was incongruent with others. These taxonomic resolving powers were in agreement with genetic divergences. Parsimony analysis showed that LSU evolved 1.97 times more rapidly than psbA and 1.07 times more rapidly than 5.8S. These results suggest that all of the tested genes except psbA can be used as taxonomic markers for centric diatoms and that LSU is the best molecular marker.  相似文献

Flow cytometric methods for recognizing several groups of eukaryotic marine phytoplankton were tested using 26 laboratory cultures. Each culture was divided into three aliquots, and these samples were analyzed for 1) Coulter volume; 2) light scatter (magnitude and polarization properties of forward scattered light and magnitude of right-angle scattered light) and autofluorescence emission (phycoerythrin and chlorophyll); and 3) autofluorescence excitation (by 488 nm and 515 nm light). Three kinds of cells could be easily distinguished from others in the culture collection: 1) The two cryptophytes and the rhodophyte had high phycoerythrin/chlorophyll ratios; 2) the two coccolithophores depolarized forward scattered light; and 3) the two pennate diatoms scattered only a relatively small amount of light in the forward direction compared with that at right angles. Mean chlorophyll fluorescence excited by blue light relative to that excited by green light was highest in the four chlorophytes, but there was overlap between some of these and some other kinds of cells. Unresolved cell types included centric diatoms, dinoflagellates, and naked coccolithophores. Forward light scatter and Coulter volume were closely related (except for the pennate diatoms) over a range of about 0.01 to 30 pL (equivalent spherical diameter about 3 to 40 microns), according to a logarithmic function.  相似文献

The electron microscopy study of algae from Lake Elgygytgyn and the water bodies of its basin has revealed 21 taxa of centric diatoms of specific and intraspecific ranks. Nine new species and varieties have been found, including Aulacoseira distans var. nivaloides, A. distans var. septentrionalis, Discostella guslyakovyi, which are rare for the flora of Russia; an endemic species (?) Pliocaenicus seckinae; and six interesting forms from the genus Aulacoseira identified only to the genus. The list of Centrophyceae of Beringia has been broadened.  相似文献

The abundance of micro-zooplankton (< 200 μm in size) was monitored weekly in Saanich Inlet throughout the winter, 1975–1976. Ciliate protozoans and metazoan nauplii were the major groups, with the former largely distributed in the top 5 m of the water column and the latter above 15 m. Average biomass throughout the winter was 3.1 mgC/m2 for ciliates above 5 m and 5.5 mgC/m2 for nauplii above 25 m, which made up 3.2 and 5.7% of the average phytoplankton biomass, respectively. They could consume over 30% of the phytoplankton production. Mesodinium sp. was an another abundant ciliate, but possibly a phyto-ciliate, with an average biomass of 8.2 mgC/m2 above 5 m. Major energy flow at lower trophic levels during the winter was characterized by a ‘nanoflagellate → micro-zooplankton’ system which was comparable with the ‘centric diatoms → macro-zooplankton’ system occuring commonly at the other seasons in the inlet.  相似文献

Besides the so-called ‘green lineage’ of eukaryotic photosynthetic organisms that include vascular plants, a huge variety of different algal groups exist that also harvest light by means of membrane intrinsic light harvesting proteins (Lhc). The main taxa of these algae are the Cryptophytes, Haptophytes, Dinophytes, Chromeridae and the Heterokonts, the latter including diatoms, brown algae, Xanthophyceae and Eustigmatophyceae amongst others. Despite the similarity in Lhc proteins between vascular plants and these algae, pigmentation is significantly different since no Chl b is bound, but often replaced by Chl c, and a large diversity in carotenoids functioning in light harvesting and/or photoprotection is present. Due to the presence of Chl c in most of the taxa the name ‘Chl c-containing organisms’ has become common, however, Chl b-less is more precise since some harbour Lhc proteins that only bind one type of Chl, Chl a. In recent years huge progress has been made about the occurrence and function of Lhc in diatoms, so-called fucoxanthin chlorophyll proteins (FCP), where also the first molecular structure became available recently. In addition, especially energy transfer amongst the unusual pigments bound was intensively studied in many of these groups. This review summarises the present knowledge about the molecular structure, the arrangement of the different Lhc in complexes, the excitation energy transfer abilities and the involvement in photoprotection of the different Lhc systems in the so-called Chl c-containing organisms.This article is part of a Special Issue entitled Light harvesting, edited by Dr. Roberta Croce.  相似文献

New for the Selenga River diatom species are discovered. The role of small-sized centric diatoms in different years and vegetation periods is assessed. 1. Contemporary limnological and palaeolimnological data from Piburger See (Eastern Alps, Austria) allowed the reconstruction of its trophic state since the late 19th century and the assessment of changes in phytoplankton biomass and species composition in relation to selected environmental parameters. 2. A radiometrically dated sediment core from Piburger See was analysed for geochemical parameters, spheroidal carbonaceous particles (SCPs), bacterial and algal pigments, and diatoms. The low SCP sediment inventory assigns Piburger See to the ‘cleaner’ sites in Europe with respect to fossil‐fuel related air pollution. The sedimentary pigment and diatom record reveals moderate eutrophication during the 20th century, followed by a slow re‐oligotrophication since the mid‐1980s because of lake restoration starting in 1970. 3. Epilimnetic temperature for Piburger See was reconstructed using air temperature records. A pronounced temperature increase has been recorded during the mid‐1940s and since the late‐20th century, both promoting algal growth and changes in species composition (e.g. increase in centric diatoms and recent bloom of Asterionella formosa). 4. Climate scenarios project additional substantial warming for this mountain lake by the end of the 21st century which will be most pronounced during the growing season. The predicted change in lake water temperature and thermal dynamics represents a key driver for the trophic and ecological status of Piburger See in the future.  相似文献

Relationships between marine diatoms and the bacterium Listeria monocytogenes have been studied by routine algological methods and high-resolution video-enhanced differential interference contrast light microscopy. The study showed that the relationship between the listeria and the benthic diatom Navicula sp. has a parasitic character, whereas the relationship between the listeria and the planktonic diatom Phaeodactylum tricornutum is protocooperative.  相似文献

Diatoms stand out among other microalgae due to the high diversity of species-specific silica frustules whose components (valves and girdle bands) are formed within the cell in special organelles called silica deposition vesicles (SDVs). Research on cell structure and morphogenesis of frustule elements in diatoms of different taxonomic groups has been carried out since the 1950s but is still relevant today. Here, cytological features and valve morphogenesis in the freshwater raphid pennate diatom Encyonema ventricosum (Agardh) Grunow have been studied using light and transmission electron microscopy of cleaned frustules and ultrathin sections of cells, and scanning electron and atomic force microscopy of the frustule surface. Data have been obtained on chloroplast structure: the pyrenoid is spherical, penetrated by a lamella (a stack of two thylakoids); the girdle lamella consists of several short lamellae. The basic stages of frustule morphogenesis characteristic of raphid pennate diatoms have been traced, with the presence of cytoskeletal elements near SDVs being observed throughout this process. Degradation of the plasmalemma and silicalemma is shown to take place when the newly formed valve is released into the space between sister cells. The role of vesicular transport and exocytosis in the gliding of pennate diatoms is discussed.  相似文献

cGametogenesis and auxospore development have been studied in detail in surprisingly few centric diatoms. We studied the development of sperm, eggs and auxospores in Actinocyclus sp., a radially symmetrical freshwater diatom collected from Japan, using LM and electron microscopy of living cultures and thin sections. Actinocyclus represents a deep branch of the 'radial centric' diatoms and should therefore contribute useful insights into the evolution of sexual reproduction in diatoms. Spermatogenesis was examined by LM and SEM and involved the formation of two spermatogonia (sperm mother-cells) in each spermatogonangium through an equal mitotic division. The spermatogonia produced a reduced 'lid' valve, resembling a large flat scale with irregular radial thickenings. Sperm formation was merogenous, producing four sperm per spermatogonium, which were released by dehiscence of the 'lid' valve. The sperm were spindle-shaped with numerous surface globules and, as usual for diatoms, the single anterior flagellum bore mastigonemes. One egg cell was produced per oogonium. Immature eggs produced a thin layer of circular silica scales before fertilization, while the eggs were still contained within the oogonium. Sperm were attracted in large numbers to each egg and were apparently able to contact the egg surface via a gap formed between the long hypotheca and shorter epitheca of the oogonium and a small underlying hole in the scale-case. Auxospores expanded isodiametrically and many new scales were added to its envelope during expansion. Finally, new slightly-domed initial valves were produced at right angles to the oogonium axis, after a strong contraction of the cell away from the auxospore wall. At different stages, Golgi bodies were associated with chloroplasts or mitochondria, contrasting with the constancy of Golgi-ER-mitochondrion (G-ER-M) units in some other centric diatoms, which has been suggested to have phylogenetic significance. Electron-dense bodies in the vacuole of Actinocyclus are probably acidocalcisomes containing polyphosphate.  相似文献

To explain photoperiodic induction of diapause in the spider mite Tetranychus urticae a new theoretical model was developed which took into account both the hourglass and rhythmic elements shown to be present in the photoperiodic reaction of these mites. It is emphasized that photoperiodic induction is the result of time measurement as well as the summation and integration of a number of successive photoperiodic cycles: the model, therefore, consists of separate ‘clock’ and ‘counter’ mechanisms. In current views involvement of the circadian system in photoperiodism is interpreted in terms of the hypothesis that the photoperiodic clock itself is based on one or more circadian oscillators. Here a different approach has been chosen as regards the role of the circadian system in photoperiodism: the possibility, previously put forward by other authors, that some aspect of the photoperiodic induction mechanism other than the clock is controlled by the circadian system was investigated by assuming a circadian influence on the photoperiodic counter mechanism. The derivation of this ‘hourglass timer oscillator counter’ model of photoperiodic induction in T. urticae is described and its operation demonstrated on the basis of a number of diel and nondiel photoperiods, with and without light interruptions.  相似文献

Light and scanning electron microscopy studies of centric diatoms of the plankton in the middle Irtysh River and waterbodies and water courses of its basin have revealed 21 taxa of Centrophyceae from 9 genera, including 14 new for the region. A revision of the species composition allowed us to specify and broaden the list of centric diatoms in the Omsk Priirtyshye up to 30 species.  相似文献

There is a considerable body of information regarding bacterially enhanced corrosion, however, this review focuses on diatoms (unicellular algae) whose contribution to biocorrosion is less well studied. The reasons why diatoms have been neglected in studies of biocorrosion in natural waters are discussed and the question whether diatoms should be considered as inert with respect of electrochemical processes is considered. A particular focus is given to the case of stainless steels (SS), which are widely used in variety of applications in natural waters. Basic information on the cell biology of diatoms is included in the review, particularly with respect to their ability to ‘sense’ and adhere to surfaces. Investigations at the nanoscale are reviewed as these studies provide information about the behavior of cells at interfaces. Recent advances include the use of atomic force microscopy (AFM), although only a few studies have been applied to diatoms. Regarding the electrochemical behavior of SS, the mechanisms by which diatoms influence the potential ennoblement process is discussed. Such studies reveal the association of diatoms, in addition to bacteria, with biocorrosion processes.  相似文献

Pollen grains from all thirteen species of the three genera to be recognised in a forthcoming revision of the Morinaceae, Acanthocalyx, Cryptothladia and Morina, have been investigated by light and scanning electron microscopy. Fresh pollen of M. longifolia has also been studied by light, scanning electron and transmission electron microscopy. Three main pollen types are described and a key provided for their identification. Each pollen type occurs throughout a genus. Further division of two of these pollen types enables groups of species to be recognised. Functional aspects of the pollen are discussed, especially their unique pre-germinative processes. A cladistic analysis of pollen characters provides a hypothesis for their evalution and supports the classification proposed in the recent revision.  相似文献
