S ummary : A method for selective counting and isolation of pectolytic clostridia in the presence of Erwinia carotovora is described; using this method pectolytic clostridia have been found in numbers of 8 × 105–1 × 108/g of rotting potato tissue in the presence of 1–4 × 108 E. carotovora /g.  相似文献

Methane-producing, cellulolytic, feather-degrading, and total anaerobic microbial populations were enumerated in four laboratory-scale (l l) thermophilic (50°C) poultry waste digesters over a 40d period. Four different operation conditions were: 5 d retention time (RT), 6% volatile solids (VS); 5 d RT, 3% VS; 10 d RT, 6% VS; and 10 d RT, 3% VS. Laying hen manure was the sole source of substrate and micro-organisms. At theoretical steady state (day 40) the biogas volumetric rate was near 3.0 l/l digester volume (l/l/d) in all but the 10 d RT, 3% VS digester which was 2 l/l/d. The total viable anaerobic population was > 106 cfu/ml digester fluid at the first sampling and stabilized at 107–108 cfu/ml between days 20 and 40 in all digesters. Methane-producing bacteria increased from ≤ 10/ml early in the sampling period to 105/ml at steady state in all but the 5 d RT, 3% VS digester which was highest at 107/ml. Cellulolytic micro-organisms were low throughout the 40 d, generally less than 10/ml. Feather-degrading micro-organisms ranged from near 102–105 at steady state and were decreasing in number near day 40 in all but the 10 d RT, 6% VS digester which maintained 105/ml after day 20. A feather-degrading culture was enriched from this digester and subsequently adapted to grow in a medium with feather as the sole source of carbon. Results of this study provide information regarding potential biological upgrading of poultry waste digesters for increased operational efficiency and potential industrial application of a feather-hydrolytic micro-organism.  相似文献

Bacteria grew to viable populations of 108–109 cfu/g during the fermentation of soybeans into tempe with the fungus, Rhizopus oligosporus. Bacillus pumilus and B. brevis were the predominant bacterial species, reaching populations of approximately 108 cfu/g during the 48 h fermentation. Species of Streptococcus faecium, Lactobacillus casei, Klebsiella pneumoniae and Enterobacter cloacae also contributed to the fermentation and achieved populations of 106–107 cfu/g. and accepted 25 May 1989  相似文献

Aflatoxin content and number of fungi in poultry feedstuffs from Indonesia   总被引:1,自引:1,他引:0  
The content of aflatoxin and associated fungi was determined in 56 samples, including 34 of corn, 10 of soybean meal, nine of rice bran and three of broken rice, collected from different poultry farms and poultry feedmills situated around Jakarta-Bogor, Indonesia.
Ninety-one per cent of the corn samples contained aflatoxins and the total concentration ranged from 22 to 6171 μ g/kg. With rice bran, 100% of the samples were positive for aflatoxin B1, ranging from 36 to 71 μ g/kg. No aflatoxin was detected in samples of soybean meal or broken rice. All the samples were contaminated by several fungi (8 times 103–5 times 106cfu/g) and further identification was limited to Aspergillus flavus and A. parasiticus. The dominant species was A. flavus (2 times 103–4 times 106cfu/g in corn samples, 1·0 times 103–1·0 times 105cfu/g in soybean meal, 2 times 104–4·4 times 105cfu/g in rice bran and 2 times 104–6 times 104cfu/g in broken rice). A simple and sensitive method was developed to replace the need for complex and laborious DNA extraction to remove inhibitory substances in potato tuber peel extract before detection of Erwinia carotovora subsp. atroseptica (Eca) by PCR. Eca was enriched by a factor of 105 when peel extract was inoculated onto a selective medium, CVP, and incubated at 27°C for 24 h. Bacterial micro-colonies which developed were suspended in 500 μl of water and the bacteria diluted in water 100-fold, or 10-fold followed by washing by centrifugation, before PCR testing. The sensitivity of detection obtained with the former was ca 101–102 cells ml−1 and with the latter ca 101 cells ml−1, when different numbers of streptomycin-resistant Eca strain were added to peel extract from three Eca-free potato cultivars. The method was validated and the sensitivity confirmed relative to two different commonly used Eca detection methods using naturally contaminated tubers.  相似文献

This was a preliminary investigation to define the conditions of colonization of a human skin equivalent (SE) model with cutaneous microorganisms. SEs of 24 mm diameter were constructed with a dermal matrix of fibrin containing fibroblasts and a stratified epidermis. Microbial colonization of the SEs was carried out in a dry environment, comparable to ' in vivo ' skin, using a blotting technique to remove inoculation fluid. The microbial communities were sampled by scrub washing and viable cells enumerated on selective growth medium. Staphylococcus epidermidis , Propionibacterium acnes and Malassezia furfur (human skin commensals) and Staphylococcus aureus (transient pathogen) were colonized at inoculum densities of 102–106 CFU SE−1 on the surface of replicate SEs. Growth of all species was supported for upto 72–120 h, with recovery densities of between 104–109 CFU SE−1. A novel, real-time growth monitoring method was also developed, using S. aureus containing a lux cassette. Light output increased from 20 to 95 h, and colonization increased from 102 to 108 CFU SE−1, as confirmed by conventional recovery. Thus, the SE model has potential to investigate interactions between resident and transient microbial communities with themselves and their habitat, and for testing treatments to control pathogen colonization of human skin.  相似文献

The number of aerobic bacteria in a blanket peat decreased with depth from 26 times 106/g dry peat in the surface layers to 0.5 times 106/g dry peat at 30–40 cm down the profile, thereafter remaining roughly constant. Obligate psychrophiles comprised <2.5% of this population. Anaerobes were most numerous, 9 times 106/g dry peat at 6–10 cm depth, decreasing to 0.5 times 106/g at 20–30 cm. Calculations indicated that these counts, 103–104-fold lower than the direct counts, substantially underestimated the active microbial population. Gram negative rods, the predominant aerobes in the surface layers, were replaced by unidentified Bacillus strains at 10–20 cm depth but became increasingly more numerous further down the profile. The Gram negative rods were the most numerous organisms/m2 but the Bacillus strains, one third of which were present as spores, made the largest contribution to the biomass/m2. Gram positive cocci, Arthrobacter and, infrequently, Nocardia were also isolated. Actinomyces -like forms were the predominant obligate anaerobes and were approximately three times more numerous than clostridia and a curved Gram negative rod.  相似文献

Relation between Aeromonas and faecal coliforms in fresh waters   总被引:2,自引:2,他引:0  
A possible correlation between the presence of mesophilic aeromonads and the number of faecal coliforms present in three fresh-water habitats subject to differing levels of faecal pollution was investigated. Concentration of Aeromonas spp. between 102–109 cfu/100 ml and faecal coliforms of between 9–107 cfu/100 ml were found in the waters. In water free from faecal pollution there was no correlation but in polluted waters there was a significant relationship between the numbers of aeromonads, faecal coliform and the concentration of organic matter measured by biological oxygen demand.  相似文献

The survival of Ralstonia solanacearum in naturally infested sandy loam soil under irrigated rice culture was investigated in Sankhu village (1400 m above sea level) in central Nepal. The experimental plot had a previous history of bacterial wilt and a range of 1.5 × 104–3 × 104 colony-forming units (CFU) per g soil was present. The survival of R. solanacearum was monitored in roots of naturally growing aquatic weeds in the rice plot and in soil before and after rice harvest. The incidence of the bacterial infection in the weeds, Dopatrium sp. and Monochoria vaginalis , were 57.5 and 10%, respectively. The bacterial population detected in soil before rice harvest was 1.5 × 104 CFU per g soil whereas a range of 7.5 × 102–1.5 × 103 CFU per g was detected after the rice harvest. Biovar typing of R. solanacearum isolated from potato plants, potato tubers, aquatic weeds, and the soil from the experimental plot yielded the diverse biovars 2 A, 3 and 4. This is the first report of the survival of these biovars in soil, which was under continuous flow of irrigation water for 3 months during rice culture.  相似文献

The dense microbial flora of the rabbit caecum consisted chiefly of bacteria (1011/g) with small numbers of yeast cells (106/g). Using strictly anaerobic techniques, 23% of the direct microscopic cell count was cultivated and 55% of the cultivatable bacteria utilized ammonia as the sole source of nitrogen. Ureolytic bacteria were isolated from the caecal lumen and mucosa and were identified as Bacteroides vulgatus, Clostridium clostridiiforme, Bacillus spp. and Staphylococcus spp. Ammonia assimilation by the bacterial flora of the caecum was by incorporation into α-oxoglutarate catalysed by NADPH-linked glutamate dehydrogenase.  相似文献

Tests were carried out to determine the effect of manufacturing procedures for a Camembert-type cheese from raw goats' milk on the growth and survival of Staphylococcus aureus organisms added to milk at the start of the process, and to study the possible presence of staphylococcal enterotoxin A in these cheeses. The initial staphylococcal counts were, respectively, 2, 3, 4, 5 and 6 log cfu ml−1. Cheese was prepared following the industrial specifications and ripened for 41 d. Detection of enterotoxins was done by the Vidas SET test and by an indirect double-sandwich ELISA technique using antienterotoxin monoclonal antibodies. Generally, numbers of microbes increased at a similar rate during manufacture in all cheeses until salting. During the ripening period, the aerobic plate count population and Staph. aureus levels remained stable and high. There was an approximately 1 log reduction of Staph. aureus in cheeses made with an initial inoculum of Staph. aureus greater than 103 cfu ml−1 at the end of the ripening period (41 d) compared with the count at 22 h. The level of staphylococcal enterotoxin A recovered varied from 1 to 3·2 ng g−1 of cheese made with an initial population of 103–106 cfu ml−1. No trace of enterotoxin A was detected in cheeses made with the lowest Staph. aureus inoculum used in this study.  相似文献

A simple, rapid and sensitive PCR-based method was developed for the detection of all five subspecies of Erwinia carotovora , including subsp. carotovora and subsp. atroseptica , and all pathovars/biovars of Erwinia chrysanthemi , on plant tissue culture material. Primers SR3F and SR1cR, based on a conserved region of the 16S rRNA gene, amplified a DNA fragment of 119 bp from all 65 such strains tested. Detection limits of the method in vitro were 2·0 × 102–3·4 × 103 cfu ml−1 (equivalent to 1–17 cfu per PCR) and, following extraction of genomic DNA from plant extract, detection limits were 2·3 × 102–1·9 × 104 cfu per microplant sample (equivalent to 5 cfu – 3·8 × 102 cfu per PCR). To improve the sensitivity of the method in planta , to obviate the need for complex and laborious DNA extractions, and to remove inhibitory substances present in the plant extract, an enrichment step was included prior to PCR. Following enrichment, the sensitivity of detection was <10 cfu per microplant sample. This method provides the first sensitive means of detecting latent infection caused by several economically important soft rot erwinias simultaneously on potato tissue culture material.  相似文献

Growth of Bacillus cereus NCIB 8579 was studied on four varieties of rice with and without tapé fermentation. Fermented and unfermented rice supported growth of B. cereus to 107–109cfu/g. With fermentation the pH fell and numbers of B. cereus remained high ( ca 108cfu/g) except on black glutinous rice where numbers declined. Cells added at different fermentation times survived less well as fermentation progressed and the pH fell. Once growth on rice is established, B. cereus is able to survive fermentation, probably as spores.  相似文献

The development of the microflora of smoked pork loin and frankfurter sausage was followed during storage in vacuum, N2 and CO2 atmospheres at 4°C. The total aerobic count on the smoked pork loin reached 107 organisms/g after 37 d in vacuum, 43 d in N2 and 49 d in CO2. The corresponding value for the sausage was 77 d in vacuum, while the growth stopped at 6 times 104 organisms/g after 98 d in N2, and at 4 times 102 organisms/g after 48 d in CO2.
The predominant organisms on the fresh products were Bacillus spp., coryneform bacteria, Flavobacterium spp. and Pseudomonas spp.
At the end of the storage time the microflora on both products in the three gas atmospheres, consisted mainly of Lactobacillus spp. and two large groups of organisms that could not be identified as any described genus. Some of the unidentified strains could be classified as a Lactobacillus sp. after subsequent subculturing on laboratory media.
The numbers of Lactobacillus spp. at the end of storage decreased in the order, CO2 > N2 > vacuum. Lactobacillus viridescens generally constituted a substantial part of the Lactobacillus flora (5–72%). On the sausages two large uniform groups of unidentifiable homofermentative Lactobacillus spp. were also found.  相似文献

The sludge from hospital waste treatment facilities is a potential source of infectious organisms. The average numbers of micro-organisms in the sludge of hospital wastewater in Taiwan were as follows: total count 8·1 × 107 cfu g−1 (dry weight of sludge), and 1·4 × 106, 3·6 × 105, 1·6 × 105, 2·2 × 105 and 5·5 × 104 cfu g−1 (dry weight of sludge) for total coliforms, faecal coliforms, faecal streptococci, Pseudomonas aeruginosa and Salmonella spp., respectively . Salmonella spp. were detected in 37% (10 of 27) of the sludges from hospital wastewaters. Therefore, the treatment of such sludge to reduce pathogenic micro-organisms should be considered.  相似文献

Four media were tested for their ability to detect the soft rot potato pathogens Erwinia chrysanthemi (Ech) and Erwinia carotovora ssp. atroseptica (Eca) in potato tubers by means of automated conductance measurements. The specificity of the conductimetric assays was determined by testing a set of different Erwinia spp. and potato-associated saprophytes, including the genera Pseudomonas, Bacillus, Enterobacter and Flavobacterium. All bacteria tested produced conductance responses in Special Peptone Yeast Extract, whereas in minimal medium with L-asparagine only Erwinia spp. and Pseudomonas spp. were able to generate large conductance responses. In minimal medium supplemented with glucose and trimethylamine- N -oxide only Enterobacteriaceae, Erwinia spp. included, generated conductance responses, while with pectate as sole carbon source only Erwinia spp. produced distinct conductance responses. The pectate medium proved to be particularly useful for specific automated conductimetric detection of Erwinia spp. in potato peel extracts. Within 48 h, the detection threshold of the conductimetric assay for Eca varied between 102 and 103 cfu per ml peel extract at both incubation temperatures of 20° and 26°C. Ech was detected at concentrations of 104–105 or 103–104 cfu ml-1 at 20° and 26°C, respectively. To eliminate 'false'-positive reactions in conductimetry caused by Erwinia carotovora ssp. carotovora , results of the conductance measurements have to be confirmed by other techniques, like serology or DNA assays.  相似文献

The Limulus lysate test (LLT) for endotoxin assay has been found to be an excellent, simple and rapid test of microbial quality of refrigerated ground beef. In fresh ground beef held at 5°C for 7–12 d, LLT titres increased from 102–105 and correlated very highly with extract-release volume (ERV) data and total viable Gram negative counts at both 5° and 30°C. The LLT was negative for fresh beef containing low numbers of bacteria and on aged beef in the absence of increasing numbers of Gram negative bacteria. Of 14 Gram negative meat isolates, all gave a positive LLT while none of eight miscellaneous Gram positive bacteria did. The use of this test provides objective information on the microbial quality of fresh refrigerated ground meats in 1 h. Based upon this study, it is suggested that a 0·1 ml inoculum from a 103 dilution of good quality ground beef should produce a negative lysate test and thus serve as an additional rapid screening test of meat microbial quality.  相似文献

Studies have been carried out in which growth patterns of a Salmonella sp. and competing micro-organisms, especially other Enterobacteriaceae, were followed during pre-enrichment in buffered peptone water (BPw) and subsequent selective enrichment in tetrathionate broth (TBB). Pre-enrichment cultures were inoculated with minced meat and three reference samples containing nalidixic acid-resistant salmonellas. Irrespective of their initial numbers in BPw, Enterobacteriaceae increased to 108/ml or more. During incubation in TBB at 43C, numbers of lactose-positive Enterobacteriaceae decreased in most enrichments which resulted in a positive salmonella isolation, but remained constant in the majority of those that did not. Levels of lactose-negative Enterobacteriaceae did not decrease in most salmonella-positive tests, but did so in half of the salmonella-negative ones. In the salmonella-positive tests the numbers of salmonellas had increased to 103–107/ml in BPw and after transfer to TBB slowly reached 104/ml or more. In all cases the numbers of salmonellas exceeded those of the competing flora on brilliant green agar (BGA). In the salmonella-negative tests the numbers of salmonellas had increased less in BPw and decreased in most of the TBB enrichments. In none of these negative tests did the numbers of salmonellas exceed those of the competing flora on BGA. Escherichia coli dominated in most of the salmonella-negative tests. The results suggest more influence of lactose-positive than lactose-negative Enterobacteriaceae on the detection of salmonellas. The effect of competing microorganisms seems to depend not only upon their initial numbers, but also upon the types that can interact with salmonellas during selective enrichment.  相似文献

Abstract Electrofusion of protoplasts of two mutant strains of Hansenula polymorpha resulted in high fusion and hybrid yields when the calcium ions present in the conventional fusion medium replaced by zinc ions. The optimal fusion conditions were an alignment field of 0.4 kV cm−1 strength and 2 MHz frequency for 30 s, followed by two consecutive pulses of 12 kV cm−1 strength and 15 μs duration. With 0.05–0.1 mM zinc ions in the fusion medium an average clone number of 104–105 clones per 108 input cells was reached. The presence of about 0.6 mM magnesium ions in the zinc fusion medium was essential.  相似文献

Abstract An examination of samples obtained from a commercial fish smoker, using seawater agar with incubation at 4°, 15° and 37°C for up to 28 days, revealed the presence of large bacterial populations in smoked fish. However, initially only low bacterial numbers, i.e., 2 × 103/g, were present in the muscle of fresh, whole haddock ( Melanogrammus aeglefinus ). With filleting, there was a sudden increase in numbers to 9.2 × 105/g. Yet immediately after smoking, the bacterial populations decreased (5 × 105/g), followed by a gradual increase with storage (e.g., 2 × 106/g after 24 h). Representative colonies were presumptively identified as Acinetobacter, Alcaligenes , coryneforms, Pseudomonas and Vibrio spp.  相似文献
