Ceratocystis fimbriata is a fungal plant pathogen that causes black rot on Ipomoea batatas. Based on inoculation studies on numerous tree species, the pathogen is known to be host specific. The closely related species, Ceratocystis manginecans, causes severe wilt on a broad range of tree hosts, including Mangifera indica, Acacia mangium and other leguminous tree species. The genetic factors underlying the pathogenicity and host specificity of Ceratocystis species have rarely been investigated. In this study, an F1 population of 70 recombinant progeny from a cross between C. fimbriata and C. manginecans was generated and the inheritance of various phenotypic traits was investigated. Results showed that colony colour, growth rate, asexual spore production and aggressiveness to I. batatas and A. mangium are all quantitative traits with high levels of heritability. However, conidia production and aggressiveness appeared to be regulated by a small number of genes. No correlation could be found between aggressiveness and other phenotypic traits, suggesting that these are inherited independently. This is the first study to consider genetic inheritance of pathogenicity and host specificity in Ceratocystis species and the results will contribute, in future, to the identification of quantitative trait loci and candidate genes associated with the traits investigated.  相似文献

The emergence of wattle wilt disease on non-native Acacia mearnsii trees in Africa, caused by the indigenous fungus Ceratocystis albifundus, has highlighted a need to better understand the diversity, ecology and distribution of Ceratocystis species in natural African environments. In this study we applied phylogenetic inference to identify and characterize isolates of Ceratocystis collected in a natural savanna ecosystem in South Africa. Three new species were recognized and are described as C. cryptoformis sp. nov. in the C. moniliformis complex, as well as C. thulamelensis sp. nov. and C. zambeziensis sp. nov., both residing in the C. fimbriata complex. Incorporating the new species into global phylogenies of Ceratocystis provided insights into the patterns of evolution and biogeography of this group of fungi. Notably, the African continent was identified as an important centre of diversification of Ceratocystis spp., from which several lineages of these fungi were shown to have radiated.  相似文献

The genus Ceratocystis includes important fungal pathogens of trees, including Eucalyptus spp. Ironically, very little is known regarding the diversity or biology of Ceratocystis species on Eucalyptus species in Australia, where most of these trees are native. The aim of this study was to survey for Ceratocystis spp., and their possible insect associates, on eucalypts in Australia and thus to establish a foundation of knowledge regarding these fungi on the continent. Collections were made in three states of Australia from wounds on trees, as well as from nitidulid beetles associated with these wounds. Ceratocystis spp. were identified based on morphology and multigene sequence comparisons. Of the 54 isolates obtained, two previously unknown species of Ceratocystis were found and these are described here as Ceratocystis corymbiicola sp. nov. and Ceratocystis tyalla sp. nov. Furthermore, the distribution of Ceratocystis pirilliformis is expanded to include Eucalyptus spp. in Tasmania.  相似文献

Twenty seven rhizobial strains associated with Acacia saligna grown in northern and southern Algeria were characterized, including generation time, host-range, the 16S rRNA gene and 16S–23S rRNA intergenic spacer restriction patterns, 16S rRNA gene sequence analysis and tolerance to salinity and drought. Cross inoculation tests indicated that 11 slow-growing isolates from northern nurseries were able to nodulate introduced Australian acacias exclusively, whereas 16 fast-growing isolates, mainly from southern nurseries, were capable of also nodulating native acacias. Restriction patterns and sequence analysis of the 16S rRNA gene showed that strains of the first group belonged to Bradyrhizobium while strains of the second group were related to Sinorhizobium meliloti and Rhizobium gallicum. Interestingly, five strains of the first group formed a distinct cluster phylogenetically close to Bradyrhizobium betae, a non-nodulating species causing tumour-like deformations in sugar beet roots. Bradyrhizobium strains were in general more sensitive to NaCl and PEG than the S. meliloti and R. gallicum representatives. Among the latter, strains S. meliloti BEC1 and R. gallicum DJA2 were able to tolerate up to 1 M NaCl and 20% PEG. This, together with their wide host-range among Acacia species, make them good candidates for developing inoculants for A. saligna and other acacia trees growing in arid areas.  相似文献

There is large potential in the establishment of Acacia auriculiformis × Acacia mangium hybrid plantations through selection of high quality parents especially with characteristics favourable for some end products like pulp and fibre. Trees altered in their lignin profile with reduced amounts of lignin or increased extractable syringyl are desirable in the pulp and paper industry. Cinnamoyl-CoA reductase (CCR) and caffeic acid O-methyltransferase (COMT) are two enzymes likely to regulate lignin content and composition in the syringyl (S)- and guaiacyl (G)-related monolignols at specific branches of the lignin biosynthetic pathway. A goal of this study was to discover the genetic variation in CCR and COMT genes in Acacia species using single nucleotide polymorphisms (SNPs). In this study, two lignin genes, CCR (3,317 bp) and COMT (2,764 bp), were isolated from the Acacia hybrid, sequenced and analysed in silico. Southern blot analysis suggested that there are one to two copies of genes encoding CCR and COMT in the Acacia hybrid. Upon genotyping 480 individuals from natural populations of A. auriculiformis and A. mangium, six CCR SNPs were found in A. auriculiformis and five CCR SNPs in A. mangium. Three COMT SNPs were found in A. auriculiformis and one COMT SNP in A. mangium. A pair of CCR SNPs showed high linkage disequlibrium (LD) with r 2 value of 0.870 in an A. mangium population from Papua New Guinea. The SNPs will be further exploited through the candidate gene-based LD mapping to identify QTLs for the Acacia marker-assisted breeding. This is the first documentation of the isolation, cloning and re-sequencing of CCR and COMT genes in the Acacia hybrid and the natural populations of A. auriculiformis and A. mangium.  相似文献

A method for the bioassay of Enthomophthora spp. against aphids is described. Twenty-four isolates comprising five species of fungi were screened for activity against Therioaphis trifolii f. maculata. The only isolates with a high level of activity were those of E. sphaerosperma obtained from aphids. The initial bioassays with E. sphaerosperma indicated that aphids, starved for 24 hr during inoculation with E. sphaerosperma primary spores, were less susceptible than those removed from the plants, for just the period of exposure to the primary spore shower. Using the latter procedure, five assays of the most pathogenic isolate gave a mean LC50 of 11.3 primary spores/mm2 while bioassays of four other isolates gave LC50 values ranging from 15.7 to 27.3 spores/mm2. The potential of E. sphaerosperma as a microbial control agent for T. trifolii f. maculata in Australia is discussed.  相似文献

Australian species of the genus Acacia are amongst the most invasive trees. As nitrogen fixers, they are able to invade oligotrophic ecosystems and alter ecosystem functioning to their benefit. We aimed to answer three questions: How does early Acacia invasion influence nitrogen and light in a sandy savanna? How does early Acacia invasion impact biodiversity? Does early invasion alter ecosystem functioning towards the dominance of Acacia? We analyzed (using generalized linear mixed models and richness estimators) paired plots focused on plants of Acacia mangium (Fabaceae) and plants of Marcetia taxifolia (Melastomataceae) by taking hemispherical photos and sampling plants, leaves and soil for measurements of light, richness, leaf nitrogen, leaf δ15N, soil nitrogen and soil coarse sand. The results suggest that early Acacia invasion alters the control of soil and of leaf nitrogen and increases shading, enabling a much wider range of light variation. The δ15N results suggest that the nitrogen taken up by Acacia is transferred to neighboring plants and influences the light environment, suggesting facilitation. The enrichment of plant species observed during early Acacia invasion is consistent with the wider range of light variation, but the forecasted leaf nitrogen conditions during the established phase of Acacia invasion might cause loss of light-demanding species because of increased shading. If early Acacia invasion turns into an established phase with highly increased shading, Acacia seedlings might be favored and ecosystem functioning might change towards its dominance.  相似文献

The growth response of Acacia mangium Willd. to inoculation with selected Bradyrhizobium strains was investigated in two field trials in the Ivory Coast (West Africa). In the first trial (Anguededou), four provenances (i.e., trees originating from seeds harvested in different geographical areas) of A. mangium were inoculated with four Bradyrhizobium strains from different origins. Six months after being transplanted in the field, the heights of all inoculated trees showed a statistically significant increase of 9 to 26% compared with those of uninoculated trees, with the most effective strain being Aust 13c. After 19 months, the positive effect of inoculation on tree growth was confirmed. The effect of A. mangium provenance on tree growth was also highly significant. Trees from the Oriomo provenance of Papua New Guinea had a mean height that was 25% greater than those of other provenances. Analysis of variance showed a highly significant effect of interaction between strain and host provenance factors. Thus, most effective strain × provenance combinations could be proposed. Immunological identification of strains clearly showed that 90 to 100% of nodules from trees inoculated with three of the four Bradyrhizobium strains or from uninoculated trees contained exclusively Aust 13c 23 months after tree transplantation. This predominance of Aust 13c in nodules was still observed 42 months after tree transplantation. The second experiment (Port-Bouët), performed with a different soil, confirmed the long-term positive effect of Aust 13c on plant growth, its high competitive ability against indigenous strains, and its persistence in soil. Strain Aust 13c should thus be of great interest for inoculating A. mangium under a wide range of field conditions.  相似文献

The cyanogenic glycoside, proacacipetalin, is reported from Acacia pachyphloia (Acacia subgenus Acacia). This represents the first record of a glycoside with an aliphatic aglycone from a species of Acacia indigenous to Australia. Actinobacillus spp. are Gram-negative bacteria associated with mucosal membranes. While some are commensals, others can cause important human and animal diseases. A. pleuropneumoniae causes severe fibrinous hemorrhagic pneumonia in swine but not systemic disease whereas other species invade resulting in septicemia and death. To understand the invasive phenotype of Actinobacillus spp., complete genomes of eight isolates were obtained and pseudogenomes of five isolates were assembled and annotated. Phylogenetically, A. suis isolates clustered by surface antigen type and were more closely related to the invasive A. ureae, A. equuli equuli, and A. capsulatus than to the other swine pathogen, A. pleuropneumoniae. Using the LS-BSR pipeline, 251 putative virulence genes associated with serum resistance and invasion were detected.To our knowledge, this is the first genome-wide study of the genus Actinobacillus and should contribute to a better understanding of host tropism and mechanisms of invasion of pathogenic Actinobacillus and related genera.  相似文献

To determine the effectiveness of rehabilitation on improving ecosystem functions, we examined net photosynthetic rate (P N), tree species composition, soil enzyme activities, and the microclimate (air and soil temperature, relative humidity) of an area on Mt. Makiling that has been rehabilitated and protected from fire for over 12 years. After it was last burned extensively in 1991, restoration was initiated by planting Acacia mangium and Acacia auriculiformis. We selected three areas to study in 2003. Two areas were rehabilitated with A. mangium and A. auriculiformis, and one was still dominated by Imperata cylindrica and Saccharum spontaneum. P N of A. mangium and A. auriculiformis showed significantly lower values than those of I. cylindrica and S. spontaneum. The Acacia plantations had more naturally regenerated tree species than the grassland. Additionally, more tree species appeared in the A. mangium plantation than in the A. auriculiformis plantation. Ficus spetica was present in all of the study sites. Dehydrogenase and phosphatase activities were significantly higher in soil under the Acacia plantations than under grassland. Grassland showed higher air temperature, relative humidity, and soil temperature as well as a larger variation per hour in these parameters compared to the Acacia plantations. The highest air temperature, relative humidity, and soil temperature were measured in April during the dry season. From the regression analysis, soil temperature was significantly correlated with air temperature. Hence plantations, as a rehabilitation activity for grassland, promote natural regeneration and stabilize the microclimate. This stabilization of the microclimate affects establishment and growth of naturally occurring tree species.  相似文献

Ceratocystis fimbriata Ellis & Halsted recently was recorded causing seed and seedling blight on Carapa guianensis Aubl. (andiroba), a tree species native to the Amazon Rainforest and prized for its valuable timber and medicinal seed oil. C. fimbriata more commonly causes wilt type diseases in woody hosts, especially on non-native host trees. However, on andiroba the disease occurs on seedlings and seeds, affecting the species regeneration. We studied 73 isolates of C. fimbriata on andiroba from three regions of the Amazon Basin to see if they represented natural or introduced populations. Analysis of ITS rDNA sequences and phylogenetic analysis of mating type genes revealed new haplotypes of C. fimbriata from the Latin American Clade that were closely related to other Brazilian populations of the fungus. In mating experiments, andiroba isolates were inter-fertile with tester strains of C. fimbriata from Brazil and elsewhere, confirming that they belong to a single biological species. Using microsatellite markers, 14 genotypes and populations with intermediate levels of genetic variability were found, suggesting that the fungus is indigenous to the Amazon Basin. Inoculation tests indicated that the andiroba isolates are host-specialized on andiroba, supporting the proposition of the special form C. fimbriata f. sp. carapa.  相似文献

The aim of this study was to determine the effectiveness of the mycorrhizal inoculation on the early development of Australian acacias in a dry tropical environment. Field experiments were carried out with an Australian Acacia species (Acacia holosericea) and four fungal strains were tested: an arbuscular mycorrhizal (AM) fungus (Glomus intraradices) and three ectomycorrhizal isolates belonging to the Pisolithus and Scleroderma genera. Mycorrhizal inoculation has significantly enhanced the growth of A. holosericea seedlings after 4 months of culturing in the greenhouse conditions in a disinfected soil. Mycorrhized and non-mycorrhized (control) plants were then transferred into the field. One of the experiments (Experiment EC5) was used to determine allometric relations based on stem diameter by destructive sampling. Two forms of biomass equations were generated and compared: (i) form 1 equation incorporated only stem diameter as the independent variable and form 2 equation incorporated stem diameter and height (product of square of stem diameter and height, D2H) as the independent variables. As no distinctive advantage of one form over the other was recorded, simplest equation (Eq. (1)) was used for above-ground biomass calculations. Mycorrhizal inoculations with one Pisolithus isolate (P. albus IR100) or G. intraradices have significantly improved the growth of A. holosericea after 18 months as well as after 30 months of plantation. These results show that mycorrhizal inoculation could be of great relevance to improvement of the reafforestation process of degraded soils with leguminous fast growing trees, more particularly in Sahelian ecozones.  相似文献

Mating is central to many fungal life cycles and is controlled by genes at the mating-type (MAT) locus. These genes determine whether the fungus will be self-sterile (heterothallic) or self-fertile (homothallic). Species in the ascomycete family Ceratocystidaceae have different mating strategies, making them interesting to consider with regards to their MAT loci. The aim of this study was to compare the composition of the MAT locus flanking regions in 11 species of Ceratocystidaceae representing four genera. Genome assemblies for each species were examined to identify the MAT locus and determine the structure of the flanking regions. Large contigs containing the MAT locus were then functionally annotated and analysed for the presence of transposable elements. Genes typically flanking the MAT locus in sordariomycetes were found to be highly conserved in the Ceratocystidaceae. The different genera in the Ceratocystidaceae displayed little synteny outside of the immediate MAT locus flanking genes. Even though species ofCeratocystis did not show much synteny outside of the immediate MAT locus flanking genes, species of Huntiella and Endoconidiophora were comparatively syntenic. Due to the high number of transposable elements present in Ceratocystis MAT flanking regions, we hypothesise that Ceratocystis species may have undergone recombination in this region.  相似文献

Human-induced disturbances and wildfires can transform areas of tropical rainforest into Imperata-dominated grassland, but it may be possible that recovery of biodiversity is facilitated by reforestation with fast-growing trees. We compared the assemblages of braconid wasps as parasitoids of taxonomically diverse groups of insects among Imperata grasslands, young and mature plantations of Acacia mangium, young secondary forests after wildfires, and old secondary forests in the lowland of East Kalimantan. The abundance and species richness of braconids, which had declined in Imperata grasslands, somewhat increased in Acacia plantations, and also the species composition of braconids in Acacia plantations was transitional between Imperata grasslands and old secondary forests. Parasitoids of detritivores and wood borers increased markedly after plantation, while those of herbivores showed a distinct turnover of species all over the range from grasslands to old secondary forests. The plantation of A. mangium had most likely facilitated the recovery of the diversity of host forest and their parasitoids, but the recovery was just at the rudimentary stage even in mature plantations. Monitoring of parasitic wasps would be useful to test the continuous recovery of forest biodiversity in plantation stands.  相似文献

The leaves of Acacia species have been found to contain homoarginine, pipecolic acid and 4-hydroxy-pipecolic acid. The nymphs of the tree locust Anacridium melanorhodon, which feed on the leaves of Acacia species, were not inhibited from feeding on palatable media containing concentrations of these amino acids equivalent to, or greater than, those found in the leaves. The graminivorous Locusta migratoria was more sensitive to these compounds, inhibitory effects being observed at concentrations comparable to those found in the leaves. The inhibitory effects of mixtures of homoarginine and pipecolic acid were additive in A. melanorhodon but not in L. migratoria. Three of the non-protein amino acids found in the seeds of Acacia species, 2,3-diaminopropionic acid, 2-amino-3-acetylaminopropionic acid and 2-amino-3-oxalylaminopropionic acid, were more effective inhibitors of feeding in Anacridium than were the leaf amino acids.  相似文献

The entomopathogenic fungi Zoophthora radicans and Pandora blunckii co-occur in field populations of Plutella xylostella and, therefore, are likely to interact during the infection process. We have investigated the possible outcomes of these interactions in the laboratory. Using four isolates, two of each fungal species, inter-specific interaction experiments were done in Petri dishes and on intact plants. In Petri dish experiments, larvae were inoculated directly using sporulating mats of mycelium, both species had the same opportunity to infect and only the relative concentration of conidia of each pathogen species applied was manipulated. In the intact plant experiments, larvae were placed onto fungus-contaminated plants, inoculation was passive and the probability of infection by either or both species of fungi depended on larval activity and proximity to inoculum. In the Petri dish experiment, the species with the largest concentration of conidia out-competed the other regardless of virulence, and results were similar in the intact plant experiment. The ecological implications for competition or co-existence of these two pathogens in the field are discussed.  相似文献

Leguminous trees play an important role in agroforestry in Ethiopia, but studies of their rhizobial symbionts are scarce. In earlier studies, we surveyed natural nodulation of native leguminous trees growing in different agro-ecological zones in Southern Ethiopia, isolated 400 rhizobia, and characterized them based on different phenotypic and genotypic methods. In the present study we characterized 18 strains belonging to the genus Mesorhizobium, isolated from nodules of Acacia abyssinica, A. senegal, A. tortilis and Sesbania sesban. Phylogenetic analysis of nearly full-length 16S rRNA gene grouped the test strains into three distinct clades separated from all currently recognized Mesorhizobium species. Three divergent strains formed separate branches while the other 15 strains formed three distinct groups, genospecies I-III. Grouping of the isolates under study based on the house-keeping genes recA, gyrB, rpoB and gltA were consistent and in agreement with that of 16S rRNA. Similarly phylogenetic relationships based on the symbiosis-related genes nodC, nodA and nifH were generally similar to those shown by the core genes, suggesting that these Acacia and Sesbania symbionts have a long history of separate evolution within Mesorhizobium. Cross inoculation experiments demonstrated a large variation in the ability of the test strains to elicit effective nodules. The Sesbania isolates, occupying a distinct clade in the nodC phylogenetic tree, formed effective nodules only with this host legume. The study strongly suggests that this collection of Mesorhizobium strains comprises several new species, and also indicates the role of the symbiotic genes in determining the host range of these bacteria.  相似文献
