On binding toVicia faba lectin, the fluorescence of 4-methylumbelliferyl-α-D-glucoPyranoside was quantitatively quenched showing that the interaction of 4-methylumbelliferyl-α-D-glucoPyranoside took Place in a binding environment. The binding of the fluorescent sugar was saccharide sPecific as evidenced by the reversal of 4-methylumbelliferyl-α-D-glucoPyranoside fluorescence quenching by D-fructose. The association constant,K a, values for the 4-methylumbelliferyl-α-D-glucoPyranoside was determined by comPetition study emPloying reversal of fluorescence quenching of 4-methylumbelliferyl-α-D-glucoPyranoside by D-fructose. TheK a value obtained for D-fructose was 1.07 ±0.03 X 104 M-1 and for 4-methylumbelliferyl-α-D-glucoPyranoside was 1.60 ±0.05 X 104 M-1 at 15°C. TheK a values of 2.51 ±0.06 X 104M-1, l.26 ±0.02 X 104 M-1 and 0.56 ±0.01 X 104M-1, resPectively at 10°, 20° and 30°C were obtained from the ChiPman equation. The relative fluorescence quenching, ΔF a, at infinite concentration of the free saccharide sites ofVicia faba lectin [P′] was 93.5% at 30°C and the binding constant for 4-methylumbelliferyl-α-D-glucoPyranoside lectin interaction as derived by Yank and Hanaguchi equation was 0.63 ±0.01 X 104M-1.  相似文献

The electron transfer reactions of horse heart cytochrome c with a series of amino acid-pentacyanoferrate(II) complexes have been studied by the stopped-flow technique, at 25°C, μ = 0.100, pH 7 (phosphate buffer). A second-order behavior was observed in the case of the Fe(CN)5 (histidine)3? complex, with k = 2.8 x 105 M?1 sec?1. For the Fe(CN)5 (alanine)4? and Fe(CN)5(L-glutamate)5? complexes, only a minor deviation of the second-order behavior, close to the experimental error (k = 3.2 × 105 and 1.6 x 105 M?1 sec?1, respectively) was noted at high concentrations of the reactants (e.g., 6 × 10?4 M). The results are in accord with recent work on the Fe(CN)64?/cytochrome c system demonstrating weak association of the reactants. The calculated self-exchange rate constants including electrostatic interactions for the imidazole,L -histidine, 4-aminopyridine, glycinate, β-alaninate, andL-glutamate pentacyanoferrate(II) complexes were 3.3 × 105, 3.3 × 105, 2.8 × 106,4.1 × 102,5.5 × 102, and 6.0 M?1 sec?1, respectively. Marcus theory calculations for the cytochrome c reactions were interpreted in terms of two nonequivalent binding sites for the complexes, with the metalloprotein self-exchange rate constants varying from 104 M?1 sec?1 (histidine, imidazole, and 4-aminopyridine complexes) to 106 M?1 sec ?1 (glycinate, β-alaninate, and L-glutamate complexes).  相似文献

Maximum levels of binding of α-bungarotoxin to foetal human brain membranes were found to remain essentially constant at 30–50 fmol/mg protein (1.1–1.5 pmol/g wet weight in whole brain) between gestational ages of 10 and 24 weeks. Equilibrium binding of α-bungarotoxin to both membranes and to detergent extracts showed saturable specific binding to a single class of sites with Kd (app) values of 3.5 × 10?9 M and 2.4 × 10?9 M respectively. Association rate constants, determined from time courses of binding of α-bungarotoxin to membranes and detergent extracts, were 2.3 × 105 M?1 sec?1 and 2.6 × 105 M?1 sec?1 respectively. Dissociation of α-bungarotoxin from both membrane and detergent extracts showed a rapid initial rate with T12 approx 15 min which, in the case of the detergent extract, was followed by a slower dissociation accounting for the remaining 20% of the bound ligand. Competition studies with a number of cholinergic ligands indicated that the α-bungarotoxin-binding sites in foetal brain display a predominantly nicotinic profile.  相似文献

The kinetics of the binding of cyanide to ferric chloroperoxidase have been studied at 25°C and ionic strength 0.11 M using a stopped-flow apparatus. The dissociation constant (KCN) of the peroxidase-cyanide complex and both forward (k+) and reverse (k?) rate constants are independent of the H+ concentration over the pH range 2.7 to 7.1. The values obtained are kcn = (9.5 ± 1.0) × 10-5 M, k+. = (5.2 ± 0.5) × 104 M?1 sec?1 and k- = (5.0± 1.4) sec-1. In the presence of 0 06 M potassium nitrate the affinity of cyanide for chloroperoxidase decreases due to the inhibition of the forward reaction. The dissociation rate is not affected. The nitrate anion exerts its influence by binding to a protonated form of the enzyme, whereas the cyanide binds to the unprotonated form. Binding of nitrate results in an apparent shift towards higher pKa values of the ionization of a crucial heme-linked acid group. Hence the influence of this group can be detected in the accessible pH range. Extrapolation to zero nitrate concentration yields a value of 3.1±0.3 for the pKa of the heme-linked acid group.  相似文献

The kinetics of double-helix formation by poly U and the complementary monomer N-6,9-dimethyladenine (m6m9A) has been measured using a new fast temperature-jump apparatus. The cooperative binding kinetics are complicated by the extensive self-association of the monomers, but a satisfactory analysis using average relaxation times was possible in terms of three different models. Application of a model which considers only monomer binding yields the upper limit for the binding rate constant of an m6m9A monomer next to an already bound monomer on a poly U strand: (2 ± 0.4) × 108 M?1sec?1. A lower limit is found by using a model which allows for binding of all m6m9A stacks to poly U with equal rate constants: (3 ± 0.3) × 107 M?1sec?1. A third model with “weighted” rate constants consistent with the data: (7.5 ± 1.0) × 107 M?1sec?1. The rate of cooperative binding of m6m9A to the trimer UpUpU has also been measured. The rate constants obtained with the trimer agree with those obtained with the polymer for each of the three models within experimental error.  相似文献

The kinetics of the reduction by aniline and a series of substituted anilines of a peroxidatically active intermediate, formed by oxidation of deuteroferriheme with hydrogen peroxide, have been studied by stopped-flow spectrophotometry. The reaction with aniline was first order with respect to [intermediate] and showed first-order saturation kinetics with respect to [aniline]. The second-order rate constant was 2.0 ± 0.2 × 105 M?1 sec?1 at 25°C (independent of pH in the range 6.60–9.68) compared with the value of 2.4 × 105 M?1 sec?1 for the reaction of aniline with horseradish peroxidase Compound I. The effect of aniline substituents upon reactivity towards the heme intermediate closely paralled those reported for reaction with the enzymic intermediate. Anilines bearing electron-donating substituents reacted more rapidly and those bearing electron-withdrawing substituents more slowly than the unsubstituted amine. The rate constants for the heme intermediate reactions (kdfh)found to be related to those for the enzymic reactions (khrp) by the equation:log kDFH= 0.65log kHRP+ 1.96 with a correlation coefficient of 0. 98.  相似文献

The anation reaction of aquopentaamminecobalt(III) by acetate has been studied in the temperature range 60–80°C and acidity range 1.0 ≦ pH ≦ 5.5 for total acetate concentrations ≦ 0.5 M and at ionic strength 1.0 M. The anation by acetic acid follows second-order kinetics (k0), whereas the kinetic results for the anation by acetate (Q1, k1) provide evidence for the formation of an ion-pair with the complex ion. Typical experimental results at 70°C are k0 = 5.33 X 10−5 M−1 sec−1, Q1 = 5.87 M−1 and k1 = 1.46 X 10−4sec−1. The activation parameters for the different reaction paths are reported and the results discussed with reference to various other anation reactions of Co(III) complexes.  相似文献

Abstract

(±)125 I-cyanopindolol (±) I CYP) was used to characterize β-adrenoceptors on rat lung and cerebral cortex membranes. The affinity of (±) ICYP was higher for lung (Kd = 64.3 pM) at 37°C. The association reaction of (±) ICYP was faster with lung (k+1 = 1.52 × 109 M?1.min?1) than with cerebral cortex β-adrenoceptors (k+1 = 1.75 × 108 M?1.min?1). In both tissues, the dissociation reaction followed a biphasic process with a fast (t ½ = 15.4 min and 5.6 min for lung and cerebral cortex respectively) and a slow component (t ½ = 474 min and 255 min for lung and cerebral cortex respectively). The thermodynamic parameters for (±) ICYP - β-adrenoceptors binding have been determined from kinetics and equilibrium studies, for the two tissues, at several temperatures between 0° and 44° C. For lung and cerebral cortex, Arrhenius plots were linear with different energies of activation. Van't Hoff plot was not linear for lung and the standard enthalpy and entropy changes of (±) ICYP - β-adrenoceptors interaction decreased linearly with temperature : the binding occured with a negative heat capacity change (ΔCp° = -368.9 cal. moles?1. K?1) at 25° C. Thermodynamic and kinetic results show that binding of (±) ICYP to lung β-adrenoceptors could involve two successive equilibria with a conformational change of the β-adrenergic receptor.  相似文献

The complexation reactions of O-phospho-DL-serine with Ni(II) or Co(II) were studied at 25°C and ionic strength 0.2 M (KNO3) by temperature-jump. The observed rate constants for formation of the Ni2+ and Co+2 monocomplexes were (1.32 ± 0.09) × 105 and (1.73 ± 0.33) × 107 M?1 sec?1, respectively. Complexation is postulated to involve formation of a monocoordinated steady state intermediate followed by rate-determining chelate ring closure.  相似文献

N-methylmyristohydroxamic acid (1) bound to polymer micelles of laurylated poly(2- and 4-vinylpyridines) (lauryl group contet: 2VP-L, 30 mol%; 4VP-L, 33 mol%) quantitatively reacted with p-nitrophenyl acetate (NpAc) within a few seconds at 30°C, pH 8.95. Second order rate constants ka were 34,000 M?1 sec?1 for 1–2VP-L and 11,400 M?1 sec?1 for 1–4VP-L at μ = 0.5, and they were pronouncedly improved by a decrease in ionic strength (ka = 27,500–80,200 M?1 sec?1 at μ = 0.08). In contrast, poly(N-ethyl-4-vinylpyridinium bromide) hardly affected the nucleophilicity of the hydroxamate ion. Therefore, the enhancement was considered to be associated with some micellar characteristics. Typical saturation phenomena of the reaction rate were observed for p-nitrophenyl hexanoate (NpOCOPe) and 3-nitro-4-acetoxybenzoic acid (NpAcCOOH). It was suggested that binding of NpOCOPe is caused by the hydrophobic interaction, while that of NpAcCOOH is probably induced by the electrostatic interaction. It is demonstrated that the cationic polymer micelle enormously activates the bound hydroxamate anion, and these complexes would be of much interest as a biomimetic system for enzyme catalysis.  相似文献

The technique of Stopped-Flow Circular Dichroism allows the simultaneous monitoring of chiroptical and absorbance transients at millisecond time resolution. In the binding of a chromophoric sulphonamide to Bovine Carbonic Anhydrase, the rapid kinetics of the induced circular dichroism and difference spectra proceed in parallel with bimolecular rate constant k1 = 5 × 106 M?1 sec?1 and apparent half reaction time of 8.7 msec for 24 μM reactants. RNA has attracted recent attention for its key role in gene expression and hence targeting by small molecules for therapeutic intervention. This study is aimed to elucidate the specificity of RNA binding affinity of parent compound of N-arylhydroxamic acids series, N-phenylbenzohydroxamic acid trivially named as PBHA,C6H5NOH.C6H5C ? O. The binding behavior was examined by various biophysical methods such as absorption, fluorescence, and viscosity measurements. Molecular docking was also done. The value of affinity constant and overall binding constant was calculated 5.79 ± 0.03 × 104 M?1 and K’ = 1.09 ± 0.03 × 105 M?1, respectively. The Stern-Volmer constant Ksv obtained was 2.28 ± 0.04 × 104 M?1. The compound (PBHA) shows a concentration-based enhancement of fluorescence intensity with increasing RNA concentration. Fluorescence quenching of PBHA–RNA complex in presence of K4 [Fe(CN)6] was also observed. Viscometric studies complimented the UV results where a continuous increase in relative viscosity of the RNA solution was observed with added optimal PBHA concentration. All the experimental evidences indicate that PBHA can strongly bind to RNA through an intercalative mode.  相似文献

Abstract: Amyloid β-peptide (Aβ) aggregates are one of the key neuropathological characteristics of Alzheimer's disease. Aβ belongs to a group of proteins that aggregate and form β-sheets, and some of these proteins bind cholesterol and other lipids. The purpose of the experiments reported here was to determine if cholesterol, fatty acids, and phosphatidylcholine (PC) would bind to Aβ1–40 and if such binding would be dependent on aggregation of Aβ1–40. Lipid binding was determined using fluorescent-labeled lipids. Incubation of Aβ1–40 for 0, 1, 3, 6, 21, and 24 h resulted in aggregation of the peptide with formation of dimers, trimers (1–24 h), and polymers (6–24 h) as determined by sodium dodecyl sulfate-gel electrophoresis. No change in the fluorescence of the lipids was observed when lipids were added to Aβ1–40 that had been incubated for 0, 1, or 3 h. However, the fluorescence intensities of cholesterol, saturated fatty acids, and PC were significantly increased (p < 0.0001) when added to Aβ1–40 that had been incubated for 6, 21, and 24 h in which Aβ1–40 polymers were detected. The binding affinity of cholesterol to Aβ1–40 polymers (KD of 3.24 ± 0.315 × 10?9M) was markedly higher as compared with the other lipids (stearic acid, 9.42 ± 0.41 × 10?8M; PC, 7.07 ± 0.12 × 10?7M). The results of this study indicate that Aβ1–40 polymers bind lipids and have a higher affinity for cholesterol than PC or saturated fatty acids. Aggregated Aβ1–40 may affect lipid transport between cells or remove specific lipids from membranes, and such effects could contribute to neuronal dysfunction.  相似文献

Abstract: Cross-reactions between dopamine D3 and σ receptor ligands were investigated using (±)-7-hydroxy-N,N-di-n-[3H]propyl-2-aminotetralin [(±)-7-OH-[3H]DPAT], a putative D3-selective radioligand, in conjunction with the unlabeled σ ligands 1,3-di(2-tolyl)guanidine (DTG), carbetapentane, and R(?)-N-(3-phenyl-1-propyl)-1-phenyl-2-aminopropane [R(?)-PPAP]. In transfected CCL1.3 mouse fibroblasts expressing the human D3 receptor, neither DTG nor carbetapentane (0.1 µM) displaced (±)-7-OH-[3H]DPAT binding. R(?)-PPAP (0.1 µM) displaced 39.6 ± 1.0% of total (±)-7-OH-[3H]DPAT binding. In striatal and nucleus accumbens homogenates, (±)-7-OH-[3H]DPAT labeled a single site (15–20 fmol/mg of protein) with high (1 nM) affinity. Competition analysis with carbetapentane defined both high- and low-affinity sites in striatal (35 and 65%, respectively) and nucleus accumbens (59 and 41%, respectively) tissue, yet R(?)-PPAP identified two sites in equal proportion. Carbetapentane and R(?)-PPAP (0.1 µM) displaced ~20–50% of total (±)-7-OH-[3H]DPAT binding in striatum, nucleus accumbens, and olfactory tubercle in autoradiographic studies, with the nucleus accumbens shell subregion exhibiting the greatest displacement. To determine directly (+)-7-OH-[3H]DPAT binding to σ receptors, saturation analysis was performed in the cerebellum while masking D3 receptors with 1 µM dopamine. Under these conditions (+)-7-OH-[3H]DPAT labeled σ receptors with an affinity of 24 nM. These results suggest that (a) (±)-7-OH-[3H]DPAT binds D3 receptors with high affinity in rat brain and (b) a significant proportion of (±)-7-OH-[3H]DPAT binding consists of σ1 sites and the percentages of these sites differ among the subregions of the striatum and nucleus accumbens.  相似文献

Abstract

Saturation experiments were performed on intact human peripheral mononuclear leucocytes (MNL) and MNL membranes with (-)125Iodocyanopindolol (125ICYP) over a large concentration range (1.5-600pmol/l). The corresponding Scatchard plots were curvilinear suggesting two saturable classes of binding sites: A high affinity binding site (Bmax1=1000±400 sites/cell, Kd1= 2.1±0.9 pmol/l for intact MNL and Bmax1=550±190 sites/cell, Kd1=4.1±0.9 pmol/l for MNL membranes)and a low affinity binding site (Bmax2=9150±3590 binding sites/cell, Kd2=440±50 pmol/l for intact MNL and Bmax2=11560±4690 sites/cell, Kd2=410±70 pmol/l for MNL membranes). Dissociation of (-)125ICYP from MNL was biphasic consisting of a slow dissociating component (dissociation rate constant k-1=(0.5±0.2)x10?3 min?1 for intact MNL and k-1=(1.0±0.1)x10?3min?1 for MNL membranes) and a fast dissociating component (k-2=(80±20)x10?3min?1 for intact MNL and k-2=(60±10)x10?3min?1 for MNL membranes). In dissociation experiments started after equilibration with various (-)125ICYP concentrations k-1 and k-2 were independent of the equilibrium concentration, whereas the percentual occupancy of the slow and the fast dissociating component varied and was similar to the estimated fractional occupancy of either binding site at the same (-)125ICYP concentrations in saturation experiments. The association rate constant was in the same order of magnitude for both binding sites. These results suggest two independent classes of binding sites for (-)125ICYP on MNL.  相似文献

Chlorogenic acid, 3’-O-caffeoyl D-quinic acid, is an inherent ligand present inHelianthus annuus L. The effect of pH on chlorogenic acid binding to helianthinin suggests that maximum binding occurs at pH 6.0. The protein-polyphenol complex precipitates as a function of time. The association constant of the binding of chlorogenic acid to helianthinin, determined by equilibrium dialysis, at 31°C has a value of 3.5 ± 0.1 × 104M−-1 resulting in a ΔG value of − 6.32 ± 0.12 kcal /mol. The association constantK ais 1.0 ± 0.1 × 104M−1 as determined by ultraviolet difference spectral titration at 25°C with ΔG° of -5.46 ± 0.06 kcal/mol. From fluorescence spectral titration at 28°C, theK avalue is 1.38 ± 0.1 × 1 0 4M−1 resulting in a ΔG of − 5.70 ± 0.05 kcal/mol. The total number of binding sites on the protein are 420 ± 50 as calculated from equilibrium dialysis. Microcalorimetric data of the ligand-protein interaction at 23°C suggests mainly two classes of binding. The thermal denaturation temperature,T mof the protein decreases from 76°C to 72°C at 1 × 10−3M chlorogenic acid concentration upon complexation. This suggests that the complexation destabilizes the protein. The effect of temperature onK aof chlorogenic acid shows a nonlinear increase from 10.2°C to 45°C. Chemical modification of both lysyl and tryptophanyl residues of the protein decreases the strength of binding of chlorogenic acid. Lysine, tryptophan and tyrosine of protein are shown to be present at the binding site. Based on the above data, it is suggested that charge-transfer complexation and entropically driven hydrophobic interaction are the predominant forces that are responsible for binding of chlorogenic acid to the multisubunit protein, helianthinin. To monitor the specificity of Staphylococcus aureus aptamer (SA‐31) against its target cell, we used enzyme‐linked aptamer assay. In the presence of target cell, horseradish peroxidase–conjugated streptavidin bound to biotin‐labeled SA‐31 showed specific binding to S   aureus among 3 different bacteria with limit of detection of 103 colony‐forming unit per milliliter. The apparent K a was 1.39 μM−1 ± 0.3 μM−1. The binding of SA‐31 to membrane proteins extracted from cell surface was characterized using isothermal titration calorimetry, and the effect of changes in binding temperature and salt concentrations of binding buffer was evaluated based on thermodynamic parameters (K a, ΔH , and ΔG ). Since binding of aptamer to its targets solely depends on its 3‐dimensional structure under experimental conditions used in selection process, the change in temperature and ion concentration changed the affinity of SA‐31 to its target on surface of bacteria. At 4°C, SA‐31 did not show an affinity to its target with poor heat change upon injection of membrane fraction to aptamer solution. However, the apparent association constants of SA‐31 slightly varied from K a = 1.56 μM−1 ± 0.69 μM−1 at 25°C to K a = 1.03 μM−1 ± 0.9 μM−1 at 37°C. At spontaneously occurring exothermic binding reactions, affinities of S  aureus aptamer to its target were also 9.44 μM−1 ± 0.38 μM−1 at 50mM, 1.60 μM−1 ± 0.11 μM−1 at 137mM, and 3.28 μM−1 ± 0.46 μM−1 at 200 mM of salt concentration. In this study, it was demonstrated that enzyme‐linked aptamer assay and isothermal titration calorimetry were useful tools for studying the fundamental binding mechanism between a DNA aptamer and its target on the outer surface of S  aureus .  相似文献

In the acridine orange–dermatan sulfate system, free and bound dye can be distinguished from each other spectroscopically. This permits the use of fluorometric methods to study the binding of acridine orange to the acid mucopolysaccharide dermatan sulfate. Experiments were conducted at 24°C in 10?3 M citrate/phosphate buffer at pH = 7.0. The binding of the dye is highly cooperative, as evidenced by considerable interaction between adjacent bound dye molecules. Analysis of the data indicates that dermatan sulfate binds 2.3 ± 0.3 mol of acridine orange per dermatan sulfate uronic acid residue with a cooperative binding constant, Kq ranging from 4.9 to 6.0 × 105 M?1 which corresponds to a free energy of 7.74 ? ΔG° ? 7.86. The cooperativity parameter q apparently increases with increasing polymer-to-dye ratio.  相似文献

The hydrolyses of p-nitrotrifluoroacetanilide catalyzed by water and imidazole were examined at 70°C. The pH-rate constant profile of the hydrolysis in H2O was examined in the pH range 0.0–11.4. The hydrolysis was independent of pH in the region from pH 1.0 to 4.5, presumably a water-catalyzed reaction. The rate constant and the D2O solvent isotope effect for this reaction were 1.0 × 10?4 sec?1 and 3.7, respectively. Both natural imidazole and imidazolium cation catalyzed hydrolysis. The rate constant of the hydrolysis catalyzed by neutral imidazole was determined to be 5.4 × 10?3M?1 sec?1 and the D2O solvent isotope effect was 1.8.  相似文献
