目的:使用反相高效液相色谱法测定色盐杆菌新种ST307的DNA G+C mol%含量.方法:以Escherichia coli DH5α为标准菌株,采用90%重蒸水10%甲醇为流动相,检测波长260nm,流速1ml·min-1,在Venusil MP C18柱上对四种碱基进行分离.结果:DNA碱基分离效果好,以外标法计算得到标准菌株DH5α的DNA G+C mol%含量为50.3%,待测菌株ST307的DNA G+C mol%含量为60.5%.结论:采用反向高效液相色谱法测定色盐杆菌的DNA G+C mol%含量准确可靠.  相似文献

应用高效液相色谱法测定了放线菌的(G+C)mol%,并就有关分析条件和DNA水解条件进行了优化选择.以Escherichia.coli DH5α作为标准菌株,实验室分离鉴定的8株红树林放线菌作为待测菌株,采用流动相为20 mmol/L KH2PO4缓冲液(pH 5.6)∶甲醇(体积比90∶10),检测波长为260 nm,硫速为1 ml/min,柱温为35℃,分析时间15 min对4种碱基进行分离.结果表明:标准碱基溶液的pH值、流动相的组成和pH值是影响各碱基色谱峰分离和峰形的主要因素.在优化选择的条件下测定,DNA碱基分离效果好,无杂峰干扰,以峰面积计算得到标准菌株Escherichia coli DH5 α的(G+C)mol%为54.9694%与已经报道的差异不显著;待测菌株的(G+C)mol%均在菌株所在科或属下的(G+C)mol%围内;采用酸水解放线菌的DNA可缩短鉴定时间,步骤简明.实验结果充分显示高效液相色谱法测定放线菌(G+C)mol%快速、准确、结果稳定,是放线菌分类鉴定的一种可靠方法.  相似文献

应用高压液相色谱对七株不同种的细菌DNA中G+C克分子百分数进行了测定。试验结果表明,用Nucleosil C作固定相,以反相分配色谱法测定核酸碱基组分,具有快速简便、灵敏精确的优点。用本法测定DNA水解物中G+C克分子百分数时,每种碱基的进样量只需0.2一1.0微克,测定时间仅为8分钟,方法的精确度为士1.5%。  相似文献

通过生理生化实验、16S rDNA序列分析和同源性杂交,将分离到的XJU-1和XJU-80菌种进行了分类鉴定。XJU-1和XJU-80具有较宽的pH生长范围(分别是pH4·5 ~12·6和pH3·8 ~12·6) ,其G C mol%含量分别是40·5mol%和42·2mol%。16S rDNA序列分析和DNA-DNA同源杂交结果表明,XJU-1和XJU-80与Bacillushalodurans C-125和Bacillus halodurans DSM497T具有较高的同源性(99 %) ;两者之间也具有85 %的相关性,但其与Bacillus halodurans C-125和Bacillus halodurans DSM497T分别具有81·3 %和71·5 %的相关性。基于以上结果,将两株分离菌株分类为Bacillus halodurans的两个新品系。  相似文献

采用反相高效液相色谱法结合液-质联用技术对漆叶提取物进行分离,并对其中的漆酚类化合物进行定性分析。结果显示:液相色谱分离得到10个峰,经质谱分析并结合文献,鉴定其中9个峰为C15或C17的饱和、单烯、二烯、及三烯漆酚类物质。首次对漆叶中漆酚类化合物进行了高效液相色谱-质谱分离和定性分析,为漆叶的临床应用提供一定的理论依据。  相似文献

针对少量且复杂蛋白质组样品,开发一种耗时短、操作简便的分离方法。以人的脑海马组织蛋白样品为研究对象,采用反相C18固相萃取小柱,采用不同乙腈浓度对酶切后样品进行梯度洗脱,与反相高效液相色谱法对比分离效果。通过比较不同乙腈梯度洗脱方案所鉴定到的谱图数、非冗余多肽数、蛋白数和各洗脱组分间重复率分析,确定一种样品量少、简单易行、分离效果好的实验方案。虽然反相C18固相萃取小柱法鉴定蛋白总数占高效液相色谱法的85.5%,但其操作简单,耗时少。通过4种不同乙腈浓度方案比较,确定乙腈洗脱浓度优化为5%、15%、20%和90%时,分离30μg人的海马多肽混合物可以得到较优的分离效果。其蛋白鉴定数为反相液相色谱法的67.0%,且重复性良好。该结果证实反相C18固相萃取小柱分离效果比反相高效液相色谱法稍差,但此方法可以分离少量复杂蛋白质组样品。该方法分离样品充分,操作易行,耗时短,是进行蛋白质组学分析中少量样品的一个简便预处理方法。  相似文献

对脂必妥片样品进行预处理,得到的混合物用薄层色谱、反相高效液相色谱法进行定性、定量分析,建立检测其含量的方法。当色谱条件为色谱柱:Kromasil(C18250 mm×4.6 mm,5μm);流动相体积比:乙腈∶水为85∶15,;流速:1.0 ml.min-1;检测波长:420 nm;柱温:30℃;测定结果表明被测峰和其它峰可完全分离,在每毫升10.21~200.03μg内具有良好的线性关系,r=0.9997,测得其中洛伐他汀含量为0.25%,回收率:97.73%,RSD:0.56%。这种方法准确、可靠,可用于含洛伐他汀药品的质量控制。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. Rasmussen’s encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and 16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis, intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8+ T cells. Our results suggest that EBV and HHV6 infection and the activation of CD8+ T cells are involved in the pathogenesis of RE.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献
