The effect of radiation of different spectral composition on axillary proliferation of lavandin (Lavandula officinalis Chaix ×Lavandula latifolia Villars cv. Grosso) was studied in combination with application of exogenous benzyladenine (BA) and putrescine (Put) and endogenous ethylene production. The effect of BA was predominant over the radiation. Continuous far-red showed a fluence rate-dependent promotion of shoot proliferation in the presence of BA. On BA-free medium, shoot number was enhanced under blue, white, and red radiation, at low photon fluence rates. BA, however, could reduce the inhibiting effect of blue and ultraviolet radiation, at high photon fluence rates. Exogenous Put stimulated axillary bud proliferation under some radiation treatments in the presence of BA. Moreover, Put, analogously to BA, could overcome the detrimental effect of ultraviolet radiation. A positive correlation between biotic ethylene production and shoot formation was evidenced under far-red at high photon fluence rate in the presence of BA, and under white, red and blue radiation at low photon fluence rate in the BA-free medium. However, when abiotic ethylene (released from the agarized medium) was stimulated by UV, no improvement of shoot formation was observed.  相似文献

Excised apical segments of etiolated rice (Oryza sativa L.) coleoptiles produced ethylene. Increasing the number of cut sites per coleoptile increased the rate of ethylene formation. Ethylene produced by an etiolated-intact seedling in the dark was about a half of that by the excised coleoptile segment. Red light of low energy as well as of continuous irradiation inhibited the production of ethylene. The inhibition by a low energy dose of red light was partly relieved, if the red light was followed immediately by a small dose of far red light. The effect of red and far red light was repeatedly reversible, indicating that ethylene production was regulated by a phytochrome system. If the exposure to far red light was preceded by a period of darkness, this photoreversibility disappeared; 50% of the initial reversibility was lost within 5 hours. Applied ethylene (10 microliters per liter) significantly promoted the growth of intact coleoptiles of either totally etiolated or red light-treated seedlings, but had no effect on the excised apical segment of coleoptile.  相似文献

Leaf discs from expanding leaves of Rumex patientia L. were exposed to 7 hours of visible plus different levels of ultraviolet radiation in the 290 to 315 nm waveband (UV-B) and then placed in darkness. Leaf disc expansion was reduced and anthocyanin production was increased in discs exposed to moderate or high levels of UV-B radiation when compared to control discs. The possibility that the inhibition of leaf expansion by UV-B radiation might be at least partially phytochrome-mediated was examined by giving discs brief red or far red irradiation following exposure to UV-B radiation. Brief red radiation (R) following treatment with moderate or high UV-B radiation did not alter the pattern of growth or anthocyanin production compared to discs placed in darkness following UV-B treatment. However, a posttreatment with far red radiation (FR) reduced the growth of discs subjected previously to either moderate UV-B or no UV-B irradiation to the level of growth of discs given high UV-B. FR posttreatment also decreased anthocyanin production in discs in moderate and high UV-B treatments. Effects of FR and UV-B radiation apparently do not involve the same mechanism. This was demonstrated by experiments in which FR following the UV-B treatments was in turn followed by R, which reversed the effects of the FR but did not alter the growth inhibition or increased anthocyanin production induced by moderate or high levels of UV-B radiation.  相似文献

Ultraviolet B (UV-B) radiation is an important environmental signal for plant growth and development, but its signal transduction mechanism is unclear. UV-B is known to induce stomatal closure via hydrogen peroxide (H(2)O(2)), and to affect ethylene biosynthesis. As ethylene is also known to induce stomatal closure via H(2)O(2) generation, the possibility of UV-B-induced stomatal closure via ethylene-mediated H(2)O(2) generation was investigated in Vicia faba by epidermal strip bioassay, laser-scanning confocal microscopy, and assays of ethylene production. It was found that H(2)O(2) production in guard cells and subsequent stomatal closure induced by UV-B radiation were inhibited by interfering with ethylene biosynthesis as well as ethylene signalling, suggesting that ethylene is epistatic to UV-B radiation in stomatal movement. Ethylene production preceded H(2)O(2) production upon UV-B radiation, while exogenous ethylene induced H(2)O(2) production in guard cells and subsequent stomatal closure, further supporting the conclusion. Inhibitors for peroxidase but not for NADPH oxidase abolished H(2)O(2) production upon UV-B radiation in guard cells, suggesting that peroxidase is the source of UV-B-induced H(2)O(2) production. Taken together, our results strongly support the idea that ethylene mediates UV-B-induced stomatal closure via peroxidase-dependent H(2)O(2) generation.  相似文献

BACKGROUND AND AIMS: The relationship between ethylene production and both seed dormancy and germination was investigated using red rice (weedy rice) as a model species. METHODS: Both fully dormant and after-ripened (non-dormant) naked caryopses were incubated with or without inhibitors of ethylene synthesis [aminoethoxyvinylglycine (AVG)] and perception [silver thiosulfate (STS)], or in the presence of the natural ethylene precursor 1-aminocyclopropane-1-carboxylic acid (ACC). The kinetics of ethylene emissions were measured with a sensitive laser-photoacoustic system. KEY RESULTS: Dormant red rice caryopses did not produce ethylene. In non-dormant caryopses, ethylene evolution never preceded the first visible stage of germination (pericarp splitting), and ethylene inhibitors completely blocked ethylene production, but not pericarp splitting. Accordingly, endogenous ACC appeared to be lacking before pericarp splitting. However, early seedling growth (radicle or coleoptile attaining the length of 1 mm) followed ethylene evolution and was delayed by the inhibitors. Wounding the dormant caryopses induced them to germinate and produce ethylene, but their germination was slow and pericarp splitting could be speeded up by ethylene. CONCLUSIONS: The findings suggest that, in red rice, endogenous ethylene stimulates the growth of the nascent seedling, but does not affect seed dormancy or germination inception. Correspondingly, this phytohormone does not play a role in the dormancy breakage induced by wounding, but accelerates germination after such breakage has occurred.  相似文献

The apical 1-cm hypocotyl of dark-grown `Clark' soybean (Glycine max [L.] Merr.) seedlings produced ethylene at rates of 7 to 11 nanoliters per hour per gram when attached to the cotyledons. Such physiologically active rates occurred prior to the deceleration of hypocotyl elongation caused by the temperature of 25 C.

Daily exposure of the etiolated seedlings to red light promoted hypocotyl elongation and prevented its lateral swelling. Red light treatment also caused a 45% decrease in ethylene production. Far red irradiation following the red treatment reversed the red effects, suggesting that the ethylene intervenes as a regulator in the phytochrome control of `Clark' soybean hypocotyl growth at 25 C.

The present study was carried out to understand the mechanism of salt stress amelioration in red pepper plants by inoculation of 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase-producing halotolerant bacteria. In general, ethylene production, ACC concentration, ACC synthase (ACS), and ACC oxidase (ACO) enzyme activities increased with increasing levels of salt stress. Treatment with halotolerant bacteria reduced ethylene production by 47–64%, ACC concentration by 47–55% and ACO activity by 18–19% in salt-stressed (150 mmol NaCl) red pepper seedlings compared to uninoculated controls. ACS activity was lower in red pepper seedlings treated with Bacillus aryabhattai RS341 but higher in seedlings treated with Brevibacterium epidermidis RS15 (44%) and Micrococcus yunnanensis RS222 (23%) under salt-stressed conditions as compared to uninoculated controls. A significant increase was recorded in red pepper plant growth under salt stress when treated with ACC deaminase-producing halotolerant bacteria as compared to uninoculated controls. The results of this study collectively suggest that salt stress enhanced ethylene production by increasing enzyme activities of the ethylene biosynthetic pathway. Inoculation with ACC deaminase-producing halotolerant bacteria plays an important role in ethylene metabolism, particularly by reducing the ACC concentration, although a direct effect on reducing ACO activity was also observed. It is suggested that growth promotion in inoculated red pepper plants under inhibitory levels of salt stress is due to ACC deaminase activity present in the halotolerant bacteria.  相似文献

Although the growth of intact plants is inhibited by irradiation with blue light, the growth rate of isolated stem segments is largely unaffected by blue light. We hypothesized that this loss of responsiveness was a result of ethylene production as part of the wounding response. However, we found no interaction between ethylene- and blue light-induced growth inhibition in dark- or red light-grown seedlings of pea (Pisum sativum L.). Inhibition of growth begins in dark-grown seedlings exposed to blue light within 3 min of the onset of blue light, as was known for red light-grown seedlings. By contrast, ethylene-induced inhibition of growth occurs only after a lag of 20 to 30 min or more (dark-grown seedlings) or 60 min (red light-grown seedlings). Also, the inhibition response of red light-grown seedlings is the same whether ethylene is present from the onset of continuous blue-light treatment or not. Finally the spatial distribution of inhibition following blue light was different from that following ethylene treatment.  相似文献

The effect of red and white light on ethylene production was investigated in several plant species. In most cases light inhibited ethylene production. However, stimulation or no effect were also observed in a few species. In those plants where light inhibited ethylene synthesis, the effect of red light was much stronger than that of white light.Both red and white light inhibited ethylene production in green and etiolated seedlings and green leaves of Impatiens balsamina L. The inhibitory effect of red light was stronger than that of white light and much more pronounced when the plants were pretreated with ACC. The effect of red light could be reversed by far-red light. These results suggest that light affects the ethylene forming enzyme (EFE) activity and that its action is mediated by phytochrome.  相似文献

The effects of heat shock (HS, 45°C) and UV-B radiation (280–320 nm, 18.3 kJ/(m2h)) and the consecutive action of the combination of these factors on ethylene production, gas exchange, and the growth of intact melon (Melo sativusSager.) seedlings were investigated. The changes in ethylene production and carbon dioxide exchange were described by a single-peaked curve. In the course of UV-B irradiation, the time of maximum ethylene and CO2evolution coincided (the first 5 min) and comprised 0.36 nl/(seedling h) for ethylene and 146.2 l/(seedling h) for carbon dioxide. After HS, the maximum of ethylene production (0.37 nl/(seedling h)) was reached within 10 min, and that of carbon dioxide production (313 l/(seedling h)), within 45 min. The rate of ethylene production (0.22 nl/(seedling h)), carbon dioxide production (97.7 l/(seedling h)), and oxygen consumption (162.5 l/(seedling h)) in the control seedlings did not change in the course of experiment. Throughout the experiment, the respiratory quotient of seedlings was ca. 0.6 regardless on the nature and duration of the acting factor. Preliminary heating at 45°C for 1 h increased the tolerance of seedlings to the subsequent UV-B radiation for 1 h. The protective effect of HS manifests itself in alleviating the inhibiting action of UV-B radiation on seedling growth and development, and this effect was preceded by an increase in ethylene production and respiration. The possible mechanisms of cross-tolerance of plants to overheating and UV-B radiation are discussed.  相似文献

Climacteric rise, ethylene production, peroxidase activity and its isozyme and their interrelationships during the ripening of tomato fruits have been studied. It was found' that there was parallelism between ethylene production and climacteric rise. The climacteric rise of tomato fruits was hastened by ethylene applied at the mature green stage. The ethylene production was inhibited by low oxygen and high carbon dioxide partial pressure. The peroxidase activity in the tomato fruits appeared to be different at three stages, higher in the half red fruits and lower in both green mature and fully red fruits. This activity was increased by ethylene and decreased by lower partial pres- sure of oxygen. The peroxidase isozymes sppeared also different at different stages of ripening. There were 4 bands in young fruits, 3 in green mature fruits, 5 in half red fruits and 3 in fully red fruits. After the application of ethylene to the tomato fruits, there appear one new band of peroxidase isozyme.  相似文献

Either ethylene or carbon dioxide stimulated the growth of oat mesocotyls in darkness, although the effect was much greater for carbon dioxide. Maximum elongation was obtained in the presence of both gases. Ethylene also induced lateral expansion of the mesocotyl: the volume of the mesocotyl was increased more in air with added ethylene than in air depleted of ethylene. Ethylene also stimulated mesocotyl growth under red light. Gibberellin only slightly increased mesocotyl length under red light and acted cooperatively with ethylene in the promotion of growth. The oat mesocotyl is a unique organ in terrestrial plants because ethylene simultaneously stimulates not only longitudinal growth but also lateral expansion. The ecological significance of oat mesocotyl growth in relation to the response to ethylene and carbon dioxide is discussed.  相似文献

Low concentrations of ethylene inhibit hypocotyl elongation of etiolated lettuce seedlings (Lactuca sativa cv. Grand Rapids), whereas red light does not inhibit it. The plumular hook tightens in response to either ethylene or red light. A combination of these two factors gives an additive response. Red light has no effect on ethylene production and red light will cause hook closure even under hypobaric pressure which removes endogenous ethylene. This suggests that ethylene and red light act independently in causing hook closure.  相似文献

Plants growing in the shade receive both low light irradiance and light enriched in far red (FR) (i.e., light with a low red (R) to FR ratio). In an attempt to uncouple the R/FR ratio effects from light irradiance effects, we utilized Stellaria longipes because this species has two distinct natural population ecotypes, alpine (dwarf) and prairie (tall). The alpine population occupies the open, sun habitat. By contrast, the prairie population grows in the shade of other plants. Both 'sun' and 'shade' ecotypes responded with increased stem elongation responses under low irradiance, relative to growth under 'normal' irradiance, and this increased growth was proportionally similar. However, only the shade ecotype had increased shoot elongation in response to a low R/FR ratio. By contrast, the sun ecotype showed increased stem elongation in response to increasing R/FR ratio. Varying the R/FR ratios had no significant effect on ethylene evolution in either sun or shade ecotype. Under low irradiance, only the sun ecotype showed a significantly changed (decreased) ethylene evolution. We conclude that R/FR ratio and irradiance both regulate growth, and that irradiance can also influence ethylene evolution of the sun ecotype. By contrast, R/FR ratio and irradiance, while having profound influences on growth of the shade ecotype, do not appear to regulate these growth changes via effects on ethylene production.  相似文献

The production of ethylene by etiolated pea epicotyls (Pisum sativum L., cv. Alaska) is confined to the plumule and plumular hook portion of the epicotyl, and occurs at a rate of about 6 μl·kg−1·hr−1. Such a rate is sufficient to give physiologically active concentrations of ethylene within the tissue. Exposure of etiolated seedlings to a single dose of red light caused a transient decrease in ethylene production and a corresponding increase in plumular expansion. Far-red irradiation following the red light treatment decreased the red effect to the level achieved by the far-red alone, suggesting that the ethylene production mechanism is controlled by phytochrome and thus that the ethylene intervenes as a regulator in the phytochrome control of plumular expansion.  相似文献

Acidic cloudwater is believed to cause needle injury and to decrease winter hardiness in conifers. During simulations of these adverse conditions, rates of ethylene emissions from and levels of 1-aminocyclopropane-1-carboxylic acid (ACC) in both red and Norway spruce needles increased as a result of treatment with acidic mists but amounts of 1-malonyl(amino)cyclopropane-1-carboxylic acid remained unchanged. However, release of significant quantities of ethylene by another mechanism independent of ACC was also detected from brown needles. Application of exogenous plant growth regulators such as auxin, kinetin, abscisic acid and gibberellic acid (each 0.1 millimolar) had no obvious effects on the rates of basal or stress ethylene production from Norway spruce needles. The kinetics of ethylene formation by acidic mist-stressed needles suggest that there is no active inhibitive mechanism in spruce to prevent stress ethylene being released once ACC has been formed.  相似文献

Abstract Growth and ethylene production by lentil root tips treated with varying concentrations of indolylacetic and indolylacrylic acids were measured. Stimulation of growth by low concentrations of auxin was accompanied by a reduced ethylene production; growth inhibition by high levels of auxin induced a diminished ethylene evolution. Application of a rhizobitoxine analogue increased growth and reduced ethylene evolution. Simultaneous application of auxin and rhizobitoxine analog reduced ethylene evolution even though growth was inhibited. This indicated the non-dependency of auxin-controlled root growth upon ethylene production.  相似文献

Tomato ( Lycopersicon esculentum Mill.) genotypes varying in intrinsic firmness were examined to determine the quantitative relationships between polygalacturonase (EC 3.2.1.15) activity, firmness and other ripening parameters including rate (days from mature-green to full red) and intensity (rate of ethylene production at climacteric peak) of ripening. Texture, respiration and ethylene production were monitored in the immature-green through the red (ripe) stages of development. Polygalacturonase activity was measured by direct assay of salt-extractable wall protein or by monitoring the release of pectins from isolated, enzymically active wall. In all fruit, polygalacturonase activity was highly correlated with pericarp softening, but only moderately correlated with softening of whole fruit (r = 0.920 and 0.757, respectively). Polygalacturonase activity was positively correlated with cell-wall autolytic activity in pink (r = 0.969) and red (r = 0.900) fruit. Firmer genotypes exhibited lower rates of respiration and ethylene production during ripening. Polygalacturonase activity in isolates prepared from fruit at the climacteric peak was positively correlated with ethylene production and respiration, and negatively correlated with days to ripening (r = 0.929, 0.805, and -0.791, respectively). The data demonstrate the importance of selecting the appropriate method of firmness determination and are consistent with the hypothesis that pectin fragments released by polygalacturonase contribute to the production of autocatalytic (system II) ethylene.  相似文献

Shade light found in ecological niches where plants are growing under a canopy or in proximity of taller neighbouring vegetation consist mainly of two separate light signals: low red to far-red ratio and low photosynthetically active radiation (PAR). The effect of the latter on the growth of 7-day old sunflower shoots was examined by assessing hypocotyl, cotyledon and leaf tissue growth under three varying PAR levels: near-normal of 1,000 μmol m?2 s?1, low of 100 μmol m?2 s?1 and very low of 10 μmol m?2 s?1. Then, the possible interaction between PAR signaling and ethylene in regulating growth of these sunflower tissues was investigated. The results showed that gradual decrease in PAR level increases hypocotyl elongation and decreases ethylene evolution. However, gradual decrease in PAR level decreases cotyledon and leaf growth and increases ethylene evolution. Thus it seems possible that PAR regulation of shoot growth is mediated by changes in ethylene evolution in tissue specific manner. This hypothesis was supported by experiments with the ethylene releasing factor, ethephon, and the ethylene biosynthesis inhibitor, AVG, as well as by transfer experiments where sunflower seedlings were transferred from one PAR regime to another with subsequent growth and ethylene measurements.  相似文献
