Plants grown in phosphorus-deficient solutions often exhibit disruption of water transport due to reduction in root hydraulic conductivity (Lpr) and enhanced ethylene production. To uncover the relationship between the reduction in Lpr and increase in ethylene production, we investigated effect of phosphorus (P) deficiency on ethylene production and Lpr in legume plants of Medicago falcata L. There was an increase in ethylene production and a reduction of Lpr of M. falcata roots when M. falcata seedlings grown in P sufficient solutions (0.5 mM H2PO42?) were transferred to P-deficient solutions (5 μM H2PO42?). Antagonists of ethylene biosynthesis, CoCl2 and aminoethoxyvinylglycine (AVG), abolished the P deficiency-induced ethylene production. Root hydraulic conductivity of M. falcata seedlings grown in P-sufficient solutions was insensitive to CoCl2 and AVG, while the two chemicals enhanced Lpr for those grown in P-deficient solutions, suggesting that P deficiency-induced decrease in Lpr can be reversed by inhibiting ethylene production. Ethylene precursor 1-amino cyclopropane-1-carboxylic acid (ACC) and ethylene donor ethephon had greater inhibitory effect on Lpr of P-sufficient seedlings than that of P-deficient seedlings. Root hydraulic conductivity of P-sufficient seedlings was more sensitive to HgCl2 than that of P-deficient seedlings. Taken together, these findings suggest that ethylene induced by P deficiency may play an important role in modulation of root hydraulic conductivity by affecting aquaporins in plants.  相似文献

Auxin is known to stimulate greatly both C2H4 production and the conversion of methionine to ethylene in vegetative tissues, while amino-ethoxyvinylglycine (AVG) or Co2+ ion effectively block these processes. To identify the step in the ethylene biosynthetic pathway at which indoleacetic acid (IAA) and AVG exert their effects, [3-14C]methionine was administered to IAA or IAA-plus-AVG-treated mung bean hypocotyls, and the conversion of methionine to S-adenosylmethionine (SAM), 1-amino-cyclopropane-1-carboxylic acid (ACC), and C2H4 was studied. The conversion of methionine to SAM was unaffected by treatment with IAA or IAA plus AVG, but active conversion of methionine to ACC was found only in tissues which were treated with IAA and which were actively producing ethylene. AVG treatment abolished both the conversion of methionine to ACC and ethylene production. These results suggest that in the ethylene biosynthetic pathway (methionine → SAM → ACC → C2H4) IAA stimulates C2H4 production by inducing the synthesis or activation of ACC synthase, which catalyzes the conversion of SAM to ACC. Indeed, ACC synthase activity was detected only in IAA-treated tissues and its activity was completely inhibited by AVG. This conclusion was supported by the observation that endogenous ACC accumulated after IAA treatment, and that this accumulation was completely eliminated by AVG treatment. The characteristics of Co2+ inhibition of IAA-dependent and ACC-dependent ethylene production were similar. The data indicate that Co2+ exerts its effect by inhibiting the conversion of ACC to ethylene. This conclusion was further supported by the observation that when Co2+ was administered to IAA-treated tissues, endogenous ACC accumulated while ethylene production declined.  相似文献

We tested that the hypothesis that root elongation might be controlled by altering the level of ethylene in intact primary roots of maize(Zea mays L.). We measured root elongation in a short period using a computerized root auxanometer. Compounds which regulate ethylene production were applied to intact primary roots in different time periods. Root elongation was stimulated by the treatment with ethylene antagonists such as Co2+, aminoethoxyvinylglycine (AVG) and L-canaline. This result suggested that root elongation was closely related to ethylene level of intact primary roots. Furthermore, IAA- and 1-aminocyclopropane-1-carboxylic acid (ACC)-induced inhibition of root elongation was reversed by treatment with Co2+. The application of ACC to roots which have been exposed to IAA and Co2+ have no significant effect on root elongation. However, the inhibition of root elongation by ACC in roots previously treated with IAA and AVG became manifest when the applied IAA concentrations were lower. These results were consistent with the hypothesis that the level of ethylene in intact roots functions to moderate root elongation, and suggested that auxin-induced inhibition of root elongation results from auxin induced promotion of ethylene production.  相似文献

Plants show various responses to phosphorus (P) deficiency. Root oxidizing capacity enhancement is one of adaptive mechanisms for rice (Oryza sativa L.) to P deficiency. However, it remains unclear how P deficiency enhances the root oxidizing capacity. In this study, rice seedlings were treated in P-deficient nutrient solution for different periods. Variations of reactive oxygen species (ROS), antioxidant enzyme activity, root lignin content, root porosity, root oxygen release, total oxidative substances and root structural changes in rice roots in response to P-sufficient and P-deficient treatments were investigated. Results indicated that P deficiency induced the production of H2O2 and O 2 ·? in roots significantly, which reached their maximum after 1- to 2-day P-deficient treatment. Interestingly, the endogenous total oxidative substances kept stable in rice roots. P deficiency increased the activities of peroxidase and superoxide dismutase by 89.5 and 51.8 % after 4-day P-deficient treatment, respectively. Moreover, one-day P deficiency elevated lignin accumulation. Root porosity of rice seedling under 2-day P-deficient treatment was 19.8 % higher than that under P-sufficient treatment. P deficiency also enhanced the release of both O2 and total oxidative substances after 1- to 4-day P deficiency. In addition, results from electronic microscopy indicated that the thickness of root cell wall tended to increase after 2-day P-deficient treatment. Taken together, our results suggested that P-deficiency-induced enhancement of root oxidizing capacity in rice roots was probably associated with ROS production, antioxidant enzyme activity increment in root tissues, and the release of O2 and oxidative substances from root inside to rhizosphere.  相似文献

Enhanced ethylene production and leaf epinasty are characteristic responses of tomato (Lycopersicon esculentum Mill.) to waterlogging. It has been proposed (Bradford, Yang 1980 Plant Physiol 65: 322-326) that this results from the synthesis of the immediate precursor of ethylene, 1-aminocyclopropane-1-carboxylic acid (ACC), in the waterlogged roots, its export in the transpiration stream to the shoot, and its rapid conversion to ethylene. Inhibitors of the ethylene biosynthetic pathway are available for further testing of this ACC transport hypothesis: aminooxyacetic acid (AOA) or aminoethoxyvinylglycine (AVG) block the synthesis of ACC, whereas CO2+ prevents its conversion to ethylene. AOA and AVG, supplied in the nutrient solution, were found to inhibit the synthesis and export of ACC from anaerobic roots, whereas Co2+ had no effect, as predicted from their respective sites of action. Transport of the inhibitors to the shoot was demonstrated by their ability to block wound ethylene synthesis in excised petioles. All three inhibitors reduced petiolar ethylene production and epinasty in anaerobically stressed tomato plants. With AOA and AVG, this was due to the prevention of ACC import from the roots as well as inhibition of ACC synthesis in the petioles. With Co2+, conversion of both root- and petiole-synthesized ACC to ethylene was blocked. Collectively, these data support the hypothesis that the export of ACC from low O2 roots to the shoot is an important factor in the ethylene physiology of waterlogged tomato plants.  相似文献

Summary This report describes the regeneration response of excised seedling roots of silktree (Albizzia julibrissin) to added ethylene precursors/generators (1-amino-cyclopropane-1-carboxylic acid [ACC], 2-chloroethylphosphonic acid [CEPA]), biosynthesis inhibitors (aminoethoxyvinylglycine [AVG], an oxime ether derivative [OED={[(ispropylidene)-amino]oxy}-acetic acid-2-(methoxy)-2-oxoethyl ester], CoCl2 [Co++]), and an ethylene action inhibitor (AgNO3 [Ag+]). When placed on B5 medium, about 50% of the control explants formed shoot buds within 15 days. Addition of ACC or CEPA (1–10 µM) to the culture medium decreased both the percentage of cultures forming shoots and the number of shoots formed per culture. In contrast, AVG and OED (1–10 µM) increased shoot formation to almost 100% and increased the number of shoots formed per culture. Likewise, both Co++ and Ag+ (1–10 µM) increased shoot regeneration, but the number of shoots produced after 30 days was less than with AVG or OED. The inhibitors of ethylene biosynthesis were partially effective in counteracting the inhibitory effect of ACC on shoot formation. Young, fully expanded leaves from 7-month-old P-deficient citrus rootstock seedlings had levels of nonprotein arginine that were 10- to 50-fold greater than those from P-sufficient control plants. Arginine content of the protein fraction increased 2- to 4-fold in P-deficient leaves. Total arginine content, which averaged 72 ± 6 micromoles per gram dry weight of P-sufficient leaf tissue (mean ± se, n = the four rootstocks) was 207, 308, 241, and 178 micromoles in P-deficient leaves from Citrus limon cv rough lemon, Poncirus trifoliata × C. sinensis cv Carrizo citrange and cv Troyer citrange, and P. trifoliata cv Australian trifoliate orange, respectively. For each rootstock, the accumulation of arginine paralleled an increase in the activity of the pathway for the de novo biosynthesis of arginine. The ratio of the nanomoles NaH14CO3 incorporated into the combined pool of arginine plus urea per gram fresh weight intact leaf tissue during a 3-hour labeling period for P-deficient to P-sufficient plants was 91:34, 49:11, 35:11, and 52:41, respectively. When P-deficient plants were supplied with P, incorporation of NaH14CO3 into arginine plus urea was reduced to the level observed for the P-sufficient control plants of the same age and arginine ceased to accumulate. Arginase and arginine decarboxylase activity were either unaffected or slightly increased during phosphorus deficiency. Taken together, these results provide strong evidence that arginine accumulation during phosphorus deficiency is due to increased activity of the de novo arginine biosynthetic pathway.  相似文献

Galactose, sucrose, and glucose (50 millimolar) applied to tobacco leaf discs (Nicotiana tabacum L. cv `Xanthi') during a prolonged incubation (5-6 d) markedly stimulated ethylene production which, in turn, could be inhibited by aminoethoxyvinylglycine (2-amino-4-(2′-aminoethoxy)-trans-3-butenoic acid) (AVG) or Co2+ ions. These three tested sugars also stimulated the conversion of l-[3,4-14C]methionine to [14C]1-amino-cyclopropane-1-carboxylic acid (ACC) and to [14C]ethylene, thus indicating that the carbohydrates-stimulated ethylene production proceeds from methionine via the ACC pathway. Sucrose concentrations above 25 mm considerably enhanced ACC-dependent ethylene production, and this enhancement was related to the increased respiratory carbon dioxide. However, sucrose by itself could directly promote the step of ACC conversion to ethylene, since low sucrose concentrations (1-25 mm) enhanced ACC-dependent ethylene production also in the presence of 15% CO2.  相似文献

White lupin (Lupinus albus L.) develops proteoid (cluster) rootsin response to phosphorus deficiency. Proteoid roots are composedof tight clusters of rootlets that initiate from the pericycleopposite protoxylem poles and emerge from every protoxylem polewithin the proteoid root axis. Auxins are required for lateralroot development, but little is known of their role in proteoidroot formation. Proteoid root numbers were dramatically increasedin P-sufficient (+P) plants by application of the syntheticauxin, naphthalene acetic acid (NAA), to leaves, and were reducedin P-deficient (-P) plants by the presence of auxin transportinhibitors [2,3,5-triiodobenzoic acid (TIBA) and naphthylphthalamicacid (NPA)]. While ethylene concentrations in the root zonewere 1.5-fold higher in -P plants, there was no effect on proteoidroot numbers of the ethylene inhibitors aminoethoxyvinvylglycine(AVG) and silver thiosulphate. Phosphonate, which interfereswith plant perception of internal P concentration, dramaticallyincreased the number of proteoid root segments in +P plants.Activities of phosphoenolpyruvate carboxylase (PEPC), malatedehydrogenase (MDH) and exuded acid phosphatase in proteoidroot segments were not different from +P controls when NAA wasapplied to +P lupin plants, but increased to levels comparableto -P plants in the phosphonate treatment. Addition of TIBAor NPA to -P plants reduced PEPC and MDH activity of -P proteoidroots to levels found in +P or -P normal root tissues, but didnot affect acid phosphatase in root exudates. These resultssuggest that auxin transport from the shoot plays a role inthe formation of proteoid roots during P deficiency. Most dicotyledonous species respond to Fe deficiency by developing some mechanisms known as Fe-deficiency responses. The role of ethylene in regulation of root ferric reductase activity of wild-type tomato (Lycopersicon esculentum L.) and its mutant Never ripe (Nr), bean (Phaseolus vulgaris L., cv. Bifeng 80-30), and cucumber (Cucumis sativus L., cv. Xintaimici) plants grown in nutrient solution without Fe supply was studied under controlled condition. The results show that: (i) the tomato mutant Nr, which is insensitive to ethylene, presented rapid increase in root ferric reductase activity after omitting Fe from the nutrient solution; (ii) the initial time for increase in root ferric reductase activity was earlier than that in ethylene production after onset of Fe deficiency in the three species; (iii) like cobalt (3 μM Co2+), an inhibitor for ethylene production, high concentration of zinc (50 μM Zn2+) and copper (5 μM Cu2+) also suppressed the increase in root ferric reductase activity of Fe-starved plants; (iv) under Fe-sufficient conditions, indol-3-butylric acid (IBA) stimulated root ferric reductase activity of cucumber and bean plants, and this stimulating effect could not be suppressed by aminoethoxyvinylglycine (AVG, an inhibitor for ethylene synthesis). These results suggested that ethylene might not be directly involved in the regulation of root ferric reductase activity of Fe-deficient dicotyledonous species.  相似文献

The promotive effect of AgNO3 and aminoethoxyvinylglycine (AVG) on in vitro shoot regeneration from cotyledons of Brassica campestris ssp. pekinensis in relation to endogenous 1-amino-cyclopropane-1-carboxylic acid (ACC) synthase, ACC, and ethylene production was investigated. AgNO3 enhanced ACC synthase activity and ACC accumulation, which reached a maximum after 3 to 7 days of culture. ACC accumulation was concomitant with increased emanation of ethylene which peaked after 14 days. In contrast, AVG was inhibitory to endogenous ACC synthase activity and reduced ACC and ethylene production. The promotive effect of AVG on shoot regeneration was reversed by 2-chloroethylphosphonic acid at 50 micromolar or higher concentrations, whereas explants grown on AgNO3 medium were less affected by 2-chloroethylphosphonic acid. The distinctive effect of AgNO3 and AVG on endogenous ACC synthase, ACC, and ethylene production and its possible mechanisms are discussed.  相似文献

In Anemia phyllitidis gametophytes two of the ethylene perception inhibitors (silver ions, Ag+; 2,5-norbornadiene, NBD) caused opposite effects on GA3-induced antheridia formation and on the increment of ACC (1-aminocyclopropane-1-carboxylic acid) content accompanying this process. Ag+ enhanced while NBD inhibited GA3-induced antheridiogenesis and each inhibitor modulated the level of ACC in a different manner. Cobalt ions (Co2+) and aminooxyacetic acid (AOA; the ethylene synthesis inhibitors), also modulated the level of GA3-induced ACC content differently. These results strongly confirm the earlier suggestion that ethylene plays a role of the second messenger in GA3-induced antheridiogenesis during “induction” and “expression” phases, and the 3rd h of the former phase is the time when elevation of ACC content induced while in the 6th h inhibited antheridiogenesis. Timing of changes in ACC content and morphogenetic effects of GA3-induced antheridiogenesis in A. phyllitidis gametophytes allowed to indicate that AOA together with NBD could participate in one while Co2+ and Ag+ in another ethylene synthesis and signaling pathway.  相似文献

It has long been known that applied ethylene can redirect the gravitropic response, but only occasionally has it been suggested that ethylene normally plays a role in gravitropism. Two inhibitors of ethylene synthesis [Co2+ and aminoethoxyvinylglycine (AVG)] and two inhibitors of ethylene action (Ag+ and CO2) were shown to delay the gravitropic response of cocklebur (Xanthium strumarium L.), tomato (Lycopersicon esculentum Mill.), and castor bean (Ricinus communis L.) stems. Gentle shaking on a mechanical shaker does not inhibit the gravitropic response, but vigorous hand shaking for 120 seconds delays the response somewhat. AVG and Ag+ further delay the response of mechanically stimulated plants. AVG delays the response of defoliated and of decapitated plants. Plants laid on their side and restricted so that they cannot bend upward store both bending energy and gravitropic stimulus; they bend immediately when released from restriction (stored energy) and continue to bend for some hours after (stored stimulus). AVG retards the storage of bending energy but not of stimulus. In gravitropism, graviperception may first stimulate ethylene evolution, which may then influence bending directly, or responses involving ethylene could be more indirect.  相似文献

Aminoethoxyvinylglycine (AVG) and cobalt ions strongly inhibit the conversion of added methionine or aminocyclopropane-1-carboxylic acid (ACC) into ethylene by green-coloured, non-stressed Norway spruce (Picea abies L.) needles but only 30%–40% of basal ethylene formation is affected by such inhibitors. In addition, free radical-mediated ACC-independent ethylene formation (AIEF) of the type released by brown-coloured spruce needles also occurs in extracts from healthy green-coloured needles. Treatment with CdCl2 (10 mM), Na2S2O5 (5 mM) or FeSO4 (10 mM) induces 3–7 fold increases in the rates of ethylene evolution from green-coloured needles. However, only Cd2+-induced ethylene formation is inhibited by AVG while ethylene induced by S2O5 2- or Fe2+ is insensitive to added AVG although increased levels of ACC have also been detected in these treatments. The role of phosphorus (P) status in root-zone CO2 utilisation for organic acid synthesis during Al3+ toxicity was assessed. Root-zone CO2 can be incorporated into organic acids via Phosphoenolpyruvate carboxylase (PEPC, EC 4.1.1.31). P-deficiency and Al3+ toxicity can induce organic acid synthesis, but it is unknown how P status affects the utilisation of PEPC-derived organic acids during Al3+ toxicity. Two-week-old Solanum lycopersicum seedlings were transferred to hydroponic culture for 3 weeks. The hydroponic culture consisted of a standard Long Ashton nutrient solution containing either 0.1 μM or 1 mM P. Short-term Al3+ toxicity was induced by a 60-min exposure to a pH-buffered solution (pH 4.5) containing 2 mM CaSO4 and 50 μM AlCl3. Al3+ toxicity induced a decline in root respiration, adenylate concentrations and an increase in root-zone CO2 utilisation for both P sufficient and P-deficient plants. However during Al3+ toxicity, P deficiency enhanced the incorporation and metabolism of root-zone CO2 via PEPC. Moreover, P deficiency led to a greater proportion of the PEPC-derived organic acids to be exuded during Al3+ toxicity. These results indicate that P-status can influence the response to Al3+ by inducing a greater utilisation of PEPC-derived organic acids for Al3+ detoxification.  相似文献

Ethylene production in rice bronzing leaves induced by ferrous iron   总被引:4,自引:0,他引:4  
Bronzing, a nutritional disorder of rice plants which is widely distributed in tropical lowlands, was induced by dipping the cut end of rice leaves into FeSO4 solution (pH 3.5). Ethylene production; the activities of peroxidase, polyphenol oxidase, and phenylalanine ammonia-lyase; and the effects of Co2+, aminoethoxyvinylglycine, Ag+, cycloheximide, and 1-aminocyclopropane-1-carboxylate, were investigated in the course of bronzing development. It was found that ethylene production could be stimulated up to about 20 times that of the control by Fe2+, and a peak could be reached at about 24 h after incubation. The Fe2+-treated leaves also had 10-fold higher peroxidase activity than the control, whereas in vitro enzyme activity was inhibited by Fe2+. Cycloheximide retarded in vivo stimulation of peroxidase, indicating that in vivo stimulation resulted from inducing de novo synthesis of the enzyme. No changes in the activities of phenylalanine ammonia-lyase and polyphenol oxidase were observed. The results, obtained from the incubation of leaves with Co2+, aminoethoxyvinylglycine, Ag+, cycloheximide, or 1-aminocyclopropane-1-carboxylate, showed that ethylene production was the effect of Fe2+ stress and that it was not involved in the process of bronzing development, which is probably an acclimation process to enable plants to cope with stress. Apples (Malus sylvestris Mill, cv Golden Delicious) were treated before harvest with aminoethoxyvinylglycine (AVG). AVG is presumed to reversibly inhibit 1-aminocyclopropane-1-carboxylic acid (ACC) activity, but not the formation of ACC synthase. AVG treatment effectively blocked initiation of autocatalytic ethylene production and ripening of harvested apples. Exogenous ethylene induced extractable ACC synthase activity and ripening in AVG-treated apples. Removal of exogenous ethylene caused a rapid decline in ACC synthase activity and in CO2 production. The results with ripened, AVG-treated apples indicate (a) a dose-response relationship between ethylene and enhancement of ACC synthase activity with a half-maximal response at approximately 0.8 μl/l ethylene; (b) reversal of ethylene-enhanced ACC synthase activity by CO2; (c) enhancement of ACC synthase activity by the ethylene-activity analog propylene.

Induction of ACC synthase activity, autocatalytic ethylene production, and ripening of preclimacteric apples not treated with AVG were delayed by 6 and 10% CO2, but not by 1.25% CO2. However, each of these CO2 concentrations reduced the rate of increase of ACC synthase activity.

The thermoinhibition at 35 and 32°C of pregermination ethylene production and germination in lettuce (Lactuca sativa L. cv Mesa 659) seeds was synergistically or additively alleviated by 0.05 millimolar kinetin (KIN) and 10 millimolar 1-aminocyclopropane-1-carboxylic acid (ACC). The synergistic effect of KIN + ACC on ethylene production and germination at 35°C was inhibited by Co2+ (44-46%) but not by aminoethoxyvinyl glycine (AVG). The uptake of ACC by the seed was not influenced by KIN. Upon slitting of the seed coats (composed of pericarp, testa and endosperm), following the uptake of chemicals, ACC was readily converted into ethylene at all temperatures, and the synergistic effects of KIN + ACC at 35°C were lost. At 35°C, KIN acted synergistically with ACC or ethephon (ETH) in alleviating the osmotic restraint. At 25°C, ETH was more active than KIN or KIN + ACC in overcoming the osmotic restraint. Thus, the integrity of the seed coats, the KIN-enhanced ACC utilization, and an interaction of KIN with the ethylene produced may be the basis for the synergistic or additive effects of KIN + ACC at high temperature.  相似文献

Co2+ promoted elongation of hypocotyl segments of light-grown cucumber (Cucumis sativus) seedlings. Time course and dose response data are presented and interactions with IAA, gibberellin, cyclohexanol, and cotyledons described. Segments without cotyledons responded to Co2+ only if grown in gas-tight vessels with IAA added. When bases of cotyledons were ringed with an inhibitor of auxin transport, Co2+ caused no growth promotion in the hypocotyl. Co2+ prevented lateral swelling of hypocotyls treated with supraoptimal IAA. Removal of ethylene from the atmosphere reduced the Co2+ response, but Co2+ did not counteract the inhibitory effect of increased ethylene levels. These results are consistent with the hypothesis that Co2+ promotes hypocotyl elongation by inhibiting ethylene production. The hypothesis was confirmed by a direct demonstration that Co2+, at growth-promoting concentrations, powerfully inhibited ethylene production in the cucumber hypocotyl.  相似文献
