During a screening programme for new medical agents, many aqueous extracts from 59 species of seaweed were found to possess bioactivity against murine immunocytes. Thirty-eight extracts (8 green, 12 brown, 18 red algae) showed suppressive effects on the mitogenic response. Furthermore, 16 extracts (2 green, 6 brown, 8 red algae) suppressed the production of Interleukin 1 (1L-1) from murine macrophage. Using the murine mixed lymphocyte reaction assay, suppressive effects were observed in 4 red algae, but none in green or brown algae. Nine seaweed extracts suppressed the production of secondary antibody (IgG, IgM). Extracts of 3 red algae suppressed strongly the proliferation of bone marrow cells, but 2 other red algae caused stimulation above 200%. This is apparently the first report showing immunosuppressive activity from marine algae.  相似文献

Antibiotic activity of lectins from marine algae against marine vibrios   总被引:6,自引:0,他引:6  
Saline and aqueous ethanol extracts of marine algae and the lectins from two red algal species were assayed for their antibiotic activity against marine vibrios. Experimental studies were also carried out on the influence of environmental factors on such activity, using batch cultures. The results indicated that many of the saline extracts of the algal species were active and that the activity was selective against those vibrios assayed. The algal extracts were active against Vibrio pelagius and the fish pathogen V. vulnificus, but inactive against V. neresis. Algal lectins from Eucheuma serra (ESA) and Galaxaura marginata (GMA) strongly inhibited V. vulnificus but were inactive against the other two vibrios. The antibacterial activity of algal extracts was inhibited by pretreatment with various sugars and glycoprotein. Extracts of the two red algae, E. serra and Pterocladia capillacea, in saline and aqueous ethanol, inhibited markedly the growth rate of V. vulnificus at very low concentrations. Culture results indicated that metabolites active against V. vulnificus were invariably produced in P. capillacea over a wide range of temperature, light intensity, and nutritional conditions. Enhanced antibacterial activity occurred when P. capillacea was grown under higher irradiance, severe nutrient stress and moderate temperature (20 °C), reflecting the specific antibiotic characteristics of this alga. The strong antibiotic activity of lectins towards fish pathogenic bacteria reveals one of the important roles played by algal lectins, as well as the potential high economic value of those marine algae assayed for aquaculture and for biomedical purposes.  相似文献

Powdered tissue from 46 species of air-dried marine algae (four green, 21 brown and 21 red algae) were screened for antitumor activity. Significant activity against Ehrlich carcinoma was found in the brown algae Scytosiphon lomentaria (69.8% inhibition), Lessonia nigrescens (60.0%), Laminaria japonica (57.6%), Sargassum ringgoldianum (46.5%), the red algae Porphyra yezoensis (53.2%) and Eucheuma gelatinae (52.1%) and the green alga Enteromorpha prolifera (51.7%). Five brown and four red algae showed appreciable antitumor activity against Meth-A fibrosarcoma. To identify specific molecules with antitumor activity, 15 kinds of polysaccharide preparations of seaweed origin and 24 kinds of lipid fractions extracted from various seaweeds were tested. Appreciable inhibition of Ehrlich carcinoma was found for fucoidan preparations from Undaria pinnatifida and Sargassum ringgoldianum, for carrageenans and for porphyran. Several glycolipid and phospholipid fractions from brown and red algae were effective against Meth-A fibrosarcoma.  相似文献

Crustose red algae induce substratum-specific settlement, attachment and metamorphosis of the planktonic larvae of Haliotis rufescens Swainson (gastropod mollusc), upon direct contact by the larvae with any of a number of algal species tested. Larvae are not induced by contact with intact foliose red, brown or green macroalgae. Geniculate red algae are only slightly active. Larval settlement and metamorphosis are shown to be triggered by a class of chemical inducers associated with macromolecules and found in extracts of all species of crustose, geniculate, and foliose red algae tested; these inducers are not found in extracts of brown or green macroalgae. The substratum specificity of larval settlement and metamorphosis is shown to result from the unique availability of these inducers at the surfaces of the crustose red algae. Using a newly-developed improved method of purification based upon size-separation by gel-filtration, followed by ion-exchange chromatography over a diethylaminoethyl (DEAE)-acrylamide matrix, the principal inducer of Haliotis larval settlement and metamorphosis has been resolved from the red algal phycobiliproteins. Sensitivity of this inducer to reduction in molecular weight by digestion with trypsin demonstrates that this inducer is associated with protein.  相似文献

The use of nitrogen‐to‐protein conversion factors (N‐Prot factors) is the most practical way of determining protein content. The accuracy of protein determination by this method depends on the establishment of N‐Prot factors specific to individual species. Experimental data are needed to allow the use of this methodology with seaweeds. The present study was designed to characterize the amino acid composition and to establish specific N‐Prot factors for six green, four brown and nine red marine algae. Mean values for individual amino acids tended to be similar among the three groups, but some differences were found. Green algae tended to show lower percentages of both aspartic acid and glutamic acid than the other two groups of algae. The percentages of both lysine and arginine were higher in red algae, while brown algae tended to show more methionine than green and red algae. The actual protein content of the species, based on the sum of amino acid residues, varied from 10.8% (Chnoospora minima, brown algae) to 23.1% (Aglaothamnion uru‐guayense, red algae) of the dry weight. Nitrogen‐to‐protein conversion factors were established for the species studied, based on the ratio of amino acid residues to total nitrogen, with values ranging from 3.75 (Cryptonemia seminervis, red algae) to 5.72 (Padina gymnospora, brown algae). The relative importance of non‐protein nitrogen is greater in red algae, and consequently lower N‐Prot factors were calculated for these species (average value 4.59). Conversely, protein nitrogen content in both green and brown algae tends to be higher, and average N‐Prot factors were 5.13 and 5.38, respectively. An overall average N‐Prot factor for all species studied of 4.92 ± 0.59 (n = 57) was established. This study confirms that the use of the traditional factor 6.25 is unsuitable for seaweeds, and the use of the N‐Prot factors proposed here is recommended.  相似文献

Summary The hypothesis that sea urchin grazing and interactions with turf-forming red algae prevent large brown algae from forming an extensive canopy in the low intertidal zone of southern California was tested with field experiments at two study sites. Experimental removal of sea urchins resulted in rapid algal recruitment. Crustose coralline algae which typically dominate the substratum in areas with dense urchin populations were quickly overgrown by several species of short-lived green, brown and red algae. The removal of urchins also significantly increased the recruitment of two long-lived species of large brown algae (Egregia laevigata and Cystoseira osmundacea at one study site and E. laevigata and Halidrys dioica at the other). The experimental plots at both sites were eventually dominated by perennial red algae.A two-factorial experiment demonstrated that sea urchin grazing and preemption of space by red algae in areas where urchins are less abundant are responsible for the rarity of large brown algae in the low intertidal of southern California. The three dominant perennial red algae, Gigartina canaliculata, Laurencia pacifica and Gastroclonium coulteri, recruit seasonally from settled spores but can rapidly fill open space with vigorous vegetative growth throughout the year. These species encroach laterally into space created by the deaths of large brown algae or by other disturbances. Once extensive turfs of these red algae are established further invasion is inhibited. This interaction of algae which proliferate vegetatively with algae which recruit only from settled spores is analogous to those which occur between solitary and colonial marine invertebrates and between solitary and cloning terrestrial plants.It is suggested that a north-south gradient in the abundance of vegetatively propagating species, in grazing intensity and in the frequency of space-clearing disturbances, may account for latitudinal variation in intertidal algal community structure along the Pacific coast of North America.  相似文献

Trypanosoma cruzi, the causative agent of Chagas' disease, replicates in mammalian cells and relies on the de novo pyrimidine biosynthetic pathway that supplies essential precursors for nucleic acid synthesis. The protozoan dihydroorotate dehydrogenase (DHOD), the fourth enzyme of the pathway catalyzing production of orotate from dihydroorotate, markedly differs from the human enzyme. This study was thus aimed to search for potent inhibitors against T. cruzi DHOD activity, and a number of methanol extracts prepared from green, brown, and red algae were assayed. The extracts from two brown algae, Fucus evanescens and Pelvetia babingtonii, yielded 59 and 58% decrease in the recombinant DHOD activity, respectively, at the concentration of 50 microg/ml. Inhibition by these extracts was noncompetitive with respect to dihydroorotate, with apparent Ki values of 35.3+/-5.9 and 10.3+/-4.4 microg/ml, respectively. Further, in an in vitro T. cruzi-HeLa cell infection system, ethanol-reconstituted F. evanescens and P. babingtonii extracts at the concentration of 1 microg/ml, respectively, decreased significantly the infection rate of host cells and the average parasite number per infected cell. These results imply that F. evanescens and P. babingtonii contain inhibitor(s) against the T. cruzi DHOD activity and against the protozoan infection and proliferation in mammalian cells. Identification of inhibitor(s) in these two brown algae and further screening of other marine algae may facilitate the discovery of new, anti-trypanosomal lead compounds.  相似文献

Extracts from 44 species of Vietnamese marine algae, including 15 Chlorophyta, 18 Rhodophyta and 11 Phaeophyta species, were examined for hemagglutination activity with a variety of different animal and human erythrocytes that were untreated or treated with enzymes. Almost all extracts showed activity toward at least one type of erythrocytes, although those from three Chlorophyta and two Rhodophyta species showed no hemagglutination with any type of erythrocytes examined. Strong activity was detected in extracts from two Chlorophyta (Anadyomene plicata and Avrainvillea erecta) and four Rhodophyta species (Gracilaria eucheumatoides, Gracilaria salicornia, Kappaphycus alvarezii, and Kappaphycus striatum) with enzyme-treated rabbit and sheep erythrocytes. The hemagglutinins of seven Chlorophyta and eight Rhodophyta species were examined for sugar-binding specificity, pH- and temperature-stability, and divalent cation-independency of hemagglutination using ammonium sulfate-precipitates prepared from their extracts. In a hemagglutination-inhibition test with various monosaccharides and glycoproteins, none of the hemagglutinins had affinity for monosaccharides, except the Codium arabicum and Gracilaria euchematoides hemagglutinins, whose activities were inhibited by both N-acetyl-d-galactosamine and N-acetyl-d-glucosamine. On the other hand, all of the hemagglutinins activities were inhibited by some glycoproteins. The inhibition profiles with glycoproteins were different depending on hemagglutinin species, and suggest the presence of lectins specific for high mannose N-glycans, complex N-glycans, or O-glycans. The activities of these algal hemagglutinins were stable over a wide range of pH and temperature, and independent of the presence of divalent cations. These results indicate that Vietnamese marine algae are a good source of novel and useful lectins.  相似文献

Stable carbon and hydrogen isotope ratios were determined for the saponifiable lipid fraction as well as the cellulose fraction (the latter after nitration to remove exchangeable hydrogens) of several species of red, brown and green algae from three locations. A significant correlation was observed between the hydrogen isotope ratios of cellulose nitrate and saponifiable lipid for red algae, but not for brown or green algae. Carbon-13/carbon-12 ratios for both fractions of red algae were in general lower than those observed for brown and green algae. Four species of red marine algae (Rhodophyceae), five species of brown marine algae (Pheophyceae) and two species of green marine algae (Chlorophyceae) were examined for the fatty acid composition of the three lipid groups separated by silica gel column chromatography (neutral lipids, glycolipids, phospholipids). The four red algae had high contents of 16:0 and C20-polyunsaturated fatty acids (PUFA), 20:5n-3 ranging from 18 to 49% of the total fatty acid content and 20:4n-6 from 1.4 to 22.5%, these fatty acids were evenly distributed in all lipid groups. The five brown algae had high contents of 18:1n-9, 18:2n-6 and 18:3n-3 but low content of 20:5n-3. No precise trend was detected for the distribution of these fatty acids in the three lipid groups. The two green algae had high contents of 16:0, 18:1n-7 and 18:3n-3 and a very low content of PUFA. They contained also large amounts of 16:4n-3 together with 16:2n-6 and 16:3n-3. While 16:2n-6 was mainly found in phospholipids, 16:4n-3 was mainly distributed in neutral lipids and glycolipids.Porphyra umbilicalis represents the richest source of 20:5n-3 whileUndaria pinnatifida can be selected when a balanced mixture of (n-6) and (n-3) PUFA is required.Author for correspondence  相似文献

A general list of the flora of Vostok Bay (Sea of Japan) compiled on the basis of original and literature data includes 161 species, among them 25 green algae, 42 brown algae, 90 red algae, and 4 species of seagrass. In the period from 2000 to 2004, 114 species of macrophytes (19 green algae, 29 brown algae, 63 red algae, and 3 species of seagrass) were found in Vostok Bay; 19 species of algae were first records for this bay. On the whole, the flora of Vostok Bay is dominated by warm-water species, among which Asian low-boreal species occupy the leading position. The flora of Vostok Bay is changing under the influence of global climatic and local anthropogenic factors. In recent years, there is a tendency for the proportion of warm-water species to increase. The local anthropogenic influence on the flora of Vostok Bay has caused changes in the species composition of plant communities.  相似文献

Twenty-one species of seaweed from the California coast were analyzed for rhenium. For the first time, high enrichment (thousandfolds) of rhenium relative to seawater was found in brown algae, but not in green or red algae. Brown algae was suggested as a biological sink of rhenium in the sea and the analogous behavior of technetium to rhenium was found in marine algae. Preliminary incubation experiments with a common brown alga (Pelvetia fastigiata) showed that algal surface is not a major accumulating locus of rhenium.  相似文献

Jordan has witnessed a rapid industrial development in the last twenty years. This has lead to the release of waste materials or pollutants into the marine environment, particularly nearby Aqaba Port. The present study investigates the levels of zinc, cadmium and lead in four brown algae, three red algae and four green algal species collected from Aqaba. Three different levels of lead and zinc concentrations were found: the highest level of both metals is exhibited among brown algae; intermediate level is exhibited among red algae and the lowest level is seen among the green algae. Very low concentrations of cadmium were found in all examined algal species. The results indicate that the brown algal species Cystosira myrica, Sargassum asperifolium, Sargassum neglectum, and Sargassum subrepandum always contain the highest concentrations of lead and zinc, but these algae are less contaminated than brown algae from industrial European seas.  相似文献

Only a few animal phyla have been screened for the presence and distribution of lectins. Probably the most intensively studied group is the mollusk. In this investigation, 22 species from 12 families of tropical sponges collected in Los Roques National Park (Venezuela) were screened for the presence of lectins. Nine saline extracts exhibited strong hemagglutinating activity against pronase-treated hamster red blood cells; five of these reacted against rabbit red blood cells, four with trypsin-treated bovine red blood cells, and five with human red blood cells regardless of the blood group type. Extracts from the three species studied from genus Aplysina (archeri, lawnosa, and cauliformis) were highly reactive and panagglutinating against the panel of red blood cells tested. The lectins from A. archeri and A. lawnosa were purified to homogeneity by ammonium sulfate fractionation, affinity chromatography on p-aminobenzyl-beta-1-thiogalactopyranoside-agarose, and gel filtration chromatography. Both lectins exhibited a native molecular mass of 63 kDa and by SDS-polyacrylamide gel electrophoresis under reducing conditions have an apparent molecular mass of 16 kDa, thus suggesting they occur as homotetramers. The purified lectins contain 3-4 mol of divalent cation per molecule, which are essential for their biological activity. Hapten inhibition of hemagglutination was carried out to define the sugar binding specificity of the purified A. archeri lectin. The results indicate a preference of the lectin for nonreducing beta-linked d-Gal residues being the best inhibitors of red blood cells binding methyl-beta-d-Gal and thiodigalactoside (Gal beta 1-4-thiogalactopyranoside). The behavior of several glycans on immobilized lectin affinity chromatography confirmed and extended the specificity data obtained by hapten inhibition.  相似文献

The Forsmark Biotest Basin is a shallow coastal ecosystem that receives brackish cooling-water discharge from a nuclear power plant. The effects of the discharge on epilithic algal communities were investigated by analysing samples taken every third week throughout one year at 11 sites differentially affected by temperature and/or flow rate enhancement. Community variation was summarized in a canonical correspondence analysis (CCA) of species abundances as a function of site and date. The temperature increase favoured blue-green algae at the expense of red and brown algae. Blue-green algae were however abundant in summer in stagnant water, whether heated or not, and some red and brown algae became abundant in winter in heated sites with flowing water. Green algae and diatoms increased in biomass in the heated sites, but not in relative cover-abundance. The absence of ice and snow cover at sites with heated and/or flowing water caused autumn species to persist into winter, because of the higher light intensity (compared with natural conditions) and the absence of the mechanical abrasion by ice. The thermal discharge lowered species diversity (Shannon-Weaver index) both in summer and winter at sites with flowing water, but not at sites with quiescent or stagnant water. CCA showed alternate periods of stability and rapid change within the seasonal cycle. Individual species were placed according to their optimum; red and brown algae in winter/spring, green algae in spring/summer, blue-green algae in summer, and diatoms at various times. Exceptions to this pattern were species endo- or epiphytic on species of a different group. Analysis of the effects of temperature, flow rate and ice cover on the seasonal pattern of particular species showed that different species respond in individualistic ways to different combinations of these environmental variables.  相似文献

Synopsis A revitalized view of feeding by herbivorous marine fishes is sought through two questions. First, What characteristics of major taxa of algae identify them as predictably high or low quality foods? Second, are marine algae valuable foods for fishes which do not mechanically disrupt cell walls and do not harbor specialized enzymes or microbes capable of lysing cell walls? Energy, ash and nutrient content of 16 species of marine algae were employed to assess food quality of fleshy red, green, brown and calcareous red algae. On the basis of ash, calories, total protein and total lipid content, fleshy algae should be superior to calcareous algae as foods for fishes; in addition, green algae should be superior to brown algae and brown algae superior to red algae. When the probable digestibility of storage and extracellular carbohydrates is considered, green and red algae are predicted superior to brown algae as food. Two species of damselfishes (Pomacentridae) from the Gulf of California,Eupomacentrus rectifraenum andMicrospathodon dorsalis, eat red and green algae and ignore brown and calcareous algae. They feed, therefore, in a fashion consistent with predictions based only on algal chemistry. These fishes absorb at least 20–24% of the biomass, 57–67% of the protein, 46–56% of the lipid and 37–44% of the carbohydrate contained in algae eaten in the wild. Since these damselfishes do not masticate their food, it appears that herbivorous fishes can digest major fractions of algal nutrients without mechanical destruction of algal cells.  相似文献

Green plants including representatives of angiosperms, gymnosperms, ferns, mosses, liverworts and green algae were shown to contain a specific sucrose phosphatase, the last enzyme in the pathway of sucrose synthesis. The enzyme from all species required Mg2+ for activity and it was partially inhibited by sucrose. Abstract Among the brown algae, species of the Fucaceae (Pelvetia, Fucus and Ascophyllum) were found to have a ‘photosynthetic buffering’ system, allowing the algae to carry out oxygen production without a concomitant uptake of inorganic carbon. This system was not found in other brown algae examined (e.g. Halidrys, Laminaria and Desmarestia) nor in 16 examined species of red and green algae. Pelvetia, Fucus and Ascophyllum belong to the littoral algae which are periodically emersed. In the Fucaceae, the meristodermal cells were found to have a special organization of organelles. Towards the outer cell wall there was a prominent layer of mitochondria while the chloroplasts were concentrated towards the inner and side walls. Between the mitochondria and the chloroplasts there was a large number of physodes. This arrangement of organelles was not found in the other brown algae examined nor in red or green algae. The significance of this organization of the mitochondria is discussed in connection with the function of the ‘photosynthetic buffering’ system.  相似文献

Summary Gibberellin-like activity was detected in extracts from Enteromorpha prolifera (Chlorophyta) and Ecklonia radiata (Phaeophyta). Material eluted from chromatograms of extracts and active in dwarf maize also brought about growth responses in the species from which the extract had been made. The responses were similar to those brought about by gibberellic acid. It is concluded that gibberellins are normally involved in regulating growth in green and brown algae.  相似文献

We examined palatability of 37 species of nonencrusting macroalgae from the Antarctic Peninsula. This represents approximately 30% of the entire antarctic macroalgal flora and 75% of the 49 nonencrusting species we collected. Organic extracts from most species were also prepared and mixed into artificial foods. We examined palatability using feeding bioassays with three common, macroalga‐consuming animals (an omnivorous antarctic rockfish, Notothenia coriiceps; an omnivorous sea star, Odontaster validus; and a herbivorous amphipod, Gondogenia antarctica). Thallus pieces from 23 of 34 macroalgal species tested with the fish (68%) were rejected. Of the 23 species rejected as thallus, organic extracts of 16 were bioassayed using the fish with 9 (56%) unpalatable. Thallus pieces from 21 of 36 macroalgal species tested with the sea star (58%) were rejected. Of the 21 species rejected as thallus, organic extracts of 20 were bioassayed using the sea stars and 14 (70%) were unpalatable. Overall, 28 of the 37 species assayed as thallus (76%) were rejected by either or both the fish and sea stars. The amphipod assay was not suitable for use with thallus but was utilized with organic extracts of 23 macroalgal species that were rejected as thallus by either or both the fish and sea stars. Of these, 14 (61%) of the species' extracts were rejected by the amphipods. Unpalatability was highest among the brown algae examined with only an ephemeral, ectocarpoid species not rejected as thallus out of 10 species tested. Of the remaining nine brown algal species, six of seven tested were also unpalatable as extracts, including all the ecologically dominant, perennial species in the area. We conclude that unpalatability to herbivores is common in antarctic macroalgae and that chemical defenses may play an important role in the unpalatability of many algal species (NSF OPP9814538, OPP9901076).  相似文献
