A method was developed for the determination of selenium concentration in serum by flow injection-hydride generation-atomic absorption spectrometry (FI-HG-AAS) following microwave digestion of serum samples and reduction of selenate to selenite. The detection limit of the method was 0.3 μg Se/L and the characteristic concentration, corresponding to the 0.0044 absorbance signal, was 0.12 μg Se/L. The results from the analysis of two Seronorm standard reference materials showed good agreement with the certified values. The method was then used to analyze selenium in sera of Austrian and Slovenian people for the calculation of dietary intakes. The selenium concentrations in sera of mothers at delivery, their neonates, and the male and female adults were 71 ± 14, 42 ± 6, 75 ± 21, and 65 ± 16 μg/L for the Austrians and 62 ± 15, 34 ± 7, 70 ± 12, and 66 ± 15 μg/L for the Slovenians. The dietary intakes of selenium of the mothers and the male and the female adults were calculated as 52, 37, and 46 μg/d for the Austrians and 45, 38, and 32 μg/d for the Slovenians.  相似文献

This article describes a study in which four trace elements (Se, Mn, Cu, and Fe) were analyzed in the blood serum of the patients with colorectal cancer from the Moravian region of the Czech Republic. Atomic absorption spectrometry with graphite furnace atomization was used for analysis of selenium and manganese and with flame atomization for analysis of copper and iron. The observed serum concentrations in adenocarcinoma colorectal patients of selenium were significantly lower (41.8 ± 11.6 μg/L) and those of manganese (16.3 ± 4.5 μg/L) and iron (2.89 ± 1.23 mg/L) were significantly higher as compared to the age-matched control group. Copper serum content (0.95 ± 0.28 mg/L) did not significantly differ as compared to healthy population.  相似文献

The effects of substrate composition and temperature on myceilal growth and sclerotium production in Grlfola umbellate (Pers.) Pilaet were Investigated In the present study. The Induction of sclerotla of G. umbellate was affected greatly by the type of medium, as well as the type of carbon source. Malt-extract agar was able to induce the production of sclerotia. The production of sclerotia was also observed when the carbon source in the GPC agar medium (glucose 20 g/L, peptone 6 g/L, corn steep liquor 10 g/L, and agar 15 g/L) was replaced with glycerol or mannitol. Altering the composition of the GPC medium with milk powder, thiamine hydrochlorlde, extract of Armlllarla mellea, active clay, dlatomite, kaolin, or arginlne did not induce the production of sclerotla. A temperature range of 18-25 ℃ was suitable for both mycellai growth and sclerotium formation. Glycerol significantly Induced slerotium formation on nutrient supplemented with sawdust substrates In bottle culture. 24S-Polyporusterone A and polyporusterone B were assayed In samples of natural and cultured sclerotla. The present study was undertaken to examine possible aluminum (Al) accumulation in the brain of rats and to investigate whether subchronic exposure to the metal leads to behavioral and neurophysiological changes in both treated and control groups. Each of the groups consisted of 10 animals. Aluminum chloride (AlCl3) at a low (50 mg/kg/d) or high (200 mg/kg/d) dose was applied to male Wistar rats by gavage for 8 wk. Al-free water by gavage was given to the control group throughout the experiment. Behavioral effects were evaluated by open-field (OF) motor activity and by acoustic startle response (ASR). Electrophysiological examination was done by recording spontaneous activity and sensory-evoked potentials from the visual, somatosensory, as well as auditory cortex. The Al content of each whole brain was determined by electrothermal atomic absorption spectrophotometry. Subchronic Al exposure slightly caused some changes in the evoked potentials and electrocorticograms and in the OF and ASR performance, but these results were not statistically significant. The brain Al levels of the control and the low and high dose of Al-exposed groups were measured as 0.717±0.208 μg/g (wet weight), 0.963±0.491 μg/g (wet weight) and 1.816±1.157 μg/g (wet weight), respectively.  相似文献

The concentrations of rare earth elements (REs) in atmospheric particles, soil, soil water, surface runoff, and different parts of corn in an experimental plot in the suburb of Beijing, China were measured and the flux of REs was estimated. The concentration of REs in air particles with diameter less than 10 μm is 36 ng/m3 and, by ratio analysis, the origin of REs in the atmosphere was likely local soil. The concentration of soluble REs is 0.69 μg/L in rainwater, 5–7 μg/L in surface runoff, as well as 1–4 μg/L in soil water and the application of RE mixture has no observed influences on their concentrations. The concentration of soluble REs in surface runoff and soil water was dominated by aqueous-solid-phase partitioning. By estimation of the flux, most of the REs applied will remain in the soil phase and continued application of REs will cause an accumulation of REs in agricultural soil.  相似文献

Selenium in serum and selenium and glycogen in erythrocytes were determined in diabetic patients divided into noninsulin-dependent (n=50) and insulin-dependent (n=31) groups according to the etiopathogenesis of their diabetes. Selenium was determined by the method of atomic absorption spectrometry. Serum level of selenium was statistically significantly different in patients with either noninsulin-dependent (59.23±12.2 μg/L) or insulin-dependent (58.23±16.7 μg/L) diabetes mellitus as compared with the control group of 62 subjects (64.2±11.5 μg/L; p<0.05). There was no statistically significant difference in the serum levels of selenium between the groups of patients with noninsulin-dependent and insulin-dependent diabetes mellitus. The levels of erythrocyte glycogen were 2.0580±1.326, 2.0380±1.735, and 2.0036±1.3537 μg/g Hb in the control group, noninsulin-dependent group, and insulin-dependent group, respectively, with no statistically significant between-group difference. The decreased levels of selenium in serum and erythrocytes of diabetic patients suggest the possible role of glutathione peroxidase activity.  相似文献

The response of Potamogeton crispus L. breakdown to controlled doses of different levels of chlorine and chlorine + ammonia was investigated over two years in outdoor experimental streams. In 1985, downstream riffles of 2 streams were dosed (observed in-stream concentrations) at ca. 10 μg/L Total Residual Chlorine (TRC), one stream at 64 μg/L TRC and one stream at 230 μg/L TRC. Two control streams were not dosed and the upstream riffles of each stream served as within stream controls. In 1986, the downstream riffle of one stream was dosed at 70 μg/L TRC and a second stream was dosed at 200 μg/L TRC. Four streams were also dosed with 2.5 mg/L NH3-N: one stream with no chlorine, one stream with ca. 10 μg/L TRC, one with 56 μg/L TRC, and one with 150 μg/L TRC. A seventh stream was dosed for 2 h at 2000 μg/L TRC and 2.5 mg/L ammonia and then allowed to recover (recovery stream). Each year, litter decomposition (degree day k values) was measured during two 35 day trials (Jun–Jul and Aug–Sep). In 1985, when streams were dosed with chlorine alone, decomposition was significantly reduced with the high (230 μg/L TRC) chlorine dose. Downstream decomposition was 27% (Jun–Jul) and 59% (Aug–Sep) of the upstream (control) rate. No other chlorine effects were found during this period. In Jun–Jul 1986, there was significantly lower decomposition in the downstream dosed sites of the 200 μg/L TRC alone stream, the 146 μg/L TRC + ammonia stream and the recovery stream; downstream decay rates were (respectively) 56%, 42% and 64% of the upstream control sites. No other up-down pairs were different in July 1986. In Aug–Sep, all three streams with chlorine + ammonia (6, 56 and 146 μg/L TRC + 2,5 mg/L ammonia) and the 70 μg/L TRC alone stream had significantly lower decomposition rates in the downstream dosed sites. For these streams, downstream decay rates ranged from 46% (high chlorine + ammonia) to 73% (low chlorine + ammonia) of the upstream control rates. No other up-down pairs were different during this trial. Up and downstream sites of the stream dosed with 2.5 mg/L ammonia alone were nearly identical for both trials (< 3% difference). Using standard and internationally validated methods,86 anthropologic characteristics were determined in 650 male(305 from urban areas and 345 from rural areas) and 704 female(331 from urban areas,373 from rural areas) Chinese Hakka adults living in Guangdong and Jiangxi.The data were used to calculate 24 anthropologic indices,which were analyzed statistically.The physical characteristics of Hakka subjects were analyzed and compared with reference ethnic data.There were four main findings of this study.First,a small proportion of Hakka adults had an eye fold on the upper eyelid,but a large proportion had a mongoloid fold.The eye slits were narrow in most adults,had a medium nasal root height and straight bridges,and most of the external angles were prominent.The nasal base was upturned in most men.The distributions of the three types of nasal base in women were similar.The proportions of subjects with middle and high alae nasi heights were high and similar.Males with a maximum nostril diameter were mostly classified as transverse and oblique,while many women were classified as transverse and had relatively wide alae nasi.The round lobe type was the most common.Upper lip skin height was mostly classified as medium.Lips were classified as thin.The hair was black,eyes were brown,and the skin was yellowish.Second,the head length was long in male Hakka.The minimum frontal breadth,face breadth,lip height,and interocular breadth were similar to those of North-Asian populations.Meanwhile,head breadth,morphological facial height,nose breadth,mouth breadth,and nose height were similar to those of South-Asian populations.Head length was long in female Hakka.The minimum frontal breadth,face breadth,lip height,and interocular breadth were similar to those of North-Asian populations.Head breadth,nose breadth,and mouth breadth were similar to those of South-Asian populations.Third,the stature of male and female Hakka in urban and rural areas was classified as medium.The proportions of male and female Hakka classified as mesocephaly(length-breadth index of the head),hypsicephalic type,metriocephalic type(breadth-height index of the head) mesorrhiny,long trunk,subbrachyskelic type,broad shoulder breadth,and narrow distance between iliac crests were higher than those of other types.Finally,principal component analyses showed that the physical characteristics of Hakka were between those of South-Asian and North-Asian ethnic populations,but were generally closer to those of North-Asian populations in China.  相似文献

Isolation of Fucoxanthin from the Rhizoid of Laminaria japonica Aresch   总被引:2,自引:0,他引:2  
Fucoxanthin was extracted from the intact rhizoid of Laminariajaponica Aresch with dimethyl sulfoxide (DMSO), and then recovered from the DMSO extract by partitioning into ethyl acetate and subsequent evaporation. Some isolation conditions such as solvent volume and extraction time were screened. The quantity and quality of the extracted fucoxanthin were determined by spectral analysis (absorption spectra and fluorescence emission spectra). The results indicated that: (1) the average total content of fucoxanthin was 122.1μg in 1 g of fresh L. japonica rhizoid; (2) in comparison with the widely used organic solvent, acetone, DMSO was much more effective for the extraction of fucoxanthin; (3) both DMSO volume and extraction time influenced extraction efficiency such as the recovery rate and purity of fucoxanthin (1 g of fresh L. japonica rhizoid treated with 4 mL DMSO for 60 min, yielded 〉 88% of the total fucoxanthin with purity 0.63); (4) when (NH4)2SO4 concentration was in the range of 0.5-1.0 mol/L, the pigments rapidly and entirely moved from DMSO into the ethyl acetate phase; (5) the ethyl acetate and DMSO were recycled using a rotary evaporator.  相似文献

Vanadium was determined in 51 solutions and drugs for intravenous administration and in 6 salt components of a multitrace element solution using electrothermal atomic absorption spectrometry. The highest V contaminations were found in albumin solutions showing values of more than 600 μg/L V. Two heparines contained 14.3 and 122 μg/L V, respectively. In the ultratrace element solution, 14.8 μg/L vanadium were determined. The most contaminated salt of this solution was Mn(II)-dl-aspartate (12.8 μg V/g). The concentrations of unbound V (V f ) in the albumin solutions were between 0.31 and 299 μg/L. The manufacturing process is the reason for V contamination. The biological half-life of V administered intravenously by albumin solutions in man was about 125 h.  相似文献

The water relations, the photosynthetic capacity and the pigment content of leaves, i.e. chlorophylls, carotenes and xanthophylls, were analysed during the summer drought and recovery after autumn rainfalls in lavender ( Lavandula stoechas L.) plants grown in Mediterranean field conditions. Summer drought caused photoinhibition of photosynthesis and significant decreases in chlorophylls (by ca 75%), β -carotene (by ca 65%), and lutein and neoxanthin (by ca 50%), although their contents remained unaltered between predawn and midday, suggesting a progressive decrease in response to drought. In contrast, the levels of violaxanthin decreased from predawn to midday, giving rise to enhanced formation of zeaxanthin and antheraxanthin in high light. Zeaxanthin and antheraxanthin formation was not induced by water deficit. Although the levels of photosynthetic pigments were severely affected by water deficit, carotenoids decreased less than chlorophylls, which resulted in increased levels of carotenoids per unit of chlorophyll. We conclude that the enhanced formation of zeaxanthin in high light and the increased levels of carotenoids per unit of chlorophyll observed in water-stressed plants may help to avoid photoinhibitory damage to the photosynthetic apparatus.  相似文献

In order to assess drought tolerance mechanism in cotton, short-term drought-induced biochemical responses were monitored in two cotton (Gossypium hirsutum L.) genotypes contrasting their tolerance to water deficit. The seeds of two genotypes, namely GM 090304 (moderately drought tolerant) and Ca/H 631 (drought sensitive), were sown in pots containing soil, sand and peat in the ratio of 1:1:1, and irrigated every alternate day up to 45 days after sowing when each genotype was subjected to a cycle of water stress by withholding irrigation for 7 days. The stress cycle was terminated by re-watering the stressed plants for 7 days. The leaf of the drought tolerant genotype (GM 090304) maintained higher relative water content under water stress than that of the drought sensitive genotype (Ca/H 631). The levels of biochemical components, such as chlorophylls, carotenoids, total protein, free proline, total free amino acids, sugars, starch and polyphenols, were measured during the stress as well as the recovery periods. The chlorophylls, carotenoids, protein and starch contents decreased in drought stressed plants as compared to control and tended to increase when the plants were recovered from stress. The degree of decrease in chlorophylls, carotenoids and protein contents under drought was higher in the sensitive genotype (Ca/H 631) as compared to the moderately tolerant genotype (GM 090304). However, proline, total free amino acids, total sugars, reducing sugars and polyphenol contents were increased in drought stressed plants and tended to decrease during the period of recovery. Drought-induced increases in total free amino acids, proline, sugars and polyphenols were significantly higher in the moderately tolerant genotype (GM 090304) than in the sensitive genotype (Ca/H 631). These results suggest that proline, sugars and polyphenols act as main compatible solutes in cotton in order to maintain osmotic balance, to protect cellular macromolecules, to detoxify the cells, and to scavenge free radicals under water stress condition.  相似文献

The effects of 0, 30, 60, and 90 mM NaCl, and 0 and 5 mM CaCl2 on certain parameters of photosynthesis and growth in alfalfa (Medicago sativa L. cv. Ghara yonjeh) plants were studied. The increasing NaCl concentration in the Hoagland nutrient solution decreased the contents of chlorophylls and the net photosynthetic rate, and increased the rate of respiration (RD) and CO2 compensation concentration in the leaves of treated plants. The contents of carotenoids (Car) were not significantly affected. The addition of 5 mM CaCl2 enhanced the RD and increased the Car contents in treated leaves. With the NaCl concentration in the culture medium increasing, the dry matter production in both root and shoot decreased, as well as the relative growth rate (RGR), net assimilation rate (NAR), and leaf area ratio (LAR). The addition of CaCl2 caused a partial elimination of the NaCl effects on the root and shoot, RGR and NAR, and it decreased the LAR.  相似文献

本文研究了不同光质和类胡萝卜素专一抑制剂 norflurazon 对欧洲赤松(Pinussylvestris)子叶叶绿体色素形成的影响,所得结果如下:1.在远红光下,类胡萝卜素相对合成速率大于叶绿素的合成速率,而在红光下恰恰相反。2.在白光下,当类胡萝卜素合成受抑制时,叶绿素的合成速率也有所降低。但是叶绿素相对积累量比类胡萝卜素大得多,叶绿素与类胡萝卜素的分子比增大到10:1,说明这两种色素的合成与积累有相当大的独立性。3.把连续在远红光下类胡萝卜素合成受抑制(含量为正常幼苗的30%)的松苗,移入强白光下4小时后,其总叶绿素和类胡萝卜素都有增加,表明只要有少量类胡萝卜素存在,光对色素合成的促进仍大于光氧化破坏。  相似文献

The contents of chlorophylls, carotenoids, flavonoids and phytohormones (IAA, ABA and other inhibitors) were determined in green and albino seedlings of cotton (Gossypium hirsutum L.) and pea (Pisum sativum L.) The growth of green and albino seedlings during 1 –2 weeks was similar. The green and albino seedlings do not differ remarkably in phytohormonal content and in the flavonoid concentration. In the etiolated seedlings of green and albino forms the content of flavonoids was rather decreased.  相似文献

Pigment contents of chloroplasts and net photosynthetic rate were dramatically reduced in maize leaves suffering from iron deficiency. However, the reduction in photosynthesis was probably not caused by decreased contents of chlorophylls and carotenoids and by photon absorption; the primary limiting factor for photosynthesis may rather be the decrease of electron transport activity in photosystem 1. Iron-deficient leaves suffered serious acceptor-side photoinhibition, and more than 60 % of absorbed photons were dissipated, while less than 40 % was used in photochemical reaction. Thermal energy dissipation depending on xanthophyll cycle and D1 protein turnover was enhanced when acceptor-side photoinhibition occurred in iron-deficient maize leaves.  相似文献

A study was conducted to examine the physiological response of contrasting mung bean (Vigna radiata) genotypes viz., T 44 & MH–96–1 (tolerant) and Pusa Baisakhi & MH–1K–24 (sensitive) under waterlogging conditions. Plants were waterlogged at vegetative stage (30 days after sowing) for 3, 6 and 9 days. Waterlogging resulted in decreased leaf area, crop growth rate, root growth and nodules number, membrane stability index, photosynthesis rate, chlorophyll and carotenoid contents, flowering rate, pod setting, yield and altered dry matter partitioning. Sensitive genotypes showed large reductions in aforementioned physiological traits and slow recovery in photosynthesis rate. On the other hand, tolerant genotypes maintained higher photosynthetic rate, chlorophylls and carotenoids, growth rate, membrane stability and fast photosynthetic recovery under waterlogging. After 9 days of exposure to waterlogging, photosynthetic rate and yield losses in most sensitive genotype (MH-1K-24) were 83 and 85 %, respectively. On an average, photosynthetic loss at 3, 6 and 9 days of waterlogging was 43, 51, and 63 %, respectively, while grain yield loss was 20, 34 and 52 % respectively.  相似文献

Phycobiliproteins are light harvesting pigments in cryptophytes, cyanobacteria, and rhodophytes that allow these organisms to absorb light in the green and orange regions of the electromagnetic spectrum. Unlike chlorophylls and carotenoids, however, phycobilins are rarely quantified as part of routine photobiological studies because they require different extraction protocols. The objectives of this study were (1) to compare 10 existing methods to determine that with the highest extraction efficiency and (2) to determine the maximum time limit for the storage of phycobilins before degradation. Cells of the cryptophyte Rhodomonas salina and the cyanobacterium Synechococcus bacillaris were harvested either by centrifugation or filtration and then subjected to lyophilization, mechanical disruption, or freeze?Cthaw techniques. The extraction efficiency for pigments in cells collected on glass fiber filters was always <32?±?5% and thus always significantly lower than in samples harvested by centrifugation, which had extraction efficiencies of 53?±?6?C98?±?11%. Disruption of cells by freezing?Cthawing and sonication both resulted in significantly higher (ANOVA, p?<?0.01) extraction efficiencies than disruption with a tissue grinder. Storage of samples at ?80°C showed no significant pigment degradation over the course of 24 weeks.  相似文献

In viviparous hypocotyls ofRhizophora apiculata Blume the triacontanol treatment enhanced the growth of root and shoot (number of primary and secondary roots, length of roots, shoot height and biomass) protein and energy contents of leaves and roots,in vivo nitrate reductase activity, contents of chlorophylls and carotenoids in leaves as well as the amount of chlorophylls present in photosystems 1 and 2 and in the light harvesting complex of chloroplasts. These promotory effects were recorded at 40 and 80 mg m-3 triacontanol, but they decreased with increasing growth regulator concentration. Communicated by Z. ŠESTáK  相似文献
