Lipase production by Candida rugosa was carried out in submerged fermentation. Plackett-Burman statistical experimental design was applied to evaluate the fermentation medium components. The effect of twelve medium components was studied in sixteen experimental trials. Glucose, olive oil, peptone and FeCl3?6H2O were found to have more significance on lipase production by Candida rugosa. Maximum lipase activity of 3.8 u mL-1 was obtained at 50 h of fermentation period. The fermentation was carried out at optimized temperature of 30oC, initial pH of 6.8 and shaking speed of 120 r/min. Unstructured kinetic models were used to simulate the experimental data. Logistic model, Luedeking-Piret model and modified Luedeking-Piret model were found suitable to efficiently predict the cell mass, lipase production and glucose consumption respectively with high determination coefficient(R2). From the estimated values of the Luedeking-Piret kinetic model parameters, α and β, it was found that the lipase production by Candida rugosa is growth associated.  相似文献

A lipase-producing bacterium strain was isolated from soil and was identified as Pseudomonas sp.. Its lipase yield was improved 2.25-fold by combined beatment of UV irradiation and NTG. The lipase fermentation condition for the mutant strain was optimized with Plackett-Burman design and Response Surface Analysis(RSA), and the formula of the optimum medium suitable for industrial scale fermentation was thereby established. A maximum yield of 87.5 U/ ml was obtained.  相似文献

To improve the fermentation yield of xylanase by optimizing the fermentation conditions for strain Xw2, a Plackett-Burman design was used to evaluate the effects of eight variables on xylanase production by strain Xw2. The steepest ascent (descent) method was used to approach the optimal response surface experimental area. The optimal fermentation conditions were obtained by central composite design and response surface analysis. The results showed that the composition of the optimal fermentation medium was corn cob + 1.5% wheat bran (1:1), 0.04% MnSO4, 0.04% K2HPO4. 3H2O, and an inoculum size of 6% in 50 mL liquid volume (pH = 6.0). The optimal culture conditions were 28oc at 150 r/min for 54.23 h. The results of this study can serve as the basis for the industrial production and application of xylanase.  相似文献

A fed-batch fermentation integrated with filtration process was proceeded in a synthetic medium with the use of a hollow-fibre membrane filter. The activity of αA interferon reached 1.4 ×109u/L,which was increased 320% over that of a control process. The integrated process was then proceeded in the synthetic medium supplemented with yeast extract during the earlier stage. The addition of yeast extract not only reduced the accumulation of acetate, but also promoted the production of αA interferon. The maximum activity achieved 1.9 ×109 u/L during the fermentation, which was increased 480% over that of a normal process.  相似文献

Two kinds of micro-organism, Arthrobacter sp. AX86(1,4-dehdrogenator)and Absidia sp. A28(11α-hydroxylator) were used in this experiment. Two different fermentation techniques were performed to accomplish the multiple conversional reactions for producing 16β-methyl-11α, 17α, 21-trihydroxy-1,4-pregnadiene-3,20-dione (Ⅲ) from 16β-methyl-3β, 17α,21-trihydroxy-5α-pregnane-20-one-21-acetate(1) 1)To produce product(Ⅲ)by means of a two-step fermentation method which were independently performed first by Arthrobacter and next by Abslaia, and 2)the product was obtained by a sequential fermentation system of aforesaid two micro-organisms in a single fermentor without isolation of the intermediates from the mixture. Our results showed that in both fermentation systems high yield of product was obtained. However, according to the technical simplicity, shorter duration of fermentation cycle and efficient yield of product, the second method is better than the first one.  相似文献

Nattokinase is a fibrinolytic enzyme that is considered to be a promising agent for thrombosis therapy. In this study, nattokinase was purified from fermentation broth of a Bacillus subtilis strain by ammonium sulfate salting-out, gel filtration chromatography, and hydrophobic interaction chromatography with a purification fold of 5.2 and at a yield of 46.3%. The purified enzyme has molecular mass of 28 kDa and fibrinolytic activity of 4 580 U/mg. Since the concentration of nattokinase on fermentation broth was quite low, we cloned nattokinase gene from B. subtilis and expressed it in E. coli BL21 (DE3). Nattokinase was actively expressed in the recombinant strain. The yield of nattokinase was increased significantly, but the activity of the protein produced by recombinant strain was low.  相似文献

DPPH-TLC and DPPH-Microplate assays were used to determine the free radical scavenging activities of methanol extracts from the fermentation broth and the mycelia of Paecilomyces cicadae P3. The results revealed that the extracts had strong free radical scavenging activities. At the concentration of 5.0mg/mL the methanol extract from fermentation broth of P3 could decrease 74.86% of 0.4mg/mL DPPH radicals after incubating at 37℃ for 10 minutes. An anti-fungal TLC assay was applied to test antimicrobial metabolites of Paecilomyces cicadae P3. Meanwhile, an anti-Candida albicans Oxford Cup assay was made to confirm their anti-fungal activities. At the concentration of 5.0mg/mL the methanol extracts from fermentation broth and mycelia of P3 had significant anti-Candida albicans activities and the diameters of inhibition zone were 21.42mm and 11.23mm respectively.  相似文献

从烟草中筛选到几株内生菌进行防治根结线虫的盆栽试验,以研究其防治效果.结果表明,内生细菌发酵液的防治效果优于内生细菌菌体,内生放线菌则相反.内生菌普遍对植物具有促生作用,但内生菌菌体的促生效果优于内生菌的发酵液.H3号内生细菌对根结线虫防治效果最好,病情指数为4.16,防效为91.68%,明显优于对照,优于阿维菌素处理,H3号内生细菌处理的烟草形态指标明显优于阿维菌素处理.
Abstract:
Several tobacco endophyte strains were screened out, and their controlling effect against tobacco root-knot nematodes was studied with pot experiment. The fermentation broth of endophytic bacteria was more effective than the bacterial thallus, while that of endophytic actino-mycetes was less effective than the actinomycete fermentation broth. All test endophytes promoted tobacco growth, and the promotion effect of their thalli was better than that of their fermentation broths. Endophytic bacterial strain H3 was the best in controlling tobacco root-knot nematode dis-ease, with 91.68% controlling effect and a disease index of 4. 16, much better than the treat-ments CK and avermectin. Moreover, the morphological feature of tobacco plant treated with H3 was better than that treated with avermectin.  相似文献

Pleurotus citrinopileatus polysaccharides ware extracted from fermentation liquid by acetone under the conditions of different solvent concentration, extracting duration and temperature. The optimal design of extraction was analysed by using orthogonal test. The results showed that under the condition of acetone concentration of 75%, extraction duration of 1.5h, and temperature of 70℃, the highest yield of Pleurotus citrinopileatus polysaccharides was obtained.  相似文献

对表达重组人血小板生成素融合蛋白(rhTPO/GST)工程菌的发酵条件进行了较详细的研究,优化了影响工程菌发酵的条件,探讨了发酵条件对工程菌表达外源蛋白量的影响。结果发现采用复合培养基分阶段流加有机营养物,用异丙基硫代-B-D-半乳糖苷(IPTG)进行诱导,使rhTPO/GST融合蛋白的湿菌重达25g/L以上,表达水平约占菌体总蛋白的30%左右。在此基础上,建立了中试发酵生产工艺流程。  相似文献

目的:利用基因工程方法对一种蛇毒锯鳞蝰素蛋白的发酵纯化工艺进行优化,以提高目的蛋白的产量和纯度。方法:对工程菌进行发酵培养并诱导表达,研究不同的培养基、不同补料方式、溶解氧浓度、培养和诱导时间对工程菌产量和目的蛋白表达量的影响,利用几丁质亲和层析纯化Ecs融合蛋白,通过合适温度和pH裂解融合蛋白得到Ecs纯品,并鉴定和检测Ecs活性。结果:经过高密度发酵优化后,菌体湿重可达110g/L,目的蛋白表达量约占总蛋白的40%;亲和层析纯化后,得到Ecs单体,得率为68mg/L发酵液。生物学活性分析显示,重组Ecs能有效抑制血小板的聚集,其活性与天然Ecs相似。结论:通过发酵和纯化工艺优化,大大提高了目的蛋白产量,为进一步规模化研究和生产奠定了基础。  相似文献

研究了大白菜乳酸菌乳链球菌DM 2 2和植物乳杆菌UM 2 2混合发酵中的生长和产酸以及环境因子的影响 ,并对混菌发酵的风味物质作了分析。结果表明 :混菌发酵中 ,DM2 2在发酵前期生长迅速 ,是优势菌群 ,而UM2 2在发酵后期占主导 ;发酵温度、发酵剂组成以及发酵液盐浓度都会显著影响混合发酵中菌的产酸代谢 ;发酵风味物质与单菌发酵区别明显。  相似文献

混菌发酵改良棉粕蛋白工艺及协同作用研究   总被引:1,自引:0,他引:1  
利用筛选到的Aspergillus niger P1和Bacillus subtilis H1混菌发酵改良棉粕蛋白,以提高其利用率。结果表明,混菌发酵的最佳发酵条件为:棉粕 40 g,硫酸铵0.2%,麸皮15%,含水量50%,接种量 15%,同时接菌且两菌接菌比例(A.niger P1 ∶ B.subtilis H1)为2 ∶ 1,发酵温度30 ℃,pH约为6.0,发酵时间 60 h。在此条件下发酵后棉粕小肽含量可提高至18.36%,平均肽链长度可降至4.23,体外消化率可提高至88.59%,显著提高了改良棉粕蛋白的效果。在相同发酵条件下比较单、混菌发酵过程的各营养指标发现:A.niger P1可分泌丰富的酸性蛋白酶,B.subtilis H1可分泌丰富的中、碱性蛋白酶,混菌发酵各种蛋白酶的活性显著提高。混菌发酵将大多数大于10个氨基酸组成的多肽降解为1~3个氨基酸组成的小肽,降解效果明显优于单菌发酵。两菌株具有很好的"协同性",可充分利用棉粕基质,协同改良棉粕蛋白。  相似文献

考察直流电磁场对类胡萝卜素产生前红酵母RG-98的生长及代谢的影响。结果表明,不同的磁场强度、处理时间及处理方式会产生不同的生物效应。将液体种子和发酵培养基一起置于0.10T电磁场处理2h,能使色素的发酵产量提高25%。比较经磁场处理与未经处理的发酵过程,发现磁场处理提高了红酵母对糖的代谢速率,并且在发酵后期促进生物量和色素含量持续地增加。  相似文献

在浙江省安吉县采集了相邻的天然灌木林和板栗林土壤,分析土壤水溶性碳(WSOC)、微生物生物量碳(MBC)、易氧化碳(ROC)、水溶性有机氮(WSON)和微生物生物量氮(MBN),并利用核磁共振方法分析土壤总有机碳的波谱特征,研究天然灌木林改造成板栗林对土壤碳库和氮库的影响.结果表明: 天然灌木林改造成板栗林后,土壤中的碱解氮、有效磷和速效钾显著增加,而WSOC、MBC、ROC、WSON和MBN显著下降.天然灌木林和板栗林土壤有机碳以烷基碳和烷氧碳为主.天然灌木林改造成板栗林后,土壤有机碳中的烷氧碳和羰基碳比例显著下降,而烷基碳和芳香碳比例以及A/O-A值和芳香度均显著增加.天然灌木林改造成板栗林并长期集约经营后,土壤活性碳库和氮库含量均显著下降,而土壤碳库的稳定性显著增加.  相似文献
