Conidiation and lytic enzyme production by Trichoderma viride at different solids concentration of pre-treated municipal wastewater sludge was examined in a 15-L fermenter. The maximum conidia concentration (5.94 × 107 CFU mL−1 at 96 h) was obtained at 30 g L−1 suspended solids. The maximum lytic enzyme activities were achieved around 12–30 h of fermentation. Bioassay against a fungal phytopathogen, Fusarium sp. showed maximum activity in the sample drawn around 96 h of fermentation at 30 g L−1 suspended solids concentration. Entomotoxicity against spruce budworm larvae showed maximum value ≈17290 SBU μL−1 at 30 g L−1 suspended solids concentration at the end of fermentation (96 h). Plant bioassay showed dual action of T. viride, i.e., disease prevention and growth promotion. The rheological analyses of fermentation sludges showed the pseudoplastic behaviour. In order to maintain required dissolved oxygen concentration ≥30%, the agitation and aeration requirements significantly increased at 35 g L−1 compared to 30 and 25 g L−1. The oxygen uptake rate and volumetric oxygen mass transfer coefficient, kLa at 35 g L−1 did not increase in comparison to 30 g L−1 due to rheological complexity of the broth during fermentation. Thus, the successful fermentation operation of the biocontrol fungus T. viride is a rational indication of its potential for mass-scale production for agriculture and forest sector as a biocontrol agent.  相似文献

Because of its novel bioactive properties the production of gymnodimine for use as a pharmaceutical precursor has aroused interest. The dinoflagellate, Karenia selliformis produces gymnodimine when grown in bulk culture using GP + selenium medium but the growth rates (μ) and levels of gymnodimine are low (μ, 0.05 days−1; gymnodimine 250 μg L−1 max). We describe the effects of organic acid additions (acetate, glycolate, alanine and glutamate additions and combinations of these) in enhancing growth and gymnodimine production in axenic cultures. The most effective organic acid combinations in decreasing order were: glycolate/alanine > acetate > glycolate. Glycolate/alanine optimised gymnodimine production by prolonging growth (maximum cell yield, 1.76 × 105 cells mL−1; gymnodimine, 1260 μg L−1; growth rate (μ), 0.2 days−1) compared to the control (growth maximum cell yield, 7.8 × 104 cells mL−1; gymnodimine, 780 μg L−1; μ, 0.17 days−1). Acetate enhanced gymnodimine by stimulating growth rate (μ, 0.23 days−1) and the large concentration of gymnodimine per cell (16 pg cell−1 cf. 9.8 pg cell−1 for the control) suggests a role for this compound in gymnodimine biosynthesis. Amending culture media with Mn2+ additions resulted in slightly decreased growth in control cultures and increased the gymnodimine while in glycolate/alanine cultures growth was stimulated but gymnodimine production decreased. The results suggest that the organic acid can enhance gymnodimine production by either enhancing growth maximum or the biosynthetic pathway.  相似文献

A bacterial flavin-containing monooxygenase (FMO) gene was cloned from Methylophaga aminisulfidivorans MPT, and a plasmid pBlue 2.0 was constructed to express the bacterial fmo gene in E. coli. To increase the production of bio-indigo, upstream sequence size of fmo gene was optimized and response surface methodology was used. The pBlue 1.7 plasmid (1686 bp) was prepared by the deletion of upstream sequence of pBlue 2.0. The recombinant E. coli harboring the pBlue 1.7 plasmid produced 662 mg l−1 of bio-indigo in tryptophan medium after 24 h of cultivation in flask. The production of bio-indigo was optimized using a response surface methodology with a 2n central composite design. The optimal combination of media constituents for the maximum production of bio-indigo was determined as tryptophan 2.4 g l−1, yeast extract 4.5 g l−1 and sodium chloride 11.4 g l−1. In addition, the optimum culture temperature and pH were 30 °C and pH 7.0, respectively. Under the optimized conditions mentioned above, the recombinant E. coli harboring pBlue 1.7 plasmid produced 920 mg of bio-indigo per liter in optimum tryptophan medium after 24 h of cultivation in fermentor. The combination of truncated insert sizes and culture optimization resulted in a 575% increase in the production of bio-indigo.  相似文献

The Late Oligocene (25.8 Ma) Enspel Fossillagerstätte in Westerwald, Germany, contains a comprehensive fossil ecosystem preserved with specimens retaining morphological detail and a concentration of organic material. Stable carbon and nitrogen isotope analyses were used to examine the lacustrine ecosystem preserved in one stratigraphic horizon. These data suggest the presence of several trophic levels, including primary producers (diatoms and higher plants), primary consumers (e.g., tadpoles and insects), and secondary consumers (e.g., the fish species Paleorutilus enspelensis). Terrigenous and aquatic plants were associated with the lowest δ13C and δ15N values (mean plant = − 26.28‰ ± 0.45, 3.18‰ ± 1.04), primary consumers such as flies are one trophic level higher, and carnivores such as fish are yet another level higher. The δ15N values for P. enspelensis also showed enrichment in 15N with increasing body length, implying a shift in diet or feeding strategy with size. P. enspelensis and tadpole (Pelobates decheni) samples showed intraorganism fractionation between ‘muscle’ and ‘bone’ tissues. Stable carbon and nitrogen isotope data from the measurement of components (shale, leaves and seeds) common to a number of different stratigraphic horizons showed significant variation between horizons. A number of the features of the stable isotopic data are similar to those relationships seen in modern ecosystems and therefore suggest that stable isotope analyses can contribute to understanding ancient ecosystems.  相似文献

Isarfelin, a peptide with inhibitory activity on mycelial growth in Rhizoctonia solani and Sclerotinia sclerotiorum and insecticidal activity toward Leucania separata, was isolated from the mycelia of Isaria felina. The IC50 value of its antifungal activity against R. solani was 3.1 μg mL−1. However, it was devoid of activity toward several bacterial species including Bacillus subtilis, E. coli and Staphylococcus aureus. The isolation procedure involved ethanol extraction, adsorption on YPR II macropore adsorption resin, ethyl acetate extraction, petroleum ether precipitation and recrystallization from ethyl acetate.  相似文献

In this work we present the synthesis, structural and spectroscopic characterisation of a series of cobalt(II) and zinc(II) coordination compounds with benzimidazole (bz) and its 2-benzimidazole derivatives: 2-aminobenzimidazole (2ab), albendazole (abz) and tris(2-benzimidazolylmethyl)amine (ntb). The compounds were evaluated for their in vitro antimicrobial activity against Staphylococcus aureus, Micrococcus luteus, Salmonella typhi, Pseudomonas aeruginosa, Escherichia coli and Proteus vulgaris. Their cytotoxic activity was also evaluated using human cancer lines, HeLa, HCT-15 and SKLU-1. The halide tetrahedral compounds [Co(bz)2Br2] 3, [Zn(2ab)2Cl2] · 0.5H2O 11, [Co(abz)Cl2(H2O)] · 3H2O 14, [Co(abz)Br2(H2O)] 15, [Zn(abz)Cl2(H2O)] · 3H2O 17 and [Zn(abz)Br2(H2O)] · H2O 18 displayed similar minimal inhibition concentration (MIC) values against Micrococcus luteus and Escherichia coli, comparable to those of amoxicillin and chloramphenicol. Additionally, 11 showed a wide range of activity towards Gram(+) and Gram(−) microorganisms. The tetradentate ntb and its trigonal bipyramidal cobalt(II) and zinc(II) compounds were active, regardless of the anion present in the complex. Compound [Co(abz)Cl2(H2O)] · 3H2O 14 showed promising activity in HeLa cells, while [Co(ntb)Br]Br · H2O 21 inhibited Hela and HCT-15 cell lines.  相似文献

The alternating current (ACt) polarographic behavior of lansoprazole (LNS) and omeprazole (OMP) was studied in Britton Robinson buffers (BRb) over the pH range 4.1–11.5. In BRb of pH 9.6 and 10.5, well-defined ACt peaks were obtained for both LNS and OMP, respectively. The current–concentration plots were rectilinear over the ranges of 0.4–20 µg mL− 1 and 0.2–10 µg mL− 1 for LNS and OMP respectively. The minimum detection limits (S/N = 2) were 0.02 µg mL− 1 (5.4 × 10− 8 M) and 0.01 µg mL− 1 (2.9 × 10− 8 M) for LNS and OMP, respectively. The proposed method was successfully applied to the analysis of the two drugs in their commercial capsules. The average percent recoveries were favorably compared to those obtained by reference methods. Co-administered drugs such as naproxen and methotrexate did not interfere with the proposed method. The proposed method was further extended to the in-vitro determination of the lansoprazole in spiked plasma, the percentage recoveries was 98.47 ± 1.29 (n = 4). The pathway for the electrode reaction for both drugs involved reduction of the sulphonyl group into the corresponding thiol group at the Dropping Mercury Electrode. The advantages of the method were time saving and more sensitive than the other published voltammetric method. Yet The present study is the first report on the use of alterating current polarography (ACt) in this respect.  相似文献

Cross-linked waxy maize (CWM) starch dispersions (STDs) of concentration 50 g kg−1 were heated in sucrose solutions containing 0–600 g kg−1 (g sucrose/kg dispersion) at 85 °C at low shear and in intermittently agitated cans at 110 °C. The STDs heated in 0–300 g kg−1 sucrose exhibited antithixotropic behavior, while those heated in 400–600 g kg−1 sucrose exhibited thixotropic behavior. The mean starch granule diameter of the starch dispersions did not show strong dependence on sucrose concentration. The dispersions, especially those with high sucrose concentrations and heated at 110 °C, exhibited G′ versus frequency (ω) profiles of gels. The STDs exhibited first normal stress differences that increased in magnitude with the concentration of sucrose. Values of the first normal stress coefficient of canned dispersions calculated from dynamic rheological data plotted against ω and experimental values plotted against shear rate of some of the STDs overlapped.  相似文献

Metal concentrations and population parameters of the seagrass Halodule wrightii were determined at three locations at Rio de Janeiro State, Brazil. The possible increase of metal availability in one of these areas, Sepetiba Bay, as a result of dredging of contaminated bottom sediments which ocurred, was evaluated by analyses of Al, Cd, Cr, Cu, Fe, Ni, Pb and Zn in root, rhizome and shoots. In addition, analyses were carried out in H. wrightii populations from non-contaminated areas located at northwestern (Cabo Frio) and southeastern (Angra do Reis) regions of Rio de Janeiro State. Concurrently, abundance and density data of the seagrass populations were obtained. It was found that concentration from Sepetiba Bay samples up to 1.6 ± 0.4 μg g−1 of Cd, 12 ± 1.0 μg g−1 of Cr, 27 ± 2.4 μg g−1 of Pb, 291 ± 47 μg g−1 of Mn, 128 ± 23 μg g−1 of Zn were significantly higher than that from two other collection sites. An increase in Cd and Zn concentration was observed in H. wrightii from Sepetiba Bay indicating that metal mobilization from contaminated sediments through dredging activities were, at least in part, transferred to the biotic compartment via accumulation by the seagrass. The populations of seagrass within the region demonstrated quite substantial changes in biomass data but not in shoot or rhizome density during the study. Such changes in biomass are to be expected, as these dynamics are typical of the small, isolated monospecific populations of H. wrightii along the Rio de Janeiro coast.  相似文献

Colonies of the seagrass Halophila ovalis are found growing adjacent to coral Acropora sp. and Seriatopora hystrix in a submarine hot spring (at 15.7 m depth, 28.6°C) at the north coast of Taketomi Island, near the southern tip of Japan. Halophila plants grow in sea water containing sulphide 930 μg S ml−1 and on the substratum with fine precipitates of the submarine hot spring which have sulphide content up to 5400 μg S g−1 DW. The accumulated sulphide concentration reaches as high as 8400 μg S g−1 DW in under ground tissues and 5700 μg S g−1 DW in above-ground tissues, respectively. It is suggested that, not the sulphide concentration but light and possibly water temperature are the limiting factors for the Halophila colonization in the submarine hot spring.  相似文献

The majority of sediment dweller foraminifera are deposit feeders. They use their pseudopodia to gather sediment with associated algae, organic detritus and bacteria. Uptake of bacteria by foraminifera have been observed but rarely quantified. We measured uptake of bacteria by the common foraminifera Ammonia tepida using 15N pre-enriched bacteria as tracers. In intertidal flats, seasonal, tidal and circadian cycles induce strong variations in environmental parameters. Grazing experiments were performed in order to measure effects of abiotic (temperature, salinity and irradiance) and biotic (bacterial and algal abundances) factors on uptake rates of bacteria. In mean conditions, A. tepida grazed 78 pgC ind− 1 h− 1 during the first eight hours of incubation, after which this uptake rate decreased. Uptake of bacteria was optimal at 30 °C, decreased with salinity and was unaffected by light. Above 7 × 108 bacteria ml wt sed− 1, uptake of bacteria remained unchanged when bacterial abundance increased. Algal abundance strongly affected algal uptake but did not affect uptake of bacteria. As uptake of bacteria represented 8 to 19% of microbes (algae plus bacteria) uptake, Ammonia seemed to be mainly dependant on algal resource.  相似文献

The rate of electron transfer between reduced cytochrome ƒ and plastocyanin (both purified from parsley) has been measured as k = 3.6 · 107 M−1 · s−1, at 298 °K and pH 7.0, with activation parameters ΔH = 44 kJ · mole−1 and ΔS = +46 J · mole−1 · °K−1. Replacement of cytochrome ƒ with red algal cytochrome c-553, Pseudomonas cytochrome c-551 and mammalian cytochrome c gave rates at least 30 times slower: k = 5 · 105, 7.5 · 105 and 1.0 · 106 M−1 · s−1, respectively.

Similar measurements made with azurin instead of plastocyanin gave k = 6 · 106 and approx. 2 · 107 M−1 · s−1 for reaction of reduced azurin with cytochrome ƒ and algal cytochrome respectively.

Rate constants of 115 and 80 M−1 · s−1 were found for reduction of plastocyanin by ascorbate and hydroquinone at 298 °K and pH 7.0. The rate constants for the oxidation of plastocyanin, cytochrome ƒ, Pseudomonas cytochrome c-551 and red algal cytochrome c-553 by ferricyanide were found to be between 3 · 104 and 8 · 104 M−1 · s−1.

The results are discussed in relation to photosynthetic electron transport.  相似文献

A novel bloom of Alexandrium minutum occurred in an inner basin of the Cape Town harbour from November 2003 to February 2004. Cellular concentrations reached a maximum of 1.4 × 108 cells l−1 during the mid-December period with corresponding chlorophyll a concentrations of 243 mg m−3. Primary productivity measurements conducted during the latter part of the bloom revealed a maximum assimilation number of 11.17 mg C mg Chl a−1 h−1 during the middle of the day. Productivity during this post-peak period was sustained largely by the reduced nitrogen species NH4 and urea (96%) as measured using 15N tracer techniques. The large subunit ribosomal DNA sequence of A. minutum isolates from Cape Town harbour was identical to conspecifics collected in Western Europe and in Australia. The composition of tetrahydropurine neurotoxins associated with paralytic shellfish poisoning (PSP) was limited to gonyautoxins (GTX1-GTX4). This profile combined with evidence of a low toxin cell quota (1.5 fmol GTX cell−1) supports a close association of this taxon with other members of the A. minutum species complex, particularly from Europe. Toxin analysis from black mussels collected during this bloom indicated that the accumulated PSP toxins originated from A. minutum and not from Alexandrium catenella as is most often the case along the South African coast.  相似文献

15N NMR studies of the interaction of 15N cyanide ion with gold(I)-thiomalate (Autm) and gold(I)-thioglucose (Autg) have been carried out at pH* 7.40. The chemical shifts of the two 15N ions containing species Au(C15N)2 and RS-Au-C15N (where RS = tm or tg) were identified at 265.94 and 260.30 ppm, respectively. From the broadened 15N NMR signals, approximate life times of the RS-Au-CN species were calculated.  相似文献

以东北森林两种典型的阔叶树种风力传播种子——花曲柳和色木槭种子为研究对象,通过室内15N尿素浸泡试验和温室盆栽试验,设置4个浓度(0、0.05、0.1和0.2 g·L-1)、3个浸泡时间(4、8和12 d)与4个叶期(2、4、6和8叶)处理,研究种子浸泡浓度、浸泡时间和幼苗叶期对种子和幼苗15N富集的影响.结果表明: 浸泡浓度和浸泡时间对两树种种子δ15N值均有显著的正反馈作用,高浓度和长时间(0.2 g·L-1+12 d)更有利于种子15N总量的富集,花曲柳和色木槭种子15N同位素最大富集倍数的浸泡浓度和浸泡时间组合分别为0.1 g·L-1+(4、8 d)和0.05 g·L-1+(4、8 d);δ15N值稀释率随幼苗株高的增加先急剧减少(2~6叶)后趋于稳定,幼苗从8叶开始叶片15N总量降低,表明6叶幼苗更适合追踪幼苗的来源;幼苗叶片δ15N值与种子浸泡浓度、浸泡时间和种子的δ15N值呈显著正相关.花曲柳和色木槭种子及幼苗均能成功富集到15N信号,采用0.1 g·L-1+8 d+6叶组合最适合追踪花曲柳和色木槭种子和幼苗.  相似文献

The potential for nutrient load (30, 100 and 350 g N m−2 per year) to alter plant performance under saline conditions (control, 4.5, 9 and 13 dS m−1) was examined in the sedge Bolboschoenus medianus. Relative growth rates (RGR) across nutrient loadings ranged from 30.2 to 41.8 mg g−1 per day in controls and were reduced to 20.9–28.5 mg g−1 per day by salinities of 13 dS m−1. Whilst higher nutrient loads generally increased RGR, the response was smaller at higher salinities. Responses to salinity and nutrient load were specific. Nutrient load increased the RGR via increases in the leaf area ratio (LAR). The LAR ranged from 1.9 to 2.1 m2 kg−1 across salinity treatments at 30 g N m−2 per year, and increased to 2.5–2.8 m2 kg−1 at 350 g N m−2 per year. Salinity reduced the RGR via a reduction in the net assimilation rate (NAR). The NAR in control plants ranged from 14.7 to 16 g m−2 per day across nutrient loadings and decreased to 11–12 g m−2 per day at 13 dS m−1. Carbon isotope discrimination of leaves decreased by 2–3‰ in response to 13 dS m−1 at the lower nutrient loadings. A prominent response of B. medianus to salinity was a change in biomass allocation from culms to tubers. In contrast, the response to nutrient load was characterised by a shift in biomass allocation from roots to leaves.  相似文献

A genetically engineered Pichia pastoris FPHY34 strain containing a 1.3 kb thermostable phytase gene (fphy) evolved by DNA shuffling was constructed and screened. Expression and purification conditions for the recombinant phytase were developed in this study. The effect of Pi on recombinant phytase expression and cell growth of P. pastoris FPHY34 was tested in shake flask culture. Optimization of carbon sources for cell growth and methanol feeding strategies for phytase expression in P. pastoris FPHY34 was carried out in a 50-L fermenter by fed-batch fermentation. The purification of phytase was investigated by micro-filtration and ultra-filtration followed by desalting, ion-exchange chromatography, and gel filtration in the ÄKTA system. It showed that the optimum inorganic phosphorus is 13.6 g L−1 and that glucose can be used as a substrate for P. pastoris cell growth instead of glycerol; the biomass yield of glycerol (YX/S) is slightly higher than that of glucose. Different profiles of lag phase and respiratory quotient (RQ) displayed between glucose and glycerol as the sole carbon source. The maximum phytase activity in per millimetre reached 2508 U mL−1 at a methanol feed rate of 3.0 mL L−1 h−1 after 80 h period of induction. A purification factor of 41.1 with a 32% yield was achieved after chromatographic purification. The specific enzyme activity was 80 U mg−1 and 3281 U mg−1 in that supernatant fraction and after gel filtration purification, respectively. The strain P. pastoris FPHY34 showed a promising application in phytase industrial production.  相似文献

Floating Pennywort (Hydrocotyle ranunculoides L. fil.) is a worldwide distributed aquatic plant. The species is native to North America and quite common also in Central and South America. In Europe, Japan and Australia it is known as an alien plant, sometimes causing serious problems for affected ecosystems and human use of water bodies. Starting from Western Europe with an eastwards directed spread, Floating Pennywort was recorded in Germany in 2004 for the first time. Since then, the species spread out and got established in western parts of Central Europe. For a definite prediction of the potential of a further spread, data about biology, in particular growth and photosynthesis are needed. Here, regeneration capacity, growth at different nutrient availabilities and photosynthesis of H. ranunculoides were investigated. In addition biomass samples were taken in the field. Results show an enormous regeneration capacity (e.g., by forming new shoots from small shoot fragments), increasing growth rates under increasing nutrient availability and a maximum increase of biomass reaching 0.132±0.008 g g−1 dw d−1. Dense populations of H. ranunculoides growing in ponds and oxbows were found at high nutrient content of the substrate, the biomass reaching there up to 532.4±14.2 g dw m−2. Gas exchange analysis showed a physiological optimum of H. ranunculoides CO2 uptake at temperatures between 25 and 35 °C and high photon flux densities (PPFD) above 800 μmol photons m−2 s−1. In comparison, native Hydrocotyle vulgaris showed an optimum of net photosynthesis at 20–30 °C and a light saturation of CO2 gas exchange at 350 μmol photons m−2 s−1.  相似文献

A group of 34 chromium-resistant bacteria were isolated from naturally occurring chromium percolated serpentine soil of Andaman (India). These isolates displayed different degrees of chromate reduction under aerobic conditions. One of the 34 isolates identified as Bacillus sphaericus was tolerant to 800 mg l−1 Cr(VI) and reduced >80% Cr(VI) during growth. In Vogel Bonner broth, B. sphaericus cells (1010 cells ml−1) reduced 62% of 20 mg l−1 of Cr(VI) in 48 h with concomitant discoloring of yellow medium to white one. Reduction of chromate was pronounced by the addition of glucose and yeast extract as electron donors. In the presence of 4.0 g l−1 of glucose, 20 mg l−1 of Cr(VI) was reduced to 2.45 mg l−1 after 96 h of incubation. Optimum pH and temperature for reduction were 6.0 and 25 °C, respectively. Increase in cell density and initial Cr(VI) concentration increased chromate reduction but was inhibited by metal ions like, Ni2+, Co2+, Cd2+ and Pb2+. Experiments with cell-free extracts indicated that the soluble fraction of the cell was responsible for aerobic reduction of Cr(VI) by this organism.  相似文献
