本文研究碳源、氮源、温度、湿度、pH值和光照等对蝉拟青霉LB菌株生长、产孢和孢子萌发的影响.结果表明,适合该菌株菌落生长和产孢的最佳碳源是可溶性淀粉和蔗糖,最佳氮源为蛋白胨;菌丝生长和孢子萌发的最适温度范围是25℃~27℃,产生分生孢子的最适温度是25℃;分生孢子萌发所需湿度范围是RH 90%~100%,当RH低于90%时很难萌发;在pH值4~10的范围内该菌能生长和产孢,菌丝生长最适pH为6,产生分生孢子和孢子萌发最适pH范围为6-7;光照处理对该菌产孢有一定的影响;分生孢子的致死条件为55℃ 10min.生物学特性显示,蝉拟青霉LB菌株是一株对营养要求不高、对环境适应能力较强的昆虫病原真菌.  相似文献

本文报道诱导茄链格孢菌(Alternaria solani)分生孢子形成的一种新技术。生长在马铃薯-葡萄糖-琼脂(PDA)上两天的茄链格孢菌琼脂块移接到玉米培养基上,置于日光灯下照射,诱发分生孢子梗生长。然后,再放在18℃下黑暗培养。12小时后,在菌丝块表面有大量的分生孢子形成。成熟的茄链格孢菌分生孢子用蒸馏水洗脱。  相似文献

短柄链蠕孢(Dendryphion nanum)为暗色丝孢菌,其分生孢子棒状,5~20个隔膜,暗褐色,串生。试验通过玻片培养观察研究了该菌分生孢子的产生方式及发育类型。结果表明:该菌的分生孢子是由产孢细胞的顶端向外突起并生长膨大而形成,因而属于全壁芽生式产孢方式。  相似文献

观察了蝉拟青霉的无性世代,在24℃下载片培养结果,分生孢子经8hr萌发,24hr普遍形成菌丝,36hr出现产孢结构和产生次代分生孢子。该菌生长合适温度24—26℃,分生孢子萌芽要求相对湿度在90%以上。pH4—12范围均见生长,但以5—6为佳。对10种碳源和9种无机氮源利用检测结果,用葡萄糖作碳源孢子产量高,用果糖作碳源菌丝体产量高。不利用菊糖、L-山梨糖、L-鼠李糖。对KNO_3利用佳,但不能利用NaNO_2,和硫脲。该菌能较强抗紫外辐射。  相似文献

西瓜炭疽病菌圆刺盘孢分生孢子产生条件的研究   总被引:1,自引:0,他引:1  
为研究圆刺盘孢Colletotrichum orbiculare的分生孢子产生条件,通过改变培养基的成分、理化性状及培养条件等方法,进行了诱导产孢试验,并对所产生的分生孢子量进行测定。结果表明,C.orbiculare在西瓜茎叶煎汁培养基上25℃恒温黑暗培养30d可产生大量分生孢子,产孢量高达7.15×109个/m L。在显微镜下观察发现,病菌在西瓜茎叶煎汁培养基上和在西瓜茎秆病斑上产生的分生孢子在形态上存在差异,前者产生的分生孢子稍短和略宽,两端钝圆;而后者产生的分生孢子相对长而细,两端较尖。试验证明寄主西瓜茎叶成分对病菌分生孢子的产生具有明显促进作用。  相似文献

芽生孢子是许多真菌通过出芽方式产生的无性孢子,在液体发酵中主要以这种形式大量繁殖。真菌杀虫剂活性成分分生孢子的大量生产使用液体发酵得到的芽生孢子作为接种物,另外,它也是当今虫生真菌遗传转化的重要受体,因而研究虫生真菌芽生孢子的形态和发生特点具有重要意义。本研究分别对液体发酵中的球孢白僵菌、粉棒束孢、蝉棒束孢、环链棒束孢、玫烟色棒束孢、细脚棒束孢、斜链棒束孢、金龟子绿僵菌、蝗绿僵菌和蜡蚧霉等10种常见虫生真菌芽生孢子的形成过程进行显微观察,了解其分生孢子产生过程的异同。结果表明,芽生孢子的产生方式有两种类型:1)蝉棒束孢在其整个生活史中以菌丝生长为主,芽生孢子产生的数量很少。2)其他各种真菌芽生孢子产生方式相似,在菌丝体形成后就开始大量以菌丝出芽或缢缩产生芽生孢子,接着还可通过芽生孢子的出芽或缢缩断裂产生新的芽生孢子。芽生孢子的产生分为3个时期:初期先由菌丝形成芽生孢子;指数期芽生孢子大量增殖,菌丝和芽生孢子都可产生芽生孢子;后期以芽生孢子产新芽生孢子为主要方式。  相似文献

分生孢子是中国被毛孢(H sinensis)生活史中极为重要的一个环节,其在冬虫夏草侵染和形成过程中有着十分关键的作用.本实验以中国被毛孢纯培养菌种为实验材料,采用显微镜观察了其菌丝体经营养生长产分生孢子过程中显微结构变化,结果表明:中国被毛孢在固体培养条件下,可以形成分生孢子,其形态和产生方式与微循环产孢过程中形成的分生孢子一致,同时,在产孢过程中,还存在瓶梗结构纵裂形成对生分生孢子的现象.  相似文献

利用LC-MS对不同培养时间蝉棒束孢菌丝体代谢物进行检测,结合多变量统计分析的主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)鉴定差异化合物。PCA和OPLS-DA表明蝉棒束孢菌丝体中的代谢物随培养时间不同有显著变化。阴、阳离子模式下我们都可以把蝉棒束孢代谢物变化分为前期、中期和后期3个时期。培养前期产生较多的细胞松弛素E、肌醇、γ-氨基丁酸、乙酰肉碱、亚油酸等与侵染宿主、早期信号传导和氧化供能有关的物质;中期产生较多的甘露醇、海藻糖、亚麻酸、柠檬酸、丙酮酸、7,8-二羟基油酸等与抗逆和氧化供能有关的物质,此外还有两种抗生素红酵母红素、δ-氨基-γ-酮戊酸达到了最大值;培养后期产生较多的脯氨酸、2,4-二氨基丁酸、3′-deoxy-hanasanagin、ulvaline、赤霉素、烟酰胺等与抗逆、抗氧化、产孢、细胞保护有关的物质。首次发现烟酰胺可能与蝉棒束孢从营养生长转化为生殖生长及产孢有关;同时赤霉素、ulvaline和δ-氨基-γ-酮戊酸可能与蝉棒束孢产孢或孢子成熟有关,尤其值得注意的是蝉棒束孢在培养后期高速合成和大量积累赤霉素,值得深入研究。根据蝉棒束孢的代谢特点,作为药品及保健品开发蝉棒束孢菌丝产品时,培养时间宜在7–11d。  相似文献

【目的】揭示蝉棒束孢居群形态性状的变异式样。【方法】通过对15个居群,75个蝉棒束孢菌株17个显微形态性状的观察和测量,采用SAS 8.1软件对蝉棒束孢显微形态性状数据进行描述性、巢式方差和Q聚类分析,获得蝉棒束孢显微形态变异数据。【结果】蝉棒束孢具有大小两型分生孢子,产孢细胞的形态可分为葫芦状和瓶状2种类型;蝉棒束孢在PDA培养基中易形成数量众多的厚垣孢子,菌丝间多能形成融合菌丝,并可观察到少量的融合菌丝在厚垣孢子与菌丝间、产孢细胞间形成。描述性分析表明,蝉棒束孢居群17个形态性状的变异系数(CV)在13.07%–104.09%之间,在物种水平上表现出丰富的变异;巢式方差分析表明,蝉棒束孢显微形态性状在居群间平均方差分量占总变异的11.29%,居群内个体间平均方差分量占总变异的15.27%,菌株内观察值间平均方差分量占总变异的73.44%。【结论】蝉棒束孢菌株形态性状变异丰富,是形态性状变异的主要来源,蝉棒束孢居群显微形态性状不存在明显的地理相关性。  相似文献

从云南高原地区的竹叶病理组织上分离到1株可产生红棕色素的丝状真菌ZL07-8。通过对分生孢子梗、分生孢子、喙等的形态学研究及对ITS和5.8S rRNA序列的分子系统学分析,发现此菌为链格孢属小孢子种群成员。然而该菌具有区别于已知种的特征,如分生孢子梗常单生,少数成簇,无分枝;分生孢子大多单生,较少链生,孢身较大;寄主为华西箭竹,故立为竹链格孢新种。干制培养物ZL07-8D保藏于山东理工大学生命科学学院实验室。  相似文献

The susceptibility of Botrytis cinerea to copper sulphate in liquid media increased when the volume, and therefore the depth, of the medium in culture bottles exceeded certain values; when the volume was 40 ml. the maximum concentration allowing growth was 300 p.p.m.
By growing mycelium in media containing progressively higher concentrations of copper sulphate a strain was produced which grew at a concentration of 750 p.p.m.
In high concentrations of copper sulphate growth always started at the edge of the liquid, and inocula grew only if they were placed in this position.
In germination tests spores from the resistant strain were more resistant to copper sulphate than were spores of the parent strain.
The resistance of mycelium, and to a lesser extent of spores, was retained after growth of the resistant strain for six months in fungicide-free media.
Spore and mycelial inocula grew in much higher concentrations of copper sulphate when nutrient media were solidified with agar.
The strain resistant in liquid media was no more resistant than the parent strain on agar media.
The resistance of the fungus was not increased after growth for long periods on agar containing high concentrations of copper sulphate. The resistance of the strain resistant in liquid media was not lost after growth on agar media for 3 months.
Attempts to produce strains more resistant than the parent to mercuric chloride were unsuccessful.
The results obtained with phenyl-mercuric acetate were essentially similar to those obtained with copper sulphate, but relatively much more resistant strains were produced.  相似文献

The fungus grew as hollow mycelial balls surrounding individual hydrocarbon droplets on n-hexadecane and as solid mycelial balls on peptone. A dense layer of fungal mycelium that showed irregular forms, fusion, and increase in hyphal size formed at the hydrocarbon-water interface. Inclusions were present in the hexadecane-grown fungus that were absent when the Penicillium sp. was grown on peptone. Sphaeropsis pyriputrescens, the causal agent of Sphaeropsis rot of pears and apples, is a recently described species. In this study the effects of culture media, temperature, water potential, pH and light on mycelial growth and pycnidial production of S. pyriputrescens were evaluated. Apple juice agar and pear juice agar were most suitable for mycelial growth of all six isolates tested. Cornmeal agar was not suitable for either mycelial growth or pycnidial production. The fungus grew from -3 to 25 C, with optimum growth at 20 C and no growth at 30 C. The fungus grew at water potential as low as -5.6 MPa on potassium chloride-amended potato-dextrose agar (PDA). Hyphal extension was not observed at -7.3 MPa after 10 d incubation, but growth resumed when the inoculum plugs were placed on PDA. The fungus grew at pH 3.3-6.3 and optimum growth was at pH 3.3-4.2. No mycelial growth was observed at pH above 7.2 after 10 d incubation, but growth resumed when the inoculum plugs were transferred onto PDA. Regardless of medium tested, few pycnidia formed at 20 C in the dark. Pycnidial production was enhanced significantly by fluorescent light, but continuous light appeared to reduce pycnidial production, depending on the medium. Oatmeal agar (OMA) was most suitable for production of pycnidia and conidia. Pycnidia that formed on 3 wk old OMA cultures at 20 C under 12 h light/12 h dark produced abundant conidia, and the technique is recommended for inoculum production.  相似文献

A new species of Phacidiopycnis associated with pome fruits is described. The fungus causes fruit rot on apples during storage and is associated with a twig dieback and canker disease of crabapple trees and dead twigs of pear trees. To characterize the biology of the fungus and compare it with Ph. piri, the type species of the genus, effects of nine media and light on mycelial growth and pycnidial production, mycelial growth in response to temperature and mode of conidial germination in response to nutrient were determined. Apple-juice agar, pear-juice agar, prune-juice agar, potato-dextrose agar (PDA) and malt-extract agar, Czapek-Dox agar and oatmeal agar (OMA) favored mycelial growth. Cornmeal agar (CMA) did not favor mycelial growth. Light effect on pycnidial formation was medium dependent. Abundant pycnidia with mature conidia formed in 14 d old PDA and OMA cultures at 20 C, regardless of light, whereas none or very few pycnidia formed on other media in the dark. Fluorescent light stimulated formation of pycnidia except on CMA. The fungus grew at -3-25 C, with optimum growth at 15-20 C. Conidia germinated either by forming germ tubes or less often by budding. Budding of conidia occurred in 1 and 10% pear-juice solutions but not in 100% pear-juice solution. Six isolates of Ph. washingtonensis from different species of pome fruits had identical ITS sequences. The sizes of the ITS region were the same for both Ph. washingtonensis and Ph. piri, and four polymorphic nucleotide sites were found in the ITS region between Ph. washingtonensis and Ph. piri. The similarity in ITS sequences between these two taxa is confirmatory evidence for the erection of the new species of Phacidiopycnis associated with pome fruits we describe here.  相似文献

Acid phosphatase activities in a culture liquid and mycelial extract were studied in submerged cultures of the filamentous fungus Humicola lutea 120-5 in casein-containingmedia with and without inorganic phosphate (Pi). The Pi-repressible influence on the phosphatase formation was demonstrated. Significant changes in the distribution of acid phosphatase between the mycelial extract and culture liquid were observed at the transition of the strain from exponential to stationary phase. Some differences in the cytochemical localization of phosphatase in dependence of Pi in the media and the role of the enzyme in the release of available phosphorus from the phosphoprotein casein for fungal growth were discussed.  相似文献

The effect of copper (II) ions on the growth of three brown-rot fungi, six white-rot fungi and one blue-stain fungus in solid medium was evaluated. The fungi were grown in malt extract agar with different concentrations of copper added, and the radial growth rate was determined. At the end of the incubation period, the mycelial biomass and the media pH were determined. The white-rot and blue-stain fungus grew up to 3 mM and 6 mM copper, respectively and the brown-rot fungi were the only ones that grew up to 10 mM, with higher growth rates than those shown by the other fungi. In general, the brown-rot fungi produced greater acidification in the culture media than the white-rot fungi and blue-stain fungus, and the acidification increased when the amount of copper was increased. The biomass production for the different species, in the absence or presence of copper, was not related to the radial growth rate, and the fungal species that produced the greatest biomass amounts did not correspond to those that presented the highest growth rates. The brown-rot fungi Wolfiporia cocos and Laetiporus sulfureus and blue-stain fungus Ophiostoma sp. demonstrated greater tolerance to high copper concentrations in solid medium than the white-rot fungi, determined as radial growth rate. On the other hand, the highest biomass producers in solid medium with copper added were the white-rot fungi Ganoderma australe and Trametes versicolor and the brown-rot fungus Gloeophyllum trabeum.  相似文献

Microorganisms associated with invertebrate hosts have long been suggested to be a source for bioactive metabolites. In this study, we reported that a sponge-associated fungus, Letendraea helminthicola, produced two antifouling compounds: 3-methyl-N-(2-phenylethyl) butanamide and cyclo(D-Pro-D-Phe). To optimize the production of these antifouling compounds, we then examined the production of compounds under different culture conditions (temperature, salinity, pH, and carbon and nitrogen sources). This fungus grew well and produced more compounds at temperatures between 18 and 30°C; the fungus grew well at 75 parts per thousand (ppt) salinity but produced the highest amount of antifouling compounds at 30 and 45 ppt. The optimal initial pH value for mycelial growth was 5.5 to 6.5, whereas the production of the antifouling compounds was maximized at pH 3.5 and 4.5. Glucose and xylose (as carbon sources) increased the production of antifouling compounds. Yeast extract and peptone (as nitrogen sources) maximized the production of mycelial biomass and antifouling compounds. Our results indicate that culture conditions greatly affect the production of bioactive compounds from mycelial fungal cultures as exemplified by strain L. helminthicola and that the conditions favorable for fungal growth may not be the best conditions for bioactive compound production.  相似文献

The California (LGCA) and Butte Sink (LGBS) strains of the sterol auxotrophic fungus Lagenidium giganteum (Oomycetes: Lagenidiales) enter the sexual cycle on media supplemented with sterols. A third isolate of this mosquito pathogen, the North Carolina strain (LGNC), requires sterols plus phospholipids to produce oospores in vitro. Enrichment of the polar and neutral lipid fractions of the LGCA and LGBS strains with unsaturated fatty acids promoted oospore induction, and increased oospore viability. With the exception of the LGCA strain, there was no consistent relationship between phospholipid supplementation in growth media and mycelial phospholipid content.  相似文献

Mycelia ofTricholoma bakamatsutake isolate No. 4 grew at temperatures ranging from 10 to 30°C, and the optimum was around 25°C. In well-buffered media of initial pH 5.0 and 6.0, No. 4 mycelia secreted gluconic acid and lowered medium pH. Mycelial growth then accelerated slightly; and with the exhaustion of glucose, growth and secretion of gluconic acid stopped. In 10 different media of initial pH 4.0–7.0, No. 4 mycelia showed higher gluconic acid secretion with higher initial pH. No. 4 mycelial grew best in pH 5.0 media, in which gluconic acid secretion was low. Mycelia of 29 isolates including No. 4 grew better in the media in which less glucose, total carbon and total nitrogen remained, and almost all isolates secreted gluconic acid. Most of the 29 isolates showed irregular colony shapes with rough mycelial fronts, brown pigmentation and aerial hypha on colony surfaces, and brown pigmentation of media under colonies. Dissimilarities were calculated with coded morphological characters on colonies, and similarity between isolates was found not to correlate with proximity of origin. Chlamydospores were observed on every colony of the 29 isolates. Chlamydospores were present on colonies of No. 4, reaching to 2 mm from the mycelial front, where brown pigmentation had not yet developed, and the numbers of chlamydospores incresed with mycelial aging.  相似文献
