以草生欧文氏杆菌LS005为指示菌,用管蝶法测定苏云金杆菌KN-11发酵液中增效物质的相对浓度。结果表明管蝶法和生物测定法在测定增效物质的代谢曲线有较好的一致性,均表明,菌株KN-11的增效物质城营养体时(约9h)开始产生,到芽孢初步形成时(约20h)已达到最高值的80%以上,之后是一个缓慢的积累过程。  相似文献

本文对烟台海域海月水母水母体阶段伞径、腕长及体重的生长规律进行了研究,并首次对其生长过程中水管系统的发育进行了观察。结果表明,在人工培养条件下,海月水母体重(W)与伞径(D:15~150㎜)呈幂函数增长;腕长(L)与伞径(D)呈线性关系;伞径生长曲线方程为:Dt = 4×10-6t4-0.0014t3 0.1087t2 0.5079t 9.428 (R2=0.9993)。。海月水母初生碟状体出现主辐管与间辐管,3日龄时碟状体出现从辐管,11日龄时出现环管、分离点。随着水母的生长,伞径增大,水管系统的分离点数与聚合点数逐渐增多,成熟时1/4伞部分离点数最多为66个,聚合点数最多为32个。  相似文献

棉铃虫颗粒体病毒增效蛋白基因2.6kb片段的表达   总被引:7,自引:2,他引:5  
以棉铃虫颗粒体病毒(Helicoverpa armigera granulosis virus,简称HaGV)基因组的DNA为模板设计引物,PCR扩增病毒增效蛋白(Enhanicn)基因,然后经BamHI/Pst I双酶切消化,得到近乎全长的约2.6kb的增效蛋白基因片段,再与pQE32质粒连接,构建了重组表达载体pQE2/En,转化大肠杆菌M15(pREP4),在IPTG诱导下表达出分子量约为102kD的融合蛋白并命名为P102,纯化的P102包涵体显示了明显的增效活性,在感染后168h时统计可提高HaNPV对棉铃虫幼虫的感染死亡率6.25%~27.09%,缩短LT5012.3h以上;在感染后72h时统计可提高Bt对棉铃虫幼虫的感染死亡率28.18%,缩短LT5o12.33h.  相似文献

利用BIOSTAT ?-CL1 5L全自动发酵罐和 2 0t不锈钢发酵罐 ,对苏云金芽孢杆菌不同菌株 (GC-91 ,MP342 ,HD-1 )发酵上清液中增效物质的生成进行了研究 ,发现增效物质于对数生长期前期开始产生并积累 ,至对数生长期末期达到高峰 ,并保持稳定 ;不同菌株的发酵上清中增效物质生成量不同 ,其中GC-91最强 (增效倍数f=6.0 ) ,MP342次之 (f=3.7) ,HD 1最弱 (f=1.5 ) ;GC-91菌株上清液中增效物质生成曲线与晶体含量 ,效价代  相似文献

目的:建立一种在150L发酵罐中提高rhIL-11蛋白产出率的控制方法。方法:研究在毕赤酵母表达rhIL-11的发酵过程中,分别采用间歇法(甲醇浓度0.5%,每24h诱导一次)、分批补料法(甲醇补料速度在8h内从2.88ml/min提高至8.64ml/min)以及恒甲醇浓度法(利用甲醇浓度传感器检测并控制甲醇补料速度使甲醇浓度始终保持0.5%左右)的不同甲醇诱导模式,对工程菌生长及目的蛋白表达的影响。结果:采用间歇法的菌浓OD_(600)可达到80、rhIL-11表达浓度可达到0.1mg/ml;分批补料法的菌浓OD_(600)可达到240、rhIL-11表达浓度可达到1.0mg/ml;恒甲醇浓度法的菌浓OD_(600)可达到300、rkIL-11表达浓度可达到1.0mg/ ml。结论:rhIL-11蛋白产出率,恒甲醇浓度法(约0.12g/d·L)比间歇法(约0.01g/d·L)和分批补料法(约0.09g/d·L)都高,分别提高了12~13倍和30~40%。  相似文献

实验旨在探讨钙/钙调蛋白依赖性蛋白激酶II(calcium/calmodulin-dependent protein kinase Ⅱ,CaMKⅡ)在脊髓背角C-纤维诱发电位长时程增强(long—term potentiation,LTP)的诱导和维持中的作用。用Western blot技术分别检测LTP形成30min和3h脊髓背角(L4-L6)CaMKⅡ的含量及其磷酸化水平。同时观察脊髓局部给予CAMKⅡ选择性抑制剂KN-93后对脊髓背角LTP和CaMKII磷酸化的影响。观察结果如下:(1)诱导LTP后30min,CaMK Ⅱ的磷酸化水平明显高于对照组,而CaMKⅡ的总量无变化;诱导LTP后3h CaMKⅡ的磷酸化水平进一步升高。而且CaMKⅡ的总量也明显增加(n=4);(2)强直刺激前30min于脊髓局部给予CaMKⅡ的特异性抑制剂KN-93(100μmol/L),可阻断LTP的诱导,同时明显抑制CaMKⅡ的磷酸化水平;(3)诱导LTP后30min给予KN-93,可显著抑制LTP的维持,同时CaMKⅡ的磷酸化水平与未用药组相比也明显降低(n=3);(4)LTP3h后给予KN-93,LTP的幅值不受影响,磷酸化的CaMKⅡ的含量与用药前相比也无差别(n=3)。根据上述实验结果可以认为,CaMKⅡ的激活参与脊髓背角C-纤维诱发电位LTP的诱导和早期维持过程。  相似文献

苏云金芽孢杆菌增效物质回收工艺的改进   总被引:2,自引:0,他引:2  
应用膜过滤技术对苏云金芽孢杆菌KN11增效物质回收工艺进行了改进,发现纳滤膜(200D)能完全截留KN11增效物质,通过3种膜过滤(0.1μm微滤膜、10000D超滤膜和200D纳滤膜)的总回收率达到85.5%;与常规回收比较,膜过滤回收工艺能显著提高浓缩液和粉的增效物质含量(分别为99.35U/mL,457.70U/g),同时除去了大部分的糖、氮等可溶性杂质,使浓缩液和粉保持了较好的理化性状。所配制的Bt高含量悬乳剂的效价在15645~19465IU/μL之间,含固量较低且流动性较好,Bt高含量原粉效价可达100646IU/mg。  相似文献

【目的】克隆小麦条锈菌钙调素依赖蛋白激酶基因Pscamk,并分析其在条锈菌侵染小麦过程中的表达特征及初步功能。【方法】基于本实验室已测序的小麦条锈菌基因组序列,利用RT-PCR方法,从小麦条锈菌生理小种CYR32中克隆Pscamk基因的cDNA序列,并利用网络数据库和生物信息学工具预测该基因编码蛋白的基本特征和保守结构;运用qRT-PCR技术分析Pscamk在不同发育及侵染阶段的表达水平,进一步通过钙调素依赖蛋白激酶(CaMK)的免疫抑制剂KN-93处理小麦条锈菌夏孢子,观察其萌发状况。【结果】获得1个1620 bp的小麦条锈菌CaMK基因Pscamk;序列分析发现,Pscamk编码蛋白包含CaMK蛋白的保守结构域,并与小麦杆锈菌该类蛋白序列相似性最高。qRT-PCR分析表明,Pscamk在条锈菌侵染初期过程中的芽管发育、初生菌丝侵染及吸器形成时期呈显著上调表达,且在条锈菌接种6 h时表达量最高,为对照夏孢子的20.74倍。在专一性免疫抑制剂KN-93处理后,随着KN-93施加浓度的增加,条锈菌夏孢子萌发率逐渐降低,当浓度为1.4μmol/L时夏孢子萌发率为8.02%,仅为对照的12%。【讨论】推测Pscamk基因参与了小麦条锈菌夏孢子萌发、芽管发育以及初期侵染结构的形成。本研究为进一步探索条锈菌细胞钙信号传导机理和致病机制奠定了基础。  相似文献

松蚜Cinara pinea Mordviko是马尾松、黑松等松属Pinus树种1—5年生枝干上的重要害虫。松蚜腹部有一对呈碟状的腹管。成虫和若蚜的腹管常有橙红色液体排出(附图),约1—2分钟后即凝固,并能粘附在腹管的端末30—60分钟,有的甚至达数小时。成虫腹管排出物直径0.4—0.5毫米左右,千粒重为42—45毫克。当蚜体受到某种刺激后,腹管也能排出上述物质。 蚜虫的腹管是昆虫身体上一种很特殊的构造。在昆虫分类上可以根据腹管的有无把蚜虫和其他昆虫区别开来;其形状和大小也很不一致,所以在形态学和分类学上都有重要意义。  相似文献

用饲料感染法建立了棉铃虫Helicoverpa rmigera(Hubmer)敏感品系(SUS1)对Bt生物农药的敏感毒力基线和区分剂量,1995年测定了五省六县棉铃虫初孵幼虫对Bt生物农药的敏感性,结果表明:山东阳谷、河北邯郸、河南新乡、安徽萧县及江苏丰县棉铃虫已产生早期抗性,抗性个体百分率为5%~10%,与敏感品系相比,LC50值稍有增加,但斜率b值明显变小;而江苏东台棉铃虫仍属敏感。这是国内外首次诊测到棉铃虫对Bt生物农药抗性。用棉叶喂饲法测定比较了转Bt基因棉花品系对不同种群棉铃虫的抗虫性效果,结果表明:用早期抗性的阳谷和新乡棉铃虫初孵幼虫接虫5d后平均死亡率较敏感品系下降16%~29%,说明棉铃虫对Bt农药与转Bt生物基因棉花品系间存在交互抗性。还讨论了Bt农药的抗性治理对策。  相似文献

Background  

Bacillus thuringiensis (Bt), an ubiquitous gram-positive spore-forming bacterium forms parasporal proteins during the stationary phase of its growth. Recent findings of selective human cancer cell-killing activity in non-insecticidal Bt isolates resulted in a new category of Bt parasporal protein called parasporin. However, little is known about the receptor molecules that bind parasporins and the mechanism of anti-cancer activity. A Malaysian Bt isolate, designated Bt18 produces parasporal protein that exhibit preferential cytotoxic activity for human leukaemic T cells (CEM-SS) but is non-cytotoxic to normal T cells or other cancer cell lines such as human cervical cancer (HeLa), human breast cancer (MCF-7) and colon cancer (HT-29) suggesting properties similar to parasporin. 参照抗生素效价生物测定法,用管碟法测定碱性蛋白酶活力,可大大提高菌种筛选的工作效率。待测样品在2%酪蛋白平板上的水解圈直径被效正后,从标准曲线上可查得相应的酶活。管碟法测得的酶活与Folin法测得的酶活相差一般小于2000u/ml。管碟法测定酶活时,操作要求认真仔细,操作误差可使一样品的水解圈直径相差2.9mm,但一般误差在0.2~0.3mm,相应酶活误差小于1000u/ml。  相似文献

 应用群落生态学方法研究了转Bt基因抗虫保铃棉(Gossypium hirsutum)‘972’、‘109B’、‘690’与‘154’种植后对江苏沿海的大丰旱作棉区与沿江的如皋水旱轮作棉区棉田杂草的影响风险。同地同期测定结果显示:不同保铃棉品系棉田杂草物种丰富度(S)、个体数量(N)、群落多样性(H′)、均匀度(J)与优势集中性(C)值在对照品种(‘泗棉3号’与‘苏棉16’)棉田上下波动,优势种及其多度基本接近。两地五期的综合分析结果显示各保铃棉品系杂草群落多样性、均匀度都略高于对照棉,而优势集中性都略低于对照棉。不同抗性保铃棉品系棉田杂草群落特征参数组分对群落多样性的直接间接作用与对照品种棉田的趋势基本一致。同一生态类型棉区的保铃棉高抗、中抗品系与对照品种棉田杂草的种相似性系数与群落相似性系数都较高。上述结果表明,转基因抗虫保铃棉品系棉田的杂草群落稳定性并不比对照品种的差,因保铃棉品系本身产生新的恶性杂草风险的可能性并不明显。  相似文献

The refuge strategy is designed to delay evolution of pest resistance to transgenic crops producing Bacillus thuringiensis Berliner (Bt) toxins. Movement of insects between Bt crops and refuges of non-Bt crops is essential for the refuge strategy because it increases chances that resistant adults mate with susceptible adults from refuges. Conclusions about optimal levels of movement for delaying resistance are not consistent among previous modeling studies. To clarify the effects of movement on resistance evolution, we analyzed simulations of a spatially explicit model based partly on the interaction of pink bollworm, Pectinophora gossypiella (Saunders), with Bt cotton. We examined resistance evolution as a function of insect movement under 12 sets of assumptions about the relative abundance of Bt cotton (50 and 75%), temporal distribution of Bt cotton and refuge fields (fixed, partial rotation, and full rotation), and spatial distribution of fields (random and uniform). The results show that interactions among the relative abundance and distribution of refuges and Bt cotton fields can alter the effects of movement on resistance evolution. The results also suggest that differences in conclusions among previous studies can be explained by differences in assumptions about the relative abundance and distribution of refuges and Bt crop fields. With fixed field locations and all Bt cotton fields adjacent to at least one refuge, resistance evolved slowest with low movement. However, low movement and fixed field locations favored rapid resistance evolution when some Bt crop fields were isolated from refuges. When refuges and Bt cotton fields were rotated to the opposite crop type each year, resistance evolved fastest with low movement. Nonrecessive inheritance of resistance caused rapid resistanceevolution regardless of movement rate. Confirming previous reports, results described here show that resistance can be delayed effectively by fixing field locations and distributing refuges uniformly to ensure that Bt crop fields are not isolated from refuges. However, rotating fields provided better insect control and reduced the need for insecticide sprays.  相似文献

The induction of alkaline phosphatase (ALP) by dibutyryl adenosine 3':5'-cyclic monophosphate (Bt2cAMP) was investigated in strain JTC-12 . P3 cells derived from monkey (Maccaca irus) kidney cortex. ALP activity was increased by Bt2cAMP in a dose-dependent manner, reaching a plateau at concentrations higher than 5 mM with the activity being about 4 times that of the controls. The concentration of Bt2cAMP required for half-maximal induction of ALP activity was about 0.8 mM. ALP activity was increased rapidly by Bt2cAMP for the first 5 days and then continued to increase gradually towards a plateau level. Removal of Bt2cAMP from the medium caused a rapid decrease in the activity, suggesting that the induction of ALP activity by Bt2cAMP is reversible. ALP activity was induced synergistically in the presence of 1 mM sodium butyrate together with Bt2cAMP at concentrations from 0.01 to 1 mM. It was also found that in the presence of 1 mM Bt2cAMP, sodium butyrate increased ALP activity in the same manner as Bt2cAMP did in the presence of 1 mM sodium butyrate. Although dexamethasone, a potent glucocorticoid, had no effect on ALP activity in control cells, the hormone suppressed the ALP activity induced by Bt2cAMP in a dose-dependent manner. At concentrations above 0.2 mM, two xanthine derivatives, theophylline and 3-isobutyl-1-methyl-xanthine (IBMX), also inhibited the induction of ALP activity by 1 mM Bt2cAMP. Inhibitors of protein synthesis, cycloheximide (1.5 micrograms/ml) and pactamycin (10 micrograms/ml), as well as inhibitors of RNA synthesis, actinomycin D (2 micrograms/ml) and alpha-amanitin (50 micrograms/ml), suppressed the induction of ALP activity.  相似文献

以小菜蛾Plutella xylostella为试虫,采用生物测定方法测定了粘虫颗粒体病毒(Pseudaletia unipuncta granulosis virus,PuGV-Ps)对苏云金杆菌(Bacillus thuringiensis,Bt)的增效作用。结果表明:不同配伍PuGV-Ps和Bt间的共毒系数在105.3至195.0之间, PuGV-Ps对Bt毒力具有增强作用,其中以Bt∶PuGV-Ps为4∶1增效作用最明显,72 h LC50为0.039 mg/mL。不同温度和pH值都影响PuGV-Ps对Bt的增效作用,16℃~20℃增效程度明显高于24℃~32℃,而碱性条件下(pH 8~9)增效作用更显著。PuGV-Ps对Bt的增效作用因小菜蛾龄期不同而变化,2、3龄幼虫死亡率较单独使用Bt分别提高了50%和30.31%,而作用于低龄(1龄)和高龄(4龄)幼虫时对Bt的增效作用不显著。PuGV-Ps饲喂2 h后再接毒Bt,小菜蛾死亡率明显提高,48 h死亡率达66.67%,较直接饲喂Bt+PuGV-Ps处理死亡率提高了53.87%,差异极显著。SDS-PAGE表明PuGV-Ps具有碱性蛋白酶的活性,离体条件下能促进δ-内毒素酶解为47 kD,60 kD和61 kD的毒性肽。  相似文献

Background

Uncertainty persists over the environmental effects of genetically-engineered crops that produce the insecticidal Cry proteins of Bacillus thuringiensis (Bt). We performed meta-analyses on a modified public database to synthesize current knowledge about the effects of Bt cotton, maize and potato on the abundance and interactions of arthropod non-target functional guilds.

Methodology/Principal Findings

We compared the abundance of predators, parasitoids, omnivores, detritivores and herbivores under scenarios in which neither, only the non-Bt crops, or both Bt and non-Bt crops received insecticide treatments. Predators were less abundant in Bt cotton compared to unsprayed non-Bt controls. As expected, fewer specialist parasitoids of the target pest occurred in Bt maize fields compared to unsprayed non-Bt controls, but no significant reduction was detected for other parasitoids. Numbers of predators and herbivores were higher in Bt crops compared to sprayed non-Bt controls, and type of insecticide influenced the magnitude of the difference. Omnivores and detritivores were more abundant in insecticide-treated controls and for the latter guild this was associated with reductions of their predators in sprayed non-Bt maize. No differences in abundance were found when both Bt and non-Bt crops were sprayed. Predator-to-prey ratios were unchanged by either Bt crops or the use of insecticides; ratios were higher in Bt maize relative to the sprayed non-Bt control.

Conclusions/Significance

Overall, we find no uniform effects of Bt cotton, maize and potato on the functional guilds of non-target arthropods. Use of and type of insecticides influenced the magnitude and direction of effects; insecticde effects were much larger than those of Bt crops. These meta-analyses underscore the importance of using controls not only to isolate the effects of a Bt crop per se but also to reflect the replacement of existing agricultural practices. Results will provide researchers with information to design more robust experiments and will inform the decisions of diverse stakeholders regarding the safety of transgenic insecticidal crops.  相似文献
