A method has been developed to measure the cell volume of theunicellular green alga Dunaliella parva 19/9 using Li+ measurementsonly. Concentrations of internal solutes can also be calculatedif they are assayed in the same samples as Li+. We found thatD. parva cells grown in 0.4 kmol m–3 NaCl have an averageaqueous cell volume of 65.1 ?2.9 µm3, a K+ concentrationof 126?6 mol m–3, a Na+ concentration of 11?11 mol m–3and a glycerol concentration of 615?27 mol m–3 (n= 12).Algae grown in 1.5 kmol m–3 NaCl have an average aqueouscell volume of 131 ?7.5 µm3, a K+ concentration of 109?4mol m–3, a Na+ concentration of 10?39 mol m–3 anda glycerol concentration of 1 425?59 mol m–3 (n = 12).These results indicate that D. parva cells adapted to high salinitieshave larger cell volumes than those adapted to lower salinities.However, there is no evidence for a significant difference ininternal Na+ concentration, despite the almost 4-fold differencein the concentration of external NaCl. The intracellular glycerolconcentration alone accounts for 65% and 54%, respectively,of the osmotic balance in low and high salt grown cells. A comparison of the methods of X-ray microanalysis,compartmental analysis and longitudinal ion profiles to estimatecytoplasnuc ion concentrations in two maize varieties.—J.exp. Bot. 39: 279-290. The ion content of compartments within plant root cells hasbeen studied by three different methods; flux analysis usingradioactive isotopes, longitudinal ion profiles and X-ray microanalysis.The data provide estimates of the concentrations of K+ and CIin the cytoplasm of roots of culture solution and salt grownmaize by three independent methods. In the cultivar LG11 grown in 50 mol m–3 NaCl X-ray microanalysis,compartmental analysis and longitudinal profiles yielded approximatelyagreeing values for cytoplasmic K+ of 90, 70 and 62 mol m–33of tissue volume respectively. However, the methods disagreedon cytoplasmic Cl where the value obtained by compartmentalanalysis was about four times that from X-ray microanalysisand longitudinal profiles. Salt Tolerance in the Triticeae: Leymus sabulosus   总被引:1,自引:0,他引:1  
Elymus dahuhcus, Leymus giganteus, L. angustus, L. sabulosusand, to a lesser extent, L. triticoides, were found to tolerate200 mol m–3 NaCl in solution culture. Elymus dahuricusdiffered from the Leymus species in its ion-uptake characteristics,showing a greater uptake of Cl and Na and a greater loss ofK from the shoots. In a more detailed experiment on Leymus sabulosusit was found that transpiration rates altered rapidly in responseto changes in external salinity whereas the accumulation ofNa and Cl in the leaves exhibited a lag of several days. Insalt stressed L. sabulosus Cl partially replaced the high levelsof nitrate found in the leaves of control plants. Glycinebetainelevels increased in the leaves from 8.0 mol m–3 plantsap in the controls to 28 mol m–3 plant sap at 250 molm–3 NaCl. The time-course of exchange of sodium and potassium ions fromroot and leaf material of the halophyte Suaeda maritima hasbeen followed and the data analysed according to the phenomenologyof efflux, or compartmental, analysis. Sodium ions were exchangedmuch more slowly (c. 4 times) from the vacuoles of leaf cellsof plants grown in sodium chloride than were potassium ionsfrom the vacuoles of leaf cells of plants grown either in similarconcentrations of potassium chloride or in low concentrationsof potassium. In plants grown in sodium chloride, sodium ionswere exchanged 9 times more slowly from the vacuoles of leafcells than from the vacuoles of root cells. The concentration of sodium ions in the cytoplasm of leaf cellsof plants growing in 340 mol m–3 sodium chloride was estimatedto be 165 mol m–3 when the average concentration in theleaf tissue was about 600 mol m–3. As measured by movement from mature to developing leaves inintact plants; there was less in vivo retranslocation of 22Naand 36CI in plants growing in sodium chloride than there wasof 86Rb in plants growing either in potassium chloride or innon-saline conditions. The results are discussed in terms of the concept and energeticsof compartmentation of ions in the cells of halophytes.  相似文献

Loading of K, Na and Cl into fibrous roots of salt-treated citrusgenotypes, Rangpur lime (Citrus reticulata var. austera hybrid?)and Etrog citron (C. medica L.) was investigated in relationto root anatomy, in particular, the differentiation of the epidermal-hypodermallayers with distance from the root tip. The influence of durationof salinity treatment on the characteristics of K, Na and Claccumulation in leaves of the two genotypes was explored intwo experiments respectively, covering the short term (14 d)and long term (12 weeks). This study focused on two regions of the fibrous root, a segment2–12 mm from the root tip, immediately basipetal to thezone of elongation and a differentiated region of the maturesuberized, fibrous root, 40–50 mm from the root tip. Inthe distal root segment (2–12 mm) the epidermis and hypodermisof both genotypes was observed as two closely packed, uniseriatelayers of living cells. In the proximal root segment (40–50mm) the differentiated hypodermis was evident as a uniseriatelayer of thick-walled lumina interspersed with ‘passagecells’ which were frequently associated with clustersof viable epidermal cells. The characteristics of Na and Cl loading in the two root zonesdiffered profoundly during the short term loading (acclimation)phase. Attainment of quasi-steady-states for Na and Cl in thedistal region (with the exception of Na in Etrog citron) wasrapid as was Na equilibration of the proximal root segmentsin both genotypes. In contrast, Cl loading in the proximal regiontook c. 14 d to reach a quasi-steady-state by which time Cllevels were 2 to 3 times higher in the proximal than in thedistal root segments. The superior tolerance of Rangpur lime to long term salinitywas highly correlated to Cl exclusion from the leaves. However,during the first 14 d of acclimation to 50 mol m–3 NaClthere was no segregation of the two genotypes based upon leafCl levels. Expression of differential accumulation of Cl inleaves appeared to be a time dependent process and was manifestonly after Cl saturation of the proximal root which representsthe bulk of the fibrous root system. The salt tolerance of Rangpurwas also associated with high selectivity of fibrous roots forK. over Na. A pronounced loss of K from cortical cells in theproximal root segment of salt-stressed Etrog citron was alsoevident by X-ray microanalysis. Salt Tolerance in the Succulent, Coastal Halophyte, Sarcocornia natalensis   总被引:2,自引:0,他引:2  
The effects of 0, 50, 100, 200, 300, 400 and 500 mol m–3NaCl on growth and ion accumulation in the succulent, coastalhalophyte Sarcocornia natalensis (Bunge ex Ung.-Sternb.) A.J. Scott were investigated. Increase in salinity from 0 to 300 mol m–3 NaCl stimulatedproduction of fresh, dry, and organic dry mass, increased succulenceand shifted resource allocation from roots to shoots. Growthwas optimal at 300 mol m–3 and decreased with furtherincrease in salinity. Water contributed to a large proportion of the increase in freshmass. Inorganic ions, especially Na+ and Cl– contributedsubstantially to the dry mass. At 300 mol m–3 NaCl inorganicions contributed to 37% of total dry mass and NaCl concentrationin the shoots was 482 mol m–3. Expressed sap osmotic potentialsdecreased from –2.10 to –3.95 MPa as salinity increasedfrom 0 to 300 mol m–3 NaCl. Massive accumulation of inorganicions, especially Na+ and Cl, accounted for 86% of theosmotic adjustment at 300 mol m–3 NaCl. Salinity treatments decreased the concentrations of K+ in shoots.Plant Na+ :K+ ratios increased steadily with salinity and reacheda maximum of 16.6 at 400 mol m3 NaCl. It is suggested that the exceptional salt tolerance of S. natalensisis achieved by massive inorganic ion accumulation which providessufficient solutes for osmoregulation, increased water fluxand turgor-induced growth. The effects of light/dark on anion fluxes in isolated guardcells of Commelina communis L. have been studied, using 82Brand 36Cl. Transfer of open guard cells from light to dark hasno effect on the 82Br influx, but produces a marked transientstimulation of 82Br or 36Cl efflux, similar to the effect ofsuch transfer on the 86Rb fluxes, and to the effects on both86Rb and 82Br fluxes of adding ABA. On return of guard cellsto light, after the transient, there is a further reductionin Cl/Br efflux. It is argued that control of a specific processof ion extrusion is important in regulating the ability of guardcells to stay open. In three out of four batches of steady-statetissue labelled with 82Br, the plasmalemma fluxes were highenough, relative to the tonoplast fluxes, for the efflux kineticsto be separable into two exponential components, allowing estimationof bromide contents in cytoplasm and vacuole (Qc and Qv), andfluxes at plasmalemma and tonoplast. With opening in light,Qc increased by 3.9 ± 0.4 pmol mm–2 µm–1and Qy by 5.2 ± 0.6 pmol mm–2 µm–1(change in content per mm2 of epidermis perµm change inaperture). Using rough estimates for the volumes of cytoplasmand vacuole these figures suggest that at 6.1 µm in thedark the concentrations were about 63 mol m–3 in the cytoplasmand 35 mol m–3 in the vacuole, rising to about 185 molm–3 in the cytoplasm and 125 mol m–3 in the vacuole,at 16.7 µm aperture in light. Neither increase can providean adequate increase in salt concentration to account for theosmotic change required, and some solute other than potassiumsalt must also be involved. In one experiment with 82Br andin the only experiment with 36Cl the plasmalemma flux was lower,and not high enough relative to the tonoplast flux to allowseparation of two phases in the efflux curves, and calculationof cytoplasmic and vacuolar contents and fluxes. We report that growth of Dunaliella salina at either 13°C/150µmol m–2s–1 or 30°C/2,500 µmol m–2s–1 results in the accumulation of comparable levels ofcarotenoids and the zeaxanthin-binding protein, Cbr. We concludethat carotenoid and Cbr abundance in this green alga respondto changes in PSII ‘excitation pressure’ ratherthan to high light per se. The effects of different NaCl concentrations on the growth andnitrogen fixation activity of white lupin (Lupinus albus [L.])was studied over a 6 d period. Plant growth parameters, photosynthesisand shoot respiration were unaffected by NaCl concentrationsup to 150 mol m–3. However, nitrogenase activity decreasedwith increased NaCl concentration up to 100 mol m–3, whilstthe O2 diffusion resistance increased with 100 mol m–3NaCl, but showed no further change when 150 mol m–3 NaClwas applied for 6 d. Increases in NaCl concentration decreasednodular starch content while increasing sucrose content, suggestingan osmotic regulation. These changes were associated with a77% decrease in sucrose synthase activity. The effect on theO2 diffusion resistance was paralleled by changes in glycoproteincontent of the nodules, as determined by immunogold localizationand ELISA. X-ray microanalysis studies of nodules showed that,following a 6 d exposure to 150 mol m–3 NaCl, Na+ ionswere largely excluded from the infected zone, whilst only lowlevels of Cl- ions penetrated into this region. Na+ entry intoroots and leaves was also at a low level. Leghaemoglobin contentdecreased with saline stress, as did superoxide dismutase; whichdecreased by 36% following exposure to 100 mol m–3 saltfor 6 d. These results are discussed in relation to the relativesalt tolerance of the Multolupa/ISLU-16 symbiosis. Ionic activity of K+ and Cl in the apoplast of the lowerepidermis of the leaf of Commelina communis was measured usingion selective micro-electrodes. Large gradients across the stomatalcomplex were observed which were related to stomatal aperture.On stomatal closure the activity of K+ and Cl in theapoplast of the guard cell rose from 3·0 mol m–3to 100 mol m–3 and 33 mol m–3 respectively. The diploid wheatgrass Thinopyrum bessarabicum was found towithstand prolonged exposure to 350 mol m–3 NaCl in hydroponicculture. During the gradual addition of salt to the externalmedium, osmotic adjustment was rapidly achieved by the accumulationof Na and Cl. Following osmotic adjustment constant leaf Naand Cl concentrations were maintained, and K was retained ata high level. Thinopyrum bessarabicum may be described as anosmoconformer, adjusting its internal osmotic pressure to 400–500mOsmol kg–1 above that of the external medium in hydroponicculture. Both slower shoot initiation and reduced leaf lengthcontributed to the reduced growth rates at higher salinities.Leaf width was not affected. Increasing salinity resulted inincreases in leaf concentrations of phosphate, glycinebetaine,sucrose and proline, and in decreases in the concentrationsof nitrate, sulphate, magnesium, calcium, total amino acidsand organic acids. The acidophilic alga Dunaliella acidophila exhibits optimalgrowth at pH 1. We have investigated the regulation of phosphateuptake by this alga using tracer techniques and by performingintracellular phosphate measurements under different growthconditions including phosphate limitation. In batch culturewith 2·2 mol m–3 phosphate in the medium the uptakeof phosphate at micromolar phosphate concentrations followeda linear time dependence in the range of minutes and rates werein the range of 1 µmol phosphate mg–1 chl h–1,only. However, under discontinuous phosphate-limited growthconditions, tracer influx revealed a biphasic pattern at micromolarphosphate concentrations: An initial burst phase resulted ina 104-fold internal phosphate accumulation and levelled offafter about 10 s. A double reciprocal plot of the initial influxrates obtained for phosphate-limited and unlimited algae exhibitedMichaelis-Menten kinetics. Phosphate limitation caused a significantactivation of the maximum velocity of uptake, yielding Vmaxup to 1 mmol mg–1 chl h–1 as compared to valuesin the order of 50 µmol phosphate mg–1 chl h–1for the second phase (this magnitude is also representativefor non-limited batch cultures). Concomitantly the Michaelisconstant was altered from 4 mmol m–3 to 0·7 mmolm–3. The rapid uptake of phosphate was inhibited by arsenateand FCCP and was not stimulated by Na+. The pH dependence oftracer accumulation and measurements of the intracellular phosphatepool under different growth conditions indicate that at lowpH and low external phosphate concentrations the high protongradient present under these conditions is utilized for a H3PO4uptake or a H+/H2PO4 cotransport. However, when the externalphosphate concentration was increased to levels sufficientlyhigh for transport to be driven by the positive membrane potential(10 mol m–3 phosphate), the pH dependence of phosphateuptake was more complex, but could be explained by the uptakeof H3PO4 or a H+/H2PO4-cotransport at low pH and a differenttype H2PO4-transport (with unknown type of ion coupling)at high pH-values. It is suggested that this flexible couplingof phosphate transport is of essential importance for the acidresistance of Dunaliella acidophila. Seedlings of cotton (Gossypium hirsutum L. cv. Acala SJ-2) weregrown in modified Hoagland nutrient solution with various combinationsof NaCl and CaCl2. Marking experiments and numerical analysiswere conducted to characterize the spatial and temporal patternsof cotton root growth at varied Na/Ca ratios. At 1 mol m–3Ca, 150 mol m–3 NaCl reduced overall root elongation rateto 60% of the control, while increasing Ca to 10 mol m–3at the same NaCl concentration restored the elongation rateto 80% of the control. Analysis of the spatial distributionof elongation revealed that the presence of 150 mol m–3NaCl in the medium shortened the growth zone by about 2 mm fromthe approximate 10 mm in the control and also reduced the relativeelemental elongation rate (i.e. the longitudinal strain rate,defined as the derivatives of displacement velocity of a cellularparticle with respect to position on root axis). Supply of 10mol m–3 Ca at the high salt condition restored partiallythe relative elemental elongation rate, but not the length ofthe growth zone. DNA polymerases were purified several hundred-fold from the10 000 x g soluble (polymerase I) and particulate (polymeraseIII) fractions prepared from virus PBCV-1 infected ChlorellaNC64A extracts. Both DNA polymerases exhibited optimal activitywith activated calf thymus DNA at pH 8.5. DNA polymerase I required3.0 mol m–3 MgSO4 and 150 to 250 mol m–3 KCl foroptimum activity whereas, DNA polymerase III required 2.0 molm–3 MgSO4 and 150 mol m–3 KCl. Both enzymes wereinhibited by pyrophosphate, actinomycin D, ethidium bromide,dideoxythymidine triphosphate, and N-ethylmaleimide but wererelatively insensitive to aphidicolin. DNA polymerase I differedfrom DNA polymerase III in its response to cations (particularlyNH4Cl), elution from a DEAE cellulose column, and molecularweight. Using NaH 14CO3 (0.1 to 3.0 mol m –3) fed to 5.0 mm ofan internodal cell of Nitella flexilis in artificial pond waterat pH 6.8 and 19–25 °C, we have found that the carbohydrates(lactose, xylose, mannose, galactoseand sucrose) are formedby photo-assimilation from 14C-DIC (dissolved organic carbon)in 1 h and are carried in the cytoplasm with amino acids (glutamineand alanine in particular) to the node attached to a tandeminternodal cell. These small sacchandes and some amino acidspassed, apparently unchanged, across the node into the sinkcell. Influx of DIC was highly sensitive to inhibitors of photosystemsI and II (at concentrations around 1.0 mol m–3) and touncouplers of phosphorylation. Most influx inhibitors, exceptfor NaN3, also reduced % transnodal transport. NH4+ (1.0 to5.0 mol m–3) appeared to reduce % transport in light (butnot in dark) with much less effect on influx. Dinitrophenoland Na citrate (at pH 8.2) also strongly reduced apparent %transport without altering cytoplasmic streaming rates. Someof the apparent reduction of transport could be due to an alterationof metabolism or of sequestering in the feed cell, but withNH4+ the latter was not detectable. Our findings support thehypothesis that transnodal transport, including that via theplasmodesmata, is at least partly ‘active’ and requiresmetabolic energy to sustain it. The aim of this study was to investigate changes in cell wallchemical composition and polymer size in the root tip of intactcotton seedlings (Gossypium hirsutum L. cv. Acala SJ-2) grownin saline environments, in order to relate the interaction betweenhigh salinity and root growth to possible changes in cell wallmetabolism. Cotton seedlings were grown in modified Hoagland nutrient solutionwith various combinations of NaCl and CaCl2. Cell walls werefractionated into four fractions (pectin, hemicellulose 1 and2, cellulose), and analysed for their total sugar content, neutralsugar composition and size of polysaccharides. At 1 mol m–3Ca, 150 mol m–3 NaCl resulted in a significant increasein the cell wall uronic acid content, but a reduction in cellulosecontent on a per unit dry weight basis. Supplemental Ca overcamethe inhibitory effect of high Na on cellulose content. The neutralsugar composition of the cell wall fractions showed no majorchanges caused by varied Na/Ca ratios. Determinations of polysaccharidepolymer size showed that high Na at 1 mol m–3 Ca led toan increase in the amount of polysaccharides of intermediatemolecular size and a decrease in that of small size in the hemicellulose1 fraction, indicating a possible inhibition of polysaccharidedegradation by high Na. This change was not observed in the10 mol m–3 Ca treatments. Two strains of Dunaliella, one halotolerant and one halophilic,were grown in batch culture at NaCl concentrations varying from500 mol m–3 to 3000 mol m–3. Measurements were madeof the following solutes: glycerol, Na+, K+, Mg2+, Cl,phosphate in the cells of logarithmic and of stationary-phasecultures. The method used was to separate the cells from thebulk of the medium by differential density centrifugation. Soluteconcentrations were calculated using Blue Dextran as a markerfor extracellular space. It was found that in log-phase cells,glycerol accounted for one-half to two-thirds of the total cellsolute concentration, the remainder being largely accountedfor by Na+ and Cl. In the stationary phase glycerol felland Na+, plus Cl, rose. Light intensity was found toaffect cell volume and solute content. The means whereby soluteconcentrations are controlled is discussed. The distribution of ions within wheat leaf mesophyll cells fromplants grown in the presence of 100 mol m–3 NaCl werestudied by X-ray micro-analysis, and the volume fractions occupiedby the analysed compartments were determined stereologically.Generally, the cells did not accumulate high ion concentrationsin their vacuoles, and the much higher Na+ and Cl concentrationsin the cell walls would produce an imbalance of cell water relations.The Na+, K+ and Cl concentrations in the cytoplasm wereoften too high to be compatible with biochemical activity. Bot. 36: 1142–1149. pH-sensitive micro-electrodes, filled with ion-exchanger resinhave been fabricated with a turgorinsensitive tip and have beenapplied to test the intracellular pH and changes thereof inroot hair cells of Sinapis alba. (1) The cytoplasmic pH of Sinapisroot hairs was determined to be 7.3 ±0.2 (at neutralexternal pH). (2) 10 mol m–3 sodium azide depolarizesthe membrane potential by about 100 mV and acidifies the cytoplasmby 0.8 pH-units. (3) The change from 1.0 mol m–3 to 10mol m–3 external potassium causes a depolarization ofabout 45 mV, but no change in internal pH. (4) At an externalpH of 5.0, sodium acetate hyperpolarizes the plasmalemma byabout 60 mV and acidifies the cytoplasmic pH by 0.2 to 0.3 units.(5) 2.0 mmol m–3 fusicoccin (FC) hyperpolarizes the plasmalemmaby 20–25 mV, acidifies the cytoplasm by 0.1 to 0.2 pH-units,and acidifies the external medium by about 0–3 pH-units.It is concluded that cytoplasmic acidification stimulates theelectrogenic proton pump in Sinapis root hairs, and it is suggestedthat the FC-induced effects, viz. hyperpolarization and externalacidification, can also be interpreted in this way. X-ray microanalysis was used to determine the distributionsof several nutrient elements between vacuoles of epidermal andmesophyll cells in barley leaves and these distributions wererelated to shoot nutrient concentrations. Under the growth conditionsused, P was found only in mesophyll vacuoles, never in the epidermis.In contrast, Cl and Ca were located almost exclusively in theepidermis while K and Na were more evenly distributed betweenthe two cell types. The compartmentation of Ca and Cl in theepidermis was maintained over a wide range of tissue concentrationsof these ions. In particular, Cl was excluded from the mesophyllof salt-grown barley until the tissue concentration reachedabout 170 mol m–3 and then it appeared in the vacuolesof these cells, but only at low concentrations. In contrast,Na was not excluded from the mesophyll of salt-grown or K-deficientbarley and there was evidence that this ion was preferentiallyaccumulated in the mesophyll. Nutrients were evenly distributedbetween the adaxial and abaxial epidermal layers, except K.which was at slightly higher concentrations in the adaxial epidermis.There was considerable variation in the concentrations of ionsin adjacent epidermal cells. The results indicate that intercellularcompartmentation of nutrients occurs in barley leaves and therole of this phenomenon in responses to nutrient deficienciesand salinity is discussed Key words: Compartmentation, barley, salinity, ions, potassium nutrition  相似文献
