A novel microorganism was isolated which is able to produce mannitol when grown in the presence of fructose and glucose as carbon sources. In flask culture in a medium containing 150 g fructose l–1, it yielded 67 g mannitol l–1 after 168 h. In fed-batch culture with 3–12% (w/v) fructose, production reached a maximum of 209 g mannitol l–1 after 200 h, corresponding to an 83% yield and a 1.03 g l–1 h–1 productivity. The isolated strain was identified as Candida magnoliae based on identical sequences in the D1/D2 domain of its 26S rDNA and a similar carbon source utilization pattern with C. magnoliae reference strains.  相似文献

The production of pigment-free pullulan by Aureobasidium pullulans in batch and fed-batch culture was investigated. Batch culture proved to be a better fermentation system for the production of pullulan than the fed-batch culture system. A maximum polysaccharide concentration (31.3 g l−1), polysaccharide productivity (4.5 g l−1 per day), and sugar utilization (100%) were obtained in batch culture. In fed-batch culture, feed medium composition influenced the kinetics of fermentation. For fed-batch culture, the highest values of pullulan concentration (24.5 g l−1) and pullulan productivity (3.5 g l−1 per day) were obtained in culture grown with feeding substrate containing 50 g l−1 sucrose and all nutrients. The molecular size of pullulan showed a decline as fermentation progressed for both fermentation systems. At the end of fermentation, the polysaccharide isolated from the fed-batch culture had a slightly higher molecular weight than that of batch culture. Structural characterization of pullulan samples (methylation and enzymic hydrolysis with pullulanase) revealed the presence of mainly α-(1→4) (∼66%) and α-(1→6) (∼31%) glucosidic linkages; however, a small amount (<3%) of triply linked (1,3,4-, 1,3,6-, 1,2,4- and 1,4,6-Glc p) residues were detected. The molecular homogeneity of the alcohol-precipitated polysaccharides from the fermentation broths as well as the structural features of pullulan were confirmed by 13C-NMR and pullulanase treatments followed by gel filtration chromatography of the debranched digests.  相似文献

Fourteen yeast strains were screened for production of 2-phenylethanol from l-phenylalanine with molasses as carbon source. Up to 1 g 2-phenylethanol l–1 was obtained. Using oleyl alcohol as a second phase for in situ product removal to enhance the production of 2-phenylethanol increased the yield to about 3 g 2-phenylethanol l–1 at 35 °C. The most productive strains were Kluyveromyces marxianus CBS 600 and CBS 397.  相似文献

Production of clavulanic acid (CA) by Streptomyces clavuligerus ATCC 27064 in shake-flask culture (28 °C, 250 rev min–1) was evaluated, with media containing different types and concentrations of edible vegetable oil. Firstly, four media based on those reported in the literature were examined. The medium containing soybean oil and starch as carbon and energy source gave the best production results. This medium, with the starch replaced by glycerol, and with various soybean oil concentrations (16, 23 and 30 g l–1) was utilized to further investigate CA production. Medium containing 23 g l–1 led to the highest CA productivity (722 mg l–1 in 120 h) and that one containing 30 g l–1 gave the highest CA titre (753 mg l–1 in 130 h). Also, substitution of corn and sunflower edible oils furnished similarly good results in terms of CA titre and productivity. It can be concluded that easily available vegetable oil is a very promising substrate for CA production, since it is converted slowly to glycerol and fatty acids, which are the main carbon and energy source for the microorganism.  相似文献

A mixed anaerobic bacterial culture decolorized Drimaren Orange K-GL, Everzol Red RBN and Everdirect Supra Yellow PG dyestuffs at 200 mg dyestuff l–1 over 24 h. Improved performance with complete decolorization within 24 h was achieved by incubation with 5 g yeast extract l–1 compared to glucose, lactose and sucrose though 50 mg yeast extract l–1 supplemented with 5 g lactose l–1 or 5 g sucrose l–1 also resulted in complete decolorization within 24 h.  相似文献

Cyclohexanone monooxygenase was produced from Acinetobacter calcoaceticus grown on a medium containing both glutamate (30 g l–1) and cyclohexanol (1 g l–1). Productivity was increased to 650 U l–1, an order of magnitude greater than previous production methods, thereby enhancing the potential commercial utility of this enzyme.  相似文献

The marine photosynthetic bacterium, Rhodovulum sp. PS88, produces RNA not only in cells but also as an extracellular polymeric substance during aerobic continuous cultivation in the dark. At a dilution rate of 0.32–0.5 h–1, the maximum RNA production was 460 mg RNA l–1 broth (200 mg RNA g–1 suspended solids) which is a value about 2–3 times more than that of yeast cells.  相似文献

The feeding of propionic acid for production of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) [P(3HB-co-3HV)] by Alcaligenes eutrophus ATCC17697 was optimized using a fed-batch culture system. The concentration of propionic acid was maintained at 3 g l–1 as growth was inhibited by propionic acid in the broth. A pH-stat substrate feeding system was used in which propionic acid was fed automatically to maintain a pH of the culture broth at 7.0. By feeding a substrate solution containing 20% (w/v) propionic acid, 4.9% (w/v) ammonia water [at a molar ratio of carbon to nitrogen (C/N molar ratio) of 10] in cell growth phase, the concentration of propionic acid in the broth was maintained at 3 g l–1 giving a specific growth rate of 0.4 h–1. To promote P(3HB-co-3HV) production, two stage fed-batch culture which consisted of the stage for the cell growth and the stage for the P(3HB-co-3HV) accumulation was carried out. When the substrate solution whose C/N molar ratio was 50 was fed in P(3HB-co-3HV) accumulation phase, the cell concentration and the P(3HB-co-3HV) content in the cells reached 64 g l–1 and 58% (w/w) in 55.5 h, respectively.  相似文献

Phaffia rhodozyma cells were treated with the mutagenic agent NTG several times and plated on yeast-malt agar containing -ionone as a selective medium. This mutagenesis of the yeast yielded a mutant (NCHU-FS501) with a total carotenoid content of 1454 g g–1 dry biomass. Temperature and pH had only a slight effect on the volumetric pigment production by the red yeast, however astaxanthin yield and specific growth rate were influenced more significantly by temperature and pH. The optimum inoculum size, temperature and air flow rate for astaxanthin formation by the mutant in a bench-top fermentor were 7.5% (v/v), 22.5°C and 3.6 vvm, respectively. Glucose (1%, w/v) as carbon source yielded the highest volumetric astaxanthin production (6.72 g ml–1). Peptone (15.8% total nitrogen) was the best nitrogen source for astaxanthin production (6.72 g ml–1). Pigment formation by the mutant was further improved by increasing the glucose concentration to 3.5%, where the astaxanthin concentration was 16.33 m ml–1. At 4.5% glucose or above astaxanthin formation was inhibited. Control of the pH of the fermentation broth did not improved pigment production.  相似文献

Aspergillus niger was explored, for the first time, for the production of 2-phenylethanol (a rose-like aroma) using L-phenylalanine as precursor. Among the strains screened, A. niger CMICC 298302 was shown to produce, in a culture medium containing 6 g L-phenylalanine l–1 and 60 g glucose l–1, 1375 mg 2-phenylethanol l–1 with a productivity of 153 mg l–1 day–1 and a molar yield of 74%. 2-Phenylethanol concentrations of 1 to 2 g l–1 led to a two-fold and ten-fold decrease, respectively, in the mycelial radial growth rate. However, 2-phenylethanol was synthesized as the sole aromatic product and accumulated in the culture broth.  相似文献

Pseudomonas oleovorans was cultivated to produce medium chain length polyhydroxyalkanoates (MCL-PHAs) from octanoic acid and ammonium nitrate as carbon and nitrogen source, respectively, by a pH-stat fed-batch culture technique. The octanoate in the culture broth was maintained below 4 g l–1 by feeding the mixture of octanoic acid and ammonium nitrate when the culture pH rose above 7.1. The final cell concentrations of 63, 55 and 9.5 g l–1, PHA contents of 62, 75 and 67% of dry cell wt, and productivities of 1, 0.63 and 0.16 g l–1 h–1 were obtained when the C/N ratios in the feed were 10, 20 and 100 g octanoic acid g–1 ammonium nitrate, respectively.  相似文献

A bacterial strain of Acinetobacter sp., which was capable of enzymatic production of pyruvate from lactate, was cultured in a 5-l reactor with a basal salt medium. After 14 h of fed-batch fermentation, 9.56 g l–1 cell concentration in the broth was obtained with 20 g l–1 (178 mM) sodium lactate and 4 g l–1 NH4Cl in the medium; and the biotransformation ability was 2.51 units ml–1. The cells were harvested from one reactor and then used for pyruvate production from lactate in the same reactor. l-lactate at a concentration about 527 mM was almost stoichiometrically converted to pyruvate in 28 h. After a total 42 h of cell culture and biotransformation, the transformative yield was about 0.72 g g–1 pyruvate from lactate and the rate of pyruvate production was calculated as 1.33 g l–1 h–1 during the process. The results suggested this simple enzymatic production of pyruvate from lactate should be a promising process and may bring a yield higher than that by microbial fermentation. By this process, the recovery of pyruvate from such a simple reaction liquid is relatively easy and inexpensive to perform.  相似文献

Summary Optimal growth conditions for Zymomonas mobilis have been established using continuous cultivation methods. Optimal substrate utilization efficiency occurs with 2.5 g l–1 yeast extract, 2.0 g l–1 ammonium sulfate and 6.0 g l–1 magnesium sulfate in the media. Catabolic activity is at its maximum with glucose uptake rates of 16–18 g l–1 h–1 and ethanol production rates of 8–9 g l–1 h–1, Qg values of 22–26 and Qp values between 11 and 13, which results in 40 g l–1 h–1 ethanol yields using a 100 g l–1 substrate feed. Any increase in these parameters goes on cost of substrate utilization efficiency. Acetone butanol ethanol (ABE) was produced in an integrated fed-batch fermentation-gas stripping product-recovery system using Clostridium beijerinckii BA101, with H2 and CO2 as the carrier gases. This technique was applied in order to eliminate the substrate and product inhibition that normally restricts ABE production and sugar utilization to less than 20 g l–1 and 60 g l–1, respectively. In the integrated fed-batch fermentation and product recovery system, solvent productivities were improved to 400% of the control batch fermentation productivities. In a control batch reactor, the culture used 45.4 g glucose l–1 and produced 17.6 g total solvents l–1 (yield 0.39 g g–1, productivity 0.29 g l–1 h–1). Using the integrated fermentation-gas stripping product-recovery system with CO2 and H2 as carrier gases, we carried out fed-batch fermentation experiments and measured various characteristics of the fermentation, including ABE production, selectivity, yield and productivity. The fed-batch reactor was operated for 201 h. At the end of the fermentation, an unusually high concentration of total acids (8.5 g l–1) was observed. A total of 500 g glucose was used to produce 232.8 g solvents (77.7 g acetone, 151.7 g butanol, 3.4 g ethanol) in 1 l culture broth. The average solvent yield and productivity were 0.47 g g–1 and 1.16 g l–1 h–1, respectively.  相似文献

Gallidermin production by Staphylococcus gallinarum was optimal using a new developed medium consisting of 50 g yeast extract l–1, 45 g CaCl2 l–1 and 5 g maltose l–1. Medium optimisation led to the cost of the medium decreasing by 93% compared to the previously used medium and in addition gained a higher product yield. A scale-up of the process based on the new medium was successfully tested up to a working volume of 200 l.  相似文献

Batch production of xylitol from the hydrolysate of wheat straw hemicellulose using Candida guilliermondii was carried out in a stirred tank reactor (agitation speed of 300 rpm, aeration rate of 0.6 vvm and initial cell concentration of 0.5 g l–1). After 54 h, xylitol production from 30.5 g xylose l–1 reached 27.5 g l–1, resulting in a xylose-to-xylitol bioconversion yield of 0.9 g g–1 and a productivity of 0.5 g l–1 h–1.  相似文献

Using lactose as an inducer, recombinant human interleukin-2 (rhIL-2) was synthesized with an N-terminus fusion partner, G3 (three tandem-arranged glucagon peptides) in fed-batch cultures at high cell concentration (60–90 g l–1) of Escherichia coli BL21(DE3) [pT7-G3IL2]. With batch additions of lactose (4 × 13.5 g), the fusion rhIL-2 was synthesized up to 9.3 g l–1. However, if all the lactose (54 g) was added at once to the culture, synthesized fusion rhIL-2 decreased to 5.4 g l–1 with a decreased cell growth rate. A statistical optimization of the production medium containing glucose, yeast extract, and lactose led to fusion rhIL-2 being produced at > 9 g l–1.  相似文献

Candida glycerinogenes is an aerobe which does not depend on sulphite for production of glycerol. With a sufficient O2 supply, up to 130 g glycerol l–1 was produced with 2.6 g acetic acid l–1 as by-product. However, with an insufficient O2 supply – with higher volumes of medium or at higher corn steep liquid concentrations – the glycerol concentration was lower because the by-products, ethanol, pyruvate and lactic acid, were produced in greater amounts, up to 45 g l–1, 4.3 g l–1, 1.6 g l–1, respectively, whereas, less acetic acid (0.6 g l–1) was produced. In addition, ethanol decreased to 0.4 g l–1 and the glycerol yield improved from 34 to 50% (w/w) by adding 50 g sulphite l–1, nevertheless, acetic acid increased to 7.8 g l–1.  相似文献

Summary Batch and continuous two-stage cultures have been conducted in order to determine the effect of yeast extract (YE) on the homolactic fermentation of whey permeate byLactobacillus helveticus. Supplementation with YE had a significant effet on lactic acid concentration, volumetric productivity, and substrate conversion, but not on lactic acid yield. Volumetric productivity in the first stage increased from 2 to 9 g l–1 per hour by increasing the YE concentration from 1.5 to 25 g l–1 At the same time conversion improved from 22% to 93% at a dilution rate of 0.2 h–1. The second stage demonstrated the effect of YE at a lower dilution rate (0.14 h–1. A high system conversion (97%) and a high final lactic acid concentration (40 g l–1) were achieved with 10 g l–1 YE.  相似文献

Poly(3-hydroxybutyrate) (PHB) was produced by fed-batch cultures of Ralstonia eutropha with phosphate limitation under different glucose concentrations. When glucose was kept at 2.5 g l–1, cell growth and PHB synthesis were limited due to the shortage of carbon source but a higher PHB content occurred in the cell-growth stage. This shows that a low glucose concentration is favorable for PHB accumulation in R. eutropha. PHB obtained with glucose at 9 g l–1 is 1.6 times that obtained with 40 g l–1. When glucose was in the range of 9 to 40 g l–1, PHB concentration and productivity decreased significantly with the increase of glucose concentration. The highest PHB productivity was obtained with glucose at 9 g l–1.  相似文献
