Abstract

C14O2 fixation in plant tissues « in vitro ». — In the present work it has been examinated the autotrophic and heterotrophic CO2 fixation of explants of « Helianthus tuberosus » « in vitro » and the photosyntetic efficiency of leaves produced from buds of « in vitro » explants of « Cichorium intybus » compared with that of mature leaves from normal plants of the same species. From our results it is evident that « in vitro » explants of « Helianthus tuberosus », grown, in the light, are able to autotrophically incorporate C14O2; the distribution of the radioactivity into the various fractions shows a large influence of the light on the neutral fraction containing sugars (50% of the total radioactivity). In the chlorophyllous explants the dark CO2 fixation is obviously of heterotrophic type: 97% of the total radioactivity is incorporated in amine acids (43%) and the organic acids (53%); on the other hand in the dark grown explants the radioactivity is differently distributed between amino acids (59%) and organic acids (39%). Mature leaves from normal plants and leaves produced from buds of « in vitro » explants of « Cichorium intybus » incorporate the same quantity of C14O2 when expressed per mg of chlorophyll; the different distribution of the radioactivity in the neutral and acid fractions could be explained in terms of a different utilization pathway of the photosynthates in the two tissues.  相似文献

The taxonomic position of strain DFH11T, which was isolated from coastal seawater off Qingdao, People’s Republic of China in 2007, was determined. Strain DFH11T comprised Gram-negative, motile, strictly aerobic spirilli that did not produce catalase. Comparative 16S rRNA gene sequence analysis revealed that strain DFH11T shared ~97.2, 93.3, 91.8, 91.7 and 91.5% sequence similarities with Oleispira antarctica, Spongiispira norvegica, Bermanella marisrubri, Oceaniserpentilla haliotis and Reinekea aestuarii, respectively. DNA–DNA hybridization experiments indicated that the strain was distinct from its closest phylogenetic neighbour, O. antarctica. The strain grew optimally in 2–3% (w/v) NaCl, at pH 5.0–10.0 (optimally at pH 7.0) and between 0 and 30°C (optimum growth temperature 28°C). The strain exhibited a restricted substrate profile, with a preference for aliphatic hydrocarbons, that is consistent with its closest phylogenetic neighbour O. antarctica. Growth of the isolate at different temperatures affected the cellular fatty acid profile. 28°C cultured cells contained C16:1ω7c and/or iso-C15:0 2-OH (50.4%) and C16:0 (19.2%) as the major fatty acids. However, the major fatty acids of the cells cultured at 4°C were C16:1ω7c and/or C16:1ω6c (40.2%), C16:0 (17.2%) and C17:1ω8c (10.1%). The G+C content of the genomic DNA was 42.7 mol%. Phylogeny based on 16S rRNA gene sequences together with data from DNA–DNA hybridization, phenotypic and chemotaxonomic characterization revealed that DFH11T should be classified as a novel species of the genus Oleispira, for which the name Oleispira lenta sp. nov. is proposed, with the type strain DFH11T (=NCIMB 14529T = LMG 24829T).  相似文献

Forty-five fungal species and two varieties belonging to the 23 genera were collected from 50 samples of diseased leaves of broad bean collected from Qena Governorate in Egypt on glucose-Czapek’s, dichloran-chloramphenicol-malt extract and dichloran-chloramphenicol-peptone agar at 28 °C. The most common genera were Alternaria, Aspergillus, Cladosporium, Fusarium, Mycosphaerella and Penicillium. The most prevalent species were Alternaria alternata, Aspergillus flavus, A. fumigatus, A. niger, Cladosporium cladosporioides, Fusarium merismoides, Mycosphaerella tassiana and Penicillium chrysogenum. Among the eight dematiaceous Hyphomycetes phytopathogenic fungi screened for their abilities to produce both exo- and endo-β-1,4-glucanase enzymes (C1 and Cx), five species had high C1 enzyme production and two species had high Cx enzyme production. However, two and three species were moderately active in producing C1 and Cx enzymes, respectively. The remaining species had low production of both C1 and Cx enzymes. The highest cellulase activity was recorded by Alternaria citri and Cochliobolus spicifer for C1 and by A alternata and A. citri for Cx enzyme. Maximum production of C1 enzyme by A. citri and C. spicifer was obtained after six days of incubation at 30 °C with an initial pH of 6 in a culture medium containing lactose and calcium nitrate as the carbon and nitrogen sources, respectively. The highest Cx enzyme production was by A. alternata and A. citri,which was recorded after eight days of incubation at 30 °C with an initial pH of 6 in a culture medium containing carboxymethyl cellulose and sodium or calcium nitrate as the carbon and nitrogen sources, respectively.  相似文献

The oleaginous yeast Rhodotorula glutinis has been known to be a potential feedstock for lipid production. In the present study, we investigated the enhancement of expression of malic enzyme (ME; NADP+ dependent; EC 1.1.1.40) from Mucor circinelloides as a strategy to improve lipid content inside the yeast cells. The 26S rDNA and 5.8S rDNA gene fragments isolated from Rhodotorula glutinis were used for homologous integration of ME gene into R. glutinis chromosome under the control of the constitutively highly expressed gene phosphoglycerate kinase 1 to achieve stable expression. We demonstrated that by increasing the expression of the foreign ME gene in R. glutinis, we successfully improved the lipid content by more than twofold. At the end of lipid accumulation phrase (96 h) in the transformants, activity of ME was increased by twofold and lipid content of the yeast cells was increased from 18.74 % of the biomass to 39.35 %. Simultaneously, there were no significant differences in fatty acid profiles between the wild-type strain and the recombinant strain. Over 94 % of total fatty acids were C16:0, C18:0, C16:1, C18:1, and C18:2. Our results indicated that heterologous expression of NADP+-dependent ME involved in fatty acid biosynthesis indeed increased the lipid accumulation in the oleaginous yeast R. glutinis.  相似文献

Increasing demand for petroleum has stimulated industry to develop sustainable production of chemicals and biofuels using microbial cell factories. Fatty acids of chain lengths from C6 to C16 are propitious intermediates for the catalytic synthesis of industrial chemicals and diesel‐like biofuels. The abundance of genetic information available for Escherichia coli and specifically, fatty acid metabolism in E. coli, supports this bacterium as a promising host for engineering a biocatalyst for the microbial production of fatty acids. Recent successes rooted in different features of systems metabolic engineering in the strain design of high‐yielding medium chain fatty acid producing E. coli strains provide an emerging case study of design methods for effective strain design. Classical metabolic engineering and synthetic biology approaches enabled different and distinct design paths towards a high‐yielding strain. Here we highlight a rational strain design process in systems biology, an integrated computational and experimental approach for carboxylic acid production, as an alternative method. Additional challenges inherent in achieving an optimal strain for commercialization of medium chain‐length fatty acids will likely require a collection of strategies from systems metabolic engineering. Not only will the continued advancement in systems metabolic engineering result in these highly productive strains more quickly, this knowledge will extend more rapidly the carboxylic acid platform to the microbial production of carboxylic acids with alternate chain‐lengths and functionalities. An actinomycete producing oil‐like mixtures was isolated and characterized. The strain was isolated from sheep faeces and identified as Streptomyces sp. S161 based on 16S rRNA gene sequence analysis. The strain showed cellulase and xylanase activities. The 1H nuclear magnetic resonance (NMR) spectra of the mixtures showed that the mixtures were composed of fatty acid methyl esters (52·5), triglycerides (13·7) and monoglycerides (9·1) (mol.%). Based on the gas chromatography–mass spectrometry (GC‐MS) analysis, the fatty acid methyl esters were mainly composed of C14‐C16 long‐chain fatty acids. The results indicated that Streptomyces sp. S161 could produce fatty acid methyl esters (FAME) directly from starch. To our knowledge, this is the first isolated strain that can produce biodiesel (FAME) directly from starch.

Significance and Impact of the Study

Nowadays, production of biodiesel is based on plant oils, animal fats, algal oils and microbial oils. Lipid mostly consists of triacylglycerols (TAG), and conversion of these lipids into fatty acid short‐chain alcohol esters (methanol or ethanol) is the final step in biodiesel production. In this study, an oil‐producing Streptomyces strain was isolated from sheep faeces. The oil was composed of C14‐C16 long‐chain fatty acid methyl esters, triglycerides and monoglycerides. This is the first isolated strain‐producing biodiesel (FAME) directly from starch. Due to showing cellulase and xylanase activities, the strain would be helpful for converting renewable lignocellulose into biodiesel directly.  相似文献

A novel actinomycete, designated strain KLBMP 4601T, was isolated from the root of the medicinal plant Curcuma phaeocaulis collected from Sichuan Province, south-west China. The strain produced extensively branched substrate and aerial hyphae that carried straight to flexuous spore chains. Chemotaxonomic properties of this strain were consistent with those of members of the genus Streptomyces. The cell wall of strain KLBMP 4601T contained ll-diaminopimelic acid as the characteristic diamino acid. The major menaquinone was MK-9(H4), with minor amounts of MK-9(H6), MK-9(H8) and MK-10(H2). The major fatty acids were C16:0, iso-C16:0, C18:1ω9c and C16:1, iso G. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain KLBMP 4601T belongs to the genus Streptomyces and is most closely related to Streptomyces armeniacus JCM 3070T (97.9 %), Streptomyces pharmamarensis PM267T (97.6 %) and Streptomyces artemisiae YIM 63135T (97.5 %). The 16S rRNA gene sequence similarity between strain KLBMP 4601T and other members of this genus were lower than 97.5 %. DNA–DNA hybridization studies of strain KLBMP 4601T with the three closest species showed relatedness values of 36.3 ± 4.2 %, 27.3 ± 0.6 %, and 30.9 ± 2.5 %, respectively. On the basis of chemotaxonomic, phenotypic and genotypic characteristics, it is evident that strain KLBMP 4601T represents a novel species of the genus Streptomyces, for which the name Streptomyces phytohabitans sp. nov. is proposed. Aims: In this work, fatty acid content and profiles were analysed in order to differentiate the species Tenacibaculum maritimum, Tenacibaculum gallaicum, Tenacibaculum discolor and Tenacibaculum ovolyticum that are pathogenic for cultured marine fish and to assess the potential of fatty acid profiles as a tool for epizootiological typing. Methods and Results: The fatty acid methylesters (FAMEs) were extracted from cells grown on marine agar for 48 h at 25°C and were prepared and analysed according to the standard protocol of the MIDI/Hewlett Packard Microbial Identification System. The cellular fatty acid profiles of Tenacibaculum strains tested were characterized by the presence of large amounts of branched (36·1–40·2%) and hydroxylated (29·6–31·7%) fatty acids. The FAME products from the four species significantly (P < 0·05) differed in the content of iso‐C15:03‐OH, iso‐C16:03‐OH, iso‐C15:1G, summed feature 3 (a component that contains C16:1ω7c and/or iso‐C15:0 2‐OH), iso‐C16:0, C17:1ω6c, C15:03‐OH, iso‐C17:03‐OH. Conclusions: Results of present study demonstrated the existence of differences in the fatty acids content between the T. maritimum isolates from different marine fish/geographical origin and between strains of T. maritimum, T. discolor, T. gallaicum and T. ovolyticum. Significance and Impact of the Study: Profiling of fatty acids may be a useful tool to distinguish T. maritimum from other Tenacibaculum species pathogenic for fish as well as for epizootiological differentiation of T. maritimum isolates.  相似文献

The carotenoid synthetic genes, crtM and crtN, derived from Staphylococcus aureus, were introduced into B. subtilis, resulting in yellow pigmentation. Absorption maxima of pigments and MALDI-TOF mass spectrometry demonstrated that the pigmented strain accumulated two C30 carotenoids, 4,4′-diapolycopene and 4,4′-diaponeurosporene. A survival test using H2O2 revealed that the pigmented strain was more resistant to oxidative stress than the strain harboring an empty-vector. These findings indicate that B. subtilis can produce carotenoids, and the strain accumulating the carotenoids, CarotenoBacillus, will become a basal host for production of C30 carotenoids and evaluation of their antioxidative effects.  相似文献

Bacterial strain Eg2T, an anaerobic, Gram-positive, non-motile, and non-spore-forming coccus, was isolated from human faeces. The optimal temperature for its growth was 37°C. Oxidase activity was negative, but catalase activity was positive. The strain was able to hydrolyze esculin and to produce acids from the fermentation of several substrates, including glucose. Lactic and acetic acids were the main products of glucose fermentation. The major fatty acids present in this strain were C16:0, C14:0, and C18:1 cis11 DMA. The G+C content was 43.4 mol%. Based on the 16S rRNA gene sequence, strain Eg2T was closely related to species of the genus Ruminococcus (96.3% similarity to R. torques and 96.2% similarity to R. lactaris), and its taxonomic position was placed within the Clostridium cluster XIVa. Based on phenotypic, chemotaxonomic, genotypic, and phylogenetic evidence, we propose that this novel strain be assigned to the genus Ruminococcus and be named Ruminococcus faecis sp. nov. The type strain is Eg2T (=KCTC 5757T =JCM 15917T).  相似文献

A moderate halophilic, Gram-negative, non-motile, rod-shape, and aerobe designated as strain HwaT was isolated from traditional fermented Korean seafood, which presented as a single cell or paired cells. Optimal growth occurred at 25°C in 10% (w/v) salts at pH 7.0–8.0; however, growth occurred in a temperature range of 10–32°C, a salts concentration of 5–25% (w/v) and pH 5.0–10.0. Tests for oxidase and catalase were positive. The cells produced poly-β-hydroxybutyric acid, but not exopolysaccharide. Based on the 16S rRNA gene sequence, not only was there low similarity between strain HwaT and all other species (94.1% similarity with H. subglaciescola DSM 4683T, 94.0% similarity with H. sulfidaeris Esulfide1T, 93.6% similarity with H. cerina SP4T and 93.0% similarity with H. halodurans DSM 5160T), but the phylogenetic analysis revealed that the isolate may be classified as a novel species belonging to the genus Halomonas in the class Gammaproteobacteria. The predominant fatty acids of strain HwaT were C18:1 ω7c, C16:0, C12:0 3-OH and C16:1 ω7c/C15:0 iso 2-OH. The DNA G+C content was calculated as 61.7 mol%. Based on phenotypic, genotypic, and phylogenetic characteristics, it is proposed that the strain designated as HwaT be assigned to the genus Halomonas as Halomonas jeotgali sp. nov. (=KCTC 22487T =JCM 15645T).  相似文献

A novel Gram-positive, strictly anaerobic, spore-forming, rod-shaped bacterium, designated strain S11-3-10T, was isolated from the pit mud used for Chinese Luzhou-flavor liquor production. Phylogenetic analysis based on 16S rRNA gene sequencing revealed that the strain formed a monophyletic clade with the closely related type strains of Clostridium cluster I and was most closely related to Clostridium amylolyticum JCM 14823T (94.38 %). The temperature, pH, and NaCl range for growth was determined to be 20–45 °C (optimum 37 °C), 4.0–10.0 (optimum pH 7.3), and 0–3.0 % (w/v), respectively. The strain was able to tolerate up to 7.5 % (v/v) ethanol. Yeast extract or peptone was found to be required for growth. Acids were found to be produced from glucose, mannose and trehalose. The major end products from glucose fermentation were identified as ethanol, acetate and hydrogen. The polar lipids were found to consist of diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol and unidentified phospholipids and polar lipids. The major fatty acids (>5 %) were identified as iso-C15:0, C16:0, C16:0 dma, C14:0, anteiso-C15:0 and iso-C13:0. No respiratory quinone was detected. The diamino acid in the cell wall peptidoglycan was identified as meso-diaminopimelic acid and the whole-cell sugars were found to include galactose and glucose as major components. The DNA G+C content was determined to be 36.4 mol%. Based on the phylogenetic, chemotaxonomic and phenotypic evidence, the isolate is considered to represent a novel species of the genus Clostridium for which the name Clostridium swellfunianum sp. nov. is proposed. The development of renewable alternatives to diesel and jet fuels is highly desirable for the heavy transportation sector, and would offer benefits over the production and use of short‐chain alcohols for personal transportation. Here, we report the development of a metabolically engineered strain of Escherichia coli that overproduces medium‐chain length fatty acids via three basic modifications: elimination of β‐oxidation, overexpression of the four subunits of acetyl‐CoA carboxylase, and expression of a plant acyl–acyl carrier protein (ACP) thioesterase from Umbellularia californica (BTE). The expression level of BTE was optimized by comparing fatty acid production from strains harboring BTE on plasmids with four different copy numbers. Expression of BTE from low copy number plasmids resulted in the highest fatty acid production. Up to a seven‐fold increase in total fatty acid production was observed in engineered strains over a negative control strain (lacking β‐oxidation), with a composition dominated by C12 and C14 saturated and unsaturated fatty acids. Next, a strategy for producing undecane via a combination of biotechnology and heterogeneous catalysis is demonstrated. Fatty acids were extracted from a culture of an overproducing strain into an alkane phase and fed to a Pd/C plug flow reactor, where the extracted fatty acids were decarboxylated into saturated alkanes. The result is an enriched alkane stream that can be recycled for continuous extractions. Complete conversion of C12 fatty acids extracted from culture to alkanes has been demonstrated yielding a concentration of 0.44 g L?1 (culture volume) undecane. Biotechnol. We demonstrate the tolerance of bacterial strain SM2014 to various unsustainable conditions and suggest its implication in waste water management. Its sustainability to reverse osmosis pressure (2.1 MPa) during desalination, and survival percentage of 73 % under hyperbaric conditions (pressure tension of 3.1 MPa under absolute oxygen atmosphere) confirmed its pressure tolerance. The growth of this strain at pH 9 or 10 and at 60 °C alone or in combination revealed its unique physiology as poly-extremotolerant strain. As an adaptive mechanism, the ratio of saturated to unsaturated fatty acids changed with growth conditions. Under poly-extreme condition long chain saturated fatty acid (C18:0, C16:0, C14:0, C12:0) predominated at the expense of unsaturated fatty acids. The nucleotide BLAST of 16S rRNA gene sequence of strain SM2014 with the NCBI gene bank sequences showed its close identity to Bacillus licheniformis with a similarity match of 94 %. The secretion of industrially valuable enzymes proteinase, lipase and amylase under such harsh conditions further signified potential of this strain as a source of extremozymes. Its unique characteristics underscore its relevance in waste water management.  相似文献

A novel endophytic actinomycete, designated strain NEAU-J3T, was isolated from soybean root (Glycine max (L.) Merr) and characterized using a polyphasic approach. Phylogenetic analysis based on 16S rRNA gene sequences suggested that strain NEAU-J3T fell within the family Micromonosporaceae. The strain was observed to form an extensively branched substrate mycelium, which carried non-motile oval spores with a smooth surface. The cell walls of strain NEAU-J3T were determined to contain meso-diaminopimelic acid and galactose, ribose and glucose were detected as whole-cell sugars. The major menaquinones were determined to be MK-9(H4) and MK-9(H6). The phospholipids detected were phosphatidylcholine and phosphatidylethanolamine. The major cellular fatty acids were determined to be C16:0, C18:1 ω9c, C18:0, C17:0, C17:1 ω7c, anteiso-C17:0, C16:1 ω7c and C15:0. The DNA G + C content was 62.5 mol%. On the basis of the morphological and chemotaxonomic characteristics, phylogenetic analysis and characteristic patterns of 16S rRNA gene signature nucleotides, strain NEAU-J3T is considered to represent a novel species of a new genus within the family Micromonosporaceae, for which the name Wangella harbinensis gen. nov., sp. nov. is proposed. The type strain of Wangella harbinensis is strain NEAU-J3T (=CGMCC 4.7039T = DSM 45747T).  相似文献

Marine microbes are a rich source of bioactive compounds, such as drugs, enzymes, and biosurfactants. To explore the bioactive compounds from our marine natural product library, an oil emulsification assay was applied to discover biosurfactants and bioemulsifiers. A spore-forming bacterial strain from sea mud was found to produce bioemulsifiers with good biosurfactant activity and a broad spectrum of antimicrobial properties. It was identified as Bacillus velezensis H3 using genomic and phenotypic data analysis. This strain was able to produce biosurfactants with an optimum emulsification activity at pH 6.0 and 2% NaCl by using starch as the carbon source and ammonium sulfate as the nitrogen source. The emulsification-guided isolation and purification procedure led to the discovery of the biosurfactant components, which were mainly composed of nC14-surfactin and anteisoC15-surfactin as determined by NMR and MS spectra. These compounds can reduce the surface tension of phosphate-buffered saline (PBS) from 71.8 to 24.8 mN/m. The critical micelle concentrations (CMCs) of C14-surfactin and C15-surfactin in 0.1 M PBS (pH 8.0) were determined to be 3.06?×?10-5 and 2.03?×?10-5?mol/L, respectively. The surface tension values at CMCs for C14-surfactin and C15-surfactin were 25.7 and 27.0 mM/m, respectively. In addition, the H3 biosurfactant exhibited antimicrobial activities against Staphyloccocus aureus, Mycobacterium, Klebsiella peneumoniae, Pseudomonas aeruginosa, and Candida albicans. Thus B. velezensis H3 is an alternative surfactin producer with potential application as an industrial strain for the lipopeptide production.  相似文献

A marine microalga, strain JPCC GA0024 was selected as high amount of neutral lipid producers from marine microalgal culture collection toward biofuel production. The strain was tentatively identified as Scenedesmus rubescens by 18S rDNA analysis. The growth of strain JPCC GA0024 was influenced by artificial seawater concentrations. The optimum growth of 0.79 g/l was obtained at 100% artificial seawater. The lipid accumulation reached 73.0% of dry cell weight at 100% artificial seawater without additional nutrients for 11 days. Gas chromatography/mass spectrometry analysis indicates that lipid fraction mainly contained hydrocarbons including mainly hexadecane (C16 H34) and 1-docosene (C22 H44). Furthermore, calorimetric analysis revealed that the energy content of strain JPCC GA0024 was 6,160 kcal/kg (25.8 MJ/kg) of calorific value, which was equivalent to the coal engery. The strain JPCC GA0024, S. rubescens, will become a promising resource that can grow as a dominant species in the seawater for the production of both liquid and solid biofuels.  相似文献

The intensitive investigations on the lipid profile of Thiobacillus ferrooxidans at various culture ages suggest some correlations of the lipid constitutents with the membrane-bound iron oxidation system. Phosphatidic acid, phosphatidyl serine and phosphatidyl ethanolamine were the major polar components; hydrocarbon, triglyceride and diglyceride were the main neutral components. Major fatty acids were C16:0, C16:1, C16:3, C18:1, C18:3, C22:1 while C20:1, C20:2, C12:0, C14:2, C18:0, C18:2, C20:0, C22:0 were found in trace amounts which also depended upon the phase of the growth. One lipoamino acid was identified as ornithine lipid in the polar fraction. Each and every component varied to some extent at different growth phasesindicating relationship of these lipids to the iron oxidation system of the strain.  相似文献

The accumulation of vitamin B6 by Pichia guilliermondii Wickerham NK–2 strain grown on hydrocarbon was investigated. Ammonium acetate was more effective than other nitrogen sources tested. Satisfactory utilization by the yeast strain was observed in n-alkanes of C10–C18, and n-pentadecane was the best for vitamin B6 production. Vitamin B6 was excreted in the cultural broth mainly in the form of pyridoxal, The maximal vitamin B6 production was approximately 25 mg per liter of the culture broth.  相似文献

A novel Gram-negative, slightly halophilic, catalase- and oxidase-positive, obligately aerobic bacterium, strain YIM-C248T, was isolated from a sediment sample collected from a salt-lake in the Qaidam Basin in Qinghai, north-west China. Cells were non-sporulating short rods, occurring singly or as doublets, motile with peritrichous flagella. Growth occurred with 1–15% (w/v) NaCl [optimum 2–4% (w/v) NaCl], at pH 6.0–10.0 (optimum pH 7.5) and at 4–35°C (optimum 25–30°C). The major cellular fatty acids were C18:1 ω7c, C12:0 3-OH, cyclo C19:0 ω8c, C16:0 and C16:1. The predominant respiratory quinone was Q-9 and the genomic DNA G + C content was 58.6 mol%. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain YIM-C248T should be assigned to the genus Halomonas. The sequence similarities between the isolate and the type strains of members of the genus Halomonas were in the range of 92.5–97.5%. The combination of phylogenetic analysis, DNA–DNA hybridization data, phenotypic characteristics and chemotaxonomic differences supported the view that strain YIM-C248T represents a new species of the genus Halomonas, for which the name Halomonas sediminis sp. nov. is proposed, with YIM-C248T (=CCTCC AA 207031 = KCTC 22167) as the type strain.
