The seasonal and diurnal variations in nitrate reductase activity (NRA) of cowpea cultivars were determined. Mean activity per gram fresh weight per hour averaged 31% higher in the 20 mg N/plant treatment than the control throughout the growing period. The highest activity occurred in the seedling stage and declined towards anthesis. Diurnal variations were marked by an increase in the NRA from 0600 to 1800 hours and then declined to a minimum at 2400 hours.  相似文献

Summary Nitrate reductase activity (NRA), nitrate content and biomass components of leaflets, leaf stalks, old stem, current-year stem and roots of ash trees (Fraxinus excelsior L.) growing in their natural habitats were investigated. In addition, NRA, total nitrogen and nitrate concentration were analyzed in the leaves and roots of ash trees from four different field sites. The highest NRA per gram biomass and also per total compartment biomass was found in the leaflets, even though root biomass was much higher than total leaflet biomass. The highest nitrate concentrations were found in the leaf stalks. Correlations between nitrate availability in the soil and NRA in leaves were not significant due to high variability of the actual soil nitrate concentrations. The seasonal variation in foliar NRA, nitrate concentration and total nitrogen concentration is much smaller in F. excelsior than reported for herbaceous species and is mainly caused by changes in the actual soil nitrate availability and by senescence of the leaves.  相似文献

Nitrate uptake, nitrate reductase activity (NRA) and net proton release were compared in five grain legumes grown at 0.2 and 2 mM nitrate in nutrient solution. Nitrate treatments, imposed on 22-d-old, fully nodulated plants, lasted for 21 d. Increasing nitrate supply did not significantly influence the growth of any of the species during the treatment, but yellow lupin (Lupinus luteus) had a higher growth rate than the other species examined. At 0.2 mM nitrate supply, nitrate uptake rates ranged from 0.6 to 1.5 mg N g(-1) d(-1) in the order: yellow lupin > field pea (Pisum sativum) > chickpea (Cicer arietinum) > narrow-leafed lupin (L angustifolius) > white lupin (L albus). At 2 mM nitrate supply, nitrate uptake ranged from 1.7 to 8.2 mg N g(-1) d(-1) in the order: field pea > chickpea > white lupin > yellow lupin > narrow-leafed lupin. Nitrate reductase activity increased with increased nitrate supply, with the majority of NRA being present in shoots. Field pea and chickpea had much higher shoot NRA than the three lupin species. When 0.2 mM nitrate was supplied, narrow-leafed lupinreleased the most H+ per unit root biomass per day, followed by yellow lupin, white lupin, field pea and chickpea. At 2 mM nitrate, narrow-leafed lupin and yellow lupin showed net proton release, whereas the other species, especially field pea, showed net OH- release. Irrespective of legume species and nitrate supply, proton release was negatively correlated with nitrate uptake and NRA in shoots, but not with NRA in roots.  相似文献

Nitrate reductase activity of soybeans (Glycine max L. Merr.) was evaluated in soil plots and outdoor hydroponic gravel culture systems throughout the growing season. Nitrate reductase profiles within the plant canopy were also established. Mean activity per gram fresh weight per hour of the entire plant canopy was highest in the seedling stage while total activity (activity per gram fresh weight per hour times the total leaf weight) reached a maximum when plants were in the full bloom to midpod fill stage. Nitrate reductase activity per gram fresh weight per hour was highest in the uppermost leaf just prior to full expansion and declined with leaf position lower in the canopy. Total nitrate reductase activity per leaf was also highest in the upper-most fully expanded leaf during early growth stages. Maximum total activity shifted to leaf positions lower in the plant canopy with later growth stages.  相似文献

Nitrate reductase activities (NRA) and nitrate concentration per unit biomass in Picea abies (L.) Karst. roots from four different soil horizons and in leaves and roots of the frequent field-layer species Oxalis acetosella L. were measured on six different irrigation and liming treatments within the Höglwald project, S-Bavaria, Germany. Liming increased and acid irrigation reduced soil nitrate availability when compared to control plots. Nitrate assimilation capacities of the respective plant compartments per unit of soil volume or ground area were calculated from the NRA per unit of biomass and from the biomass distribution on the various treatments.Mean NRA per unit of biomass in Picea abies roots ranged between 0.23 and 0.09 mol NO 2 - g-1 d.w. h-1 without significant effects of soil horizon or treatment. Limed and non-limed treatments showed for Picea different root distributions within the soil profile, but root biomass per unit of ground area (295 to 220 g d.w. m-2) was not affected by the various treatments. Thus, nitrate assimilation capacity of Picea roots per unit of ground area ranged between 19.5 and 11.4 mol NO 2 - m-2 h-1 without major treatment effects.In laminae of Oxalis acetosella mean NRA per unit of biomass ranged between 2.91 and 0.27 mol NO 2 - g-1 d.w. h-1 and, in contrast to Picea abies, treatment effects were found with NRA on limed plots increased and on acid irrigated plots reduced when compared to control plots. Mean leaf biomass of Oxalis per unit of ground area ranged between 9.57 and 0.66 g d.w. m-2 and responded in a similar manner to the various treatments. Thus, for the Oxalis leaf NRA per unit of ground area (27.85 to 0.18 mol NO2 m-2 h-1) a cumulative response to the variations in nitrate availability was found.The different responses of Picea abies and Oxalis acetosella to changes in soil nitrate availability are discussed with respect to their suitability to prevent soil nitrate leaching.  相似文献

Summary Using a continuous flow technique the relationship between growth rate and substrate concentration was investigated with glucose as the limiting factor of a culture of Escherichia coli. Graphical and numerical analysis of the experimental data demonstrated that the application of the Michaelis-Menten equation produced erroneous results, whereas, the constants obtained from the Teissier equation were in agreement with the experimental data. On this basis, new equations defining the steady state cell and substrate concentration in continuous flow cultures were developed and tested against experimental data.Comparison of the specific growth rates, substrate uptake rates and oxygen consumption rates demonstrated that all were directly proportional to each other and could be related to each other by mathematical equations. Specifically it was shown that as the growth rate increased from 0.06 to k m =0.76 the substrate uptake rate increased from 134 to 1420 mg glucose per gram cell weight per hour and the oxygen consumption rate increased from 48.6 to 505 mg O2 per gram cell weight per hour. Independent of the growth rate 37% of the carbohydrate consumed were oxidized. The yield factor varied from 0.44 at low growth rates to 0.54 at high growth rates. Analysis of the growth rate-substrate uptake rate relationship indicated that a minimum substrate uptake rate of 55 mg glucose per gram cell weight per hour existed below which cell reproduction would cease. This was supported by the fact that steady state conditions could not be maintained in the culture at D values below 0.02 when the substrate supply rate decreased below 45 mg glucose per gram cell weight per hour.Material contained in this paper was submitted as a thesis in partial fulfillment of the requirements for the Ph. Levels of nitrate reductase activity (EC 1.9.6.1.) as high as 11 μmoles nitrite produced/hour gram fresh weight were found in barley (Hordeum vulgare cv. Compana) roots grown under low oxygen conditions. Roots of plants given identical treatment under sterile conditions did not develop the high levels of nitrate reductase activity. The results suggest that the buildup of particulate, reduced viologen-utilizing nitrate reductase reported in barley roots may be caused by bacterial contamination. The nitrate reductase activity in roots grown under low oxygen conditions was not specific for reduced nicotinamide adenine dinucleotide like the assimilatory nitrate reductase (EC 1.6.6.1.) normally found in aerated plant roots.  相似文献

Activities of nitrate reduction enzymes, nitrate reductase activity (NRA) and nitrite reductase activity (NiRA) from roots and nodules of 5 mutant genotypes and one commercial cultivar (Alameda) of faba bean ( Vicia faba L. var. minor) grown in the presence of N2 alone or with additional NO3 in the medium have been studied. A naturally occurring mutant (VFM109) with impaired ability to reduce nitrate in its nodules is described. All the other cultivars of V. faba showed nodule NRA, although the range was very wide, from almost negligible (VFM72) up to 2 μmol h−1 (g FW)−1. This activity was entirely of plant origin. Root NRA also ranged widely accross cultivars. However, the level of activity expressed as well as the response of NRA to nitrate followed a pattern opposite to that observed in nodules. Roots and nodules of all cultivars showed very high rates of NiRA, respectively 50 and 150-fold higher than NRA, thus precluding accumulation of nitrite in these tissues. Root enzymes were significantly stimulated by nitrate while negative (NRA) or little effect (NiRA) was found for nodules. Nitrate and nitrite reduction are carried out by inducible enzymes in roots of V. faba and by constitutive enzymes in nodules, indicating that there may be different forms of these enzymes in each tissue. Differences in the plant genotype were a major cause of the variability in nitrate and nitrite reduction by nodulated root systems of V. faba .  相似文献

Abstract. Composition and density of the soil seed banks, together with seedling emergence in the field, were examined on Svalbard. 1213 soil samples were collected from six drymesic habitats in three regions representing various stages of colonization from bare moraines to full vegetation cover and spanning a range of typical nutrient and thermal regimes. Of the 165 vascular plant species native to Svalbard, 72 were present as mature plants at the study sites and of these 70% germinated seed. Proglacial soil had 12 seedlings per m2, disturbed Dryas heath 131, intact Dryas heath 91, polar heath 715, thermophilic heath 3113, and a bird cliff 10437 seedlings. Highest seed bank species richness was at the thermophilic heath (26 species). Seedlings of 27 species emerged in the field, with fewer seedlings in disturbed habitats (60 seedlings per m2) than in intact Dryas heath (142), suggesting that an absence of ‘safe sites’ limited seedling establishment in disturbed habitats. Measurement of seedling emergence in the field increased awareness of which species are able to germinate naturally. This may be underestimated by up to 31% if greenhouse trials alone are used, owing partly to unsuitability of greenhouse conditions for germination of some species and also to practical limitations of amount of soil sampled. Most thermophilic species failed to germinate and some species present at several sites only germinated from the thermophilic heath seed bank, suggesting that climate constrains recruitment from seeds in the High Arctic.  相似文献

It is not known why some species accumulate high concentrations of sucrose in leaves during photosynthesis while others do not. To determine the possible basis, we have studied 10 species, known to differ in the accumulation of sucrose, in terms of activities of sucrose hydrolyzing enzymes. In general, acid invertase activity decreased as leaves expanded; however, activities remaining in mature, fully expanded leaves ranged from low (<10 micromoles per gram fresh weight per hour) to very high (>100 micromoles per gram fresh weight per hour). In contrast, sucrose synthase activities were low and relatively similar among the species (4-10 micromoles per gram fresh weight per hour). Importantly, leaf sucrose concentration, measured at midafternoon, was negatively correlated with acid invertase activity. We propose that sucrose accumulation in vacuoles of species such as soybean and tobacco is prevented by acid invertase-mediated hydrolysis. Initial attempts were made to characterize the relatively high activity of acid invertase from mature soybean leaves. Two apparent forms of the enzyme were resolved by Mono Q chromatography. The two forms had similar affinity for substrate (apparent Km [sucrose] = 3 millimolar) and did not interconvert upon rechromatography. It appeared that the loss of whole leaf invertase activity during expansion was largely the result of changes in one of the enzyme forms. Overall, the results provide a mechanism to explain why some species do not accumulate sucrose in their leaves. Some futile cycling between sucrose and hexose sugars is postulated to occur in these species, and thus, the energy cost of sucrose production may be higher than is generally thought.  相似文献

A simple, rapid, accurate and sensitive method is proposed for the simultaneous determination of nitrite and nitrate in human saliva. Nitrite and nitrate present in the human saliva were determined after 10- to 100-fold dilution with ion chromatography (IC) using suppressed conductivity detection. Recoveries of nitrite and nitrate were found to be ranged between 95% and 101%. The method was linear (r2=0.9991) over the concentration working range. The detection limits were found to be 15.0 μg/l and 33.5 μg/l, for nitrite and nitrate, respectively. Ions that are present in human saliva and several other ions that are suspected to affect nitrite and nitrate determination were checked. It was found that most of the ions did not cause any interference in the determination. The method allows simultaneous determination of nitrite and nitrate in human saliva.  相似文献

Aims For many terrestrial plants, nitrate is the most important form of available soil nitrogen for growth. However, many plant species, which grow on acidic, ammonium-dominated soils, exhibit a constantly low level of nitrate reductase activity (NRA). Little is known about NRA in high-mountain vascular plants in similar conditions. Abstract. The wet to moist bryophyte‐dominated vegetation of Sassendalen, Svalbard, was classified into seven communities. These communities were grouped into (1) Cardamino nymanii‐Saxifragion foliolosae marsh; (2) Caricion stantis fen; (3) Luzulion nivalis snowbed – including manured vegetation corresponding to moss tundras. All communities have a basically arctic distribution. Marshes are developed in habitats with a water table above the bryophyte vegetation surface and fens on sites with a water table level high above the permafrost but below the bryophyte surface. Moss tundras normally have no standing water table, but in Sassendalen they have a low water table due to their development on less steep slopes than in their normal habitat near bird cliffs. CCA confirms that the standing water level is the prime differentiating factor between the alliances, while aspect favourability and permafrost depth differentiate between the fen communities and temporary desiccation is important for the Catoscopium nigritum community. Carex subspathacea is a characteristic fen species in the absence of other Carex species dominating elsewhere in the Arctic. Arctic marshes are linked to an extremely cold environment. They have a very low species diversity with a few species dominating; Arctophila fulva, Pseudocalliergon trifarium, Scorpidium scorpioides and Warnstorfia tundrae are character species. Moss tundra as defined here appears to be restricted to Svalbard and, probably, neighbouring Novaya Zemlya. This may be due to the absence of rodents and the high seabird density, which is related to the mild sea currents reaching further to the north here and which implies manuring of surrounding ecosystems. Manuring in a very cold environment produces moss carpets with a thin active layer and accumulation of thick peat layers without a standing water level. In Sassendalen the role of arctic seabirds is replaced by Svalbard reindeer which are nonmigratory and are concentrated to favourable grazing areas where their manuring effect is intense. Their long‐term manuring effect probably explains the occurrence of moss tundras in this weakly rolling landscape where seabird colonies are absent.  相似文献

The synthesis and accumulation of nitrite has been suggested as a causative factor in the inhibition of legume nodules supplied with nitrate. Plants were grown in sand culture with a moderate level of nitrate (2.1 to 6.4 millimolar) supplied continuously from seed germination to 30 to 50 days after planting. In a comparison of nitrate treatments, a highly significant negative correlation between nitrite concentration in soybean (Glycine max [L.] Merr.) nodules and nodule fresh weight per shoot dry weight was found even when bacteroids lacked nitrate reductase (NR). However, in a comparison of two Rhizobium japonicum strains, there was only 12% as much nitrite in nodules formed by NRR. japonicum as in nodules formed by NR+R. japonicum, and growth and acetylene reduction activity of both types of nodules was about equally inhibited. In a comparison of eight other NR+ and NRR. japonicum strains, and a comparison of G. max, Phaseolus vulgaris, and Pisum sativum, the concentration of nitrite in nodules was unrelated to nodule weight per plant or to specific acetylene reduction activity. The very small concentration of nitrite found in P. vulgaris nodules (0.05 micrograms NO2-N per gram fresh weight) was probably below that required for the inhibition of nitrogenase based on published in vitro experiments, and yet the specific acetylene reduction activity was inhibited 83% by nitrate. The overall results do not support the idea that nitrite plays a role in the inhibition of nodule growth and nitrogenase activity by nitrate.  相似文献

Background : The Snowy Mountains contain Australia's longest-lasting snowpatches. Because of climate change, their longevity has declined, with the loss of some specialist vegetation in the underlying snowbeds.

Aims: To characterise the current status of the vegetation associated with the longest-lasting snowpatches in Australia and its association with abiotic factors.

Methods: We assessed plant composition, soil depth, moisture and nutrients and subsurface temperatures in five zones of increasing vegetation height and cover in snowbeds.

Results: The zone beneath the middle of snowpatches was characterised by little vegetation cover and lower species richness, later emergence from snow, skeletal soils, and lower mean soil temperatures than zones further downslope where soils increased in depth and nutrient levels. Vegetation beneath these snowpatches no longer occurs in distinct communities. Plants have not simply migrated upslope, instead, areas that have deep soil that used to have snowpatch specialist species are being colonised upslope by grasses and downslope by tall alpine herbfield species that prefer bare ground.

Conclusions: Reduced longevity of Australia's longest-lasting snowpatches has led to the loss of distinct snowpatch plant communities. With limited soils beneath the centre of current snowpatches, and a lack of other suitable sites there is no location for these plant communities to migrate to.  相似文献

The activity of the enzyme nitrate reductase (NRA) was measuredin natural populations of phytoplankton in the Oslofjord, Norway,and in culture experiments with 10 different species of phytoplankton.The NRA values from the culture experiments and from the naturalsamples were of the same order of magnitude. The NRA values,when expressed on a chlorophyll a basis, were highly species-variable.In general they were correlated to the nitrogen nutrient concentrationsin the water. In order to use NRA as a guide to an understandingof the assimilation of nitrate, it is important to know thespecies composition of the phytoplankton.  相似文献

The green-fruited Lycopersicon hirsutum Humb. and Bonpl. accumulated sucrose to concentrations of about 118 micromoles per gram fresh weight during the final stages of development. In comparison, Lycopersicon esculentum Mill. cultivars contained less than 15 micromoles per gram fresh weight of sucrose at the ripe stage. Glucose and fructose levels remained relatively constant throughout development in L. hirsutum at 22 to 50 micromoles per gram fresh weight each. Starch content was low even at early stages of development, and declined further with development. Soluble acid invertase (EC 3.2. 1.26) activity declined concomitant with the rise in sucrose content. Acid invertase activity, which was solubilized in 1 molar NaCl (presumably cell-wall bound), remained constant throughout development (about 3 micromoles of reducing sugars (per gram fresh weight) per hour. Sucrose phosphate synthase (EC 2.4.1.14) activity was present at about 5 micromoles of sucrose (per gram fresh weight) per hour even at early stages of development, and increased sharply to about 40 micromoles of sucrose (per gram fresh weight) per hour at the final stages of development studied, parallel to the rise in sucrose content. In comparison, sucrose phosphate synthase activity in L. esculentum remained low throughout development. The possible roles of the sucrose metabolizing enzymes in determining sucrose accumulation are discussed.  相似文献

Summary Using dilution count methods the presumptive population of nitrate forming and nitrite forming organisms was estimated for 25 different fields taking monthly samples for 27 months. The results obtained would appear to indicate that the nitrite forming population was fairly constant with an approximate level of 106 to 107 per gram dry soil; that the level of nitrate formers was definitely more variable and that, in general, the nitrite forming population was about 10 to 100 times greater than the nitrate forming population. Neither population showed anything that could be regarded as a seasonal variation, but results only covered a period of 2 years.  相似文献

Aim Current evidence from temperate studies suggests that ectomycorrhizal (ECM) fungi require overland routes for migration because of their obligate symbiotic associations with woody plants. Despite their key roles in arctic ecosystems, the phylogenetic diversity and phylogeography of arctic ECM fungi remains little known. Here we assess the phylogenetic diversity of ECM communities in an isolated, formerly glaciated, high arctic archipelago, and provide explanations for their phylogeographic origins. Location Svalbard. Methods We generated and analysed internal transcribed spacer (ITS) nuclear ribosomal DNA sequences from both curated sporocarp collections (from Svalbard) and soil polymerase chain reaction (PCR) clone libraries (from Svalbard and the North American Arctic), compared these with publicly available sequences in GenBank, and estimated the phylogenetic diversity of ECM fungi in Svalbard. In addition, we conducted coalescent analyses to estimate migration rates in selected species. Results Despite Svalbard’s geographic isolation and arctic climate, its ECM fungi are surprisingly diverse, with at least 72 non‐singleton operational taxonomic units (soil) and 109 phylogroups (soil + sporocarp). The most species‐rich genera are Thelephora/Tomentella, Cortinarius and Inocybe, followed by Hebeloma, Russula, Lactarius, Entoloma, Sebacina, Clavulina, Laccaria, Leccinum and Alnicola. Despite the scarcity of available reference data from other arctic regions, the majority of the phylogroups (73.4%) were also found outside Svalbard. At the same time, all putative Svalbard ‘endemics’ were newly sequenced taxa from diverse genera with massive undocumented diversity. Overall, our results support long‐distance dispersal more strongly than vicariance and glacial survival. However, because of the high variation in nucleotide substitution rates among fungi, allopatric persistence since the Pliocene, although unlikely, cannot be statistically rejected. Results from the coalescent analyses suggest recent gene flow among different arctic areas. Main conclusions Our results indicate numerous recent colonization events and suggest that long‐distance, transoceanic dispersal is widespread in arctic ECM fungi, which differs markedly from the currently prevailing view on the dispersal capabilities of ECM fungi. Our molecular evidence indicates that long‐distance dispersal has probably played a major role in the phylogeographic history of some ECM fungi in the Northern Hemisphere. Our results may have implications for studies on the biodiversity, ecology and conservation of arctic fungi in general.  相似文献

The onset and cessation of the synthesis of denitrification enzymes of Pseudomonas stutzeri were investigated by using continuous culture and defined dissolved oxygen levels covering the full range of transition from air saturation to complete anaerobiosis. Expression of nitrate reductase, nitrite reductase (cytochrome cd1), and N2O reductase was controlled by discrete oxygen levels and by the nature of the nitrogenous oxide available for respiration. N2O reductase was synthesized constitutively at a low level; for enhanced expression, oxygen concentrations were required to decrease below 5 mg of O2 per liter. The threshold values for synthesis of nitrate reductase and cytochrome cd1 in the presence of nitrate were ca. 5 and ca. 2.5 mg of O2 per liter, respectively. With nitrous oxide as the respiratory substrate, nitrite reductase was again the most sensitive to oxygen concentration; however, thresholds for all denitrification enzymes shifted to lower oxygen levels. Whereas the presence of nitrate resulted in maximum expression and nearly uniform induction of all reductases, nitrite and nitrous oxide stimulated preferably the respective enzyme catalyzing reduction. In the absence of a nitrogenous oxide, anaerobiosis did not induce enzyme synthesis to any significant degree. The accumulation of nitrite seen during both the aerobic-anaerobic and anaerobic-aerobic transition phases was caused by the differences in onset or cessation of synthesis of nitrate and nitrite reductases and an inhibitory effect of nitrate on nitrite reduction.  相似文献
