Behavioral and physiological responses to hypoxia were examined in three sympatric species of sharks: bonnethead shark Sphyrna tiburo, blacknose shark, Carcharhinus acronotus, and Florida smoothhound shark, Mustelus norrisi, using closed system respirometry. Sharks were exposed to normoxic and three levels of hypoxic conditions. Under normoxic conditions (5.5–6.4mg l–1), shark routine swimming speed averaged 25.5 and 31.0cm s–1 for obligate ram-ventilating S. tiburo and C. acronotus respectively, and 25.0cm s–1 for buccal-ventilating M. norrisi. Routine oxygen consumption averaged about 234.6 mg O2kg–1h–1 for S. tiburo, 437.2mg O2kg–1h–1 for C. acronotus, and 161.4mg O2 kg–1 h–1 for M. norrisi. For ram-ventilating sharks, mouth gape averaged 1.0cm whereas M. norrisi gillbeats averaged 56.0 beats min–1. Swimming speeds, mouth gape, and oxygen consumption rate of S. tiburo and C. acronotus increased to a maximum of 37–39cm s–1, 2.5–3.0cm and 496 and 599mg O2 kg–1 h–1 under hypoxic conditions (2.5–3.4mg l–1), respectively. M. norrisi decreased swimming speeds to 16cm s–1 and oxygen consumption rate remained similar. Results support the hypothesis that obligate ram-ventilating sharks respond to hypoxia by increasing swimming speed and mouth gape while buccal-ventilating smoothhound sharks reduce activity.  相似文献

In order to maintain axenic seedstock cultures axenically of thecommercially important red seaweed, Porphyra yezoensis, aprocedure was developed for axenic isolation and culture of conchocelis andmonospores. For axenic isolation of the conchocelis, contaminated microalgaewere most effectively removed by filtering contaminated samples through a100-m mesh after sonication. Removal of bacteria and otheralgaewas accomplished using a mixture of 5 agents (0.02% chitosan, 100 gml–1 GeO2, 10 gml–1 ampicillin, 40 gml–1 kanamycin and 200 gml–1 streptomycin). Axenic single colonies wereisolatedfrom a semi-solid medium prepared from 1% transfer gel. After collectingmonospores from the 40–50% density layer on a percoll-gradient, removalofbacteria and fungi from the monospores was accomplished using a mixture of 5antibiotics (3.5 g ml–1 nystatin, 2 mgml–1 ampicillin, 400 gml–1 kanamycin, 50 gml–1 neomycin and 800 gml–1 streptomycin). A method of measuring CO2gas exchange (caused, for example, by microalgal photosynthesis on emersed tidal mudflats) using open flow IR gas analyzers is described. The analyzers are integrated in a conventional portable photosynthesis system (LI-6400, LI-COR, Nebraska, USA), which allows manipulation and automatic recording of environmental parameters at the field site. Special bottomless measuring chambers are placed directly on the surface sediment. Measurements are performed under natural light conditions and ambient CO2concentrations, as well as under different CO2concentrations in air, and various PAR radiation levels produced by a LED light source built into one of the measurement chambers. First results from tidal channel banks in a north Brazilian mangrove system at Bragança (Pará, Brazil) under controlled conditions show a marked response of CO2assimilation to CO2concentration and to irradiance. Photosynthesis at 100molmol–1CO2in air in one sample of a well-developed algal mat was saturated at 309mol photons m–2s–1, but increased with increasing ambient CO2concentrations (350 and 1000mol mol–1CO2) in the measuring chamber. Net CO2assimilation was 0.8mol CO2m–2s–1at 100mol mol–1CO2, 5.9mol CO2m–2s–1at 350mol mol–1CO2and 9.8mol CO2m–2s–1at 1000mol mol–1CO2. Compensation irradiance decreased and apparent photon yield increased with ambient CO2concentration. Measurements under natural conditions resulted in a quick response of CO2exchange rates when light conditions changed. We recommend the measuring system for rapid estimations of benthic primary production and as a valuable field research tool in connection with certain ecophysiological aspects under changing environmental conditions.  相似文献

Summary The objectives of this study included directin vivo measurements of circulating blood gases, pH, heart rate, and blood pressure during voluntary dives of unrestrained Nile monitor lizards. A Radiometer flow-through cuvette was employed for continuous recording of arterial PO2, PCO2 and pH. Hematological properties revealed no particular adaptations for diving. Mean values were: hematocrit = 24%; hemoglobin concentration = 7.1 g %; oxygen capacity = 9.3 vol %; red cell dimensions = 22×12 ; red cell count = 0.67 million/l. The respiratory properties of the blood, studiedin vitro andin vivo, show distinct adaptations to habitual diving. Oxygen affinity of blood is low (P50 = 42.4 mm Hg at pH 7.45, 25 °) and the dissociation curve is markedly sigmoid (n = 3.1). These features, coupled with a Bohr factor ( logP 50/pH) of –0.48, ensure increased utilization of oxygen while maintaining relatively high tissue PO2. Arterial pH decreases during diving from about 7.5 to 7.1 due to combined respiratory and metabolic acidosis. High plasma bicarbonate (30 mM/l at PCO2 = 25 mm Hg) and a buffering capacity of H C3 O/ pH = 18.9 mM/l increase the tolerance to this acidosis and prolong diving time. Thein vivo oxygen dissociation curve shows a 90 % depletion of arterial oxygen content during typical dives. Diving elicited a rapidly developing bradycardia with maximum of 85 % reduction in heart rate. The temperature sensitivity of HbO2 binding was very low (H = –3kcal). This would minimize the HbO2 affinity increase accompanying the decrease in body temperature likely to occur in lizards going from sun basking to submergence in water.Supported by a grant from the Danish Natural Science Research Council.  相似文献

We investigated whether amyloid--peptide (A1–42) has an effect on the elevations of the intracellular concentration of Ca2+ ions ([Ca2+]i) induced by depolarizations of NG108-15 cells and on related Ca2+ channels. A1–42 (10-1000 nM) had no immediate effect on depolarization-induced [Ca2+]i elevations. [Ca2+]i increases were slightly diminished in cells grown in the presence of 100 or 1000 nM A1–42. Nifedipine (1 M) reduced these elevations equally in cells grown in the absence or presence of A1–42. In contrast, the ability of -conotoxin GVIA to diminish the depolarization-induced [Ca2+]i responses became lost in cells grown in the presence of 100 nM A1–42. This indicates that the influx of calcium through the N-type Ca2+ channels was compromised by the chronic exposure of cells to a submicromolar concentration of A1–42, presumably because of impairement of their function or diminished expression. This may be important in the pathogeny of Alzheimer's dementia in view of the pivotal role of N-type Ca2+ channels in neurotransmitter release.  相似文献

Summary The enzymatic transformation of desacetyl-lanatoside A (DLA) to its secondary glycoside, digitoxin, in solutions of -and -cyclodextrins is effected using of -glucosidase from barley. Due to the interaction of cyclodextrins (CyDs) with desacetyl-lanatoside A, an increase in solubility of the latter of 24.5 and 230 times was observed for -cyclodextrin and -cyclodextrin, respectively. Kinetic studies of the enzymatic transformation gave for -glucosidase the values KM=3.3×10–4 mol. dm–3 and Vmax=0.557 mol mg–1 min–1 when the substrate was the deacetyl-lanatoside A complex with -cyclodextrin, while in the case of the complex with -cyclodextrin these values were KM=5.45×10–4 mol dm–3 and Vmax=0.896 mol mg–1 min–1.  相似文献

Evidence from ventricular preparations of cat, sheep, rat and dog suggests that both 1-adrenoceptors (1AR) and 2-adrenoceptors (2AR) mediate positive inotropic effects but that only 1AR do it through activation of a cAMP pathway. On the other hand, our evidence has shown that both 1 AR and 2 AR hasten relaxation of isolated human myocardium consistent with a common cAMP pathway. We have now investigated in the isolated human right atrial appendage, a tissue whose -AR comprise around 2/3 of 1AR and 1/3 of 2AR, whether or not 2AR-mediated effects occur via activation of a cAMP pathway. We carried out experiments on atria obtained from patients without advanced heart failure undergoing open heart surgery. To activate 2AR, we used the 2AR-selective ligand zinterol. Experiments were carried out on paced atrial strips (1 Hz) and tissue homogenates and membrane particles. Zinterol caused positive inotropic and lusitropic (i.e. reduction of t1:2 of relaxation) effects with EC50 values of 3 and 2 nM, respectively. The zinterol-evoked effects were unaffected by the AR-selective antagonist CGP 20712A (300 nM) but blocked surmountably by the 2AR-selective antagonist ICI 118551 (50 nM) which reduced both EC50 values to 1 M. Zinterol stimulated adenylyl cyclase activity with an EC50 of 30 nM and intrinsic activity of 0.75 with respect to (–)-isoprenaline (600 M); the effects were resistant to blockade by CGP 20712A (300 nM) but antagonised surmountably by ICI 118551 (50 nM). Zinterol bound to membrane PAR labelled with (–)-[125I] cyanopindolol with higher affinity for 2AR than for - 1 AR; the binding to 2AR but not to - BAR was reduced by GTPyS (10 M). In the presence of CGP 20712A (300 nM) (–)-isoprenaline (400 M); (to activate both 1AR and 2AR maximally) and zinterol (10 M); increased contractile force 3.4-fold and 2.5-fold respectively and reduced relaxation tut by 32% and 18% respectively. These effects of (–)-isoprenaline and zinterol were associated (5 min incubation) with phosphorylation (pmol P/mg supernatant protein) of troponin I and C-protein to values of 8.4 ± 2.0 vs 12.4 ± 2.3 and 10.1 ± 2.5 vs 8.6 ± 1.6 respectively. (–)-Isoprenaline and zinterol also caused phosphorylation of phospholamban (1.8 ± 0.3 vs 0.4 ± 0.1 pmol P/mg respectively) specifically at serine residues. We conclude that in human atrial myocardium activation of both 1AR and 2AR leads to cAMP-dependent phosphorylation of proteins involved in augmenting both contractility and relaxation.  相似文献

Connective tissue of the freshwater pulmonateLymnaea stagnalis was shown to contain fucosyltransferase activity capable of transferring fucose from GDP-Fuc in 1–2 linkage to terminal Gal of type 3 (Gal1–3GalNAc) acceptors, and in 1–3 linkage to GlcNAc of type 2 (Gal1–4GlcNAc) acceptors. The 1–2 fucosyltransferase was active with Gal1–3GalNAc1-OCH2CH=CH2 (K m=12 mM,V max=1.3 mU ml–1) and Gal1–3GalNAc (K m=20 mM,V max=2.1 mU ml–1), whereas the 1–3 fucosyltransferase was active with Gal1–4GlcNAc (K m=23 mM,V max=1.1 mU ml–1). The products formed from Gal1–3GalNAc1-OCH2CH=CH2 and Gal1–4GlcNAc were purified by high performance liquid chromatography, and identified by 500 MHz1H-NMR spectroscopy and methylation analysis to be Fuc1–2Gal1–3GalNAc1-OCH2CH=CH2 and Gal1–4(Fuc1–3)GlcNAc, respectively. Water uptake and transport were studied in eight liana species in a seasonally dry tropical forest on Barro Colorado Island, Panama. Stable hydrogen isotope composition (D) of xylem and soil water, soil volumetric water content (v), and basal sap flow were measured during the 1997 and 1998 dry seasons. Sap flow of several neighboring trees was measured to assess differences between lianas and trees in magnitudes and patterns of daily sap flow. Little seasonal change in v was observed at 90–120 cm depth in both years. Mean soil water D during the dry season was –19 at 0–30 cm, –34 at 30–60 cm, and –50 at 90–120 cm. Average values of xylem D among the liana species ranged from –28 to –44 during the middle of the dry season, suggesting that water uptake was restricted to intermediate soil layers (30–60 cm). By the end of the dry season, all species exhibited more negative xylem D values (–41 to –62), suggesting that they shifted to deeper water sources. Maximum sap flux density in co-occurring lianas and trees were comparable at similar stem diameter (DBH). Furthermore, lianas and trees conformed to the same linear relationship between daily sap flow and DBH. Our observations that lianas tap shallow sources of soil water at the beginning of the dry season and that sap flow is similar in lianas and trees of equivalent stem diameter do not support the common assumptions that lianas rely primarily on deep soil water and that they have higher rates of sap flow than co-occurring trees of similar stem size.  相似文献

A microbiosensor for hydrogen peroxide (H2O2) was constructed by immobilizing catalase in a polyacrylamide gel on the tip of a Clark-type oxygen microelectrode. The outer tip diameter was 15–40 m. The sensors had response times of 0.7–1.2 s, and could detect as little as 2–4 M H2O2. They could measure with a spatial resolution of about 100m and remained operational for up to three weeks.  相似文献

The effect of lead on the filtration rate of the zebra musselDreissena polymorpha was investigated, together with the accumulation of Pb in the soft tissues of the mussels. The NOEC-filtration was 116 g.l–1 (0,56 mol.l–1) and the EC50-filtration was 370 g.l–1 (1.79 mol.l–1). The NOEC-accumulation was the concentration found in the control water (1.4g.l–1). These experiments show that the EC50-filtration for Pb is similar to that for Cd, higher than that for Cu and lower than that for Zn. The water quality criteria for lead allow 25 g Pb.l–1 in surface water. This will not cause short-term effects. Long-term effects may, however, occur, since an accumulation of Pb as low as 16 g.l–1 was recorded in this study.  相似文献

We have previously reported (Kim et al., J. Dairy Res., 1999, in press) three novel angiotensin-I-converting enzyme (ACE) inhibitory peptides, Tyr-Pro-Glu-Arg, Tyr-Tyr-Pro-Gln-Ile-Met-Gln-Tyr and Asn-Asn-Val-Met-Leu-Gln-Trp, isolated from the tryptic digest of recombinant human s1-casein. A series of C-terminal di- and tripeptides from these three peptides have now been synthesized and their ACE inhibitory activities have been measured. Among the peptides tested, the tripeptide Leu-Gln-Trp had the highest ACE inhibitory activity (IC50=3.8 mol l–1).  相似文献

Etiolated sunflower cotyledons developed in complete darkness and lacking photosystem (PS) 2 were exposed to continuous 200 µmol(photon) m–2 s–1 white light for 1, 3, 6, 12, and 18 h prior to evaluations of excitation-energy dissipation using modulated chlorophyll a fluorescence. Photochemical potential of PS2, measured as the dark-adapted quantum efficiency of PS2 (FV(M)/FM), and thermal dissipation from the antenna pigment-protein complex, measured as the Stern-Volmer non-photochemical quenching coefficient (NPQ), increased to 12 h of irradiation. Following 12 h of irradiation, thermal dissipation from the antennae pigment-protein complex decreased while the efficiency of excitation capture by PS2 centers (FV/FM) and light-adapted quantum efficiency of PS2 (PS2) continued to increase to 18 h of irradiation. The fraction of the oxidized state of QA, measured by the photochemical quenching coefficient (qP), remained near optimal and was not changed significantly by irradiation time. Hence during the development of maximum photochemical potential of PS2 in sunflower etioplasts, which initially lacked PS2, enhanced thermal dissipation helps limit excitation energy reaching PS2 centers. Changes of the magnitude of thermal dissipation help maintain an optimum fraction of the oxidized state of QA during the development of PS2 photochemistry.  相似文献

In this paper we use a dynamical systems approach to prove the existence of a unique critical value c * of the speed c for which the degenerate density-dependent diffusion equation u ct = [D(u)u x ] x + g(u) has: 1. no travelling wave solutions for 0 < c < c *, 2. a travelling wave solution u(x, t) = (x - c * t) of sharp type satisfying (– ) = 1, () = 0 *; '(*–) = – c */D'(0), '(*+) = 0 and 3. a continuum of travelling wave solutions of monotone decreasing front type for each c > c *. The effects of eight microelements (I, BO3 3–, MoO4 2–, Co2+, Cu2+, Mn2+, Fe2+, Zn2+) on the biosynthesis of camptothecin and the growth of suspension cultures of Camptotheca acuminata were studied. The increase of I to 25 M l–1, Cu2+ to 1 M l–1, Co2+ to 2 M l–1 and MoO4 2– to 10 M l–1 in Murashige and Skoog (MS) medium resulted in 1.66, 2.84, 2.53 and 2.04 times higher of camptothecin yield than that in standard MS medium respectively. Combined treatment of I (25 M l–1), Cu2+ (1 M l–1), Co2+ (2 M l–1) and MoO4 2– (10 M l–1) lead to improve cell dry weight, camptothecin content, and camptothecin yield to 30.56 g l–1, 0.0299%, and 9.15 mg l–1, respectively, which were 20.2, 208.9 and 273.8% increment respectively when compared with those of control.  相似文献

In an attempt to detect differences in the thermal effect of infrared irradiation of different wavelengths, transient sweating response to infrared irradiation in various spectral regions was examined. In Series 1, the ventral or dorsal surface of the nude subject was irradiated repetitively for a period of 4 min (2 min on, 2 min off) by each of three kinds of infrared heaters with main emissivity in near-infrared (NIR; 0.7–2.8 m), intermediate-infrared (MIR; 1.5–5.8 m), and far-infrared (FIR; 2.8–25 m) regions. The sweating response on a non-irradiated area tended to be the greatest with MIR, while the magnitude of the sweating response on the irradiated area showed no consistent differences among various wavelengths. The results infer that MIR stimulated cutaneous thomoreceptors most effectively, while its direct effect on local sweat gland activity was minimal. In Series 2, the effects of 9–12 min irradiations in more restricted ranges of wavelength were compared by the combination of the three kinds of heaters with filters (translucent to wavelength ranges of 1.3–2.7, 2.7–3.5, 3.6–8.0 m, respectively). The sweating response on a remote area was predominantly greater with the range of 2.7–3.5 m than with the other wavelength ranges, while the local effect on sweating was minimal with this range. The results of Series 2 reinforce those of Series 1, indicating that the degree of stimulation of cutaneous thermoreceptors and of direct thermal effect on sweat gland activity differ with spectral regions incident on the skin, thus affecting local and remote effects on the sweating response.  相似文献

The radiotracer13N was used to undertake compartmental analyses for NO 3 in intact non-mycorrhizal roots ofPicea glauca (Moench) Voss. seedlings. Three compartments were defined, with half-lives of exchange of 2.5 s, 20 s, and 7 min. These were identified as representing surface adsorption, apparent free space, and cytoplasm, respectively. Influx, efflux, and net flux as well as cytoplasmic and apparent-free-space nitrate concentrations were estimated for three different concentration regimes of external nitrate. After exposure to external NO 3 for 3 d, influx was calculated to be 0.09 mol·g–1·h–1 (at 10 M [NO 3 ]o), 0.5mol·g–1·h–1 (at 100 M [NO inf3 sup– ]o), and 1.2 mol · g–1· h–1 (at 1.5 mM [NO 3 ]o). Efflux increased with increasing [NO 3 ]o, constituting 4% of influx at 10 M, 6% at 100 M, and 21% at 1.5 mM. Cytoplasmic [NO 3 ] was estimated to be 0.3 mM at 10 uM [NO 3 ]o, 2mM at 100 M [NO 3 ]o, and 4mM at 1.5 mM [NO 3 ]o, while free-space [NO 3 ] was 16 M, 173 M, and 2.2 mM, respectively. A series of experiments was carried out to confirm the identity of the compartments resolved by efflux analysis. Pretreatment at high temperature or application of 2-chloro-ethanol, sodium dodecyl sulphate or hydrogen peroxide made it possible to distinguish the metabolic (cytoplasmic) phase from the remaining two (physical) phases. A vast number of lakes developed in the abandoned opencast lignite mines of Lusatia (East Germany) contain acidic waters (mmolSm–2a). Potential Fe(III) reduction measured by the accumulation of Fe(II) during anoxic incubation yielded similar rates in both types of sediments, however, the responses towards the supplementation of Fe(III) and organic carbon were different. Sulfate reduction rates estimated with 35S-radiotracer were much lower in the slightly acidic sediment than in the pH-neutral sediment (156 v.s. 738mmolSO4 2–m–2a–1). However, sulfate reduction rates were increased by the addition of organic carbon. Severe limitation of sulfate-reducing bacteria under acidic conditions was also reflected by low most probable numbers (MPN). High MPN of acidophilic iron- and sulfur-oxidizing bacteria in acidic sediments indicated a high reoxidation potential. The results show that potentials for reductive processes are present in acidic sediments and that these are determined mainly by the availability of oxidants and organic matter.  相似文献

In the Mammoth Springs (Yellowstone National Park) waters with near neutral pH and soluble sulfide (H2S, HS, S2–) of over 1–2 mg/liter (30–60M) are characterized by substrate covers of phototrophic bacteria (Chloroflexus and aChlorobium-like unicell) above 50C and by a blue-green alga (Spirulina labyrinthiformis) below this temperature.Synechococcus. Mastigocladus, and other blue-green algae typical of most hot springs of western North America are excluded, apparently by sulfide. The sulfide-adaptedSpirulina photosynthesized at maximum rates at 45C and at approximately 300 to 700Ein/m2/sec of visible radiation. Sulfide (0.6–1.2 mM) severely poisoned photosynthesis of nonadapted populations, but those continuously exposed to over 30M tolerated at least 1 mM without inhibition. A normal14C-HCO3 photoincorporation rate was sustained with 0.6–1 mM sulfide in the presence of DCMU (7M) or NH2OH (0.2 mM), although both of these photosystem II inhibitors prevented photoincorporation without sulfide. Other sulfur-containing compounds (S2O3 2– SO3 2–, S2O4 2– thioglycolic acid cysteine) were unable to relieve DCMU inhibition. The lowering of the photoincorporation rate by preferentially irradiating photosystem I was also relieved by sulfide. The most tenable explanation of these results is that sulfide is used as a photo-reductant of CO2, at least when photosystem II is inhibited. It is suggested that in some blue-green algae photosystem II is poisoned by a low sulfide concentration, thus making these algae sulfidedependent if they are to continue photosynthesizing in a sulfide environment. Presumably a sulfidecytochrome reductase enzyme system must be synthesized for sulfide to be used as a photo-reductant.  相似文献
