局限青霉生淀粉水解酶的产生条件及酶一般性质的研究   总被引:1,自引:0,他引:1  
生淀粉水解酶产生菌局限青霉(Penicillium restrictum)127-2是由土壤中分解得到的,具有较强的降解生淀粉能力,水解1g生淀粉需8.0国际单位酶量.Sasaki等曾报道草酸青霉1-9、变幻青霉的培养液能分解生淀粉,但未报道酶的形成条件;Takao等研究了罗尔伏革菌生淀粉酶的形成条件和酶的一般性质.本文介绍局限青霉127-2生淀粉水解酶的形成条件和酶的一般性质.1 材料和方法1.1 菌种培养1.1.1 斜面培养:用查氏琼脂斜面培养基在28—30℃培养5—7天.1.1.2 三角瓶振荡培养:于250ml三角瓶中加40ml发酵培养基,经1kg/cm~2 30分钟灭菌,冷却后接种,置于28—30℃的摇床(200r/min)振荡培养4—5天.1.2 酶活力测定  相似文献

脓毒症在外科临床工作中较常见,治疗相当困难;本文主要概述了一氧化氮合酶的基因定位、结构特点以及一氧化氮合酶与脓毒症的关系,进一步说明由一氧化氮合酶介导的一氧化氮生成与脓毒症关系密切,而选择性一氧化氮合酶抑制剂在脓毒症各阶段恰当的应用可能是有效治疗脓毒症、降低病死率的一个重要途径,也将成为今后研究的热点。  相似文献

The transition from unicellular to multicellular life forms requires the development of a specialized structural component,the extracellular matrix(ECM).In Metazoans,there are two main supportive systems,which are based on chitin and collagen/hyaluronan,respectively.Chitin is the major constituent of fungal cell walls and arthropod exoskeleton.However,presence of chitin/chitooligosaccharides has been reported in lower chordates and during specific stages of vertebrate development.In this study,the occurrence of chitin synthases(CHSs) was investigated with a bioinformatics approach in the cephalochordate Branchiostoma floridae,in which the presence of chitin was initially reported in the skeletal rods of the pharyngeal gill basket.Twelve genes coding for proteins containing conserved amino acid residues of processive glycosyltransferases from GT2 family were found and 10 of them display mosaic structures with novel domains never reported previously in a chitin synthase.In particular,the presence of a discoidin(DS) and a sterile alpha motif(SAM) domain was found in nine identified proteins.Sequence analyses and homology modelling suggest that these domains might interact with the extracellular matrix and mediate protein-protein interaction.The multi-domain putative chitin synthases from B.floridae constitute an emblematic example of the explosion of domain innovation and shuffling which predate Metazoans.  相似文献

Cysteine occupies a central position in plant metabolism because it is a reduced sulfur donor moleculeinvolved in the synthesis of essential biomolecules and defense compounds. Moreover, cysteine per se and its deriva-tive molecules play roles in the redox signaling of processes occurring in various cellular compartments. Cysteine issynthesized during the sulfate assimilation pathway via the incorporation of sulfide to O-acetylserine, catalyzed byO-acetylserine(thiol)lyase (OASTL). Plant cells contain OASTLs in the mitochondria, chloroplasts, and cytosol, resultingin a complex array of isoforms and subcellular cysteine pools, in recent years, significant progress has been made inArabidopsis, in determining the specific roles of the OASTLs and the metabolites produced by them. Thus, the dis-covery of novel enzymatic activities of the less-abundant, like DES1 with L-cysteine desulfhydrase activity and SCSwith S-sulfocysteine synthase activity, has provided new perspectives on their roles, besides their metabolic functions.Thereby, the research has been demonstrated that cytosolic sulfide and chloroplastic S-sulfocysteine act as signalingmolecules regulating autophagy and protecting the photosystems, respectively. In the cytosol, cysteine plays an essentialrole in plant immunity; in the mitochondria, this molecule plays a central role in the detoxification of cyanide, which isessential for root hair development and plant responses to pathogens.  相似文献

胸苷酸合成酶(thymidylate synthase, TS)是催化生物体内胸苷酸合成所必需的酶, 多年来一直作为肿瘤化疗的靶点. 研究表明, TS是一种RNA结合蛋白, 可以与其自身mRNA的2个位点结合, 使mRNA翻译受阻. 本文以mRNA体外展示技术进行了由大容量多肽库(>1013)中筛选胸苷酸合成酶mRNA亲和多肽的研究, 对随机库进行了12轮循环的选择及扩增. 结果表明, 与初始库相比, 经选择循环之后, 碱性氨基酸及芳香族的苯丙氨酸含量明显增加, 它们在TS RNA与蛋白质的相互作用中扮演着重要角色. 按其理化特性对每一随机位点的氨基酸进行分类, 并与初始库比较, 发现位点1, 12, 17和18具有明显的带正电荷的特性, 表明碱性侧链参与了与RNA的结合. 二级结构预测表明, 随着筛选的进行, 与TS mRNA 亲和的多肽显示出明显的螺旋倾向, 而且形成螺旋的区域富含碱性氨基酸. 凝胶迁移及体外翻译实验证实, 选择循环之后的多肽能够与TS mRNA高度亲和, 并能抑制TS mRNA的翻译. 本研究表明mRNA体外展示方法得到的亲和多肽可以用作新的TS RNA的翻译抑制剂, 并有可能成为一类新型的抗肿瘤药物.  相似文献

It has been known that arginine is used as the basic amino acid in the ?subunit of cytochrome b559 (Cyt b559) except histidine. However, previous studies have focused on the function of histidine in the activities of photosystem (PS) Ⅱ and there are no reports regarding the structural and/or functional roles of arginine in PSII complexes. In the present study,two arginine18 (R18) mutants of Chlamydomonas reinhardtii were constructed using site-directed mutagenesis, in which R18 was replaced by glutamic acid (E) and glycine (G). The results show that the oxygen evolution of the PSII complex in the R18G and R18E mutants was approximately 60% of wild-type (WT) levels and that, after irradiation at high light intensity, oxygen evolution for the PSII of mutants was reduced to zero compared with 40% in WT cells. The efficiency of light capture by PSII (Fv/Fm) of R18G and R18E mutants was approximately 42%-46% that of WT cells. Furthermore, levels of the ?subunit of Cyt b559 and PsbO proteins were reduced in thylakoid membranes compared with WT. Overall, these data suggest that R18 plays a significant role in helping Cyt b559 maintain the structure of the PSII complex and its activity,although it is not directly bound to the heme group.  相似文献

Gelatinization temperature (GT) is an important parameter for evaluating the cooking and eating quality of rice besides amylose content (AC). The inheritance of the genes affecting GT has been widely studied and is considered to be controlled by a major gene. Here, we report the map-based cloning of rice ALK that encodes the soluble starch synthase II (SSSII). Comparison between the DNA sequences from different rice varieties, together with the results obtained with digestion of the rice seeds in alkali solution, indicates that the base substitutions in coding sequence of ALK may cause the alteration in GT.  相似文献

开花期是水稻最重要的农艺性状之一,水稻的花期决定着水稻的地区适应性和最终产量。人工选择使水稻从短日照向长日照、低纬度向高纬度扩张,因此水稻已逐渐进化出适应长日照条件下的开花调控机制。目前,虽然鉴定了一些影响水稻长日照的开花基因如SDG724、RFT1、EHD4、DTH2,但是挖掘水稻长日照开花基因还十分有限。本研究通过筛选水稻突变体库,获得一批在长日照下花期有显著差异的突变体材料,其中一份突变体lfm1(late-flowering mutant1),在长日照条件下开花延迟,在短日照条件下开花时间正常。通过图位克隆,将Lfm1基因初定位至第8染色体端粒附近。进一步的精细定位将Lfm1基因定位于分子标记8-0.269和与8-0.283之间,范围为12 kb,该区域包括3个候选基因。经测序分析发现,在突变体lfm1中,LOC_Os08g01420基因的第六外显子2800处缺失9个碱基,突变体lfm1等位于已报道的突变体ehd3。在适度(中日照条件下,~12 h/12 h)的光照条件下,突变体lfm1表现为穗粒数增多,生育期略延长,具有应用于生产的潜力。Lfm1基因的克隆为培育适应不同生态区域的水稻材料提供了重要的基因资源。  相似文献

Gelatinization temperature (GT) is an important parameter in evaluating the cooking and eating quality of rice. Indeed, the phenotype, biochemistry and inheritance of GT have been widely studied in recent times. Previous map-based cloning revealed that GT was controlled by ALK gene, which encodes a putative soluble starch synthase II-3. Complementation vector and RNAi vector were constructed and transformed into Nipponbare mediated by Agrobacterium. Phenotypic and molecular analyses of transgenic lines provided direct evidence for ALK as a key gene for GT. Meanwhile, amylose content, gel consistency and pasting properties were also affected in transgenic lines. Two of four nonsynonymous single nucleotide polymorphisms in coding sequence of ALK were identified as essential for GT. Based on the single nucleotide polymorphisms (SNPs), two new sets of SNP markers combined with one cleaved amplified polymorphic sequence marker were developed for application in rice quality breeding.  相似文献

A rice semidwarfing gene, sd-1, known as the "green revolution gene," was isolated by positional cloning and revealed to encode gibberellin 20-oxidase, the key enzyme in the gibberellin biosynthesis pathway. Analysis of 3477 segregants using several PCR-based marker technologies, including cleaved amplified polymorphic sequence, derived-CAPS, and single nucleotide polymorphisms revealed 1 ORF in a 6-kb candidate interval. Normal-type rice cultivars have an identical sequence in this region, consisting of 3 exons (558, 318, and 291 bp) and 2 introns (105 and 1471 bp). Dee-Geo-Woo-Gen-type sd-1 mutants have a 383-bp deletion from the genome (278-bp deletion from the expressed sequence), from the middle of exon 1 to upstream of exon 2, including a 105-bp intron, resulting in a frame-shift that produces a termination codon after the deletion site. The radiation-induced sd-1 mutant Calrose 76 has a 1-bp substitution in exon 2, causing an amino acid substitution (Leu [CTC] to Phe [TTC]). Expression analysis suggests the existence of at least one more locus of gibberellin 20-oxidase which may prevent severe dwarfism from developing in sd-1 mutants.  相似文献

Rice blast, caused by the fungal pathogen Magnaporthe grisea, is one of the most devastating diseases in rice worldwide. The dominant resistance gene, Pi-d2 [previously named Pi-d(t)2], present in the rice variety Digu, confers gene-for-gene resistance to the Chinese blast strain, ZB15. Pi-d2 was previously mapped close to the centromere of chromosome 6. In this study, the Pi-d2 gene was isolated by a map-based cloning strategy. Pi-d2 encodes a receptor-like kinase protein with a predicted extracellular domain of a bulb-type mannose specific binding lectin (B-lectin) and an intracellular serine-threonine kinase domain. Pi-d2 is a single-copy gene that is constitutively expressed in the rice variety Digu. Transgenic plants carrying the Pi-d2 transgene confer race-specific resistance to the M. grisea strain, ZB15. The Pi-d2 protein is plasma membrane localized. A single amino acid difference at position 441 of Pi-d2 distinguishes resistant and susceptible alleles of rice blast resistance gene Pi-d2. Because of its novel extracellular domain, Pi-d2 represents a new class of plant resistance genes.  相似文献

在水稻基因组序列中发现类似MYC序列的同源序列,并设计引物成功克隆了一个水稻OsMYC基因(GenBank登录号:AY398581),并对其进行了分子结构分析。OsMYC基因具有典型的DNA结合结构域:碱性区域/螺旋-环-螺旋(bHLH)基序,与其他MYC类似基因的蛋白序列比对结果表明,OsMYC基因与AtMYC2、MYC7E和PG1等氨基酸序列一致性分别是78%、48%、46%,而在bHLH区的一致性分别为95%、84%、77%,N端保守区的一致性分别为81%、54%、52%;位于bHLH区域内的核定位信号区则完全一致;系统进化树分析结果表明,该基因与AtMYC2、MYC7E和PG1等位于同一亚类。此外,该基因主要在营养器官中表达,茎秆中表达最强,根和叶片中较弱,并且能被外源ABA或Fe^3 所诱导,这与AtMYC2、MYC7E基因的表达模式基本相同。该基因是水稻MYC转录因子家族的一个新成员。  相似文献
