The brown planthopper (BPH), Nilaparvata lugens (Stål), which periodically erupted in tropical Asian rice before the 1960s, became a major threat after farmers adopted green revolution technologies in the 1960s. Management and policy changes in the 1980s and 1990s emphasized non-insecticidal tactics to avert BPH outbreaks. However, insecticides have resurfaced as the primary means for controlling rice insect pests and tropical Asian countries have recently experienced planthopper outbreaks in record numbers. Our review of factors that have contributed to the outbreaks points to insecticides as the most tangible outbreak factor primarily because of their harmful effects on natural enemies. BPH resistance to insecticides and especially imidacloprid has increased the probability of outbreaks as farmers have applied increasing quantities of insecticide in an attempt to combat resistant populations. Similarly, heavy use of nitrogen fertilizer, especially on hybrid rice, has increased the potential for outbreaks. Other factors triggering outbreaks are less documented, but we discuss the possibility that the high outbreak synchrony in geographically separated populations of BPH may suggest a “Moran effect” such as climate that promotes an environment favoring above-average increases in BPH populations. Also, we hypothesize that BPH functions as a metapopulation and, as such, periodic outbreaks could be a natural phenomenon requiring resupply of planthoppers into vacant areas to ensure genetic linkage among subpopulations. We conclude with a series of recommendations for research and policy changes aimed at better understanding the cause of BPH outbreaks and for developing sustainable management practices to prevent future outbreaks.  相似文献

Identifying the molecular markers for complex quantitative traits in natural populations promises to provide novel insight into genetic mechanisms of adaptation and to aid in forecasting population dynamics. In this study, we investigated single nucleotide polymorphisms (SNPs) using candidate gene approach from high‐ and low‐fecundity populations of the brown planthopper (BPH) Nilaparvata lugens Stål (Hemiptera: Delphacidae) divergently selected for fecundity. We also tested whether the population fecundity can be predicted by a few SNPs. Seven genes (ACE, fizzy, HMGCR, LpR, Sxl, Vg and VgR) were inspected for SNPs in N. lugens, which is a serious insect pest of rice. By direct sequencing of the complementary DNA and promoter sequences of these candidate genes, 1033 SNPs were discovered within high‐ and low‐fecundity BPH populations. A panel of 121 candidate SNPs were selected and genotyped in 215 individuals from 2 laboratory populations (HFP and LFP) and 3 field populations (GZP, SGP and ZSP). Prior to association tests, population structure and linkage disequilibrium (LD) among the 3 field populations were analysed. The association results showed that 7 SNPs were significantly associated with population fecundity in BPH. These significant SNPs were used for constructing general liner models with stepwise regression. The best predictive model was composed of 2 SNPs (ACE‐862 and VgR‐816) with very good fitting degree. We found that 29% of the phenotypic variation in fecundity could be accounted for by only two markers. Using two laboratory populations and a complete independent field population, the predictive accuracy was 84.35–92.39%. The predictive model provides an efficient molecular method to predict BPH fecundity of field populations and provides novel insights for insect population management.  相似文献

 We used a mapping population of 131 doubled-haploid lines, produced from a cross between an improved indica rice variety (IR64) and a traditional japonica variety (Azucena), to detect quantitative trait loci (QTLs) for resistance to the brown planthopper (BPH), Nilaparvata lugens. We evaluated the parents and mapping population with six tests that measure varying combinations of the three basic mechanisms of insect host plant resistance, i.e., antixenosis, antibiosis, and tolerance. To factor-out the effect of the major resistance gene Bph1 from IR64, the screening was done with two BPH populations from Luzon Island, The Philippines, that are almost completely adapted to this gene. A total of seven QTLs associated with resistance were identified, located on 6 of the 12 rice chromosomes. Individual QTLs accounted for between 5.1 and 16.6% of the phenotypic variance. Two QTLs were predominantly associated with a single resistance mechanism: one with antixenosis and one with tolerance. Most of the QTLs were derived from IR64, which has been shown to have a relatively durable level of moderate resistance under field conditions. Abstract Two sympatric populations of brown planthopper (BPH), one from rice and the other from Leersia hexandra were collected from each of five locations in Malaysia. All the tested malathion-resistant individuals of the rice BPH population and F1 generation (cross between malathion-resistant [usually caught on rice] and malathion-susceptible [usually caught on Leersia]) showed high esterase activity, while all malathion-susceptible individuals on L. hexandra showed low esterase activity. In the F2 generation, all the individuals tested against malathion were approximately 75% resistant and 25% susceptible and the inheritance pattern of esterase activity (high and low esterase activity) segregated in the same manner to a 3: 1 ratio. This confirms that resistance to malathion is mono-factorial and inheritance pattern of esterase activity is also linked to malathion resistance. Carboxylesterase or total esterase activity in BPH is inherited in a simple Mendelian fashion that is encoded by a single dominant gene. For the total esterase assay, average esterase activity levels in the rice-infesting population ranged from 17.64 to 19.37 nmoles 1-napthol/mg protein while that in the Leersia-infesting population ranged from 5.29 to 6.11 nmoles 1-napthol/mg protein. In terms of esterase activity, the two sympatric Nilaparvata lugens populations separated into two distinct groups. Results based on the tube color intensity test showed 96% and 98% resistant and susceptible individuals were present in the rice- and Leersia-infesting populations, respectively. In a filter paper test, the rice-infesting population had 94% with high esterase activity while the Leersia-infesting population had 96% with low esterase activity.  相似文献

Brown planthopper (BPH) [Nilaparvata lugens (Stål) (Hemiptera: Delphacidae)] is a major pest in rice [Oryza sativa L. (Poaceae)] production. Identification of resistance genes and development of BPH‐resistant varieties is an economical and effective way to control this pest. In this study, BPH honeydew excretion, survival rate, and emergence rate were used as indicators to detect the antibiotic level, whereas the relative growth rates of plant height (RH) and fresh weight (RW), and the number of days until yellowing were used to identify the level of tolerance to BPH in rice varieties. Rice varieties Swarnalata and B5, which showed high levels of antibiosis and tolerance to BPH, thus were highly resistant in the seedling bulk test; Mudgo and T12, which showed moderate resistance to the insects, had a high level of tolerance and moderate antibiosis to BPH. Varieties Rathu Heenati, ARC 10550, and Chin Saba were identified to be susceptible to BPH, showing a moderate level of tolerance and no antibiosis. In comparison to the evaluation methods of BPH resistance, the honeydew excretion and survival rate could be used to detect the antibiotic level, and the RH, RW, or leaf yellowing days could be employed as indicators to evaluate the rice varieties’ tolerance. Overall, a combined application of these indicators can effectively identify the levels of antibiosis and tolerance to BPH in rice varieties, and BPH‐resistance levels of the varieties were mainly determined by the antibiosis level. The results should help in understanding BPH‐resistance categories of rice varieties and for resistance breeding.  相似文献

褐飞虱是水稻生产中最严重的害虫之一,从野生稻中发掘抗虫基因,有利于培育具有抗虫能力强的水稻新品种。该研究通过对云南野生稻进行温室和大田抗虫鉴定以及9个已知抗褐飞虱基因的PCR鉴定,发现云南野生稻对褐飞虱表现出不同程度的抗性,尤其疣粒野生稻和药用野生稻对褐飞虱表现出高抗,可作为抗虫基因发掘的优良抗源材料;不同褐飞虱抗性的云南野生稻中含有的抗褐飞虱基因差异很大,3种野生稻中均不含Bph1和Bph18(t)抗病基因,景洪普通野生稻和元江普通野生稻可能含bph2基因,东乡普通野生稻可能含bph2、Bph15和Bph27(t)基因,疣粒野生稻中可能含bph2和bph19(t)基因,药用野生稻和药用野生稻(宽叶型)中可能含bph2和Bph6基因,药用野生稻F1中可能含bph2、Bph14和bph20(t)基因,药用野生稻F2中可能含bph2和Bph27(t)基因或者其同源基因。该研究为快速发掘利用云南野生稻中的抗虫基因奠定了理论基础。  相似文献

1 Control failures of insecticides used against the tomato leafminer Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) in Brazil led to the investigation of the possible occurrence of resistance of this insect pest to abamectin, cartap, methamidophos and permethrin. 2 The insect populations were collected from seven sites in the states of Minas Gerais, Rio de Janeiro, and São Paulo. These populations were subjected to concentration–mortality bioassays using insecticide‐impregnated filter papers. 3 We were unable to obtain a single population which provided a susceptibility standard for all insecticides tested. Therefore, the resistance levels were estimated in relation to the most susceptible population to each insecticide. Resistance to abamectin and cartap were observed in all populations when compared with the susceptible standard population, with resistance ratios ranging from 5.2‐ to 9.4‐fold and from 2.2‐ to 21.9‐fold for abamectin and cartap, respectively. Resistance to permethrin was observed in five populations with resistance ratios ranging from 1.9‐ to 6.6‐fold, whereas resistance to methamidophos was observed in four populations with resistance ratios ranging from 2.6‐ to 4.2‐fold. 4 The long period and high frequency of use of these insecticides against this insect pest suggest that the evolution of insecticide resistance on them has been relatively slow. Alternatively, the phenomenon might be widespread among Brazilian populations of T. absoluta making the finding of suitable standard susceptible populations difficult and leading to an underestimation of the insecticide resistance levels in this pest. 5 Higher levels of resistance to abamectin, cartap and permethrin are correlated with greater use of these compounds by growers. This finding suggests that local variation in insecticide use was an important cause of variation in susceptibility.  相似文献

San Jose scale, Diaspidiotus perniciosus (Comstock), is a serious pest in Chilean apple tree orchards, and a number of organophosphate insecticides were used to control them for decades. Recently, control failures with these insecticides were reported and linked to insecticide resistance development. In this study, 40 San Jose scale field populations were collected and their susceptibility to two commonly used organophosphate insecticides, that is chlorpyrifos and methidathion, was assessed. The obtained bioassay data suggest moderate levels of resistance to both insecticides when compared to a reference susceptible strain. The highest resistance ratio (RR) detected for chlorpyrifos and methidathion was 31‐fold and 11‐fold, respectively. The bioassay results suggest the occurrence of a significant cross‐resistance between both compounds. Biochemical measurements revealed a role for esterases in conferring resistance to organophosphates, but not modified acetylcholinesterase. The spatial spread and extend of insecticide resistance were also evaluated. Our result shows that no autocorrelation can be assumed, and then, insecticide resistance is caused by random factors.  相似文献

The brown planthopper (Nilaparvata lugens Stål; BPH) is one of the most serious rice pests worldwide. Growing resistant varieties is the most effective way to manage this insect, and wild rice species are a valuable source of resistance genes for developing resistant cultivars. BPH27 derived from an accession of Guangxi wild rice, Oryza rufipogon Griff. (Accession no. 2183, hereafter named GX2183), was primarily mapped to a 17-cM region on the long arm of the chromosome four. In this study, fine mapping of BPH27 was conducted using two BC1F2 populations derived from introgression lines of GX2183. Insect resistance was evaluated in the BC1F2 populations with 6,010 individual offsprings, and 346 resistance extremes were obtained and employed for fine mapping of BPH27. High-resolution linkage analysis defined the BPH27 locus to an 86.3-kb region in Nipponbare. Regarding the sequence information of rice cultivars, Nipponbare and 93-11, all predicted open reading frames (ORFs) in the fine-mapping region have been annotated as 11 types of proteins, and three ORFs encode disease-related proteins. Moreover, the average BPH numbers showed significant differences in 96–120 h after release in comparisons between the preliminary near-isogenic lines (pre-NILs, lines harboring resistance genes) and BaiR54. BPH growth and development were inhibited and survival rates were lower in the pre-NIL plants compared with the recurrent parent BaiR54. The pre-NIL exhibited 50.7 % reductions in population growth rates (PGR) compared to BaiR54. The new development in fine mapping of BPH27 will facilitate the efforts to clone this important resistant gene and to use it in BPH-resistance rice breeding.  相似文献

Six rice genotypes showing susceptible and resistant reactions to brown planthopper (BPH), Nilaparvata lugens were studied for feeding‐induced changes in defence enzymes and pathogenesis‐related (PR) proteins. The high resistant genotypes PTB 33, ADT 45 and ASD 7 and moderately resistant genotypes CO 43 and KAU 1661 recorded the greater expression of defence enzymes peroxidase, polyphenol oxidase, phenylalanine ammonia lyase, total phenol and β‐1,3 glucanase in response to N. lugens feeding at 1 day after infestation (DAI) compared with susceptible genotype TN1. The greater activity of chitinase was observed in resistant cultivars at 3 DAI and the activity was sustained for more than 1 week compared with susceptible TN1. In conclusion, the current study revealed that these defence enzymes and PR proteins might attribute to the resistance mechanisms in rice plants against BPH infestation.  相似文献

采用成虫浸叶生测法分别测定了福建省福州、漳州、龙岩、三明、南平、宁德等地烟粉虱田间种群对13种杀虫剂的抗性.结果表明,福建省6个烟粉虱田间种群对氯氟氰菊酯、甲氰菊酯、氯氰菊酯、溴氰菊酯、乙酰甲胺磷、毒死蜱产生了高水平抗性,抗性倍数分别达838.38~2460.52、244.64~834.29、116.02~266.35、81.75~124.18、425.18~875.56和54.53~78.43倍,对乐果产生了中等水平抗性,抗性倍数达14.16~17.66倍,对敌敌畏产生了中等偏低水平的抗性,抗性倍数达6.23~11.25倍,对灭多威的抗性水平较低,抗性倍数仅为4.07~5.66倍.漳州种群已对烟碱类杀虫剂产生了中等水平抗性,对吡虫啉、啶虫脒和噻虫嗪的抗性倍数分别达23.08、10.32和24.60倍,而其它地区烟粉虱种群对吡虫啉、啶虫脒和噻虫嗪的抗性水平较低,甚至不明显,抗性倍数分别仅为1.31~3.28、1.82~7.23和1.39~5.45倍.福建省各地区烟粉虱种群对阿维菌素尚未产生明显的抗药性.  相似文献

Abstract

Brown planthopper (Nilaparvata lugens Stal.) is one of the most destructive insect pests in rice. Development of near-isogenic lines (NILs) is an important strategy for genetic analysis of brown planthopper (BPH) resistance (R) genes and their deployment against diverse BPH populations. A set of 25 NILs with 9 single R genes and 16 multiple R gene combinations consisting of 11 two-gene pyramids and 5 three-gene pyramids in the genetic background of the susceptible indica rice cultivar IR24 was developed through marker-assisted selection. The linked DNA markers for each of the R genes were used for foreground selection and confirming the introgressed regions of the BPH R genes. Modified seed box screening and feeding rate of BPH were used to evaluate the spectrum of resistance. BPH reaction of each of the NILs carrying different single genes was variable at the antibiosis level with the four BPH populations of the Philippines. The NILs with two- to three-pyramided genes showed a stronger level of antibiosis (49.3–99.0%) against BPH populations compared with NILs with a single R gene NILs (42.0–83.5%) and IR24 (10.0%). Background genotyping by high-density SNPs markers revealed that most of the chromosome regions of the NILs (BC3F5) had IR24 genome recovery of 82.0–94.2%. Six major agronomic data of the NILs showed a phenotypically comparable agronomic performance with IR24. These newly developed NILs will be useful as new genetic resources for BPH resistance breeding and are valuable sources of genes in monitoring against the emerging BPH biotypes in different rice-growing countries.

As important signal molecules, jasmonates (JAs) and green leaf volatiles (GLVs) play diverse roles in plant defense responses against insect pests and pathogens. However, how plants employ their specific defense responses by modulating the levels of JA and GLVs remains unclear. Here, we describe identification of a role for the rice HPL3 gene, which encodes a hydroperoxide lyase (HPL), OsHPL3/CYP74B2, in mediating plant‐specific defense responses. The loss‐of‐function mutant hpl3‐1 produced disease‐resembling lesions spreading through the whole leaves. A biochemical assay revealed that OsHPL3 possesses intrinsic HPL activity, hydrolyzing hydroperoxylinolenic acid to produce GLVs. The hpl3‐1 plants exhibited enhanced induction of JA, trypsin proteinase inhibitors and other volatiles, but decreased levels of GLVs including (Z)‐3‐hexen‐1‐ol. OsHPL3 positively modulates resistance to the rice brown planthopper [BPH, Nilaparvata lugens (Stål)] but negatively modulates resistance to the rice striped stem borer [SSB, Chilo suppressalis (Walker)]. Moreover, hpl3‐1 plants were more attractive to a BPH egg parasitoid, Anagrus nilaparvatae, than the wild‐type, most likely as a result of increased release of BPH‐induced volatiles. Interestingly, hpl3‐1 plants also showed increased resistance to bacterial blight (Xanthomonas oryzae pv. oryzae). Collectively, these results indicate that OsHPL3, by affecting the levels of JA, GLVs and other volatiles, modulates rice‐specific defense responses against different invaders.  相似文献

The flavone, tricin (5,7,4′‐trihydroxy‐3′,5′‐dimethoxyflavone), is a valuable secondary metabolite that is common in gramineous plants, including cultivated rice (Oryza sativa). It can defend the rice plant against infestation by the brown planthopper (BPH), Nilaparvata lugens Stål, one of the most important pests of rice. This study evaluated the tricin concentration in infested and non‐infested rice plants. The results of the liquid chromatography coupled to tandem mass spectrometry (LC–MS/MS) quantitative analysis showed that the tricin concentration in rice leaves was significantly higher than in the stems and roots. The mass concentration of tricin in the leaves at the leaf stage was significantly higher than at the tiller and booting stages. The relationship between rice variety, BPH resistance and tricin concentration was investigated. There was a significant negative correlation between tricin concentration and the injury severity scores for rice varieties. Moreover, BPH infestation caused variations in tricin concentration among rice plants. High BPH infestation levels can significantly reduce the tricin concentration in rice plants. However, there is no significant effect of the length of infestation times on tricin concentrations in rice leaves. These results suggest that there may be an elicitor in BPH saliva, which is injected into rice plants during BPH infestation and triggers the tricin metabolic system. Future studies need to identify the elicitor and clarify the mechanism underlying tricin reduction in infested rice plants.  相似文献

This article reports the current status of ethiprole resistance in Nilaparvata lugens Stål in the central region of Thailand, together with the associated resistance mechanisms. A resistance survey found that a field population had developed 308.5-fold resistance to ethiprole. Further selection with ethiprole for nine generations in the laboratory led to 453.1-fold ethiprole resistance. However, following this selection procedure, the resistance of N. lugens to other insecticides decreased to about one-third of its original resistance. This result implies that there is no cross-resistance between ethiprole and other kinds of insecticides in this pest. In an in vivo study of synergisms, triphenyl phosphate (TPP) exhibited a strong synergism (SR 4.2) with ethiprole in the resistant hoppers, piperonyl butoxide (PBO) also showed significant synergistic effects with ethiprole (1.6), but diethyl maleate (DEM) did not show any obvious synergism with ethiprole (1.2). An in vitro biochemical study indicated that esterase activity increased with ethiprole resistance in N. lugens, that P450 monooxygenase activity also increased significantly with high resistance, but that glutathione S-transferase activity did not. These results reveal that increases in esterase activity and P450 monooxygenase activity cause the ethiprole resistance observed in the field populations of N. lugens. Whether the mechanisms for ethiprole resistance involve target-site sensitivity is not yet known; further molecular analysis is required. However, an analysis of insecticide cross-resistance and the insecticide application history of the resistant populations indicated that target resistance was present and that rotation between insecticides with different modes of action will provide a key countermeasure to maintain the efficacy of ethiprole.  相似文献
