石鸡 (Alectorischukar)是我国北方重要的猎鸟 ,由于栖息地片断化和人类狩猎 ,陇东黄土高原上的石鸡数量正日益减少。本文用PCR直接测序的方法 ,测定了陇东黄土高原 8个石鸡种群mtDNA控制区I区和部分II区的4 91个碱基 ,探讨其遗传多样性。 78个样本共发现 2 4个变异位点 (占所测序列的 4 .89% )和 2 5种单倍型 (占所测样本的 32 .0 5 % )。 8个种群中 ,铜川种群的序列变异率、单倍型多样性和核苷酸多样性都最高 ,分别是 0 .4 7、0 .82和 0 .0 0 2 9;而红回种群的最低 ,仅分别为 0 .10、0 .2 8和 0 .0 0 0 8,这与红回种群受奠基者效应、遗传隔离和自然选择的作用有关。 8个种群共享 1种单倍型C1,说明它们来自共同祖先 ,是 1个单系群 ,属于 1个进化显著单元 ,但它们聚成两个集群。两集群间单倍型相似性指数仅 0 .15 ,遗传距离达 0 .4 3% ,单因素方差分析显示遗传变异差异显著 (F =5 .0 2 >F0 .0 5(14 ,1) =4 .0 6 ) ,分别有 13种和 10种单倍型为两个集群所特有。基于遗传差异性 ,陇东黄土高原的石鸡应分为两个管理单元进行保护 ,尤其对遗传变异和遗传多样性最高的铜川种群应进行重点保护。  相似文献

隶属于鸟纲鸡形目雉科的石鸡是一个多型种,在我国已有7个亚种被报道,其中石鸡华北亚种是我国的特有鸟。用石鸡华北亚种(Alectoris chukar pubescens)12个地理种群112个样本的线粒体DNA控制区(mt DNA CR)1154 bp序列的信息研究了石鸡华北亚种的种群历史动态。112个样本中共发现28个变异位点,定义了29种单倍型,其中12个地理种群的50个样本共享单倍型H1,8个地理种群的16个样本共享单倍型H4。地理种群间存在较大的基因流,且种群间没有由于地理距离产生隔离的证据。负的Tajima(D=-1.336,P0.05)和Fu(Fs=-1.720,P0.05)统计检验值及错配分布的单峰模式都支持石鸡华北亚种经历了种群扩张。石鸡华北亚种大部分种群的错配分布与种群过去的扩张相一致,扩张发生在晚更新世中期的第五寒冷期(0.027—0.06 Ma)。推测其扩张的原因可能为:1)更新世期间我国北方地区没有发生大规模的冰川,2)青藏高原的隆升使我国北方干旱化和荒漠化加剧利于石鸡种群的扩散。武都(WD)位于东洋界,而石鸡是典型的古北界的鸟种,表明WD种群可能是石鸡在东洋界的一个建群种,因此需要给予充分的关注。  相似文献

【目的】为推测中国橘小实蝇Bactrocera dorsalis向北方扩散的路径和北方地区的传播来源。【方法】本研究对橘小实蝇15个地理种群的144个样本的mtDNA COⅠ序列进行测定;依据mtDNA COⅠ序列利用相关软件对橘小实蝇不同地理种群的遗传多样性、遗传分化等进行分析。【结果】在橘小实蝇144个样本的mtDNA COⅠ序列中获得了75个单倍型,其中单倍型H_7为7个地理种群所共享,北方地理种群与原发生地区种群间存在共享单倍型,绝大部分为种群独享单倍型。15个橘小实蝇地理种群总体表现出高水平的核苷酸多样性(Pi=0.02076)和单倍型多样性(Hd=0.99413),不同橘小实蝇地理种群间遗传分化较大。系统进化树显示,橘小实蝇各地理种群中的单倍型分布格局较为混杂,未形成明显的系统地理结构;系统进化树分为2支,湖北武汉、重庆北碚、福建福州、湖南衡阳、江苏南京、海南海口、河北保定、河南郑州、广东广州的地理种群聚为一支,海南三亚、广西南宁、云南丽江、湖南邵阳、山东泰安、山东临沂的地理种群聚为一支,北方新发生地理种群在两个分支中均有分布。橘小实蝇整体错配分析结果显示为单峰,表明橘小实...  相似文献

大石鸡(Alectorismagna)是中国西北部的特有种。我们测定了大石鸡兰州亚种(A.m.lanzhouensis)8个地理种群106个样本的线粒体DNA控制区5′端458bp序列,研究其种群遗传结构和遗传多样性。27个变异位点共确定25种单倍型,其中单倍型M2广泛分布,而许多单倍型为一些地方种群特有。单倍型分布沿着南北方向变化,存在明显的地理结构。8个种群中核苷酸多样性最高的是定西种群,0.0069,最低的是海原种群,0.0028;单倍型多样性最高的是武山种群,0.86,最低的是北道种群,0.52。北方种群比南方种群具有更高的遗传多样性。系统发生树和单倍型分布表明,大石鸡兰州亚种存在两个明显的分支。溯祖理论、更新世冰期和花粉支持兰州亚种起源于兰州盆地,这个盆地是其遗传多样性的中心。  相似文献

河流可能会阻碍陆生动物的迁移并导致其遗传分化,很多研究证明了河流屏障假说,但是也有少数研究不支持这个假说。本研究采集了云南省384只黄胸鼠Rattus tanezumi样本,分别来自于被5条主要河流隔开的6个地理区域。通过对mt DNA和IRBP两个基因的测序分析,检验云南省主要河流对黄胸鼠种群遗传格局的影响。共鉴定得到81个mt DNA单倍型和135个IRBP单倍型,其中很多单倍型都被2个或2个以上区域共享。AMOVA分析显示,大多数的变异源于种群内部或种群之间,只有少数变异(15%)源于6个区域之间。SAMOVA分析将mt DNA种群分成了3组,但每一组都被2个或2个以上的区域共享;对IRBP的分析则没有检测到明显的谱系地理结构。结果表明,河流屏障对黄胸鼠的遗传格局几乎无影响。  相似文献

为了明确我国北方不同地理种群甜菜夜蛾Spodoptera exigua遗传多样性与种群遗传结构,阐明该种害虫的种群历史动态,首次对采自我国北方8省17县(市)304头甜菜夜蛾样品进行mt DNA Cytb基因序列测定与分析,利用Dna SP 5.0和Arlequin 3.0软件分析种群遗传多样性、遗传结构、遗传分化与分子变异,基于MP、ML与贝叶斯法构建单倍型系统发育树,与此同时,基于Median-joining法对所有个体构建单倍型网络关系图。结果表明,在所分析的304个序列样本中,共检测出19个单倍型,其中,包括9个共享单倍型,单倍型Hap6为所有种群所共享。总群体具有较低的遗传多样性(Hd=0.422±0.035,π=0.00119±0.00011)与较小的遗传分化(F_(ST)=0.108,P0.001)。单倍型系统发育分析与网络关系图结果表明,虽然19个单倍型被分为2个分支,但各单倍型相互散布在不同种群中,未形成明显谱系地理格局。AMOVA分析表明,甜菜夜蛾遗传变异主要来自种群内(89.18%),种群间变异水平较低(10.82%)。中性检验(Tajima's D=-1.897,P0.05;Fu's FS=-4.424,P0.05)与错配分布分析表明,我国北方地区甜菜夜蛾种群曾经历过种群的近期扩张。  相似文献

采用聚合酶链式反应(PCR)和直接测序法分析雉鸡甘肃亚种的种群遗传结构。测序获得了8个种群79个样本的长819bp的线粒体细胞色素b(Cytb)基因序列,发现7个变异位点,界定了6种单倍型(Haplotype),其中Hap-1是共享单倍型,占79.75%。在8个种群中,除ZJC和CHX种群外,其它种群的核苷酸多样性和单倍型多样性都很低,种群间遗传分化小,基因流大。ZJC种群由于亚种间的基因渗透,导致其遗传多样性最高。CHX种群受徽成盆地周边山地的森林隔离作用,单倍型最多。分子方差分析(AMOVA)表明,遗传变异主要发生在种群内,占86.44%。种群间和地理种群组间差异不显著。地理种群组的核苷酸多样性(π)同形态上的变异规律一致。FS值检测表明,雉鸡甘肃亚种曾发生过快速扩张。  相似文献

研究以线粒体Cyt b基因为分子标记,对赤水河两种荷马条鳅属(Homatula)鱼类(红尾荷马条鳅和短体荷马条鳅)的遗传多样性及种群结构进行了分析;同时,结合了对在长江上游其他几个水系分布的同种鱼类进行比较,分析其生物地理学过程。遗传多样性分析结果表明,5个水系135尾红尾荷马条鳅Cyt b基因序列共检测出42个单倍型,单倍型多样性和核苷酸多样性分别为0.936和0.00493;其中,赤水河种群的分别为0.891和0.00208。3个水系52尾短体荷马条鳅Cyt b基因序列共检测出12个单倍型,单倍型多样性和核苷酸多样性分别0.821和0.01105;赤水河种群的分别为0.646和0.00390。基于ML和BI法构建的单倍型分子系统发育树结果表明,两物种各自构成单系,且得到较强支持。红尾荷马条鳅各种群地理格局分布明显,赤水河种群为并系类群位于分支基部。在短体荷马条鳅支系中,岷江与沱江两个水系的个体相互聚类在一起,而赤水河群体聚成一个分支。赤水河与其他水系不存在共享单倍型,表现了明显的隔离和差异性的地理分布格局。由于这些水系之间地理位置相距较远,推测这种格局的形成不是地质运动造成的水系隔离,而是历史时期水位的高低变化造成鱼类种群的扩散和隔离。错配分析支持赤水河两物种种群扩张的推断,但中性检验却并非全部支持,显示种群历史相对复杂。  相似文献

【目的】通过对四纹豆象Callosobruchus maculatus Fabricius不同地理种群mtDNA-Cytb和COⅠ基因部分序列进行比较, 分析其不同地理种群间的遗传分化情况, 为揭示其与生物入侵的关系及入侵过程中种群系统发育地理格局与演变机制提供依据。【方法】用PCR产物直接测序法对分别来自中国海南、喀麦隆、韩国和泰国的四纹豆象4个地理种群的mtDNA-Cytb和COⅠ序列进行测序, 运用软件MEGA3.1对四纹豆象不同地理种群mtDNA-Cytb和COⅠ序列进行序列分析, 以绿豆象C. chinensis为外群构建了不同单倍型的分子系统树。【结果】34条420 bp Cytb序列中共检测到14个多态位点和5种单倍型, 33条822 bp COⅠ序列中检测到28个多态位点和9种单倍型, 其中4种单倍型为独享单倍型, 其余为全部或部分种群的共享单倍型。AMOVA分析结果显示, 四纹豆象4个地理种群间的遗传结构差异并不明显, 遗传差异主要发生在地理种群内。对4个地理种群进行了Fst值和基因流动统计, 结果表明4个地理种群间既存在着一定数量的基因交流, 也存在一定程度的遗传分化。【结论】根据单倍型分布格局初步推测, 中国不可能是四纹豆象的原产地, 而喀麦隆有可能是原产地之一, 并且喀麦隆种群与泰国种群之间的基因交流比较充分, 而中国种群与其他种群之间的遗传分化相对较大。  相似文献

黄胫小车蝗Oedaleus infernalis Saussure是一种在我国分布广泛、 对农牧业生产危害严重的经济害虫。本文对黄胫小车蝗10个地理种群的线粒体COI基因序列进行测序和分析, 利用DnaSP 5.0和Arlequin 3.5.1.2软件对该蝗虫种群间的遗传多样性、 遗传分化程度、 基因流水平及分子变异进行了分析, 建立了单倍型贝叶斯系统发育进化树和单倍型网络图。结果表明: 在所分析的144个序列样本中, 共检测到21种单倍型, 其中1种单倍型为10个地理种群所共享。总群体的单倍型多样性指数为0.653, 各地理种群单倍型多样度范围在0.423~0.790之间。总群体和各种群的Tajima’s D检验结果皆不显著, 说明该种害虫在较近的历史上未经历群体扩张。总群体的遗传分化系数Gst为0.04436, 固定系数Fst为0.05255, 基因流Nm为9.01。AMOVA分子方差分析结果表明, 黄胫小车蝗的遗传分化主要来自种群内部, 种群间的遗传变异水平较低。各地理种群的遗传距离的大小与其地理距离间没有显著的相关性。贝叶斯系统发育进化树与单倍型网络图显示, 黄胫小车蝗各地理种群中的单倍型散布在不同的分布群中, 分布格局较为混杂, 未形成明显的系统地理结构。研究结果揭示, 黄胫小车蝗各种群间的基因交流并未受到地理距离的影响。  相似文献

This study used polymerase chain reaction (PCR) and dideoxy direct sequencing methods to analyze the genetic structure of the chukar partridge (Alectoris chukar) in the Longdong Loess Plateau, northwestern China. A total of 491 nucleotides of mitochondrial DNA from the control region gene were sequenced for 78 chukar partridges from eight different populations. The 24 variable positions defined 25 haplotypes. The high gene flow (Nm=3.75) implied little genetic differentiation between the eight populations. All the populations shared the haplotype C1, which suggested that they all came from a common ancestor. The 25 haplotypes were spread through the populations, but could be clustered into two groups. The haplotype similar index between the two groups was only 0.15, with a genetic distance of 0.43%. The genetic variation was significantly different between the two groups, which had been isolated for all of recorded history. The genetic structure of chukar partridge populations in the Plateau appears to be the result of the synergistic effect of a glacier, along with debris flow and human activities since the middle of the Pleistocene.Communicated by F. Rusty-necklaced partridges (Alectoris magna) are found in Qinghai, Ningxia and Gansu provinces of northwestern China, distributed in mountainous areas that were heavily affected by cyclical climate changes throughout the Pleistocene. We investigated the potential impact of Pleistocene range shifts on phylogeographic patterns using 458 nucleotides of mitochondrial control-region from 102 individuals sampled from 10 populations across the current range. We found 31 haplotypes defined by 33 polymorphic positions. Phylogenetic analyses revealed two robustly supported clades, one corresponding to the Chaidamu Basin and the other including all individuals from all other populations. At the population level we also found significant evidence of differentiation. The phylogeographic structure of the rusty-necklaced partridge is consistent with the "phylogenetic discontinuity, spatial separation" pattern outlined by Avise et al. [Avise, J.C., Arnold, J., Ball, R.M., Bermingham, E., Lamb, T., Niegel, J.E., Reeb, C.A., Saunders, N.C., 1987. 在六盘山地区发现鹑类的两种鸟类石鸡和大石鸡进行杂交,本实验采用PCR和RFLP的方法,分析了六盘山西侧20个大石鸡和东侧36个石鸡的mtDNA基因,发现宁夏海原5个和甘肃庄浪8个的大石鸡具有石鸡的基因型,说明这可能是雌性石鸡与雄性大石鸡杂交的结果,在六盘山东侧石鸡种群中没有发现大石鸡的基因型,其基因是从石鸡到大石鸡的单向流动。杂交后代的形态与大石鸡相似且体型比双亲都大。这种渐渗杂交可能是杂交个体与雄性大石鸡回交的结果。根据分子钟推测,这种不对称的基因流动,可能是由于冰期隔离后次级相遇形成的。杂交种可与雄性大石鸡回交。虽然还需要加大样本量来进一步研究这种不对称基因流动,但是持续的渐渗杂交会导致大石鸡基因的灭绝  相似文献

Two hundred and sixty eight DNA sequences (hypervariable region 1 of the mitochondrial control region) were obtained from chimpanzees ( Pan troglodytes ) in 19 natural populations within the range of the easternmost subspecies, P. t. schweinfurthii. Methods of phylogenetic reconstruction were applied at both the haplotype and population levels. Chimpanzee haplotypes do not sort into location-specific clades on any haplotype trees, indicating that the subspecies is free of major phylogeographic subdivisioning. Trees of populations in which geographic structure was imposed on the data lacked phylogenetic resolution in that interpopulational relationships were poorly supported statistically. These results indicate either a near simultaneous origin for the chimpanzee populations sampled, or an obscuring of interpopulational phylogenetic relationships by gene flow. In contrast, area cladograms of the forests from which chimpanzees were sampled (constructed using lists of endemic taxa) were robust and statistically well-supported. Chimpanzee population history is apparently decoupled from the history of the forests which the populations inhabit. Eastern chimpanzee data are also used to draw phylogenetic and molecular evolutionary comparisons to humans.  相似文献

Recent advances in molecular ecology allow better assessment of the role of ecological and evolutionary processes in determining the dynamics of genetic diversity in natural populations. The chukar partridge ( Alectoris chukar ) is abundant and continuously distributed in the mesic Mediterranean regions of Israel, becoming scarce and patchily distributed in the arid Negev Desert. This demographic transition occurs across a main environmental transition, the Northern Negev ecotone. Populations of chukars along this gradient have been shown to differ from other populations in morphology, bilateral asymmetry, and in allozyme variation. The aim of this study is to infer whether historical (i.e. secondary contact of formerly vicariant populations) or recurrent demographic events (i.e. restriction to gene flow across the ecotone) have shaped the observed patterns of genetic diversity in chukar populations in Israel. We analysed mtDNA control-region sequences in 216 chukars collected from 28 localities (five distinct geographical regions). Results show that (1) average haplotype diversity is high ( h  = 0.93) while haplotype divergence is low (π= 0.8%), suggesting that extant populations diversified recently; (2) mtDNA gene diversity does not significantly differ across the distribution range; and (3) haplotype distribution differs among the sampling locations ( amova ; P  < 0.00031). Mantel test and autocorrelation analyses indicate that isolation-by-distance may explain the geographical partitioning of genetic diversity. These patterns have likely been produced by complex dynamics between past fragmentation of northern vs. intermediate and southern populations. Late Pleistocene climatic changes, fostering cycles of range contraction and expansion, may have led to genetic differentiation in allopatry, followed by restricted gene flow across secondary contact zones.  相似文献

Mediterranean red-legged (Alectoris rufa) and rock (Alectoris graeca) partridge populations are affected by genetic pollution. The chukar partridge (Alectoris chukar), a species only partly native to Europe, is the most frequently introgressive taxon detected in the genome of hybrid partridges. Both theoretical (evolutionary) and practical (resources management) matters spur to get insight into the geographic origin of the A. chukar hybridizing swarm. The phenotypic A. rufa populations colonizing the easternmost part of the distribution range of this species, the islands of Elba (Italy) and Corsica (France), were investigated. The analysis of both mitochondrial (mtDNA: Cytochrome-b gene plus Control Region: 2,250 characters) and nuclear (Short Tandem Repeats, STR; Random Amplified Polymorphic DNA, RAPD) genomes of 25 wild (Elba) and 20 captive (Corsica) partridges, disclosed spread introgression of chukar origin also in these populations. All mtDNA haplotypes of Elba and Corsica partridges along with those we obtained from other A. rufa (total, = 111: Italy, Spain, France) and A. graeca (= 6, Italy), were compared with the mtDNA haplotypes of chukars (= 205) sampled in 20 countries. It was found that the A. chukar genes detected in red-legged (= 43) and rock partridges (= 4) of Spain, France and Italy as well as in either introduced (Italy) or native (Greece, Turkey) chukars (= 35) were all from East Asia. Hence, a well-defined geographic origin of the exotic chukar genes polluting the genome of native Mediterranean A. rufa and A. graeca (inter-specific level) as well as A. chukar (intra-specific level), was demonstrated.  相似文献

采用聚合酶链式反应(PCR)和mtDNA细胞色素b基因314 bp核苷酸序列测定等方法,证明在大石鸡与山石鸡的异域种群之间存在广泛的差异.然而采自六盘山地区与山石鸡相邻分布的大石鸡种群的样本中却存在山石鸡的mtDNA基因型,这种基因型与其它大石鸡个体的基因型有4.4%核苷酸差异.在与山石鸡相邻分布的大石鸡种群中有25%的个体具有山石鸡的mtDNA基因型,而采自六盘山接触地带以西约200 km处兰州市郊的大石鸡标本中则没有发现这种基因型.此发现支持山石鸡和大石鸡之间存在或曾经有过杂交的假设.作者还提出了一种限制性内切酶分析方法,可以快速地检测这种mtDNA基因型.在此之前还没有过关于山石鸡和大石鸡之间基因流动的报道.  相似文献
