木本曼陀罗内生真菌抗菌活性的筛选研究   总被引:1,自引:0,他引:1  
从木本曼陀罗植物内生真菌中筛选高效抗菌活性的菌株。选择与人类和植物相关的36株病原微生物,分别对分离自木本曼陀罗(Datura arborea L.)植物的内生真菌77株进行了发酵代谢产物的抗菌活性筛选研究。结果显示:对细菌病原菌、皮肤致病真菌、植物致病真菌有抑制作用的内生真菌分别有24,9,17株,其中5株内生真菌对10种以上的供试病原菌有明显的抑制作用,活性最好的1株对20种病原菌有较强抑制活性,最大抑菌圈直径达48mm。这说明木本曼陀罗植物内生真菌抗菌能力较强,抗菌谱较广。  相似文献

通过抗菌活性初步筛选,从采自云南元江县的印楝(Azadirachta indica A.Juss)植物茎和果实中已分离到的372株内生真菌中筛选出80株作为复筛菌株,经显微形态特征观察鉴定为5目、6科、29个属。选择16种病原微生物作为指示菌检测复筛菌株发酵产物的抗菌活性,结果表明,其中29株内生真菌对细菌、植物病原真菌和皮肤致病真菌中的一种或多种病原微生物有抑制生长作用,活性菌株比例占复筛菌株的36.25%,并显示种群多样性,其中7株内生真菌显示较强的广谱抗菌作用,活性较好的菌株主要分布在曲霉属和交链孢属。  相似文献

从云南元江野生仙人掌的肉质茎中分离出28株内生真菌,经形态学分类鉴定归于3个目,3个科,10个属。同时选择30种病原微生物作为指示菌进行抑菌试验,研究其抑菌活性。结果表明,3株元江仙人掌内生真菌分别对细菌、植物病原真菌和皮肤致病真菌多种病原微生物有较为明显的抑制生长作用。  相似文献

为研究药用植物金荞麦、款冬及黄皮树中内生菌的抗菌活性,筛选具有广谱抗菌效果的内生菌。以导致人畜及植物致病的15种病原微生物为拮抗菌株,采用改进K-B纸片法、生长速率法及琼脂二倍稀释法进行抑菌试验。结果从三种药用植物中共分离到内生菌38株,其中FE-R9、TE-R7、PE-S11三株内生真菌发酵液提取物对10种人畜指示菌的最低抑菌浓度(M IC)分别在7.50~15.00 g.L-1、3.13~12.50 g.L-1、0.39~15.00 g.L-1之间。FE-R9、TE-R7、PE-S11对多种病原微生物具有显著抑制生长的作用,这可为寻找天然抗菌物质提供资源。  相似文献

从三尖杉,南方红豆杉及香榧中分离出172株内生真菌,对其进行抗菌活性检测,结果表明共90株内生真菌对一种或多种植物病原真菌,如红色面孢霉(Neurospora sp.),木霉(Trichoderma sp.),镰刀菌(Fusarium sp.)等有抑制作用,来自三尖杉、南方红豆杉和香榧的抗菌活性菌株比例分别为40%,54.2%及57.1%。其中平板抑菌圈直径大于15mm的高抗菌株有35株。按Ainsworth等鉴定系统和方法进行鉴定,具有抗菌活性的内生真菌主要分布于心青霉属、镰孢菌属等18个属中。  相似文献

为进一步了解贵州黔西南薏苡(Coix lacryma-Jobi)可培养内生真菌的多样性组成及其抗菌活性。通过组织块分离法,选取薏苡根、叶和种仁为分离对象,进行内生真菌分离,并通过形态学观察,分子生物学特征对所分离内生真菌进行鉴定。采用平板菌块对峙法对薏苡内生真菌进行抗菌活性筛选。从薏苡根、叶和种仁中共分离纯化获得76株薏苡内生真菌,根据形态和分子生物学特征,它们属于10个目、14个科、26个属和1个不明属以及未知菌;以大肠杆菌(Escherichia coli)、金黄色葡萄球菌(Staphyloccocus aureus)、枯草芽胞杆菌(Bacillus subtilis)、酿酒酵母(Saccharomyces cerevisiae)为指示菌株,对薏苡内生真菌进行抗菌活性筛选。其中有25株对大肠杆菌具有抗菌活性,占分离菌株的比例为32. 9%;有34株对金黄色葡萄球菌具有抗菌活性,其比例为44. 7%;有24株对枯草芽胞杆菌具有抗菌活性,比例是31. 6%;有23株对酿酒酵母具有抗菌活性,占比例为30. 3%;对4种标准菌株都有抗菌活性的有13株,占总株数的17. 1%。薏苡的不同组织结构中存在丰富的内生真菌资源,部分内生真菌具有抑制其他微生物生长的活性,具有产生天然活性产物成分的潜力,为工业应用奠定了资源基础,具有进一步发掘和研究的价值。  相似文献

本文采用TLC-生物自显影-MTT法检测了3株滇重楼内生真菌即芬芳镰刀菌、季氏毕赤氏酵母和烟曲霉正丁醇提取物对大肠杆菌、枯草芽孢杆菌、白色念珠菌和稻瘟病菌的抗菌活性成分,以芬芳镰刀菌和季氏毕赤氏酵母菌液提取物中含有的抗菌活性成分为多.同时与打孔药剂扩散法的检测结果进行了比较,为进一步快速分离内生真菌中抗菌活性成分提供了依据.  相似文献

灯台树内生放线菌多样性及抗菌活性评价   总被引:1,自引:0,他引:1  
从云南西双版纳采集7份灯台树样品,经过表面消毒,用4种分离培养基分离得到105株内生放线菌。经16S rRNA基因序列分析鉴定,分布于7个科9个属。利用5种植物病原真菌指示菌对所有菌株的发酵液进行抗菌活性检测。结果显示,有12.4%、14.3%、11.4%、12.4%、8.6%的菌株分别对镰刀霉、疫霉、赤星霉、苹果炭疽、白色念珠菌有抗性。对3株具有广谱抗菌活性的菌株进行再次发酵和抗菌活性复筛,结果显示这3株的抗菌活性稳定,并可能含有生物碱类化合物。  相似文献

从川乌(Radix aconiti)植物的根和茎中分离获得内生真菌61株,经显微形态观察进行内生真菌的分类鉴定,同时采用纸片法进行抗菌活性筛选。结果有29株内生真菌显示抗菌活性,占菌株数的47.54%;毛壳菌属是抗菌活性菌株的优势种属;活性高且抗菌谱广的菌株有3株,主要分布在毛壳菌属和束丝菌属。结果表明:川乌植物内生真菌中广泛分布着有抗菌活性的菌株。  相似文献

Aim:  The aim of this study was to investigate the nature and bioactivities of endolichenic fungi in three abundant lichens, Pseudocyphellaria sp., Usnea sp. and Parmotrema sp. in the lower elevation of Hakgala montane forest in Sri Lanka.
Methods and Results:  Endolichenic fungal strains, fungi that live asymptomatically in the lichen thallus, much the same way as endophytic fungi live within healthy plant tissues, were isolated from three abundant lichen species, Pseudocyphellaria sp., Usnea sp. and Parmotrema sp., at Hakgala montane forest in Sri Lanka, using the surface sterilization method. Nine endolichenic fungal strains were isolated from Parmotrema sp. and Usnea sp. separately, while 11 endolichenic fungi were recovered from the lichen Pseudocyphellaria sp. Isolation of endolichenic fungus Chrysosporium sp. 2 was common to all three lichen species. Substrate utilization patterns and antifungal activities of eight endolichenic fungal species were evaluated and the results revealed that all the test fungi were able to produce at least one enzyme to utilize the test substrates. Nigrospora sp., Chrysosporium sp. 1 and 2 and Cladosporium sp. showed antifungal activities on growth of some selected plant pathogenic fungi.
Conclusions:  Endolichenic fungal strains (29) were isolated from the lichens Parmotrema sp., Usnea sp. and Pseudocyphellaria sp. in Sri Lanka. Chrysosporium sp. 2 was common in all three lichens. Some of these endolichenic fungal strains showed antifungal activities against common plant pathogenic fungi and they are capable of utilizing the substrates by producing specific enzymes.
Significance and Impact of the Study:  The diversity and prevalence of the endolichenic fungi have not been studied extensively and this is the first report of isolation and identification of endolichenic fungi in lichens available in Sri Lanka.  相似文献

从80株乳酸菌中筛选出45株具有抗灰葡萄孢霉菌活性的乳酸菌菌株,其中10株具有较强抗灰葡萄孢霉菌活性。对这10株乳酸菌菌株的抗植物致病真菌谱进行了研究,这10株乳酸菌对番茄早疫病菌,甜瓜疫霉菌,甜瓜枯萎病菌,苹果炭疽病菌的生长均有较强的抑制作用。其中1株具有广谱抗植物致病真菌活性的乳酸菌菌株BX6-4为植物乳杆菌。经番茄离体叶片接种试验发现,植物乳杆菌BX6-4的发酵液能够在体外强烈地抑制灰葡萄孢霉菌的生长。  相似文献

Several Trichoderma strains have been reported to be effective in controlling plant diseases, and the action of fungal hydrolytic enzymes is considered as the main mechanism involved in the antagonistic process. Strain Trichoderma harzianum T334 is a potential biocontrol agent against plant pathogenic fungi with the ability to produce low levels of proteases constitutively. To improve its fungal antagonistic capacity, mutagenetic program was undertaken for the construction of protease overproducing derivates. The mutant strains were obtained by means of UV-irradiation and were selected for p-fluorophenyl-alanine resistance or altered colony morphology. It was revealed by means of specific chromogenic protease substrates that both trypsin-like and chymotrypsin-like protease secretion was elevated in most of the mutant strains. The profiles of isoenzymes were different between the mutants and the wild-type strain, when examined by gel filtration chromatography. Certain mutants proved to be better antagonists against plant pathogens in in vitro antagonism experiments. This study suggests the possibility of using mutants with improved constitutive extracellular protease secretion against plant pathogenic fungi.  相似文献

Abstract A total of 17 presumptive killer yeast strains were tested in vitro for growth inhibitory and killing activity against a range of fungal pathogens of agronomic, environmental and clinical significance. Several yeasts were identified which displayed significant activity against important pathogenic fungi. For example, isolates of the opportunistic human pathogen, Candida albicans , were generally very sensitive to Williopsis mrakii killer yeast activity, whilst killer strains of Saccharomyces cerevisiae and Pichia anomala markedly inhibited the growth of certain wood decay basidiomycetes and plant pathogenic fungi. Results indicate that such yeasts, together with their killer toxins, may have potential as novel antimycotic biocontrol agents.  相似文献

目的 检测极端环境分离出的真菌拮抗临床病原真菌的活性.方法 选用极端环境分离出的24株真菌菌株上清液及菌丝体的提取液,来进行临床病原真菌(孢子丝菌和犬小孢子菌)的拮抗试验,采用M38-A2产孢丝状真菌抗真菌药物敏感性试验方案和E-test纸片扩散法.结果 抑菌圈直径≥1.0 cm,具有较强拮抗孢子丝菌活性的极端环境真菌有8株;抑菌圈直径≥2.0 cm,具有极强拮抗犬小孢子菌的活性的极端环境真菌有1株.两种临床菌株的拮抗试验,90%的拮抗结果相一致.结论 70%极端环境真菌菌株上清液及菌丝体提取液具有拮抗临床病原真菌孢子丝菌和犬小孢子菌的活性.  相似文献

This study was carried out to identify pathogenic bacteria and fungi on mistletoe (Viscum album L.) and investigate their potential use in biological control of this parasitic plant. For this purpose, a total of 48 fungal isolate and 193 bacterial strains were isolated from contaminated V. album during the summers 2005–2006. The isolated bacterial strains and fungal isolates were identified by using the Sherlock Microbial Identification System (MIS; Microbial ID, Newark) and microscopic methods, respectively. The bacterial strains that induced hypersensitive reaction (HR) on tobacco (Nicotiana tabacum L.) and fungal isolates were tested for pathogenicity on young shoots of mistletoe by using injection methods. The pathogenic bacterial strains and fungal isolates were also tested for their activity against mistletoe using spray methods. Five bacterial strains (two Burkholderia cepacia, one each of Bacillus megaterium, Bacillus pumilus and Pandoraea pulminicola) were HR and pathogenicity positive when injected but none of them when sprayed on mistletoe. When fungi were injected, 32 isolates were pathogenic but only thirteen when sprayed on mistletoe. Alternaria alternata VA?‐202, VA?‐205, VA?‐217 and Acremonium kiliense VA‐11 fungal isolates were the most effective ones and caused strong disease symptoms on mistletoe. The present study is the first report on the efficiency of potential biocontrol agents against mistletoe in Turkey.  相似文献

Double-stranded RNAs (dsRNAs) are widespread in plant pathogenic fungi, but their functions in fungal hosts remain mostly unclear, with a few exceptions. We analyzed dsRNAs from Nectria radicicola, the causal fungus of ginseng root rot. Four distinct sizes of dsRNAs, 6.0, 5.0, 2.5, and 1.5 kbp, were detected in 24 out of the 81 strains tested. Curing tests of individual dsRNAs suggested that the presence of 6.0-kbp dsRNA was associated with high levels of virulence, sporulation, laccase activity, and pigmentation in this fungus. The 6.0-kbp dsRNA-cured strains completely lost virulence-related phenotypes. This 6.0-kbp dsRNA was reintroduced by hyphal anastomosis to a dsRNA-cured strain marked with hygromycin resistance, which resulted in the restoration of virulence-related phenotypes. These results strongly suggest that 6.0-kbp dsRNA up regulates fungal virulence in N. radicicola. Sequencing of several cDNA clones derived from 6.0-kbp dsRNA revealed the presence of a RNA-dependent RNA polymerase (RDRP) gene. Phylogenetic analysis showed that this gene is closely related to those of plant cryptic viruses. Biochemical analyses suggested that the 6.0-kbp dsRNA may regulate fungal virulence through signal-transduction pathways involving cyclic AMP-dependent protein kinase and protein kinase C.  相似文献

Lipids created via microbial biosynthesis are a potential raw material to replace plant-based oil for biodiesel production. Oleaginous microbial species currently available are capable of accumulating high amount of lipids in their cell biomass, but rarely can directly utilize lignocellulosic biomass as substrates. Thus this research focused on the screening and selection of new fungal strains that generate both lipids and hydrolytic enzymes. To search for oleaginous fungal strains in the soybean plant, endophytic fungi and fungi close to the plant roots were studied as a microbial source. Among 33 endophytic fungal isolates screened from the soybean plant, 13 have high lipid content (>20 % dry biomass weight); among 38 fungal isolates screened from the soil surrounding the soybean roots, 14 have high lipid content. Also, five fungal isolates with both high lipid content and promising biomass production were selected for further studies on their cell growth, oil accumulation, lipid content and profile, utilization of various carbon sources, and cellulase production. The results indicate that most strains could utilize different types of carbon sources and some strains accumulated >40 % of the lipids based on the dry cell biomass weight. Among these promising strains, some Fusarium strains specifically showed considerable production of cellulase, which offers great potential for biodiesel production by directly utilizing inexpensive lignocellulosic material as feedstock.  相似文献
