Three species of cyprinids, i.e., Capoeta antalyensis (n = 112), C. caelestis (n = 99) and C. angorae (n = 178) were collected from the Anatolian Region of Turkey to carry out this investigation. A total of 389 specimens of Capoeta were examined for Allocreadium isoporum infections throughout the four different seasons of 2012. The prevalence and mean intensity of infection of A. isoporum was investigated in relation to the host species, the seasons across the year, host size, and sex. Two‐hundred and twenty‐two specimens of a trematode identified as A. isoporum were found in these fishes. One‐hundred seventy‐eight C. angorae were caught, and 70 A. isoporum were recorded in 8 of 178 fish examined, with prevalence and mean intensity of infection of 4.49% and 8.7% parasites per infected fish, respectively. In the 112 specimens of C. antalyensis, 73 individuals of A. isoporum were found, with a prevalence of 19.6% and a mean intensity of 3.3. As for the infection in C. caelestis, a total of 79 specimens of A. isoporum were found in 17 of the 99 fish examined, with prevalence and mean intensity of infection of 17.1% and 4.6%, respectively. Prevalence and mean intensity values in relation to host size and sex, as well as the seasonality of infections was analyzed and the results were compared with other studies. The trematode A. isoporum is reported for the first time parasitising three endemic species of Capoeta spp. in Turkey, adding to the knowledge of geographical distribution and host range of this trematode species.  相似文献

The development of molecular methodologies for targeting pathogens such as the Microsporidia has greatly improved our monitoring capabilities and initiatives. This study analyzed samples collected from five locations in Pensacola, Florida, USA for the presence of Microsporidian pathogens. To circumvent various impediments associated with water collection and filtration, we utilized biofilms as sentinels for detection of Microsporidia. We implemented membrane‐dissolution and sample purification in a single confined step followed by real‐time PCR to confirm pathogen presence. The results of this study demonstrate that microsporidia are present in environmental water sites in the Florida panhandle and that biofilms may serve as another alternative mode to circumvent filtration methods for their detection.  相似文献

Microsporidia are opportunistic pathogens in nature infecting all animal phyla. There is a potential risk of microsporidian spores transmission from urban rooks inhabiting some metropolitan cities to people through casual interactions. The aim of this study was to identify microsporidia species in the droppings of rooks in Wroclaw, Poland. A total of 15 collective sets of droppings were examined using nested‐PCR method. Amplification of ITS rRNA gene revealed the presence of Enterocytozoon bieneusi D, Peru 6, and Encephalitozoon hellem 1A genotypes. This study indicates that excreta of urban rooks can be an important source of human infection with these pathogens.  相似文献

Short‐rotation woody biomass crops (SRWC) have been proposed as a major feedstock source for bioenergy generation in the Northeastern US. To quantify the environmental effects and greenhouse gas (GHG) balance of crops including SRWC, investigators need spatially explicit data which encompass entire plantation cycles. A knowledge gap exists for the establishment period which makes current GHG calculations incomplete. In this study, we investigated the effects of converting pasture and hayfields to willow (Salix spp.) and hybrid‐poplar (Populus spp.) SRWC plantations on soil nitrogen (N) cycling, nitrous oxide (N2O) emissions, and nitrate (NO3?) leaching at six sites of varying soil and climate conditions across northern Michigan and Wisconsin, following these plantations from pre conversion through their first 2 years. All six sites responded to establishment with increased N2O emissions, available inorganic N, and, where it was measured, NO3? leaching; however, the magnitude of these impacts varied dramatically among sites. Soil NO3? levels varied threefold among sites, with peak extractable NO3? concentrations ranging from 15 to 49 g N kg?1 soil. Leaching losses were significant and persisted through the second year, with 44–112 kg N ha?1 leached in SRWC plots. N2O emissions in the first growing season varied 30‐fold among sites, from 0.5 to 17.0 Mg‐CO2eq ha?1 (carbon dioxide equivalents). N2O emissions over 2 years resulted in N2O emissions due to plantation establishment that ranged from 0.60 to 22.14 Mg‐CO2eq ha?1 above baseline control levels across sites. The large N losses we document herein demonstrate the importance of including direct effects of land conversion in life‐cycle analysis (LCA) studies of SRWC GHG balance. Our results also demonstrate the need for better estimation of spatial variability of N cycling processes to quantify the full environmental impacts of SRWC plantations.  相似文献

Replacement of fossil fuels with sustainably produced biomass crops for energy purposes has the potential to make progress in addressing climate change concerns, nonrenewable resource use, and energy security. The perennial grass Miscanthus is a dedicated energy crop candidate being field tested in Ontario, Canada, and elsewhere. Miscanthus could potentially be grown in areas of the province that differ substantially in terms of agricultural land class, environmental factors and current land use. These differences could significantly affect Miscanthus yields, input requirements, production practices, and the types of crops being displaced by Miscanthus establishment. This study assesses implications on life cycle greenhouse gas (GHG) emissions of these differences through evaluating five Miscanthus production scenarios within the Ontario context. Emissions associated with electricity generation with Miscanthus pellets in a hypothetically retrofitted coal generating station are examined. Indirect land use change impacts are not quantified but are discussed. The net life cycle emissions for Miscanthus production varied greatly among scenarios (?90–170 kg CO2eq per oven dry tonne of Miscanthus bales at the farm gate). In some cases, the carbon stock dynamics of the agricultural system offset the combined emissions of all other life cycle stages (i.e., production, harvest, transport, and processing of biomass). Yield and soil C of the displaced agricultural systems are key parameters affecting emissions. The systems with the highest potential to provide reductions in GHG emissions are those with high yields, or systems established on land with low soil carbon. All scenarios have substantially lower life cycle emissions (?20–190 g CO2eq kWh?1) compared with coal‐generated electricity (1130 g CO2eq kWh?1). Policy development should consider the implication of land class, environmental factors, and current land use on Miscanthus production.  相似文献

Pathogens and the potential risk they present to public health in recreational waters are of continual public concern. The focus of this study was a year‐long sampling campaign to document the presence of Microsporidia and protozoan pathogens in the Bayou Texar waterway in Pensacola, Florida. We used biofilms as sentinel indicators for trapping pathogens in five different locations in Pensacola, Florida. Of the 34 biofilm samples, 16 were positive for pathogens. Of these samples, 13 were positive for Enchephalitozoon spp. (mostly E. cuniculi), 11 were positive for Enterocytozoon bieneusi, and two were positive for Cryptosporidium parvum. The data demonstrate that Microsporidia were easily recovered and primarily present in water during summer months.  相似文献

Production of energy crops is promoted as a means to mitigate global warming by decreasing dependency on fossil energy. However, agricultural production of bioenergy can have various environmental effects depending on the crop and production system. In a field trial initiated in 2008, nitrate concentration in soil water was measured below winter wheat, grass‐clover and willow during three growing seasons. Crop water balances were modelled to estimate the amount of nitrate leached per hectare. In addition, dry matter yields and nitrogen (N) yields were measured, and N balances and energy balances were calculated. In willow, nitrate concentrations were up to approximately 20 mg l?1 nitrate‐N during the establishment year, but declined subsequently to <5 mg l?1 nitrate‐N, resulting in an annual N leaching loss of 18, 3 and 0.3 kg ha?1 yr?1 N in the first 3 years after planting. A similar trend was observed in grass‐clover where concentrations stabilized at 2–4 mg l?1 nitrate‐N from the beginning of the second growing season, corresponding to leaching of approximately 5 kg ha?1 yr?1 N. In winter wheat, an annual N leaching loss of 36–68 kg ha?1 yr?1 was observed. For comparison, nitrate leaching was also measured in an old willow crop established in 1996 from which N leaching ranged from 6 to 27 kg ha?1 yr?1. Dry matter yields ranged between 5.9 and 14.8 Mg yr?1 with lowest yield in the newly established willow and the highest yield harvested in grass‐clover. Grass‐clover gave the highest net energy yield of 244 GJ ha?1 yr?1, whereas old willow, winter wheat and first rotation willow gave net energy yields of 235, 180 and 105 GJ ha?1 yr?1. The study showed that perennial crops can provide high energy yields and significantly reduce N losses compared to annual crops.  相似文献

Enteric illnesses remain the second largest source of communicable diseases worldwide, and wild birds are suspected sources for human infection. This has led to efforts to reduce pathogen spillover through deterrence of wildlife and removal of wildlife habitat, particularly within farming systems, which can compromise conservation efforts and the ecosystem services wild birds provide. Further, Salmonella spp. are a significant cause of avian mortality, leading to additional conservation concerns. Despite numerous studies of enteric bacteria in wild birds and policies to discourage birds from food systems, we lack a comprehensive understanding of wild bird involvement in transmission of enteric bacteria to humans. Here, we propose a framework for understanding spillover of enteric pathogens from wild birds to humans, which includes pathogen acquisition, reservoir competence and bacterial shedding, contact with people and food, and pathogen survival in the environment. We place the literature into this framework to identify important knowledge gaps. Second, we conduct a meta‐analysis of prevalence data for three human enteric pathogens, Campylobacter spp., E. coli, and Salmonella spp., in 431 North American breeding bird species. Our literature review revealed that only 3% of studies addressed the complete system of pathogen transmission. In our meta‐analysis, we found a Campylobacter spp. prevalence of 27% across wild birds, while prevalence estimates of pathogenic E. coli (20%) and Salmonella spp. (6.4%) were lower. There was significant bias in which bird species have been tested, with most studies focusing on a small number of taxa that are common near people (e.g. European starlings Sturnus vulgaris and rock pigeons Columba livia) or commonly in contact with human waste (e.g. gulls). No pathogen prevalence data were available for 65% of North American breeding bird species, including many commonly in contact with humans (e.g. black‐billed magpie Pica hudsonia and great blue heron Ardea herodias), and our metadata suggest that some under‐studied species, taxonomic groups, and guilds may represent equivalent or greater risk to human infection than heavily studied species. We conclude that current data do not provide sufficient information to determine the likelihood of enteric pathogen spillover from wild birds to humans and thus preclude management solutions. The primary focus in the literature on pathogen prevalence likely overestimates the probability of enteric pathogen spillover from wild birds to humans because a pathogen must survive long enough at an infectious dose and be a strain that is able to colonize humans to cause infection. We propose that future research should focus on the large number of under‐studied species commonly in contact with people and food production and demonstrate shedding of bacterial strains pathogenic to humans into the environment where people may contact them. Finally, studies assessing the duration and intensity of bacterial shedding and survival of bacteria in the environment in bird faeces will help provide crucial missing information necessary to calculate spillover probability. Addressing these essential knowledge gaps will support policy to reduce enteric pathogen spillover to humans and enhance bird conservation efforts that are currently undermined by unsupported fears of pathogen spillover from wild birds.  相似文献

Previous phylogenetic studies of the grape downy mildew pathogen, Plasmopara viticola, revealed five cryptic species in eastern North America that differed in their host range and geographic distribution. Preliminary comparative studies also documented differences in temperature responses during infection between certain cryptic species, indicating the biological relevance of knowing which cryptic species of the pathogen are present in a given region. However, limited information is available regarding the presence, prevalence and dynamics of cryptic species of P. viticola in the southeastern United States. Here, 301 P. viticola isolates obtained from cultivated grape species in five distinct grape‐growing regions of Georgia and Florida were subjected to cleaved amplified polymorphic sequence analysis and multilocus sequencing (internal transcribed spacer region of the rDNA, actin and β‐tubulin) to identify cryptic species and infer phylogenetic relationships. Three cryptic species, P. viticola clade aestivalis (Pva), clade vinifera (Pvv) and clade vulpina (Pvu), were identified in Georgia, whereas two, Pva and Pvv, were found in Florida; all three cryptic species are reported here for the first time in Georgia, whereas Pva is reported for the first time in Florida. Pva was the most prevalent cryptic species (72.1% of isolates) and was distributed widely from the North Georgia Mountains to Mid‐Florida, whereas Pvv (27.2%) and Pvu (0.7%) were found only in the Coastal Plain region of the two states. Interestingly, Pvu was obtained from French American hybrid Blanc du Bois and could be subcultured on Vitis vinifera Chardonnay, suggesting a broader host range than only the wild species Vitis vulpina reported previously.  相似文献

Dimethylsulfoniopropionate (DMSP) is produced by many species of marine phytoplankton and has been reported to provide a variety of beneficial functions including osmoregulation. Dinoflagellates are recognized as major DMSP producers; however, accumulation has been shown to be highly variable in this group. We explored the effect of hyposaline transfer in Gambierdiscus belizeanus between ecologically relevant salinities (36 and 31) on DMSP accumulation, Chl a, cell growth, and cell volume, over 12 d. Our results showed that G. belizeanus maintained an intracellular DMSP content of 16.3 pmol cell?1 and concentration of 139 mM in both salinities. Although this intracellular concentration was near the median reported for other dinoflagellates, the cellular content achieved by G. belizeanus was the highest reported of any dinoflagellate thus far, owing mainly to its large size. DMSP levels were not significantly affected by salinity treatment but did change over time during the experiment. Salinity, however, did have a significant effect on the ratio of DMSP:Chl a, suggesting that salinity transfer of G. belizeanus induced a physiological response other than DMSP adjustment. A survey of DMSP content in a variety of Gambierdiscus species and strains revealed relatively high DMSP concentrations (1.0–16.4 pmol cell?1) as well as high intrageneric and intraspecific variation. We conclude that, although DMSP may not be involved in long‐term (3–12 d) osmoregulation in this species, G. belizeanus and other Gambierdiscus species may be important contributors to DMSP production in tropical benthic microalgal communities due to their large size and high cellular content.  相似文献

Banana (Musa spp.) is severely damaged by Fusarium wilt caused by Fusarium oxysporum f. sp. cubense (Foc). Biocontrol by inducing systemic resistance has been considered as one of the most important strategies to improve plant health. Very few studies have investigated appropriate reference gene selection for RT‐qPCR (quantitative real‐time polymerase chain reaction) analysis suitable for conditions of systemic activated resistance. In this study, we assessed over a time‐course the expression of seven candidate reference genes (EF1, TUB, ACT1, ACT2, L2, RPS2 and RAN) for Cavendish cultivar Brazilian (Musa spp. AAA) and dwarf banana cultivar Guangfen No. 1 (Musa spp. ABB) that were inoculated by Bacillus subtilis strain TR21 and Foc. We choose these plants because they are commonly planted in Southern China. Expression stability of the candidate genes was evaluated using various software packages (GeNorm, NormFinder and BestKeeper). L2 and TUB genes displayed maximum stability in Guangfen No. 1. In Brazilian, ACT1 and TUB were the most stable genes. To further validate the suitability of the reference genes identified in this study, the expression of pathogenesis‐related 1 (PR1) gene under TR21 and Foc strains Foc004/Foc009 treatments was also studied. Identified reference genes in this work that are most suitable for normalizing gene expression data in banana under Fusarium wilt resistance induction conditions will contribute to the understanding of disease resistance mechanisms induced by biocontrol strains in banana.  相似文献

Samples of sugarcane leaves were collected from different commercial fields and breeding stations in Egypt. Aetiology of sugarcane phytoplasma disease was investigated using nested PCR. Phytoplasma‐specific primers (P1/P7 and R16F2n/R16R2) were used to amplify a fragment of the 16S rRNA gene. Sequencing and restriction fragment length polymorphism analyses revealed that the tested phytoplasmas belonged to the 16SrI (aster yellows phytoplasma) group. Phylogenetic analyses of 60 screened accessions of 16S ribosomal RNA gene sequences of Candidatus phytoplasmas comprising those collected from Egypt (this study) and those extracted from GenBank showed that they split into two distinct clusters. All the phytoplasmas form a stable phylogenetic subcluster, as judged by branch length and bootstrap values of 100% in the 16S group cluster. Results of phylogenetic analyses indicated that these phytoplasmas are closely related and share a common ancestor. Conversely, based on the analysis of the 16S‐23S region, examined isolates segregated into four different clusters suggesting a notable heterogeneity between them. These results are the first record of the presence of phytoplasma in association with sugarcane yellow leaf in Egypt.  相似文献

As the human population has increased, so too has the demand for biotically pollinated crops. Bees (Apoidea) are essential for pollen transfer and fruit production in many crops, and their visit patterns can be influenced by floral morphology. Here, we considered the role of floral morphology on visit rates and behaviour of managed honey bees (Apis mellifera) and wild bumble bees (genus Bombus), for four highbush blueberry cultivars (Vaccinium corymbosum L.). We measured five floral traits for each cultivar, finding significant variation among cultivars. Corolla throat diameter may be the main morphological determinant of visit rates of honey bees, which is significantly higher on the wider flowers of cv. ‘Duke’ than on ‘Bluecrop’ or ‘Draper’. Honey bees also visited cv. ‘Duke’ legitimately but were frequent nectar robbers on the long, narrow flowers of cv. ‘Bluecrop’. Bumble bees were infrequent (and absent on cv. ‘Draper’) but all observed visits were legitimate. Crop yield was highest for the cultivar with the highest combined (honey bee + bumble bee) visit rate, suggesting that aspects of floral morphology that affect pollinator visit patterns should be considered in crop breeding initiatives.  相似文献

Drought entails important effects on tree physiology, which may result in short‐ to long‐term radial growth decreases. While the majority of studies have focused on annual drought‐related variability of growth, relatively little is known about sustained growth decreases following drought years. We apply a statistical framework to identify climatic factors that induce abrupt growth decreases and may eventually result in tree mortality. We used tree‐ring data from almost 500 standing dead trees and 200 living trees in eight sites of the Swiss network of strict forest reserves, including four of the most important Central European tree species (Abies alba, Picea abies, Fagus sylvatica and Quercus spp.). First, to assess short‐term growth responses to drought under various climate and site conditions, we calculated correlations and linear mixed‐effects models between ring‐width indices (RWIs) and drought based on the Standardized Precipitation Evapotranspiration Index (SPEI). Second, to quantify drought effects on abrupt growth decreases, we applied distributed lag nonlinear models (DLNMs), which account for both delayed effects and the nonlinear relationship between the SPEI and the occurrence of abrupt growth decreases. Positive correlations between RWIs and the SPEI indicated short‐term growth responses of all species, particularly at arid sites. Results of the DLNMs revealed species‐specific growth responses to drought. For Quercus spp., abrupt growth decreases were more likely to occur several years following severe drought, whereas for P. abies, A. alba, and F. sylvatica abrupt growth decreases started frequently immediately in the drought year. We conclude that the statistical framework allows for quantifying the effects of drought intensity on the probability of abrupt growth decreases, which ultimately contributes to an improved understanding of climate impacts on forest community dynamics.  相似文献

Maintaining effective immune response is an essential factor in the survival of small populations. One of the most important immune gene regions is the highly polymorphic major histocompatibility complex (MHC). We investigated how a population bottleneck and recovery have influenced the diversity and selection in three MHC class II loci, DLA‐DRB1, DLA‐DQA1 and DLA‐DQB1, in the Finnish wolf population. We studied the larger Russian Karelian wolf population for comparison and used 17 microsatellite markers as reference loci. The Finnish and Karelian wolf populations did not differ substantially in their MHC diversities ( = 0.047, P = 0.377), but differed in neutral microsatellite diversities ( = 0.148, P = 0.008). MHC allele frequency distributions in the Finnish population were more even than expected under neutrality, implying balancing selection. In addition, an excess of nonsynonymous compared to synonymous polymorphisms indicated historical balancing selection. We also studied association between helminth (Trichinella spp. and Echinococcus canadensis) prevalence and MHC diversity at allele and SNP level. MHC‐heterozygous wolves were less often infected by Trichinella spp. and carriers of specific MHC alleles, SNP haplotypes and SNP alleles had less helminth infections. The associated SNP haplotypes and alleles were shared by different MHC alleles, which emphasizes the necessity of single‐nucleotide‐level association studies also in MHC. Here, we show that strong balancing selection has had similar effect on MHC diversities in the Finnish and Russian Karelian wolf populations despite significant genetic differentiation at neutral markers and small population size in the Finnish population.  相似文献

In plant–ant–hemipteran interactions, ants visit plants to consume the honeydew produced by phloem‐feeding hemipterans. If genetically based differences in plant phloem chemistry change the chemical composition of hemipteran honeydew, then the plant's genetic constitution could have indirect effects on ants via the hemipterans. If such effects change ant behavior, they could feed back to affect the plant itself. We compared the chemical composition of honeydews produced by Aphis nerii aphid clones on two milkweed congeners, Asclepias curassavica and Asclepias incarnata, and we measured the responses of experimental Linepithema humile ant colonies to these honeydews. The compositions of secondary metabolites, sugars, and amino acids differed significantly in the honeydews from the two plant species. Ant colonies feeding on honeydew derived from A. incarnata recruited in higher numbers to artificial diet, maintained higher queen and worker dry weight, and sustained marginally more workers than ants feeding on honeydew derived from A. curassavica. Ants feeding on honeydew from A. incarnata were also more exploratory in behavioral assays than ants feeding from A. curassavica. Despite performing better when feeding on the A. incarnata honeydew, ant workers marginally preferred honeydew from A. curassavica to honeydew from A. incarnata when given a choice. Our results demonstrate that plant congeners can exert strong indirect effects on ant colonies by means of plant‐species‐specific differences in aphid honeydew chemistry. Moreover, these effects changed ant behavior and thus could feed back to affect plant performance in the field.  相似文献
