Allozyme and morphological differentiation patterns within the Carex flava complex in Fennoscandia were described by means of multivariate analyses. As a result of these studies, and considering also data on ecology, distribution and hybridization, the following taxa were recognized: C. flava with no infraspecific taxa, C. lepidocarpa with two subspecies, ssp. lepidocarpa and ssp. jemtlandica, C. demissa with no infraspecific taxa, and C. oederi with three varieties, var. oederi , var. bergrothii , and var. pulchella. Carex lepidocarpa ssp. jemtlandica was allozymically divergent from ssp. lepidocarpa , indicating a long period of isolation of the two subspecies. A poor morphological separation in some localities is apparently due to secondary contacts in connection with recolonization of Fennoscandia after the last ice age, resulting in the formation of fertile hybrids and back-crosses. The subdivision of C. oederi into three varieties is supported by ecological data and it is suggested that they have originated by parallel ecotype formation under similar selective regimes in different areas. However, the varietal limits are not supported by differentiation in neutral (allozyme) characters. A phylogenetic hypothesis was constructed by means of neighbour-joining based allozyme data and it was indicated that C. demissa may be more closely related to C. lepidocarpa than to C. oederi. Previous treatments that have reduced the C. flava complex to one or two species may have been based on inappropriate species concepts, too coarse methods of analysis of morphological data, ignorance of patterns of co-occurrence and hybridization, or a combination of these factors.  相似文献

Chromosome numbers from 17 Finnish populations of six taxa in the Carex flava complex were examined. Special attention was given to taxa claimed to be endemic to Fennoscandia. The meiotic counts for C. flava n = 30, C. viridula var. viridula n = 35 and var. pulchella n = 35 are in accordance with previous reports. The counts for C. jemtlandica n = 34, C. bergrothii n = 35, and C. kotilaini n = 34 are new. The hybrid C. flava × kotilaini reveals severely disturbed meiosis with many uni- and multivalents. The results are briefly discussed in relation to morphological data. Carex jemtlandica should evidently be included in C. lepidocarpa. Carex bergrothii and C. kotilaini belong to the polymorphic C. viridula , but the latter probably consitute a specific cytotype.  相似文献

The morphological variation in the Carex flava complex (sect. Ceratocystis ) in Finland, particularly taxa described by Alvar Palmgren as endemic to northern Europe, was studied using numerical analyses. Discriminant analyses were used to maximize the separation of the taxa. However, the separation of the taxa described by Palmgren was weak. The taxonomic treatment of the C. flava complex in Finland based on morphology, ecology, distribution and hybridization of the taxa is presented. Four species are recognized: Carex flava (the lectotype is selected here), C. lepidocarpa, C. demissa and C. viridula. C. lepidocarpa includes ssp. lepidocarpa and ssp. jemtlandica . In the polymorphic C. viridula , three taxa are recognized: var. viridula , var. bergrothii and var. pulchella . A key to the taxa is presented. The distribution of the taxa in Finland is presented, and their ecology is briefly discussed.  相似文献

The organization of genetic variation in Phlox drummondii was investigated using both allozyme electrophoresis and quantitative genetics. Variation at five polymorphic enzyme loci was characterized in nine populations, and variation in 16 morphological and life-history characters was examined using an analysis of full- and half-sibs in seven populations. Significant levels of genetic variation were found at enzyme loci and for metric characters. Significant heritabilities were observed for 15 of the 16 characters examined. Genetic differences among populations were revealed both by Nei's genetic distance and by phenotypic differences, summarized by discriminant analysis. Partitioning variance in allozyme frequencies among hierarchical levels of genetic organization indicated that 94% of this variance lay within populations, 4% between populations within varieties, and 2% between varieties. Partitioning phenotypic variance for metric characters indicated that 73% lay within populations, 24% lay between populations within varieties, and 3% lay between varieties. Thus, both electrophoretic and metric characters indicated that despite extensive genetic differentiation among populations, most of the evolutionary potential of the species lies within populations.  相似文献

Morphometric and allozyme data were collected from 11 populations of Dactylorhiza majalis s.l. in the Ardennes (Belgium, France) and North Jutland (Denmark). All study populations were growing in obviously acid habitats. Genetic distances were calculated and illustrated by UPGMA dendrograms, while principal components analyses (PCAs) were used to summarize morphological variation patterns. Based on the PCAs, the probability of each character to distinguish correctly between the main groups of populations was estimated. The study populations from the Ardennes and from the Danish island of Læsø seem to agree with populations from Sweden previously studied by Hedrén—and to belong to D. majalis subsp. sphagnicola (comb. nov.). It should be noted that allozyme data suggest a polytopic origin of this taxon. The study populations from the Danish region of Thy differ morphologically from the others and exhibit much less genetic variation. They are described as a new subspecies, D. majalis subsp. calciñigiens. The evolution and biogeography of the two taxa are discussed, and brief taxonomic accounts are provided.  相似文献

Lathyrus vernus (L.) Bernh. is a diploid, long-lived perennial and insect-pollinated herb with no special adaptation to long-distance dispersal. It occurs on neutral soil in deciduous forests throughout western Eurasia. Due to specific habitat preferences,L. vernus has a fragmented distribution with isolated populations. We investigated allozyme variation at eleven loci in 20 populations ofL. vernus from one geographically central region (the Czech Republic and the Slovak Republic) and two geographically marginal regions (southern and central Sweden) in the species present-day distribution. There was a clear differentiation between the three regions and the genetic distance between the populations was highly correlated with geographic distance. The total genetic diversity (HT) was 0.354. The proportion of genetic diversity due to differentiation between regions, and to differentiation between populations within regions, accounted for 10% each. There was no difference in level of genetic diversity between the three regions. No significant difference in level of genetic diversity was found between small and large populations. The genetic diversity inL. vernus may either be a result of the long generation-time of the species or peculiarities in the post-glacial migration species, e.g. survival only in refugia far east of the sampled populations and/or migration as a continuous process not involving founder-events.  相似文献

Within Scandinavia, Carex capitata s.l, is usually divided into two taxa: the lowland to lowalpine C. capitata s.s., and the alpine C. arctogena. The systematic status of these taxa is uncertain, reflected by their taxonomic treatment in different floras. In this study, allozymes were used to assess the degree of genetic differentiation between and within taxa. Ten populations of Carex capitata s.s. and eight of C. arctogena, from Norway and Sweden, were included in the study. Sixteen loci in eleven different enzyme systems were analysed: five were variable within or between taxa, three could be interpreted in all plants. In all, 20 different alleles were found. Four of these alleles were confined to C. capitata s.s., and two to C. arctogena. In C. capitata s.s., three loci (18%) were polymorphic, while all loci were monomorphic in C. arctogena. The number of alleles per polymorphic locus was 2.2 in C. capitata s.l. The taxa had completely different alleles at two loci {Pgi-2, Pgm-1). Nei's genetic identity was 0.86 between taxa, and ranged between 0.95 and 1 in pairwise comparisons of populations of Carex capitata s.s., while it was 1 in all comparisons between populations of C. arctogena. The distinction of C. capitata s.s. and C. arctogena as separate species agrees well with the amount of differentiation found between other congeneric species. It is suggested that the low genetic variation observed in both taxa is due to ancient founder events. The difference in genetic variability between taxa may be due to differences in immigration history: whereas C. capitata s.s. may have colonized Scandinavia from several close refugia after the last glaciation, C. arctogena may have reached Scandinavia after long-distance dispersal of a limited number of propagules.  相似文献

Patterns of variation within and between Carex gynodynama and C. mendocinensis were investigated by studying allozyme and chromosome variation in natural populations and structural variation using herbarium specimens. Multivariate analyses of structural data demonstrated that C. gynodynama is clearly distinct from C. mendocinensis, and that sterile specimens similar to C. mendocinensis are intermediate between that species and C. gynodynama. The mean genetic distance between the two species, based on allozyme phenotypes at 17 enzyme-coding loci, was 0.22 ± 0.12. The sterile putative hybrids had the expected heterozygous pattern at three enzyme-coding loci at which the parental species were fixed for different alleles. Chromosome numbers are reported for the first time for both species and their putative hybrid. Carex mendocinensis had a different number in each of the three populations examined with n = 28, n = 29, or n = 30. Chromosome counts from one population of C. gynodynama revealed five plants with n = 25 and one with n = 26. Putative hybrids from this population exhibited irregular pairing at meiosis with 2n = ca. 55–57. Patterns of allozyme variation also suggest that C. mendocinensis has an outcrossing or mixed mating system but that C. gynodynama is an inbreeding species. Carex gynodynama exhibited very little variation in structure, habitat, or at the enzyme-coding loci examined, suggesting that it may have experienced a genetic bottleneck relatively recently. Carex mendocinensis had higher levels of variation both within and between populations at enzyme-coding loci and in structural features. This pattern of variation and a geographic distribution centered in serpentine areas of the Klamath–Siskiyou region, with disjunct smaller populations in serpentine areas farther south, suggest that C. mendocinensis once may have been a more widespread species.  相似文献

We studied the structure of genetic variation (at both ramet- and genet-level) and clonal diversity within and among populations in the four closely related arctic clonal sedges Carex bigelowii, C. ensifolia, C. lugens and C. stans by use of allozyme markers. Compared to other sedges and arctic plants, the studied taxa all had high levels of genetic variation, both within populations and taxa. These taxa contained most of the total gene diversity (H(T)) within populations and a small part of the diversity among populations (G(ST) ranged 0.05--0.43). Carex bigelowii had genetic variation (H(S) = 0.173, mean for populations) at a comparable level to other outbreeding arctic plants and to other widespread, rhizomatous and mainly outbreeding Carex species. In contrast, C. ensifolia (H(S) = 0.335), C. lugens (H(S) = 0.339) and C. stans (H(S) = 0.294) had within-population variations that were higher than in most other studied Carex species and for arctic plants in general. Genetic variation was not related to any tested environmental variable, but it was lower in areas deglaciated only 10,000 years BP compared to areas deglaciated 60,000 years BP or not glaciated at all during the Weichselian. All the populations were multiclonal, except for two populations of C. stans that were monoclonal. In contrast to genetic variation, clonal diversity decreased with latitude and did not differ between areas with different times of deglaciation. In accordance with previous studies, C. bigelowii and C. lugens were found to be outbreeding, while C. ensifolia and C. stans had mixed mating systems.  相似文献

Allozyme variation at 25 genetic loci was assayed in twelve indigenous cultivars (land races) of barley from Iran. For these loci the average probability that two gametes drawn at random from one population would differ at a locus was 0.082. In the collection as a whole, this measure of genetic diversity was 0.161, so that about half the total diversity was distributed between populations. Of the total of 31 allozyme variants, about half were common (frequency >0.10) in only one or two regions. The results were compared with diversity estimates based on spike morphological polymorphisms; with allozyme polymorphism in two composite crosses (CC21 and CC34) of cultivated barley; and with previous results for allozyme polymorphism in Israel populations of wild barley. In the land race samples, the diversity of spike types was a poor index of allozyme diversity. The total allozyme diversity in this collection of land races was intermediate between the moderate levels in composite crosses and the high levels in Hordeum spontaneum from Israel. These results emphasize the role of land races as valuable genetic resources for plant breeding. They support sampling strategies which, by taking samples of moderate size from many sites, emphasize the collection of locally common alleles, as against strategies framed to capture the rare, conspicuous morphological variant by intensive or biased sampling, or strategies based on collecting the extreme ecotypes from a cline. Coelomactra antiquata is a commercially exploited bivalve, and its populations have been severely declining in the coast of China during the last two decades. In order to provide guidelines for conservation strategies and management programs, four populations of C. antiquata, representing the most important populations in China, were analysed morphologically and genetically to reveal the genetic structure by using 7 polymorphic allozyme loci and 12 morphological variables. The mean allele richness ranged from 2.79 to 3.53, while the mean heterozygosity ranged from 0.326 to 0.519. All genetic distance metrics revealed high level of genetic differentiation between the three northern populations and one southern population. The marked genetic differentiation can be explained by the freshwater outflow of Yangtze River and upwelling of Zhejiang province. The morphological analyses were in accordance with the allozyme data, which also disclosed a high degree of differentiation between the northern and southern populations. The results obtained in this study indicate that the northern and southern populations in C. antiquata should be treated as separate units for conservation management.  相似文献

Although sexual reproduction appears to be rare in many clonal plant species, the majority of clonal species show similar levels of genetic variation to non-clonal plant species. Aegopodium podagraria is a noxious garden weed and has become a successful invader in many natural habitats. Earlier studies have demonstrated population differentiation in life-history traits between different A. podagraria populations in Sweden. In this study, we used three methods to assess genetic variation in this species. Using analyses of molecular markers (chloroplast DNA PCR-RFLP and allozyme electrophoresis) we did not detect any variation between different A. podagraria clones. However, a multivariate analysis of leaflet shape in five populations from central Sweden revealed considerable variation both within and between populations. The variation found in leaflet shape is suggested to be genetically based because the leaves were collected from plants grown from seed under similar conditions in a common garden environment. These relatively high levels of variation within and between populations indicate that there is likely to be repeated seedling recruitment within established populations. 1  相似文献

Factors leading to endemism, and the evolutionary implications of endemism, can be explored by studying closely related taxa with variously restricted distributions. Such a model is provided by Carex section Ceratocystis (Cyperaceae); Carex cryptolepis, Carex sp. nov., and C. lutea are North American endemics with broad, restricted, and highly restricted distributions, respectively. The prediction that levels of genetic diversity are a consequence of distribution size was tested within a phylogenetic context using population level genetic variation at 18 allozyme loci. In contrast to expectations, mean proportion of loci polymorphic, number of alleles per polymorphic locus, and expected heterozygosity were significantly greater in C. lutea than either C. cryptolepis or Carex sp. nov. Although the possibility of a shift in breeding system, past introgression, or progenitor-derivative relationships could explain the relatively high levels of variation observed in C. lutea, these were dismissed on the basis of allozyme and nuclear ribosomal sequence data. We conclude that C. lutea maintains levels of genetic diversity typical of caespitose carices despite its narrow endemism and that the low levels of genetic variation in C. cryptolepis and Carex sp. nov. are likely the result of population fluctuations during Pleistocene glacial-interglacial cycles.  相似文献

Allozyme investigation of the five Cimicifuga taxa in Korea was conducted to assess genetic and clonal diversity within populations and genetic divergence among populations and taxa. Levels of allozyme variation maintained in Korean Cimicifuga taxa were comparable to those for most herbaceous perennials. In general, samples excluding copies of the same multilocus genotype maintained higher levels of genetic diversity than the total samples within populations. Copies of homozygous genotypes at several loci resulting from clonal spread lead to decreased levels of genetic diversity within populations, indicating that clonal reproduction found in Cimicifuga affects population genetic structure. In general, more widely distributed species such as C. dahurica and C. japonica harbored higher levels of allozyme diversity than the other taxa examined. Although two varieties of C. heracleifolia are geographically and reproductively isolated, the genetic and clonal structure of var. bifida seems to resemble var. heracleifolia, indicating that the two varieties may have had a similar evolutionary history. However, the allozyme data strongly indicate that the two morphological types (Groups I and II) of C. simplex should be treated as separate species.  相似文献

Buffalograss, Buchloë dactyloides, is widely distributed throughout the Great Plains of North America, where it is an important species for rangeland forage and soil conservation. The species consists of two widespread polyploid races, with narrowly endemic diploid populations known from two regions: central Mexico and Gulf Coast Texas. We describe and compare the patterns of allozyme and RAPD variation in the two diploid races, using a set of 48 individuals from Texas and Mexico (four population samples of 12 individuals each). Twelve of 22 allozyme loci were polymorphic, exhibiting 35 alleles, while seven 10-mer RAPD primers revealed 98 polymorphic bands. Strong regional differences were detected in the extent of allozyme polymorphism: Mexican populations exhibited more internal gene diversity (He= 0.20, 0.19) than did the Texan populations (He= 0.08, 0.06), although the number of RAPD bands in Texas (n= 62) was only marginally smaller than in Mexico (n= 68). F-statistics for the allozyme data, averaged over loci, revealed strong regional differentiation (mean FRT=+ 0.30), as well as some differentiation among populations within regions (mean FPR=+ 0.09). In order to describe and compare the partitioning of genetic variation for multiple allozyme and RAPD loci, we performed an Analysis of Molecular Variance (AMOVA). AMOVA for both allozyme and RAPD data revealed similar qualitative patterns: large regional differences and smaller (but significant) population differences within regions. RAPDs revealed greater variation among regions (58.4% of total variance) than allozymes (45.2%), but less variation among individuals within populations (31.9% for RAPDs vs. 45.2% for allozymes); the proportion of genetic variance among populations within regions was similar (9.7% for RAPDs vs. 9.6% for allozymes). Despite this large-scale concordance of allozyme and RAPD variation patterns, multiple correlation Mantel techniques revealed that the correlations were low on an individual by individual basis. Our findings of strong regional differences among the diploid races will facilitate further study of polyploid evolution in buffalograss.  相似文献

The Lycaeides butterfly species complex in North America consists of two nominal, morphologically defined species. These butterflies are ecologically diverse and appear to be distributed as a geographically complex mosaic of locally differentiated populations that may be undergoing adaptive radiation. We asked whether patterns of molecular genetic variation within the species complex are congruent with currently recognized morphological species and whether the distribution of molecular variation is consistent with the hypothesis that Pleistocene climate changes contributed to the process of differentiation within the genus. Variation in the form of the genitalia from 726 males from 59 populations clearly distinguishes both species with only six populations containing morphologically intermediate or ambiguous individuals. However, partitioning of molecular variance in a 236 bp section of the mitochondrial AT-rich region from 628 individuals (57 populations) surveyed using single strand conformation polymorphism analysis (SSCP) indicates that only 26% of the total genetic variation is distributed along nominal species boundaries as defined by morphology. Instead, three phylogeographical groups were detected, represented by three major haplotype clades, which account for 90% of the total genetic variance. Pleistocene glaciations appear to have fostered divergence during glacial maxima, while post-glacial range expansions created opportunities for gene exchange and reticulation along suture zones between geographical groups. Data presented here allow us to make inferences about the history of the species complex. However, evidence of ancestral polymorphism and reticulation limit our ability to define species boundaries based on mitochondrial DNA sequence variation.  相似文献

An unusual chromosomal hybrid zone of the house mouse, Mus musculus domesticus, exists in Upper Valtellina, Northern Italy, consisting of four Robertsonian (Rb) races and the standard (all-acrocentric, or 2n = 40) race, all hybridizing freely within 10 km2. The hybrid zone in Valtellina provides an excellent opportunity to study the role of Rb fusions in reproductive isolation and speciation. This hybrid zone has already been well studied for the distribution of Rb fusions and the fertility of hybrids, but in order to understand the dynamics of the zone, a basic understanding of the origin and genetic similarity of the chromosomal races is necessary. This paper presents the results of three different methods of measuring genetic differentiation: multivariate analysis of morphological traits and analyses of allozyme variation and mitochondrial DNA sequences. The standard race is clearly distinguishable from the three Rb races by all three methods, but the Rb races are not distinguishable from one another. This provides strong evidence for our previous suggestions that the well-established Rb races in Valtellina are closely related, and that the standard race was introduced into the valley more recently from a distant source. The fact that the Rb races are indistinguishable is also consistent with our hypothesis that a within-village speciation event involving two of the races (Hauffe & Searle, 1992) was a recent occurrence. The low level of allozyme heterozygosity among the Rb races suggests that these populations are the products of at least one bottleneck. The present article substantially extends earlier studies and provides the first detailed morphological and molecular analysis of this complex hybrid zone.  相似文献

Relationships between allozyme differentiation, habitat variation and individual reproductive success were examined in local populations of a perennial herb, Gypsophila fastigiata, on the Baltic island of Öland (Sweden). Relatively little (c. 2%) of the total allozyme diversity in this largely outcrossing species is explained by differentiation between sites tens of kilometres apart. The low level of geographic differentiation suggests that gene flow between sites is, or has recently been, extensive. Yet the component of allozyme diversity due to differentiation between plots (only tens of meters apart) within sites is 3 times larger than the between-site component of diversity. Allozyme variation, especially at the Pgi-2 locus, is significantly associated with habitat variation within sites. Different allele x habitat combinations for the Pgi-2 locus are associated with differences in individual reproductive fitness. Differential selection in different local habitats may thus contribute to the fine-scale structuring of genetic diversity within sites.  相似文献

The warm-temperate vegetation of Korea, currently limited to southern coastal areas, shifted during the Last Glacial Maximum (LGM) towards glacial refugia, putatively located in southern Japan. We hypothesized two scenarios of post-glacial re-colonization of warm-temperate species in relation to current levels of genetic diversity within their populations. If extant Korean populations originated from a single source (a single glacial refugium), we expect significantly lower levels of genetic diversity relative to those from Japan due to founder effects. Alternatively, if they were derived from multiple source populations, levels of genetic diversity within Korean populations will not be significantly reduced compared to those of Japanese ones. To test which of these scenarios is more likely, we investigated the patterns of genetic diversity in 14 populations (seven from southern Korea and seven from southern Japan) of the broad-leaved evergreen tree Machilus thunbergii, employing 11 allozyme loci. High levels of genetic variation in M. thunbergii were found both in southern Korea and southern Japan, with a considerable genetic homogeneity not only between the two regions but also between populations within the two regions. These results suggest a pattern of re-colonization after the LGM fitting the second scenario (immigration from multiple refugia), probably through multiple waves and/or with large founder populations.  相似文献
